Abstract:
The abnormal aggregation of fibrillar α-synuclein in Lewy bodies plays a critical role in the pathogenesis of Parkinson’s disease. However, the molecular mechanisms regulating α-synuclein pathological effects are incompletely understood. Here we show that α-synuclein binds phosphoinositide-3 kinase enhancer L (PIKE-L) in a phosphorylation-dependent manner and sequesters it in Lewy bodies, leading to dopaminergic cell death via AMP-activated protein kinase (AMPK) hyperactivation. α-Synuclein interacts with PIKE-L, an AMPK inhibitory binding partner, and this action is increased by S129 phosphorylation through AMPK and is decreased by Y125 phosphorylation via Src family kinase Fyn. A pleckstrin homology (PH) domain in PIKE-L directly binds α-synuclein and antagonizes its aggregation. Accordingly, PIKE-L overexpression decreases dopaminergic cell death elicited by 1-methyl-4-phenylpyridinium (MPP+), whereas PIKE-L knockdown elevates α-synuclein oligomerization and cell death. The overexpression of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) or α-synuclein induces greater dopaminergic cell loss and more severe motor defects in PIKE-KO and Fyn-KO mice than in wild-type mice, and these effects are attenuated by the expression of dominant-negative AMPK. Hence, our findings demonstrate that α-synuclein neutralizes PIKE-L’s neuroprotective actions in synucleinopathies, triggering dopaminergic neuronal death by hyperactivating AMPK.

AbstractKenneth L. Pike’s tagmemic explanation of his etic-emic equivalence corresponds to the notion of “approximate” translation. According to a weaker version of the Sapir-Whorf hypothesis, Pike’s cross-cultural and multilingual perspective of Bible translation approximates the duality and triadicity of Peirce’s immediate/emotional, dynamical/energetic, and final/logical interpretants. Pike’s astronautical examples of the artificial language Kabala-X translated into English and the science fiction story of the Earthmen who invaded Mars are fictional and creative artifacts of human-alien cryptography leading, as argued here, to false semio-logical reasoning.

Abstract:
The article presents the results of a study of surface waters and fish (perch (Perca fluviatilis L.), carp (Cyprinus carpio L.), bream (Abramis brama L.), pike (Esox lucius L.) of the Irtysh river. The content of Cu and Pb in the organs and tissues of fish living in the Irtysh River within the city of Semey was determined by the ditizon photometric method. The ecological assessment of the results indicates that the concentrations of Cu and Pb in the studied fish specimens do not exceed the MPC. A significant difference in the specific features of the accumulation of heavy metals has been established, depending on the type of fish nutrition and the time of year. The Pb content in various organs of perch, carp, bream and pike ranges from 0.062 mg/kg to 0.083 mg/kg. The highest concentration of Cu is observed in predatory fish (3.27 mg/kg), the lowest – in planktonivores (2.82 mg/kg). Pb also mainly accumulates in pike (0.073 mg/kg). In spring, the content of Pb in the gills of perch and pike increases, Cu – in the gills of bream and pike. The maximum concentrations of Cu in fish were observed in summer, which is associated with an increase in the food supply. The Pb content in summer, on the contrary, decreases due to its adsorption by suspended solids and deposition with them into bottom sediments.

Abstract:
Changes in blood lactate, pH, Pco2, and plasma [Formula: see text] and glucose were measured during recovery from exercise–handling stress in chronically cannulated northern pike (Esox lucius L.). Exercise was induced by lifting pike out of water (three 30-s periods of emersion alternated with 30-s periods of water submersion), while they were inside tubular plastic chambers. Large depressions in blood pH and plasma [Formula: see text] occurred within minutes after exercise. Initially, the acidosis was of mixed respiratory (elevated Pco2) and metabolic (H+ release from the muscle) origin. Although the respiratory acidosis was corrected in 2 h, 8 h was required to return blood pH to preexercise levels. A large accumulation of lactate anion in the blood occurred after exercise, the increase by 2 h (12.8 mmol L−1), being approximately equal to the metabolic acid load. These results conflict with previous reports on muskellunge (Esox masquinongy M.), in which blood lactate elevation following exercise was small (2.5 mmol L−1) and greatly exceeded by the metabolic H+ load (7.0 mmol L−1). By 2 h after exercise the plasma glucose level of pike had increased by 8.8 mmol L−1 and the return to preexercise levels was almost complete by 12 h. Intraarterial infusion of a large sodium lactate load elevated plasma glucose level by only 1.7 mmol L−1, indicating that postexercise hyperglycemia in pike is a response to stress and not simply a consequence of lactate removal by conversion into glucose.

