Abstract:
Potential intensity (PI) is a metric for climate model evaluation of TC-related thermodynamic conditions. However, PI is utilized usually for assessing basin-wide TC-related thermodynamic conditions, and not for evaluating TC passage to a certain region. Here we evaluate model-simulated PI over the passage of TCs affecting South Korea (KOR PI) as well as the PI over the entire western North Pacific basin (WNP PI) using 25 CMIP5 and 27 CMIP6 models. In terms of pattern correlations and bias-removed root mean square errors, CMIP6 model performances for KOR PI are found to be noticeably improved over CMIP5 models in contrast to negligible improvement for WNP PI, although it is not in terms of normalized standard deviations. This implies that thermodynamic condition on the route of TCs affecting South Korea is likely better captured by CMIP6 models than CMIP5 models.

Cette reaction fait partie des processus quasi-elastiques, caracterises par de petits moments transferes, et donc non interpretables par la chromodynamique quantique. Le modele de glauber, utilise avec succes dans le cadre de la diffusion elastique, est exploite ici pour l'etude de la reaction d'echange de charge. Une interaction effective entre quarks, incluant un terme central et un terme spinorbite, est employee afin de rendre compte de l'echange du meson rho. Les resultats obtenus permettent de conclure a la nature diffractive de la reaction etudiee, mais ne permettent pas de determiner avec precision les rayons moyens des densites de quarks interagissants

Abstract:
This is a report on the possibility of using a Raspberry Pi as an intrusion detection system in a home environment to increase network security. The focus of this study was on how well two different generations of Raspberry Pi would be able to  handle network traffic while acting as an intrusion detection system. To examine this a testing environment was set up containing two workstation computers connected to a Raspberry Pi, each computer hosting a virtual machine. Tests measuring the network throughput as well as the CPU and memory usage were performed on each of the Raspberry Pi devices. Two models of Raspberry Pis were used; Raspberry Pi model B+ and Raspberry Pi 2 model B; each of them running the operating system Arch Linux ARM. The results of these tests were that both of the Raspberry Pis could be used as an intrusion detection system but has some limitations that could impede usage depending on the requirements of the user. Raspberry Pi 2 model B show benefits of its updated hardware by suffering lower throughput degradation than Raspberry Pi model B+, while using less of it's total CPU and memory capacity.
Den här rapporten behandlar möjligheten att använda en Raspberry Pi som ett intrångdetekteringssystem i en hemma miljö för att öka nätverkssäkerheten. Fokusen i den här studien ligger på hur väl de två senaste generationerna av Raspberry Pi skulle kunna hantera nätverkstrafik samtidigt som den undersöker nätverkstrafiken och söker efter hot. För att kontrollera hur väl en Raspberry Pi kan fungera som ett intrångdetekteringssystem har en laborationsmiljö upprättats bestående av två fysiska maskiner som vardera används för att virtualisera en virtuell maskin. Tester för att mäta datagenomströmning, processor och minnesbelastning utfördes på var och en av Raspberry Pi. Två modeller av Raspberry Pi användes; Raspberry Pi model b+ och Raspberry Pi 2 model b, både körde operativsystemet Arch Linux ARM. Resultatet av testerna visade att det går att använda båda enheterna för att upprätta ett intrångdetekteringssystem, men det finns vissa begränsningar i enheterna vilket kan begränsa implementationsmöjligheterna. Raspberry Pi 2 model B uppvisade bättre resultat i form av att den är lägre belastad och har en högre datagenomströmning till skillnad från Raspberry Pi model B+. Raspberry Pi 2 model B har nyare och snabbare hårdvara vilket är den troliga orsaken till att den presterar bättre.

Abstract:
Conjugated Polymers have attracted a great deal of research interest in recent years due to their optoelectronic properties which makes them suitable for applications in organic light-emitting devices (OLEDs) and organic photovoltaics. Their properties are strongly dependent on the electron-electron and electron-nuclear interactions as well as the disorder which is present in almost all systems at finite temperatures. In this thesis the optical properties of electronically neutral conjugated polymers will be investigated. The results obtained are general and applicable to a wide range of parameters. In order to compare these to experiment the optical properties of poly( paraphenylene), poly(para-phenylene vinylene), and derivatives have been calculated. In these polymers the primary photoexcitations are Frenkel excitons which can be described by the Frenkel-Holstein Hamiltonian, which explicitly takes into account the exciton-nuclear coupling. Disorder can be introduced into this model both as diagonal and off-diagonal disorder within the Hamiltonian. First the optical transitions in ordered, linear conjugated polymers are investigated. It is found that the length of the polymer has a direct spectroscopic signature in the emission spectrum. When off-diagonal disorder is introduced the excitons localise on portions of the chain and the length of these portions, the conjugation length, then shows a clear emission signature. As such, a disordered polymer can be described theoretically as a chain of shorter segments, which define chromophores in a polymer context. Following from these calculations the role of conformation was investigated and effects were observed that greatly determine the optical properties of non-linear polymers. Most notable the Herzberg-Teller effect, which renders symmetrically forbidden transitions weakly allowed and greatly affects the absorption and emission spectra. The signatures observed in these spectra allow the determination of the (coarse grained) conformation of the polymer, something that has been difficult to measure directly.

