Abstract:
The scanning electron microscope and a backscattered electron detector were used for the first time to identify calcium oxalate in leaf tissues after labelling crystals with silver. Freehand sections fixed in formalin – acetic acid – alcohol or glutaraldehyde were treated with 5% acetic acid to remove background calcium carbonate and calcium phosphate and were silver stained by calcium-substitution techniques. Two standard staining techniques, Pizzolato (AgNO3–H2O2) and Yasue (AgNO3 – rubeanic acid) tests, were compared. Druses and raphides were identified morphologically and histochemically in the leaves of Anthurium scherzeranum and Dieffenbachia maculata. The Pizzolato procedure was the best method for staining calcium oxalate for scanning electron microscope studies. Excessive deposition of granular material in the Yasue staining method tended to obscure crystal morphology in scanning electron microscope preparations.

Poucos são os estudos desenvolvidos no sentido de caracterizar o administrador esportivo de clubes, federações e entidades de administração do Esporte (Bastos, 2003, 2004; Medalha,1982) e descrever as características organizacionais e de gestão de equipes brasileiras de Voleibol (Durieux, 2005; Pizzolato, 2004). A presente pesquisa, descritiva e exploratória, teve como objetivo descrever o perfil do gestor e as características organizacionais de equipes de Voleibol do Brasil participantes da Superliga de Voleibol 2007-2008, através de estudo de casos múltiplos. Para tanto, foi aplicado questionário junto aos gestores das equipes, composto de questões abertas e fechadas. Concluiu-se que as equipes possuem gestor profissional que exerce funções gerenciais estratégicas, e que a estrutura organizacional do Voleibol brasileiro de alto rendimento se caracteriza por ser fortemente calcada na instituição clubística, amparada prioritariamente pela iniciativa privada e com apoio do setor público municipal

Abstract:
Histochemical and cristallographic studies were performed in light microscopy and scanning electron microscopy on crystal-bearing cystidia of several Inocybe spp. (Agaricales) and confirmed the excretory function of these sterile cells of the hymenium. At the apex of these bottle-like cells, the wall had an axial polysaccharidic matrix rich in carboxyl groups. It also presented an enhanced permeability and a local concentration of Ca2+. Functioning as an hydathode, the cystidium secreted, at its apex, an aqueous droplet into which twin crystals developed. In addition, crystal sand were found in the cystidium wall. These two crystalline products were calcium oxalate as confirmed with anthracene green and the Pizzolato test. A detailed crystallographic study showed that these twin crystals undoubtedly belong to the monoclinic system, i.e., the monohydrated form of the calcium oxalate or whewellite. In the case of Inocybe asterospora Quél., these grouped crystals were formed regularly by two pairs of twin crystals. The identification and localization of the Ca2+ ions have been determined with the scanning electron microscope by means of the X-emission pictures and the resulting energy dispersive X-spectrum for the latter element. Abstract:
Spiritual development is an epistemological journey of seeking to make meaning of life’s activities, order, and the relationship between events (Love, 2002). This process occurs when students experience a degree of dissonance that pushes them to question what they know, how they know it, and expand their understanding based on new experiences and information learned (Bakari, 2000; Chaudhari & Pizzolato, 2008). Studying abroad provides opportunities for students to encounter disequilibrium as they interact with other cultures that cause them to consider alternative viewpoints, and enable other’s practices and beliefs to influence the formation of their own views, beliefs and practices (Chaudhari & Pizzolato, 2008). This article utilizes Fowler’s (1981) stages of faith development to understand the meaning-making and spiritual development of study abroad participants. Multiple semi-structured interviews (pre-, during, and post-study abroad) were conducted with 25 African-American students who participated in long-term, immersive, study-abroad programs in 13 different countries on 5 different continents. During pre-study abroad interviews, three participants disclosed a faith-background and expressed intention to connect with a community of believers during their time abroad. These participants were in what Fowler would consider a synthetic-conventional stage of faith development, characterized by conventional practices influenced from home and parental influences. While abroad, five students shared that they had independently researched local churches in their host countries, and regularly attended services alone or with another study abroad participant, because they needed the support of a community of believers to empower them during their time abroad. Additionally, numerous participants addressed differences in how faith is discussed and practiced in their host country and America. One-third of participants discussed studying abroad having an impact on their faith and religious beliefs. In addition, participants discussed specific lessons and practices learned from their community of believers in their host countries, which they intended to incorporate in their faith practices and beliefs upon return to the U.S. Particularly interesting was a distinction noted between the role faith plays in the U.S. and abroad: participants described their faith communities in the U.S. placing a strong emphasis on what God can do for people, whereas in faith communities abroad, the emphasis was placed on believer’s responsibility to worship God rather than on what God does or does not provide. Another key finding was the connection between student development stages of identity development and stages of faith development. Participants in later stages of identity development demonstrated a transition from Fowler’s synthetic-conventional stage of faith development to the individuative-reflective stage of faith development. In the individuative-reflective stage, students break free from parental and community influenced practices to explore their own thoughts and experiences of God and determine for themselves how they will practice their faith. A similar process occurs in practices and behaviors when a student explores and forms a solidified identity – they shift from parental and communal influences on identity formation to forming beliefs and actions based on personal experiences and thoughts.

