Abstract:
Approved for public release; distribution is unlimited.
360-degree feedback is a personal development and appraisal tool designed to quantify the competencies and skills of fellow employees by tapping the collective experience of their superiors, subordinates, and peers. Substantially better than the hierarchical, single-source assessments employees are familiar with, this multi-source system provides participants with a comprehensive interpretation of their performance from numerous perspectives within the organization. The objective of this thesis is to develop a 360-degree feedback system tailored specifically for the Dean position, Graduate School of Business and Public Policy, Naval Postgraduate School. This thesis presents a literature review, a case description involving the Dean position, and discusses a series of interviews conducted with key groups of organizational stakeholders. With the results of this research confirming the need for and potential content of a feedback system, this thesis culminates by presenting 360- degree feedback procedures and documents created specifically for the Dean position.

Simon, Elliot. "Determination of an Optimum Sector Size for Plan Position Indicator Measurements using a Long Range Coherent Scanning Atmospheric Doppler LiDAR." Thesis, Uppsala universitet, Institutionen för geovetenskaper, 2015. http://urn.kb.se/resolve?urn=urn:nbn:se:uu:diva-295081.

Abstract:
As wind energy plants continue to grow in size and complexity, advanced measurement technologies such as scanning Doppler LiDAR are essential for assessing site conditions and prospecting new development areas.   The RUNE project was initiated to determine best practices for the use of scanning LiDARs in resource assessments for near shore wind farms. The purpose of this thesis is to determine the optimum configuration for the plan position indicator (PPI) scan type of a scanning LiDAR. A task specific Automated Analysis Software (AAS) is created, and the sensitivity of the integrated velocity azimuth process (iVAP) reconstruction algorithm is examined using sector sizes ranging from 4 to 60 degrees. Further, a comparison to simultaneous dual Doppler measurement is presented in order to determine the necessity of deploying two LiDARs rather than one.    DTU has developed a coordinated long range coherent scanning multi-LiDAR array (the WindScanner system) based on modified Leosphere WindCube 200S devices and an application specific software framework and communication protocol. The long range WindScanner system was deployed at DTU’s test station in Høvsøre, Denmark and measurement data was collected over a period of 7 days. One WindScanner was performing 60 degree sector scans, while two others were placed in staring dual Doppler mode. All three beams were configured to converge atop a 116.5m instrumented meteorological mast.   A significant result was discovered which indicates that the accuracy of the reconstructed measurements do not differ significantly between sector sizes of 30 and 60 degrees. Using the smallest sector size which does not introduce systematic error has numerous benefits including: increasing the scan speed, measurement distance and angular resolution.   When comparing collocated dual Doppler, sector scan and in-situ met-mast instrumentation, we find very good agreement between all techniques. Dual Doppler is able to measure wind speeds within 0.1%, and 60 degree sector scan within 0.2% on average of the reference values. For retrieval of wind direction, the sector scan approach performs particularly well. This is likely attributable to lower errors introduced by the assumption of flow field homogeneity over the scanned area, in contract to wind direction which tends to be more non-uniform. For applications such as site resource assessments, where generally accurate 10 minute wind speed and direction values are required, a scanning LiDAR performing PPI scans with a sector size of between 30 and 38 degrees is recommended. The laser’s line of sight path should be directed parallel to the predominant wind direction and at the lowest elevation angle possible.

Abstract:
The study sheds light on pupils individual plans of development and portfolios from the perspective of children. It has an ethnographic standpoint. It was carried out on children and pupils in pre-school, pre-school class and third grade. By further investigating how children and pupils use their individual development plans and portfolios, in which settings and to whom they communicate them, this study is able to grasp the discourse that is created around the instrument itself. This study is also interested in and aims to find the theoretical frame, in essence, which envelops both the construction of childhood and children’s own activities within the limitations and opportunities that this structure offers. By focusing on micro, hereby meaning the thoughts and perceptions of children and pupils themselves, and on macro, such as the structure around the children and pupils, the aim of finding the previously mentioned theoretical space is fulfilled. The study was carried out by means of an ethnographic method, with conversations, interviews, surveys, observations and filming. The results show that children and pupils view their individual developmental plans and portfolios as an affordance, both in a physical sense and as an offer of a symbolic space, a space shared by others but with the same significance for all. It is also apparent that the children and pupils use their individual development plans to ascertain their positions, to show who they are.

A pension fund is defined as an investment product in which the members of the scheme pay contributions in order to receive an income at retirement. This is a very simplistic definition when compared to the United Kingdom's Pension funds, which are complex and very specific financial products. This report is a result of an internship at the Willis Towers Watson Lisbon office, in the area of retirement, where the main scope of the work was in UK pension schemes' Actuarial Valuations. The main goal of this project is to study the different ways of measuring the funding position of a pension scheme, regarding the setting of assumptions. This will result in a practical demonstration of the impact of the defined assumptions on the results obtained. In order to do that, two pension schemes will be used, from companies that have been the focus of the internship. For academic purposes, this internship report contributes to a better understanding on measuring the funding position of pension schemes, and also on the application of knowledge obtained in the master's, for example on survival models, life annuities, financial markets and other subjects/topics.

