Over the years, cyclodextrin uses have been widely reviewed and their proprieties provide a very attractive approach in different biomedical applications. Cyclodextrins, due to their characteristics, are used to transport drugs and have also been studied as molecular chaperones with potential application in protein misfolding diseases. In this study, we designed cyclodextrin polymers containing different contents of β- or γ-cyclodextrin, and a different number of guanidinium positive charges. This allowed exploration of the influence of the charge in delivering a drug and the effect in the protein anti-aggregant ability. The polymers inhibit Amiloid β peptide aggregation; such an ability is modulated by both the type of CyD cavity and the number of charges. We also explored the effect of the new polymers as drug carriers. We tested the Doxorubicin toxicity in different cell lines, A2780, A549, MDA-MB-231 in the presence of the polymers. Data show that the polymers based on γ-cyclodextrin modified the cytotoxicity of doxorubicin in the A2780 cell line.

Protein aggregation into highly ordered supramolecular aggregates is the hallmark of many degenerative diseases including the neurological disorders (Parkinson’s, Alzheimer’s, and Huntington’s diseases), type II diabetes, systemic amyloidosis, spongiform encephalopathies, etc. One of the simplest and effective methods for the identification and characterization of amyloid fibrils in vitro and the visualization of amyloid inclusions in vivo is based on the use of probes sensitive to the beta-pleated motifs. In the attempt to design new amyloid-sensing dyes or to optimization the existing molecules, it is crucial to have the sufficient knowledge of the molecular and atomic levels interactions in the binding sites. Among the especially useful methods available to provide the atomic-level insights into the mechanisms of various types of biomolecular interactions is molecular docking technique. In the present study, the molecular docking tool has been employed to investigate the interactions between the monomethine cyanine dyes and the lysozyme amyloid fibrils constructed from the K-peptide of lysozyme, GILQINSRW (residues 54–62 of the wild-type protein). Using the AutoDOCK and the protein-ligand interaction profiler PLIP it was found: i) monomethines interact with the fibril surface (with the aromatic residues on the top of β-sheet or with the edges of the β-sheet); ii) the dye binding is governed by the hydrophobic interactions, salt bridges and the hydrogen bonds between the aliphatic substituents on the nitrogen atom of benzothiazole part of dye molecules and the lysozyme amyloid fibril; iii) the variations in the cyanine structure and in the lysozyme amiloid twisting didn’t insert significant effect on the binding mode of cyanines.


 La actividad física (AF) afecta de manera positiva en la salud de las personas a cualquier edad, de acuerdo con los resultados de diferentes estudios en cuanto a la salud cognitiva de las personas mayores, los beneficios son más evidentes. Por esa razón, se ha realizado una revisión sistemática de la literatura científica que aborda los efectos de la AF en las funciones cognitivas (FC) de las personas mayores. Se consultaron cuatro bases de datos: Scopus, Web of Science, ScienceDirect, y Pub Med, y el buscador Scholar Google. Tras aplicar los criterios de selección, quedaron 31 artículos, que fueron analizados en cuanto a) país, cantidad de individuos y edad, b) variables, y c) enfoque de investigación, duración y resultados. En general, la AF mejora diversas dimensiones de las funciones cognitivas de las personas mayores, donde median factores genéticos y ambientales que tienen que ver con el estilo de vida del individuo. Los tipos de AF se comportan de manera diferencial en cuanto a la dimensión de la función cognitiva que mejoran. La determinación de marcadores bioquímicos (BDNF, dopamina, péptido β-amiloide, gamma gap), de imagenología por resonancia magnética nuclear (RMN) y cambios en la actividad eléctrica del cerebro, soportan la hipótesis de que la AF produce cambios en la forma y actividad cerebral, sin que ello suponga cambios significativos en la conducta observada y medible mediante test estandarizados de salud mental. 
 Abstract. Physical activity (PA) positively affects people's health at any age, and according to the results of different studies on the cognitive health of older people, the benefits are more evident. For this reason, a systematic review of the scientific literature addressing the effects of PA on the cognitive functions (CF) of older people has been conducted. Four databases were consulted to locate relevant literature: Scopus, Web of Science, Science Direct and Pub Med, and the Scholar Google search engine. After applying the selection criteria, 31 articles were left, which were analyzed in terms of a) country, number of individuals and age, b) variables, and c) research approach, duration and results. In general, PA improves several dimensions of cognitive functions of older people, where genetic and environmental factors that have to do with the individual's lifestyle are mediated. Types of PA behave differently in terms of the dimension of cognitive function they improve. The determination of biochemical markers (BDNF, dopamine, amyloid peptide β, gamma gap), nuclear magnetic resonance imaging (NMR) and changes in the electrical activity of the brain support the hypothesis that PA produces changes in brain shape and activity, without significant changes in behavior observed and measurable by standardized tests of mental health.

