Abstract:
The BTB domain is a protein–protein interaction motif found throughout eukaryotes. Here we report the identification of two closely related BTB domain-containing proteins, POZ/BTB CONTAINING-PROTEIN 1 (POB1) and POB2, as negative regulators of defense against pathogens. In yeast two-hybrid assays, POB1 and POB2 dimerize through their BTB domains. The pob1–1 pob2–1 double mutant plants exhibited enhanced resistance against the fungal pathogen Botrytis cinerea and the oomycete pathogen Hyaloperonospora arabidopsidis Noco2. Double knockout, but not single mutants of pob1–1 and pob2–1, displayed enhanced sensitivity to growth inhibition by jasmonic acid (JA). In addition, expression of the JA-responsive defensin gene PDF1.2 is enhanced in the pob1–1 pob2–1 double mutant. Our data suggest that POB1 and POB2 function redundantly as negative regulators of JA-mediated pathogen resistance responses.

Abstract:
Odorant-binding proteins (OBPs) are a group of small and soluble proteins present in both vertebrates and insects. They have a high level of structural stability and bind to a large spectrum of odorant molecules. In the environmental field, benzene is the most dangerous compound among the class of pollutants named BTEX (benzene, toluene, ethylbenzene, and xylene). It has several effects on human health and, consequently, it appears to be important to monitor its presence in the environment. Commonly, its detection requires the use of very sophisticated and time-consuming analytical techniques (GC-MS, etc.) as well as the presence of specialized personnel. Here, we present the application of an odorant-binding protein (pOBP) isolated from pigs as a molecular recognition element (MRE) for a low-energy impedenziometric biosensor for outdoor and real-time benzene detection. The obtained results show that the biosensor can detect the presence of 64 pM (5 µg/m3) benzene, the limit value of exposure for human health set by the European Directive 2008/50/EC.

Abstract:
The fission yeast pob1 gene encodes a protein of 871 amino acids carrying an SH3 domain, a SAM domain, and a PH domain. Gene disruption and construction of a temperature-sensitivepob1 mutant indicated that pob1 is essential for cell growth. Loss of its function leads to quick cessation of cellular elongation. Pob1p is homologous to two functionally redundant Saccharomyces cerevisiaeproteins, Boi1p and Boi2p, which are necessary for cell growth and relevant to bud formation. Overexpression of pob1inhibits cell growth, causing the host cells to become round and swollen. In growing cells, Pob1p locates at cell tips during interphase and translocates near the division plane at cytokinesis. Thus, this protein exhibits intracellular dynamics similar to F-actin patches. However, Pob1p constitutes a layer, rather than patches, at growing cell tips. It generates two split discs flanking the septum at cytokinesis. The pob1-defective cells no longer elongate but swell gradually at the middle, eventually assuming a lemon-like morphology. Analysis using the pob1-ts allele revealed that Pob1p is also essential for cell separation. We speculate that Pob1p is located on growing plasma membrane, possibly through the function of actin patches, and may recruit proteins required for the synthesis of cell wall.

ABSTRACT The bacterium Acinetobacter baylyi uses the branched β-ketoadipate pathway to metabolize aromatic compounds. Here, the multiple-level regulation of expression of the pca-qui operon encoding the enzymes for protocatechuate and quinate degradation was studied. It is shown that both activities of the IclR-type regulator protein PcaU at the structural gene promoter pcaIp, namely protocatechuate-dependent activation of pca-qui operon expression as well as repression in the absence of protocatechuate, can be observed in a different cellular background (Escherichia coli) and therefore are intrinsic to PcaU. The regulation of PcaU expression is demonstrated to be carbon source dependent according to the same pattern as the pca-qui operon. The increase of the pcaU gene copy number leads to a decrease of the basal expression at pcaIp, indicating that the occupancy of the PcaU binding site is well balanced and depends on the concentration of PcaU in the cell. Luciferase is used as a reporter to demonstrate strong repression of pcaIp when benzoate, a substrate of the catechol branch of the pathway, is present in addition to substrates of the protocatechuate branch (cross-regulation). The same repression pattern was observed for promoter pcaUp. Thus, three promoters involved in gene expression of enzymes of the protocatechuate branch (pobAp upstream of pobA, pcaIp, and pcaUp) are strongly repressed in the presence of benzoate. The negative effect of protocatechuate on pobA expression is not based on a direct sensing of the metabolite by PobR, the specific regulator of pobA expression.

ABSTRACT Acinetobacter PobR and PcaU are transcriptional activators that closely resemble each other in primary structure, DNA-binding sites, metabolic modulators, and physiological function. PobR responds to the inducer-metabolite p-hydroxybenzoate and activates transcription of pobA, the structural gene for the enzyme that converts p-hydroxybenzoate to protocatechuate. This compound, differing fromp-hydroxybenzoate only in that it contains an additional oxygen atom, binds to PcaU and thereby specifically activates transcription of the full set of genes for protocatechuate catabolism. Particular experimental attention has been paid to PobR and PcaU fromAcinetobacter strain ADP1, which exhibits exceptional competence for natural transformation. This trait allowed selection of mutant strains in which pobR function had been impaired by nucleotide substitutions introduced by PCR replication errors. Contrary to expectation, the spectrum of amino acids whose substitution led to loss of function in PobR shows no marked similarity to the spectrum of amino acids conserved by the demand for continued function during evolutionary divergence of PobR, PcaU, and related proteins. Surface plasmon resonance was used to determine the ability of mutant PobR proteins to bind to DNA in the pobA-pobR intergenic region. Deleterious mutations that strongly affect DNA binding all cluster in and around the PobR region that contains a helix-turn-helix motif, whereas mutations causing defects in the central portion of the PobR primary sequence do not seem to have a significant effect on operator binding. PCR-generated mutations allowing PobR to mimic PcaU function invariably caused a T57A amino acid substitution, making the helix-turn-helix sequence of PobR more like that of PcaU. The mutant PobR depended on p-hydroxybenzoate for its activity, but this dependence could be relieved by any of six amino acid substitutions in the center of the PobR primary sequence. Independent mutations allowing PcaU to mimic PobR activity were shown to be G222V amino acid substitutions in the C terminus of the 274-residue protein. Together, the analyses suggest that PobR and PcaU possess a linear domain structure similar to that of LysR transcriptional activators which largely differ in primary structure.

ABSTRACT Transcriptional control of carbon source preferences by Acinetobacter sp. strain ADP1 was assessed with a pobA::lacZ fusion during growth on alternative substrates. The pobA-encoded enzyme catalyzes the first step in the degradation of 4-hydroxybenzoate, a compound consumed rapidly as a sole carbon source. If additional aromatic carbon sources are available, 4-hydroxybenzoate consumption is inhibited by unknown mechanisms. As reported here, during growth on aromatic substrates, pobA was not expressed despite the presence of 4-hydroxybenzoate, an inducer that normally causes the PobR regulator to activate pobA transcription. Growth on organic acids such as succinate, fumarate, and acetate allowed higher levels of pobA expression. In each case, pobA expression increased at the end of the exponential growth phase. Complex transcriptional regulation controlled 4-hydroxybenzoate catabolism in multisubstrate environments. Additional studies focused on the wild-type preference for benzoate consumption prior to 4-hydroxybenzoate consumption. These compounds are degraded via the catechol and protocatechuate branches of the β-ketoadipate pathway, respectively. Here, mutants were characterized that degraded benzoate and 4-hydroxybenzoate concurrently. These mutants lacked the BenM and CatM transcriptional regulators that normally activate genes for benzoate catabolism. A model is presented in which BenM and CatM prevent pobA expression indirectly during growth on benzoate. These regulators may affect pobA expression by lowering the PcaK-mediated uptake of 4-hydroxybenzoate. Consistent with this model, BenM and CatM bound in vitro to an operator-promoter fragment controlling the expression of several pca genes, including pcaK. These studies provide the first direct evidence of transcriptional cross-regulation between the distinct but analogous branches of the β-ketoadipate pathway.

