Abstract:
Abstract Pulp and paper bio-sludge (PPBS) has low economic value and is therefore often composted or incinerated. The purpose of this work is to evaluate the feasibility of using PPBS to breed black soldier fly (Hermetia illucens) larvae (BSFL), so improving resource efficiency and supplying protein and fat to the animal feed market. BSFL were reared on PPBS in a climate chamber on a laboratory scale in order to assess nutrient deficiency, inhibition and whether the BSFL recycle the PPBS well or not. The PPBS used came from a chemi-thermomechanical pulp/groundwood pulp mill. The effect of supplementing nutrient solution added either into the PPBS or as a free liquid surface were studied. Prepupae dry weight, survival rate until prepupae stage and residual dry PPBS were measured. The addition of reference diet leachate into the PPBS did not have a significant impact on the weight of the prepupae (2.0 mg) but the survival rate was significantly higher (16.0%). On the other hand, the addition of reference diet leachate as a free liquid surface had a significant impact on both the weight of the prepupae (4.8 mg) and the survival rate (25.7%). The bio-conversion and PPBS—reduction was as best 0.4 and 3.2% respectively. This study has proven that the nutrients in PPBS are not readily available to the larvae. Addition of reference diet leachate increase the survival rate which opens up for possibilities of co-digestion. However, it is clear that under the tested conditions, BSFL is not recycling PPBS well and BSFL as a method for recycling of PPBS need further research. Graphic Abstract

Abstract:
Protoplasts derived from a non- regenerable soybean cell line provide an excellent experimental system for studying plant preprophase bands (PPBs). Cultured protoplasts developed PPBs in high frequencies, permitting a detailed analysis of PPB development. From observations of thousands of PPBs, six distinct developmental stages were identified. This classification should prove useful in recognizing developmental stages of PPBs and in comparing results among different tissues and species. Perinuclear fluorescence appeared when PPBs were well developed. It consisted of microtubule strands radiating from the nuclear surface at its early stages and more extensive arrays on the surface at later stages. All protoplast cultures from 0.5 day to 5 days had a PPB index (ratio between number of PPBs and number of phragmoplasts) greater than unity, suggesting that in this cell line a high frequency of PPBs is apparently not related to the potential for organized growth (e.g. embryogenesis). Different methods of quantifying the occurrence of PPBs were evaluated. The PPB value (ratio between % of cells with PPB and the mitotic index) was compared with the PPB index and results indicate that it is a useful new parameter for studies of the distribution of PPBs in plant cells.

In order to improve the budget system, a number of approaches and techniques have been adopted in public sector organizations such as Program and Performance Budgeting System (PPBS), Performance Based Budgeting System (PBBS) and Zero-Based Budgeting (ZBB). The present study is an extension in the line of very few researches which had been conducted in developing countries in regard to implementation of “One Budgetary Approach” which is known as “Program and Performance Budgeting System PPBS”. The study concentrates its focus on the support which it may find in case of “budgetary format” is adopted by the “Public Sector Organizations” in the kingdom of Saudi Arabia. The study explores several dimensions such as familiarity, acceptability and adoptability of PPBS, “degree of contribution of Accounting System followed by “public sector organizations” to adopt PPBS”, the benefits that might be realized and the obstacles that probably might be faced if this approach of budgeting is adopted by Public Sector Organizations” in Al-Kharj region. The study came up with the following main findings; there is a fair familiarity and understanding of PPBS by financial managers and accountants working in the “public sector organizations”, the accounting system followed by “public sector organizations” contributes to adopt PPBS successfully, there are certain benefits could be obtained while adopting PPBS by public sector organizations, and finally certain obstacles have been discovered which are standing as stumbling-stone to adopt PPBS in “public sector organizations” in Al-Kharj region effectively.

