Abstract:
Electron transport systems exist in the plasma membranes of all cells. Although not well characterised they play roles in cell growth and proliferation, hormone responses and other cell signalling events, but perhaps their most important role, especially in erythrocytes, is enabling the cell to respond to changes in both intra- and extracellular redox environments. Human erythrocytes possess a transmembrane electron transport capability that mediates the transfer of reducing equivalents from reduced intracellular species to oxidised extracellular species and is concomitant with proton extrusion. In the work for this thesis I showed that erythrocyte membranes contain a transmembrane WST-1 (water soluble tetrazolium-1) reductase activity that uses reducing equivalents from intracellular NADH to reduce extracellular WST-1. The rate of WST-1 reduction was increased by the presence of phenazine methosulfate and, although of low activity, it showed similar properties to a previously reported transmembrane NADH-oxidase activity. 1H NMR experiments showed that WST-1 was reversibly bound to the membrane and/or proteins in the membrane within the timeframe of the NMR experiment, confirming the location of the WST-1 reduction. Preliminary attempts to purify NADH:WST-1 reductase and NADH:ferricyanide reductase activities from the erythrocyte plasma membrane were inconclusive. The protein(s) responsible for the reduction of these oxidants appear to be of low abundance in the plasma membrane and may be part of a larger protein complex. Further work on the isolation of these redox activities is required before the protein(s) involved can be identified with any confidence. The ability of cells to export electrons suggests that an electron import mechanism might also exist to re-establish the cell�s redox-buffering equilibrium under conditions of oxidative stress. This hypothesis was tested in glucose-deprived erythrocytes using reduced glutathione and NADH as extracellular electron donors. It was shown that neither reduced glutathione nor NADH donated reducing equivalents through a transmembrane redox system. Extracellular NADH was, however, able to produce profound changes in starvation metabolism and methaemoglobin reduction rates. The addition of extracellular NADH caused a six-fold increase in the rate of lactate production above that observed in glucose-starved controls, together with a concomitant decrease in pyruvate production. In erythrocytes containing high levels of methaemoglobin, extracellular NADH increased the rate of methaemoglobin reduction in both the presence and absence of glucose. These results were explained by the leakage of lactate dehydrogenase from erythrocytes due to an admittedly low level of haemolysis. This caused the displacement of the intracellular pseudo-equilibrium of the lactate dehydrogenase reaction via transmembrane exchange of lactate, allowing the conversion of extracellular pyruvate to lactate and resulted in an increase in intracellular NADH concentrations. The latter increased the rate of methaemoglobin reduction. In conclusion, the work described in this thesis showed that erythrocyte membranes do not contain mechanisms for importing electrons or reducing equivalents from extracellular reduced glutathione or NADH. Erythrocytes do, however, contain an electron export system which can reduce extracellular oxidants such as WST-1 and the activity of this system depends on an intricate balance between intracellular antioxidants and enzyme activities. There is much still to be learnt about plasma membrane redox systems, little is known, for example, about the protein composition, mechanism of action, and the in vivo conditions under which these systems are most active.

Cette these presente un composant a pixel d'un type nouveau, qui pourra etre utilise pour la construction de detecteurs de trajectoires a grande resolution, pour les experiences de physique des particules. Ce pixel pmos a ete etudie dans le cadre de la recherche et developpement de l'experience belle a kek. Le pixel pmos, situe sur la face ohmique d'une diode pin desertee, se compose d'un puits-n ou un transistor pmos double drains est implante. Les sorties aux bornes des drains des pixels pmos du reseau, sont reliees entre elles par des strips de lecture. Ce pixel pmos a des caracteristiques interessantes: une amplification interne qui permet de detecter des particules chargees avec un signal-sur-bruit important, et l'avantage d'une lecture bi-dimensionnelle de type strip pour un composant simple face. Nous avons concu et fabrique des prototypes simplifies du pixel pmos, de differents types. A l'aide d'un faisceau infrarouge, nous avons verifie le principe de fonctionnement d'un pixel compose d'un transistor pmos double drains implante dans l'electrode d'une diode pin, ainsi que la lecture bi-dimensionnelle de type strip. L'amplification et les performances de bruit du pixel pmos ont ete estimees. Un test sur faisceau de particules au minium d'ionisation nous a permis de mesurer la distribution du depot d'energie et la resolution de position, en accord avec les simulations effectuees

The objective of this work was to identify what are the main divergences found between what the theory preconizes as scope and approach of the Project Management Offices (PMOs) and what really happens when practice is observed on site. Such intend was achieved with the support of a case study analysis. The theme may be considered relevant based on the fact that it provides a better comprehension about how the PMOs structures really operate. Specially regarding to scope, that is, whether the PMO is a Program-Project, a Departmental PMO, or a Corporate PMO, and regarding its range, that is, if its action is operational, tactical or strategic. Qualitative methodology was used in this research, through a case study approach. A large size manufacturing company not structured by projects was chosen to be evaluated. This company has a complex organizational structure and such characteristic makes possible a comprehensive study. The performance of the PMOs addressed in the case study was discussed taking in consideration the literature and also the information collected from the participant observation. The results of this work allows to conclude that the assumption of the research, that there are divergences between what the theory preconizes as scope and approach of the PMOs and what really happens on practice, is confirmed through the case study. Beyond that, there is not a clear causality relation between the amplitude of action of those PMOs and its hierarchic position within the organization. Also there is not a pattern of action of the PMOs when compared to functions that they perform. Finally, recommendations were made related to the case study presented. The main ones are shown as follows. It is important to hold a multi case study for a better comprehension of the theme with at least one company structured by projects and other one non-structured by projects in order to compare the performance of their PMOs aiming to better understand the theme. To include more stakeholders among the group of the interviewed people within the case study. To hold a survey with various companies of various segments inside the national territory. Also to perform a study where it may be verified whether different organizational typologies may affect the practice of PMOs in the organizations.
Neste trabalho, o objetivo é identificar, a partir da análise de um estudo de caso, quais são as principais divergências encontradas quando se compara o que a teoria preconiza como o escopo e a abordagem dos Project Management Offices - PMOs e o que acontece na prática, no que diz respeito à real atuação destas estruturas. O tema pode ser considerado relevante devido ao fato de fornecer uma melhor compreensão sobre como as estruturas de PMO realmente atuam, principalmente no que diz respeito ao escopo, isto é, se o PMO é um Programa-Projeto, um PMO Departamental ou um PMO Corporativo e relativamente a sua amplitude, ou seja, se sua atuação é operacional, tática ou estratégica. Para este estudo foi utilizada a metodologia qualitativa, por meio da abordagem de estudo de caso. Foi selecionada uma empresa de grande porte do setor de manufatura não estruturada por projeto para avaliação. A empresa do estudo de caso possui uma estrutura organizacional complexa que possibilita um estudo abrangente. A atuação dos PMOs do estudo de caso foi discutida à luz da revisão de literatura e levando-se em conta, também, as informações colhidas a partir da observação participante. Os resultados deste trabalho permitem concluir que o pressuposto defendido, que é a existência de divergências entre o que a teoria preconiza como escopo e abordagem do PMO e a real atuação dos PMOs, foi confirmado através do estudo de caso; além disso, não existe relação de causalidade clara entre o escopo de atuação dos PMOs e sua posição hierárquica dentro da empresa; e não existe uma atuação padrão destas estruturas, quando comparadas às funções que elas exercem. Por fim, são apresentadas recomendações relativas ao estudo de caso realizado, ou seja, é importante realizar um estudo multicasos entre ao menos uma empresa não estruturada por projeto e outra estruturada por projeto e comparar a atuação dos seus PMOs para melhor compreensão do tema, incluir mais stakeholders no grupo de entrevistados dentro do estudo de caso, realizar um survey com várias empresas de vários segmentos dentro do território nacional e um possível estudo onde seja verificado se tipologias diferentes de estruturas organizacionais podem afetar a atuação práticas dos PMOs.