Abstract Spt16 and Pob3 form stable heterodimers in Saccharomyces cerevisiae, and homologous proteins have also been purified as complexes from diverse eukaryotes. This conserved factor has been implicated in both transcription and replication and may affect both by altering the characteristics of chromatin. Here we describe the isolation and properties of a set of pob3 mutants and confirm that they have defects in both replication and transcription. Mutation of POB3 caused the Spt− phenotype, spt16 and pob3 alleles displayed severe synthetic defects, and elevated levels of Pob3 suppressed some spt16 phenotypes. These results are consistent with previous reports that Spt16 and Pob3 act in a complex that modulates transcription. Additional genetic interactions were observed between pob3 mutations and the genes encoding several DNA replication factors, including POL1, CTF4, DNA2, and CHL12. pob3 alleles caused sensitivity to the ribonucleotide reductase inhibitor hydroxyurea, indicating a defect in a process requiring rapid dNTP synthesis. Mutation of the S phase checkpoint gene MEC1 caused pob3 mutants to lose viability rapidly under restrictive conditions, revealing defects in a process monitored by Mec1. Direct examination of DNA contents by flow cytometry showed that S phase onset and progression were delayed when POB3 was mutated. We conclude that Pob3 is required for normal replication as well as for transcription.

Rho GTPases regulate the actin cytoskeleton in all eukaryotes. Fission yeast Cdc42 is involved in actin cable assembly and formin For3 regulation. We isolated cdc42-879 as a thermosensitive strain with actin cable and For3 localization defects. In a multicopy suppressor screening, we identified pob1+as suppressor of cdc42-879 thermosensitivity. Pob1 overexpression also partially restores actin cables and localization of For3 in the mutant strain. Pob1 interacts with Cdc42 and this GTPase regulates Pob1 localization and/or stability. The C-terminal pleckstrin homology (PH) domain of Pob1 is required for Cdc42 binding. Pob1 also binds to For3 through its N-terminal sterile alpha motif (SAM) domain and contributes to the formin localization at the cell tips. The previously described pob1-664 mutant strain (Mol. Biol. Cell. 10, 2745–2757, 1999), which carries a mutation in the PH domain, as well as pob1 mutant strains in which Pob1 lacks the N-terminal region (pob1ΔN) or the SAM domain (pob1ΔSAM), have cytoskeletal defects similar to that of cdc42-879 cells. Expression of constitutively active For3DAD* partially restores actin organization in cdc42-879, pob1-664, pob1ΔN, and pob1ΔSAM. Therefore, we propose that Pob1 is required for For3 localization to the tips and facilitates Cdc42-mediated relief of For3 autoinhibition to stimulate actin cable formation.

Abstract:
Background: The authors have sometimes encountered postoperative new-onset pseudoparalysis (PONP) after arthroscopic repair for large to massive rotator cuff tear (ARCR). As there are insufficient data regarding PONP, the authors aimed to determine whether such a condition is characterized by obvious risk factors, as well as to assess the conditions under which it can be reversed. Purpose: To evaluate the predictors of PONP development after ARCR and its reversal and to integrate these predictors into a scoring system that can be applied in clinical practice. Study Design: Case-control study; Level of evidence, 3. Methods: The authors retrospectively analyzed 430 consecutive ARCRs performed between March 2010 and May 2016. Patients were stratified according to the occurrence of PONP. To determine the risk factors of PONP, the authors compared the PONP and control groups (patients who did not experience PONP) in terms of demographic, clinical, and operative characteristics. Results were expressed in terms of odds ratios (ORs). Results: The incidence of PONP was 6.0% (26 of 430). Predictors of PONP included torn subscapularis (OR, 7.875; 95% CI, 1.780-34.956; P = .007), ≥30-mm retraction of the torn supraspinatus (OR, 4.657; 95% CI, 1.537-14.203; P = .007), age ≥65 years (OR, 3.865; 95% CI, 1.465-10.040; P = .006), and preoperative stiffness (OR, 2.954; 95% CI, 1.212-7.159; P = .017). The reversal rate of PONP was 65.4% (17 of 26), and the mean time to PONP reversal was 10 months. The occurrence and reversal of PONP were not related to retear. Fatty infiltration of the supraspinatus lower than Goutallier grade 3 was the sole predictor of reversal (OR, 22.000; 95% CI, 1.857-260.648; P = .014). Conclusion: Although the incidence of PONP after ARCR was low (6.0%), the risk of PONP was higher in patients with a torn subscapularis, larger supraspinatus tear size, older age, and preoperative stiffness. The mean time from PONP onset to reversal was 10 months, and PONP reversal was affected solely by fatty infiltration of the supraspinatus. Therefore, surgeons should consider the risk for PONP and attempt to identify high-risk patients before rotator cuff repair.

Introduction: Resorbable synthetic scaffolds are promising for different indications, especially in the context of bone regeneration. However, they require additional biological components to enhance their osteogenic potential. In addition to different cell types, autologous blood-derived matrices offer many advantages to enhance the regenerative capacity of biomaterials. The present study aimed to analyze whether biologization of a PCL-mesh coated using differently centrifuged Platelet rich fibrin (PRF) matrices will have a positive influence on primary human osteoblasts activity in vitro. A polymeric resorbable scaffold (Osteomesh, OsteoporeTM (OP), Singapore) was combined with differently centrifuged PRF matrices to evaluate the additional influence of this biologization concept on bone regeneration in vitro. Peripheral blood of three healthy donors was used to gain PRF matrices centrifuged either at High (710× g, 8 min) or Low (44× g, 8 min) relative centrifugal force (RCF) according to the low speed centrifugation concept (LSCC). OP-PRF constructs were cultured with pOBs. POBs cultured on the uncoated OP served as a control. After three and seven days of cultivation, cell culture supernatants were collected to analyze the pOBs activity by determining the concentrations of VEGF, TGF-β1, PDGF, OPG, IL-8, and ALP- activity. Immunofluorescence staining was used to evaluate the Osteopontin expression of pOBs. After three days, the group of OP+PRFLow+pOBs showed significantly higher expression of IL-8, TGF-ß1, PDGF, and VEGF compared to the group of OP+PRFHigh+pOBs and OP+pOBs. Similar results were observed on day 7. Moreover, OP+PRFLow+pOBs exhibited significantly higher activity of ALP compared to OP+PRFHigh+pOBs and OP+pOBs. Immunofluorescence staining showed a higher number of pOBs adherent to OP+PRFLow+pOBs compared to the groups OP+PRFHigh+pOBs and OP+pOBs. To the best of our knowledge, this study is the first to investigate the osteoblasts activity when cultured on a PRF-coated PCL-mesh in vitro. The presented results suggest that PRFLow centrifuged according to LSCC exhibits autologous blood cells and growth factors, seem to have a significant effect on osteogenesis. Thereby, the combination of OP with PRFLow showed promising results to support bone regeneration. Further in vivo studies are required to verify the results and carry out potential results for clinical translation.