Abstract:
Objective: The study aimed to evaluate the preventive effects of hydroxycitric acid(HCA) for stone formation in the glyoxylate-induced mouse model. Materials and methods: Male C57BL/6J mice were divided into a control group, glyoxylate(GOX) 100 mg/kg group, a GOX+HCA 100 mg/kg group, and a GOX+HCA 200 mg/kg group. Blood samples and kidney samples were collected on the eighth day of the experiment. We used Pizzolato staining and a polarized light microscope to examine crystal formation and evaluated oxidative stress via the levels of malondialdehyde (MDA), superoxide dismutase (SOD), and glutathione peroxidase (GSH-Px). Quantitative reverse transcriptase-polymerase chain reaction (qRT-PCR) was used to detect the expression of monocyte chemotactic protein-1(MCP-1), nuclear factor-kappa B (NF κ B), interleukin-1 β (IL-1 β) and interleukin-6 (IL-6) messenger RNA (mRNA). The expression of osteopontin (OPN) and a cluster of differentiation-44(CD44) were detected by immunohistochemistry and qRT-PCR. In addition, periodic acid Schiff (PAS) staining and TUNEL assay were used to evaluate renal tubular injury and apoptosis. Results: HCA treatment could reduce markers of renal impairment (Blood Urea Nitrogen and serum creatinine). There was significantly less calcium oxalate crystal deposition in mice treated with HCA. Calcium oxalate crystals induced the production of reactive oxygen species and reduced the activity of antioxidant defense enzymes. HCA attenuated oxidative stress induced by calcium oxalate crystallization. HCA had inhibitory effects on calcium oxalate-induced inflammatory cytokines, such as MCP-1, IL- 1 β, and IL-6. In addition, HCA alleviated tubular injury and apoptosis caused by calcium oxalate crystals. Conclusion: HCA inhibits renal injury and calcium oxalate crystal deposition in the glyoxylate-induced mouse model through antioxidation and anti-inflammation.

Peroxisome proliferator-activated receptor- (PPAR-) γ is a ligand-dependent transcription factor, and it has become evident that PPAR-γ agonists have renoprotective effects, but their influence and mechanism during the development of calcium oxalate (CaOx) nephrolithiasis remain unknown. Rosiglitazone (RSG) was used as a representative PPAR-γ agonist in our experiments. The expression of transforming growth factor-β1 (TGF-β1), hepatocyte growth factor (HGF), c-Met, p-Met, PPAR-γ, p-PPAR-γ (Ser112), Smad2, Smad3, pSmad2/3, and Smad7 was examined in oxalate-treated Madin-Darby canine kidney (MDCK) cells and a stone-forming rat model. A CCK-8 assay was used to evaluate the effects of RSG on cell viability. In addition, intracellular reactive oxygen species (ROS) levels were monitored, and lipid peroxidation in renal tissue was detected according to superoxide dismutase and malondialdehyde levels. Moreover, the location and extent of CaOx crystal deposition were evaluated by Pizzolato staining. Our results showed that, both in vitro and in vivo, oxalate impaired PPAR-γ expression and phosphorylation, and then accumulative ROS production was observed, accompanied by enhanced TGF-β1 and reduced HGF. These phenomena could be reversed by the addition of RSG. RSG also promoted cell viability and proliferation and decreased oxidative stress damage and CaOx crystal deposition. However, these protective effects of RSG were abrogated by the PPAR-γ-specific inhibitor GW9662. Our results revealed that the reduction of PPAR-γ activity played a critical role in oxalate-induced ROS damage and CaOx stone formation. RSG can regulate TGF-β1 and HGF/c-Met through PPAR-γ to exert antioxidant effects against hyperoxaluria and alleviate crystal deposition. Therefore, PPAR-γ agonists may be expected to be a novel therapy for nephrolithiasis, and this effect is related to PPAR-γ-dependent suppression of oxidative stress.