AbstractObjectivesAlzheimer’s disease is complex and involves several proteins. Most affected are Tau protein and amyloid precursor protein (APP) which, when cleaved by the enzymes β-secretase (BACE1) and γ-secretase (Nicastrin), yield the amyloid peptide. Although these processes take place mainly in neurons, it is not exclusive of them, as glia cells also contribute to these processes. The objective of this study was to evaluate the effect of nimesulide, resveratrol and citalopram on C6 (glioma) cells when exposed to Lipopolysaccharide (LPS).MethodsExpression levels of the proteins APP, BACE1, COX-2, Nicastrin and Tau-p were evaluated by Western-blot and ELISA in C6 cells by effect of LPS, and the drugs citalopram, nimesulide and resveratrol.ResultsIt was found that LPS is able to hyperphosphorylate Tau in this cell model and the drugs decrease hyperphosphorylation. We also found that the drugs increase the expression of APP, decrease BACE1 and promote the expression of Nicastrin. COX-2 decreases its expression when nimesulide is used.ConclusionsOur results suggest that C6 cell line is useful to analyze the effect of pro-inflammatory molecules on tau phosphorylation and APP expression in vitro. The beneficial effect on the reduction of tau hyperphosphorylation shown by citalopram, nimesulide and resveratrol should be taken with caution due to the limitations of the present study and further research on these compounds is needed to determine their therapeutic use in neurodegenerative diseases such as Alzheimer’s disease.AmaçAlzheimer hastalığı karmaşıktır ve birkaç protein içerir. En çok etkilenenler, u-sekretaz (BACE1) ve γ-sekretaz (Nicastrin) enzimleri tarafından parçalandığında amiloid peptidi veren Tau proteini ve amiloid öncü proteinidir (APP). Bu süreçler esas olarak nöronlarda gerçekleşmesine rağmen, glia hücreleri de bu süreçlere katkıda bulunduğundan, bunlardan ayrı değildir. Amaç: Bu çalışmanın amacı, LPS’ye maruz kaldığında nimesulid, resveratrol ve sitalopramın C6 (glioma) hücreleri üzerindeki etkisini değerlendirmektir.Gereç ve YöntemAPP, BACE1, COX-2, Nicastrin ve Tau-p proteinlerinin ekspresyon seviyeleri, LPS’nin etkisi ile C6 hücrelerinde Western-blot ve ELISA ve sitalopram, nimesulid ve resveratrol ilaçları ile değerlendirildi.BulgularBu hücre modelinde LPS’nin Tau’yu hiperfosforilat edebildiği ve ilaçların hiperfosforilasyonu azalttığı bulundu. Ayrıca ilaçların APP ekspresyonunu arttırdığını, BACE1’i azalttığını ve Nicastrin ekspresyonunu teşvik ettiğini bulduk. Nimesulid kullanıldığında COX-2 ekspresyonunu azaltır.SonuçlarSonuçlarımız, pro-enflamatuar moleküllerin tau fosforilasyonu ve in vitro APP ekspresyonu üzerindeki etkisini analiz etmek için C6 hücre hattının yararlı olduğunu göstermektedir. Sitalopram, nimesulid ve resveratrol tarafından gösterilen tau hiperfosforilasyonunun azaltılması üzerindeki yararlı etki, mevcut çalışmanın sınırlamaları nedeniyle dikkatle alınmalı ve bu bileşikler üzerinde, Alzheimer hastalığı gibi nörodejeneratif hastalıklarda terapötik kullanımlarının belirlenmesi için daha fazla araştırmaya ihtiyaç vardır.

Nella patogenesi della malattia di Alzheimer, il livello di espressione della proteina precursore del peptide β-amiloide (APP) riveste un ruolo determinante. E’ stato evidenziato che mutazioni che incrementano la sintesi proteica di APP, inclusa la duplicazione genica del locus di APP in alcune famiglie e nella trisomia del cromosoma 21, che contiene il gene APP, sono sufficienti per indurre lo sviluppo della patologia in forma precoce. Per questo motivo, la regolazione del gene APP è stata oggetto di intensi studi riguardanti sia la caratterizzazione del promotore, che le regioni 5’ e 3’ non tradotte dell’RNA messaggero. Negli ultimi anni, il meccanismo di regolazione post-trascrizionale dell’espressione genica mediato dai microRNA è stato estesamente indagato in diversi meccanismi biologici e in vari tipi cellulari. I microRNA modulano finemente la traduzione di proteine necessarie alla risposta a stimoli di origine extracellulare, ed in altri casi definiscono spazialmente e temporalmente i profili di espressione di geni coinvolti nello sviluppo e differenziamento. Lo svolgimento di questa tesi sperimentale ha trovato il suo avvio nella domanda se il sistema dei microRNA fosse coinvolto in maniera significativa nella regolazione dell’espressione dell’APP. Tramite RNAi, è stata silenziato il gene Argonauta 2, componente fondamentale del complesso ribonucleoproteico che media l’azione dei microRNA, ed è stato osservato un aumento della proteina APP. Mediante analisi bioinformatica sono stati individuati i possibili microRNA interagenti con APP ed è stato scelto il miR-101, arricchito nel sistema nervoso. Tramite analisi dei profili di espressione in ippocampo di ratto, tra gli 8 giorni e i 6 mesi di età, è stata osservata una correlazione inversa tra i livelli di miR-101, che aumenta, e APP, che diminuisce nel corso del tempo. Utilizzando un costrutto reporter, è stata dimostrata la capacità del miR-101 di inibire la traduzione di APP attraverso l’interazione diretta con il 3’UTR del messaggero. Bloccando l’azione del miR-101 in colture primarie ippocampali, è stato osservato un significativo aumento della proteina APP endogena, confermando l’azione regolatoria del microRNA nel contesto neuronale fisiologico, mentre la over-espressione del miR-101 si è rivelata efficace nell’indurre una repressione a livello post-trascrizionale dell’APP. E’ stato inoltre osservato che la stimolazione di neuroni ippocampali con Interleuchina-1β, principale mediatore della neuroinfiammazione e fattore di rischio per AD, induce una modulazione coordinata nel tempo di APP e del miR-101. I dati ottenuti dimostrano quindi l’esistenza di un’attività repressiva del miR-101 sull’APP in condizioni fisiologiche, mentre la perturbazione di tale regolazione è sufficiente ad indurre un aumento della proteina APP, condizione associata alla patologia di Alzheimer. Considerando studi recenti, in cui il miR-101 è stato visto down-regolato in cervelli di casi sporadici di malattia di Alzheimer, e i dati qui ottenuti, miR-101 può essere visto come un regolatore con attività protettiva verso una eccessiva produzione del precursore del peptide β-amiloide. La caratterizzazione della via di trasduzione del segnale, neuroinfiammatorio, che coinvolge il miR-101, e la sua possibile associazione con la patologia di Alzheimer potrebbero aprire nuovi scenari nella eziopatogenesi di questa malattia neurodegenerativa.