Abstract:

Abstract:
Abstract Gender is an important stratification and prognostic factor in oncology in the response to treatments, particularly to the immunotherapy. The anti-PD-1/PD-L1 immune-checkpoint inhibitors (Pembrolizumab, Nivolumab, Atezolizumab) have improved the prognosis of non-small cell lung cancer (NSCLC) patients in 20-35% cases, but with a lower efficacy in females. The different steroid hormones - estrogens, progesterone or androgens - present in both gender have often contrasting effects on the activity of the host immune system, partly explaining the differences observed. It is not known if metabolic features of cancer cells of male and female patients may impact on the immune system response and immunotherapy efficacy. We set up an analytical pipeline based on patient-derived samples, cell lines and humanized male and female mice bearing gender-matched NSCLC xenografts. NanoString analysis in a cohort of 42 NSCLC patients of both genders treated with Pembrolizumab, identified the estrogen receptor α (ERα)/ESR1 gene as one of the most significantly associated with the response to Pembrolizumab, particularly in female patients. A panel of 30 human NSCLC cell lines of female and male origin, that reproduced the ERα/gender association observed in clinical samples, the amount of ERα and 17-β-estradiol was directly related to the expression of PD-L1, suggesting a potential direct linkage. ERα transcriptionally up-regulated CD274/PD-L1 gene, with higher effects in females. EGFR downstream effectors Akt and ERK1/2, increased the active form phospho(Ser118)ERα, upregulating PD-L1. Inhibiting the estrogen synthesis or switching off EGFR activity down-regulated PD-L1 levels. Consistently, the aromatase inhibitor letrozole increased the efficacy of Pembrolizumab in humanized NSCLC xenografts, by reducing tumor growth and tumor-associated PD-L1, increasing the infiltrating CD8+T-lymphocytes, NK cells and Vγ9Vδ2 anti-tumor T-lymphocytes. The benefit was maximal in 17-β-estradiol/ERα high female xenografts, intermediate in 17-β-estradiol/ERα medium female and male xenografts, minimal in 17-β-estradiol/ERα low male xenografts. Additionally, an untargeted lipidome analysis revealed that letrozole decreased the saturated fatty acids (SFAs)/mono- or poly-unsaturated fatty acids (MUFAs-PUFAs) ratio, and the cholesterol/cholesterol esters ratio. These changes increased plasma-membrane fluidity, an event that in turns reduced the affinity between PD-L1 and PD-1. As further proof of concept, ω6- or ω3-PUFA treated cells, displayed an increased membrane fluidity and a change in lipid metabolism that affect the response to anti-PD-1/PD-L1 as letrozole did. We propose a prototypical in vitro 3D spheroids culture gender-tailored platform that allows to investigate the metabolic circuitries causing such differences and to repurpose metabolic modifiers as novel immune-adjuvant agents in oncology. Abstract:

Abstract:

Abstract:
Abstract Thomas Watson’s critics have suggested that The Hekatompathia, Or Passionate Centurie of Love ambitiously aspired to be a pedagogical text, but if this work is designed to teach, then this essay suggests Watson’s manipulations of genre, style, and intertexts combine to offer a pedagogy for poets, a compilation of rhetorical postures one may employ to simultaneously deliver and disguise socio-political satire in Elizabethan England. This essay first discusses how Hekatompathia additionally signals its satirical aims by participating in the pasquinade tradition, and positioning a “pasquine piller” at the volta of this sequence of one hundred passions. Next, it shows how Watson’s “passions” intertextually recall Pierre de Ronsard’s Discours des Misères de ce Temps, a collection of lyrics satirizing the French factionalism that has led to civil war, as well as Thomas Jeney’s later English translation that turns a mirror to princes toward Queen Elizabeth. Upon recognizing the Ronsardian subtexts of courtly factionalism and civil unrest associated with Watson’s “passions,” one may see how they are compounded as the poet sets them forth in the “pathetical style” of Seneca and Lucan. The civil wars of ancient Rome and subsequent imperial tyranny are frequently held up as a cautionary tales for early modern English and European rulers, but Watson’s simultaneous translation of the French Wars of Religion relocates these civil broils in England, implicating Elizabethan court dissidence and hypocrisy.

Abstract:
2013 - 2014
If you want to assign a place to Pasquale De Luca in Italian Literature, first you must leave out all his Italian Literature production, so pleasant, from 1900 onwards. This is generally the judgement of the few, who have dealt with him. Different Opinions you may have about the participation of Pasquale De Luca to the Verismo Movement, a regionalist production that, without doubt, must be included in. When Pasquale De Luca moved to Milan, he had already given the best of himself. Later he will elaborate and improve the two most challenging novels AlleportedellaFelicità( At the door of Happiness)and Le Ambiziose (The ambitious women), written long before. It is worth remembering now just this Verismo production, unjustly forgotten, also framing De Luca in the short Neapolitan glimpse of the ‘80s, with his fervid personality and creative imagination. Everyone who worked with him and for him, loved and praised him. Lots of great critics and writers, not only Italian but also foreign ones, gave admired judgements of him. We can find the proof of esteem and affection towards Pasquale De Luca in the pages of A Pasquale De Luca nelsuo primo GiubileoLetterario, where colleagues and friends thought to manifest their affection and their regret at his departure from the direction of the journal Nature and Art, that he had brilliantly directed, spreading with it the culture and the good taste in Italian families to launch himself in the journalism with a new review which would have become his favourite creature. FEDERICO, LUCA. "L'apprendistato letterario di Raffaele La Capria." Doctoral thesis, Università degli studi di Genova, 2020. http://hdl.handle.net/11567/1005664.

Full text
Abstract:

Abstract:
Leopold “wildlifer” Dan Thompson and his research partner were driving south on US Highway 51 on Thursday, April 22, 1948. “When the news of Aldo Leopold’s death came over the radio, we gasped and blankly looked at each other. I turned off the radio, and we drove on for another thirty minutes of silence.” Dan’s thoughts drifted back to the howl of a single wolf he had heard the evening before. He wondered “if the wolf had sensed the passing of a kindred spirit of the wilderness.” Aldo Leopold turned the American Dream upside down. He was born in a mansion and died at a shack; yet the Shack was the home of his greatest riches. Some have dubbed it the most famous chicken coop in the world. Upon his retirement, Aldo had planned to build a year- round home on the property, but instead the Shack was passed on as it was to new generations far beyond his family. Leopold’s book about the Shack and other adventures, “Great Possessions,” was in jeopardy after his death of ever getting published. But comments on the essays were called in from key students, friends, and family members, Luna served as the editor for his father’s writings, and the book was released by Oxford University Press under the title A Sand County Almanac and Sketches from Here and There (1949). In it, Aldo Leopold invited his readers to sit with him on his bench at the Shack, watch as he cut wood, walk with him in the woods and prairies, and loiter at his campfires to think about the question: How can we live on the land without spoiling it? And now even broader: How can we live as part of the earth, and its systems, without spoiling it? The urgency of his questions has only heightened since his death. Leopold anticipated our crises and left us the practical and philosophical wisdom to set our compasses and choose paths toward healthier vistas.
