A recombinant thermostable lipase (Pf2001Δ60) from the hyperthermophilic ArchaeonPyrococcus furiosus(PFUL) was immobilized by hydrophobic interaction on octyl-agarose (octyl PFUL) and by covalent bond on aldehyde activated-agarose in the presence of DTT at pH = 7.0 (one-point covalent attachment) (glyoxyl-DTT PFUL) and on glyoxyl-agarose at pH 10.2 (multipoint covalent attachment) (glyoxyl PFUL). The enzyme’s properties, such as optimal temperature and pH, thermostability, and selectivity, were improved by covalent immobilization. The highest enzyme stability at 70°C for 48 h incubation was achieved for glyoxyl PFUL (around 82% of residual activity), whereas glyoxyl-DTT PFUL maintained around 69% activity, followed by octyl PFUL (27% remaining activity). Immobilization on glyoxyl-agarose improved the optimal temperature to 90°C, while the optimal temperature of octyl PFUL was 70°C. Also, very significant changes in activity with different substrates were found. In general, the covalent bond derivatives were more active than octyl PFUL. TheEvalue also depended substantially on the derivative and the conditions used. It was observed that the reaction of glyoxyl-DTT PFUL using methyl mandelate as a substrate at pH 7 presented the best results for enantioselectivityE=22and enantiomeric excess (ee (%) = 91).

Abstract:
Irrigation scheduling and programming are very effective tools for efficient water use in a plant factory with artificial lighting (PFAL). In order to confirm optimal irrigation schemes for the production of cucumber scions and rootstocks in a PFAL, in this study, four different start points of irrigation were applied by measuring the weight of the plug tray to compare the growth of cucumber scions and rootstocks cultivated in a PFAL. Additionally, the growth characteristics of cucumber seedlings grafted with scions and rootstocks cultivated between in a greenhouse and in a PFAL were investigated. Although the growth of cucumber scions and rootstocks was highest when irrigation was conducted at 70% of water content in a medium, the growth of grafted cucumber seedlings before and after transplanting was not significantly different among the irrigation treatments in a PFAL. However, water use efficiency (WUE) during cucumber scions and rootstock production in a PFAL was higher at 60% than at 70%. Considering seedling growth and the efficiency of irrigation such as WUE and irrigation schedule, the optimal start point of irrigation during the production of cucumber scions and rootstocks in a PFAL was determined as 60% of water content in a medium. When the optimal irrigation regime was applied to the production of cucumber scions and rootstocks in a PFAL, the morphological characteristics of cucumber scions and rootstocks cultivated in a PFAL were more suitable for grafting compared with that of the cucumber scions and rootstocks cultivated conventionally in a greenhouse. The favorable environmental conditions during the cultivation of cucumber scions and rootstocks in a PFAL also positively affected the flowering response of cucumber grafted seedlings after transplanting.

Pelvic fracture associated urethral injury (PFAUI) is a rare and challenging sequel of blunt pelvic trauma. Treatment of iatrogenic false urethral passage (FUP) remains as a challenge for urologists. In this case report we reviewed the iatrogenic FUP caused by wrong procedures performed in the treatment of a patient with PFAUI and the treatment of posterior urethral stricture with transperineal bulbo-prostatic anatomic urethroplasty in the management of FUP. A 37-year-old male patient with PFAUI had undergone a laparotomy procedure for pelvic bone fracture, complete urethral rupture, and bladder perforation 8 years ago. After stricture formation, patient had undergone procedures that caused FUP. Following operations, he had a low urinary flow rate, and incontinence and urgency even with small amounts of urine. FUP was diagnosed by voiding cystourethrography and retrograde urethrography. FUP was fixed with open urethroplasty with the guidance of flexible antegrade urethtoscopy. False passage should always be taken into account in the differential diagnosis of patients with persistent symptoms that underwent PFAUI therapy. In addition, we believe that in the evaluation of patients with PFAUI suspected for having a false passage, bladder neck and urethra should be assessed by combining routine voiding cystourethrography and retrograde urethrography with preoperative flexible cystoscopy via suprapubic route.