The main target of this work is to project and make a simple repeater working in 446 MHz tape called PMR. This repeater is made from functional station PMR and repeating program unit, which was constructed by the help of the system for recording and replaying sound. The repeater is managed by microcontroller Atmel from line AVR. The repeater records minimal and maximal temperature during its operation and its date and time is still actual thanks to transceiver DCF. It is possible to connect the transceiver with a computer by the serial link.

Thermo-oxidative aging produces a non-uniform degradation state in PMR-15 resin. A surface layer, usually attributed to oxidative degradation, forms. This surface layer has different properties from the inner material. A set of material tests was designed to separate the properties of the oxidized surface layer from the properties of interior material. Test specimens were aged at 316°C in either air or nitrogen, for durations of up to 800 hours. The thickness of the oxidized surface layer in air aged specimens, and the shrinkage and coefficient of thermal expansion (CTE) of nitrogen aged specimens were measured directly. The nitrogen-aged specimens were assumed to have the same properties as the interior material in the air-aged specimens. Four-point-bend tests were performed to determine modulus of both the oxidized surface layer and the interior material. Bimaterial strip specimens consisting of oxidized surface material and unoxidized interior material were constructed and used to determine surface layer shrinkage and CTE. Results confirm that the surface layer and core materials have substantially different properties.

This study evaluates the Periodic Mesopores Organosilicas (PMO) synthesized by co-condensation of 1,4-bis(trietoxysilil)benzene (BTEB) and feniltrietoxysilane (FTE) in different mole fractions, under acidic conditions using triblock copolymer Pluronic P123 as template and in the presence of inorganic salt (KCl), to adsorption of aromatic compounds in aqueous synthetic solution. The molar composition of the synthesis followed the synthesis equation: xFTE :29,58-xBTEB: 1.0 P123: 226.75 KCl: 38HCl: 12583H2O, where x ranged from 0 to 11.83, as designating the solids obtained as PMO1,0; PMO0,8; and PMO0,6, whose index indicates the fraction of BTEB in the composition of the mixture. Structurally, it was observed that incorporation of FTE to process generates PMOs with reduced thermal stability, with lower values of surface area and pore volume and lower hydrophilic character. The measures for small angle of X-ray diffraction, the transmission microscopy and the isotherm of nitrogen adsorption and desorption showed that the products are hexagonally ordered cylindrical pores and has dimensions of meso/macro complementary. The tests for removal of aromatic compounds dissolved in water, the PMO1,0 and PMO0,6 exhibited efficient adsorption capacity of benzene, toluene, p-xylene and o-Xylene (BTXs). In the adsorption experiments with PMO0,6, in batch study, the capacity of removing benzene (0,84 mg.g-1), toluene (0,93 mg.g-1), p-xilene (1,00 mg.g-1) e o-xilene (0,86 mg.g-1) were lower than those obtained in the tests adsorption column: benzene (1,15 mg.g-1), toluene (1,26 mg.g-1), p-xilene (1,25 mg.g-1) e o-xilene (1,15 mg.g-1). The kinetic mechanism of adsorption is primarily identified as pseud-second order and the isotherms can be represented by Redlich-Peterson and Temkin models.

Estruturalmente, observou-se que a incorporação do FTE ao processo gera PMOs com estabilidade térmica reduzida, com menores valores de área superficial e de volumes de poros além de menor caráter hidrofílico. As medidas de difração de raios X a baixo ângulo, a microscopia eletrônica de transmissão e a isoterma de adsorção e dessorção de nitrogênio revelaram que os produtos estão hexagonalmente ordenado, possuem poros cilíndricos e de dimensões meso/macro complementares. Os testes para remoção dos compostos aromáticos dissolvidos em água, o PMO1,0 e PMO0,6 exibiram eficientes capacidades de adsorção para benzeno, tolueno, p-xileno e o-xileno (BTXs). Nos ensaios de adsorção com PMO0,6, em batelada, as capacidades de remoção do benzeno (0,84 mg.g-1), tolueno (0,93 mg.g-1), p-xileno (1,00 mg.g-1) e o-xileno (0,86 mg.g-1) foram menores que as obtidas nos testes de adsorção em coluna: benzeno (1,15 mg.g-1), tolueno (1,26 mg.g-1), p-xileno (1,25 mg.g-1) e o-xileno (1,15 mg.g-1). O mecanismo cinético de adsorção é, prioritariamente identificado como de pseud-segunda ordem e as isotermas podem ser representadas pelos modelos de Redlich-Peterson e Temkin.

As medidas de difraÃÃo de raios X a baixo Ãngulo, a microscopia eletrÃnica de transmissÃo e a isoterma de adsorÃÃo e dessorÃÃo de nitrogÃnio revelaram que os produtos estÃo hexagonalmente ordenado, possuem poros cilÃndricos e de dimensÃes meso/macro complementares. Os testes para remoÃÃo dos compostos aromÃticos dissolvidos em Ãgua, o PMO1,0 e PMO0,6 exibiram eficientes capacidades de adsorÃÃo para benzeno, tolueno, p-xileno e o-xileno (BTXs). Nos ensaios de adsorÃÃo com PMO0,6, em batelada, as capacidades de remoÃÃo do benzeno (0,84 mg.g-1), tolueno (0,93 mg.g-1), p-xileno (1,00 mg.g-1) e o-xileno (0,86 mg.g-1) foram menores que as obtidas nos testes de adsorÃÃo em coluna: benzeno (1,15 mg.g-1), tolueno (1,26 mg.g-1), p-xileno (1,25 mg.g-1) e o-xileno (1,15 mg.g-1). O mecanismo cinÃtico de adsorÃÃo Ã, prioritariamente identificado como de pseud-segunda ordem e as isotermas podem ser representadas pelos modelos de Redlich-Peterson e Temkin.
This study evaluates the Periodic Mesopores Organosilicas (PMO) synthesized by co-condensation of 1,4-bis(trietoxysilil)benzene (BTEB) and feniltrietoxysilane (FTE) in different mole fractions, under acidic conditions using triblock copolymer Pluronic P123 as template and in the presence of inorganic salt (KCl), to adsorption of aromatic compounds in aqueous synthetic solution. The molar composition of the synthesis followed the synthesis equation: xFTE :29,58-xBTEB: 1.0 P123: 226.75 KCl: 38HCl: 12583H2O, where x ranged from 0 to 11.83, as designating the solids obtained as PMO1,0; PMO0,8; and PMO0,6, whose index indicates the fraction of BTEB in the composition of the mixture. Structurally, it was observed that incorporation of FTE to process generates PMOs with reduced thermal stability, with lower values of surface area and pore volume and lower hydrophilic character. The measures for small angle of X-ray diffraction, the transmission microscopy and the isotherm of nitrogen adsorption and desorption showed that the products are hexagonally ordered cylindrical pores and has dimensions of meso/macro complementary. The tests for removal of aromatic compounds dissolved in water, the PMO1,0 and PMO0,6 exhibited efficient adsorption capacity of benzene, toluene, p-xylene and o-Xylene (BTXs). In the adsorption experiments with PMO0,6, in batch study, the capacity of removing benzene (0,84 mg.g-1), toluene (0,93 mg.g-1), p-xilene (1,00 mg.g-1) e o-xilene (0,86 mg.g-1) were lower than those obtained in the tests adsorption column: benzene (1,15 mg.g-1), toluene (1,26 mg.g-1), p-xilene (1,25 mg.g-1) e o-xilene (1,15 mg.g-1). The kinetic mechanism of adsorption is primarily identified as pseud-second order and the isotherms can be represented by Redlich-Peterson and Temkin models.