<br>In the pathogenesis of Alzheimer's disease, the expression level of the β-amyloid precursor protein (APP) plays a crucial role. It has been shown that mutations which increase APP protein synthesis, including the duplication of the locus of APP gene in certain families and trisomy of chromosome 21 (containing the APP gene), are sufficient to induce the early form of pathology. Therefore APP gene regulation has been extensively studied and both the promoter and 5’ and 3’ untranslated regions of mRNA have been charachterised. In recent years, the post-transcriptional mechanism of gene expression mediated by microRNAs has been discovered and investigated in several biological mechanisms and cell types. MicroRNAs have been shown to finely modulate the translation of proteins needed for the response to extracellular stimuli, and to define the spatial and temporal expression profiles of genes involved in neuronal development and differentiation. This experimental thesis focused on the question whether microRNAs are significantly involved in APP regulation. Through RNAi , Argonauta-2 gene, a key component of the ribonucleoprotein complex mediating the action of microRNAs, was silenced and APP protein increase was observed. Through bioinformatic analysis, microRNAs potentially interacting with APP have been identified and miR-101, which is enriched in the nervous system, was further investigated. Through expression analysis in rat hippocampus, from 8 days up to 6 months of age, it was observed an inverse correlation between miR-101 levels, which increase, and APP expression, which decreases over time. Using a reporter construct, the ability of miR-101 to inhibit the translation of APP through direct interaction with the messenger 3’UTR was demonstrated. Through inhibition of miR-101 in hippocampal primary cultures, a significant increase of endogenous APP protein was observed, confirming the miRNA regulatory function in the neuronal context. Instead miR-101 over-expression was effective in inducing APP repression at post-transcriptional level. It was also observed that stimulation of hippocampal neurons with IL-1, the first player of neuroinflammation and risk factor for Alzheimers Disease, induces coordinated modulation of APP and miR-101. These data demonstrate the repressive effect of miR-101 on APP under physiological conditions, while the alteration of this regulation is sufficient to induce an increase of APP protein, a condition which is associated to familial Alzheimer’s disease. In view of previous studies, in which miR-101 was shown to be down-regulated in the brains of sporadic cases of Alzheimer's disease, and of data obtained in this work, miR-101 may be a protective regulator of excessive production of β-amyloid precursor protein. The characterization of the signal transduction pathway activated by neuroinflammation, involving miR-101, and the relationship between this pathway and Alzheimer's disease might open a new perspective on the pathogenesis of this neurodegenerative disease.

In questa tesi sono stati affrontati tre progetti indipendenti, accomunati dall’uso della modellistica molecolare e in particolare della dinamica molecolare. Nel primo progetto è stato studiato il dominio Sec14-PH della neurofibromina (NF1). I domini sec14 sono stati scoperti in numerose proteine dai procarioti all’uomo come scambiatori di lipidi tra membrane, per mezzo di una tasca la cui apertura è legata al movimento di una specifica alpha-elica (elica lid). La struttura cristallina del dominio Sec14 di NF1 (sia del wild type sia di mutanti associati all’insorgenza della patologia neurofibromatosi) ha rivelato la sua particolarità di essere strutturalmente accoppiato ad un dominio PH che interagisce fortemente con l’elica lid tramite un suo loop (detto lid-lock loop). Su questa base è stato formulato un meccanismo di apertura della tasca del Sec14 che coinvolgerebbe un movimento concertato del lid-lock loop, ma questo movimento non è mai stato osservato o dimostrato. Guidati da dati sperimentali sulla denaturazione termica del Sec14-PH di NF1, sia del wild type sia di alcuni mutanti, diverse simulazioni ad alta temperatura sono state effettuare per comparare la dinamica del dominio wild type con un mutante patologico associato all’insorgenza della patologia neurofibromatosi. Con le nostre simulazioni è stato possibile proporre un meccanismo di funzionamento dell’apertura dell’elica lid e fornire delle basi strutturali e dinamiche dell’insorgenza della patologia nel caso del mutante specifico studiato. Nel secondo progetto è stato affrontato lo studio di una proteina chiamata EtfAB che catalizza un processo recentemente scoperto noto come biforcazione elettronica basata sulle flavine. Questo meccanismo è sfruttato solo da alcuni microrganismi anerobici come terza via di accoppiamento energetico e finora si conoscono quattro famiglie di proteine, evolutivamente non correlate, in grado di catalizzarlo. Una di queste è EtfAB, della quale non è chiaro come possa avvenire il trasferimento elettronico tra le due molecole di FAD ad essa legate. Infatti, la distanza tra questi due FAD osservata nella struttura cristalline di EtfAB è di 18 Å, mentre si ritiene più plausibile che i trasferimenti elettronici in biologia non avvengano a distanze maggiori di 14 Å. Per questo è stato suggerito un possibile meccanismo che potrebbe avvicinare le due molecole di FAD. Usando la dinamica molecolare è stato possibile testare, e smentire, il meccanismo proposto. Inoltre, con il Density Functional Theory (DFT), è stato possibile fornire un’interpretazione ad alcuni dati spettroscopici riguardo il possibile trasferimento elettronico tra le due molecole di FAD. Nel terzo progetto, ho collaborato con il Prof. Luca Bertini ad un progetto sulla produzione e propagazione di alcune specie reattive dell’ossigeno (ROS) nel contesto del peptide amiloide beta coinvolto nella patogenesi dell’Alzheimer. Nell’ambito dell’ipotesi amiloide sull’insorgenza della patologia di Alzheimer, un ruolo importante è stato attribuito ai danni causati dai ROS, prodotti da un complesso metallico all’interno del peptide amiloide stesso, in particolare dal radicale ossidrilico (OH.