Abstract:
Abstract. A study of Pc-5 magnetic pulsations using data from the Combined Release and Radiation Effects Satellite (CRRES) was carried out. Three-component dynamic magnetic field spectrograms have been used to survey ULF pulsation activity for the approximate fourteen month lifetime of CRRES. Two-hour panels of dynamic spectra were examined to find events which fall into two basic categories: 1) toroidal modes (fundamental and harmonic resonances) and 2) poloidal modes, which include compressional oscillations. The occurence rates were determined as a function of L value and local time. The main result is a comparable probability of occurence of toroidal mode oscillations on the dawn and dusk sides of the magnetosphere inside geosynchronous orbit, while poloidal mode oscillations occur predominantly along the dusk side, consistent with high azimuthal mode number excitation by ring current ions. Pc-5 pulsations following Storm Sudden Commencements (SSCs) were examined separately. The spatial distribution of modes for the SSC events was consistent with the statistical study for the lifetime of CRRES. The toroidal fundamental (and harmonic) resonances are the dominant mode seen on the dawn-side of the magnetosphere following SSCs. Power is mixed in all three components. In the 21 dusk side SSC events there were only a few examples of purely compressional (two) or radial (one) power in the CRRES study, a few more examples of purely toroidal modes (six), with all three components predominant in about half (ten) of the events. Prostate cancer is the most frequently diagnosed malignancy in males in the United States and most other industrialized Western countries. It kills more than 30,000 American men yearly and it is one of the top 10 deadliest cancers in Taiwan. There have been no therapeutic agents that provide a survival advantage in prostate cancer. Therefore, it is necessary to develop novel agents for the treatment of hormone-refractory prostate cancer. In the first part of this study, we used human hormone-resistant prostate cancer cell line PC-3 to investigate the anti-tumor effect and mechanism of compound LYF-5 (3’-methoxyl-2-phenyl-6-methyl-1,8-naphthyridine-4-one). LYF-5 inhibited cell growth in a dose-dependent manner with an IC50 of 0.12 μM by sulforhodamine B (SRB) assays. Using FACScan flow cytometric analysis, the data showed that LYF-5 induced the arrest of cells in G2/M phase of the cell cycle and a subsequent increase of hypodiploid sub-G1 cells. The apoptotic cell death was also identified by TUNEL reaction technique. Immunofluoresence microscopic examination indicated that LYF-5 had an action target on tubulin/microtubule. Several apoptosis-related proteins were examined in this study. After the detection of expression of Bcl-2 family members by Western blotting, the data showed that LYF-5 induced phosphorylation of Bcl-2 and Bcl-xL, and Mcl-1 down-regulation. In addition, the cleaved forms of Mcl-1 was also apparent. Moreover, LYF-5-induced apoptosis was associated with processing of pro-caspase-8, pro-caspase-3 and pro-caspase-9, indicating the involvement of caspase activation. The expression of XIAP and cIAP2 were decreased after treating with LYF-5. Furthermore, we used immunofluoresence microscopic examination to examine the release reaction of cytochrome c. The data showed that LYF-5 significantly induced the release of cytochrome c from mitochondria. Taken together, it is suggested that LYF-5 induces apoptotic cell death through the target on tubulin/microtubule, the induction of mitotic arrest of the cell cycle, phosphorylation and cleavage of anti-apoptotic Bcl-2 family protein members, the activation of caspase and the release of cytochrome c. In the second part of this study, the structure-activity relationship of a series of anthracenedione derivatives on anticancer activity and their action mechanisms in human hormone-resistant prostate cancer PC-3 cells were investigated. By using SRB assays, we found that several compounds displayed anticancer activity and amongst, WRC-76 was the most effective one with an IC50 of 0.28 μM. Accordingly, WRC-76 was selected for the mechanism study. The data showed that WRC-76 induced the arrest of the cell cycle in S and G2/M phase and a subsequent increase of hypodiploid sub-G1 phase. Immunofluoresence microscopic examination indicated that WRC-76 had no effect on tubulin/microtubule function. Several apoptosis-related proteins were examined in this study. After the detection of expression of Bcl-2 family members by Western blotting, the data showed that the expression of Mcl-1 was decreased after theatment with WRC-76. Moreover, WRC-76-induced cell apoptosis was associated with processing of pro-caspase-8, pro-caspase-3 and pro-caspase-9. The expression of XIAP but not survivin was inhibited by WRC-76. Caspase-12, CHOP and GRP78 are three ER stress markers and the expression of caspase-12 and CHOP were significantly increased by WRC76. Furthermore, JNK and p38 MAP kinases involved pathways were also activated in cells responsive to WRC-76. Taken together, it is suggested that WRC-76 induces apoptotic cell death through the activation of caspase-8, -9 and -3. WRC-76 also induces the ER stress of increase of CHOP expression and caspase-12 activity. Furthermore, the activation of JNK and p38 MAPK may, at least partly, explain WRC-76-induced apoptotic cell death.