Abstract:
Purpose: Pneumonia is a serious respiratory disease among children with high mortality and morbidity all over the world. Long non-coding RNAs have been proven to play a vital role in many inflammatory diseases including pneumonia. In the present study, the protective impact of lncRNA PFAL on cell viability, cell apoptosis and secretion of inflammatory cytokines, as well as the underlying molecular mechanism in TNF-α-induced inflammatory injury model of pneumonia were investigated.Methods: WI-38 cell line was treated with 20 ng/ml TNF-α to establish an inflammatory injury model of pneumonia. LncRNA PFAL or miR-18a was up- or down-regulated in the WI-38 cells by transfection procedure. Cell viability was assessed using CCK-8 assay, while the rate of cell apoptosis was measured by utilizing flow cytometry. The mRNA expression levels of lncRNA PFAL, miR-18a, apoptosis-related and JNK pathway genes were determined with RT-qPCR. Moreover, the production of inflammatory cytokines such as IL-6 and MCP-1 were detected by using Western blot analysis.Results: The results indicated that cell viability was significantly (P<0.05) reduced, while the rate of cell apoptosis was increased in the TNF-α-induced WI-38 cells. Also, TNF-α treatment enhanced the expression of inflammatory cytokines that included IL-6 and MCP-1 in WI-38 cells. Overexpression of PFAL suppressed the injury induced by TNF-α and miR-18a was positively regulated by PFAL. Moreover, the inhibition of miR-18a weakens the effect of PFAL overexpression in TNF-α-induced cell injury. Furthermore, PFAL and miR-18a were involved in the regulation of JNK pathway.Conclusion: Overexpression of PFAL suppresses TNF-α-induced WI-38 cell injury by up-regulating miR-18a via the inactivation of JNK signaling pathway. Purpose The product family assembly line (PFAL) is a mixed model-assembly line, which is widely used in mass customization and intelligent manufacturing. The purpose of this paper is to study the problem of PFAL, a flexible (evolution) planning method to respond to product evolution for PFAL, to focus on product data analysis and evolution planning method. Design/methodology/approach The evolution balancing model for PFAL is established and an improved NSGA_II (INSGA_II) is proposed. From the perspective of data analysis, dynamic characteristics of PFAL are researched and analyzed. Especially the tasks, which stability is considered, can be divided into a platform and individual task. In INSGA_II algorithm, a new density selection and a decoding method based on sorting algorithms are proposed to compensate for the lack of traditional algorithms. Findings The effectiveness and feasibility of the method are validated by an example of PFAL evolution planning for a family of similar mechanical products. The optimized efficiency is significantly improved using INSGA_II proposed in this paper and the evolution planning model proposed has a stronger ability to respond to product evolution, which maximizes business performance over an effective period of time. Originality/value The assembly line designers and managers in discrete manufacturing companies can obtain an optimal solution for PFAL planning through the evolution planning model and INSGA-II proposed in this paper. Then, this planning model and optimization method have been successfully applied in the production of small wheel loaders.

Assembly line balancing not only directly determines production efficiency but also influences precision and quality of key assemblies or even the overall performance of final products. Driven by market demand and development of science and technology, product family must evolve constantly, which necessitates frequent adjustment and rebalancing of product family assembly line (PFAL). In order to maintain production efficiency, improve assembly quality and precision, and reduce costs for adjustment, the evolution balancing problem of PFAL for small-sized wheel loader is solved in this paper. Firstly, the evolution balancing model of PFAL is put forward. Then, with minimizing the number of workstations, the in-station and between-station load indexes and adjustment costs, and maximizing relevancy between activities as optimization objectives, meanwhile regarding product platform planning, modularity design and the critical chain technology in concurrent engineering as constraint conditions, the evolution balancing problem of PFAL is optimized using improved genetic algorithm (IGA). Finally, the whole analysis procedure is demonstrated by the small-sized wheel loader PFAL case study and the effectiveness of the proposed method is verified.

Abstract. Positive Feedback Adiabatic Logic (PFAL) shows the lowest energy dissipation among adiabatic logic families based on cross-coupled transistors, due to the reduction of both adiabatic and non-adiabatic losses. The dissipation primarily depends on the resistance of the charging path, which consists of a single p-channel MOSFET during the recovery phase. In this paper, a new logic family called Improved PFAL (IPFAL) is proposed, where all n- and pchannel devices are swapped so that the charge can be recovered through an n-channel MOSFET. This allows to decrease the resistance of the charging path up to a factor of 2, and it enables a significant reduction of the energy dissipation. Simulations based on a 0.13µm CMOS process confirm the improvements in terms of power consumption over a large frequency range. However, the same simple design rule, which enables in PFAL an additional reduction of the dissipation by optimal transistor sizing, does not apply to IPFAL. Therefore, the influence of several sources of dissipation for a generic IPFAL gate is illustrated and discussed, in order to lower the power consumption and achieve better performance.

