Abstract:
RESUMO Objetivou-se ajustar e avaliar modelos para predição da composição física e química corporal de borregas a partir de medidas obtidas da seção da 9ª à 11ª costelas (seção 9-11) ou da 12ª costela. Foram confinadas em baias individuais 36 borregas mestiças alimentadas com níveis crescentes de concentrado (0; 20; 40; 60 e 80%) até atingirem 37,70 ± 10,23kg para o abate. No abate, a meia-carcaça direita de cada animal foi dissecada para determinação da proporção de músculo (PMC), gordura (PGC), osso (POC), proteína bruta (PPBC) e extrato etéreo (PEEC). A seção 9-11 e a 12ª costela foram obtidas na meia-carcaça esquerda com objetivo similar. Os modelos para predição foram ajustados como possíveis variáveis preditoras a PMC, PGC, POC, PPBC e PEEC na seção 9-11 (PM9-11, PG9-11, PO9-11, PPB9-11 e PEEC9-11, respectivamente) ou na 12ª costela (PM12, PG12, PO12, PPB12 e PEE12, respectivamente). Foi observada correlação positiva entre a PM9-11 e PMC (r = 0,69; P<0,001). A PG9-11 foi correlacionada com a PGC (r = 0,93; P<0,001). A PO9-11 foi positivamente correlacionada com a POC (r = 0,92; P<0,001). A PG12 gerou modelo acurado para a predição da PGC (R2 = 0,92; P<0,001). A equação de predição para estimativa da POC a partir da PO9-11 foi mais precisa e acurada (R2= 0,86; P<0,001) em relação à PO12 (R2 = 0,55). É possível gerar modelos de predição de componentes físicos e químicos corporais de borregas a partir da seção 9-11 e 12ª costela.

Abstract. Rising ocean temperatures will likely increase stratification of the water column and reduce nutrient input into the photic zone. This will increase the likelihood of nutrient limitation in marine microalgae, leading to changes in the abundance and composition of phytoplankton communities, which in turn will affect global biogeochemical cycles. Calcifying algae, such as coccolithophores, influence the carbon cycle by fixing CO2 into particulate organic carbon (POC) through photosynthesis and into particulate inorganic carbon (PIC) through calcification. As calcification produces a net release of CO2, the ratio of PIC / POC determines whether coccolithophores act as a source (PIC / POC > 1) or a sink (PIC / POC < 1) of atmospheric CO2. We studied the effect of phosphorus (P-) limitation and temperature stress on the physiology and PIC / POC ratios of two subspecies of Coccolithus pelagicus. This large and heavily calcified species (PIC / POC generally > 1.5) is a major contributor to calcite export from the photic zone into deep-sea reservoirs. Phosphorus limitation did not influence exponential growth rates in either subspecies, but P-limited cells had significantly lower cellular P-content. A 5 °C temperature increase did not affect exponential growth rates either, but nearly doubled cellular P-content under both high and low phosphate availability. The PIC / POC ratios did not differ between P-limited and nutrient-replete cultures, but at elevated temperature (from 10 to 15 °C) PIC / POC ratios decreased by 40–60%. Our results suggest that elevated temperature may intensify P-limitation due to a higher P-requirement to maintain growth and POC production rates, possibly reducing abundances in a warmer ocean. Under such a scenario C. pelagicus may decrease its calcification rate relative to photosynthesis, resulting in PIC / POC ratios < 1 and favouring CO2-sequestration over release. Phosphorus limitation by itself is unlikely to cause changes in the PIC / POC ratio in this species.

Abstract:
Poor knowledge and adherence to point-of-care (POC) HIV testing standards have been reported in rural KwaZulu-Natal (KZN), a high HIV prevalent setting. Improving compliance to HIV testing standards is critical, particularly during the gradual phasing out of lay counsellor providers and the shifting of HIV testing and counselling duties to professional nurses. The main objective of this study was to identify priority areas for development of POC diagnostics curriculum to improve competence and adherence to POC diagnostics quality standards for primary healthcare (PHC) nurses in rural South Africa. Method: PHC clinic stakeholders were invited to participate in a co-creation workshop. Participants were purposely sampled from each of the 11 KwaZulu-Natal Districts. Through the Nominal Group Technique (NGT), participants identified training related challenges concerning delivery of quality point of care diagnostics and ranked them from highest to lowest priority. An importance ranking score (scale 1–5) was calculated for each of the identified challenges. Results: Study participants included three PHC professional nurses, one TB professional nurse, one HIV lay councilor, one TB assistant and three POC diagnostics researchers, aged 23–50. Participants identified ten POC diagnostics related challenges. Amongst the highest ranked challenges were the following:absence of POC testing Curriculum for nurses, absence of training of staff on HIV testing and counselling as lay counsellor providers are gradually being phased out,. absence of Continuous Professional Development opportunities and lack of Staff involvement in POC Management programs. Conclusion: Key stakeholders perceived training of PHC nurses as the highest priority for the delivery of quality POC diagnostic testing at PHC level. We recommend continual collaboration among all POC diagnostics stakeholders in the development of an accessible curriculum to improve providers’ competence and ensure sustainable quality delivery of POC diagnostic services in rural PHC clinics.