Interaction of pioglitazone (PGZ) and macrophages (Mps) in renal crystal formation remains unclear. In the present study, we investigated the possible mechanisms involved with Mps of PGZ in suppressing renal crystal formation. Crystal formation in the mouse kidney was detected using polarized light optical microscopy and Pizzolato staining. Gene expression was detected by Western blot analysis, quantitative RT-PCR, immunohistochemistry, immunofluorescence, and ELISA. Mp phenotypes were identified by flow cytometric analysis. Cell apoptosis was detected with TUNEL assay, and tubular injury was detected with periodic acid-Schiff staining. Interaction of peroxisome proliferator-activated receptor (PPAR)-γ and promoter was determined by chromatin immunoprecipitation assay. Luciferase reporter assay was performed to authenticate target genes of miRNA-23 (miR-23). Recombinant adenovirus was used to elucidate the role of miR-23 in vivo. Renal crystal formation, inflammation, tubular injury, and cell apoptosis were significantly marked in glyoxylic acid-treated groups and significantly decreased in PGZ-treated groups. PGZ significantly reduced Mp infiltration and M1 Mp polarization in the kidney. In vitro, PGZ shifted crystal-stimulated M1-predominant Mps to M2-predominant Mps, which were anti-inflammatory. PPAR-γ could directly bind to one PPAR-γ regulatory element in the promoter of pre-miR-23 to promote expression of miR-23 in Mps. We identified two downstream target genes of miR-23, interferon regulatory factor 1 and Pknox1. Moreover, miR-23 decreased crystal deposition, M1 Mp polarization, and injury in the kidney. This study has proven that PGZ decreased renal calcium oxalate crystal formation and renal inflammatory injury by suppressing M1 Mp polarization through a PPAR-γ-miR-23-interferon regulatory factor 1/Pknox1 axis. PGZ is liable to be a potential therapeutic medicine for treating urolithiasis.

Luisa Prandi (born in 1952) has showed a great interest in Classics and the Ancient World even from her earliest age. Her love for the Ancient Greek world arose during her studies at the University of Milan. As a student of distinguished scholars such as Marta Sordi (1925-2009), Luigi F. Pizzolato (1939-), Giuseppe Billanovich (1913-2000) and Orsolina Montevecchi (1911-2009), she learned from each of them a devotion for the study of the ancient sources that have made, with a great merit, that she becomes one of the best and most recognized scholars on the ancient world. She is, also, a main name in the research of the topic of the Historians of Alexander or Alexandrographers. These authors are fundamental for the knowledge of the Macedonian king: as she uses to argue, we cannot write a single word about Alexander overlooking them”. Writing about Alexander is a such a Herculean task, since there is no other figure of whom there are more historians with extreme contradictions among their accounts. It is almost impossible to say something new about Alexander, but to say about his historians, about whom so few fragments are available, is even more difficult. Prof. Luisa Prandi’s brilliant versatility is quite a strange attribute among academics: she has showed many times her abilities to work directly with all kinds of sources (especially papyri), but always with a sober, serious, and noncontroversial approach. Readers will find in her a person of carefully taste concerning what to say and how to stress her opinions and views. That seriousness is reflected and witnessed when she writes, since she belongs to that wonderful school of researchers who are aware that theories will pass but the facts (sources) are the ones that remain. From that humility and that certainty, Luisa Prandi presents herself to our readers of Karanos.

