Au, Lucy. "Assessing the Potential Needs for Telehealth in Papua New Guinea (PNG)." Thesis, University of Canterbury. Health Sciences Centre, 2009. http://hdl.handle.net/10092/4656.

Full text
Abstract:
Papua New Guinea has the highest infant and maternal morbidity and mortality rates in the Western Pacific Regions and 50% of hospital admissions are from vaccine preventable diseases. About 85% of 6 million inhabitants live in rural PNG where basic services are lacking or non-existent. Difficulties such as lack of infrastructures like road network and communication, geographical barriers like big mountain ranges, large rivers and swamps, shortage of skilled health professionals and higher concentration of health workers in cities pose great challenges in delivering health services effectively across the country. Telehealth may play an important role in reaching health services to the underserved population in PNG. As part of this study, it is essential to assess the potential of telehealth to enhance the delivery of health services. Specifically, this research aims to report the knowledge, attitudes and skills towards information and communication technology of health care providers in PNG. The study used a cross sectional method of health professionals working at the time of the survey. It collected 174 questionnaires from four hospitals and used SPSS (version17.0) to analyse the data. The results showed that younger male physicians, paramedics, those with gadgets, higher educational qualifications and exposed to ICT resources possess better knowledge and skills than female, older age group and those without gadgets or under exposed to ICT. Currently, the health care professionals in general have leap frogged the technology by focusing on handheld devices such as cell phones rather than landlines. This represents scope for growth and willingness by health workers to adopt and expand telehealth in PNG.

Abstract:
Assembly of neuronal networks with distinct patterns of connectivity during nervous system development involves the growth, extension and branching of axons and dendrites. Over the years genetic and biochemical studies in model organisms have contributed significantly in identifying mechanisms regulating axon growth and extension. However the molecular mechanisms underlying axon branching remain unclear. The egg-laying neuronal circuitry in C. elegans has proven to be a robust system for identifying and characterizing novel genes involved in neuronal morphology. This circuitry which mediates egg-laying behavior in nematodes is composed of two families of motorneurons, HSNs and VCs, which are among the most branched neurons in C. elegans. A genetic screen for axon branch defects in the egg-laying neurons identified png-1 to disrupt neuronal morphology including axon branching. png-1 encodes a Peptide: N-glycanase (PNGase), a conserved cytosolic enzyme that removes N-linked sugar moieties from glycoproteins. In this thesis I present my work characterizing and examining png-1 and its role in mediating axon branching. Mutations in png-1 resulted in excessive ectopic axon branching in the VC4 and VC5 egg-laying neurons as well as branching in the normally unbranched AVL and DVB neurons. Behavioral analysis in these mutants revealed defects in egg-laying behavior and mild in-utero egg retention phenotypes. Cellular characterization shows ubiquitous expression of png-1 in many tissues including vulva cells, muscles, gonads, and neurons. My analysis also shows that png-1 acts both cell-autonomously and cell non-autonomously from neurons and epithelial cells to restrict axon branching around the vulva. Using a candidate gene approach I identified a deletion allele of the DNA repair gene, rad-23, to display axon branching defects and interact with png-1 within a common pathway to regulate axon branching. Additionally, through a genetic modifier screen for enhancers and suppressors of VC4-5 branching defects in png-1, I identified a new allele of sax-2 as an enhancer mutation. sax-2 encodes a scaffolding protein that regulates the activity and localization of sax-1, an NDR kinase. Examination of neuronal phenotypes in sax-1 and sax-2 mutants revealed similar png-1 like defects in VC4-5. Genetic analysis of the double mutants png-1;sax-1 and png-1;sax-2 revealed strong synergistic phenotypes suggesting that png-1 and sax-1/sax-2 function in parallel pathways to regulate axon branching. In summary, this thesis reveals novel components and pathways in the regulation of neuronal branching.

Abstract:
In this thesis I examine the contemporary nexus between White women and the raced and classed institution of White womanhood. More specifically, I focus on White Australian women who are middle class, rich in cultural capital, and generally consider themselves to be progressive; that is race privileged women but women who are not usually associated with overt racism. My analysis unfolds White Australian women in the discursive context of the ideologies of feminism and feminist-influenced anti-racist politics, as well as the ideologies of femininity. The thesis shows how this nexus is enacted through a vision of White women as Good as expressed in the political commitments, mentalities, relationships, narratives and corporeality of such women. The research problem that I identified and worked through in the thesis is as follows: for middle class White women, (who can be seen and see themselves as generic 'women'), Whiteness has been seen and played out as Goodness. Further, in the playing out of this Goodness White women accumulate and defend the prestige and privileges of Whiteness. Specifically, I argue that Whiteness is reproduced in some of the discourses and practices of White feminism, by the progressive White women involved in anti-racist politics, and in the femininity industry and the ways it is taken up. The nub of the problem I identify is that White women's involvement in the structures and narratives that support Whiteness is often grounded in the very qualities of character and conduct that emerge from the colonial and class-constructed ideal of White womanhood and which have historically distinguished them from denigrated others. These qualities- notably virtue, innocence and self-restraint- whilst differently nuanced in other contexts are an ongoing expression of the uses made of White womanhood as the visible sign of race and class superiority. The work examines four key periods: the Australian colony of PNG during the decolonising 1960's and 1970's; the high years of 1970's and 1980's feminism; the race debates of the 1990's; and the bodily practices of present day White women gripped by fears of fat and aging. I explore the ways in which White women's Whiteness is played out in benevolent Black/White relationships, the over-reach of difference feminism, particular kinds of anti-racist identities and activism, and body-improvement practices. In all these cultural sites, White women's Whiteness is often represented as a kind of moral being and deployed as moral authority in ways that are consonant with the raced and classed construction of White women as moral texts. My research approach was determined by the research problem I identified. Given my argument that White women mis-recognise Whiteness as Goodness in a race-structured society, then the collecting of data through interviews or surveys would have yielded material subject to this blindness. Instead, I explored sites and material where moral claims were being pressed, and case studies where 'women' were enacting themselves or being represented or interpellated as moral texts. My selection of primary source material ranges from feminist newsletters, women's and other magazines, literature, film, event programs and flyers, radio and television broadcasts, newspapers and websites, as well as reflections on my own experiences. Secondary source material includes feminist theoretical texts as well as texts drawn from a range of other disciplines, and other historical background materials. I lay out and support my arguments using a technique not dissimilar to collage, aiming to construct a picture that is compelling in its detail as well as coherent in its overall effect. This thesis is a contribution to the de-naturalisation of Whiteness. Navigating a course between the opposing hazards of essentialising Whiteness and understating its effects in contemporary Australian society, I have brought into clearer view some of the strategies which maintain the authority of Whiteness.