Abstract:

In this thesis we explore in particular the dynamics of a special type of quasi-particle in pi-conjugated materials termed polaron, the origin of which is intimately related to the strong interactions between the electronic and the vibrational degrees of freedom within these systems. In order to conduct such studies with the particular focus of each appended paper, we simultaneously solve the time-dependent Schrödinger equation and the lattice equation of motion with a three-dimensional extension of the famous Su-Schrieffer-Heeger (SSH) model Hamiltonian. In particular, we demonstrate in Paper I the applicability of the method to model transport dynamics in molecular crystals in a region were neither band theory nor perturbative treatments such as the Holstein model and extended Marcus theory apply. In Paper II we expand the model Hamiltonian to treat the revolution of phenylene rings around the sigma-bonds and demonstrate the great impact of stochastic ring torsion on the intra-chain mobility in conjugated polymers using poly[phenylene vinylene] (PPV) as a model system. Finally, in Paper III we go beyond the original purpose of the methodology and utilize its great flexibility to study radiationless relaxations of hot excitons.

Abstract:
PTEN (phosphatase and tensin homolog deleted on chromosome ten) is a potent tumor suppressor. PTEN’s tumor suppressor action is rooted in its phosphatase function on the lipid substrate phosphatidylinositol-(3,4,5)-trisphosphate (PI(3,4,5)P3). PTEN’s enzymatic activity is specific for the third position of the inositol headgroup. PI(3,4,5)P3 is a second messenger that is a part of the PI3K-Akt pathway, and its dysregulation leads to constitutively activated AKT. The result of AKT activation is cell cycle progression, motility, cell growth, and proliferation, and consequently, overaction leads to neoplastic growth and tumorigenesis. PTEN antagonizes this pathway by regulating PI(3,4,5)P3 population through its phosphatase activity which produces the lipid PI(4,5)P2 (phosphatidylinositol-(4,5)-bisphosphate). A result of PTEN’s function is that its activity must be localized at the PM (plasma membrane) since this is where its substrate resides. Additionally, the mole percent of the phosphoinositide family of lipids is small. From highest percent composition to lowest the phosphoinositide species in the PM rank as PI(4,5)P2 (~2%), PI(4)P (~1%), and PI(3,4,5)P3 (~0.02%). For PTEN to turn over its substrate, it must first translocate from the cytosol to the PM and then search through the plasma membrane for this rare but high in demand lipid. This is at the center of the scarcity paradox. This work explores how PTEN may overcome this paradox by using its multiple lipid binding domains to interact with multiple lipid partners to efficiently localize it toward a region with a high probability of having PI(3,4,5)P3. This hypothesis is tested using two kinetic methodologies. First, we use pre- steady state stopped-flow spectrometry to determine the rates that govern PTEN-lipid binding. Second, we use single-molecule total internal reflectance fluorescence (smTIRF) microscopy to resolve the diffusion coefficients and dwell times of bound PTEN on SLBs supported lipid bilayers (SLBs). We test PTEN against various lipid compositions to determine how the bilayer structure in addition to the chemistry of the lipid influences the enzyme’s binding. These compositions include PI(4,5)P2, PI phosphatidylinositol (PI), phosphatidylserine (PS), PI(4,5)P2/PI and PI(4,5)P2/PS. In addition to this kinetic work, we will also present a novel model membrane platform that takes advantage of a microfluidic device to develop lateral lipid gradients in SLBs. This microfluidic platform, in the future, will allow for the investigation of the dynamic behavior of proteins interacting with lipids but with a bilayer that has a structure recapitulating polarized membranes like in chemotaxing cells.

To calculate realistic models of objects with Ni in their atmospheres, accurate atomic data for the relevant ionization stages need to be included in model atmosphere calculations. In the context of white dwarf stars, we investigate the effect of changing the Ni IV-VI bound-bound and bound-free atomic data on model atmosphere calculations. Models including photoionization cross-section (PICS) calculated with AUTOSTRUCTURE show significant flux attenuation of up to similar to 80 per cent shortward of 180 angstrom in the extreme ultraviolet (EUV) region compared to a model using hydrogenic PICS. Comparatively, models including a larger set of Ni transitions left the EUV, UV, and optical continua unaffected. We use models calculated with permutations of these atomic data to test for potential changes to measured metal abundances of the hot DA white dwarf G191-B2B. Models including AUTOSTRUCTURE PICS were found to change the abundances of N and O by as much as similar to 22 per cent compared to models using hydrogenic PICS, but heavier species were relatively unaffected. Models including AUTOSTRUCTURE PICS caused the abundances of N/O IV and V to diverge. This is because the increased opacity in the AUTOSTRUCTURE PICS model causes these charge states to form higher in the atmosphere, more so for N/O V. Models using an extended line list caused significant changes to the Ni IV-V abundances. While both PICS and an extended line list cause changes in both synthetic spectra and measured abundances, the biggest changes are caused by using AUTOSTRUCTURE PICS for Ni.

Abstract:
This project relates the structure of the bacteriocin molecule (which is genetically determined) to its antimicrobial function. We have sequenced the 19,542 bp pediocin plasmid pMD136 and developed a genetic transfer system for pediococci. The pediocin A operon is complex, containing putative structural, immunity, processing, and transport genes. The deduced sequence of the pediocin A molecule contains 44 amino acids and has a predicted PI of 9.45. Mechanistic studies compared the interaction of pediocin PA-1 and nisin with Listeria monocytgenes cells and model lipid systems. While significant nisin-induced intracellular ATP depletion is caused by efflux, pediocin-induced depletion is caused exclusively by hydrolysis. Liposomes derived from L. monocytogenes phospholipids were used to study the physical chemistry of pediocin and nisin interactions with lipids. Their different pH optima are the results of different specific ionizable amino acids. We generated a predicted 3-D structural model for pediocin PA-1 and used a variety of mutant pediocins to demonstrate that the "positive patch" at residues 11 and 12 (and not the YGNGV consensus sequence) is responsible for the binding step of pediocin action. This structure/function understanding gained here provides necessary prerequisites to the more efficacious use of bacteriocins to control foodborne pathogens.