Abstract:
The aim of this study is to describe, from a discourse theoretical perspective, the prevailing discourse and the subject positions available within this discourse among sevens pupils attending higher education preparatory programmes at Upper Secondary Schools in Sweden. The discourse emerges in qualitative interviews where the pupils talk about their experiences of individual educational plans. The result shows that concordance prevails among the pupils about the fact that their individual educational plans have been drawn up because they have fallen behind in their school work, and because they run the risk of having grade F. Furthermore, the result shows that concordance does not prevail among the pupils when it comes to why they have fallen behind in their school work or how the individual educational plan has helped to improve the results. The result shows that in the pupils’ descriptions several subject positions appear as accessible within the discourse. All pupils accept the subject position of a pupil running the risk of getting an F, likewise they all accept a subject position that signifies that he or she is not an unusual or special pupil.

Abstract:

The aim of this essay is to analyze how students are constituted as subjects in local curricula. The theoretical perspective is grounded in Michel Foucault’s notion of governmentality and power/knowledge and his critique of a sovereign subject. From this theoretical perspective I use Foucault’s concept of formation of objects and Ernesto Laclau and Chantal Mouffe’s idea of subject positioning as tools for a discourse analysis. The analyzed documents are local curricula from fourteen Swedish municipalities. The documents are divided in to two equally sized groups based on the political governing in the municipality.

The analysis presents five themes by which statements that constitute students as subjects can be categorized. These five themes are; lifelong learning, desire, essential traits, students responsibility and life and health. The differences between local curricula from political right wing and political left wing governed municipalities are rather small, but there can be seen differences in the technologies of liberal governing. The subject that is constituted in local curricula is primarily a self-governing subject who governs it self in relation to a expertise knowledge.

This paper presents the study, manipulation and control of a two degrees of freedom system, known as planar pendulum, using reaction wheels to achieve a certain position, the basic concept of this system being the dynamics of an inverted pendulum. The classic PID controller was proposed as well as a Fuzzy control. The mathematical model was generated to be used in MatLab numerical simulations of these controls, using the Simulink tool which allows a self-adjustment of the controller of the system. The PID control was built according to the self-tuning of Simulink and also using the Ziegler Nichols method. The differences between both are discussed. Fuzzy control was designed, creating the bank of rules and looking for a control strategy more suitable for the system. Friction in joints, and drives were taken in account in the mathematical modeling. Finally a real model was built using a data acquisition board in conjunction with LabView for control and a microcontroller board called Arduino, as well as an encoder for the acquisition of angles. Measurement errors and unbalance of the system are problems that could not be completely eliminated, but were kept to a minimum. Numerical and experimental results were compared for each control and for each assembly and their differences were discussed.

Peng, Cheng. "Position-weighted template matching for measuring in-plane dynamics of microdevices." Thesis, University of British Columbia, 2015. http://hdl.handle.net/2429/55611.

Full text
Abstract:
The measurement of in-plane dynamics of microdevices is crucial to analyzing their dynamic characteristics under certain excitations. It has become more and more important to enable precise measurements and visual means to characterize dynamic microstructures, as the designs of moving micro-electro-mechanical systems (MEMS) are rapidly becoming more and more complex. And the visualization and measurement of the dynamics of MEMS structures are of considerable significance to the development of more effective and advanced microdevices. This thesis investigates the problem of visualizing, measuring and analyzing the in-plane dynamics of microdevices. We propose a novel object position tracking algorithm, called position-weighted template matching, improving the traditional template matching technique. The newly proposed algorithm effectively addresses the position "jump" problem that typically happens for object tracking in planar microdevices, where similar sub-patterns may exist in a single structure. We have incorporated the parabola fitting interpolation technique into our algorithm to achieve a higher, sub-pixel resolution level. We have implemented our proposed methods into a software module, associated with a LabVIEW Graphical User Interface (GUI). Several comparative experiments were carried out to demonstrate the effectiveness of our algorithm. In addition, the procedure was also used for performing a system identification on a fabricated MEMS resonator. Our implemented LabVIEW GUI can be potentially interfaced with low-cost hardware to enable visualization and measurement of in-plane motion of microdevices.

Ramsay, Heather. "The correlation of evolutionary rate and pathway position in plant terpenoid synthesis." Thesis, University of British Columbia, 2008. http://hdl.handle.net/2429/12671.

