PASTORIZIA ANTICA E MODERNA NELL'ITALIA CENTRALE: UNO STUDIO INTERDISCIPLINARE NEI MONTI DEL CICOLANOQuest'articolo è un contributo al dibattito attuale, tra archeologi, di come sia possibile ricostruire la pastorizia antica attraverso i dati archeologici. I monti del Cicolano oggi vengono sfruttati ogni estate dalle gente dei villaggi circostanti per tre scopi diversi: agricoltura, pastorizia stanziale, pastorizia transumante. Il progetto comprende uno studio geomorfologico, una ricognizione archeologica, una ricerca documentaria, e studi etnoarcheologici. Il regolare insediamento stagionale cominciò nella tarda preistoria e nel periodo romano i monti furono usati da agricoltori e da pastori forse transumanti. L'agricoltura arativa e la pastorizia risultano ambedue nei documenti medioevali e postmedioevali e si possono riconoscere nel materiale archeologico raccolto. Maè assai difficile identificare differenti tipi di pastorizia nei dati archeologici forniti dalla ricognizione. L'archeologia dell'insediamento, la cultura materiale portatile, la fauna, i residui botanici creati dagli agricolturi e pastori di oggi nel Cicolano mostrano le difficoltà nel distinguere i diversi tipi di uso dei terreni alti. Ma, almeno, ci danno utili suggerimenti per sviluppare le future metodologie.

Abstract:
The focus of this research is the area known as Sorgenti del Brembo di Carona (sources of river Brembo of Carona), which is located in the Orobie Alps (province of Bergamo, Italy). The current archaeological activities in the area, carried out by the Civico Museo Archeologico di Bergamo, are site-specific and mainly focused on Iron Age rock engravings and on a medieval dwelling excavation. The present study aims at a wider approach to upland archaeology, more focused on landscape evolution rather than on single evidence. The starting point is the methodology developed in other alpine contexts, like the ALPES (Alpine Landscapes: Pastoralism and Environment of Val di Sole) project. The data, gathered through extensive field survey activities, assessed the presence of a complex landscape, with pastoral evidence, iron mining facilities and charcoal production sites, dating from Early Middle Ages to the present. All the collected data are managed through a GIS in order to maintain their spatial reference. Therefore, it was possible to easy cross-reference them with several historical documents (cartography, cadastres, archives) and also to perform quantitative and spatial analysis. This method allowed us to reconstruct a diachronic evolution of human activities impact on the landscape formation. An inductive predictive modelling based on the integration with ethnoarchaeology was also implemented using modern pastoral sites. The results shed light on the complex dynamics of the human approach to high-altitude regions and on the alpine environment constraints to human activities. On the other hand, it was also possible to asses both the strengths and biases of the current application of predictive models to Alpine cultural heritage. The methodology developed during this research, following and implementing previously developed methods, can be a step forward on the definition of a common archaeological approach to upland contexts.
Il progetto di ricerca nasce a seguito delle indagini archeologiche condotte dal Civico Museo Archeologico di Bergamo nel comune di Carona (BG), situato in alta val Brembana, sulle Alpi Orobie, che hanno permesso di identificare un sito cultuale con incisioni rupestri dell'età del Ferro e un villaggio minerario con fasi altomedievali e medievali. L'obiettivo principale della presente ricerca è stato ampliare la conoscenza storico-archeologica di tutto il territorio alla testata del Brembo di Carona, senza focalizzarsi su singoli siti e applicando le metodologie sviluppate all'Università di Trento nell'ambito del progetto ALPES (Alpine Landscapes: Pastoralism and Environment of Val di Sole), che prevedono un approccio al paesaggio montano in una prospettiva diacronica, inquadrabile nell'ambito della Landscape Archaeology. Le attività di ricerca sul campo hanno rappresentato il fulcro del progetto, permettendo l'individuazione di centinaia di evidenze antropiche. I dati raccolti sul campo sono stati contestualizzati attraverso l'analisi di diverse tipologie di fonti e materiali, non solo di tipo archeologico ma anche inquadrabili in ambiti storico-archivistici e topografici, con un’impostazione della ricerca in senso marcatamente interdisciplinare. L'elaborazione di un modello predittivo etnoarcheologico ha avuto il duplice obiettivo di fornire uno strumento di interpretazione delle strutture presenti sul territorio e di validare la stessa metodologia prognostica impiegata, già elaborata in ambito trentino. I dati raccolti e i risultati della loro analisi hanno permesso la ricostruzione diacronica di un paesaggio complesso, caratterizzato dalla compresenza di differenti attività economiche (pastorizia, attività minerarie e sfruttamento forestale), attraverso le quali si è espressa l'azione umana nell'ambiente montano lungo l'arco di più di un millennio. La metodologia proposta, in quanto sintesi di diverse esperienze di ricerca in ambito alpino, potrebbe porre le basi per una più ampia riflessione riguardo possibili approcci condivisi e comuni ad una "archeologia di montagna", che sempre più si sta delineando come una disciplina autonoma.