To verify the productivity of the autotrophic transplant production method (ATPM), a novel propagation method in a plant factory with artificial lighting for transplant production (T-PFAL), strawberry transplants were produced by the ATPM for 365 days. A total of 3497 transplants were produced by the ATPM over 365 days with nine propagules in a cultivation area of 3.6 m2 in the T-PFAL. When the simulated results were fit with the measured results, the propagation cycle timescales from planting propagules to producing the first, second, and third runner plants were 15, 27, and 43 days, respectively. The cumulative number of transplants (CNT) produced from 5, 10, and 20 initial propagules in a cultivation area of 36 m2 over 365 days was simulated by the verified program along with the propagation cycles, and these values were 27,970, 30,010, and 31,900, respectively. The simulated CNTs from nine initial propagules in 18 and 72 m2 over 365 days were 15,950 and 55,940, respectively. These results indicate that the ATPM is an appropriate propagation method to produce transplants rapidly in a T-PFAL, especially when the number of propagules or propagules is limited.

Classification analysis of grain yield data identified 9 genotypic groups and 5 environmental groups with similar patterns in yield performance within each group. Genotypes with similar origin and pedigree were similar in yield performance across environments in Australia and the WANA region. Environmental conditions across both regions showed similarity in discriminating genotypes for their yield performance. Genotypes Attila, Spear, Excalibur, and 82Y:1186 were highest yielding in Australia and the WANA region. However, genotypes Nesser, Pfau/Seri//Bow, Hartog, Vulcan, and Sunland showed wide adaptation across both regions. Genotypes Cranbrook, Genaro 81, Seri 82, Kauz, SUN 190A, and Pgo/Seri 82 showed specific adaptation to favourable environments. Genotypes in each group, based on yield performance, showed differing degrees of yield stability. Pfau/Seri//Bow has the potential for release for commercial production in Australia and the WANA region. The genotypes Pfau/Seri//Bow, Hartog, Sunland, and Vulcan could be used in trials in both regions as indicator varieties to evaluate new breeding lines for mutual exchange between the 2 sets of breeding programs.

Previously, authors have proposed CPLAG and MCPLAG circuits extracting benefits of CPL family implemented based upon semiadiabatic logic for low power VLSI circuit design along with gating concept. Also authors have communicated RCPLAG circuits adding another dimension of reconfigurability into CPLAG/MCPLAG circuits. Moving ahead, in this paper, authors have implemented/reconfigured RCPLAG universal Nand/And gate and universal Nor/Or gate for extracting behavior of dynamic positive edge triggered DFF. Authors have also implemented Adder/Subtractor circuit using different techniques. Authors have also reported modification in PFAL semiadiabatic circuit family to further reduce the power dissipation. Functionality of these is verified and found to be satisfactory. Further these are examined rigorously with voltage, Cload, temperature, and transistor size variation. Performance of these is examined with these variations with power dissipation, delays, rise, and fall times associated. From the analysis it is found that best operating condition for DFF based upon RCPLAG universal gate can be achieved at supply voltage lower than 3 V which can be used for different transistor size up to 36 μm. Average power dissipation is 0.2 μW at 1 V and 30 μW at 2 V at 100 ff Cload 25°C approximately. Average power dissipated by CPLAG Adder/Subtractot is 58 μW. Modified PFAL circuit reduces average power by 9% approximately.

Abstract German Sign Language (DGS) displays variation in the simple plural, the form of which is conditioned by classes of phonological features within the lexicon. Pfau & Steinbach (2005) report a number of ‘alternative pluralization strategies’ available as repairs for this underspecification, including classifier constructions, spatial localization, and number and quantifier phrases. I propose a previously undescribed mechanism for plural marking, the ‘classifier-based plural morpheme’ (CLP), grammaticalized from the classifier system into a morpheme in the grammars of individual DGS signers. Elicitation data show that this morpheme attaches only to nouns which are specified for phonological features that restrict the realization of the canonical plural marker, adding a new option to the range of pluralization strategies available.