-). Tuttavia, i dettagli su come questi radicali propaghino e reagiscano non sono ancora stati chiariti. Mentre i calcoli DFT del Prof. Bertini affrontavano le capacità ossidative del radicale ossidrilico e i possibili prodotti di reazione nel contesto del peptide amiloide beta, con i miei calcoli di dinamica molecolari è stata fornita una panoramica su quali possibili bersagli del radicale ossidrilico, coordinato allo ione Cu del complesso, possano effettivamente reagire entrando in contatto con il radicale ossidrilico a causa dei moti dinamici del peptide.<br>In this thesis, three independent projects were addressed, sharing the computational approach based on molecular modeling and in particular molecular dynamics. In the first project, the Sec14-PH domain of neurofibromin (NF1) was investigated. The Sec14 domains have been identified in many different proteins, from prokaryotes to humans, serving as exchangers of lipid molecules between membranes, by means of a pocket whose opening is allowed by the motion of a specific alpha-helix (called lid helix). The crystal structure of the NF1-Sec14 domain (of both the wild type and some mutants associated with the onset of neurofibromatosis pathology) has revealed its peculiarity of being structurally coupled to a PH domain that strongly interacts with the lid helix through a long loop (called lid-lock loop). On this basis, a mechanism for the opening of the Sec14 lipid pocket was formulated which would involve a concerted movement of the lid-lock loop, but this movement has actually never been shown. Guided by available experimental data on the thermal denaturation of Sec14-PH domain of NF1, both on the wild type and some neurofibromin-related mutants, several simulations at high temperature were carried out to compare the dynamics of the wild type domain with a pathological mutant associated with the onset of neurofibromatosis. Our simulations lead us to suggest an opening mechanism for the lid helix and provide a hypothesis for the structural and dynamic basis of the onset of the disease in the case of the specific mutant. The second project addressed the study of a protein called EtfAB which catalyzes a recently discovered process known as Flavin-Based Electron Bifurcation (FBEB). This mechanism is only exploited by some anaerobic microorganisms as a third way of energy coupling. So far, four unrelated protein families are known that are able to catalyze FBEB. Among these, EtfAB, catalyzes the electron transfer between the two FAD molecules bound to it. Surprisingly, the distance between these two FADs, as observed in the crystal structure of EtfAB, is 18 Å, whereas biological electron transfer is considered more likely to occur at a maximal distance of 14 Å. To explain this, a possible mechanism has been suggested that could bring the two FAD molecules closer together. Using molecular dynamics, it was possible to test, and discard, the proposed mechanism. Furthermore, with the Density Functional Theory (DFT), it was possible to provide an interpretation to some spectroscopic data regarding the possible electron transfer between the two FAD molecules. In the third project, I collaborated with Prof. Luca Bertini on a project on the production and propagation of some reactive oxygen species (ROS) in the context of the amyloid-beta peptide involved in the pathogenesis of Alzheimer's. In the amyloid hypothesis on the onset of Alzheimer's disease, an important role has been attributed to the damage caused by ROS, produced by a metal ion coordinated to the amyloid peptide itself, in particular by the hydroxyl radical (OH.-). However, the details of how these radicals propagate and react have not yet been clarified. While Prof. Bertini's DFT calculations addressed the oxidative capacities of the hydroxyl radical and the possible reaction products in the context of the amyloid-beta peptide, my molecular dynamics simulations provided an overview on which possible targets of the hydroxyl radical, coordinated to the ion Cu of the complex, could actually react with the hydroxyl radical due to the dynamic motions of the peptide.

La malaltia d’Alzheimer es caracteritza per l’acumulació de plaques extracel.lulars amiloides, formades pels anomenats pèptids amiloides (Aβ40). L’homeòstasi del coure (Cu) es veu afectada en l’etiologia de la malaltia d’Alzheimer, encara que el seu rol és un fet controvertit. Per a l’estudi de la influència del Cu(II) en l’agregació del pèptid amiloide, la morfologia i l’estructura secundària dels agregats amiloides formats en presència de Cu(II) s’han utilitzat AFM, TEM, SEM, SAXS, FTIR i espectrometria de fluorescència. A més, els efectes tòxics d’aquests agregats s’han estudiat en cèl.lules neuronals. Els resultats obtinguts mostren que aquests agregats són amorfos i presenten una toxicitat més alta que les fibres. Per a l’estudi dels dendrímers de maltosa com a possibles moduladors de l’agregació i de la toxicitat del pèptide amiloide. S’ha confirmat que aquests dendrimers no són tòxics en cèl.lules neuronals i que són capaços de modular l’agregació i toxicitat del pèptid amiloide. Aquests resultats permeten considerar als dendrimers de maltosa com a possibles eines per a reduir la toxicitat dels pèptids amilodies.<br>Senile plaques of Alzheimer’s disease patients are composed primarily of the amyloid-β-peptide (Aβ). Recent studies implicate Cu(II) in the aetiology of AD. The role of Cu(II) in ADis currently highly disputed. Influence of Cu(II) on Aβ aggregation and amyloidogenic properties of glycodendrimers were investigated in this thesis. AFM, TEM, SEM, SAXS, FTIR and fluorescence spectroscopy were used to study a morphology and a secondary structure of Aβ-Cu(II) aggregates. The toxic effects of Aβ40-Cu(II) amorphous aggregates was confirmed for neuronal cell lines. It was shown that maltose glycodendrimers can be efficiently used to modulate Alzheimer’s amyloid peptide aggregation and inhibit cell toxicity by facilitating the clustering of amyloid fibrils. These results show that glycodendrimers are promising non-toxic agents in the search for anti-amyloidogenic compounds. It was also suggested that fibril clumping may be anti-amyloid toxicity strategy.