Pastorinha é uma expressão teatral cuja realização está relacionada à celebração do Natal. A proposta desta dissertação é apresentar uma descrição etnográfica e análise antropológica tanto para a encenação quanto para os grupos que a fazem, em Belém, na contemporaneidade. Após a apresentação de atores sociais e espaços e de explicação acerca de seu passado histórico e seu formato artístico, começa a descrição do ciclo ritual, tendo por referência, principalmente, informações resultantes de observação direta em duas pastorinhas, A Filha de Sion e Os Filhos de Judá. Eles são grupos temporários, constituídos prioritariamente por adolescentes e dirigidos por mulheres não-casadas maridos e sem filhos pequenos. O processo ritual em questão não só é marcado por caráter anti-estrutural como também proporciona aprendizagem aos menores, no que se refere tanto ao saberes referentes à própria encenação quanto àqueles que as organizadoras julgam necessários para a participação na vida social.
Pastorinha is a theatrical expression whose fulfillment is connected to the Christmas' celebration. This dissertation's proposal is to show ethnographic description and anthropological analysis as much for the staging as for the groups that do it, in the city of Belém in ours days. After the presentation of social actors and spaces and an explanation about its historic past and its artistic shape, the ritual cycle's description starts. To do this, the most important information's source is the direct observation of two pastorinhas: A Filha de Sion and Os Filhos de Judá. They are temporary groups, constituted, mainly, by teenagers and their leaders are women without husband or children. It´s striking the antistructural character of the ritual process. Further on, it proportions apprenticeship as much about the knowledge required to do the staging as other, that the leaders think that are necessary to the inclusion on the social life.

VrD1 (Vigna radiata defensin 1) is a member of the plant defensin family, containing 46 amino acids and four pairs of disulfide bonds. Isolation of a cDNA encoding a small cysteine-rich protein designated VrCRP (then also known as VrD1) from a bruchid-resistant mungbean revealed the first discovered plant defensin exhibiting both in vitro and in vivo insecticidal and antifungal activities. However, the molecular and structural basis of this unique insecticidal activity of VrD1 is still not fully understood. This thesis is divided into two parts: I. Optimized preparation of VrD1 mutants expressed in Pichia pastoris: Based on the structural and sequence alignment, it is suggested that VrD1, in addition to γ-thionins and several amylase inhibitors, is highly homologous to scorpion toxins, especially the short toxins. We have deduced that VrD1 may utilize a newly found cluster of basic residues on one side of VrD1 molecule to achieve its insecticidal function, whereas another cluster of previously identified basic residues located on the other side of the molecule, which is conserved for all γ-thionins, should be used to achieve the antibacterial/antifugual activities for VrD1 and for all other plant defensins. In order to understand the roles of this newly found cluster of conserved basic residues, we have constructed several expression strains for VrD1 mutants and purified these mutant proteins using the Pichia pastoris system. However, the recent re-examination of our VrD1 expression system showed that some of the previous constructions contained unexpected base frame-shift. On the other hand, regarding the parts with correct sequences, the yields of the consequent protein purification were rather low. This may give rise to the difficulty in reaching sufficient sample size in the experiments of whole-cell recordings performed previously. Therefore, in the present study, we focus our work on two aspects. (1) Reconstruction of the appropriate VrD1 mutants with correct sequences. (2) Increasing the expression levels via optimized preparation of VrD1 mutants expressed in pichia. In the past, we have utilized G418-resistance as a selection marker for high gene copy numbers and potentially for a high expression level. However, this strategy could not ensure the satisfactory expression yields. In the present study, we combine the small-scale expression and the western blotting as checkpoints to improve the screening strategy by exclusion of the false-positive G418-resistant transformants. After transformation of the VrD1-pPIC9K plasmids for R26A and K24A mutants into P. pastoris (SMD1168), seven well-grown transformants on 0.5 mg/mL G418/YPD plates were selected for high copy number and high expression level strains. Using this screening strategy, we obtained Y38 and Y43 for the expression of VrD1-R26A. The western blot results demonstrated that the colony Y38 has significantly higher target protein expression than the colony Y43. Results from the large-scale expressions are in line with those from small-scale expressions. Moreover, using the site-directed mutagenesis, we have successfully generated VrD1-K7A from wild-type VrD1. This will be a convenient method in the preparation for other mutant strains with higher insecticidal and antifungal activities in the future. II. Investigation on the membrane target(s) of VrD1 in insect cells and the molecular mechanism of VrD1 insecticidal activity: Base on sequence and structural alignment, we have postulated that VrD1 may utilized a similar interaction mode as short scorpion toxins to act on insect cell membranes with K+-channel or Cl--channels as molecular targets. Preliminary data has excluded Cl--channels for candidates based on electrophysiological experiments. To study the protein-protein interaction for VrD1 and cell membranes, two major aspects need to be considered. 【1】 Generation of large amounts of VrD1 protein, at least a few milligrams for each experiment. Instead of using pichia expression system, we purify native VrD1 from mung bean seeds. About 5~6 milligrams of VrD1 protein can be produced from 500 grams of mung bean seeds. Mass spectrum has confirmed the correctness of VrD1. 【2】 Choice of appropriate cell-lines that allow VrD1 to interact with its membrane target(s). Previous reports showed that VrD1 can lead to a significant inhibitory effect on Sf21 or Sf9 (Spodoptera frugiperda 21 or 9). However, due to the lack of Spodoptera frugiperda genome database, it will be very difficult to identify unknown VrD1-target candidates from membrane extracts. Therefore, Drosophila S2 cell-line was chosen in this study to perform cell survival assay before the investigation with membrane proteomics.