Full text
Abstract:
Genes are expected to face stronger selective constraint and to evolve more slowly if they encode enzymes upstream as opposed to downstream in metabolic pathways, since upstream genes are more pleiotropic, being required for a wider range of end-products. However, few clear examples of this trend in evolutionary rate variation exist. We examined whether genes involved in plant terpenoid biosynthesis exhibit such a pattern, using data for 45 genes from four fullysequenced angiosperms, Vitis, Arabidopsis, Populus and Ricinus. Taking a bioinformatic approach, we used phylogenetic trees of proteins from each genome to determine orthologs and count paralogs in each genome, using proteins from the Oryza genome as an outgroup. Our results show that dN/dS does in fact correlate with pathway position along pathways converting glucose to the terpenoid phytohormones abscissic acid, gibberellic acid and brassinosteroids. Upstream versus downstream rate variation is particularly strong in the gibberellic acid pathway, but absent in the pathway to lutein, another terpenoid derivative. In contrast, we found no apparent variation of dN/dS with gene copy number. We also introduce a new measure of pathway position, the Pathway Pleiotropy Index (PPI), which counts groups of enzymes between pathway branchpoints. We found that this measure is superior to pathway position in explaining variation in dN/dS along each pathway. Kendall's τ for the correlation of PPI with dN/dS was 0.24 - 0.54 with a mean of 0.4. We further show that variation in dN/dS is due to differences in selective constraint, not positive selection. Therefore, our results are consistent with the prediction that selective constraint is progressively relaxed along metabolic pathways, showing plant terpenoid synthesis to be a robust example of positional rate variation.

Abstract:

Urban middle school students experience poor self-efficacy and poor attitudes toward school climates after being retained. Previous research has indicated that grade-level retention in primary and secondary education might cause long-term achievement gaps, school failure, and high school dropout rates. However, current research has yet to examine relationships between archival data retrieved on retained middle school students’ achievement outcomes and perceptions of school climate. The purpose of this nonexperimental, quantitative study was to assess the relationships between retained middle school students’ self-efficacy as measured by the School Climate Survey and their performance outcomes as measured by PowerSchool®. Bandura’s theory of self-efficacy maintains that an individual must have the belief, motivation, determination, and drive to persevere when challenged. The archival data were collected from 1 northeastern urban middle school in the United States representing underachieving participants (N = 45) enrolled in the Postive Academic and Behavioral Support Program during the academic school years of 2017 and 2018. Population groups of female and male students ranged in age between 11–14 years old. A repeated measure design analyzed the same participants over a 6-month period by measuring archival data on achievement outcomes from GPAs, attendance, and demographics (sex and age). Results showed significant increases in GPAs and significant increases in males’ positive perceptions of school over the school years of 2017 and 2018. The results of this study can be used to promote positive social change for education professionals working in urban school districts providing support services to at-risk students facing school failure.

Urban middle school students experience poor self-efficacy and poor attitudes toward school climates after being retained. Previous research has indicated that grade-level retention in primary and secondary education might cause long-term achievement gaps, school failure, and high school dropout rates. However, current research has yet to examine relationships between archival data retrieved on retained middle school students' achievement outcomes and perceptions of school climate. The purpose of this nonexperimental, quantitative study was to assess the relationships between retained middle school students' self-efficacy as measured by the School Climate Survey and their performance outcomes as measured by PowerSchool®. Bandura's theory of self-efficacy maintains that an individual must have the belief, motivation, determination, and drive to persevere when challenged. The archival data were collected from 1 northeastern urban middle school in the United States representing underachieving participants (N = 45) enrolled in the Postive Academic and Behavioral Support Program during the academic school years of 2017 and 2018. Population groups of female and male students ranged in age between 11-14 years old. A repeated measure design analyzed the same participants over a 6-month period by measuring archival data on achievement outcomes from GPAs, attendance, and demographics (sex and age). Results showed significant increases in GPAs and significant increases in males' positive perceptions of school over the school years of 2017 and 2018. The results of this study can be used to promote positive social change for education professionals working in urban school districts providing support services to at-risk students facing school failure.

Abstract:
This thesis analyzes the problem of motion stability of submarines in free positive buoyancy ascent under casualty conditions such as control surface jam and loss of propulsion system response. We employ fully nonlinear, coupled six degree of freedom equations of motion and we allow response to occur in combined vertical and horizontal planes. Continuation and homotopy theory techniques are utilized to trace all possible steady state solutions in six degrees of freedom, while local perturbation reveals their stability properties. Vehicle geometric properties and control surface deflections are used as primary bifurcation parameters. Regions in parameter spaces are identified where extreme sensitivity of solutions to geometric properties and hydrodynamic modeling is present