The Amyloid β-peptide (Aβ) is accumulated in several diseases including GNE myopathy and Alzheimer’s disease (AD). GNE myopathy is a skeletal muscle disorder caused by biallelic mutations in the UDP-N-acetylglucosamine 2-epimerase/N-acetylmannosamine kinase (GNE) gene, which codifies for a key enzyme in sialic acid biosynthesis. The mutated GNE gene encodes a hypofunctional enzyme causing a decrease in cellular sialic acid. We have found that hyposialylation favors Aβ endocytosis in skeletal muscle cells, which is dependent on clathrin and heparan sulfate proteoglycan. Furthermore, we have observed that intracellular Aβ induces apoptosis in skeletal muscle cells through the impairment of Akt signaling pathway. Accordingly, Akt phosphorylation is reduced and apoptosis is enhanced in GNE myopathy myoblasts. Finally, through a genome-wide screen in Saccharomyces cerevisiae we have identified novel modulators of Aβ toxicity including components of the mitochondrial respiratory chain and members of the Ca2+ signaling pathways. In conclusion, this study provides a better understanding of the Aβ relevance in the pathophysiology of GNE myopathy and AD.<br>El pèptid β-amiloide (Aβ) s’acumula en diverses malalties com la miopatia de GNE i la Malaltia d’Alzheimer (MA). La miopatia de GNE és una malaltia del múscul esquelètic causada per mutacions al gen UDP-N-acetylglucosamine 2-epimerase/N-acetylmannosamine kinase (GNE), que codifica un enzim clau en la biosíntesis de l’àcid siàlic. El GNE mutat produeixen un enzim hipofuncional i una conseqüent disminució de l’àcid siàlic cel·lular. Hem vist que la hiposialització afavoreix l’endocitosi de l’Aβ en cèl·lules musculars, la qual és dependent de clatrina i de l’heparà sulfat proteoglicà. A més, hem observat que l’Aβ intracel·lular indueix l’apoptosi en cèl·lules musculars mitjançant la inhibició de l’Akt. Així, la fosforilació de l’Akt es troba reduïda i l’apoptosi induïda en mioblasts d’un pacient amb la miopatia de GNE. Finalment, a través d’un cribratge del genoma complert de Saccharomyces cerevisiae, hem identificat nous moduladors de la toxicitat per Aβ que inclouen components de la cadena respiratòria mitocondrial i membres de la via de senyalització del Ca2+. En resum, aquest estudi ofereix una millor comprensió del rol de l’Aβ en la miopatia de GNE i la MA.

Nitric oxide (NO) is a molecule that has pleiotropic effects in brain and vascular system. Physiologically, NO induces the translation of the GluN2B subunit of N-methyl Daspartate receptor (NMDARc) by derepressing it 5’untranslated region (5’UTR) effect. This pathway is due to the activation of the heme regulated eIF2α (HRI) kinase and prevents an excess of GluN2B levels, especially at the extrasynaptic areas, where it can trigger excitotoxicity. Pathologically, NO in a pro-oxidant environment such as in Alzheimer’s Disease (AD) reacts with superoxide anion producing peroxynitrite, which can nitrotyrosinates proteins. There are other concomitant oxidative processes that affect AD patient like protein glycation. Therefore the albumin, the most abundant plasmatic protein, in AD patients is more nitrotyrosinated and glycated, which affects its structure. Modified albumin has a reduced ability as an osmolarity buffer and it is hardly uptaken by hepatoma cells. Moreover, modified albumin binds more Aβ, contributing to maintain higher amount of amyloid in brain and plasma.<br>El óxido nítrico (NO) es una molécula con efectos pleyotrópicos en cerebro y sistema vascular. Fisiológicamente, induce la traducción de la subunidad GluN2B del N-methyl D-aspartate receptor (NMDARc) al revertir la represión de su 5’untranslated region (5’UTR). Este efecto se debe a la activación de la heme regulated eIF2α kinase (HRI) y previene el exceso de GluN2B, especialmente en regiones extrasinápticas, donde desencadena excitotoxicidad. Patológicamente, el NO en un ambiente pro-oxidatvio como el dado en la enfermedad de Alzheimer (AD) reacciona con el anión superóxido produciendo peroxinitrito, y causando entre otros efectos la nitrotirosinación de proteínas. Simultaneamente, las proteínas de pacientes con AD padecen otros procesos oxidativos como la glicación. Por tanto, la albúmina, la proteína plasmática más abundante, en estos pacientes está más nitrotirosinada y glicada, afectando su estructura. La albúmina modificada presenta menos capacidad para tamponar la osmolaridad y apenas es digerida por las células de hepatoma. Además, une más Aβ, contribuyendo a mantener más alta la carga amiloidogénica en cerebro y plasma.

Memory impairment occurs when important synapses are modified. Alzheimer s disease (DA) is the most common cause of dementia. DA is characterized by cognitive damage, accumulation of the pathogenic amyloid-β (Aβ) peptide, and cholinergic dysfunction. Moreover, oxidative stress is associated with DA. Therapies used for dementia are still palliative rather than curative. Consequently, new therapies are urgently required. Biological effects of tellurium compounds have been studied, leading to a set of interesting and promising applications. Accordingly, 2-phenylethinyl-butyltellurium (PEBT), an organotellurium compound, has been reported as antioxidant. The purpose of this study was to characterize PEBT as a promising alternative for memory improvement and prevention of cognitive deficits, using experimental models of DA in mice. Initially, the present study was conducted to evaluate the effect of a single oral administration (p.o.) of PEBT at a dose of 10 mg/kg on memory, employing the step-down inhibitory avoidance task. PEBT administered 1 h before training, immediately after training or 1 h before the test session of the step-down inhibitory avoidance task increased the step-down latency time in comparison to the control mice, improving acquisition, consolidation, and retrieval of memory, respectively. The glutamate uptake, but not glutamate release, by cerebral cortex and hippocampal slices of mice was inhibited after 1 h of treatment with PEBT. After 24 h of PEBT exposure, the inhibition of cerebral cortex glutamate