Abstract:
Background: Prediction of post-pancreaticoduodenectomy (PD) morbidity is difficult, especially in the early postoperative period when CT (Computed Tomography) scans are not available. Elevated serum amylase and lipase in postoperative day 0 or 1 may be used to define postoperative acute pancreatitis (POAP), but the existing literature does not agree on whether POAP is significantly associated with postoperative pancreatic fistula (POPF). Methods: We analyzed the data obtained from a previously published randomized controlled trial. POAP was defined as elevations in serum amylase above 110 U/L on postoperative day 0 or 1. Clinically relevant POAP (CR-POAP) was defined as elevations in C-reactive protein level (CRP) on postoperative day 2 in those with POAP. Postoperative complications including severe complications (Clavien–Dindo ≥ IIIa), POPF, and clinically relevant POPF (CR-POPF) were analyzed. Results: In 246 patients, POAP did not show significant associations with total postoperative complications (odds ratio (OR) 0.697; 95% CI, 0.360–1.313; p = 0.271), severe complications (OR 0.647; 95% CI, 0.258–1.747; p = 0.367), and CR-POPF (OR 0.998; 95% CI, 0.310–3.886; p = 0.998) in multivariable analysis. Conclusions: In patients undergoing PD, POAP was not significantly associated with postoperative complications including POPF. Caution should be taken when using POAP as a predictor of POPF.

Abstract:
The aim: To determine the effectiveness of the combination of the percutaneous transluminal rotational atherectomy (PTRA) techniques with plain old balloon angioplasty (POBA) and POBA monotherapy (POBA-mono) in endovascular treatment (ET) of patients with occlusive-stenotic lesions below the knee (BTK) arteries. Materials and methods: We studied two groups, the main included patients (n=9) aged who underwent PTRA and POBA of BTK arteries, and the comparative group (POBA-mono) of patients (n=24) who underwent POBA monotherapy of BTK arteries. Results: Thrombosis of the reconstructed segments in the target arteries after 12 months was diagnosed in 95% of patients of the control and 1 – main groups (Ft, p=0.00001). Healing of trophic ulcers of the foot in the first month after ET was observed in 78% (n=7) of patients of the main group and in 13% (n=3) of the control group (χ2 (1, n=33) = 10.2961; p=0.0013), and after 3 months – in 100% (n=9) and 83% (n=20) of patients in the respective groups. Conclusions: The combination of PTRA with POBA reduces the probability of repeated violations of the patency of the target artery. In addition, it increases the healing efficiency of trophic ulcers of the LE compared to POBA monotherapy.

The porphyrin derivative grafted with TiO2 and the composite (PORBP-TiO2) was further confirmed with different analytical techniques like diffuse reflectance spectra, XRD, FT-IR, XPS, SEM analysis. PORBP-TiO2 was checked for photodegradation of methylene blue in visible light condition and degradation results were compared with bare TiO2. Our newly synthesized photocatalyst showed a very fast and efficient photocatalytical activity in comparison with bare TiO2. Fluoresence life time results also supported the strong photocatalytic activity of PORBP-TiO2. The reusability studies also showed that PORBP-TiO2 is having very good stability upto 10 cycles also. Finally we proposed a suitable catalytic diagram for PORBP-TiO2 photocatalysis.

Objective To determine the efficacy of a preoperative bleeding questionnaire (POBQ) and coagulation screening in predicting hemorrhage associated with adenotonsillectomy. Study Design Retrospective study. Subjects and Methods Between January 1998 and December 2003, 7730 tonsillectomy and adenotonsillectomy patients were administered the POBQ preoperatively. Further coagulation screening was based on POBQ responses. Results A total of 232 (3.0%) of 7730 had postoperative bleeding; 184 (3.2%) of 5782 patients who had negative questionnaires bled postoperatively compared with 48 (2.5%) of 1948 patients with positive questionnaires ( P = 0.126). Of 1948 patients with positive questionnaires, 141 (7.2%) had abnormal preoperative coagulation screens and 9 (6.4%) of 141 bled; of the 1807 (92.8%) with negative coagulation screens, 39 (2.2%) bled ( P = 0.005). Conclusion The POBQ is an effective tool for identifying patients who are at potential risk for post-tonsillectomy bleeding. Patients with both a positive POBQ and coagulation screen had a statistically higher likelihood of postoperative bleeding than other patients. The POBQ allowed the identification of individuals with bleeding disorders to be treated before surgery, likely decreasing the risk of bleeding in these patients.

Populations of cells lacking POB1 accumulate abnormally high numbers of enlarged large-budded cells with a single nucleus at the neck of the bud. The average DNA content in a population of cells lacking POB1 is shifted toward the G2 value. These two phenotypes indicate that while the bulk of DNA replication is completed without POB1, mitosis is delayed. Deleting POB1 also causes elevated levels of both chromosome loss and genetic recombination, enhances the temperature sensitivity of cells with mutant DNA polymerase alpha genes, causes increased sensitivity to UV radiation in cells lacking a functional RAD9 checkpoint gene, and causes an increased probability of death in cells carrying a mutation in the MEC1 checkpoint gene. The sequence of the POB1 gene indicates that it is identical to the CTF4 (CHL15) gene identified previously in screens for mutations that diminish the fidelity of chromosome transmission. Introduction. Altered ocular perfusion and vascular dysregulation have been reported in glaucoma. The aim of this paper was to evaluate the vascular response to a hypercapnic stimulus.Methods. Twenty normal tension glaucoma (NTG) patients and eighteen age- and gender-matched controls had pulsatile ocular blood flow (POBF) measurements, systemic cardiovascular assessment, and laser Doppler digital blood flow (DBF) assessed. Measurements were taken at baseline, after 10-minutes rest, in the stable sitting and supine positions and following induction and stabilization of hypercapnia, which induced a 15% increase in end-tidalpCO2. The POBF response to hypercapnia was divided into high (>20%) and low responders (<20%).Results. 65% of NTG patients had a greater than 41% increase in POBF followingCO2rebreathing (high responders). These high responders had a lower baseline POBF, lower baseline DBF, and a greater DBF response to thermal stimulus.Conclusion. NTG patients that have a greater than 20% increase in POBF after a hypercapnic stimulus have lower baseline POBF and DBF values. This suggests that there is impaired regulation of blood flow in a significant subgroup of NTG patients. This observation may reflect a generalised dysfunction of the vascular endothelium.

ABSTRACT Pseudomonas aeruginosa efficiently intoxicates eukaryotic cells through the activity of the type III secretion-translocation system (TTSS). Gene deletions within the translocation operon pcrGVH-popBD abolish pore-forming activity of P. aeruginosa strains with macrophages and TTSS-dependent hemolysis. Here we investigated the requirements for PcrV, PopB, and PopD in pore formation by analyzing specific mutants using red blood cells (RBCs) and fibroblasts expressing green fluorescent protein fused to actin. Simultaneous secretion of three proteins, PopB, PopD, and PcrV, was required to achieve wild-type hemolysis and effector translocation. Deletion of pcrV in a cytotoxic strain did not affect secretion of PopB and PopD but abolished hemolytic activity and translocation of effectors into fibroblasts. Notably, the PcrV-deficient mutant was not capable of inserting PopD into host cell membranes, whereas PopB and PopD, but not PcrV, were readily found within membranes of wild-type-infected RBCs. Immunoprecipitation experiments performed by using a liposome model of pore assembly revealed a direct interaction between PopD and PopB but not between PopD and PcrV. Consequently, PcrV is necessary for the functional assembly of the PopB/D translocon complex but does not interact directly with pore-forming Pop proteins.