Abstract:
Students who exhibit disruptive and inappropriate behaviors are a challenge for schools, as teachers and administrators are most often held responsible for managing these behaviors. This study was designed to evaluate School-Wide Positive Behavior Supports (SWPBS) when used in a rural middle school setting. Research indicates that using SWPBS in an individual school over a period of time can decrease the amount of disruptive behaviors in the school while also increasing attendance and academic scores. However, little longitudinal research exists concerning the use of SWPBS within middle school settings across a system. Through the use of quantitative methods, this study included examining overall effect on office disciplinary referrals (ODRs), suspensions, and expulsions under the 3 categories of schools as well as faculty and student perceptions (current year only) of SWPBS. While there was no significant difference found among ODRs, expulsions, or suspensions among the 3 school categories through ANOVA analysis, the study did highlight the need for further evaluation of how schools implement SWPBS. A one-way, chi-square analysis of faculty perceptions did not show a significance among the three school categories, while student perceptions did show significance and a follow-up pairwise comparison was conducted. The student survey analysis suggests that students in either a school with the SWPBS plan or without a plan are 3 times more likely to have a negative attitude toward the behavioral school policies than schools with a partial SWPBS plan. The analysis on perceptions indicates the need for further faculty and student questioning in future research. The primary significance of this study is that it addresses the use of SWPBS in various middle school settings and in various stages of use (full plan, partial plan, or no plan). This study also evaluates data from two years before the SWPBS implementation as opposed to just one year. The study highlights issues related to middle school students and how school systems might better serve those students.

Abstract:
This thesis is dedicated to the study of three problems of complex and almost complex hyperbolicity. Its first part is dedicated to the research of a quantitative consequence to Kobayashi hyperbolicity, which is a qualitative property. The result we obtain has the form of an isoperimetric inequality that suggests Ahlfors' inequality, the central result of the theory of covering surfaces. Its proof uses only riemannian tools. The second part of the thesis is dedicated to the proof of an almost complex version of Borel's theorem, which says that an entire curve in the compex preojective plane missing four lines in general position is degenerate. In an almost compex context, we can obtain a similar result for entire J-curves just by replacing projective lines by J-lines. The proof of this result uses central projections and Ahlfors' theory of covering surfaces. The last part is dedicated to the proof of an almost complex version of Bloch's theorem, which says that given a sequence of holomorphic discs in the projective plane, either it is normal, either it converges in some sens to a reunion of three lines. Our result will show in particular that the complementary set of four J-lines in general position is hyperbolic modulo three J-lines

Full text
Abstract:
The present research explores and examines positive affect, which varies in approach motivational intensity, and its influence on creative cognition in the context of computer game play. Cognitive functions of positive affect and computer game play are a growing concern for educational practitioners and parents. Little is known about how 'out-of-school' use of computer games influences affect and creativity. This research explores the effects of positive affect elicited by computer games on various forms of creative cognition (including breadth of attention, productivity and originality) from the perspectives in a theoretical framework, emphasising approach motivational dimension of positive affect. This framework predicts that high approach- motivated positive affect may narrow breadth of attention, and reduce productivity and originality. Accordingly, low approach-motivated positive affect may broaden breadth of attention, and enhance productivity and originality. Three within-participants experiments were conducted to examine these predictions. Results were mixed in their support of theoretical predictions. In Study 1, as expected, playing an exciting computer game versus viewing a humorous film elicited a stronger degree of high (desire and eagerness) versus low (relaxation and amusement) approach-motivated positive affective states respectively. The same results of affective states were replicated in Study 2. However, unexpectedly, these positive affective states varying in approach motivation did not show differences in their effects on breadth of attention as there were no significant differences of response times on a global-local visual processing attentional task used to measure breadth of attention. The asymmetric transfer effect inherent in the within-participant research design and an absence of global precedence effect on the attentional task representing a weak validity of the measurement for breadth of attention were found. In Study 3, the degree of high approach-motivated positive affect [i.e., desire) reported after playing an exciting computer game with high arousal did not differ from that reported after playing a relaxing computer game with low arousal. Unexpectedly, originality performed after playing the exciting computer game was better than that performed after playing the relaxing computer game; nevertheless, playing these two types of games did not differ in productivity. The present research supports the view that positive affective states can vary in approach motivation as well as the state of arousal. Results are discussed with reflections on methodology and their implications for the theoretical framework. A new research proposal based on a reflected theoretical framework is suggested fo r future research on how positive affect elicited by computer game play may influence creative cognition. I.

The presente study focuses in teh capacity of positive emotions being able to create valuable resources for music students, but also by the fact that they can function as na essential agente for the instrumental efficiency and its continuity. From Barbara Fredrickson’s Broaden-and-Build Theory we established a parallel with the pedagogical tool play-along with CD, as a potentially motivating tool and fosterer of valuable resources and skills for the performers, as well as of the individuals’ thought-action repertoire. This tool used together with strategies based on positive emotions becomes indispensable for first and second grade students, who are at the beginning of their musical studies as well as of their intrinsic motivation building, namely, in na instrument with so many complexities such as the bassoon.

