Full textPhosphorus is an essential element for life. This study focuses on the behaviours of phosphate minerals in natural waters. We have experimentally determined the dissolution and precipitation kinetic laws of the main phosphate minerals, at several temperatures and 1 atm. The studied phases were struvite (MgNH4PO4,6H2O), fluorapatite Ca5(PO4)3F, variscite (AlPO4,2H2O), strengite (FePO4,2H2O) and REE-phosphates as rhabdophane (LaPO4,2H2O and NdPO4,2H2O). These dissolution rates, normalized to a constant BET surface area, follow the approximate order struvite > fluorroapatite > variscite > rhabdophane. Since the phosphate group passes directly into the aqueous solution, it is reasonable to assume that the rate controlling process is related to the progressive disruption of metal oxygen bonds holding the PO4 tetrahedra together in the mineral structure. The precipitation rates were measured at different temperatures and acid conditions. During the experimental stages, aluminium and iron phosphates precipitated as amorphous phases, becoming crystalline as temperature rises and reaction time progresses. Rhabdophane, however, quickly precipitated directly from the solution as a crystalline phase. The low solubility and the big reactivity of phosphate minerals limit phosphate availability in natural waters. At constant solution composition, the precipitated phosphate-bearing solid phase depends on the induced pH conditions. At acid conditions, variscite is the phosphate dominant phase, while at moderately to high alkaline conditions apatite formation occurs. Further basic pH conditions combined with high ammonium concentrations results in struvite precipitation. , at medium pHs is apatite and at basic pH in presence of ammonium is struvite

Full textThe synthesis via aqueous medium of apatitic tricalcium phosphate Ca9(HPO4)(PO4)6(OH), which leads to anhydrous β TCP Ca3(PO4)2 above 700°C, was studied with the help of Doelhert experimental designs. Under an accurate control of temperature and pH, the experimental conditions allowing the reproducible synthesis of pure apatitic TCP are a temperature of 30°C, a pH between 6,5 and 7,0 and a ripening time of 10 hours. Characterization methods using ATD and DRX were required to validate the synthesis process and to control the purity of the as-synthesized powders by detection of very small quantities of secondary phases (β CPP, HA). Crystallographic changes of β TCP with temperature seems analogue to a progressive compensation of structural alterations affecting its hexagonal lattice at low temperature. Reversibility of the phenomenon involved is not insured without restrictive conditions of additional heat treatment. Flexural strength and indentation toughness of pure β TCP were measured on fully dense materials obtained by hot pressing for 30 minutes at 1100°C. Optimal values are, respectively, 85 MPa and 1,3 MPa. M1/2. Macroporous bone substitutes were prepared by immersion of polymeric foams in β TCP or biphasic mixture 65% HA/35% β TCP slurries. After milling, BCP and TCP slurries are shear thinning at low shear rate whereas they exhibit a linear rheological behaviour at high shear rates. After sintering at 1100°C for 2 hours, samples of β TCP were dense with a microstructure free from porosity for a total macroporosity of 74% and a compressive strength of 2,1 MPa. Samples of BCP exhibit, after sintering at 1350°C for 1 hour and a secondary firing step at 1000°C during 48 hours, a total porosity of 78,5% made of 80% of macropores and 20% of micropores, for a compressive strength of 0,4 MPa. Cellular materials obtained have a controlled and interconnected porosity: macropores diameters between 330 and 690 µm and interconnexion diameters between 50 and 250 µm. TCP and BCP processed parts have a specific area of, respectively, 0,9 m²/cm3 and 0,7 m²/cm3

Full textMoreover, we characterized the metabolic adaptations that breast cancer cells undergo in the deprivation of glutamine or when mitochondria are defected. We conducted metabolic flux analysis using metabolomics and fluxomics approaches and we employed Systems Biology approaches in order to estimate a global view of flux alterations in different culture conditions. We observed an increased pyruvate cycle with glutamine deprivation, thus indicating that targeting the enzymes of this pathway such as malic enzyme could be a promising approach combined with inhibition of glutaminase enzyme. On the other hand, we observed that mimicking hypoxia by oligomycin treatment redirected breast cancer cells to increase reductive carboxylation. Considering that hypoxia is a common condition in the tumor environment, targeting reductive carboxylation mechanism could be a novel strategy to fight against cancer. Collectively, all the results provided in this thesis demosntrate the importance of metabolism in cancer cell proliferation and survival. This work also highlights the importance of Systems Biology approaches to comprehend the molecular mechanisms underlying complex multifactorial diseases in order to point out new potential therapeutic targets

Full textThe mineral phase of bone is similar to a poorly crystallized carbonated apatite. The ageing of the population leads the surgeons to use synthetic bone substitutes. Among these biomaterials, there are Biphasic Calcium Phosphates (BCP), consisting in a mixture of hydroxyapatite (HA) and -tricalcium phosphate ( β-TCP). In a first step, thanks to NMR 31P and XRD, we detected the incorporation of sodium within the β-TCP lattice, also its localisation and influence on lattice parameters. . . . . . . . . . . . . This study contributes to use Rietveld refinement for the determination of lattice parameters, crystallite size as well as the percentage of HA and β-TCP phases in the BCP. This works allowed to answer to problems applied to this class of bioceramics (improvement of the ceramics mechanical properties, study of a method of synthesis to obtain bone substitute materials). Moreover, this study allows to better understand fundamental knowledge on crystallographic structures of calcium phosphate materials used as bone substitutes