uptake disappeared. The improvement of memory by PEBT seems most likely to be mediated through an interaction with the amino acid transporters of the glutamatergic system. Thereafter, a subchronic PEBT treatment (1 mg/kg, p.o., for 10 days) after injection of Aβ(25-35) (3 nmol/3 μl/per site, intracerebroventricular) reversed Aβ-induced learning and memory deficits in the Morris water maze and step-down inhibitory avoidance tasks. In addition, PEBT (10 mg/kg, p.o.), administered 30 min before scopolamine (1 mg/kg, intraperitoneal), ameliorated memory deficit induced by this amnesic agent in the Morris water maze. Further, scopolamine was given 30 min before training and test or immediately post-training of step-down inhibitory avoidance task, inducing damage on acquisition, retrieval, and consolidation of memory, respectively. PEBT, administered 30 min before scopolamine, improved consolidation and retrieval stages, but not acquisition. General locomotor and exploratory activities, evaluated in the open-field test, were similar in all mice. Finally, the antiapoptotic effect of PEBT was evaluated. Human retinal pigment epithelial cells (ARPE-19 cell line) were exposed to in vitro oxidative stress by 10 ng/ml tumor necrosis factor-α and 600 μM H2O2. One hour PEBT incubation at concentrations of 7.5 and 10 μM attenuated the apoptosis induced by oxidative stress. This effect lasted up to 6 hours after oxidative stress induction. PEBT (5 and 10 μM) inhibited oxidative stress-induced poly (ADP-ribose) polymerase (PARP) cleavage and restored extracellular-signal-related kinase (ERK) phosphorylation decreased by oxidative stress. The protective mechanism exerted by PEBT against oxidative stress may involve PARP cleavage, regulation of ERK pathway, as well as its known antioxidant properties. In conclusion, the finds of the present thesis point out the ameliorative effect of PEBT on memory stages (acquisition, consolidation and retrieval). Likewise, PEBT improved memory impairment in mice. These effects seem to be due to strengthen the physiological glutamatergic tonus by PEBT and the antiapoptotic effect of PEBT. Therefore, PEBT could be considered a candidate for the prevention of memory deficits such as those observed in DA.<br>memória inclui pelo menos três tipos de processamento relacionados entre si: aquisição, consolidação e evocação. A memória é afetada quando as sinapses encarregadas de fazer ou evocar memórias encontram-se alteradas. A doença de Alzheimer (DA) é a causa mais comum de demência. A DA é caracterizada por danos cognitivos, acúmulo de peptídeo β-amiloide (Aβ) e disfunção colinérgica. Além disso, o estresse oxidativo está associado à DA. Uma vez que ainda não há cura para a DA e as terapias atuais são apenas paliativas, torna-se importante a busca de novos compostos para melhorar danos cognitivos. Com o estudo dos efeitos biológicos de compostos de telúrio, muitas aplicações estão sendo descobertas. Neste sentido, o composto orgânico de telúrio 2-feniletinil-butiltelúrio (PEBT) apresenta efeito antioxidante. O objetivo deste estudo foi caracterizar o PEBT como uma alternativa promissora para a melhora e prevenção de danos cognitivos, usando modelos experimentais da DA em camundongos. Primeiramente, avaliou-se o efeito de uma única dose oral (p.o) de PEBT (10 mg/kg) na memória, utilizando a tarefa da esquiva inibitória. O tratamento com PEBT 1 h antes do treino, imediatamente após o treino ou 1 h antes do teste da esquiva inibitória aumentou a latência comparada com os animais controles, melhorando a aquisição, consolidação e evocação da memória, respectivamente. A captação de glutamato, mas não a liberação deste neurotrasmissor, foi inibida em córtex e hipocampo de camundongos após 1 h de tratamento com PEBT. Após 24 h, a inibição da captação de glutamato no córtex não foi mais evidenciada. A melhora da memória causada pelo PEBT parece ser mediada através da interação com os transportadores de glutamato. Além disso, o tratamento subcrônico com PEBT (1 mg/kg, p.o., por 10 dias) após a injeção de Aβ(25-35) (3 nmol/3 μl/per site, intracerebroventricular) reverteu o prejuízo no aprendizado e na memória causados por Aβ nas tarefas do labirinto aquático de Morris e na esquiva inibitória. O PEBT (10 mg/kg, p.o.), adminstrado 30 min antes da escopolamina (1 mg/kg, intraperitoneal), também protegeu do dano de memória causado por este agente anticolinérgico no labirinto aquático de Morris. Quando a escopolamina foi administrada 30 min antes do treino ou teste, ou imediatamente após o treino da esquiva inibitória houve dano na aquisição, evocação e consolidação da memória, respectivamente. O PEBT, administrado 30 min antes da escopolamina, protegeu do dano na consolidação e evocação da memória, mas não na aquisição. Não houve diferença nas atividades locomotora e exploratória dos animais tratados com o PEBT no teste do campo aberto. Finalmente, o efeito antiapoptótico do PEBT foi avaliado. Células do epitélio pigmentado da retina humana (linhagem ARPE-19) foram expostas ao estresse oxidativo induzido pelo fator de necrose tumoral-α (10 ng/ml) e H2O2 (600 μM). O PEBT (7.5 e 10 μM), quando pré-incubado por 1 h, protegeu contra a apoptose induzida pelo estresse oxidativo e este efeito permaneceu até 6 h após a indução. O PEBT (5 e 10 μM) inibiu a clivagem da poli(ADP-ribose) polimerase (PARP) induzida por estresse oxidativo e, também, restaurou a fosforilação da quinase regulada por sinal extracelular (ERK). O efeito protetor do PEBT contra o estresse oxidativo parece envolver a clivagem da PARP e a regulação da fosforilaçao de ERK, além de sua atividade antioxidante. Nesse sentido, os resultados apresentados nesta tese destacam o efeito do PEBT na melhora das três fases da memória, bem como em modelos de dano cognitivos em camundongos. Estes resultados parecem estar relacionados ao aumento do tônus glutamatérgico causado pelo PEBT e seu efeito antiapoptótico. Assim sendo, estes dados sugerem que o PEBT poderá, futuramente, ser considerado candidato para a prevenção de danos de memória, como aqueles observados na DA.