Abstract:
The purpose of this study was to explore how the therapist-provided conditions of congruence, unconditional positive regard, and empathic understanding were experienced and conveyed in child-centered play therapy (CCPT). Although the therapist-provided conditions are considered essential to the therapeutic process in CCPT, a gap exists between child-centered theory and empirical exploration of the process and dynamics of these relational variables in CCPT. Due to the limited research in this area, a grounded theory approach was utilized to explore how the three variables emerge in CCPT. Participants included four advanced doctoral students, all Caucasian females with extensive training in CCPT, and 12 children ranging from 4 to 8 years of age receiving weekly, individual CCPT. One individual CCPT session was observed and video-recorded for each therapist-child dyad (n = 12). Following each observation, play therapists were interviewed regarding the observed play session (n = 12). During each interview, the researcher and therapist watched the recorded play session in its entirety and discussed noteworthy interactions between the child and therapist. The video-recorded play therapy sessions and therapist interviews were analyzed using a multiphasic, constant comparative method. Results of the analysis included a process-model of the therapist-provided conditions in CCPT, examples of play therapists’ internal experiences and external behaviors associated with the presence and absence of the therapist-provided conditions, and a model of the process play therapists utilize to respond to breaks and barriers to congruence, unconditional positive regard, empathic understanding, and unconditional positive self-regard in CCPT. Implications for clinical practice, training, supervision, and research are discussed.

Abstract:

Abstract:
Bibliography: pages 174-182.
This dissertation investigates the machine vision grading of flue-cured Virginia tobacco by means of digital processing of tobacco leaf images. With reference to international grading standards and to modem image processing techniques, two classifiers are designed. The colour classifier uses seven features extracted from each leaf image to grade the leaf into one of five official colour classes. It does this with an expected correct classification rate of 93.5%. The plant position classifier identifies the position on the stalk from which a leaf was reaped, using ten size and shape features to classify the leaf into one of six plant position categories. It has a correct classification rate of 70%. Average colours for each colour class and archetypal shapes for each plant position category are derived from the digital leaf data. These should be of value to tobacco graders as objective representations of typical leaves within each class.

In this study the author attempts to identify factors that are determinants for why some pupils in Swedish grade school have high status in comparison with their peers. The study takes on the characteristics of both an ethnographic and a literature study by comparing the findings of interviews carried out with students of a first grade class with previous research. The study identify several tools for analyzing of how status may be understood in specific situations. The study also finds that important factors for gaining high status amongst children in aschool classes might be the ability to apply access strategies to get accepted in social interactions, such as game play, have the ability to create games that are enjoyed by peers and social skills, to name a few. Overall the study shows that children's game play with each otherare crucial for their social interactions and their social position. The study also show that children that have earlier established relationships with other children in a specific group alsohave a better opportunity to establish a high status position.

Chotprakaikiat, Warayut. "Development of a novel plant expressed conjugate vaccine for HER2-positive breast cancer." Thesis, University of Southampton, 2016. https://eprints.soton.ac.uk/400684/.

Full text
Abstract:
Overexpression HER2/neu correlates with poor prognosis in many cancers, including breast cancer. HER2 -specific monoclonal antibodies including Herceptin® have had significant impact on breast cancer survival rates. However, there are a number of limitations with their use, including tumour recurrence, the requirement for multiple doses and high -cost production. Vaccination, however, can provide long-term tumour specific antibodies and cellular-mediated immune responses. Here the aim was to develop a potent vaccine to target HER2. Self-tumour antigen HER2 is poorly immunogenic due to tolerance. Previous studies using DNA vaccines demonstrated that conjugation of tumour antigens to Fragment C of tetanus toxin (FrC) broke tolerance. Here to target HER2 a novel HER2 portion ED44 was generated and conjugated to FrC. The HER2-targeting vaccine subunits were expressed using an efficient plant-based manufacturing process 'magnifection' and were available through collaboration (ICON Genetics, Germany). Both rat and human versions were generated to study the mechanisms or for clinical application respectively. They were tested in several complementary breast cancer models including non-tolerant (TUBO, D2F2/E2) and tolerant (Balb/neuT) settings. In non-tolerant settings unconjugated ED44 was effective at inducing of protective antibody. In the tolerant model, only ED44-FrC conjugate was effective in inducing high levels of antibody and suppressing already developed tumours. Only ED44-FrC conjugate vaccine induced antibody of high affinity of a wide range of isotypes. FrC-specific T cell help induced by the conjugate vaccine was critical for effective anti-HER2 antibody. The human ED44-FrC induced sera were able to inhibit HER2-mediated downstream signalling and induced growth inhibition of two HER2positive cell lines. Hence, the novel vaccine is a promising candidate for HER2 positive cancer treatment. In the last chapter a novel vaccine design that included plant virus particle (PVP) was used to induce anti-HER2 cytotoxic T cells. This sets the foundation for development of this platform in the future.