Abstract:
This thesis studies the effect of using delta encoding to compress archivesof images. The results are compared with two types of lossless PNGcompression. The tests show that the most sophisticated PNG methodtested compresses the archives around 2-12% higher than the delta encodingmethods, but at the cost of taking more than 10 times as long. The conclusion is therefore drawn that delta encoding could be a usefulmethod for compressing image archives in environments where speed ismore important than storage.

Abstract:
The study combined qualitative methods (observation, interview and case study techniques) and quantitative methods (use of questionnaire and survey instruments) methods to (a) identify and describe current teaching practices in Papua New Guinea (PNG) secondary science laboratory classrooms; (b) investigate the effect of specific teaching practices on academic success on an external science achievement examinaton; and (c) investigate whether an educational productivity model for investigating factors associated with learning is applicable to a developing country context, namely, PNG. Analysis of data from 3,182 Papua New Guinea secondary students, generated revealed similar science laboratory learning environments across most high schools with Open-Endedness having the lowest score. Overall students' attitudes towards science were favourable, with boys having a more favourable attitudes than girls. Multivariate analyses showed that science academic achievement, science practical achievement and attitudes were related to quality and quantity of instruction, the science laboratory learning environment and gender. As in similar studies in other countries, male students performed significantly better than female students in external science achievement examinations. But female students achieved significantly better on a practical science process test. Finally, the study identified some specific aspects of current teaching practices in a developing country context, involving science learning environments and students' attitudes towards science.

Full text
Abstract:
This survey study on students attitudes towards science was conducted in Papua New Guinea. It involved 200 randomly selected grade six students in eight selected community (primary) schools (4 urban and 4 rural) of four provinces. There were six research questions to focus the study. The instruments consisted of 9 Students Background Questions, 9 interview questions, 43 Students Science Attitude Questionnaire Items, and a 20-item cognitive test. The former three sets of instruments were trialled in two schools. The test questions were not trialled as they were trialled previously in several schools within PNG. The 43 Likert-type items were divided into five categories - curiosity, enjoyment, science learning, perceived achievement, and relevance. Each category was assigned 10 items, with the exception of perceived achievement which had three items. About half of the the statements in a category were negatively worded. A student had to put a tick in a box next to a statement according to his or her feelings. The choices were given by the five-point scale - Strongly Agree (SA), Agree (A), Hot Sure (NS), Disagree (D), and Strongly Disagree (SD) along with three laces (smiling, uncertain and frowning) which were added as interest, From the twenty five students in a study school, a group of 6 students (3 males and 3 females) was randomly selected for face to face interview. The responses in the items associated with the five categories (dependent variables) were used to assess various student characteristics (independent variables) of parent and home background (i.e. parent educational levels and language use at home), gender (male and female), biogeographical location (urban and rural), and level of performance in standardized cognitive test (above average, average and below average). Statistical techniques of correlation coefficient, analysis of variance and factor analysis were used in analysing the responses but the results were not clear due insufficient trialling and test of reliability with the limited period in which the data had to be collected. As consequence, chi-square test (contingency tables) was used as alternative statistical test to determine significant differences in responses to the items at 0.05 level. With the exception of perceived achievement, student responses to the items on the categories were generally and satisfactorily positive. The mean levels of attitudes in the categories were not as high as one may expect (Table 5.1). Relatively, few of the items associated with student independent variables showed significant differences. There was generally no significant differences in parent and home backgrounds (i.e. language use and parent educational background). Urban students tended to be more positive than the rural students, but. this not could be generalised due to a small number of items with significant differences. Male students showed more positive attitudes to science than female students, but this was limited to a small number of items. The students with above average scores in the cognitive test showed more positive attitudes to science than those of average and below verage scores. However, the number of items were quite small, and therefore could not be generalised. Points of improvement in the practice of teaching and future research in attitudes to science are recommended. For instance, the present officially prescribed time of 40 minutes per week for science should be changed to 1.5 to 2 hours after further investigation. The reason for this is that it requires some preparation before actual lessons begin (e.g. collecting materials by groups) and the students are generally slow.