This study aims to evaluate the influence of palm oil empty fruit bunch (EFB) fibers on flexural strength performance of concrete in the presence of palm oil biomass clinker (POBC). This study considered various proportions of palm oil EFB fibers as 0%, 0.2%, 0.4%, and 0.6% in concrete with fixed amount of POBC as 10%. It was investigated that there is substantial influence of palm oil EFB fibers on properties of concrete containing 10% POBC as sand replacement. The experimental findings of this study indicated that the workability of fresh mix concrete decreases as palm oil EFB fiber content increased. Besides that, hardened properties of concrete were found to be improved as the amount of palm oil EFB fibers increased in the concrete. It was noticed that flexural strength was improved with addition of 0.2% palm oil EFB fibers that act as reinforcement and deliver growth in flexural strength for concrete containing 10% of POBC. Hence, it was concluded that palm oil EFB fiber could be utilized as fiber reinforcement in concrete to improve flexural strength performance of the concrete. Abstract Abstract 3344 Poster Board III-232 Background: Hepatic VOD is a life-threatening complication following SCT with a high incidence in children. Development of VOD is one of the most common causes of early death after SCT. Busulfan (BU), an alkylating agent with a very narrow therapeutic index is a commonly used conditioning agent in pediatric stem cell transplantation (SCT) with a strong correlation between AUC and both efficacy and toxicity. Oral BU (poBU) has significant age-related and interpatient pharmacokinetic differences and was linked with an increased risk for VOD. IV busulfan (ivBU) yielded promising results in some studies to be associated with low toxicity profile, especially with a reduced incidence of VOD. Methods: Patients <18 years with myeloablative SCT were included in a prospective multicenter phase II/III trial to evaluate the efficacy of Defibrotide (DF). Eligibility criteria included conditioning with BU (iv and po) and melphalan (MEL). Pts were prospectively randomized to the control arm or to receive DF. Primary endpoint was the incidence of hepatic VOD by D+30 using modified Seattle criteria (2 or more of the following: bilirubin > 2 mg/dL, hepatomegaly, ascites and/or unexplained weight gain > 5%). VOD was assessed by physical exam; hepatomegaly and ascites were confirmed by ultrasound. A blinded IRC of 3 expert hematologists confirmed the diagnosis of VOD. Although the study was not powered to assess mortality, a composite score was assessed as a secondary endpoint that incorporated VOD-associated toxicity (respiratory failure, renal failure, encephalopathy) and mortality. The additional analysis of the influence of BU on VOD was not planned and is therefore explorative. Results: 360 pts were enrolled between January 2006 and January 2009 by 28 centers in the EU and Israel. An Intent-to-Treat (ITT) analysis was performed on all pts who signed informed consent (n=356). 251 (71%) pts from the ITT population were conditioned with BU (64% ivBU; 36% poBU). 202 (55%) were treated with BU and Melphalan (MEL) (60% ivBU; 40% poBU). In 49 (14%) BU was used without MEL (80% ivBU; 20% poBU). In children <2yrs 44 (12%) were conditioned with BU/MEL (59% ivBU; 41% poBU) and 26 (7%) were conditioned without MEL (85% ivBU; 15% poBU). The median age of patients with iv BU was 3.65 yrs and 5,13 yrs with poBU; 28% infants, 50% children (ages 2-11 yrs) and 22% adolescents (evenly distributed). 45% female, 55% male (evenly distributed). Allo-SCT was performed in 69% with ivBU and 46% with poBU (remaining with auto-SCT). The diagnoses were evenly distributed between poBU and ivBU. Except in AML 18% were conditioned with ivBU and 32% with poBU. Preexisting liver disease was present in 23% ivBU and 9% poBU pts, of which 46% (17/37) ivBU and 37% (3/8) poBU had elevated transaminases. Overall VOD was experienced by 23% of the infants, 14% of the children and 13% of the adolescents. The overall incidence of VOD in pts treated with BU was 18%. VOD was diagnosed in 24% ivBU vs 8% poBU pts. In pts treated with BU/MEL VOD was diagnosed in 16%. VOD in ivBU/MEL was 21% (26) vs 8% (6) in poBU/MEL. In BU without MEL VOD was diagnosed in 27% (13/49). 31% (12/39) in ivBU pts and 10% (1/10) in poBU pts. In infants treated with BU/MEL the incidence of VOD in the ivBU group was 23% (6/26) and 11% (2/18) in poBU. In BU without MEL in infants the incidences were 41% (9/22) ivBU versus none (0/4) in poBU. The diagnosis of VOD independent of severity was associated with a higher mortality and equaled 24.6% (14/57) compared to 7% in pts without VOD (21/299). Respiratory failure was observed in 10% (16/161) of ivBU vs 2% (2/90) of poBU pts (9% (11/122) vs 1% (1/80) iv vs po BU/MEL); renal failure in 5% (8/161) ivBu vs 1% (1/90) poBU (5% (6/122) vs none in iv vs po BU/MEL). The incidence of multi-organ-failure (MOF) by day +100 was 12% (19/161) in ivBU vs 3% (3/90) in poBU (p=0.022). Compared to all other pts the risk to develop VOD in infants treated with allo-SCT and ivBU is 2.4 times higher (p=0.003). Conclusions: Although the scope of this trial was not to assess the influence of BU on the incidence of VOD and there was an imbalanced distribution of liver diseases and a potential bias for other risk factors the incidence of VOD in ivBU was unexpectedly high especially in infants. Objective To characterize and identify risk factors for 30-day surgical site infections (SSIs) in patients with head and neck cancer who underwent microvascular reconstruction. Study Design Cross-sectional study with nested case-control design. Setting Nine American tertiary care centers. Subjects and Methods Hospitalized patients were included if they underwent head and neck cancer microvascular reconstruction from January 2003 to March 2016. Cases were defined as patients who developed 30-day SSI; controls were patients without SSI at 30 days. Postoperative antibiotic prophylaxis (POABP) regimens were categorized by Gram-negative (GN) spectrum: no GN coverage, enteric GN coverage, and enteric with antipseudomonal GN coverage. All POABP regimens retained activity against anaerobes and Gram-positive bacteria. Thirty-day prevalence of and risk factors for SSI were evaluated. Results A total of 1307 patients were included. Thirty-day SSI occurred in 189 (15%) patients; median time to SSI was 11.5 days (interquartile range, 7-17). Organisms were isolated in 59% of SSI; methicillin-resistant Staphylococcus aureus (6%) and Pseudomonas aeruginosa (9%) were uncommon. A total of 1003 (77%) patients had POABP data: no GN (17%), enteric GN (52%), and antipseudomonal GN (31%). Variables independently associated with 30-day SSI were as follows: female sex (adjusted odds ratio [aOR], 1.6; 95% CI, 1.1-2.2), no GN POABP (aOR, 2.2; 95% CI, 1.5-3.3), and surgical duration ≥11.8 hours (aOR, 1.9; 95% CI, 1.3-2.7). Longer POABP durations (≥6 days) or antipseudomonal POABP had no association with SSI. Conclusions POABP without GN coverage was significantly associated with SSI and should be avoided. Antipseudomonal POABP or longer prophylaxis durations (≥6 days) were not protective against SSI. Antimicrobial stewardship interventions should be made to limit unnecessary antibiotic exposures, prevent the emergence of resistant organisms, and improve patient outcomes.