Both P-limitation and elevated temperature significantly increased coccolith malformations. Our results suggest that a temperature increase may intensify P-limitation due to a higher P-requirement to maintain growth and POC production rates, possibly reducing abundances in a warmer ocean. Under such a scenario C. pelagicus may decrease its calcification rate relative to photosynthesis, thus favouring CO2 sequestration over release. It seems unlikely that P-limitation by itself causes changes in the PIC / POC ratio in this species.

Here, we present data collected from the Great Calcite Belt, a calcite-rich band across the Southern Ocean surface, to compare the transformation of POC in the euphotic and mesopelagic zones of the water column. The 234Th-derived export fluxes and size-fractionated concentrations of POC, particulate inorganic carbon (PIC), and biogenic silica (BSi) were measured from the upper 1000 m of 27 stations across the Atlantic and Indian sectors of the Great Calcite Belt. POC export out of the euphotic zone was correlated with BSi export. PIC export was not, but did correlate positively with POC flux transfer efficiency. Moreover, regions of high BSi concentrations, which corresponded to regions with proportionally larger particles, exhibited higher attenuation of > 51 μm POC concentrations in the mesopelagic zone. The interplay among POC size partitioning, mineral composition, and POC attenuation suggests a more fundamental driver of POC transfer through both depth regimes in the Great Calcite Belt. In particular, we argue that diatom-rich communities produce large and labile POC aggregates, which not only generate high export fluxes but also drive more remineralization in the mesopelagic zone. We observe the opposite in communities with smaller calcifying phytoplankton, such as coccolithophores. We hypothesize that these differences are influenced by inherent differences in the lability of POC exported by different phytoplankton communities.

Background: The high burden of disease in South Africa presents challenges to public health services. Point-of-care (POC) technologies have the potential to address these gaps and improve healthcare systems. This study ascertained the acceptability and impact of POC CD4 testing on patients’ health and clinical management. Methods: We conducted a qualitative survey study with patients (n = 642) and healthcare providers (n = 13) at the Lancers Road (experienced POC) and Chesterville (non-experienced POC) primary healthcare (PHC) clinics from September 2015 to June 2016. Results: Patients (99%) at Lancers and Chesterville PHCs were positive about POC CD4 testing, identifying benefits: No loss/delay of test results (6.4%), cost/time saving (19.5%), and no anxiety (5.1%), and 58.2% were ready to initiate treatment. Significantly more patients at Chesterville than Lancers Road PHC felt POC would provide rapid clinical decision making (64.7% vs. 48.1%; p < 0.0001) and better clinic accessibility (40.4% vs. 24.7%; p < 0.0001) respectively. Healthcare providers thought same-day CD4 results would impact: Clinical management (46.2%), patient readiness (46.2%), and adherence (23.0%), and would reduce follow-up visits (7.7%), while 38.5% were concerned that further tests and training (15.4%) were required before antiretroviral therapy (ART) initiation. Conclusion: The high acceptability of POC CD4 testing and the immediate health, structural, and clinical management benefits necessitates POC implementation studies.

To investigate in prepubertal obese children (POC) the profile of chronic low-grade systemic inflammation (CLGSI) and its relation to homocysteinemia, 72 POC were evaluated for serum C-reactive protein (CRP) and amyloid A (SAA) levels, both markers of CLGSI, and plasma levels of total homocysteine (tHcy), an independent risk factor for adult atherosclerosis, in comparison to 42 prepubertal lean children (PLC). The main observations in POC were higher CRP levels compared to PLC, positive association of SAA levels to CRP levels, no association of CRP or SAA levels to tHcy levels. Thus, in POC, positively interrelated to each other, elevated CRP and unaltered SAA levels reveal a unique profile of the CLGSI, not explaining homocysteinemia-induced risk for future atherosclerosis.