Abstract 第一部分 In search for potential anticancer drug candidates in natural products, we have carried out a large scale screening test on cytotoxic effects using MTT assay. The compound of CMB, from Cesearia sp., was the most effective one on the induction of apoptosis in human androgen-independent prostate cancer cells (PC-3). CMB induced cell apoptosis based on TUNEL-response and negative release reaction of lactate dehydrogenase (LDH). Furthermore, its cytotoxic effect was additive in combination with paclitaxel. Using Western blotting analysis, we found that CMB compound induced the release of cytochrome c from mitochrodria to cytosol and markedly induced the phosphorylation of a p38 mitogen-activated protein kinase (MAPK) in a time-dependent manner; besides, SB203580, a selective p38 MAPK inhibitor, reversed CMB compound-induced apoptosis suggesting the involvement of p38 MAPK-mediated pathway. CMB also caused the activation of caspase-3 in PC-3 cells. We examined the expression of Bcl-2 family proteins, such as Bcl-2 and Bax. CMB caused a downregulation of the expression of Bcl-2 and showed little influence on the expression of Bax. In this study, we also examined the anti-angiogenic activity of CMB in an in vivo model. The data demonstrated that CMB inhibited vascular endothelium growth factor (VEGF)-induced angiogenesis in a concentration-dependent manner. In summary, it is suggested that CMB-induced apoptosis is through a p38 MAPK-involved and Bcl-2 downregulation-cytochrome c release-caspase-3 activation signaling pathway. Abstract 第二部分 We have carried out a large anticancer screening test in numerous natural products by MTT assay. We found that several prostanoids from the Japanese stolonifer Clavularia sp., G4c-series showed effective cytotoxic activities in human androgen-independent prostate cancer cells (PC-3) and G4c-f8-5-1 was the most potent one. Besides, G4c-f8-5-1 induced cell apoptosis based on strong positive TUNEL-response. G4c-f8-5-1 triggered the lengthening of sub G1 phase of the cell cycle in a concentration- and time-dependent manner with an IC50 value of 0.5 μM. Using several pharmacological inhibitors to block biological functions, our data showed that the regulation of the generation of free radical, and the activition of p38-MAPK, protein kinase C, and Ras were not involved in the G4c-f8-5-1-induced apoptotic response. However, G4c-f8-5-1 induced a significant increase of caspase-3 activity. Furthermore, there is little alteration of the Bcl-2 protein expression level after the exposure of G4c-f8-5-1 to PC-3 cells. On the contrast, G4c-f8-5-1 caused an increase of the Bax protein expression in a concentration-dependent manner. It is suggested that G4c-f8-5-1 is the most potent prostanoid that induces apoptosis in PC-3 cells via a Bax upregulation- and caspase-3 activation-involved mechanism in this study.

Abstract:
In this thesis we examine the generation mechanisms of Pc 5 ULF waves during geomagnetic storms. Also, we study the interaction between Pc 5 ULF waves and energetic particles in the radiation belts and the observed energetic particle flux modulation by Pc 5 ULF waves is verified using particle simulations. Firstly, we present case studies of Pc 5 pulsations using ground-based magnetometer and satellite data during geomagnetic storm times, specifically we selecting three storm time events which show a brief increase in Dst in the main phase of the storms. By studying these events, we attempt to identify the generation mechanisms responsible for the geomagnetic pulsations. The observed pulsations exhibit the characteristic features of a Field Line Resonance. Our results also show evidence for the penetration of ULF wave power in the Pc 5 band to much lower L-shells than normal, suggesting significant reduction of the local Alfven eigenfrequency continuum as compared to non-storm times. This may have considerable significance for the interaction between ULF waves and MeV electrons in the outer radiation belt during storms. Secondly, based on the hypothesis that Pc 5 ULF waves may play an important role in energetic particle dynamics in the radiation belt and ring current, we investigated the relationship between Pc 5 pulsations and energetic particle flux oscillations. We observed very strong Pc 5 oscillations during the great magnetic storm of March 24, 1991 [Lee et al., 2007] and electron flux simultaneously oscillating with the same frequencies in the time domain. We also characterize two more events and present an examination of the relationship between the electron flux modulation and Pc 5 ULF pulsations. Based on our observations, the modulation of energetic particles might be associated with a drift-resonance interaction, or the advection of an energetic particle density gradient. Finally, we numerically calculate the trajectories and energy change of charged particles under the influence of model ULF wave electric fields. This modeling work is used to help to explain the observations and provides evidence which supports the modulation mechanisms such as advection of a flux gradient and drift resonance.