‘Pseudo–profound bullshit’ (PPBS) is a class of meaningless statements designed to appear profound. Profundity ratings for PPBS have been found to be negatively related to analytical thinking and positively related to epistemically suspect beliefs (e.g. belief in the paranormal). Conceptually similar traits within the Openness/Intellect (O/I) domain form a simplex, whereby traits are arranged along a single dimension from intelligence to apophenia (i.e. observing patterns or causal connections were none exist). Across two studies (total N = 297), we attempted to replicate the O/I simplex and determine how it relates to perceiving PPBS as profound. Participants completed questionnaires measuring traits from the O/I simplex and provided profundity ratings for PPBS. Profundity ratings of PPBS tended to correlate negatively with intelligence and positively with apophenia. The association with intelligence generally reflected a greater ability to discriminate the profound from the pseudo–profound, whereas the association with apophenia reflected poorer discrimination in Study 1, with less conclusive results in Study 2. In both studies, the O/I simplex was closely replicated. The results suggest a link between the O/I domain and perceiving PPBS as profound and tentatively support the theory that intelligence may protect against apophenia. © 2018 European Association of Personality Psychology

Abstract:
Pancreatic ductal adenocarcinoma (PDAC) is one of the most aggressive cancers with a low response to treatment and a five-year survival rate below 5%. The ineffectiveness of treatment is partly because of an immunosuppressive tumor microenvironment, which comprises tumor-supportive pancreatic stellate cells (PSCs). Therefore, new therapeutic strategies are needed to tackle both the immunosuppressive PSC and pancreatic cancer cells (PCCs). Recently, physical cold atmospheric plasma consisting of reactive oxygen and nitrogen species has emerged as a novel treatment option for cancer. In this study, we investigated the cytotoxicity of plasma-treated phosphate-buffered saline (pPBS) using three PSC lines and four PCC lines and examined the immunogenicity of the induced cell death. We observed a decrease in the viability of PSC and PCC after pPBS treatment, with a higher efficacy in the latter. Two PCC lines expressed and released damage-associated molecular patterns characteristic of the induction of immunogenic cell death (ICD). In addition, pPBS-treated PCC were highly phagocytosed by dendritic cells (DCs), resulting in the maturation of DC. This indicates the high potential of pPBS to trigger ICD. In contrast, pPBS induced no ICD in PSC. In general, pPBS treatment of PCCs and PSCs created a more immunostimulatory secretion profile (higher TNF-α and IFN-γ, lower TGF-β) in coculture with DC. Altogether, these data show that plasma treatment via pPBS has the potential to induce ICD in PCCs and to reduce the immunosuppressive tumor microenvironment created by PSCs. Therefore, these data provide a strong experimental basis for further in vivo validation, which might potentially open the way for more successful combination strategies with immunotherapy for PDAC.

Abstract:
Abstract Purpose Based on recent scientific evidence, bariatric surgery is more effective in the management of morbid obesity and related comorbidities than conservative therapy. Pylorus preserving surgical procedures (PPBS) such as laparoscopic single-anastomosis duodeno-jejunal or duodeno-ileal bypass with sleeve gastrectomy are modified duodenal switch (DS) surgical techniques. The duodeno-jejunal bypass liner (DJBL) is a novel surgical method in the inventory of metabolism focused manual interventions that excludes duodeno-jejunal mucosa from digestion, mimicking DS procedures without the risk of surgical intervention. The aim of this article is to summarize and compare differences between safety-related features and weight loss outcomes of DJBL and PPBS. Methods A literature search was conducted in the PubMed database. Records of DJBL-related adverse events (AEs), occurrence of PPBS-related complications and reintervention rates were collected. Mean weight, mean body mass index (BMI), percent of excess of weight loss (EWL%), percent of total weight loss (TWL%) and BMI value alterations were recorded for weight loss outcomes. Results A total of 11 publications on DJBL and 6 publications on PPBS were included, involving 800 and 1462 patients, respectively. The baseline characteristics of the patients were matched. Comparison of DJBL-related AEs and PPBS-related severe complications showed an almost equal risk (risk difference (RD): −0.03 and confidence interval (CI): −0.27 to 0.21), despite higher rates among patients having received endoscopic treatment. Overall AE and complication rates classified by Clavien-Dindo showed that PPBS was superior to DJBL due to an excess risk level of 25% (RD: 0.25, CI: 0.01–0.49). Reintervention rates were more favourable in the PPBS group, without significant differences in risk (RD: −0.03, CI: −0.27 to 0.20). However, PPBS seemed more efficient regarding weight loss outcomes at 1-year follow-up according to raw data, while meta-analysis did not reveal any significant difference (odds ratio (OR): 1.08, CI: 0.74–1.59 for BMI changes). Conclusion Only limited conclusions can be made based on our findings. PPBS was superior to DJBL with regard to safety outcomes (GRADE IIB), which failed to support the authors’ hypothesis. Surgical procedures showed lower complication rates than the incidence of DJBL-related AEs, although it should be emphasized that the low number of PPBS-related mild to moderate complications reported could be the result of incomplete data recording from the analysed publications. Weight loss outcomes favoured bariatric surgery (GRADE IIB). As the DJBL is implanted into the upper gastrointestinal tract for 6 to 12 months, it seems a promising additional method in the inventory of metabolic interventions.