473 Background: Treatment strategies in resectable gastric cancer (GC) patients include perioperative chemotherapy (PEC), postoperative chemoradiation therapy (POCR), and postoperative chemotherapy (POC). Treatment delay can lead to metastases with unfavorable outcomes. We aim to identify the distribution of patients upstaged to pathologically node positive disease (pLN+) stratified by time and the optimal treatment in this cohort. Methods: We analyzed resected GC patients in the National Cancer Database (2004-2016) with clinically node negative disease (cLN-). Time from diagnosis to first definitive therapy was stratified as ≤2, >2 to ≤4, and >4 weeks. The distribution of patients upstaged from cLN- to pLN+ disease, the concordance between cLN- and pathologically lymph node negative (pLN-) disease based on treatment strategy, and time to first definitive therapy was analyzed. POC and POCR were compared to evaluate their effect on overall survival (OS) in patients with cLN- upstaged to pLN+ disease. Kaplan-Meier test, log-rank test, multivariable Cox proportional hazards analysis (MVA) were performed. Results: Of the 4,828 gastric cancer patients with cLN- disease, 514 (10.65%) patients received PEC and 4,314 (89.35%) patients received upfront surgery followed by POCR/POC. Distribution of patients stratified by time between diagnosis to first definitive treatment - chemotherapy (PEC) or surgery (POC/POCR), for ≤2 weeks was 9.5% vs. 33%, >2 - ≤4 weeks was 28.4% vs. 24.2% and >4weeks was 62% vs 42.7%, respectively. Patients upstaged from cLN- to pLN+ disease in the PEC group was 43.37%(n=206), and 70%(n=2574) in the upfront surgery group (POC/POCR). Of the 206 (43.37%) patients upstaged from cLN- to pLN+ in the PEC cohort, the upstage rate was 7.77% when chemotherapy was started within 2 weeks of diagnosis, compared to 27.67% with >2 to ≤4 weeks, and 64.56% with >4 weeks. Of the 2,574 (70%) patients upstaged in the upfront surgery cohort (POCR/POC), 30.61% were upstaged in ≤2 weeks, 24.95% in >2 to ≤4 weeks, and 44.44% in ≥4 weeks. On MVA, patients with cLN- disease that were pLN- (POCR=766, POC=341) had no significant difference in OS when treated with POCR (HR 1.11, p=0.39) compared to POC. Patients with pLN+ (POCR=2300, POC=907) disease had an association with improved OS when treated with POCR (HR 0.78, p<0.001) compared to POC. Conclusions: When resectable gastric cancer patients undergoing upfront surgery are upstaged from clinically node negative to pathologically node positive disease, postoperative chemoradiation therapy may be the optimal treatment strategy compared to postoperative chemotherapy. Clinical nodal status may not be an optimal predictor of nodal disease.

Abstract. The ongoing rise in atmospheric pCO2 and the consequent increase in ocean acidification have direct effects on marine calcifying phytoplankton which potentially translates into altered carbon export. To date it remains unclear first, how nutrient ratio, in particular from coccolithophores preferred phosphate limitation, interacts with pCO2 on particulate carbon accumulation. Second, how direct physiological responses on the cellular level translate into a net population response. In this study cultures of Emiliania huxleyi were full-factorially exposed to two different N:P ratios (Redfield and high N:P) and three different pCO2 levels. Effects on net population particulate inorganic and organic carbon (PIC, POC) were measured after E. huxleyi cultures reached stationary phase. Thereby cell sizes and total cell abundance were taken into account. Corresponding to literature results show a significant negative cellular PIC and POC response which, however, was strongest under high N:P ratio. In contrast, net population PIC and POC accumulation was significantly attenuated under high N:P ratio. We suggest that less cellular nutrient accumulation allowed for higher cell abundances which compensated for the strong negative cellular PIC and POC response to pCO2 on the population level. Moreover, the design of this study also allowed following natural alteration of carbon chemistry through changing DIC and alkalinity. Our results suggest that at high initial pCO2 natural alteration of pCO2 during the experimental runtime was regulated by algal biomass. In contrast, at low initial pCO2 the PIC/POC ratio was responsible for changes in pCO2. Our results point to the fact that the physiological (i.e. cellular) PIC and POC response to ocean acidification cannot be linearly extrapolated to total population response and thus carbon export. It is therefore recommended to consider effects of nutrient limitation on cell physiology and translate these to net population carbon accumulation when predicting the influence of coccolithophores on both, the atmospheric pCO2 feedback and their function in carbon export mechanisms.