El Desarrollo Sostenible es un concepto que exige acciones concretas que transformen sus principios e interpretaciones, en acciones prácticas con las cuales contribuir de modo mensurable para un futuro distinto. La intervención al nivel urbano es, reconocidamente, uno de los mejores enfoques en la aproximación de la sociedad presente al paradigma del desarrollo sostenible. En este contexto territorial, los ciudadanos pueden contribuir de modo efectivo para la realidad de global, como elementos individuales o colectivos. El modo como se actúa a nivel urbano depende en gran medida del modo como las Autoridades Locales desarrollan sus actividades, a estas les compete actuar de modo decisivo y convincente en la introducción de políticas de sostenibilidad, definiendo políticas, planos y programas. En Portugal, los principales instrumentos de actuación a nivel local son los Planes Municipales de Ordenación Territorial, parte integrante del Sistema de Gestión Territorial. Estos planes asumen permiten una intervención normativa, en especial en la ordenación territorial, a distintos escalas de intervención. Os microrganismos são responsáveis por diversos processos biológicos essenciais ao ambiente, sendo estes intimamente relacionados com as taxas de decomposição da matéria orgânica e com a persistência da fertilidade nos solos. Apesar da importância e grande diversidade, a identificação de táxons envolvidos em processos específicos está geralmente restrita a uma pequena fração da microbiota que pode ser isolada e cultivada. Sendo assim, pouco se sabe sobre os microrganismos que atuam no ciclo do carbono no solo, como, aqueles que participam da oxidação do metano (CH4), por exemplo. Estes, chamados metanotróficos exercem papel importante no controle da emissão desse gás de efeito estufa para a atmosfera podendo servir como um filtro de metano e mitigar suas emissões. A Terra Preta Antropogênica (TPA) é um importante ecossistema na região amazônica e contém fragmentos cerâmicos e frações orgânicas, como o carvão (biocarvão), que foram incorporados em períodos pré-colombianos. Isso resultou em solos sustentáveis com elevada fertilidade, apresentando cerca de três vezes mais matéria orgânica, setenta vezes mais biocarvão e diversidade microbiana maior quando comparados com os solos adjacentes. Com o presente trabalho, objetivou-se avaliar o efeito do enriquecimento atmosférico com metano sobre a abundância da comunidade bacteriana total e de metanotróficas nos solos de Terra Preta da Amazônia sob floresta e cultivo (TPA Floresta e TPA Cultivada) e seus respectivos solos adjacentes (ADJ Floresta e ADJ Cultivado), coletados Estação Experimental do Caldeirão (Iranduba, AM). Para tanto, foi realizado um experimento de microcosmo no qual os solos foram incubados com atmosfera contendo 10% de metano e meio de cultura NMS (do inglês, Nitrate mineral salts), utilizado para crescimento de metanotróficas, a fim de avaliar a resposta dessas comunidades ao longo de 21 dias. A variação da concentração de metano na atmosfera dos frascos foi monitorada através de cromatografia gasosa e o DNA do solo recuperado nos tempos de coleta durante o experimento foi extraído para utilização na técnica de PCR quantitativo (qPCR), a qual possibilitou a quantificar o número de cópias dos genes 16S rRNA Bacteria e pmoA nas amostras. O solo de Terra Preta da Amazônia se mostrou um potencial dreno de CH4 atmosférico Comparando as respostas dos solos com floresta (TPA Floresta e ADJ Floresta) e cultivados (TPA Cultivada e ADJ Cultivado), notou-se uma menor variação da abundância da comunidade metanotrófica presente nestes últimos, o que indica que alteração do uso do solo afeta a capacidade do mesmo em retirar metano da atmosfera. Os solos Adjacentes apresentaram resposta diferente dos solos de TPA, indicando que a história de formação, ocupação e uso do solo também influenciam na capacidade do solo em drenar o metano da atmosfera.
Microorganisms are responsible for several biological processes essential to the environment, which are closely related to the rates of decomposition of organic matter and with the persistence of fertility in soils. Despite the importance and high diversity, identification of taxa involved in specific processes is usually restricted to a small fraction of the microbiota that can be isolated and cultivated. Thus, little is known about the microorganisms that act on the carbon cycle in the soil, such as those participating in the oxidation of methane (CH4), for example. These, known as methanotrophs play an important role in controlling the emission of greenhouse gas into the atmosphere and may serve as a methane filter and mitigate their emissions. Amazonian Dark Earth (ADE) is an important ecosystem in the Amazonian region and contains ceramic fragments and organic amendments, such as charcoal (biochar), which were incorporated in Pre-Columbian periods. This resulted in sustainable soils with high fertility, presenting about three times more organic matter, seventy times more biochar and higher microbial diversity when compared to adjacent soils. The present work aimed to evaluate the effect of atmospheric methane enrichment on the abundance of the bacterial and methanotrophic community in ADE soils under forest and cultivation (ADE Forest and ADE Cultivated) and their respective adjacent soils (ADJ Forest and ADJ Cultivated), sampled at Caldeirão Experimental Station (Iranduba, AM). For this purpose, a microcosm experiment was performed in which the soils were incubated under an atmosphere containing 10% of methane and NMS (Nitrate mineral salts) culture medium used for methanotrophic growth in order to evaluate the response of these communities over 21 days. The variation of methane concentrations in the atmosphere of the vials was measured by gas chromatography and the soil DNA recovered in the collection time during the experiment was extracted for use in the technique of quantitative PCR (qPCR), which made it possible to quantify the number of copies of 16S rRNA Bacteria and pmoA on samples. The Amazonian Dark Earth soil showed a potential sink for atmospheric CH4. Comparing atmospheric responses of forest soils (ADE Forest and ADJ Forest) and cultivated soils (ADE Cultivated and ADJ Cultivated), noted a minor variation in the abundance of methatroph community in these last, indicating that land use change affects the ability of it to sink the methane atmosphere. Adjacent soils had different responses of ADE soils, indicating that the history formation, occupation and land use also influence the capacity of the soil to drain methane from the atmosphere.