Abstract:
Education is often viewed as a key approach to address sexual-health issues; the current concern is the burgeoning HIV/AIDS epidemic. This ethnographic study investigates the gender practices associated with high-risk sexual behaviour in Papua New Guinea as viewed by educators there. A number of practices, including gender inequality and associated sexual behaviours have been highlighted by male and female participants as escalating PNG’s HIV/AIDS epidemic. The study finds that although participants were well-informed concerning HIV/AIDS, they had varying beliefs concerning the prevailing gender/sexual issues involved in escalating highrisk behaviour and how to address the problem. The study further examines the behavioural beliefs and intentions of the educators themselves. Subsequently, within the data a number of underpinning factors, pertaining to gender, education and life experience, were found to be related to the behaviour beliefs and intentions of participants towards embracing change with regard to behaviours associated with gender equality in PNG. These factors appeared to encourage participants to adopt healthier gender and sexual behavioural intentions and, arguably, could provide the basis for ways to help address the gender inequality and high-risk behaviours associated with HIV/AIDS in PNG.

Abstract:
Guided missiles are among the most effective threats against air platforms. Aircraft and helicopter losses in the last decades were mostly due to guided missiles, 70% of which were infrared guided missiles. Today, there are as many as 500,000 shoulder-fired missiles in military arsenals around the world, whose guidance algorithms enable them to track the desired trajectories very precisely. In this thesis, main focus is on defining infrared missile guidance and control algorithms in order to study on various target-missile scenarios for the effectiveness of these algorithms. First, a mathematical model of a generic missile is given. Then the flight control system of the missile is created by using LQR and PID controllers. Different kinds of PNG algorithms applied to an infrared missile are presented. The seeker part of the infrared missile is also discussed. The effectiveness of guidance algorithms are studied based on different target &ndash
missile scenarios and the responses of them to IR countermeasures are also observed. This study shows that different guidance algorithms can be used for different scenarios. If suitable algorithms are combined and suitable constants are applied, the guided missile can track the target very precisely. In addition, the seeker part has to be improved with tracking algorithms in order to recognize IR-countermeasures and not to follow them.

Signal compression is necessary part for ECG scanning, because of relatively big amount of data, which must be transmitted primarily wirelessly for analysis. Because of the wireless sending it is necessary to minimize the amount of data as much as possible. To minimize the amount of data, lossless or lossy compression algorithms are used. This work describes an algorithm SPITH and newly created experimental method, based on PNG, and their testing. This master’s thesis there is also a bank of ECG signals with parallel sensed accelerometer data. In the last part, modification of SPIHT algorithm, which uses accelerometer data, is described and realized.

Petrone, Rosalie Catherine. "Using Commercially Available Hormones to Enhance Swine Reproductive Efficiency in Batch Management Systems." Thesis, Virginia Tech, 2015. http://hdl.handle.net/10919/54594.

Full text
Abstract:
The U.S. hog industry's shift to vertically-integrated, intensively-managed operations brought about a variety of management systems for breeding herds, including batch farrowing. In this system, groups of sows are weaned in 2- to 5- week intervals, making estrus synchronization of new gilts and sows critical to maintaining reproductive efficiency in the herd. The use of commercially available hormones to synchronize estrus in this system has not been extensively studied. This experiment was conducted to determine whether the use of commercially available hormones (MATRIX® and P.G. 600®; Merck Animal Health, De Sota, KS) in a 5-week batch management system had a positive impact on reproductive efficiency in gilts and sows over the course of 3 parities. Gilts were allocated to an Entry Group (A, B, or D) and then assigned to a treatment, Hormone-Assisted (HA) (5 mL P.G. 600 injection 5 days and/or fed 15 mg/day of MATRIX for 14 consecutive days prior to the breeding week) or Control (no exogenous hormones). Gilts and sows were checked daily for estrus with a mature boar, and a group was bred using AI during a 7-day breeding period every 5 weeks and allowed to farrow up to 3 parities. Among groups, there was a tendency (P = 0.08) for more HA than control gilts to display estrus and be mated on schedule. For gilts within Group A, more (P < 0.01) HA than control females displayed estrus and were mated. Body weight at first service for gilts in Groups A and B were higher than Group D (P < 0.01). Overall, there was no effect of treatment on (P = 0.20) non-productive days. There was a strong tendency for HA sows to have a greater (P = 0.07) number of parities completed than control sows; Total pigs born (P < 0.05) and total pigs born alive (P < 0.05) were greater for HA sows than control sows. In Parity 1, Group D sows had a lesser number of pigs born (P < 0.01) and pigs born alive (P < 0.02) than Groups A and B; The number of pigs weaned differed between entry groups (P < 0.05) (Group B > Group A > Group D); Control sows weaned more pigs (P < 0.02) and had a greater litter weaning weight (P < 0.01) than HA sows; HA sows had a lower (P < 0.05) wean-to-estrus interval than control sows. No significant effects of group or treatment were observed in Parities 2 and 3. The use of exogenous hormones to synchronize estrus had a positive impact on reproductive efficiency in HA gilts/sows in a 5-week batch management system.

Abstract:
MapReduce is a wide-spread programming model for processing big amounts of data in parallel. PACT is a generalization of MapReduce, based on the concept of Parallelization Contracts (PACTs). Writing efficient applications in MapReduce or PACT requires strong programming skills and in-depth understanding of the systems’ architectures. Several high-level languages have been developed, in order to make the power of these systems accessible to non-experts, save development time and make application code easier to understand and maintain. One of the most popular high-level dataflow systems is Apache Pig. Pig overcomes Hadoop’s oneinput and two-stage dataflow limitations, allowing the developer to write SQL-like scripts. However, Hadoop’s limitations are still present in the backend system and add a notable overhead to the execution time. Pig is currently implemented on top of Hadoop, however it has been designed to be modular and independent of the execution engine. In this thesis project, we propose the integration of Pig with another framework for parallel data processing, Stratosphere. We show that Stratosphere has desirable properties that significantly improve Pig’s performance. We present an algorithm that translates Pig Latin scripts into PACT programs that can be executed on the Nephele execution engine. We also present a prototype system that we have developed and we provide measurements on a set of basic Pig scripts and their native MapReduce and PACT implementations. We show that the Pig-Stratosphere integration is very promising and can lead to Pig scripts executing even more efficiently than native MapReduce applications.