Abstract:
BACKGROUND: Lichen planus (LP) is a mucocutaneous inflammatory chronic disease of unknown etiology. Alterations in the innate immune response such as the pathogen-associated molecular pattern (PAMPs) and damage-associated molecular pattern (DAMPs) can lead to chronic inflammation and contribute to the pathogenesis of LP. OBJECTIVE: Evaluate the effect of the activation trough the DAMP S100A8 and the Toll-like receptor 4 (TLR-4) on the Natural killer cells (NK) and cytotoxic TCD8 cells and their memory / effector subsets in LP disease. METHODS: We selected 25 patients with LP (22 women, 3 men) with a mean age of 43.46 years ± 8.46 and a control group of 25 subjects (22 women, 3 men) with a mean age of 42 ± 5, 5. The transcriptional determination and protein expression by immunohistochemistry of DAMPs, S100A8 and HMGB-1 as well as TLR-4 and RAGE was performed on biopsies of skin lesions from patients with LP, and serum levels of S100A8, HMGB-1, MICA and MICB were determined by ELISA. Peripheral blood mononuclear cells (PBMCs) were assessed by flow cytometry to evaluate the frequency of TNF, IL-1beta and the degranulation marker CD107a in CD8+ T cells and CD56 + NK cells and their subsets. The evaluation of the TLR signaling pathway in purified CD8 + T cells activated with S100A8 were analyzed by PCR array and the determination of mRNA expression of inflammasome components on CD8 + T cells activated by S100A8 was measured by real time PCR. RESULTS: It was shown in the LP individuals an increased expression of the S100A8 protein in the cutaneous lesions and HMGB-1, TLR-4 and RAGE in the dermis, in parallel to increased level of mRNAs for S100A8 and S100A9 and decreased expression of RAGE. Moerover, increased serum levels of the dimer S100A8 / A9 was detected in patients compared to controls, in contrast to DAMP HMGB1 that revealed similar levels in both groups. The influence of S100A8 in CD8 + T cells and NK cells, was analyzed in PBMC activating with lipopolysaccharide and recombinant protein S100A8, both ligands of TLR-4. It was detected in LP individuals, an increased cytotoxic response of CD8+ T lymphocytes and CD56bright NK cells trough CD107a degranulation marker expression. The S100A8 protein was able to induce the pro-inflammatory genes expressions such as IL-1beta, TNF and IL-6 in CD8 + T cells of LP patients in contrast to healthy subjects who promoted IL-10 expression and type I IFN. CD8 + T cells of LP individuals activated or not with S100A8 are able to express NLRP1, NLRP3 and AIM-2 and IL-1beta production at similar levels to healthy controls. Moreover, CD8 + T cells activated with S100A8 showed increased expression of TLR3, TLR5, TLR7 and TLR8 in LP compared to biopsies from healthy controls. The increased CD8 + T cells cytotoxic response was mediated by the subtype of effector memory (TEM CD45RA- CCR7). The increased baseline expression of activating receptor NKG2D and the inhibitory NKG2A in the NK CD56dim cells in LP individulas, and the similar level of MICB soluble in both groups. CONCLUSION: These results shows that innate immunity components, such as S100A8 protein may contribute to the maintenance of LP inflammatory profile

Abstract:
The purpose of this Master’s thesis was to evaluate coaches’ program delivery in Golf Canada’s national youth programming, Learn to Play, regarding two components: (a) the consistency of teaching life skills and (b) the quality of program delivery. The Learn to Play program has recently been updated to include an explicit approach to teaching life skills through golf. Data collection included a pre-season interview, three in-season observations, a post-season questionnaire, and a post-season interview. Two articles were written. Using the implicit/explicit continuum of life skills development and transfer (Bean, Kramers, Forneris, & Camiré, 2018), Article One qualitatively explored coaches’ (N = 13; Mage = 38) consistency of life skills delivery across the season. The findings revealed five profiles, with coaches categorized as being either inconsistent (i.e., progressive, overenthusiastic, unaware) or consistent (i.e., non-intentional, intentional) in their approach to teaching life skills. Using the Program Quality Assessment in Youth Sport observational measure (Bean, Kramers, Camiré, Fraser-Thomas, & Forneris, 2018), Article Two quantitatively examined program quality with two groups of coaches (N = 14; Mage = 40) for two purposes: (a) to examine whether implementation of the original (n = 5) versus the updated (n = 9) program led to differences in program quality and (b) to assess whether there was consistency or discrepancy between researcher observation scores and coach self-report scores of program quality. The findings revealed that coaches using the updated program delivered higher quality programming than coaches using the original program. Coaches also reported higher program quality scores than the researchers observed, identifying discrepancies between coach self-perceptions and researcher observations. Collectively, the findings from this Master’s thesis are a novel contribution to the literature by providing a nuanced exploration into the teaching of life skills and quality of delivery of Golf Canada’s Learn to Play programming.

Basinger, Jason C. "Perspective and Practices to Address Rough-and-Tumble Play in the School Setting: A Survey of School-Wide Positive Behavior Support Teams in Utah." DigitalCommons@USU, 2012. https://digitalcommons.usu.edu/etd/1382.