Projetos são precursores de novos produtos, serviços e processos organizacionais. Sendo assim, a gestão de projetos aparece como uma grande oportunidade para organizações nos diversos ramos de atividade. O aumento da complexidade do mundo dos negócios e a crescente competitividade fazem com que as empresas tenham que responder cada vez mais rápida e eficientemente aos estímulos externos. Neste contexto, as falhas na execução dos projetos muitas vezes têm resultados desastrosos. Esta dissertação tem por objetivo identificar e analisar os fatores críticos de sucesso para a operação de Escritórios de Gerenciamento de Projetos (Project Management Office – PMO) nas organizações. O PMO permite às empresas usufruir dos benefícios da integração de processos, assim como informar a alta administração sobre o andamento do seu portfólio de projetos, buscando competitividade no mercado com o sucesso de seus projetos. A pesquisa qualitativa (espontânea) teve participação de especialistas brasileiros e estrangeiros, reconhecidos pelo PMI como pessoas destacadas (consultores especializados) em gerenciamento de projetos. Na pesquisa Delphi, quarenta e três especialistas de vários países foram submetidos a um questionário de vinte e cinco perguntas minuciosamente preparadas, a fim de extrair o máximo possível de informações consideradas críticas nas atividades atuais e tendências de PMOs nas empresas. A pesquisa Delphi foi realizada em duas rodadas e concluída com recomendações efetivas, validadas internacionalmente. Como resultado do trabalho, foi desenvolvido um estudo que poderá ser utilizado como referência para a operação de Escritórios de Gerenciamento de Projetos (PMO) nas empresas sem distinção de segmento.
Projects are precursors of new products, services and organizational processes. Therefore, the project management appears as a great opportunity for organizations in different industries. The increasing complexity of business and increased competition mean that companies have to answer even more quickly and efficiently to external stimuli. In this context, the flaws at project implementation often have bad results. This dissertation aims to identify and analyze the critical success factors for the operation of Project Management Office (PMO) organizations. The PMO allows businesses to enjoy the benefits not only at the integration process but also to inform senior management about the progress of your project portfolio, seeking competitiveness in the market with the success of their projects. Qualitative research (spontaneous) had Brazilian and foreign specialists, recognized by PMI as prominent people (consultants) in project management. In the Delphi survey, forty-three experts from several countries were submitted to a questionnaire of twenty-five questions thoroughly prepared in order to extract the maximum information considered critical for current activities and trends from PMOs at their companies. The Delphi survey was conducted in two rounds and concluded with effective recommendations, internationally validated. As a result of the work, it had developed a study that could be used as a reference for operation of the Project Management Office (PMO) at companies regardless of segment.

PMOs are complex organisational-specific entities. Companies are increasingly establishing PMOs in search of more favourable outcomes for their projects. However, these PMO organisational structures are rarely found to survive and are often disbanded or transformed every few years. The PMOs implemented in the public sector are faced with the task of justifying their value to business over time. These PMOs function in particularly stressful environments as they run complex projects that are driven by politics. The public sector provides a unique context in that organisational transformation is likely to occur every few years with the election of a new political cabinet. The notion of a PMO is a relatively new phenomenon with limited research available at present. There are few practical and theoretical guidelines identified in the research literature. The public sector environment has received even lesser research despite offering a unique organisational structure that commonly undertakes large, complex and political-focussed projects. The purpose of this study is to seek a better understanding of the contribution made to public sector PMO value in terms of efficiency and effectiveness. An understanding of the efficiency and effectiveness factors involved can aid managers in identifying where to focus their efforts to ensure value-add of new services and products are achieved in the public sector. A mixed method research approach was employed to investigate the public sector PMO organisational environment case study, using interviews and a complementary survey. A model of the factors that contribute to the efficiency and effectiveness of a PMO was derived from the research literature. This was followed by an in-depth case study of a public sector PMO. The factors relevant to the case study were captured using interviews and then analysed using thematic analysis. The results were then compared to the model. The study revealed that most of the public sectors PMOs (77%) was found to be structured as virtual PMO offices with varying levels of capacity and authority. The results of the survey found that most of the PMOs focus in terms of efficiency was on productivity (60%), planning (20%), and profit (20%). In terms of effectiveness, the foci were mainly on responsiveness (40%) and flexibility, adaption and innovation (60%). It was concluded that project management maturity could be significantly improved by focussing on the areas that were lacking for these PMOs, i.e. benchmarking, hiring external consultants with PM knowledge and linking with external consultants.

Abstract:
Ce travail est consacre a l'etude des degradations du transistor pmos soumis aux injections de porteurs hautement energetiques generes par l'accroissement des champs electriques lies a la miniaturisation de sa geometrie. Ces mecanismes de degradations se manifestent par les derives temporelles des parametres electriques representatifs des performances du transistor, en raison de piegeage de charges dans l'oxyde et de la generation d'etats electroniques a l'interface oxyde-silicium. Cette etude presente tout d'abord un bref rappel des parametres de fonctionnement du transistor ainsi qu'un modele simple de la generation des porteurs chauds par le processus d'ionisation par impact. Nous presentons l'analyse des degradations du transistor pmos a l'aide de methodes complementaires de caracterisations electriques en courant-tension puis par une technique de pompage de charges qui nous permettront de dissocier les types de defauts induits. A l'aide de ces deux premieres techniques nous developpons des methodes de prediction de duree de vie qui tiennent compte des aspects specifiques du vieillissement du transistor pmos. Enfin, ces techniques de mesures couplees a une technique dite de la grille flottante et a des simulations bi-dimensionnelles nous permettront d'elaborer une etude approfondie des defauts induits par des charges fixes d'oxyde jusque dans la region de recouvrement grille-drain de la structure du transistor pmos

Abstract:
O objetivo desse estudo foi o de investigar os efeitos do treinamento físico resistido e/ou da suplementação com glutamina sobre as vias de sinalização que regulam a síntese e a degradação de proteínas no músculo esquelético de ratos. Os animais foram divididos nos seguintes grupos: 1) controle (C), 2) treinado (T), 3) suplementado com glutamina (G) e 4) treinado e suplementado com glutamina (TG). Não houve alteração na massa do músculo EDL, mas observou-se aumento significativo na área de secção transversa da fibra (AST). Houve aumento no conteúdo das proteínas Raptor e Deptor e TSC1. O grau de fosforilação da Akt, 4EBP-1, p70SK e S6 foram aumentados nos grupos suplementados com glutamina. A atividade do proteassoma 26S diminuiu no grupo exercitado em relação ao controle. Os resultados deste estudo são sugestivos de que o protocolo de exercício físico utilizado causou hipertrofia por aumento da fosforilação de 4EBP-1. Na suplementação com glutamina, foi observado aumento da fosforilação da 4EBP-1 e de S6 elevando a AST no músculo EDL.
The effect of physical exercise training and/or glutamine supplementation on the signaling pathways that regulate synthesis and degradation of proteins in rat skeletal muscles was investigated. The following groups were examined: 1) control (C); 2) physically trained (T); 3) supplemented with glutamine (G); and 4) physically trained and supplemented with glutamine (GT). EDL muscle mass was not changed but a significant increase in cross-sectional area of the fibers was observed. There was an increase in the protein content of the Raptor and Deptor and TSC1. The levels of phosphorylated S6 and 4EBP-1 were significantly increased in the groups supplemented with glutamine. 26S proteasome activity was decreased in the exercised as compared to the control group. The physical exercise protocol used herein caused hypertrophy via phosphorylation of 4EBP-1. Glutamine supplementation led to an increase of the phosphorylation state of 4EBP-1 and S6 raising the AST of the EDL muscle.