Abstract:
The effectiveness of the PEG ratio as a valuation tool has been a topical debate between market commentators ever since being popularised by Lynch (1989). This study examines the appropriateness of the fair value criteria of 1.0 (PEGL) in comparison with a time-series based share specific benchmarking model (PEGT). Furthermore, influencing factors of analyst forecasting accuracy, namely: the number of analyst contributions, forecast dispersion and forecast horizon, were tested and compared using sub-set portfolios for each category with the objective of identifying a possible optimal PEG trading rule strategy. The outcome showed a consistent outperformance of PEGT portfolios compared to PEGL portfolios and the market benchmark. Unexpected results were obtained for the impact of analyst forecasts on the performance of the PEG ratio with additional literature review providing possible reasons that analyst optimism may have a more influencing impact on the PEG ratio than forecasting accuracy. Finally, an optimised PEG trading rule strategy delivered annual abnormal returns of 5.4% (CAGR: 19.7%) for a PEGL portfolio, versus that of 13.7% (CAGR: 28.5%) for a PEGT portfolio. The ensuing methodology appeared to single out small cap firms with above market growth prospects. Copyright

Abstract:
The developmental stage of certain diseases is closely linked to the concentration of various proteins in biological samples. A sensitive detection of these so-called biomarkers can thus significantly contribute to an early diagnosis. In the present work, structured, fluorogenic probes were developed that offer the possibility of a sensitive, direct and in real-time detection of target proteins in a homogeneous process. The peptidic recognition sequence for the target protein was thereby flanked by two self-complementary PNA segments. As a result, the probes possessed a hairpin-type arrangement, in which suitable appended labels are forced into close proximity and guaranteed a minimal background signal in the unbound state. By interacting with the target protein a reorganization of the probe structure occured, which could be followed by fluorescence spectroscopy. To embed the fluorogenic units different approaches were developed and the resulting architectures were validated relating to their use as a sensitive detection system. As target proteins the intracellular SH2-domains of the Src and Lck kinase and the extracellular matrix metalloprotease MMP-7, a proteolytic biomarker for cancer, were investigated. In particular, the new In-Stem Hairpin Peptide Beacons (IS-HPBs), in which fluorogenic pseudo nucleic acids were incorporated into the PNA-stem region, proved as sensitive protease reporters with an up to 50-fold signal amplification. By using an excimer-signaling IS-HPB and a time-resolved fluorescence method the direct detection of MMP-7 with a critical concentration of 1 nM within complex human blood serum was achieved. A possible application in medical diagnostics was thus confirmed. Furthermore, initial indications were obtained using thermodynamic studies that the structure of a peptide-based probe contributes to increased selectivity.

Abstract:
Pigs transgenic for hDAF were created by microinjection of human DNA into fertilized pig oocytes. Ten control hearts, eight heterozygous hDAF transgenic hearts and one homozygous hDAF transgenic heart, were transplanted heterotopically into the abdomen of non-immunosuppressed cynomolgus monkeys. Fifteen heterozygous transgenic and five control hearts were then transplanted heterotopically into the abdomen of immunosuppressed cynomolgus monkeys. Of the transgenic hearts ten were immunosuppressed with a high cyclophosphamide (CyP) regimen (group I) and five were immunosuppressed with a low CyP regimen (group 2). All control hearts were immunosuppressed with the high CyP regimen. The hDAF transgenic hearts in the high CyP group had a median beating time of 40 days (range 6-62 days) compared with 9 days (range 8-34 days) for those in the low CyP group. This difference was statistically significant (p <0.05). The control grafts had a median beating time of 55 minutes (range 2-180 minutes). The difference between the control grafts and the high CyP transgenics was statistically highly significant (p <0.001). The difference between the control grafts and the low CyP transgenics was also statistically significant (p <0.05). All control hearts were hyperacutely rejected. Only two hearts in the high CyP group were lost due to AVR, on days 6 and 62: all other hearts showed no evidence of rejection. All hDAF transgenic hearts in the low CyP group underwent AVR. These results confirm that HAR can be abrogated with the hDAF transgene in this pig-to-primate model and that prolonged survival can be achieved. Perioperative CyP would appear to be an essential component of this regimen, without which, graft loss due to antibody-mediated AVR is encountered. Finally, hDAF transgenic pig hearts were transplanted both heterotopically, and orthotopically into immunosuppressed baboons. No graft underwent HAR and survival of heterotopic pig-to-baboon heart xenografts for up to 21 days was achieved.

Abstract:
The purpose of this paper is to examine teachers' approach to popular culture in preschool. The questions we want answered are: What approach do educators have to popular culture in preschool and what underlies that approach? Is there a high - and low culture in preschool? If educators bring popular culture into the preschool, how then is it used? Do children’s culture influence everyday life in preschool?  We have employed qualitative methods to seek answers to our questions. We conducted seven interviews with educators from four different preschools. The starting point for our study was the socio-cultural perspective because we examined people's cultures and how the meeting between them unfolds.  We concluded that educators have a very ambivalent attitude towards popular culture in preschool. Respondents could see popular culture as positive for children's learning, because it makes learning meaningful and enjoyable for the children. But at the same time, they had difficulty relating to popular culture because of its commercialism, the strongly gender-coded roles that frequently occur and the status created around owning the products. We also observed a strong high culture that contrast with popular culture in preschool. High culture was linked to educators' own experiences and values, and to how instructive and useful cultural phenomenon were for the children. Popular culture needs to be connected with a clear learning objective for teachers to consider taking it into the preschool. During the investigation we discovered that teachers enjoy great power over what is brought into the preschool and over what is held out of children's reach. Children's influence over their own culture is limited in preschool. The most prominent aspect of the study was educators' own ambivalence towards popular culture.