Acompanhando o processo de harmonização contabilística e de relato financeiro desenvolvido pela União Europeia (UE), foi criado, em Portugal, um novo corpo de normas coerentes com as normas internacionais de contabilidade em vigor na UE. O Sistema de Normalização Contabilística (SNC), que entrou em vigor no nosso país, em 2010, surge na linha de modernização contabilística ocorrida na UE, e baseia-se em princípios e não em regras, possibilitando, assim, ser um sistema aderente ao modelo do International Accounting Standards Board (IASB) adotado pela UE. As regras de contabilidade baseadas no Plano Oficial de Contabilidade (POC) foram substituídas pelas Normas Contabilísticas e de Relato Financeiro (NCRF) patentes no SNC. As NCRF trouxeram algumas alterações a nível contabilístico mas também a nível fiscal. A existência de novas terminologias e novas regras levou a que se tornasse útil analisar as mudanças ocorridas e o seu impacto nas Demonstrações Financeiras (DF’s) das empresas. Assim, neste trabalho analisa-se o POC e SNC e enumeram-se as diferenças entre os dois normativos. Posteriormente, é feita uma análise fiscal percebendo-se de que forma as regras do Sistema Tributário foram influenciadas pela entrada em vigor do SNC e as alterações, nele, ocorridas. Following the harmonization process of accounting and financial reporting developed by the European Union (EU) was created in Portugal, a new set of rules consistent with the international accounting standards in force in the EU. The Accounting Standardization System (SNC), which became effective in our country in 2010 arises in line with the accounting modernization occurred in the EU, and is based on principles instead of rules; thus enables a system adherent to the International Accounting Standards Board (IASB) model adopted by the EU. The accounting rules based on the National Plan of Accounts (POC) were replaced by the Accounting and Financial Reporting Standards (NCRF) as defined in the SNC. The NCRF brought some changes both in accounting and fiscal systems. The existence of new terminology and rules justified the analysis of the changes occurred and their impact on the Financial Statements of the companies. Thus, this thesis analyzes the POC and SNC and lists the differences between the two standards. Thereafter, a fiscal analysis will be undertaken to understand in what extent the Tax System was influenced by the entry into force of the SNC and the changes that took place. Finally, it is developed an empirical study, where is analyzed and perceived the impact of the changes brought by the SNC into the Financial Statements of the Portuguese entities.

A informação contabilística desde sempre foi e será uma ferramenta crucial no apoio à tomada de decisão para qualquer organização. Por isso, também no Setor Público Administrativo (SPA), um sistema de informação contabilístico útil e oportuno, ajuda os seus utilizadores avaliar e a tomar decisões de maneira mais apropriada. Este estudo objetiva identificar se a informação contabilística resultante da aplicação obrigatória do POCP no SPA e do POC-Educação no ensino superior público em particular, é utilizada como apoio à gestão de uma instituição de ensino superior público, quer através da sua análise direta quer de indicadores de gestão calculados a partir dela. Para a concretização do objetivo proposto, realizou-se uma pesquisa bibliográfica e exploratória, cuja abordagem teve preponderância qualitativa via de estudo de caso. O instrumento de recolha de dados utilizado foi um inquérito, respondido pelos Órgãos de Gestão e Unidades de caráter Científico ou Pedagógico da ESCE do IPS, que teve um nível de respostas de 54%. Os resultados obtidos evidenciaram que todos os respondentes reconhecem importância aos indicadores apresentados no inquérito que são calculados com base na informação contabilística, bem como a necessidade desses indicadores serem comparados entre Escolas do IPS e também a nível nacional, nas Instituições de Ensino Superior. Porém, no desempenho dos seus cargos de gestão, no respetivo Órgão/Unidade, apenas cerca de 33% dos respondentes dispõem de informação com base na informação contabilística e a 61,5% dos respondentes são disponibilizados indicadores cujo cálculo, para ser efetuado, não necessita de informação contabilística.
Abstract: Since ever the accounting information was and will be a crucial tool in the support for decision in any company. On this, also in the public administrative sector, having a useful and timely accounting system information can help the users of this information evaluate and take decisions in more appropriate way. The present study has the propose to identify if the accounting information resultant from the mandatory application of POCP (The Oficial Plan of Public Accounts) in the public administrative sector and the POC-Education (the official plan of public accounts – education) in the public high education, in particular, is used as support to management of one institution of high education, through either a direct analysis through either management indicators calculated from this. To achieve the main objective was conducted a literature research and exploratory, whose approach had a qualitative preponderance through a case study. The data collection instrument was a survey and the answers were given by the Governing Bodies Management and character Units Scientific or Pedagogical ESCE IPS, which had a level of 54% answers. The obtained results showed that: The results showed that all the respondents to the inquiry recognize the importance of the indicators presented in the inquiry which are calculated on the basis of accounting information, as well as the need of those indicators be compared between IPS schools and also at national level, in higher education institutions. However, in the performance of its management positions, in the respective Body/ Unit, only about 33% of the respondents has information based on data accounting (or accounting information) and the 61.5% of respondents disposes of indicators that for its calculation be done, do not need accounting information.