Full text
Abstract:
Prior research of rough-and-tumble play (RTP) has shown mixed results—different operational definitions, varying functions, and positive and negative outcomes. Few researchers have studied interventions to address RTP in school settings. With unclear evidence of RTP outcomes and the extent school interventions are addressing RTP in school settings, this study explored the extent and effectiveness of intervention programs being implemented to prevent/reduce negative outcomes of RTP in elementary schools. A survey was created and conducted with 30 school problem-solving teams in a western state to obtain information concerning RTP in elementary school settings. Teams provided estimated percentages of RTP leading to beneficial and problematic behaviors, types of benefits or problems resulting from RTP, specific prevention/intervention programs that teams report implementing to address RTP concerns, percentage estimations of students responding to implemented interventions, and training needs to address interventions for RTP concerns. Survey responses showed different medians for estimated percentages for problematic outcomes (80%) versus beneficial outcomes (10%). Further, the number of problem items (M = 9.57, SD = 1.87) was more highly endorsed by teams than the number of items listing benefits (M = 4.43, SD = 3. 39), suggesting RTP was more often problematic than beneficial. Interventions estimated to be effective in treating negative RTP outcomes with 80% or greater response rates are reward systems, social skills trainings, active supervision, and bully prevention. These study findings are different from previous research, which concluded that RTP was harmless and/or beneficial to students, and might be due to environmental differences (school vs. community). It might be estimated that schools should monitor or prevent RTP to avoid problems, such as aggression, bullying, and poor peer relationships. Programs frequently used by teams targeted skill acquisition through social skills training, anger management, and bully prevention. This study provides understanding to the extent RTP should be addressed in schools. School problem-solving teams report that RTP can be problematic in school settings; however, it can be prevented with school-wide intervention and intervened with individual and small-group interventions.

Kruger, Jacques. "An autoethnographic exploration of “play at work” / Jacques Kruger." Thesis, North-West University, 2011. http://hdl.handle.net/10394/8460.

Full text
Abstract:
This research brings together two concepts that are often depicted as polar opposites. Sutton-Smith (2001) however suggests that the opposite of play is not work, but depression, and moreover echoes other scholars in reclaiming play as an essential human expression, even for adults. This study, therefore, argues that, given the precarious wellness territory our workplaces are in, something about work is not working. It is furthermore proposed that, given all the evidence of the therapeutic potential inherent to play, there is indeed something nutritious at play in play. Despite these well-supported arguments, play remains hidden away in the academic shadows of more serious industrial psychological preoccupations. Surprisingly, the same conspicuous absence is even mirrored in Positive Psychology, a bustling field that claims to celebrate glee, fun, and happiness (Seligman, 2002a). Entitled “An autoethnographic exploration of play at work,” this dissertation leans on the metaphor of “exploration”, or more specifically, exploratory play. This results in two distinct yet interwoven dimensions to the research study. Firstly, the research approaches the phenomenon of play and play-based methods in workshop contexts through the lived experience of the researcher. Secondly, the research project in itself is conceptualised as work, and the methodology of autoethnography is conceptualised as a playful approach to this work of conducting research. Aside from widening the research scope, this also appropriately matches research methodology to the research domain. Aside from being about play at work, this research also is play at work. Autoethnography, as a recent development in qualitative research, remains unconventional and somewhat controversial in the South African social sciences. Autoethnography, as an offspring of ethnography, offers a method to reflexively incorporate the researcher’s own lived experience in the study of culture as a primary source of rich phenomenological data. Instead of minimising the emotive and subjective, this research amplifies and celebrates it. Given a fair degree of unfamiliarity in terms of autoethnography as well the accusation of being overly self-centred, the experience of the researcher is then complemented by the views of a number of co-creators to the culture being studied. This is done through external data-gathering in the forms of a focus group as well as number of semistructured, dyadic interviews. While therefore leaning more toward postmodern themes, this research also incorporates what has been termed analytical autoethnography (Anderson, 2006), wherein the researcher is a full-member of the setting being studied, is portrayed as such and is committed to theoretical analysis. This study can therefore be summarised as an autoethnographic case study that balances evocative and analytical styles (Vryan, 2006) while emanating from the philosophical assumptions of interpretivism and subjectivism. Internal realities and meaning-creation are thus emphasised rather than the received views of positivism. The central research question being explored is how play and play-based methods promote work-related well-being. To answer this question, firstly, play and play-based methods are explored, both from a theoretical and practical point of view. From within workshop (pedagogical) contexts, the play-based methods considered throughout this study include metaphor and story, creative-arts-based play, physical-body play and also the uncelebrated yet essential methods of icebreakers and games. A preliminary taxonomy is proposed for play-based methods to offer description and to facilitate reflection and learning. Descriptive elements in this taxonomy include interactive vs. solitary, competitive vs. cooperative, motor-sensory vs. cognitive-mind, participative vs. vicarious and rule-bound vs. improvisational. Building on this exploration of play-based methods, the second aspect explored in more detail has to do with the more internal and subjective experiences of participants, or players, if you like. These experiences are then related to prominent concepts encountered in Positive Psychology to, by proxy, understand how they relate to work-related well-being. Significant themes that emerge from this include play as fun, play as mind-body integration, play as authenticity, play as community, and play as stress-relief and resilience. This is then woven into a creative non-fiction, in accord with a trend in qualitative research called creative analytical practices (CAP) (Richardson, 2000). This creative non-fiction, detailed in Chapter 4, forms a key autoethnographic output that animates all these themes in a way that is accessible, evocative and playful. Chapter 5 complements this chapter with an in-depth exploration of the research journey as a confessional tale. While adopting the metaphor of hiking in mountains (exploring nature), this confessional tale clarifies the research process and incorporates an in-depth analysis of the themes, both in terms of research data as well as literature. This is supported by a number of separate appendixes, including interview transcripts, depictions of the interview analysis as well as a number of photos from the field. In terms of its uniqueness and unconventionality, this research joins in the choir of related work to incorporate more contemporary research genres into the social sciences in South Africa. By doing so, it opens up doors to phenomena that simply resist being studied with the ontological and epistemological assumptions of conventional modern science. Furthermore, the effect and impact of this research is that it provides accessible and practical ideas as to how a synthesis of play and work can help us renew and rejuvenate our work and workplaces. That is, how we can come alive in the work contexts that risk becoming sterile, clinical and inhuman in the wake of Taylorist reductionism and efficiency. Given that state of work and workplace, and the productive and therapeutic potential in play, indeed, we are too busy not to play.