Abstract:
Background: Incidence and prevalence of diabetes have been steadily increasing with a raise of global prevalence about 8.5%. The major types of diabetes are differentiated by insulin deficiency versus insulin resistance. SGLT2 inhibitors are a new class of drugs that act by inhibiting glucose reabsorption in the proximal renal tubules. Remogliflozin a prodrug of remogliflozin, which is a potent and selective sodium-glucose co-transporter-2 inhibitor was used for the study. The objective of the study was to evaluate the efficacy and safety of Remogliflozin etabonate in reducing HbA1C and serum glucose in type II diabetics.Methods: This was a prospective observational study was done for 3 months. HbA1C, FBS and PPBS readings were noted and then the subjects were introduced to Remoglifozin 100 mg twice a day. At the end of 12 weeks HbA1C, FBS and PPBS were noted. The obtained data was analysed for its efficacy and safety.Results: The study included 22 male subjects and 28 females. Before Remoglifzoin was given the mean HbA1C level was 8.23± 0.798, mean FBS was 179± 29.79 and PPBS was 299.38± 24.21. Remoglifozin 100 mg was given and the mean HbA1C level was 7.52± 0.765, mean FBS was 166.30± 32.13 and mean PPBS was 249.46± 18.21. Post 3 months of Remoglifzoin induction a reduction in HbA1C, FBS and PPBS was seen.Conclusions: This study concludes Remoglifozin etabonate of 100 mg when given twice daily reduced the HbA1C levels, FBS and PPBS levels significantly.

Purpose To extend our knowledge about factors influencing early vocabulary development for infants with cochlear implants (CIs), we investigated the impact of positive parenting behaviors (PPBs) from the Indicator of Parent Child Interaction, used in parent–child interactions during everyday activities. Method Implantation age for the sample recruited from CI clinics in Australia ranged from 6 to 10 months for 22 children and from 11 to 21 months for 11 children. Three observation sessions at three monthly intervals were coded for use of PPBs. Children's productive vocabulary, based on the MacArthur–Bates Communicative Development Inventories parent checklist, was collected approximately 6 and 9 months later. A repeated-measures negative binomial generalized linear mixed-effects model was used to investigate associations between the total PPBs per session, covariates (maternal education, gender, and time since implant), and the number of words produced. In follow-up analyses with the PPBs entered separately, variable selection was used to retain only those deemed informative, based on the Akaike information criterion. Results As early as Session 1, associations between the PPBs and vocabulary were identified. Time since implant had a positive effect. For different sessions, specific PPBs (descriptive language, follows child's lead, and acceptance and warmth) were identified as important contributors. Conclusions Complementing previous findings, valuable information was identified about parenting behaviors that are likely to impact positively the early vocabulary of infants with CIs. Of importance is providing parents with information and training in skills that have the potential to help create optimal contexts for promoting their child's early vocabulary development.