Chandra, Fiona Adriani. "Limits and Tradeoffs in the Control of Autocatalytic Systems." Thesis, 2013. https://thesis.library.caltech.edu/7825/4/image72.png.

Full text
Abstract:
Despite the complexity of biological networks, we find that certain common architectures govern network structures. These architectures impose fundamental constraints on system performance and create tradeoffs that the system must balance in the face of uncertainty in the environment. This means that while a system may be optimized for a specific function through evolution, the optimal achievable state must follow these constraints. One such constraining architecture is autocatalysis, as seen in many biological networks including glycolysis and ribosomal protein synthesis. Using a minimal model, we show that ATP autocatalysis in glycolysis imposes stability and performance constraints and that the experimentally well-studied glycolytic oscillations are in fact a consequence of a tradeoff between error minimization and stability. We also show that additional complexity in the network results in increased robustness. Ribosome synthesis is also autocatalytic where ribosomes must be used to make more ribosomal proteins. When ribosomes have higher protein content, the autocatalysis is increased. We show that this autocatalysis destabilizes the system, slows down response, and also constrains the system’s performance. On a larger scale, transcriptional regulation of whole organisms also follows architectural constraints and this can be seen in the differences between bacterial and yeast transcription networks. We show that the degree distributions of bacterial transcription network follow a power law distribution while the yeast network follows an exponential distribution. We then explored the evolutionary models that have previously been proposed and show that neither the preferential linking model nor the duplication-divergence model of network evolution generates the power-law, hierarchical structure found in bacteria. However, in real biological systems, the generation of new nodes occurs through both duplication and horizontal gene transfers, and we show that a biologically reasonable combination of the two mechanisms generates the desired network.

Abstract:
Computation technology has dramatically changed the world around us; you can hardly find an area where cell phones have not saturated the market, yet there is a significant lack of breakthroughs in the development to integrate the computer with biological environments. This is largely the result of the incompatibility of the materials used in both environments; biological environments and experiments tend to need aqueous environments. To help aid in these development chemists, engineers, physicists and biologists have begun to develop microfluidics to help bridge this divide. Unfortunately, the microfluidic devices required large external support equipment to run the device. This thesis presents a series of several microfluidic methods that can help integrate engineering and biology by exploiting nanotechnology to help push the field of microfluidics back to its intended purpose, small integrated biological and electrical devices. I demonstrate this goal by developing different methods and devices to (1) separate membrane bound proteins with the use of microfluidics, (2) use optical technology to make fiber optic cables into protein sensors, (3) generate new fluidic devices using semiconductor material to manipulate single cells, and (4) develop a new genetic microfluidic based diagnostic assay that works with current PCR methodology to provide faster and cheaper results. All of these methods and systems can be used as components to build a self-contained biomedical device.

Letsou, William Peter. "Noncommutative Biology: Sequential Regulation of Complex Networks and Connected Matter." Thesis, 2018. https://thesis.library.caltech.edu/10966/1/fig1-1.png.

Full text
Abstract:

During animal development from zygote to adult, a limited set of regulatory molecules are autonomously deployed in the service of tissue-specific gene expression (reviewed in chapter 1). Inherent in the process is the tension that single cells sample heterogeneous expression states while robustly maintaining a collective final outcome. This thesis addresses theoretical issues that help resolve the paradox that one cell simultaneously contains the fate information of many.

Previous models of development have likened cell fate to minima on a smooth potential energy surface. Such static pictures can be misleading because they suggest the egg knows the path it will take to the adult before it divides even once. Recognition that the potential analogy is an oversimplification has led others to propose that the surface is actually nonsmooth. Chapter 2 reviews the theoretical basis for smooth potentials and resolves these problems by appealing to the tangent space of gene expression. It is then shown that if the potential difference is sufficient to characterize the difference between egg and adult, then the tangent space controls on gene expression are one-dimensional. Furthermore, a shortcoming of models ignoring the connectivity and common origin of dividing cells is that they erect artificial barriers between alternative fates. A fundamentally different picture is sketched wherein the difference between egg and adult is schematized as the shape of the locus of equipotential fates accessible at the same point in time. The conjugacy of space and time is invoked to explain how the requirement that each fate be on a line of equipotential is the same as requiring that each alternative fate move the same distance down the surface at each step. The developmental trajectory is deterministic but not known in advance because it needs to be ascertained at each step which way cells "turn" in order to maintain their equipotential relationship. Chapters 3 and 4 refine this sequential model of collective development with specific examples.

A simple solution to the problem of cell-type specific gene expression is combinatorial binding of transcription factors at promoters. It is shown in chapter 3 that such models result in substantial information bottlenecks, because all cell fate information is concentrated at the start. We explore a novel, noncommutative model of gene regulation—known as sequential logic—that spreads the information out over time. It is shown using time sequences of noncommutative controllers that targets which otherwise would have been activated together can be regulated independently. We derive scaling laws for two noncommutative models of regulation, motivated by phosphorylation/neural networks and chromosome folding, respectively, and show that they scale super-exponentially in the number of regulators. It is also shown that specificity in control is robust to loss of a regulator. Consequently, sequential logic overcomes the information bottleneck in complex problems and enables novel solutions through roundabout strategies. The theoretical results are connected to real biological networks demonstrating specificity in the context of promiscuity.

