Abstract:
A recombinant thermostable lipase (Pf2001Δ60) from the hyperthermophilic ArchaeonPyrococcus furiosus(PFUL) was immobilized by hydrophobic interaction on octyl-agarose (octyl PFUL) and by covalent bond on aldehyde activated-agarose in the presence of DTT at pH = 7.0 (one-point covalent attachment) (glyoxyl-DTT PFUL) and on glyoxyl-agarose at pH 10.2 (multipoint covalent attachment) (glyoxyl PFUL). The enzyme’s properties, such as optimal temperature and pH, thermostability, and selectivity, were improved by covalent immobilization. The highest enzyme stability at 70°C for 48 h incubation was achieved for glyoxyl PFUL (around 82% of residual activity), whereas glyoxyl-DTT PFUL maintained around 69% activity, followed by octyl PFUL (27% remaining activity). Immobilization on glyoxyl-agarose improved the optimal temperature to 90°C, while the optimal temperature of octyl PFUL was 70°C. Also, very significant changes in activity with different substrates were found. In general, the covalent bond derivatives were more active than octyl PFUL. TheEvalue also depended substantially on the derivative and the conditions used. It was observed that the reaction of glyoxyl-DTT PFUL using methyl mandelate as a substrate at pH 7 presented the best results for enantioselectivityE=22and enantiomeric excess (ee (%) = 91).

Abstract:
Irrigation scheduling and programming are very effective tools for efficient water use in a plant factory with artificial lighting (PFAL). In order to confirm optimal irrigation schemes for the production of cucumber scions and rootstocks in a PFAL, in this study, four different start points of irrigation were applied by measuring the weight of the plug tray to compare the growth of cucumber scions and rootstocks cultivated in a PFAL. Additionally, the growth characteristics of cucumber seedlings grafted with scions and rootstocks cultivated between in a greenhouse and in a PFAL were investigated. Although the growth of cucumber scions and rootstocks was highest when irrigation was conducted at 70% of water content in a medium, the growth of grafted cucumber seedlings before and after transplanting was not significantly different among the irrigation treatments in a PFAL. However, water use efficiency (WUE) during cucumber scions and rootstock production in a PFAL was higher at 60% than at 70%. Considering seedling growth and the efficiency of irrigation such as WUE and irrigation schedule, the optimal start point of irrigation during the production of cucumber scions and rootstocks in a PFAL was determined as 60% of water content in a medium. When the optimal irrigation regime was applied to the production of cucumber scions and rootstocks in a PFAL, the morphological characteristics of cucumber scions and rootstocks cultivated in a PFAL were more suitable for grafting compared with that of the cucumber scions and rootstocks cultivated conventionally in a greenhouse. The favorable environmental conditions during the cultivation of cucumber scions and rootstocks in a PFAL also positively affected the flowering response of cucumber grafted seedlings after transplanting.

Abstract:
Pelvic fracture associated urethral injury (PFAUI) is a rare and challenging sequel of blunt pelvic trauma. Treatment of iatrogenic false urethral passage (FUP) remains as a challenge for urologists. In this case report we reviewed the iatrogenic FUP caused by wrong procedures performed in the treatment of a patient with PFAUI and the treatment of posterior urethral stricture with transperineal bulbo-prostatic anatomic urethroplasty in the management of FUP. A 37-year-old male patient with PFAUI had undergone a laparotomy procedure for pelvic bone fracture, complete urethral rupture, and bladder perforation 8 years ago. After stricture formation, patient had undergone procedures that caused FUP. Following operations, he had a low urinary flow rate, and incontinence and urgency even with small amounts of urine. FUP was diagnosed by voiding cystourethrography and retrograde urethrography. FUP was fixed with open urethroplasty with the guidance of flexible antegrade urethtoscopy. False passage should always be taken into account in the differential diagnosis of patients with persistent symptoms that underwent PFAUI therapy. In addition, we believe that in the evaluation of patients with PFAUI suspected for having a false passage, bladder neck and urethra should be assessed by combining routine voiding cystourethrography and retrograde urethrography with preoperative flexible cystoscopy via suprapubic route.