O estudo em apreço pretende analisar as implicações para os TOC decorrentes das recentes alterações à normalização contabilística em Portugal. Para uma melhor percepção e enquadramento da questão de investigação “Do POC ao SNC: Que implicações para os TOC?”, realizaram-se pesquisas de artigos científicos na área do comportamento dos profissionais de contabilidade, que serviram de base à elaboração do inquérito por questionário. O inquérito por questionário foi elaborado numa aplicação informática disponível via Internet, permitindo responder à questão de investigação e hipóteses formuladas. O mesmo foi aplicado a profissionais da área das Ciências Empresariais. Com este estudo espera-se conseguir perceber quais as principais dificuldades e implicações na profissão de TOC decorrentes da adopção do SNC, de forma a no futuro próximo apoiar, ajustar, informar, consciencializar, organizar e preparar melhor os profissionais para situações idênticas. Este assunto reveste-se de particular importância para os TOC, para a OTOC e para a comunidade académica, atendendo ao facto de que as mudanças são significativas e que importa saber como é que os profissionais mais implicados estão a reagir. Os resultados obtidos através do estudo empírico revelam, essencialmente, que os TOC sentiram uma grande mudança com a entrada em vigor do SNC, que necessitaram de assistir a muitas mais horas de formação do que em anos anteriores, que os conceitos se alteraram, a forma de pensar se alterou, que as formações ministradas pela OTOC são de qualidade, que as áreas temáticas onde se sentem mais dificuldades são na aplicação do justo valor e nos testes de imparidade, que o SNC permitirá maior liberdade de pensamento e que o TOC deve ser o parceiro privilegiado do empresário. Contudo, os profissionais da contabilidade demonstram uma capacidade de adaptação positiva, pois na sua maioria sentem poucas dificuldades na transição do POC para o SNC. The present study aims to analyze the implications for the Accountants from the recent changes to accounting standards in Portugal.For a better understanding and framing of the research question "From POC to the SNC: What Implications for the Accountants?", there were research of scientific articles in the field of accounting professionals behaviour, that were the basis for development the surveyquestionnaire. The survey questionary was designed in a computer application available on the Internet, allowing answerin the research question and hypotheses. The same was applied to professionals in the field of Management Sciences. With this study it is expected to understand the main difficulties and implications in the accountant profession of resulting from the adoption of the SNC in order to support, adjust, inform, raise awareness, organize and better prepare professionals to similar situations in the near future. This issue is of particular importance to the Accountants, the Accountants Association and the academic community, given the fact that the changes are significant and it is important to know how the professionals most involved are responding. The results obtained from the empirical study shows, essentially, that the Accountants felt a big change with the entry into force of the SNC, which were needded many training hours than in previous years, the concepts that have changed, the wayof thinking has changed, that the training provided by Accountants Association have quality, the thematic fields where they feel more difficulties is the application of fair value and impairment tests, the SNC will allow greater freedom of thought and the Accountants should bethe privileged partner of the entrepreneur. However, the accounting professionals demonstrate a positive ability to adapt, because mostly they feel minor difficulties in the transition of the POC to the SNC.

Abstract:
This dissertation consists of three studies dealing with particulate organic carbon (POC). The first study describes the temporal and spatial variability of particulate matter (PM) and POC, and physical processes that affect the distribution of PM and POC with synchronous remote sensing data. The purpose of the second study is to develop POC algorithms in the Gulf of Mexico based on satellite data using numerical methods and to compare POC estimates with spectral radiance. The purpose of the third study is to investigate climatological variations from the temporal and spatial POC estimates based on SeaWiFS spectral radiance and physical processes, and to determine the physical mechanisms that affect the distribution of POC in the Gulf of Mexico. For the first and second studies, hydrographic data from the Northeastern Gulf of Mexico (NEGOM) study were collected on each of 9 cruises from November 1997 to August 2000 across 11 lines. Remotely sensed data sets were obtained from NASA and NOAA using algorithms that have been developed for interpretation of ocean color data from various satellite sensors. For the third study, we use the time-series of POC estimates, sea surface temperature (SST), sea surface height anomaly (SSHA), sea surface wind (SSW), and precipitation rate (PR) that might cause climatological variability and physical processes. The distribution of surface PM and POC concentrations were affected by one or more factors such as river discharge, wind stress, stratification, and the Loop Current/Eddies. To estimate POC concentration, empirical and model-based approaches were used using regression and principal component analysis (PCA) methods. We tested simulated data for reasonable and suitable algorithms in Case 1 and Case 2 waters. Monthly mean values of POC concentrations calculated with PCA algorithms. The spatial and temporal variations of POC and physical forcing data were analyzed with the empirical orthogonal function (EOF) method. The results showed variations in the Gulf of Mexico on both annual and inter-annual time scales.