Abstract:
L’industrie microélectronique arrive à concevoir des transistors atteignant dimensions de l’ordre de la dizaine de nanomètres. Et ce faisant elle tend atteindre ses limites en terme de réduction des dimensions des transistors CMOS. Or à ces dimensions, la fiabilité et la variabilité des dispositifs prennent une ampleur critique en ce qui concerne les prédictions de durée de vie et de garantie des composants. Parmi les aspects critiques, la dégradation NBTI (Négative Bias Temperature Instability) représente l’un des plus gros défis en termes de fiabilité. Cette dégradation tire son origine d’un piégeage de charge dans l’oxyde de grille et est responsable pour une grande partie de la dégradation des transistors. A l’aide d’un important travail expérimental, nous avons caractérisé à l’aide de mesure rapide les cinétiques de dégradation et de relaxation de la dégradation NBTI, puis nous avons travaillé sur la modélisation des phases de stress et de relaxation. Nous sommes parvenues à créer un modèle pour le stress et la relaxation que nous avons éprouvé sur un certain nombre de nœuds technologiques allant du 14nm FDSOI au 180nm Bulk. Nous avons aussi évalué l’impact de certains changements de procédées de fabrication sur la dégradation NBTI.Enfin nous proposons une étude poussée de la variabilité induite par le NBTI et du modèle DCM (Defect centric Model) permettant de modéliser cette variabilité. Nous proposons alors une correction mathématique de ce modèle, et la possibilité de le réécrire afin de pouvoir l’utiliser pour un plus grand nombre de défauts. Enfin nous mettrons ce modèle en échec sur les prédictions qu’il fait de défauts et nous proposons un nouveau modèle sous la forme d’un DCM à deux défauts ou DDCM (Dual Defect Centric Model).Mots-clés : Microélectronique, FDSOI, Bulk, variabilité, NBTI, caractérisation électrique, modélisation
The microelectronics industry is able to design transistors reaching dimensions of the order of ten nanometers. And doing this, we reaching the limits in terms of size reduction of CMOS transistors. At these dimensions, the reliability and variability of the devices is critical in terms of lifetime prediction and component warranty. Among the critical aspects, NBTI (Negative Bias Temperature Instability) degradation represents one of the biggest challenges in terms of reliability. This degradation coming from a charge trapping in the gate oxide is responsible for a large part of the degradation of the transistors. Performing a huge experimental work based on the characterization of the kinetic of degradation and relaxation of the NBTI degradation with rapid measurements, allowing us to work on the modeling of the stress and relaxation phases of NBTI degradation. We have successfully create a model for stress and relaxation of the NBTI degradation. These models were then tested on several technological nodes from 14nm FDSOI to 180nm Bulk. We also study the impact of some process changes on NBTI degradation. Finally, we propose a detailed study of the variability induced by the NBTI and the DCM model (Defect centric Model) allowing to model this variability. We also propose a mathematical correction of this model but also another mathematical expression of this model allowing to use it for a large number of defects. Enfin, nous prouvons que DCM est défectueux dans sa prédiction du nombre de défauts et nous proposons un nouveau modèle sous la forme d'un DCM avec deux défauts ou DDCM (Dual Defect Centric Model)

O objeto do estudo foi elaborar uma modelo estrutural completo dos fatores que influenciam o sucesso da atuação do PMO e que benefícios essa atuação positiva traria para a gestão de projetos no Sistema Eletrobras e a partir disso, realizar um aperfeiçoamento do modelo estrutural por meio de métodos estatísticos. Para isso foi realizado uma pesquisa de campo (survey) no intuito de levantar dados junto aos profissionais envolvidos em Gestão de Projetos dentro do Sistema Eletrobras, a partir do modelo proposto e foram evidenciados os fatores de sucesso na atuação do PMO (Project Management Office) que possibilitam o escritório de gerenciamento de projetos tornar-se um vetor de gestão por excelência na empresa. Para alcançar o objetivo idealizado, realizou-se (a) Levantamento bibliográfico para conceituar gerenciamento de projetos e PMOs, (b) identificou-se os fatores que influenciam o sucesso de atuação do PMO, (c) identificou-se as abordagens teóricas dos fatores diretos que mais contribuem para que o PMO da Eletrobras possa atuar como um vetor de gestão por excelência, otimizando o aumento da eficiência da sua atuação e o sucesso na gestão de projetos. Averiguou-se quais abordagens teóricas, a partir do sucesso de atuação do PMO, traziam benefícios para a gestão de Projetos no Sistema Eletrobras. Assim, para realizar os trabalhos estatísticos para a avaliação de adequação do modelo, foi utilizado o software SPSS Statistics, para a execução da AFE (Análise Fatorial Exploratória), em busca de testar o modelo baseado na teoria previamente apresentada. Foram obtidas 116 respostas válidas preenchidas por colaboradores com envolvimento em gerenciamento de projetos no sistema Eletrobras. No início o modelo teórico era composto por vinte abordagens teóricas e três fatores independentes. Após as etapas do SEM, obteve-se dezessete abordagens e dois fatores independentes, diferentes do modelo proposto. Com isso, a hipóteses H1 e H2 não foram rejeitadas, pois apresentaram fator significante com (P < 0,05). A hipótese H3 teve que ser excluída, pois na AFE suas abordagens teóricas compuseram um único conjunto com a hipótese H2, visto que a Matriz da Variância Total Explicada demonstrou ser o modelo mais ajustado com somente dois fatores. Todas as análises foram validadas pelo alpha de cronbach.
The object of study was preparing a full structural model of the factors that influence the success of the PMO acting and that this positive performance would bring benefits for project management in the Eletrobras System and from this, carry out an improvement of the structural model through methods Statistical. For it was conducted a field survey (survey) in order to collect data with professionals involved in Project Management within the Eletrobras System, from the proposed model and were shown the success factors in the performance of the PMO (Project Management Office) which enable the project management office become a vector of management excellence by the company. To achieve the idealized goal, there was (a) Bibliographic survey to conceptualize project management and PMOs, (b) identified the factors that influence the PMO acting success, (c) identify the theoretical approaches of the factors direct that contribute most to the PMO Eletrobras can act as a vector management par excellence, optimizing the increased efficiency of its operations and success in project management. It was found that theoretical approaches, from the PMO acting success, bringing benefits to the Project management in the Eletrobras System. Thus, to perform statistical work for the evaluation of the adequacy of the model, SPSS Statistics software was used, for the implementation of EFA (Exploratory Factor Analysis), seeking to test the model based on the theory presented previously. 116 valid responses were obtained filled by employees with involvement in project management in the Eletrobras System. Earlier the theoretical model was composed of twenty theoretical approaches and three independent factors. After the stages of SEM, seventeen approaches was obtained and two independent factors, other than this model. With this, the H1 and H2 hypotheses were not rejected because they presented with significant factor (P <0.05). The hypothesis H3 had to be excluded, as their theoretical approaches the AFE comprised a single set with the hypothesis H2, whereas Matrix Total Variance Explained proved to be the most adjusted model with only two factors. All analyzes were validated by Cronbach's alpha.