Abstract:
Video games are an almost universal adolescent leisure activity. Though some in the scholarly field are concerned about the effects of violent video game content on adolescent social outcomes, others believe that the benefits of video game play have been overlooked. A literature review was conducted to examine the evidence for positive and negative outcomes of video game play on adolescents. There were 14 articles identified and subsequently reviewed. Findings demonstrated a unique link between video game play and antisocial (e.g., aggression), prosocial (e.g., generosity), and pragmatic (e.g., problem-solving skills) outcomes. However, the results also suggest that much of this research is atheoretical and does adequately consider the roles of relational context. Discussion of the potential roles of relationships, theoretical applications, strengths and limitations of the research, and future directions are provided.

Gavriilidou, Margarita. "What role do psychosocial factors play in influencing HIV positive people's compliance with medical treatment?" Thesis, Manchester Metropolitan University, 2013. http://e-space.mmu.ac.uk/315703/.

Full text
Abstract:
Antiretroviral therapy has given hope and expectations for a better life to HIV positive individuals, however, HIV medication cannot be effective without HIV positive individuals’ compliance to it. This study investigated the ways in which living with HIV and taking medication is located within the psychological, social and cultural context of everyday life and relationships in Greece. It also examined gender and identity issues, which make compliance/non-compliance understandable from the HIV positive peoples’ perspective. In addition, emphasis was given to locating compliance to medical regimes in which the perspectives of HIV positive persons were prioritised and understood in relation to relationships with health care professionals. A mixed methods approach was undertaken to provide understanding of compliance and non-compliance factors to HIV medication in a holistic way. A self-completed questionnaire was used to examine the psychosocial factors underpinning compliance to medication. Face-to-face semi-structured interviews were used to explore issues of identity, gender, relationship between doctors and patients and social understandings of HIV. Finally, self-completed weekly diaries were used to document compliance actions, thoughts and feelings in order to reveal the ways medical regimes fit into everyday life. The study was conducted in three Public Hospitals, one Governmental Hospice and one Non-governmental Organization. Eighty (63 males and 17 females) Greek HIV positive patients completed the questionnaire. Interview sample consisted of 7 and 3 males and females respectively. Finally, 6 Greek HIV positive males and 3 females completed the diaries of the research. The questionnaire data was analysed using descriptive statistics via SPSS 11. In addition, a range of non-parametric tests (Mann Whitney and Kruskal Wallis) were used in order to check if ordinal variables influence compliance with HIV medication. Finally linear regression analysis was used in order to establish the influence of factors on compliance with HIV medication. Interviews and the diaries data were analysed though thematic analysis, focusing on identification of patterns and behaviours which were then interpreted in terms of themes. The findings of the study indicated that, when support was given from life partners compliance with HV medication was increased. However, when support was given from family members, compliance with HIV medication was decreased. According to the findings, family dynamics have changed in several cultures over recent decades, partner roles have changed especially in the west and in Mediterranean societies. In regards to 6 medicalization in everyday life, the study showed that when individuals were experiencing side effects, or had fears of future side effects, religious issues (punishment for homosexuality), loss of one’s freedom due to medication, non-compliant behaviours could occur. Finally, the study indicated that some HIV positive individuals perceived their health levels as good and believed that not taking medication once or twice a week was a compliant behaviour. Hence, false perceptions regarding health levels and compliance issues could lead to non-compliant behaviours. A further examination on the communication patterns of the family system and its impact on HIV positive individuals is recommended as it is clearly not very helpful any more. Further exploration of the general socio-cultural positioning of Greece is recommended as certain HIV positive individuals coped with HIV diagnosis and taking medication, by rejecting it. Finally, the need for psychological support is recommended as it is very rarely provided within the Greek health care system.