Noncommutative sequential logic has improved storage capacity, but it does not specify who or what supplies the sequences of input that determine cell fate. Chapter 4 offers a solution by way of the seemingly unrelated problem of looping in twisted strings. Cells and strings obey a set of common space-time constraints, ultimately due to the conservation of energy. It is argued that the most parsimonious allocation of energy from the straight to strained string is the one in which each segment sees the same share of the total. Planar looping is shown to be a consequence of the parsimony principle and the Euler-Poincaré equations for rotational motion in the presence an applied torque. We then solve the problem for the looping of a twisted string; with two strains, the Euler-Poincaré predict a different answer than the classical Frenet-Serret equations. Using the results of chapter 2, it is concluded that the Frenet-Serret curvatures assigned ahead of time are not guaranteed to generate space curves that conserve energy: the predicted string has localized strains the Euler-Poincaré solution lacks. Rotational dynamics of strings are connected to developing organisms by postulating conserved RNA polymerase as an analog of angular momentum, and transcriptional activity as energy. Alternative fates along a one-dimensional "string" of dividing cells are possible by finding the RNAP distribution that conserves transcriptional activity along a curve of constant developmental potential. Consequently, each alternative fate samples a different sequence of changes to the distribution as it follows a local gradient downhill from high to low developmental potential over time.

In conclusion, regulation in the tangent space of gene expression resolves the paradox that development has a unique solution specified in the DNA of the egg which cannot be determined with certainty until completion of the adult. Noncommutative sequential logic generates complexity that cannot be realized at the start, while interdependent cells (and strings) require time to ensure that each fate is at the same potential difference from a common ancestor. This fundamental reimagining of the Waddington framework can be tested using new multiplexed mRNA imaging technologies that preserve the spatial context of cells in developing tissue.

Leke, Daniel Kaki. "Vocational education and training in secondary schools: investigation into a VET model for Papua New Guinea." Thesis, 2010. http://hdl.handle.net/2440/64581.

Full text
Abstract:
Vocational education and training (VET) was introduced into the secondary school curriculum in many developed countries because of its economic and social benefits. Papua New Guinea (PNG), a developing nation, appeared to overlook VET as an important tool for economic and social development since independence in 1975. Its secondary schools have provided only an academic pathway for the top quarter of its students. Thousands of secondary school students become school leavers at Grade 10 and 12 levels, without having VET knowledge and employability skills/attributes. This student transition problem negatively impacted on PNG society through high level youth unemployment and unrest. The purpose of this study was to investigate and determine the characteristics of a VET program model at the secondary school level which might address the current student transition problem in PNG. The following question was used to direct this research: How can a VET model address and minimise the current student transition problem at the secondary school level in Papua New Guinea? This study applied the positivist (qualitative) and descriptive (quantitative) mixed methodology. The use of mixed/triangulation methods was considered important because one data collection form would supply strengths to offset the possible weaknesses of the others. The data was gathered using applied discourse/text analysis, questionnaires and semi-structured interviews from purposely selected PNG stakeholders: students, teachers, policy-makers/school principals and business/industry personnel. The results illustrated how VET in PNG was given little priority at the policy level. Most stakeholders thought that academic subjects would assure a better future than VET subjects. Although VET subjects and employability skills and attributes were considered to have some importance by most stakeholders, almost always they expressed some reservations. Most students and teachers thought that VET subjects were less valuable than academic subjects. Similarly, most business/industry personnel expressed that postgraduate students and other tertiary graduates possessed better skills for work than the secondary school graduates. This was because VET programs were restricted to post-secondary schools or TVET colleges. Most stakeholders failed to realise that the academic subjects could be studied only by a minority students. Teachers and school principals were trying their best to manage their schools within their means. Most did not have the required teaching and learning facilities to implement the new VET subjects at secondary level. These reforms were put in jeopardy because of the lack of resources, especially classroom space, training facilities and qualified VET teachers. Although the stakeholders were positive about the importance of VET, there was little evidence of actual participation in VET programs at the secondary school level. The results illustrated a need to embrace both VET and general academic subjects as an integral part of secondary school level in PNG. Therefore, the study concluded that PNG needs to consider and implement an integrated academic/VET program model at the secondary school level. This integrated academic/VET model would enhance many students' pathways for further education and training, employment and improvement of life in PNG.

With the convenience and fast speed on the Internet, exchanges of multimedia become more and more frequent nowadays. PNG images are used in a wide variety of applications. In this study, we propose several methods for data hiding applications via PNG images, including copyright protection, secret sharing, and covert communication. For copyright protection, we propose a block-based method for file authentication, which can authenticate the integrity and fidelity of PNG images by adjusting pixel values to meet some constraints in the three color channels (red, green, and blue) and the alpha channel. For secret sharing, we propose a method by solving three simultaneous equations to generate n shares from n 讣汹轨 cover images, and embedding the shares in the alpha channels of the color images. We can recover the secret by collecting all the shares. For covert communication, we propose two data hiding methods which are based on the proposed authentication method and the proposed secret sharing method. We also apply the two methods to yield a combined data hiding and authentication method to authenticate hidden data. Good experimental results show the feasibility of the proposed methods.