Abstract:
Purpose: Pneumonia is a serious respiratory disease among children with high mortality and morbidity all over the world. Long non-coding RNAs have been proven to play a vital role in many inflammatory diseases including pneumonia. In the present study, the protective impact of lncRNA PFAL on cell viability, cell apoptosis and secretion of inflammatory cytokines, as well as the underlying molecular mechanism in TNF-α-induced inflammatory injury model of pneumonia were investigated.Methods: WI-38 cell line was treated with 20 ng/ml TNF-α to establish an inflammatory injury model of pneumonia. LncRNA PFAL or miR-18a was up- or down-regulated in the WI-38 cells by transfection procedure. Cell viability was assessed using CCK-8 assay, while the rate of cell apoptosis was measured by utilizing flow cytometry. The mRNA expression levels of lncRNA PFAL, miR-18a, apoptosis-related and JNK pathway genes were determined with RT-qPCR. Moreover, the production of inflammatory cytokines such as IL-6 and MCP-1 were detected by using Western blot analysis.Results: The results indicated that cell viability was significantly (P<0.05) reduced, while the rate of cell apoptosis was increased in the TNF-α-induced WI-38 cells. Also, TNF-α treatment enhanced the expression of inflammatory cytokines that included IL-6 and MCP-1 in WI-38 cells. Overexpression of PFAL suppressed the injury induced by TNF-α and miR-18a was positively regulated by PFAL. Moreover, the inhibition of miR-18a weakens the effect of PFAL overexpression in TNF-α-induced cell injury. Furthermore, PFAL and miR-18a were involved in the regulation of JNK pathway.Conclusion: Overexpression of PFAL suppresses TNF-α-induced WI-38 cell injury by up-regulating miR-18a via the inactivation of JNK signaling pathway. Purpose The product family assembly line (PFAL) is a mixed model-assembly line, which is widely used in mass customization and intelligent manufacturing. The purpose of this paper is to study the problem of PFAL, a flexible (evolution) planning method to respond to product evolution for PFAL, to focus on product data analysis and evolution planning method. Design/methodology/approach The evolution balancing model for PFAL is established and an improved NSGA_II (INSGA_II) is proposed. From the perspective of data analysis, dynamic characteristics of PFAL are researched and analyzed. Especially the tasks, which stability is considered, can be divided into a platform and individual task. In INSGA_II algorithm, a new density selection and a decoding method based on sorting algorithms are proposed to compensate for the lack of traditional algorithms. Findings The effectiveness and feasibility of the method are validated by an example of PFAL evolution planning for a family of similar mechanical products. The optimized efficiency is significantly improved using INSGA_II proposed in this paper and the evolution planning model proposed has a stronger ability to respond to product evolution, which maximizes business performance over an effective period of time. Originality/value The assembly line designers and managers in discrete manufacturing companies can obtain an optimal solution for PFAL planning through the evolution planning model and INSGA-II proposed in this paper. Then, this planning model and optimization method have been successfully applied in the production of small wheel loaders.

Assembly line balancing not only directly determines production efficiency but also influences precision and quality of key assemblies or even the overall performance of final products. Driven by market demand and development of science and technology, product family must evolve constantly, which necessitates frequent adjustment and rebalancing of product family assembly line (PFAL). In order to maintain production efficiency, improve assembly quality and precision, and reduce costs for adjustment, the evolution balancing problem of PFAL for small-sized wheel loader is solved in this paper. Firstly, the evolution balancing model of PFAL is put forward. Then, with minimizing the number of workstations, the in-station and between-station load indexes and adjustment costs, and maximizing relevancy between activities as optimization objectives, meanwhile regarding product platform planning, modularity design and the critical chain technology in concurrent engineering as constraint conditions, the evolution balancing problem of PFAL is optimized using improved genetic algorithm (IGA). Finally, the whole analysis procedure is demonstrated by the small-sized wheel loader PFAL case study and the effectiveness of the proposed method is verified.