Abstract Northern pike (Esox lucius L.) is of great importance to inland commercial and recreational fisheries in Poland. Thanks to northern pike’s substantial size, strength, the possibility of using a variety of fishing methods and techniques and various baits, and many other values, lake anglers value this species above all others. Additionally, for over a decade, northern pike has been the most widely stocked species in Polish inland waters of lakes, rivers and dam reservoirs. The aim of this work was to analyze in detail the size and characteristics of northern pike commercial fisheries and angling catches in Polish inland waters over the long term. The analyses were based on a compilation of traditional scientific papers, primarily analyses of commercial catches, and a review of the results of more than 40 years of studies on the size of northern pike angling catches. The analysis of research material indicated a steady decrease in commercial catches of this species in the years State Fisheries Enterprises (SFE) were in operation followed by relative stability in catches over the past two decades. The paper describes the methods used to estimate angling catches, namely questionnaire and catch-return (the so-called register) research. An increasing disproportion between the sizes of pike catches made with angling and commercial fishing gear was documented. Despite the problems presented in the paper related to estimating total pike catches, the analysis revealed that anglers catch eightfold more northern pike than do commercial fishers.

Abstract:
The work submitted in this thesis describes three investigations of the relationship between stimuli, the pike and the pike's inclination to feed. Consideration is given to the importance of relating internal and external factors to the causation of behaviour. The effects of individual pike and the time lapsed after feeding on predatory behaviour are firstly assessed. Use of multivariate analysis determined key behaviours in the repertoire of pike that could be successfully employed in analysing prey-capture sequences. The modifying influence of hunger, as categorised by stomach content, on prey capture sequences is presented and discriminant function analysis is used as a method of describing differences in predatory behaviour. A simple treatment of bout criteria is also given. The interaction of the components of visual stimuli and its influence on predatory response revealed that particular movements and shape of a stimulus altered the attack behaviour of pike. The relative strength of a visual cue, for example, how "fish-like" a stimulus is, has dependence on how inclined an individual is to feed. A view of how these factors influence the behaviour of pike in a wider context is expressed.

Abstract:
Otolith increments are used extensively to gain information on age and growth of fish. This is based on two fundamental assumptions. First, that the primary increments are produced at a known and constant periodically, and secondly, that the distance between the consecutive otolith increments is proportional to the growth of the fish. This study proposes to investigate the validity of the second assumption on a daily and weekly scale. There was a significant correlation between otolith size and fish size in Atlantic salmon (Salmo solar L.) and haddock (Melanogrammus aeglefinus L.). However, no correlation was found between somatic growth and otolith growth in individually measured yolk-sac haddock larvae or demersal juvenile haddock. This supports the suggestion that there is no functional relationship between otolith accretion rate and somatic growth rate. Different back-calculation models were employed to illustrate how this apparent 'uncoupling' of somatic growth and otolith growth can increase the error in size and age predictions. The possibility that there is a relationship between one or more components of metabolic rate and otolith accretion was investigated, in individual demersal juvenile haddock, Atlantic salmon, and pike (Esox, lucius L.). Variations in otolith daily increment width and somatic growth were compared with experimentally induced variations in resting metabolic rate, specific dynamic action (the increase in metabolism during digestion) and activity metabolism. Salmon and haddock were subjected to acute variations in temperature to vary resting metabolic rate. Pike were fed a single meal to elicit specific dynamic action. Activity levels in salmon were manipulated by exploiting the territoriality of salmon in a fluvial aquarium. An experimental change in resting metabolic rate resulted in a change in increment width that was uncoupled from somatic growth. Metabolic rate and increment width increased simultaneously after feeding in pike. In both experiments there was a less than isometric relationship between the two parameters. No correlation was found between increment width and the activity level of salmon.