Abstract:
Researchers have documented the prevalence of alcohol use disorder among various populations. For example, researchers have argued that those who are sexual minorities, gender minorities, or racial or ethnic minorities may be disproportionately affected by alcohol use disorder. Furthermore, researchers have explored minority stress-related factors and minority strengths-related factors as predictors of alcohol use disorder among these populations. However, researchers in few studies have described the prevalence of alcohol use disorder among LGBTQ+ People of Color who may have sexual minority, gender minority, and racial or ethnic minority identities. The purpose of the current study was two-fold: (1) to address existing literature gaps by documenting prevalence of alcohol use disorder among LGBTQ+ People of Color, and (2) to explore whether minority-stress and minority-strengths factors predicted alcohol use via an intersectional framework. I recruited a sample of 267 LGBTQ+ POC and had individuals fill out the following measures: LGBT-POC Microaggression Scale, The Multidimensional Scale of Perceived Social Support, and the Collective Identity Self-Esteem Scale, and a Likert-type measure of LGBTQ+ POC Community Connectedness designed for this study. I conducted a hierarchical logistic regression analysis to determine whether LGBTQ+ POC minority stress and LGBTQ+ POC minority strengths predicted alcohol use. Neither LGBTQ+ POC minority stress nor the LGBTQ+ POC minority strengths variables were significant predictors in the current study. The findings from this study may help describe the specific social context for alcohol use among LGBTQ+ People of Color which would assist researchers and practitioners with their knowledge of how alcohol use affects LGBTQ+ People of Color.