Lettuce is one of the most suitable plant species to grow indoor or in a Plant Factory with Artificial lighting (PFAL) system, due to its short height and the relatively low light saturation point. Red lettuces are highly cultivated in PFALs, highly appreciated for its unique pigmentation and antioxidant activity, especially, the anthocyanin content that gives it the red color. However, red lettuces cultivated with PFAL in controlled environments face the challenge of having low anthocyanin content due of the lack of ultraviolet (UV) light that is necessary for its production. In this study, two red-lettuce cultivars ‘Annapolis’ and ‘Salanova’ were grown in a plant factory with LEDs (light-emitting diode) and UV light treatments to compare their growth trajectory and nutritional characteristics. Other characteristics such as Fresh Weight (FW) and total leaf area were measured to evaluate the influence of UV light in the lettuce growth. The nutritional quality of lettuces was assessed by measuring anthocyanins and polyphenols contents using a microplate reader and their ascorbic acid content measured via a reflectometer. Fresh Weight (FW), leaf area and leaf thickness under the conditions of this study, showed no effect (p>0.05) on the plants growth with the incorporation of UV radiation. However, the incorporation of UV light increased the functional components in ‘Annapolis’ and ‘Salanova’ lettuces. The polyphenol and anthocyanins contents in both lettuce cultivars showed significant differences (p<0.05). For ascorbic acid content, a significant difference was found only for ‘Annapolis’ lettuce. The incorporation of UV light can increase the functional ingredients such as polyphenols and anthocyanins without growth suppression for ‘Salanova’ cultivar. UV light increased polyphenol and anthocyanins contents in both cultivars, making them suitable cultivars for PFAL under the growth conditions used in this experiment. These results taken together can be used to improve the accumulation of functional ingredients in red lettuces without growth suppression incorporating UV light and that could be suitable for production in PFALs.

Avaliaram-se a influência do ambiente em 20 genótipos de trigo, no rendimento e qualidade de grãos, a adaptabilidade e a reação a doenças, em regiões onde a irrigação se faz necessária para permitir o bom desempenho agronômico de um genótipo. Instalaram-se os experimentos em Tatuí, Votuporanga, Ribeirão Preto e Mococa, Estado de São Paulo, no período de 1995-98. Avaliaram-se, também, a tolerância dos genótipos ao alumínio tóxico em solução nutritiva e a qualidade industrial para panificação. Com bom rendimento de grãos destacaram-se: em Tatuí, os genótipos IAC 351, IAC 335, IAC 289 e Mochis; em Votuporanga, IAC 289, TUI"S" e SERI*3/BUC; em Ribeirão Preto e Mococa, o IAC 370. No conceito de genótipo ideal, o IAC 370 apresentou alta capacidade produtiva, foi responsivo à melhoria do ambiente e sensível às condições desfavoráveis do ambiente. Entre as doenças, a ferrugem-da-folha foi a de abrangência generalizada com maior incidência em Tatuí. Anahuac, IAC 287, CAL/CHKW//VEE"S" e IAC 370 foram os mais suscetíveis ao oídio. Os genótipos Anahuac, IAC 287, JCAM//EMU"S"/YACO"S", PFAU e IAC 339 foram os mais sensíveis à toxicidade por Al3+. Os genótipos Anahuac, IAC 24, IAC 287, IAC 289, IAC 334, PFAU, TUI"S", IAC 339, IAC 370 e IAC 351 apresentaram características de farinha com bom potencial para panificação.

This article examines the work of a diverse group of photographers who in the late 1960s and 1970s employed mapping techniques and devices as a means of artistic creation. Products of photography’s unprecedented growth, photographers John Pfahl, Michael Bishop, Kenneth Josephson and the participants of the Rephotographic Survey Project employed cartographic and topographic strategies as part of their exploration of the history of their medium and the American West. These artist-photographers, moreover, responded to the nineteenth-century surveys of the West as well as its relation to other, better-known contemporary movements like ‘New Topographics’. In all, this article provides the first exploration of this distinctive group of American photographers which may be collectively termed: ‘new cartographics’.

Abstract. For the first time carbon nanotube (CNT) transistor based adiabatic logic (AL) was analyzed in this work and compared to CNT based static CMOS (CCNT). Static CCNT inverters are used as a reference and compared to inverters in the AL families Efficient Charge Recovery Logic (ECRL) and Positive Feedback Adiabatic Logic (PFAL) in terms of energy dissipation. Energy savings by adiabatic logic in dependence of operating frequency, supply voltage and number of nanotubes per transistor are reviewed. It is shown that CNT based AL circuits provide high energy saving factors even for high frequencies compared to CNT based static CMOS circuits.

We make a detailed study on carrier sensing of 802.11 in Nakagami fading channels. We prove that to maximize sensing accuracy, the optimal channel accessing probability is solely determined by the path-loss SIR (Signal to Interference Ratio). We define pfail-interference range and pbusy-carrier sensing range for fading channels and prove that their scaling laws in Nakagami fading channels are similar to those in the static channel. The newly derived theoretical results show a unified property between the static and fading channels. By extensive simulations, we reveal that fading depresses the probability of a dominating transmission state, and therefore it can mitigate severe hidden and exposed terminal problems, but fading harms the average sensing accuracy for an optimally adjusted carrier sensing threshold.