Este trabajo de investigación, presenta una propuesta de Caso de Negocio para el proyecto de implementación de una nueva versión de la actual PMO de la empresa AtekPC, basados en la información disponible en el caso de Harvard 308-S52 (El Departamento de Gestión de Proyectos de AtekPC). En este caso de negocio, se realiza un análisis tanto de los costos totales del ciclo de vida del producto, como los beneficios esperados que generará la PMO en su primera fase la de una PMO Básica (Según la tipología de Gerard Hill). El alcance de la PMO Básica será de dar soporte a los proyectos de Operaciones, ya que es el área que más efecto tiene sobre la productividad. La PMO dará soporte también, al área de TI, sin embargo para esta primera fase no se incluirá en su metodología a los proyectos ágiles. La madurez que vaya adquiriendo la PMO respecto a su propósito, responsabilidades y autoridad, redundará en mayores beneficios para la organización, optimizando el desempeño de los proyectos y mejorando la integración interfuncional entre las áreas de la organización, que permitirá afrontar con mayor eficiencia los nuevos y complejos proyectos de acuerdo al planeamiento estratégico de la empresa.
This research paper presents a Business Case proposal for the implementation project of a new version of the current PMO of the company AtekPC, based on the information available in the case of Harvard 308-S52 (The Project Management Department of AtekPC). In this business case, an analysis is made for the total costs of the life cycle of the product and the expected benefits that the PMO will generate in its first phase of Basic PMO. The scope of the Basic PMO will be to give support to the Operations projects, since it is the area that has more effect on productivity. The PMO will also give support to the IT area, however for this first phase, agile projects will not be included in its methodology. The maturity that the PMO acquires with respect to its purpose, responsibilities and authority, will result in greater benefits for the organization, optimizing the performance of the projects and improving the inter-functional integration between the areas of the organization, which will allow to face with greater efficiency the new and complex projects according to the strategic planning of the company.

Le réseau de radio communications mobiles professionnelles (PMR), qui est utilisé pour le fonctionnement de la sécurité publique, doit évoluer pour les solutions à large bande pour satisfaire les demandes des utilisateurs à l’avenir. Dans les technologies à large bande actuels, Long Term Evolution (LTE), développé par le 3GPP (3rd Generation Partnership Project), est considéré comme l’un des candidats potentiels pour la prochaine génération de PMR. Malgré le fait que la technologie LTE en charge la transmission de données à haute vitesse et prend en charge différentes tailles de paquets en utilisant la modulation et le codage adaptatifs (AMC), le LTE n’est pas encore optimisée pour la communication vocale à bas débit, en particulier en cas d’utilisation LTE pour la radio communications mobiles professionnelles (PMR ) contexte. Par conséquent, dans cette thèse, nous présentons des solutions pour renforcer la capacité de la voix de la technologie LTE dans le cadre PMR à la fois la liaison montante et la transmission de liaison descendante. Les nouvelles propositions, basées sur la technologie de norme LTE existant avec des adaptations, permettent la réduction de les frais généraux de données et les frais généraux de contrôle sur LTE (VoLTE) dans le contexte PMR
The Professional Mobile Radio (PMR) network, which is used for public safety operation, has to evolve to the broadband solutions to satisfy the user demands in the future. In the current broadband technologies, Long Term Evolution (LTE) standard, developed by the 3GPP (3rd Generation Partnership Project), is considered one of the potential candidates for the next generation of PMR. Despite the fact that LTE supports high-speed data transmission and supports different packet sizes by using Adaptive Modulation and Coding (AMC), LTE is not yet optimized for low bit rate voice communication, especially in case of using LTE in Professional Mobile Radio (PMR) context. Therefore, in this dissertation, we present several new solutions for enhancing the voice capacity of LTE in the PMR context for both uplink and downlink transmission. The new propositions, based on existent LTE standard technology with adaptations, allow reducing both control and data overhead issues of Voice over LTE (VoLTE) in PMR context

The role of neurofibromin, the product of the Neurofibromatosis Type 1 (NF1) gene, in cell physiology is still not well understood. Considering the different traits associated to NF1 it is clear that neurofibromin participates in processes of proliferation and differentiation of different cell types. The Ras-GAP activity of neurofibromin is its best characterized biochemical function, which is regulated by the alternative splicing of exon 23a (E23a). In this thesis we intended to better understand the role of alternative splicing of E23a during neuronal differentiation, with the aim that in the future it could provide information on the learning and cognitive issues related to this disease. Due to the large size of neurofibromin, the difficulty of manipulating it in vitro and the necessity to mimic, as much as possible, the physiological conditions of the cell, we used Phosphorodiamidate Morpholino Oligomers (PMOs) to modify the exonic composition of NF1 mRNA while preserving the endogenous neurofibromin expression levels. We developed a PMO-based system that successfully allowed to force the expression of type II (+E23a) or type I (-E23a) isoforms without altering the physiological expression levels of NF1 mRNA in PC12 cells, a neuronal differentiation model, in the presence or absence of Nerve Growth Factor (NGF). This PMO system could be used in other cellular models of NF1, and could be reproduced for studying the regulation of the alternative splicing in other genes. Furthermore, we set up a group of functional assays for assessing proliferation, differentiation, signaling and apoptosis in this PC12 neuronal model. Our results showed that forcing type I (-E23a) isoform was not sufficient for inducing PC12 neuronal differentiation in the absence of NGF. However, the results also demonstrate that any alteration of the NGF-induced ratio between type I/II isoforms, either in a quantitative or time-dependent manner, interfered with the correct neuronal differentiation process, in particular, altering the correct formation of neurites, as well as the proper regulation of the RAS/MAPK and cAMP/PKA signalling pathways. Moreover, the alteration of the natural E23a alternative splicing also impeded the proper neuronal differentiation process in other neuronal models, such as the H19-7 hippocampus cells. Our results also showed that the depletion of neurofibromin in PC12 induce a generalized process of apoptosis; suggest the existence of an early negative feed-back on the function of neurofibromin when type I isoform is abnormally expressed, and shows that the regulation of the cAMP/PKA pathway is also dependent on the GAP domain of neurofibromin. All together, the results of this thesis indicate that the regulation of the alternative splicing of exon 23a of the NF1 gene allows the fine-tuning of the RAS/MAPK and cAMP/PKA pathways through its GAP activity in a coordinate and opposite way along the time-dependent process of neuronal differentiation.
La neurofibromina és el producte del gen NF1, que mutat causa la Neurofibromatosis de tipus 1. Tot i que en l'actualitat, encara ens cal entendre millor el rol d'aquesta proteïna en la fisiologia cel•lular, l'activitat Ras-GAP és la funció bioquímica més ben caracteritzada de la neurofibromina. Aquesta funció està regulada per l'splicing alternatiu de l'exó 23a (E23a). En aquesta tesi ens vàrem proposar comprendre millor el paper d'aquest splicing alternatiu durant el procés de diferenciació neuronal, amb l'objectiu de proporcionar nova informació sobre els problemes cognitius i d'aprenentatge associats a aquesta malaltia. Degut a la gran grandària de la neurofibromina i a la dificultat de manipular-la in vitxo, es varen utilitzar Phosphorodiamidate Morpholino Oligomers (PMOs) per modificar la composició exònica del mARN (per tant l'estructura resultant de la neurofibromina) sense alterar les condicions fisiològiques d'expressió del gen NF 1 . Es va desenvolupar un sistema basat en PMOs per forçar l'expressió de l'isoforma tipus II (+E23a) o tipus I (-E23a) del gen NF1 en cèl•lules PC12, un model de diferenciació neuronal, en presència o absència de Nerve Growth Factor (NGF). A més, per entendre la importància de 1'E23a es va establir un grup de metodologies i assajos funcionals per poder determinar diferents respostes cel•lulars i valorar la funció d'aquest en el procés de diferenciació neuronal. Els nostres resultats van mostrar que forçar l'isoforma tipus I (-E23a) no era suficient per induir la diferenciació de les cèl•lules PC12 en absència de NGF. No obstant això, qualsevol alteració en la relació entre les isoformes tipus I/II en presència de NGF, ja sigui d'una manera quantitativa o dependent del temps, interferia en el correcte procés de diferenciació neuronal, en particular, alterant la correcta formació de neurites, així com l'adequada regulació de les vies de senyalització RAS/MAPK i cAMP/PKA. En conjunt, els resultats d'aquesta tesi indiquen que la regulació de l'splicing alternatiu de 1'E23a del gen NF1 permet un ajust fi de les vies RAS/MAPK i cAMP/PKA a través de la activitat GAP de la neurofibromina, d'una forma oposada i coordinada al llarg del temps durant el procés de diferenciació neuronal.