BACKGROUND The relationship between liver enzymes like alanine aminotransferase (ALT), aspartate aminotransferase (AST), Alkaline phosphatase (ALP) and diabetes has been studied, but the results of these are inconsistent. Several prospective studies have reported that ALT was associated with incident diabetes. We wanted to study the levels of serum alanine aminotransferase (ALT), serum aspartate aminotransferase (AST) and serum alkaline phosphatase (ALP) in patients of Type 2 DM. METHODS This is a hospital based observational study which was conducted in Narayan Medical College and Hospital, Jamuhar, Sasaram, Rohtas, Bihar. Alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), glycosylated haemoglobin (HbA1c), FBS, PPBS, total bilirubin, and hepatitis B surface antigen (HBsAg) were determined in all study participants. RESULTS The mean value was 96.35, 67.37, 152.78 and 1.098 for AST, ALT, ALP and total bilirubin. On multivariate analysis for effect of FBS, PPBS, HbA1c on the value of AST weak correlation was found with R square of 0.11. On the effect of FBS, PPBS, HbA1c on the value of ALT we found weak correlation with an R square of 0.079. CONCLUSIONS There is a weak correlation between deranged liver enzyme (AST, ALT and ALP) with HbA1c, FBS and PPBS. But still, liver functions should be monitored in diabetic patients. The purpose of this report is to determine whether the products of the Planning, Programming and Budgeting System (PPBS) are worthwhile, they must be measured against some form of output. The prepositioned War Reserve (PWR) of the Marine Corps is a measure of sustainability: a desired output of the PPBS. This thesis investigated the PPBS, the Marine Corps programming methodology and ammunition requirement generation to determine whether these processes artificially constrain ammunition purchases. This thesis suggests that the constraints placed on ammunition requirements are related to the lack of long-range strategic goals, inadequate planning in the PPBS and the inherent weaknesses of program budgeting. Abstract:
This thesis provides an analysis of the causal factors leading to the increased influence and authority of the CINCs in the defense resource decision and allocation process. A discussion is provided on the various Joint Chiefs of Staff (JCS) reforms which lead to the enhancement of the CINCs' role in the Planning, Programming, and Budgeting System. Major issues, constraints, control and implementation problems currently confronting the CINCs are explored. A brief summary of the initiatives begun by Deputy Secretary of Defense (DEPSECDEF) Taft to increase the involvement of the CINCs in the defense programming process also is provided. The policy issues related to implementation of the Goldwater-Nichols Department of Defense (DoD) reorganization Act of 1986 are reviewed along with some of the positive and negative aspects of the increased demand for CINC participation in PPBS. Conclusions and recommendations for further study are furnished

During the early 1960s, the US Department of Defense, under Secretary Robert McNamara implemented for the first time in national government a Planning-Programming and Budgeting System (PPBS) in order to improve effectiveness and efficiency in defence program management. McNamara sought improved effectiveness through a formal five-year program designed to reduce costs. He also sought efficient methods of managing joint service strategy coordination, requirements' analysis and planning, and improved alignment between the choice of requirements and the size and nature of the acquisition program. The Australian Defence Organisation (ADO) and the UK Ministry of Defence later sought to implement their own forms of PPBS. Recently, both have introduced program management innovations that seek to achieve more reliably effective and efficient outcomes. The thesis has reviewed program management theory with a particular focus on its implementation challenges relating to strategic management, program review, personnel management and program coordination. It has sought to answer the research problem: Which specific management designs could offer better outcomes for Australian defence technological edge programs? The thesis' central proposition is that the greatest opportunities for improving defence program outcomes occur when classic PPBS concepts are implemented within a Program Management System that incorporates Zero- Base budgeting (ZBB), Management by Objectives (MBO), and Matrix structural systems. All of these systems, either alone or in combination seek to enhance program quality, scheduling, financial management and evaluation. The research used in-depth case study research based on qualitative data found within a selection of recent Australian National Audit Office reports, and other public records. The central proposition is subjected to dynamic reliabilityrelated contingency analysis and evaluation. The thesis concludes with the proposition that if managers were to implement a contingency based integrated mixture of the above-mentioned systems they could expect improved technological edge program outcomes.