Resumo: Xavante é uma língua da família Jê, falada por aproximadamente 13.000 indígenas que vivem no estado do Mato Grosso. O presente trabalho descreve a fonologia segmental desta língua, utilizando a abordagem fonêmica encontrada no livro Phonemics (PIKE 1971[1947]). Embora a fonologia do Xavante já tenha sido tratada por outros autores, a análise apresentada aqui contém uma variedade de observações e interpretações novas, apresentando soluções para alguns problemas que não foram resolvidos em estudos anteriores. O primeiro capítulo descreve a metodologia usada na pesquisa. Dados foram coletados através da utilização de publicações anteriores, que serviram como guia no desenvolvimento de questionários delineados para solicitar tipos específicos de dados lingüísticos. A pesquisa baseia-se em grande parte em dados solicitados a um informante, indivíduo alfabetizado em Xavante e em Português, que foram comparados com a fala de outras pessoas da mesma região dialetal. O capítulo 2 contém um breve sumário do contexto histórico e lingüístico do povo Xavante e uma revisão da literatura lingüística relevante, composta principalmente de trabalhos feitos por missionários do Summer Institute of Linguistics (SIL) e da Missão Salesiana de Mato Grosso, além da dissertação de Quintino (2000). O terceiro capítulo começa com um sumário da abordagem teórico-metodológica encontrada no livro Phonemics (1971[1947]) de Kenneth Pike. Esta abordagem foi escolhida em parte porque as análises anteriores da fonologia Xavante foram feitas a partir desta perspectiva, e os problemas fonológicos na língua podem ser claramente compreendidos quando vistos à luz das virtudes e limitações da abordagem Pikeana. Uma análise abrangente de todos os fonemas segmentais da língua é apresentada em seguida, com base na perspectiva de Pike e em trabalhos anteriores, além de dados e novas análises. Os fonemas /z,?,?/ e os segmentos de coda [p,m,b] e [j,~j] são discutidos em detalhe e interpretados de uma forma diferente dos autores anteriores. A estrutura silábica é também analisada do ponto de vista Pikeano. O capítulo 4 descreve a evolução dos três diferentes sistemas ortográficos atualmente usados entre os Xavantes, mostrando que algumas das dificuldades enfrentadas pelos criadores destas ortografias refletem problemas na análise fonológica da língua. O capítulo 5 apresenta uma análise original dos segmentos de coda em Xavante. Um aspecto problemático da abordagem de Pike, decorrente do pressuposto de que fonemas são entidades indivisíveis, é discutido neste capítulo, bem como as tentativas de Burgess (1971) e Quintino (2000) de utilizar abordagens teóricas alternativas para analisar as codas em Xavante. Em seguida, apresenta-se o argumento de que as duas codas possíveis na língua, manifestadas respectivamente pelo conjunto neutralizado [p,m,b] e os alofones [j,~j], representam dois segmentos fonológicos em contraste fonêmico na posição final da sílaba. O capítulo 6 resume criticamente o conteúdo dos capítulos anteriores e um apêndice trata dos problemas não resolvidos relacionados ao acento, ao alongamento de vogais e às alterações morfofonológicas
Abstract: The present work reanalyzes the phonology of Xavante (Jê family, 13,000 speakers, Mato Grosso State, Brazil). Based on several previous analyses and the author's own fieldwork, the segmental phonology of the language is presented from the viewpoint of Pike's Phonemics. Syllable structure is defined with reference to the distribution of segments at phrase and morpheme boundaries. The phonological problems confronted by the creators of a spelling system for the language are also described. The distributional analysis of the syllable codas in the language is used to illustrate the problems with Pike's interpretation of neutralization. The attempts by Burgess (1971) and Quintino (2000) to use alternative theoretical approaches to analyze Xavante codas are discussed, and an original solution to the problem is presented. It is argued that the two possible codas in the language, manifested respectively by the neutralized group [p,b,m] and the allophones [j,~j], represent two phonological segments in phonemic contrast in syllable-final position. A brief discussion of unresolved problems related to accent, vowel length, and morphophonological alteration is also included