Abstract Background Proteasome inhibition with bortezomib has revolutionized the treatment of multiple myeloma, but the vast majority of patients eventually develop clinical bortezomib resistance through poorly understood mechanisms. One of the most conserved cellular responses to proteasome inhibition is to up-regulate proteasome subunit expression, presumably with the goal of enhancing proteasome activity and restoring intracellular protein homeostasis. We therefore hypothesized that proteasome inhibitor resistance could be associated with enhanced proteasome assembly, and that suppression of this assembly process could help restore drug sensitivity. The current studies focused on POMP, which is involved in addition of catalytically active b subunits to the hemiproteasome ring initially formed by structural a subunits. Methods We studied ANBL-6, KAS-6/1, OPM-2, and RPMI 8226 multiple myeloma cell lines which had acquired bortezomib resistance through prolonged exposure to increasing drug concentrations, and compared them to their drug-naïve, vehicle-treated counterparts. In addition, we evaluated primary cells derived from patients with myeloma, and examined an in vivo murine xenograft model of human myeloma. Results Bortezomib-resistant (V10R) ANBL-6, KAS-6/1, OPM-2, and RPMI 8226 cell lines showed enhanced levels of POMP mRNA by quantitative (q) PCR compared to their drug-sensitive counterparts, which was associated with higher POMP protein levels seen by immunoblotting. Exogenous over-expression of POMP in drug-naïve OPM-2 and KAS-6/1 cells was sufficient by itself to induce resistance to both bortezomib and carfilzomib. Conversely, suppression of POMP with one of two different Lentiviral small hairpin (sh) RNAs restored sensitivity in OPM-2 and KAS-6/1 V10R cells to bortezomib and carfilzomib. Since no known pharmaceuticals directly target POMP, we examined its promoter region, and found a consensus binding site for nuclear factor (erythroid-derived 2)-like (NRF) 2. Consistent with a role of NRF2 in POMP expression, NRF2 mRNA and protein were increased in V10R myeloma cells, and in drug-naïve cells treated with bortezomib. Moreover, transfection of cells with NRF2 cDNA activated a POMP promoter reporter, while chromatin immunoprecipitation with anti-NRF2 antibodies preferentially precipitated sequences near the POMP promoter. Also, NRF2 over-expression induced POMP and enhanced proteasome chymotrypsin-like activity, while its suppression had the opposite effects. All trans-retinoic acid (ATRA) blocked nuclear accumulation of NRF2 in OPM-2 and KAS-6/1 V10R cells, and reduced expression of POMP. Combinations of bortezomib with ATRA showed enhanced activity against these drug-resistant cell lines in association with greater proteasome inhibition, and were synergistic in drug-naïve cells. In primary samples, ATRA with bortezomib induced a greater reduction in viability than did either treatment alone. Finally, in a murine xenograft model with OPM-2 V10R cells, neither ATRA nor bortezomib showed substantial activity, while the combination regimen, by comparison, retained efficacy. Conclusions Taken together, our data support the hypotheses that NRF-2-influenced POMP over-expression contributes to proteasome inhibitor resistance in multiple myeloma, while approaches targeting POMP hold promise in overcoming resistance. Moreover, they provide a framework for translation of proteasome inhibitors with ATRA to the clinic to enhance activity, and to overcome resistance to this important class of anti-myeloma agents. We performed a retrospective analysis to evaluate the survival on first line biologic drug of rheumatoid arthritis (RA) patients with potential occult HBV infection (pOBI). We analysed longitudinal data of 486 consecutive RA patients starting a first biological drug in a time frame from 1st January 2008 to 31st December 2014. Demographic and disease related characteristics were collected at baseline and at the last observation visit. Baseline serological markers of HBV infection and causes of treatment discontinuation were also recorded. Primary endpoint was the influence of pOBI on drug survival, estimated by Kaplan-Meier life table analysis. Estimates hazard ratios (HRs) of drug discontinuation, adjusted for disease characteristics, biological drug class and HBcAb status were computed by Cox-regression models. The retention rate was significantly lower in pOBI positive patients (58.2%) when compared to pOBI negative ones (67.8%) and this data was confirmed also when only discontinuation due to ineffectiveness was considered (pOBI positive 66.4% vs pOBI negative 75.3%, long rank 7.93, p=0.005). Cox regression models showed a significant association between HBcAb-neg (HR 0.58, 0.41-0.84), higher ESR-DAS28 at baseline (HR 1.07, 1.03-1.11) or RF/ACPA-neg (HR 1.46, 1.04-2.06) and drug discontinuation. Occult HBV infection seems to influence negatively the effectiveness of biological therapies in RA patients.

Abstract In this study, palm oil was the first time to convert into a novel polyester plasticizer for polyvinyl chloride (PVC). In the first stage, palm oil was converted into palm oil monoglyceride (POM) by alcoholysis with glycerol. Next, a novel palm oil monoglycerides based polyester plasticizer (POMP) was synthesized from POM and maleic anhydride through esterification and condensation reaction. The structure of POMP was characterized with FTIR, 1H NMR and GPC. Then PVC blends were prepared using POMP as a plasticizer, melting behavior, thermal property, compatibility, mechanical properties and mechanism of plasticization of PVC blends were systematically studied. Melting behavior indicated that POMP could decrease the torque and the melt viscosity of PVC blends that was conducive to process. With the content of POMP increasing from 5 g to 15 g in PVC blends, the plasticized PVC blends demonstrated better compatibility, the degradation temperature (Td) increased from 252.6°C to 257.0°C, the glass transition temperature (Tg) decreased from 55°C to 49.5°C. Plasticization was put into effect by interaction of the electron cloud between the PVC chain and POMP molecule. This study may lead to the development of new type of PVC plasticizer based on vegetable oil.

The growth of the palm oil industry has resulted in an increase in the production of solid waste, created from the extraction of fresh fruit bunches, which can take the form of palm oil boiler ash (POBA). POBA can be used to modify asphalt binder and asphalt mixtures to reduce the harmful effect of this waste on the environment. The objective of each modification is to increase the strength, stiffness, durability, workability and constructability of asphalt mixtures while reducing the environmental effects. This study examines the physical and chemical properties of 60/70 penetration-grade asphalt binder, modified using POBA and warm mix asphalt (WMA) additive. Ranges of modified binder were prepared by adding 2% of the warm additive Rediset with different POBA contents (3%, 5%, 7% and 9%) throughout the wet mixing process. Physical properties of modified binder were obtained from penetration, softening point, ductility and rotational viscosity tests. Molecular components and structures of the modified binder were identified using Fourier transform infrared (FTIR) and scanning electron microscopy (SEM). Thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC) were used to determine the thermal properties of modified asphalt binder. The addition of 7% POBA in WMA binder showed the best characteristics in the tested consistency of its physical properties. As a modifier, POBA showed no chemical interaction with the molecules and structures of the asphalt binder and did not significantly change the physicochemical transitions. From the results, it can be concluded that using POBA in WMA binder for pavement construction is a viable option.