Aquesta tesi doctoral té com a objectiu el desenvolupament de diversos biosensors que operen sense necessitat de marcatge addicional basats en dispositius plasmònics òptics per a la detecció directa de medicaments o biomarcadors relacionats amb diferents malalties i que són analitzats directament en mostres humanes com plasma, sèrum, orina o esput. Aquests dispositius biosensors ofereixen un sens fi de beneficis com és la seva alta sensibilitat, facilitat d’operació, l’obtenció de dades quantitatives, detecció sense marcatge en temps real, i comunament només necessiten d’un petit volum de mostra. Tot això converteix els biosensors plasmònics en eines analítiques molt adequades per al diagnòstic de malalties, el control de la medicació o el seguiment de teràpies personalitzades. El nostre grup d’investigació ha demostrat amb èxit la implementació de biosensors òptics basats en plasmònica i en fotònica de silici, inclòs el desenvolupament complet de bioaplicaciones, el que ha aplanat el camí de la seva futura transferència tecnològica per a la seva implementació com a dispositius Point-of-Care ( POC). Els biosensors desenvolupats en aquesta Tesi inclouen la seva optimització i validació completa amb mostres reals, exemplificant alguns desafiaments clínics en els quals aquests biosensors plasmònics poden superar importants limitacions de les tècniques d’anàlisi convencionals actuals, mostrant el seu potencial i versatilitat com a futurs dispositius POC per ser usats en les unitats d’atenció primària en salut o fins i tot en l’entorn domèstic per al propi autocontrol per part dels pacients. La tesi està organitzada en sis capítols. El capítol 1 conté la introducció dels conceptes bàsics i l’estat de l’art sobre els avenços actuals en les tècniques de diagnòstic i control de malalties i / o teràpies i el paper que exerceixen els biosensors per millorar-los. El capítol 2 inclou una descripció detallada de les plataformes biosensoras emprades i una descripció general dels processos metodològics. El Capítol 3 descriu el desenvolupament d’un dispositiu nanoplasmónico per al control terapèutic de l’medicament acenocumarol, un anticoagulant comunament administrat directament en plasma humà. El Capítol 4 es centra en el desenvolupament d’un biosensor plasmónico que serveixi com a control de la dieta lliure de gluten que han de portar els pacients celíacs. El Capítol 5 descriu les estratègies desenvolupades per a la detecció de dos biomarcadors per al diagnòstic primerenc de tuberculosi en mostres d’esput. Finalment, el Capítol 6 explora la detecció de quatre autoanticossos específics associats amb l’aparició de l’tumor directament en el sèrum humà com biomarcadors potencials per al diagnòstic primerenc de el càncer colorectal.
Esta Tesis Doctoral tiene como objetivo el desarrollo de diversos biosensores que operan sin necesidad de marcaje adicional basados en dispositivos plasmónicos ópticos para la detección directa de medicamentos o biomarcadores relacionados con diferentes enfermedades y que son analizados directamente en muestras humanas como plasma, suero, orina o esputo. Estos dispositivos biosensores ofrecen un sinnúmero de beneficios como es su alta sensibilidad, facilidad de operación, la obtención de datos cuantitativos, detección sin marcaje en tiempo real, y comúnmente sólo necesitan de un pequeño volumen de muestra. Todo esto convierte a los biosensores plasmónicos en herramientas analíticas muy adecuadas para el diagnóstico de enfermedades, el control de la medicación o el seguimiento de terapias personalizadas. Nuestro grupo de investigación ha demostrado exitosamente la implementación de biosensores ópticos basados en plasmónica y en fotónica de silicio, incluido el desarrollo completo de bioaplicaciones, lo que ha allanado el camino de su futura transferencia tecnológica para su implementación como dispositivos Point-of-Care (POC). Los biosensores desarrollados en esta Tesis incluyen su optimización y validación completa con muestras reales, ejemplificando algunos desafíos clínicos en los que dichos biosensores plasmónicos pueden superar importantes limitaciones de las técnicas de análisis convencionales actuales, mostrando su potencial y versatilidad como futuros dispositivos POC para ser usados en las unidades de atención primaria en salud o incluso en el entorno doméstico para el propio autocontrol por parte de los pacientes. La tesis está organizada en seis capítulos. El Capítulo 1 contiene la introducción de los conceptos básicos y el estado del arte sobre los avances actuales en las técnicas de diagnóstico y control de enfermedades y/o terapias y el papel que desempeñan los biosensores para mejorarlos. El Capítulo 2 incluye una descripción detallada de las plataformas biosensoras empleadas y una descripción general de los procesos metodológicos. El Capítulo 3 describe el desarrollo de un dispositivo nanoplasmónico para el control terapéutico del medicamento acenocumarol, un anticoagulante comúnmente administrado directamente en plasma humano. El Capítulo 4 se centra en el desarrollo de un biosensor plasmónico que sirva como control de la dieta libre de gluten que deben llevar los pacientes celíacos. This Doctoral Thesis aims to the development of several label-free biosensing analytical strategies integrated within optical plasmonic devices for the direct detection of drugs or biomarkers related to different diseases in biological samples such as plasma, serum, urine, and sputum. These biosensor devices offer several benefits like their high sensitivity, ease of operation, quantitative data, label-free operation, and real-time detection, and commonly require a small sample volume. All this turn plasmonic biosensors into well-suited analytical tools for diagnosing diseases, monitoring medication, or for personalized therapies follow-up. Our research group has extensively demonstrated the successful conjunction of novel in-house optical biosensor configurations (like plasmonic and photonic-based designs) with the full demonstrations of bioapplications, which has paved the way for their potential technological transfer as Point-of-Care devices (POC) for clinical diagnostics. The biosensor assays here implemented, which include their full optimization and validation with real samples, exemplify clinical challenges where such biosensors can overcome limitations of current conventional analytical techniques. The results show the potential and versatility that plasmonic biosensors can offer as future POC devices placed in primary healthcare units or even in the household environment for patients’ self-monitoring. This thesis is organized into six chapters. Chapter 1 is the introductory one, which explains the basic concepts and the state of the art of the current advances in diagnosis and monitoring techniques of diseases and/or therapies and the role of biosensors to improve them. Chapter 2 includes a detailed description of the biosensor platforms employed and a general description of the methodological processes. Chapter 3 is related to the development of a nanoplasmonic device for the therapeutic monitoring of the drug acenocoumarol, a commonly administered anticoagulant, directly in human plasma. Chapter 4 focuses on the implementation of a plasmonic biosensor that monitors the gluten-free diet in urine in celiac patients. Chapter 5 describes the biosensing strategies developed for the detection of two biomarkers for the early diagnosis of tuberculosis in sputum samples. Finally, Chapter 6 explores the detection of four specific autoantibodies associated with the tumor onset directly in human serum as potential biomarkers for the early detection of colorectal cancer.