Abstract:
Drug development for conventional delivery targets is the major challenge for therapeutic progress. PI3K signaling has shown a prominent role in progression of meningioma which subsequently makes these receptors a target for efficient treatment approach. Loss or alteration of NF2 (neurofibromin 2) with respect to its gene product merlin is crucially implicated in tumor development eventually leads to the meningioma. However, the interactions of meningioma pathophysiology (merlin) to many pathways makes it complicated. Different signalling pathways interacting with merlin are the hippo pathway, PI3K /Akt route and mTORC pathway. We thesis on the inhibitory approach for P13K pathway and phosphorylation of Akt to terminate merlin polyubiquitination and promote its expression and association with PIKE-L thereby activating tumor suppressive action. Our work emphasizes on exploring biological compounds that target the PI3K pathway using the in-silico method. Here, we implemented 50 phytochemicals from various plant sources against PI3K for molecular docking and assessed as a potential anticancer drug. The docking was performed showing binding energy, drug likeness and affinity of different phytochemicals. The following study demonstrated various phytochemicals (such as Leachianone A, withaferin A, kurarinol and gardenoilic acid B) along with comparing their ADME score, bioavailability radar and score that provides insights on possible therapeutic approach of these biological compounds.

Abstract:
Within my diploma thesis I have been trying to resolve the possibilities of optimization of artificial incubation of eggs and embryos in northern pike (Esox Lucius L.) under controlled conditions during three different experiments. In the first experiment was investigated the effect of three different semination solutions to the success of incubation of eggs and embryos in northern pike. I watched the survival of eggs and embryos, larvae hatching abnormalities and biometrics of larvae. It was found that the best overall results were achieved with semination solution composed of 15 ml of chemical crystalic urea per 1 liter of water, with an appropriate amount of abnormalities and highest larvae survival. In the second experiment were performed manipulation with developing eggs at given time intervals after fertilization of eggs. I watched the survival of eggs and embryos, abnormality and biometrics of hatched larvae. It was found that the lowest survival, abnormalities, and smallest TL and W were reached during manipulations within 24 hours after fertilization. In the third experiment was investigated the effect of water temperature on the length of the incubation of eggs and embryos in northern pike. With regard to the overall survival, larvae hatching, abnormalities and size of larvae achieved the best results temperatures 6 and 10 ° C. Conversely, it is entirely unsuitable for incubation temperature of 3 ° C (lowest survival, large amounts deformities). Different incubation temperatures have also confirmed the effect of water temperature on biometrics of hatched larvae.

Abstract:
The main objective of this thesis is to describe and experimentally evaluate the possibility of adaptation of pike (Esox lucius L.) in controlled housing conditions and income artificial pelleted feed. The thesis is mainly based on the effect of an initial larval density of pike for adaptation and survival and the growth of larvae reared in the rearing tanks. The experiment consisted of four different densities of larvae (10, 20, 40, 80 pc of the larvae l??, which were always repeated three times. Two experiments were performed (M, H), with a length of rearing of five days. Rearing itself was conducted in twelve circular tanks with a capacity of 180 l??, the recirculation system connected to the experimental rearing facility in Vodňany FROV JU. The temperature during rearing was constant. For the first experiment the temperature was 22.3 ? 03 °C. For the second one, the average temperature was 21.5 ? 0.6 °C. The frequency of feeding was continuous (24 h) with a combination of hand feeding and feeders with a timer (at night). The illumination of the aquarium tanks was provided by fluorescent lamps, for each tank individually. The light intensity was high (210 lux) and constant during the rearing. Both experiments showed the optimal density at the initial feeding from 20 to 40 units . l??. Also, we have implemented made out performed- the basic biometric analysis of W (g) and TL (mm), the body length, size of eyes, head, jaw length, prenatal length.

Abstract:
The aim of this diploma thesis is to compare quality of northern pike sperm collected by different methods. First method is collection of stripped sperm by abdominal massage of the belly. Second method is collection of stripped sperm with special catheter to eliminate sperm contamination by urine. The last method is collection of testicular sperm. Differently collected sperm was evaluated and compared its quality. The main observed parameters were sperm volume, spermatozoa concentration, spermatozoa motility and velocity and osmolality of seminal fluid. Sperm samples were used for eggs fertilization. In fertilized eggs, the fertility of eggs and larvae hatching rate were observed.