This study examined the local perception on the climate changes and how these changes combined with anthropogenic pressures generated by the forms of use of the forest resources, impact the sustainability of the forests of Pobè and Dogo-Kétou in southeast Benin. The data were collected through an investigation of 148 and 139 local populations sampled respectively in the municipalities of Pobè and Kétou in the studied area. An interview guide and a structured research questionnaire were used to collect the data which were analyzed using the software Ri386 3.5.1. The mains results revealed that more than 98% of the local population living around these forests perceive the effects of climate change and anthropogenic activities are the main causes. The consequences of climate change from the social perception are diversified, such as the violent winds and the scarcity of rains. The prolonged droughts are also confirmed by 61% of local population in Kétou and 81% in Pobè. In these conditions, trees have a harder time of reproduction with a poor capacity of regeneration. The analysis of the vulnerability of these forests in relation to the use values (UV) revealed that the populations living near to Dogo-Kétou forest use so much the forest resources (UV>2.5) than those of Pobè (UV<2.5). So, moderate pressure was notified on Pobè forest. The forest of Dogo-Kétou is therefore more vulnerable than the forest of Pobè from the view of the pressures linked to the categories of uses relative to food, energy wood, service wood, non-timber forest products and medicinal use. For adaptation strategies, the local population revealed the enrichment of the forests by indigenous forest species (32% in Kétou and 19% in Pobè), the reforestation (76% in Kétou and 84% in Pobè) and the creation of private plantations (39% in Kétou and 57% in Pobè). However, the establishment of ethnobotanical family gardens, the reduction of wood consumption for energy to the detriment of domestic gas, sensitization, prayer (church and vodoun) are ways to better adapt a good resilience to climate change.Keywords: Climate change, adaptation strategies, reforestation, resilience, forests, Benin.

Although hydroelectric power plants generate most electricity in Brazil, the dams provide environmental barriers which also include the interruption of fish migration. Current study, conducted prior to the construction of São Domingos HPP on the shaft of the Cachoeira Branca (waterfall) on the Verde river, state of Mato Grosso do Sul, Brazil, evaluated the genetic diversity of two natural populations of dourado (Salminus brasiliensis) located downstream (Population A - PopA) and upstream (Population B - PopB) the waterfall. Eight primers were employed to analyze 56 specimens (PopA: 26; PopB: 30) and observed in 102 fragments, of which 86 were polymorphic (84.3%). Low frequency fragments (PopA: 2; PopB: 1), three limiting fragments (PopA) and three exclusive fragments (PopB) were identified. The intra-population genetic variability calculated the Shannon index, whilst the percentage of polymorphic fragments showed high rate variability within each population (PopA: 0.300 and 60.80% and PopB: 0.411 and 79.40%, respectively). The distance and genetic identity revealed high genetic differentiation (0.076 and 0.927, respectively). Results reveal that populations have high intra-population genetic variability and genetic differentiation between them, with low gene flow.

Abstract This study examines the physical properties of a 60/70 penetration grade bitumen modified using warm mix asphalt (WMA) additive, Rediset and palm oil boiler ash (POBA) as a modifier. Modified binders were prepared by adding 2% of Rediset with different POBA contents (3, 5, 7, and 9%) through wet mixing process. Physical properties of modified bituminous binder were obtained from penetration, softening point, rotational viscosity and ductility tests. The addition of 7% POBA in WMA binder has the best characteristics in term of its physical properties. The penetration values, softening point temperatures and ductility characteristics show inconsistent patterns. However, the rotational viscosity of each combination had decreased with the increment of temperature even though the trend was not significantly constant. Penetration index (PI) showed an increment with 3 and 5% and decrement pattern on 7 and 9% as well as penetration viscosity number (PVN) showing an inconsistent decrement with the addition of POBA. From the results, it can be concluded that using POBA in WMA binder for pavement construction is a viable option and can be further investigated.

ABSTRACT The FACT complex of vertebrate cells, comprising the Cdc68 (Spt16) and SSRP1 proteins, facilitates transcription elongation on a nucleosomal template and modulates the elongation-inhibitory effects of the DSIF complex in vitro. Genetic findings show that the related yeast (Saccharomyces cerevisiae) complex, termed CP, also mediates transcription. The CP components Cdc68 and Pob3 closely resemble the FACT components, except that the C-terminal high-mobility group (HMG) box domain of SSRP1 is not found in the yeast homolog Pob3. We show here that Nhp6a and Nhp6b, small HMG box proteins with overlapping functions in yeast, associate with the CP complex and mediate CP-related genetic effects on transcription. Absence of the Nhp6 proteins causes severe impairment in combination with mutations impairing the Swi-Snf chromatin-remodeling complex and the DSIF (Spt4 plus Spt5) elongation regulator, and sensitizes cells to 6-azauracil, characteristic of elongation effects. An artificial SSRP1-like protein, created by fusing the Pob3 and Nhp6a proteins, provides both Pob3 and Nhp6a functions for transcription, and competition experiments indicate that these functions are exerted in association with Cdc68. This particular Pob3-Nhp6a fusion protein was limited for certain Nhp6 activities, indicating that its Nhp6a function is compromised. These findings suggest that in yeast cells the Cdc68 partners may be both Pob3 and Nhp6, functioning as a bipartite analog of the vertebrate SSRP1 protein.

This review, which is divided into three parts, concerns electrochemical synthesis, spectroscopic characterization, and formation mechanisms of poly(o-aminophenol) (POAP) film electrodes. The first part of this review is devoted to describing the electropolymerization process of o-aminophenol (o-AP) on different electrode materials and in different electrolyte media by employing both potentiodynamic and potentiostatic methods. The evolution of the voltammogram during the electrosynthesis of POAP and the assignation of the voltammetric peaks to different species, according to the formation mechanism formulated by each author, are described. The effects of some chemical substances and electrochemical pretreatments of the electrode surface, on the electropolymerization process of o-AP, are also considered in this part of the review. The synthesis of POAP in neutral media, which yields a nonconducting polymer, and its use as component of biosensors and protective layer in corrosion processes, are analyzed at the end of the first part. The second part of this review refers to spectroscopic studies carried out by different authors to both identify the products of the o-AP electro-oxidation and elucidate the chemical structure of POAP film electrodes. This second part of the review also describes the different spectroscopic methods employed to study the redox process of POAP, which allows the demonstration of the existence of transient species during the transition of the completely oxidized form of POAP to the completely reduced one. The third part of this review shows the different mechanisms formulated to interpret the POAP film formation from both acid and basic solutions of o-AP. Also, some electrochemical and spectroscopic data which allowed to propos the corresponding formation mechanisms, especially in basic media, are described.

Dynamically dimensioned search (DDS) is a well-known optimization algorithm in the field of single solution-based heuristic global search algorithms. Its successful application in the calibration of watershed environmental parameters has attracted researcher’s extensive attention. The dynamically dimensioned search algorithm is a kind of algorithm that converges to the global optimum under the best condition or the good local optimum in the worst case. In other words, the performance of DDS is easily affected by the optimization conditions. Therefore, this algorithm has also suffered from low robustness and limited scalability. In this work, an improved version of DDS called DDS-POBL is proposed. In the DDS-POBL, two effective methods are applied to improve the performance of the DDS algorithm. Piecewise opposition-based learning is introduced to guide DDS search in the right direction, and the golden section method is used to search for more promising areas. Numerical experiments are performed on a set of 23 classic test functions, and the results represent significant improvements in the optimization performance of DDS-POBL compared to DDS. Several experimental results using different parameter values demonstrate the high solution quality, strong robustness, and scalability of the proposed DDS-POBL algorithm. A comparative performance analysis between the DDS-POBL and other powerful algorithms has been carried out by statistical methods by using the significance of the results. The results show that DDS-POBL works better than PSO, CoDA, MHDA, NaFA, and CMA-ES and gives very competitive results when compared to INMDA and EEGWO. Moreover, the parameter calibration application of the Xinanjiang model shows the effectiveness of the DDS-POBL in the real optimization problem.