Idag är det relativt utbrett bland företag att använda flera försäljningskanaler. Flera kanaler möjliggör att företaget kan nå flera kundsegment och på så vis öka sin lönsamhet. POC Sweden AB designar och marknadsför alpina skyddsprodukter och har idag planer på att använda flera försäljningskanaler. Deras potentiella försäljningskanaler utgörs av fysiska återförsäljare, konceptbutiker och e-handel. POC:s mål med de potentiella försäljningskanalerna är att öka sin lönsamhet utan att varumärkets image reduceras. När ett företag använder sig av flera försäljningskanaler bör företaget vara medvetna om deras för- respektive nackdelar och hur de påverkar företagets verksamhet. POC har därför framfört ett intresse av att författarna upprättar en prioriteringsordning av företagets potentiella försäljningskanaler utifrån deras mål. För att göra detta möjligt har författarna använt sig av teorier som berör distributionsområdet samt inhämtat praktisk information genom två kvalificerade intervjuobjekt. Efter att författarna analyserat ovanstående information har de kommit fram till följande prioriteringsordning; (1) Fysiska återförsäljare, (2) E-handel, (3) Konceptbutiker. Fysiska återförsäljare i form av sportkedjor har alltså första prioritet då de möjliggör högst lönsamhet utan att varumärkets image reduceras.


Today it’s relatively widely spread among companies to use many sales channels because it makes it possible for a company to reach more customer segments, and in that way increase revenues. POC Sweden AB, which designs and markets alpine safety products, currently has plans to use multiple channels. Their potential sales channels consist of physical retailers, concept stores, and e-commerce. The company’s goal with their potential sales channels is to increase their revenues while not reducing their brand image. When a company uses multiple sales channels, they should be aware of the channel’s positive and negative characteristics, and how these characteristics will affect the company. POC has therefore requested that the authors establish a ranking of the company’s potential sales channels based on their goal of increasing revenue while not reducing their brand image. To make this possible the authors have used theories which cover the distribution area and collected practical information from two qualified interviewees. After analyzing the information above, the authors have come to the conclusion of the following priority ranking; (1) Physical Retailers, (2) E-Commerce, (3) Concept stores. Physical retailers in form of sport chains have first priority, as they make it possible to generate the highest revenue without reducing the brand image.

Abstract:
Many ditransitive events can be expressed in German using either the indirect object construction (IOC) or the prepositional object construction (POC). While most previous research on ditransitives in German has focused on the IOC, very little attention has been paid to the much rarer POC or to individual verb biases for the IOC vs. the POC. We investigated structural preferences of native speakers of German using elicited production and acceptability judgment studies. The results show that the POC is not as restricted as previously thought, and that different modalities (i.e., production vs. comprehension) reveal different aspects of verb biases, which can be easily missed when relying on one type of data.

Basic principles and main characteristics of three approaches for structure synthesis of robot mechanisms (the screw theory-based approach, the displacement subgroup-based approach and the approach based on position and orientation characteristic (in short, POC) ) are studied and compared in this paper. The comparison deals with the mathematical tools, the symbolic representation of mechanism topological structure, the mathematical representation of POC of the output motion link with respect to the frame link, the basic equations for structure synthesis of serial and parallel mechanisms and relevant operation rules, and the characteristics of their synthesized mechanisms, etc. This comparative study shows that the POC-based approach is totally different from the other two approaches: (1) the POC-based approach requires only simple mathematical tools (such as vector algebra, set theory, etc), (2) the POC-based approach is conceptually simpler and therefore easier to understand and to use, and (3) the POC-based approach is more general.

Abstract Early and accurate detection of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) at the point-of-care (POC) is crucial for reducing the transmission of coronavirus disease 2019 (COVID-19). To address this need, we have developed a valve-enabled lysis, paper-based RNA enrichment, and RNA amplification device (VLEAD) for detecting SARS-CoV-2. We have combined VLEAD with a smart coffee mug for sample preparation, nucleic acid isothermal amplification, and colorimetric detection using a smartphone camera or a naked eye. VLEAD enables two critical functions required for POC testing: sample preparation and detection. Since the reagents can be pre-packaged in the device, all operational steps can be carried out at POC. We have demonstrated the functions of the device by analyzing samples spiked with heat-inactivated SARS-CoV-2, which were obtained from BEI Resources. This rapid and highly sensitive POC platform for SARS-CoV-2 detection has a potential to help reduce COVID-19 transmission.