Oxidative stress and inflammatory response are closely related to nephrolithiasis. This study is aimed at exploring whether rosiglitazone (ROSI), a regulator of macrophage (Mp) polarization, could reduce renal calcium oxalate (CaOx) deposition by ameliorating oxidative stress and inflammatory response. Male C57 mice were equally and randomly divided into 7 groups. Kidney sections were collected on day 5 or day 8 after treatment. Pizzolato staining and polarized light optical microscopy were used to detect crystal deposition. PAS staining and TUNEL assay were performed to assess the tubular injury and cell apoptosis, respectively. Gene expression was assessed by immunohistochemistry, immunofluorescence, ELISA, qRT-PCR, and Western blot. The reactive oxygen species (ROS) level was assessed using a fluorescence microplate and fluorescence microscope. Hydrogen peroxide (H2O2), malonaldehyde (MDA), and glutathione (GSH) were evaluated to determine oxidative stress. Lactic dehydrogenase (LDH) activity was examined to detect cell injury. Adhesion of CaOx monohydrate (COM) crystals to HK-2 cells was detected by crystal adhesion assay. HK-2 cell death or renal macrophage polarization was assessed by flow cytometry. In vivo, renal crystal deposition, tubular injury, crystal adhesion, cell apoptosis, oxidative stress, and inflammatory response were significantly increased in the 7-day glyoxylic acid- (Gly-) treated group but were decreased in the ROSI-treated groups, especially in the groups pretreated with ROSI. Moreover, ROSI significantly reduced renal Mp aggregation and M1Mp polarization but significantly enhanced renal M2Mp polarization. In vitro, ROSI significantly suppressed renal injury, apoptosis, and crystal adhesion of HK-2 cells and markedly shifted COM-stimulated M1Mps to M2Mps, presenting an anti-inflammatory effect. Furthermore, ROSI significantly suppressed oxidative stress by promoting the Nrf2/HO-1 pathway in HK-2 cells. These findings indicate that ROSI could ameliorate renal tubular injury that resulted from oxidative stress and inflammatory response by suppressing M1Mp polarization and promoting M2Mp polarization. Therefore, ROSI is a potential therapeutic and preventive drug for CaOx nephrolithiasis.

The Brazilian Symposium on Virtual and Augmented Reality (SVR) celebrated its 10th edition in 2008, in the city of João Pessoa. During these last 10 years, SVR has earned its space and magnitude as a truly consolidated event within the Brazilian research community. The papers in this special issue of RITA were selectively chosen among the 24 full papers presented in SVR 2008. They are extended and revised versions of those originally presented in the symposium. The material in this special issue is representative of the wide scope of the areas covered by the symposium. The first paper, the only one not presented at the symposium, was written by the editors jointly with Luciano Soares and Veronica Teichrieb. It provides a rich panorama of current Brazilian research in Virtual Reality (VR) and related areas, based on an analysis of the 124 full papers presented over the last four SVR editions. The second paper, by Silvano Malfatti, Selan dos Santos, Luciane Fraga, Claudia Justel, Paulo Rosa, and Jauvane Oliveira, describes some interesting VR work. Its authors present an engine (EnCIMA) aimed at the quick development of VR applications. The third paper, by Fábio Miranda, Romero Tori, Cláudio Bueno, and Lucas Trias, presents research results from the Augmented Reality (AR) area. This paper, which received the best paper award in the symposium, presents a projection-based AR X-Ray tool to allow visual exploration of internal details of walls, as an illustration. The fourth paper, by Leandro Fernandes, Vitor Pamplona, João Prauchner, Luciana Nedel, and Manuel Oliveira, explores 3D user interaction. It presents the implementation of a data glove that uses a camera to track visual markers at finger tips. Abstract Introduction There is a significant incidence of cats with renal disease (RD) and calcium oxalate (CaOx) kidney uroliths in domesticated cats. Foods which aid in the management of these diseases may be enhanced through understanding the underlying metabolomic changes. Objective Assess the metabolomic profile with a view to identifying metabolomic targets which could aid in the management of renal disease and CaOx uroliths. Method This is a retrospective investigation of 42 cats: 19 healthy kidney controls, 11 with RD, and 12 that formed CaOx nephroliths. Cats were evaluated as adults (2 through 7 years) and at the end of life for plasma metabolomics, body composition, and markers of renal dysfunction. Kidney sections were assessed by Pizzolato stain at the end of life for detection of CaOx crystals. CaOx stone presence was also assessed by analysis of stones removed from the kidney at the end of life. Results There were 791 metabolites identified with 91 having significant (p < 0.05, q < 0.1) changes between groups. Many changes in metabolite concentrations could be explained by the loss of renal function being most acute in the cats with RD while the cats with CaOx stones were intermediate between control and RD (e.g., urea, creatinine, pseudouridine, dimethylarginines). However, the concentrations of some metabolites differentiated RD from CaOx stone forming cats. These were either increased in the RD cats (e.g., cystathionine, dodecanedioate, 3-(3-amino-3-carboxypropyl) uridine, 5-methyl-2′-deoxycytidine) or comparatively increased in the CaOx stone forming cats (phenylpyruvate, 4-hydroxyphenylpyruvate, alpha-ketobutyrate, retinal). Conclusions The metabolomic changes show specific metabolites which respond generally to both renal diseases while the metabolomic profile still differentiates cats with RD and cats with CaOx uroliths.