Abstract:
In May, 2005, the Papua New Guinea (PNG) National Department of Health (NDoH) determined that the Integrated Management of Adult and Adolescent Illness (IMAI) Chronic HIV Care training program, modified for the PNG context, would serve as the basis for the development of teams for the Rapid Scale-Up of HIV/AIDS Care and Treatment. The IMAI Chronic HIV Care training program was developed as a joint effort carried out in a working partnership involving WHO, Geneva, the WHO Regional Office for Africa (AFRO), as well as many international collaborating institutions. Trials and implementation of the program have occurred predominantly on the African continent and to date there has been no research undertaken to examine the effectiveness of the IMAI program as a learning modality for preparing registered nurses to provide comprehensive HIV care. The purpose of this hermeneutic informed interpretive research study is to explore and describe how registered nurses in Papua New Guinea make sense of their learning and implementation of the IMAI training program in providing Comprehensive Care and treatment to people living with HIV/AIDS. The study is a mixed methods study. The rationale for this approach was to better understand the research problem by converging both broad numerical trends from quantitative research and the detail of qualitative research with the intent of producing a richer and more complete picture of the investigated phenomenon. Data collection included a quantitative survey, semi-structured interviews and a focus group. These data informed the development of descriptive accounts that allowed for the subsequent identification of common and divergent themes reflective of factors that influenced nurses learning and implementation of IMAI training. The findings from the quantitative survey revealed that all respondents had a positive impression of the IMAI program and expressed the view that the IMAI program had a positive effect on various aspects of patient care and their learning and experience. Overall, the survey identified that registered nurses who participated in the IMAI Chronic HIV Care training program perceived the program to be beneficial for improving the way HIV care is provided. Three major themes were identified from the qualitative analysis, and under these major themes data was arranged as sub-themes. The three major themes identified were Overcoming Personal Anxiety, Actioning Learning and Identifying Challenges. The qualitative analysis revealed that overwhelmingly, the fear of anxiety of HIV was significant for all participants of the semi-structured interviews. Nurses in the focus group felt that the overt expression of fear given by the interview participants more accurately reflected the reality for the majority of nurses in Papua New Guinea. Most nurses also identified that their participation in the IMAI program contributed to their personal change process and helped them to identify and understand their fear of HIV. Expert patient trainers (EPTs) were a highly valued component of the flexible IMAI training methodology and were seen by nurses as being important both for learning and for assisting them to overcome their personal anxiety created by the fear of HIV. The findings also identified that the key challenge faced by nurses in attempting to implement their learning was that of system issues including a lack of medical supplies, including antiretroviral drugs (ART), lack of staff and an absence of continuing professional education. These issues require enhanced commitment from the NDoH if nurses are to be effective in implementing their learning to provide HIV care and treatment. It is also important that NDoH give support to enable the establishment and implementation of continuing education for nurses providing HIV care and treatment and that the development of processes to ensure the ongoing maintenance of quality in HIV care and treatment training be implemented as a matter of priority. Finally this study has provided an in-depth understanding of the experiences of how registered nurses in Papua New Guinea make sense of their learning and implementation of the IMAI training program in providing comprehensive HIV care and treatment to people living with HIV/AIDS. The current multitude of factors fueling the HIV/AIDS epidemic in Papua New Guinea are explored as is the current status of HIV/AIDS in New Guinea, and the multitude of cultural patterns, social, and political factors influencing the spread of HIV within the country. This exploration also undertakes a comprehensive discussion of cultural issues related to gender inequality including a rich description of changing cultural patterns and values in Papua New Guinea.

Abstract:
Research Doctorate - PhD Human Geography
Abstract: This thesis is a call for greater reflexivity on the role of emotions in development. It argues that emotions are integral in both reproductions of Orientalisms and in producing resistances to Orientalisms. The framework for this research is a combination of Postcolonial Studies and Emotional Geographies, and a case study of HIV and AIDS development work in Papua New Guinea (PNG). The methods used are several forms of critical discourse analysis applied to: mainstream development research; development policy (at several levels); in-country experience; and the middle ground between policy and policy enactment. The data sources were: 30 mainstream HIV and AIDS development research articles; 4 policy documents; 20 in depth interviews; and a free writing journal documenting 3 months of participant observations in a PNG HIV and AIDS development organisation. Results reveal evidence of emotions, of attempts to avoid unpleasant emotions, and of attempts to elicit emotions in others in all aspects of the development process. In the case of PNG HIV and AIDS Development work, it seems that emotions, or the attempts to influence emotions, have the potential to promote or resist Orientalisms. Of particular note are feelings of uncertainty, confusion and disorientation. If one is reflexive, these emotions may be powerful in generating resistance to Orientalisms, but without reflexivity they can reproduce them in powerful ways. I conclude this thesis by claiming that a reflexive understanding of the roles of emotions can uncover covert and persistent forms of Orientalisms in development work. Emotional reflexivity can also help find new ways of thinking and being that can move the ‘West’ beyond Orientalisms. In particular, I call for more reflexive ways of embracing and accepting the uncertainty that in inherent in the development process.

Abstract:
My thesis is an exploration of how human rights are understood and experienced by Papua New Guinean people living with HIV, men with diverse sexualities and transgender women. I examine the emergence and progress of two organisations established by these communities: Igat Hope and Kapul Champions. I consider the ways these organisations have, while still pursuing their missions to limit the impacts of HIV on their members and all Papua New Guineans, been used to help promote the human rights of constituents. Through this examination, I reflect on my own history with these organisations and the community movements which have built them, and on my roles as advocate and researcher. I find that the human rights of people with HIV, men with diverse sexualities and trans women in PNG are regularly violated across many areas of life. I find these communities have an understanding of human rights and that they support their promotion, particularly rights analyses that recognise the value they place on community connection and that emphasise their inclusion. I see human rights in PNG as a dynamic space, with an indigenisation of the rights framework that will facilitate its application. I find both Igat Hope and Kapul Champions to have significant records of achievement, including in human rights. I also find a way to be comfortable in my multiple roles, suggesting some ways in which my research might support local advocacy efforts.