An involvement of N-Methyl-D-Aspartate receptors (NMDAR) in AD is proposed, since its inhibition attenuates some aspects of AD s neuropathology. In this regard, polyamines, like spermidine and spermine, positive modulators of NMDARs, have been shown to have both concentration and synthesis increased by Aβ. Using the novel object recognition task we showed that negative modulation of polyamine system, been trough blockade of its binding site at NMDAR by arcaine (0.02 nmol/site), traxoprodil (0.002 nmol/site), or inhibition of polyamine synthesis by DFMO (2.7 nmol/site), reverses Aβ25-35-induced memory impairment in mice. The activation of polyamine binding site at NMDAR located at extrasynaptic sites might underlie the cognitive deficits of Aβ25-35-treated mice, since incubation of hippocampal neuron cultures with spermidine (400 μM) or Aβ25-35 (10 μM) significantly increased nuclear accumulation of jacob protein, a marker of extrasynaptic NMDAR activation. Moreover, traxoprodil (4nM), arcaine (4 μM) or DFMO (5 μM) blocked the Aβ-induced jacob nuclear translocation. Activation of extrasynaptic NMDAR in neurons leads to striping of synaptic contacts and simplification of neuronal cytoarchitecture. Incubation of hippocampal neuron cultures with traxoprodil (4 Nm), arcaine (4 μM) or DFMO (5 μM) reversed the deleterious effects of Aβ25-35 on dendritic spine number and spine morphology. We also evaluated the involvement of A-type K+ currents on the Aβ25-35-induced memory impairment. Administration of Tx3-1 (3 100 pmol/site), a selective IA blocker, restored memory of mice injected with Aβ25-35 and tested on the novel object recognition task The reversal of memory impairment and the protective effect on dendritic spine alterations exerted by the modulators of the polyamine system suggest the polyamine binding site at extrasynaptic NMDAR a potential player in Aβ-induced cognitive deficit.<br>O peptídeo β-amiloide (Aβ), reconhecido como agente tóxico na Doença de Alzheimer (DA) é implicado como causador de danos cognitivos e sinápticos, apesar de os mecanismos não serem completamente compreendidos. O envolvimento do receptor N-metil-D-aspartato (NMDA) na DA é sugerido, visto que o seu bloqueio atenua alguns aspectos neuropatológicos da DA. Nesse contexto, tem sido demonstrado que as poliaminas, como espermidina e espermina, moduladores positivos do receptor NMDA, possuem níveis e síntese elevada tanto no cérebro de pacientes com DA como em preparações in vitro utilizando o peptídeo Aβ. Neste estudo demonstrou-se que a modulação do sistema das poliaminas, através do bloqueio do seu sítio de ligação no receptor NMDA por arcaína (0,02 nmol/sítio), traxoprodil (0,002 nmol/sítio) ou da inibição de sua síntese por DFMO (2,7 nmol/sítio), reverte o déficit cognitivo induzido pela injeção de Aβ25-35 em camundongos testados na tarefa de reconhecimento de objetos. A ativação do sítio de ligação das poliaminas em receptores NMDA extrassinápticos pode subjazer o déficit cognitivo de camundongos injetados com Aβ25-35, visto que a incubação de culturas primárias de neurônios hipocampais com espermidina (400 μM), NMDA (200 μM) ou Aβ25-35 (10 μM) aumenta o acúmulo nuclear de jacob, um marcador de ativação de receptores NMDA extrassinápticos, de maneira significante. Ademais, traxoprodil (4 nM), arcaína (4 μM) ou DFMO (5 μM) bloquearam o acúmulo nuclear de jacob induzido por Aβ. A ativação de receptores NMDA extrassinápticos em neurônios leva a simplificação da citoarquitetura neuronal e a diminuição de contatos sinápticos. Aqui demonstrou-se que a incubação de culturas de neurônios hipocampais com traxoprodil (4 nM), arcaína (4 μM) ou DFMO (5 μM) reverte as alterações na a densidade e morfologia de espinhas dendríticas induzido pela incubação com Aβ25-35. Ainda, também avaliou-se o envolvimento de correntes de K+ do tipo A no déficit cognitivo induzido pela injeção i.c.v. de Aβ25-35. A administração de Tx3-1 (3 100 pmol/sítio), um bloqueador seletivo de correntes IA, reverteu o prejuízo de memória de camundongos injetados com Aβ25-35 e testados na tarefa de reconhecimento de objetos. A reversão dos danos cognitivos e sinápticos induzidos por Aβ25-35 através da modulação do sistema das poliaminas sugere a estimulação do sítio de ligação das poliaminas no receptor NMDA, possivelmente extrassínaptico, como um dos mecanimos por trás do déficit cognitivo induzido pelo peptídeo Aβ.

Afeta essencialmente os idosos e está associada a alterações a nível da memória e da orientação espacial. Descrita pela primeira vez em 1907 pelo médico Aloysius Alzheimer, caracteriza-se pela deposição de peptídeo β-amiloide (Aβ) e proteína tau hiperfosforilada, sob a forma de placas senis e de aglomerados neurofibrilares (NFTs), respetivamente. A deteção destas proteínas anormais é essencial no diagnóstico da doença, uma vez que os sintomas da DA são semelhantes aos encontrados noutras patologias. Uma minoria dos doentes desenvolve a doença devido à presença específica de mutações genéticas na proteína precursora amiloide (APP) ou nas presenilinas 1 e 2 (PSEN1/PSEN2). Para os restantes casos estão já identificados alguns fatores de risco, contudo a causa exata é ainda desconhecida. A hipótese de que a DA tenha uma causa infeciosa encontra-se atualmente sob intensa investigação. De facto, alguns microorganismos têm a capacidade de permanecer no organismo na forma latente, causando uma infeção crónica. Ao longo da vida podem sofrer várias reativações, o que origina um estado inflamatório recorrente. O estado inflamatório está diretamente relacionado com a DA, pela ativação de mecanismos que contribuem para a deposição proteica. Por outro lado, verificou-se que o peptídeo Aβ poderá exercer atividade antimicrobiana contra vários agentes patogénicos, tais como os vírus Herpes Simples do tipo 1 e 2 (HSV-1/ HSV-2) ou Influenza, entre outros. A teoria antimicrobiana tenta explicar a etiologia da DA, associando o aumento da produção de peptídeo Aβ e a sua consequente deposição em resposta à presença crónica de determinados microorganismos no cérebro. A patologia amiloide observada na DA inicia-se vários anos antes do aparecimento dos primeiros sintomas, facto que se apresenta em concordância com esta teoria.<br>Alzheimer's Disease (AD) is a neurodegenerative disease, considered the leading cause of dementia worldwide. It primarily affects the elderly and is associated with changes in memory and spatial orientation. First described in 1907 by the doctor Aloysius Alzheimer, it is characterized by the deposition of β-amyloid peptide (Aβ) and hyperphosphorylated tau protein in the form of senile plaques and neurofibrillary agglomerates (NFTs), respectively. Detection of these abnormal proteins is essential in the diagnosis of the disease, since the symptoms of AD are similar to those found in other pathologies. A minority of patients develop the disease because of the specific presence of genetic mutations in the amyloid precursor protein (APP), or in presenilin’s 1 and 2 (PSEN1 / PSEN2). For the remaining cases, some risk factors have been identified, but the exact cause is still unknown. The hypothesis that AD has an infectious cause is currently under intense investigation. Indeed, some microorganisms have the ability to remain in the body in a latent form causing a chronic infection. Throughout an individual’s life they can undergo several reactivations, which gives rise to a recurrent inflammatory state. The inflammatory state is directly related to AD by the activation of mechanisms that contribute to protein deposition. On the other hand, the Aβ peptide has been shown to be able to exert antimicrobial activity against various pathogens, such as Herpes Simplex-1 and 2 (HSV-1 / HSV-2) or Influenza, among others. The antimicrobial theory attempts to explain the etiology of AD by associating the increase in Aβ production and its consequent deposition with the chronic existence of certain microorganisms in the brain. The amyloid pathology observed in AD begins several years before the appearance of the first symptoms, which is in agreement with this theory.