Die Genexpressionsmaschinerie in Chloroplasten und Mitochondrien und die ihrer prokaryotischen Vorläufer sind konserviert. Innerhalb eines bakteriellen Grundgerüsts entwickelte sich darüber hinaus ein komplexer RNA-Metabolismus. In der vorliegenden Arbeit wird eine neue Klasse kleiner RNAs (15-50nt) mit plastidärem und mitochondrialen Ursprung beschrieben. Diese kurzen RNAs überlappen mit Bindestellen von RNA-bindenden Proteinen, die mRNAs gegen exonukleolytischen Verdau beschützen. Diese stabilisierende Funktion wird vermutlich hauptsächlich von PPR (Pentatricopeptid repeat) Proteinen und verwandten Proteine bewerkstelligt. Die kleinen RNAs repräsentieren dabei minimale nuklease-resistente Bereiche, sogenannte RNA-Bindeprotein footprints. Solche footprints finden sich in fast jedem intergenischen Bereich, der Prozessierung aufweist. Durch transkriptomweite Untersuchungen von kleinen RNAs in Mutanten von RNA-Bindeproteinen konnte für diese eine Reihe von Bindestellen identifiziert werden. Nuklease-resistente kleine RNAs fehlen in entsprechenden Mutanten. Der Vergleich neu identifizierter Ziele einzelner RNA-Bindeproteine führte dabei zu neuen Erkenntnissen über den Mechanismus der RNA-Erkennung durch PPR Proteine. Im Gegensatz zu Plastiden befinden sich kleine RNAs in Mitochondrien überwiegend an den 3‘ Enden von Transkripten, deren Stabilität vermutlich maßgeblich von diesen RNA-Bindeproteinen beeinflusst wird. Für das chloroplastidäre Ribonukleoprotein CP31A konnte gezeigt werden, dass es an der Stabilisierung der ndhF mRNA beteiligt ist. Die Interaktion mit der ndhF mRNA, die eine zentrale Komponente des NDH-Komplexes kodiert, wird dabei über die 3‘ untranslatierte Region vermittelt. Zusätzlich konnte gezeigt werden, dass CP31A die Stabilität einiger antisense Transkripte beeinflusst. Weiterhin wurden zehn neue Cytidin Desaminierungungen durch die Analyse von RNA-Seq Datensätzen in der Modellpflanze Arabidopsis thaliana identifiziert.
Chloroplasts and mitochondria are of endosymbiotic origin. Their basic gene expression machineries are retained from their free-living prokaryotic progenitors. On top of this bacterial scaffold, a number of organelle-specific RNA processing steps evolved. In this thesis, a novel class of organelle-specific short (15-50nt) RNAs is described on a transcriptome-wide scale. The small RNAs are found at binding sites of PPR (Pentatricopeptide repeat) and PPR-like proteins, which protect mRNAs against exonucleolytic decay. The small RNAs represent minimal nuclease resistant RNAs, so called PPR footprints. Small RNAs were identified in almost every intergenic region subjected to intergenic processing. This finding suggests that accumulation of processed transcripts in plastids is mostly due to protection by highly specific RNA-binding proteins. Small RNA sequencing identified a number of nuclease insensitive sites missing in mutants of RNA-binding proteins. Analysis of multiple small RNAs representing target sites of single PPR proteins expands the knowledge of target specificity. In mitochondria, accumulations of small RNAs predicts that at least two thirds of mitochondrial mRNAs are stabilized by RNA-binding proteins binding in their 3’UTR. In sum, small organellar RNAs turned out to be instrumental in elucidating the hitherto enigmatic intercistronic processing of organellar RNAs and allowed novel insights into the function of the dominant family of organellar RNA binding proteins, the PPR proteins. A chloroplast ribonucleoprotein CP31A is shown to be involved in stabilization of an mRNA for a central component of the NDH-complex by interaction with its 3’UTR. In addition, CP31A represents the first factor described that influences the accumulation of chloroplast antisense transcripts. Finally, ten novel plastid C to U RNA-editing sites were identified in the model plant Arabidopsis thaliana, using a novel RNA-Seq based approach.

Pneumatic positioning systems (PPS) are widely used in fields were mechatronics applications exist. In order to improve the performance and robustness, either by designing new controllers or studying other characteristics of PPSs under different load conditions, a test bench with a load simulator is required. Hydraulic load simulators (HLS) are one of the most significant applications of hydraulic force control systems. The HLS aims to control the forces according to static and dynamic requirements. Typically, a force control system interacts with the external environment such that a good performance and robustness are not easily achieved. Different approaches are being discussed in the literature consisting on increasing the system compliance. System compliance can be controlled or modified in active or passive ways. Active compliance is obtained by force-based feedback control, allowing more flexibility to perform tasks where compliance is required. On the other hand, passive compliance is typically obtained by introducing mechanical elastic components between the actuator and the environment. In this paper, the influence of PPS compliance in the force controlled tuning of HLS is discussed. Because HLS is applying a controlled force against to a very compliant system like a pneumatic system, stable force responses could be expected. However, the dynamic interaction between the HLS with the PPS effected the force responses. This paper focuses in analyzing these effects, highlighting the design of PI and QFT controllers for force control based on these characteristics. Both of Systems are analyzed mathematically via simulation.

The force-moment capabilities of branch-redundant planar-parallel manipulators (PPMs) are investigated. A previously developed explicit methodology for generating the force-moment capabilities of redundant PPMs is used on three different PPM architectures. The results for the 4-RRR, 4-RPR, 4-PRR layouts (where the underline denotes the actuated joint in each branch) are presented and discussed. For the revolute-actuated layouts, it was shown that the force-moment capabilities for the 4-RRR were in general better than those of the 4-RPR for the chosen manipulator parameters. The presented analysis is an effective tool for designing PPMs to determine the largest forces and moments that can be applied at any point within the workspace.