Project based learning (PoBL) is a constructivism learning approach that focuses around student centred learning. PoBL has been found to be effective in different tertiary institutions of learning across the globe. However, in spite the effectiveness of PoBL, it has not been adopted in Nigerian tertiary institutions. Therefore, the purpose of this study is to investigate the effective strategies for integrating PoBL in teaching and learning Woodwork Technology Education (WTE) at tertiary institutions in Nigeria. Three research questions were posed in this study. A mixed method approach involving both quantitative and qualitative method was employed for the study. The sample of the study comprised 50 in-service postgraduate students from Nigerian tertiary institutions studying Technical Vocational Education and Training (TVET) in Universiti Teknologi Malaysia for the quantitative aspect, and 9 in-service post graduate students for the qualitative part. A 16-item structured questionnaire was used for quantitative data collection while semi structured interview was used for qualitative data collection. Data was analyse using the SPSS software version 24 to analyse quantitative data and NVIVO 12 was used for qualitative data analysis. The findings of the study revealed that traditional instructional approach is the predominant method used in teaching and learning WTE at tertiary institutions in Nigeria. It was also revealed from the findings that PoBL encourage student ownership of learning, enhances students critical thinking, effective collaboration and communication. The findings from the study also revealed that restructuring of WTE curriculum at tertiary institutions in Nigeria to focus on a learner centred approach, as well as organising seminars and workshops for WTE lecturers to be acquainted with the application of PoBL in WTE teaching and learning were among the effective strategies for integrating PoBL in teaching and learning WTE at tertiary institutions in Nigeria. Finally, this paper calls for the integration of PoBL into the teaching and learning of WTE at Universities, Polytechnics and Colleges of education in Nigeria for effectiveness in skills acquisition.

Penelitian ini bersifat non eksperimental yang dilakukan secara eksploratif; menggunakan 20 ekor sapi berumur antara 32 sampai 36 bulan. Parameter yang diamati meliputi kualitas semen, kondisi organ reproduksi sapi betina dan angka kebuntingan. Hasil penelitian menunjukkan, kualitas semen sapi POBA rendah atau jelek berdasarkan hasil analisis secara mikroskopis maupun makroskopis; namun memiliki libido yang cukup tinggi yaitu rata-rata sekitar 4 menit 25 detik dari mulai mengendus atau didekatkan dengan ternak pemancing (teaser) sampai ejakulasi. Secara umum kondisi organ reproduksi sapi POBA betina normal, sehingga siap dijadikan indukan yang produktif. Sebanyak 66,67% menunjukkan perilaku estrus yang tampak terutama keluar lendir yang terlihat jelas menggantung di bibir vagina bahkan diantaranya mengejar dan/atau menaiki sapi-sapi lain. Namun hasil pemeriksaan kebuntingan (PKB) melalui palpasi rektal menunjukkan tidak ada sapi betina yang bunting. Dapat disimpulkan bahwa sapi POBA jantan infertil, yang diindikasikan melalui hasil pemeriksaan semen dan tidak adanya sapi betina yang berhasil bunting melalui perkawinan alam selama 3 (tiga) bulan. Sedangkan sapi betina POBA memiliki organ reproduksi normal serta menunjukkan ekspresi estrus yang baik. (The evaluation of cattle reproduction status between crosses Ongole Crossbreed with Bali cows) ABSTRAK. The purpose of this research was to determine the reproductive status of the crossbred PO and Bali cattle (hereinafter referred to as POBA cattle) through several field activities. This research is non-experimental and carried out exploratory using 20 cattle aged 32 to 36 months. Parameters observed included semen quality, cows' reproductive organ condition, and pregnancy rate. The quality of POBA bulls semen was low or bad as indicated by the results of microscopic and macroscopic analysis; but had pretty high libido, which was an average of about 4 minutes and 25 seconds, starting to sniff or being brought near to teaser until ejaculation. In general, the reproductive organs of POBA cows were normal so that they were ready to become productive sires. As many as 66.67% showed estrus behavior, especially visible mucus that was hanging on the lips of the vagina, even among them chasing and/or riding other cattle. However, the results of pregnancy examination through rectal palpation showed that there were no pregnant cows. It can be concluded that POBA bulls are infertile, which was indicated by the results of semen examination and the absence of successfully conceived cows through natural mating for 3 (three) months. Meanwhile, POBA cows have normal reproductive organs and show good estrus expression.

Four horizontal slices with a thickness of 1 mm were cut, representing the mid-portion of the root. The specimens (n = 480) were irrigated with 17% ethylenediaminetetraacetic acid(EDTA) for 60 seconds, then twice with distilled water (DW) for 30 s each. The canals were filled with either AH Plus (Dentsply Sirona, Konstanz, Germany) or BioRoot RCS (Septodont, St. Maur-des-Fossés, France) (n = 240). Separated into four groups per type of sealer (n = 60), the specimens were incubated at 37 °C covered with gauze moistened in DW or phosphate-buffered saline (PBS) for either one or eight weeks. Dislodgement resistance was measured and POBS was calculated. Statistical analysis was performed using the analysis of variance (ANOVA) test and the Student–Newman–Keuls test (p = 0.05). AH Plus showed higher POBS when stored in PBS compared to DW, irrespective of the incubation period (p < 0.05). BioRoot RCS displayed higher POBS when stored in DW compared to PBS after eight weeks of incubation (p < 0.05). No difference was found after one week of incubation (p > 0.05). Irrespective of the sealer or the immersion solution, POBS decreased from one week to eight weeks (p < 0.05). Mixed failure modes were found in all groups irrespective of sealer, immersion medium, or immersion period. POBS decreased after a longer incubation time in both immersion solutions. Duration of immersion and the type of immersion solution had a significant impact on the outcome of the POBS testing protocol.

This study demonstrated that polymerization behavior of plant oil-based acrylic monomers (POBMs) synthesized in one-step transesterification reaction from naturally rich in oleic acid olive, canola, and high-oleic soybean oils is associated with a varying mass fraction of polyunsaturated fatty acid fragments (linoleic (C18:2) and linolenic (C18:3) acid esters) in plant oil. Using miniemulsion polymerization, a range of stable copolymer latexes was synthesized from 60 wt.% of each POBM and styrene to determine the impact of POBM chemical composition (polyunsaturation) on thermal and mechanical properties of the resulted polymeric materials. The unique composition of each plant oil serves as an experimental tool to determine the effect of polyunsaturated fatty acid fragments on POBM polymerization behavior and thermomechanical properties of crosslinked films made from POBM-based latexes. The obtained results show that increasing polyunsaturation in the copolymers results in an enhanced crosslink density of the latex polymer network which essentially impacts the mechanical properties of the films (both Young’s modulus and toughness). Maximum toughness was observed for crosslinked latex films made from 50 wt.% of each POBM in the monomer feed.

Plastic waste derived from plastic dog poop bags (DPBs) could be considered a negligible source of plastic waste. However, it is estimated that this kind of waste contributes to 0.6% of the total plastic waste generated worldwide, and it is expected to increase in the following years. Plastic dog poop bags can be replaced with biodegradable and bio-based alternatives. These alternatives can biodegrade in various environments such as composting, soil, or water and thus allow for end-of-life management without the risk of contaminating the environment with microplastics. However, not all biodegradable bags are always compostable. In this study, we tested composting as the end-of-life management of selected biodegradable dog poop bags (i.e., prototypes of bags and commercially available bags). We analyzed the biodegradation of selected biodegradable plastic dog poop bags during the composting of green waste in laboratory composting reactors after 4 weeks and 90 days of composting. All the investigated DPBs decomposed 100% after 90 days of composting. However, the fresh compost obtained after the 90-day composting of green waste mixed with the investigated bags containing dog poop did not demonstrate high quality.