Introdução: A doença de Alzheimer é multifatorial ou poligênica e corresponde a cerca de 50 a 75 % de casos de demência nos seres humanos. A proteína TAU é importante na patogenia da demência frontotemporal e para o citoesqueleto neuronal. Essa proteína consegue interagir com a α- e βglobulina para estabilizar os microtúbulos que são essenciais no transporte axonal, na plasticidade sináptica e na manutenção da estrutura neural. Objetivos: Esse trabalho teve como objetivo conhecer as proteínas e os genes promotores no desenvolvimento da doença de alzheimer. Metodologia: Foi realizada uma busca simples nos bancos de dados da SciELO, PubMed e Periódicos Capes, por meio de palavras-chave: “doença de Alzheimer”, “proteína TAU” e “demência frontotemporal”. Foram revisados artigos publicados nos anos de 2009 à 2021. Resultados: A proteína TAU é encontrada montada na doença de alzheimer e os MNFs possuem filamentos emparelhados nos FHA. Os fatores de risco são: a presença do alelo ε4 do gene da apolipoproteína E APOE, do gene da proteína APP no cromossomo 21 e dos genes Presenilinas 1 e 2 nos cromossomos 14 e 1. A TAU faz sua própria polimerização e inibe a despolimerização rápida da tubulina, esse processo é regulado pelo estado de fosforilação da proteína TAU, que compreende aproximadamente 79 sítios de fosforilação em resíduos de serina e treonina. O equilíbrio entre fosforilação e desfosforilação dos epítopos promove alterações que influenciam como a proteína TAU interage com a α- e β-tubulina que estabilizam os microtúbulos nos neurônios. Várias proteínas quinases e fosfatases estão envolvidas na regulação da fosforilação da TAU, sendo a enzima glicogênio sintase quinase 3β GSK3β a mais importante TAU quinase nos neurônios. A redução da expressão de certas fosfatases também é identificada nos tecidos cerebrais de pacientes com alzheimer. Conclusão: A TAU hiperfosforilada intraneural pode ser encontrada no cérebro de indivíduos com demência muito leve, não acompanhada de patologia β-amiloide. A hiperfosforilação de TAU pode ser um evento precoce na fisiopatologia do alzheimer, enquanto outros mecanismos patológicos, incluindo superprodução da Aβ e ativação de cascatas inflamatórias e estresse oxidativo, podem ser secundários à disfunção geral na homeostase neuronal.

INTRODUÇÃO: A biocompatibilidade dos biosenssores, tem sido cada vez mais evidenciados em aplicações por meio de nanoestruturas com uma maior afinidade. Biomoléculas de DNA, enzimas, anticorpos e antígenos, são amplamente utilizadas devido a sua alta afinidade e estabilidade, reconhecendo com eficiência diferentes moléculas e organismos. A barreira hematoencefálica desempenha função principal relaciona a inibição da entrada de substâncias em seu interior, visando inibir a entrada de células indesejadas, íons e cloretos. Doenças de acometimento neurológico possuem uma deficiência em relação ao seu diagnóstico precoce, contudo, a utilização de biosenssores visa propiciar agilidade, economia e melhora relacionada a qualidade da saúde global. A detecção de biomarcadores em processos neuroinflamatórios, é demonstrada através de polímeros com impressão molecular em bases naturais. OBJETIVO: Revisar a literatura científica quanto a utilização de biosenssores nanoestruturados em doenças de perfil neuroinflamatórias e neurodegenerativas, visando o potencial de ação e sensibilidade mediante ao declive fisiológico. MÉTODOS: Foram utilizados artigos científicos para a oriundos das bases de dados da saúde: PubMed, Science Direct e SciELO. A data de publicação (2015-2021). RESULTADOS: A utilização recente de nanoestruturas com biocompatibilidade, tem-se mostrado promissora para o diagnóstico de doenças relacionadas ao envelhecimento. Biossensores são capazes de reconhecer fluídos biológicos, o que possibilita a identificação e grau da doença. Ainda, foi possível mensurar a presença de glutamato através de bioeletrodos, mediante a imobilização de um peptídeo mimético em eletrodos de grafite em pacientes com Doença de Alzheimer. Neste sentido, a utilização da enzima acetilcolinesterase em biosenssores, demostraram resultados significativos relacionados a detecção de ácido acético e acetilcolina, em pacientes com demência. Ainda, foi possível identificar suas respectivas concentrações, grau e nível da doença. CONCLUSÃO: A perspectiva futurista da utilização de biosenssores nanoestruturados por meio do avanço biotecnológico demonstra ser promissora, não somente pelo custo, mas devido a sua maior sensibilidade de identificação e quantificação. Biosensores que visam rastrear doenças neurológicas detectam biomarcadores importantes como: Glutamato, acetilcolina e a proteína β-amiloide. Contudo, apesar de demonstrar eficiência em relação à seletividade e sensibilidade, estudos com maior profundidade devem ser desenvolvidos na área para melhorar a robustez e eficiência analítica.