Full textPhosphate (Pi) is a macroelement essential to plant development. During Pi deficiency, the expression of several genes is deregulated, allowing the plant to cope with this type of abiotic stress. In Arabidopsis thaliana, Pi deficiency strongly induces PS2;1 (At1g73010) and PS2;2 (At1g17710), two HAD-type phosphatases. The biochemical functions of these two proteins were previously characterized in vitro, although their in vivo roles have not yet been established.Here, the functions of these two proteins were examined in plants using reverse genetics approaches. Overexpression or inactivation of their expression levels had no effect on the Pi content of the plant cell, or on classic markers of Pi deficiency. Furthermore, PS2;1 and PS2;2 affect phosphocholine levels in planta (and potentially phosphoethanolamine), two compounds identified as their potential substrates by in vitro assays.In contrast, these findings do not suggest any involvement of PS2;1 or PS2;2 in the degradation of pyrophosphate, another potential substrate according to previous in vitro assays. In conclusion, these results suggest that PS2;1 and PS2;2 are involved in the remodeling of membrane lipids triggered by Pi deficiency, allowing the conversion of phospholipids to galactolipids or sulfolipids. Specifically, these enzymes should allow the recycling of Pi from phosphocholine and phosphoethanolamine polar heads, byproducts of the degradation of phospholipid phosphatidylcholine and phosphatidylethanolamine

Full textThe present thesis deals with the synthesis of new microporous gallophosphates and fluorogallophosphates. In this work, a new synthesis route for gallophosphate materials has been studied. It consists in the in situ generation of the structure-directing agent (SDA) from alkylformamide. These molecules decompose into alkylamines in the synthesis media. The in situ release of the amine during the synthesis appears to be a key step in the crystallization of the gallophosphate materials. Indeed, experiments using alkylamine in the starting mixture directly did not lead to the crystallization of the same materials. The in situ generation of the alkylamine has been very successful, several new materials have been obtained i. E. Mu-30, Mu-34, Ea-TREN GaPO, Mu-35, Mu-37 and Mu-38. The materials obtained were then characterized by powder XRD, SEM, elemental and thermal analyses and solid state NMR spectroscopy. Some structures were solved from single crystal XRD, or from XRD powder pattern by using Rietveld method

Full textSoluble phosphorus (P) fertilizers added to soil rapidly precipitate, forming sparingly soluble phosphates, not available to plants. Furthermore, the excessive use of chemical fertilizers to compensate soil P deficiency is not considered sustainable and it leads to costly and potentially harmful applications. Many reports confirmed that beneficial soil microorganisms, including phosphate-solubilizing bacteria (PSB), have a significant influence on soil fertility and crop productivity. Indeed, PSB can also improve phosphate rock (PR) efficiency when directly applied to soil. However, most published works on PSB overlooked the possible interaction between PSB and arbuscular mycorrhizal fungi (AMF), which are ubiquitous in cultivated plants. A new approach based on the trapping of PSB strongly attached to the hyphosphere of AMF Rhizophagus irregularis (Ri) DAOM 197198, previously inoculated with microbial soil suspensions was developed to isolate relevant PSB able to mobilize P from a low reactive igneous PR more efficiently than those directly isolated from the same rhizosphere soil samples. An in vitro study demonstrated that the synergism between hyphobacteria (Burkholderia anthina Ba8 and Rhizobium miluonense Rm3) and Ri hyphae highly improved the solubilisation of PR. Our results go beyond the existing studies and showed specific mechanisms involved on PSB-AMF interactions. Indeed, hyphobacteria, mainly B. anthina Ba8, strongly adhere to Ri hyphal surfaces and PR particles forming a structured biofilm. Under greenhouse conditions, the direct application of PSB and AMF Ri as biostimulants for sustainable corn production showed that these beneficial microorganisms improve growth and P uptake of corn fertilized with superphosphate or Quebec PR.

Full textThis work is devoted to the crystallographic study and the electrochemical and optical properties of cadmium phosphates and fluorophosphates. The investigation of the NaF - CdO - NaPO3 system revealed a new fluorophosphate of formula Na2CdPO4F and the polymorphism of the phosphate NaCdPO4. The crystal structures of these two phases were solved using XRD powder diffraction data, from materials obtained by a ceramic route. The characterization of the fluorophosphate Na2CdPO4F was also carried out by infrared spectroscopy, Raman scattering and Nuclear Magnetic Resonance. The electrochemical study, carried out on solid solutions of the Na2(Cd/Mn)PO4F type, revealed that the simultaneous presence of two divalent cations was beneficial for the improvement of the sodation/desodation rates. The luminescence properties of the red emitters NaCdPO4: Eu3+ and Na2CdPO4F: Eu3+ are closely correlated with the structural properties. Some formulations of NaCdPO4: Eu3+ type are of interest for application as red components in light emitting devices. The study of the LiF - CdO - NaPO3 system revealed the existence of an original fluorophosphate of formula Li4Na41Cd30P30O117F20. Its crystal structure, determined on monocrystal, is characterized by the presence of a novel entity PO3F2, where the phosphorus admits a five-fold coordination