Oko, Jerome. "An investigation into factors affecting Grade 10 and 12 students’ mathematics performance in Port Moresby, Papua New Guinea (PNG)." Thesis, 2021. https://hdl.handle.net/2440/134190.

Full text
Abstract:
Though Papua New Guinea (PNG) has great development potential with its natural resources, the country faces challenges with its education system. One of these challenges is the poor mathematics performance of students at secondary schools. This poor performance is evident both through the significant number of students who cannot continue to Grade 11, and through the simultaneous decline in student enrolment in science related degrees at the university level. That being the case, this study aims to examine the school, teacher and student-level factors and their interrelationships that affect Grade 10 and 12 students’ mathematics results. A quantitative-dominant mixed-method approach is employed in this study, which has 729 student and 41 teacher participants. The instruments used in this study are: survey questionnaires, mathematics test, and interview questions. The survey questionnaires for students and teachers are adopted from international studies such as Trends in International Mathematics and Science Study (TIMSS) and Programme for International Student Assessment (PISA) and are modified according to the context of this study. The mathematics test questions are adopted and modified from past Grade 10 and 12 national mathematics examination papers, respectively. These survey questionnaires and tests are validated and verified through confirmatory factor analysis and Rasch analysis. Rasch analysis scores obtained from the survey questionnaires and tests are used to examine the relationships between the independent and dependent variables at the teacher and student-levels through the use of Structural Equation Modelling, respectively. Hierarchical Linear Modelling is also employed to examine the direct effects from teacher and student-levels and cross-interaction effects between variables at these two different levels. The interview data at the teacher-level is analysed through thematic approach to capture information that might have been missed in teachers’ survey questionnaires to complement the findings of quantitative (survey) data. A number of significant factors (attitude, motivation, fathers’ occupation, private schools, students’ gender, teachers’ with mathematics major and quality of teaching) that are influencing students’ mathematics results are identified by this study, with mothers’ highest education level found to be the most critical factor. The study concludes that more attention should be given to the factors identified in this study, in order to improve Grade 10 and 12 students’ mathematics results. Overall, this study contributes theoretical, methodological, and practical knowledge to teaching and learning in mathematics education.

Majer, Günter, and Alexander Southan. "PFG-NMR studies of ATP diffusion in PEG-DA hydrogels and aqueous solutions of PEG-DA polymers." 2017. https://ul.qucosa.de/id/qucosa%3A31633.

Full text
Abstract:
Adenosine triphosphate (ATP) is the major carrier of chemical energy in cells. The diffusion of ATP in hydrogels, which have a structural resemblance to the natural extracellular matrix, is therefore of great importance to understand many biological processes. In continuation of our recent studies of ATP diffusion in poly(ethylene glycol) diacrylate (PEG-DA) hydrogels by pulsed field gradient nuclear magnetic resonance (PFG-NMR), we present precise diffusion measurements of ATP in aqueous solutions of PEG-DA polymers, which are not cross-linked to a three-dimensional network. The dependence of the ATP diffusion on the polymer volume fraction in the hydrogels, φ, was found to be consistent with the predictions of a modified obstruction model or the free volume theory in combination with the sieving behavior of the polymer chains. The present measurements of ATP diffusion in aqueous solutions of the polymers revealed that the diffusion coefficient is determined by φ only, regardless of whether the polymers are cross-linked or not. These results seem to be inconsistent with the free volume model, according to which voids are formed by a statistical redistribution of surrounding molecules, which is expected to occur more frequently in the case of not cross-linked polymers. The present results indicate that ATP diffusion takes place only in the aqueous regions of the systems, with the volume fraction of the polymers, including a solvating water layer, being blocked for the ATP molecules. The solvating water layer increases the effective volume of the polymers by 66%. This modified obstruction model is most appropriate to correctly describe the ATP diffusion in PEG-DA hydrogels.

Döhler, Christian. "Komnzo: A language of Southern New Guinea." Phd thesis, 2016. http://hdl.handle.net/1885/107178.

Full text
Abstract:
This thesis provides an introduction to Komnzo, a Papuan language of Southern New Guinea. Komnzo is spoken by around 200 people in the village of Rouku and a couple of adjacent hamlets. Komnzo belongs to the Tonda subgroup of the Yam language family, which is also known as the Morehead Upper-Maro group. This grammar provides the first comprehensive description of a Yam language. It is based on 16 months of fieldwork. The primary source of data is a text corpus which the author recorded and transcribed between 2010 and 2015. The corpus adds up to ten hours of text including narratives, procedurals and naturally occuring social interaction. The sequence of chapters follows the well-established order: phonology (2), word classes (3), nominal morphology (4), verb morphology (5) and TAM marking (6), noun phrase syntax (7), clausal syntax (8), interclausal syntax (9) and information structure (10). These chapters are supplemented by an anthropological, historical and sociolinguistic introduction (1), and they are rounded off by a chapter on lexicology (11). The appendix includes a 70-page dictionary with 1850 entries and three sample texts. The entire text corpus is accessible online. Komnzo provides many fields of future research, but the most interesting aspect of its structure lies in the verb morphology, to which the two largest chapters of the grammar are dedicated. Komnzo verbs may index up to two arguments showing agreement in person, number and gender. Verbs encode 18 TAM categories, valency, directionality and deictic status. Morphological complexity lies not only in the amount of categories verbs may express, but also in the way how these are encoded. Komnzo verbs exhibit what may be called `distributed exponence', that is single morphemes are underspecified for a particular grammatical category. Therefore, morphological material from different slots has to be integrated first, and only after this integration, one can arrive at a particular grammatical category. The descriptive approach in this grammar is theory-informed rather than theory-driven. Comparison to other Yam languages and diachronic developments are taken into account whenever it seems helpful.