The crystal structure is reported of p-hydroxybenzoate hydroxylase (PobA) from Pseudomonas putida, a possible drug target to combat tetracycline resistance, in complex with flavin adenine dinucleotide (FAD). The structure was refined at 2.2 Å resolution with four polypeptide chains in the asymmetric unit. Based on the results of pairwise structure alignments, PobA from P. putida is structurally very similar to PobA from P. fluorescens and from P. aeruginosa. Key residues in the FAD-binding and substrate-binding sites of PobA are highly conserved spatially across the proteins from all three species. Additionally, the structure was compared with two enzymes from the broader class of oxygenases: 2-hydroxybiphenyl 3-monooxygenase (HbpA) from P. nitroreducens and 2-methyl-3-hydroxypyridine-5-carboxylic acid oxygenase (MHPCO) from Mesorhizobium japonicum. Despite having only 14% similarity in their primary sequences, pairwise structure alignments of PobA from P. putida with HbpA from P. nitroreducens and MHPCO from M. japonicum revealed local similarities between these structures. Key secondary-structure elements important for catalysis, such as the βαβ fold, β-sheet wall and α12 helix, are conserved across this expanded class of oxygenases.

Pozyskiwana jest ona za pomocą gruntowych pomp ciepła (GPC). Instalowanie gruntowych pomp ciepła z pionowymi wymiennikami jest korzystne z punktu widzenia efektywności ekonomicznej, niewielkiego wpływu na środowisko oraz poszerzania zakresu wykorzystania odnawialnych źródeł energii. W artykule podano najważniejsze uwarunkowania środowiskowe: gradient geotermiczny, naturalny ziemski strumień ciepła, przewodność cieplną skał oraz lokalne warunki hydrogeodynamiczne. Wskazane zostały podstawowe warunki techniczne dla instalacji gruntowych pomp ciepła. Zwrócono również uwagę na znaczenie parametrów termicznych skał. W Polsce liczba zainstalowanych gruntowych pomp ciepła jest niższa w porównaniu z innymi krajami europejskimi. Jednym z problemów w zastosowaniu tych urządzeń jest niewystarczające rozpoznanie specyficznych warunków gruntowo-wodnych pod pionowe gruntowe wymienniki ciepła, co wpływa na ekonomiczną efektywność tego typu inwestycji. Artykuł jest przeglądem publikacji poświęconych zarysowanemu problemowi.

The PoAP polymer was found to affect the redox process of copper and pyrocatechin. The use of polymer of different thicknesses showed that the obtained film has a dense, nonporous structure. The redox process of the examined substances may be considered as proceeding on the polymer surface. The PoAP polymer obtained in the described conditions takes part in charge transfer.

This paper describes a one-step chemically produced multiwalled carbon nanotubes/poly-ortho-aminophenol (MWCNTs/PoAP) nanocomposite film production process using in-situ oxidative polymerization. The CoCl 2 -doped nanocomposites exhibit good stability and increased magnetic performance. The morphological analysis revealed that PoAP deposits on the surface of the nanoparticle result in the creation of a fibrous crystal-like structure. The vibrational spectra show that there is a substantial interaction between PoAP and MWCNT/MWCNT[-COOH]. The introduction of nanocrystalline MWCNTS increases PoAP’s UV absorption and causes a blue shift in π-polaron absorption. The magnetic study of Co-doped MWCNTs/PoAP nanocomposites has shown a marked reduction in the hysteresis behavior, indicating significant hole doping associated with the oxygen functional groups on the surface of xidized carbon ’nanotubes. The syntheses of nanocomposites are silent features of the work with implications in crystal engineering and nanocomposite design for various applications.

Psoriatic onycho-pachydermo-periostitis (POPP) is an unfamiliar and poorly recognised condition first described in 1989 by Fournie et al. It is a variant of psoriatic arthritis comprising the triad of onycholysis, soft tissue thickening and radiographically apparent periostitis. Whilst typically affecting the great toe, any of the digits of the hands and feet may be affected. A ‘drumstick’ appearance to the digits of the foot is the most common clinical presentation and can be extremely painful. Nail changes are another hallmark of POPP and can be mistakenly diagnosed as fungal infection leading to lengthy periods of incorrect treatment. In this article, we will outline the clinical presentation, imaging features, pathogenesis and treatment options for POPP. Currently, the existence of POPP is not widely known. Awareness of this unusual condition will allow early appropriate treatment and can aid in the diagnosis of indeterminate seronegative disease.

Rebamipide can be appointed for prevention and treatment of erosive and ulcer injuries of a stomach, a small intestine with dyspepsia, both with inhibitors of a proton pomp, and in the form of monotherapy for the purpose of a gastroprotection to patients when use of inhibitors of a proton pomp is inexpedient. The gastroprotection algorithm depending on a condition of a gastric acid secretion of patients is offered.

The inhibitive performance of 2-[(5-phenyl-1,3,4-oxdiazol-2-yl)methyl]-2H-benzo[b] [1,4] thiazin-3-one (POBT) for low carbon steel corrosion in 1 M H2SO4 has been evaluated by non-electrochrmical, and electrochemical measurements. Efficiency of the inhibitor enhanced with inhibitor concentration and declined with rise in temperature. 92.39% inhibition efficiency was attained using 0.1[Formula: see text]mM of POBT. Anticorrosive effect of the inhibitor is owing to the adsorption of POBT molecules on the metal surface. Mono layer adsorption of inhibitor was favored by observing the best fit with Langmuir isotherm. Polarization measurements reveal that the inhibitor behaves as mixed type. Surface analysis by scanning electron microscopy (SEM) and energy dispersive X-ray spectrometry (EDX) confirm the existence of a protective film of POBT on low carbon steel surface. Quantum chemical parameters obtained using density functional theory (DFT) method complements the above results.

The increment of peat soil productivity meets through ameliorant addition. Ameliorant sources can be obtained from oil palm plantation and electric steam power station waste. The study aimed at investigating the ability of the oil palm fruit empty bunch (OPFEB) biochar, palm oil boiler ash (POBA) and coal fly ash (CFA) as alternative ameliorants besides compost to improve nutrient availability in peat soil of Central Kalimantan. Treatments tested were OPFEB biochar, POBA, CFA, compost, OPFEB biochar+compost, POBA+compost, and CFA+compost. The seven treatments were arranged in a completely randomized design of single factor with four replications. The results revealed that the ameliorants increased peat soil pH at a rate of 0.7 compared to those in initial soil used and available P of 135.7% even though there was no significant increment of exchangeable cations of K, Ca and Mg. Mixing of biochar, POBA or CFA with compost improved nutrients availability in peat soil.

Abstract POBA cattle is a crossbred cattle from Peranakan Ongole (PO) and Bali cattle, which is developed by the Beef Cattle Research Institute (BCRI), Ministry of Agriculture of the Republic of Indonesia. This study aimed to identify the genetic and phenotypic characteristics of POBA cattle raised in the BCRI. Genetic diversity of FSH-β gene was also determined in order to identify a possible marker for reproductive status in POBA cattle. Rambon cattle collected from Banyuwangi regency were used as comparison. A 313 bp fragment of the FSH-β gene was amplified using the polymerase chain reaction (PCR). The PCR products were sequenced and aligned to detect polymorphism. As results, a polymorphism (SNP g.2583C/T) in the FSH-β gene, which produced three genotypes (TT, CC, and CT) was detected. The frequency of each allele was 0.13 (allele C) and 0.87 (allele T). However, the FSH-β gene polymorphism did not significantly affect sperm quality, body weight, body size, and cervical condition of POBA cattle.