En los tiempos actuales el proyecto es una herramienta válida para lograr los objetivos planteados en los planes estratégicos de la organización y lograr así la sustentabilidad en el tiempo, sin embargo no siempre se logra obtener un buen desempeño desde el punto de vista de plazos, recursos y expectativas de los clientes. Muchas organizaciones buscan mejorar sus prácticas desarrollando metodologías y procesos más rigurosos para abordar de mejor forma los proyectos. Dentro de estas muchas organizaciones que buscan mejorar sus procesos y lograr más eficiencia a través de una forma estructurada de gestión de proyectos se encuentra Methanex, ya que debido a su acelerado ritmo de crecimiento en el nivel de producción, debe ejecutar varios proyectos en forma simultánea, es así que nace la idea de formar la PMO (Project Management Office) de manera tal de homogeneizar procesos, definir procedimientos comunes, determinar diferentes metodologías para abordar las distintas áreas de conocimiento: nueve de acuerdo a la metodología PMI (Project Management Institute), se trata también de aprovechar las economías de escala y capitalizar lecciones aprendidas. Abstract:
La resine polyimide pmr-15 est principalement utilisee comme matrice pour materiaux composites dans le domaine aeronautique et constitue une reference car elle offre d'excellentes proprietes mecaniques a haute temperature. Cependant, l'evolution chimique du melange initial est l'une des causes conduisant a la formation de composites de mauvaise qualite. Un controle de la polymerisation de ce thermostable in-situ et en continu s'avere donc necessaire. La spectroscopie ultrasonore apparait comme l'une des rares techniques potentielles pour ce suivi : en particulier, la methode novatrice utilisant un implant piezoelectrique et developpee est pressentie pour l'application sur site industriel. En raison de la complexite du mecanisme reactionnel de la resine pmr-15, sa caracterisation par spectroscopie ultrasonore classique est abordee par une approche sur composes modeles : les deux systemes choisis representent les deux principales reactions en competition au cours de la polymerisation. Alliee a des analyses d'infrarouge a transformee de fourier, de chromatographie d'exclusion sterique et de viscosimetrie, l'aptitude de la spectroscopie ultrasonore classique a suivre la polymerisation de composes modeles de la resine pmr-15 est demontree. La meme demarche est adoptee pour l'etude de cette derniere et les bonnes correlations obtenues par analyses ftir, sec et viscosimetrie permettent d'attribuer aux evolutions de la vitesse ultrasonore durant la polymerisation, les evenements apparaissant au cours de la transformation de la resine : fusion des monomeres, reaction d'imidation, reticulation thermique. L'etude ultrasonore du preimpregne pmr-15 est ensuite realisee et les etapes essentielles de la polymerisation sont identifiees. Enfin, la technique de l'implant piezoelectrique, pressentie pour l'application sur site industriel, est utilisee lors du suivi de la resine et du preimpregne pmr-15 : pour la premiere fois, cette methode est utilisee lors d'un suivi de polymerisation a temperature variable, et sur une large gamme 20-340c. Les resultats confirment ceux obtenus par spectroscopie ultrasonore classique. La validation de cette technique, facile a mettre en uvres, lui confere un avenir prometteur pour l'application sur site industriel pour laquelle un appareil portable a ete concu.

L'objectif de cette etude est d'analyser les proprietes thermomecaniques du polyimide pmr-15 pour comprendre le comportement mecanique des composites thermostables, fibres de carbone/resine pmr-15. Pour atteindre cet objectif, l'etude demande au prealable la fabrication de plaques de la resine thermodurcie. Etudiee sans succes jusqu'alors, la mise en uvre de la resine polyimide a ete realisee par un procede, brevete, faisant appel aux mecanismes reactionnels des constituants de base et a un plan d'experiences des proprietes mecaniques propre a definir les conditions de durcissement optimales. Apres avoir etabli un bilan global des proprietes du materiau sur un grand intervalle de temperature (-200c, +300c), l'analyse et la modelisation du comportement ont ete effectuees a chaud aux limites de l'etat vitreux du polymere. Sous des conditions de sollicitation variees, les resultats mecaniques montrent les qualites du materiau et mettent en evidence son caractere thermostable.

Abstract:
Duchenne muscular dystrophy (DMD) is a fatal, muscle-wasting disorder due to mutations/deletions in the dystrophin gene. Whilst improvements in palliative care have increased the life expectancy of patients, cardiomyopathy and respiratory complications are still the leading causes of death. A potential therapy for the treatment of DMD is antisense oligonucleotides (AOs), which modulate dystrophin pre-mRNA splicing to restore the dystrophin reading frame and generate a truncated functional protein. Conjugation of AOs to cell penetrating peptides (CPP), such as Pip5e-, significantly improves delivery to skeletal muscles and to the heart, which is imperative given the impact of cardiomyopathy to mortality. However, it should be noted that the contribution of skeletal muscles, such as the core respiratory muscle, the diaphragm, in dystrophic cardiopulmonary function is poorly understood. The specific aims of the work in this thesis were to (i) understand the effect of the diaphragm on cardiac function using magnetic resonance imaging (MRI), (ii) screen a number of derivatives of Pip5e (Pip6) in an effort to discover further promising peptides and define the properties integral to heart penetrating capacity, and (iii) assess whether Pip6-PMOs restore cardiac function (MRI) following a repeat, low dose regimen. In short, the specific restoration of dystrophin in the diaphragm of the dystrophic mouse model, the mdx mouse, did not improve cardiac function, highlighting the importance of a body-wide therapy. The screening of multiple Pip5e-PMO derivatives revealed 3 promising peptides with improved cardiac splicing capacity; however, serial deletions of amino acids from the central core resulted in the diminution of dystrophin restoration, possibly due to a reduction in hydrophobicity. Finally, the Pip6-PMO treatment regimen substantially restored dystrophin protein (28% in heart) and stabilised cardiac function, even with an increased work load. In conclusion, this study illustrates the importance of a body-wide treatment, such as the CPP strategy (Pip-PMO). These Pip-PMO conjugates demonstrate high dystrophin restoration in a number of muscles, including cardiac muscle, and have a beneficial effect on cardiac function.