Student development has connections to important academic purposes in higher education (King, Baxter Magolda, Barber, Kendall Brown & Lindsay, 2009). In particular, a growing body of work on self-authorship, a social-constructive theory of development, has demonstrated relevance to the purposes of higher education (Baxter Magolda, 2001; King & Baxter Magolda, 2004). The conditions which support self-authorship development in academic settings have been studied in detail, drawing attention to what King et al. (2009) frame as developmentally effective educational experiences. Explorations of self-authorship development in academic settings have focused on students’ experiences and outcomes. The classroom experiences of faculty, particularly those working outside institutional initiatives, to support self-authorship have received less attention. This study used a theory-driven (Baxter Magolda, 2001; Pizzolato, 2005), practice-based research framework, to explore a faculty-student affairs collaboration through participant observation as the collaborators sought to align their teaching practices with the tenets of self-authorship development in the context of a senior undergraduate course in Service-Learning. Four themes emerged, which have relevance for those who wish to consider student personal and academic development concurrently. We argue that individual faculty members can collaborate with student affairs professionals and use self-authorship theory to expand their constructions of what it means to be a “good professor” by approaching teaching as a mirror image of the self-authorship journey travelled by students. Les programmes de perfectionnement des étudiants sont liés aux objectifs académiques importants de l’enseignement supérieur (King, Baxter Magolda, Barber, Kendall Brown & Lindsay, 2009). En particulier, les travaux de plus en plus nombreux qui portent sur l’épistémologie personnelle (self-authorship), une théorie constructive sociale de développement, ont démontré leur pertinence par rapport à l’enseignement supérieur (Baxter Magolda, 2001; King & Baxter Magolda, 2004). Les conditions qui soutiennent le développement de l’épistémologie personnelle en milieu universitaire ont été étudiées en détail et attirent l’attention sur ce que King et al (2009) formulent comme des expériences éducatives efficaces de développement. Les explorations du développement de l’épistémologie personnelle en milieu universitaire se sont généralement concentrées sur les expériences et les résultats des étudiants. Les expériences des professeurs en salle de classe, en particulier celles des enseignants qui oeuvrent en dehors des initiatives institutionnelles dans le but de soutenir l’épistémologie personnelle, ont été beaucoup moins examinées. Cette étude a été menée dans un cadre de recherche guidé par la théorie et axé sur la pratique. Elle explore la collaboration entre professeurs et affaires étudiantes par le biais de l’observation des participants alors que les collaborateurs tentent de faire correspondre leurs pratiques d’enseignement avec les principes du développement de l’épistémologie personnelle, dans le contexte d’un cours de premier cycle de niveau avancé d’apprentissage du service communautaire. Quatre thèmes ont été mis en lumière. Ceux-ci sont pertinents pour ceux et celles qui souhaitent tenir compte à la fois du développement personnel et du développement académique des étudiants. Nous soutenons que les professeurs peuvent collaborer avec les professionnels des affaires étudiantes et faire usage de la théorie de l’épistémologie personnelle pour élargir leur compréhension de ce que cela signifie d’être « un bon professeur » en abordant l’enseignement comme le pendant du chemin d’épistémologie personnelle suivi par les étudiants.