El presente artículo desarrolla una propuesta para el diseño y despliegue de una Oficina de Dirección de Proyecto o PMO (Project Management Office) como apoyo para mejorar la dirección de proyectos de servicios de tecnología.
The way organizations execute their strategies can be the difference between success and failure in achieving their objectives such that the strategies must be translated into relevant, feasible and measurable projects. Many organizations already work in a project management environment and have project management professionals but organizations need to do more with their already limited resources, focusing on projects that generate value, executing projects more efficiently and better controlled. This article develops a proposal for the design and deployment of a project management office (PMO) as a way to improve project management in technology services.

No âmbito específico da gestão de projetos é um garante que o portfólio de projetos está alinhado com os objetivos da organização, e é entregue de forma eficiente e sustentável suportando os meios pelos quais a gestão e outras importantes partes interessadas, recebem a informação oportuna, relevante e fiável (APM, 2004). No presente trabalho é usada para classificar os modelos postos em prática pelo Project Management Office, no apoio a projetos ou programas através dos processos de partilha de conhecimento (Garland, 2009). Na medida em que estudos anteriores referem consistentemente lacunas na investigação nesta área, o objetivo da presente dissertação será assim de examinar o papel estratégico da governança do Project management office e qual a sua verdadeira influência nos processos de partilha do conhecimento entre projetos. For the Project Management Institute (PMI), "governance is defined as the process of development, communication, implementation and monitoring, ensuring the policies, procedures, organizational structures and practices associated with a particular program." In the organizational context, provides a framework for decision making and ethical management actions based on transparency, responsibility and well-defined roles (Too & Weaver, 2014b). In the specific scope of project management it is a guarantee that the project portfolio is aligned with the organization's goals, and is delivered in an efficient and sustainable manner, supporting the means by which management and other key stakeholders receive timely, relevant and reliable information (APM, 2004). In this study it is used to classify the implemented models of the Project Management Office, to support projects or programs in knowledge sharing processes (Garland, 2009). As previous studies consistently refer research gaps in this area, the purpose of this dissertation will be to examine the governance strategic role of the Project Management Office and what is its real influence on knowledge sharing processes between projects. Both obstacles and motivations will also be observed for the real knowledge transfer, in the perception how project managers understand the implemented policies.

This thesis is about the standardization of the planning phase in a project management office (PMO). The purpose of the research is to find the way to standardize the planning phase in PMO. This purpose is motivated by the relevance of planning phase in projects. Planning is about to forecast, and the consequences of a bad forecasting can be critical for the developing of the project. The research question of the study is the following one:➢ How can the planning phase be standardized in a Project Management Office?To answer this question five interviews have been performed. The interviewees were senior project managers from different companies in Sweden. In order to keep the confidentiality, the names of the managers and the companies which they belong to are omitted. A literature review has been done to set up a theoretical framework and validate the results from the interviews. The study concludes that the only part in planning which can be easily standardized is the work breakdown structure. The results showed that standardization of planning phase as a whole is a big challenge for PMO and also that specialization helps in great measure the standardization of planning.

Cette thèse a été préparée à l'IMT atlantique dans le contexte du projet FITNESS. Ce projet Européen vise à moderniser les radios PMR (Professional Mobile Radio), utilisées par les services de sûretés. L'objectif est de développer un nouveau récepteur compatible avec les 4 normes européennes, tout en ajoutant de nouvelles fonctionnalités. Les contributions de cette thèse à FITNESS concernent la rétrocompatibilité du nouveau récepteur avec les standards actuels. Un filtre de sélection de canal de fréquence centrale reconfigurable a été utilisé pour gérer cette interopérabilité. Toutefois on notera que sa bande passante est trop large et que plusieurs canaux de communications insuffisamment atténués serons présents au niveau des convertisseurs. Pour relâcher les contraintes de linéarité et les marges de numérisation, nous proposons de rendre le récepteur conscient de son environnement spectral.L'intérêt de cette méthode a été développé dans une première partie de la thèse. Une méthode de détection aveugle basée sur les propriétés cyclostationnaires du signal de communication a été proposée. Nous utilisons ensuite cet outil pour détecter la présence d'harmoniques indésirables créés lors d'un fonctionnement en régime non linéaire du récepteur. Nous montrons qu'il est ainsi possible de détecter très tôt le mode de fonctionnement du récepteur. Enfin, basé sur les deux études précédentes nous proposons une méthode permettant d'adapter automatiquement les paramètres de la chaine analogique. Cette preuve de concept basé sur une boucle de rétroaction permet une amélioration significative du taux d'erreur binaire tout en relâchant les contraintes au niveau analogique
This thesis was prepared at IMT Atlantique, in the context of the FITNESS project. This European project aims to modernize PMR (Professional Mobile Radio) radios, used by security services. The objective is to develop a new receiver compatible with the 4 European standards, while adding new features. The contributions of this thesis to FITNESS concern the backwards compatibility of the new receiver with current standards. A reconfigurable central frequency channel selection filter was used to manage this interoperability. However, it should be noted that its bandwidth is too large and that several insufficiently attenuated communication channels are present at the A/D converters input. To relax linearity constraints and digitization margins, we propose to make the receiver aware of its spectral environment.The interest of this method has been developed in a first part of this document. A method for blind detection based on the cyclostationary properties of the communication signal has been proposed. We then use this tool to detect the presence of unwanted harmonics created during non-linear operation of the receiver. We show that it is possible to detect the operating mode of the receiver before the useful signal degradation. Finally, based on the two previous studies, we propose a method to automatically adapt the parameters of the analog receiver. This proof of concept based on a feedback loop allows a significant improvement in the bit error rate while relaxing constraints at the analog level

Resumo: A busca por padronização e melhorias dos processos produtivos se acentua em períodos marcados por uma crise econômica mundial. Procurar alternativas com o intuito de se manter presente e competitivo, frente às novas exigências do mercado, foi a reação de um instituto de pesquisas, atuante no ramo de TI. Este cenário desencadeou a motivação para realização do trabalho descrito nesta dissertação. Este instituto foi escolhido para um estudo de caso, que tem como objetivo observar se houve uma elevação dos índices de conformidade ao processo de gestão de projetos, com foco no departamento de hardware, que se avaliou ser decorrente da implantação de um Escritório de Gerenciamento de Projetos (PMO). As ações realizadas para estruturação desse PMO, após serem observadas, estão documentadas ao longo desta dissertação, assim como a correlação dos dados sobre aderência ao processo de dois anos de projetos realizados nesta instituição, comprovando o impacto da atuação do PMO no desempenho dos projetos do departamento foco da análise
Abstract: The search for standardization and improvements for productive processes gets sharp during periods that are marked by a global economic crisis. To look for alternatives aiming at staying present and competitive, considering new market requirements, was the reaction of a research and development institute that works with Information Technology. This scenario triggered the motivation to perform the effort described in this thesis. This institution was chosen for a case study that aims at observing if there was an increase of compliance rates to the project management process, with the focus on the hardware department. The compliance increase was determined being the installation consequence of a Project Management Office (PMO). The performed actions for the PMO structuring, after being observed, are documented in this document, as well as the correlation of process compliance data from a two year period of developed projects in this institution, proving the impact of the PMO performance on the projects outcomes of the target department
