Full textAs the long term effects of atmospheric particles (PM) are misunderstood, we develop an experimental strategy using primary culture of human bronchial epithelial repeatedly exposed to PM to investigate (1) whether particles could contribute to airway remodelling by inducing sustained inflammation and mucus secretion (2) the impact of the epithelial secretions on the proliferation and the differentiation of the bronchial fibroblasts. Observations revealed that ambient particles are still present in the bronchial epithelium 5 weeks after treatments, and a part of the organic compounds of particles were metabolized by cells. We also showed an induction of the inflammatory response after particles treatment which persists with the secretion of several pro-inflammatory cytokines. The secretion of growth factors ligand was observed after the end of treatments, and this secretion may contribute to the mucous differenciation. Moreover, epithelial secretions may induce an increase of the number of fibroblasts which in co-culture. We showed that EGFR pathway and these ligands were involved in fibroblasts proliferation. Our results suggest that bronchial epithelial cells repeatedly exposed to ambient particles exhibit a sustained pro- inflammatory response and evolve towards a mucous phenotype. The respiratory epithelium exposed to particles would so contribute to the remodeling airways through the secretion of growth factors which could be involved in the fibroblast proliferation

Full textThis work fits into a research program on the deposit of fibrous particles in a pulmonary pattern with several branching. The first step consisted in elaborating a manufacturing method of a bronchial pattern according to WEIBEL's one. For the aeraulic study, we undertook to carefully study the characteristics of the outflow in inspiratory and expiratory phases for the instantaneous flows usually used in pulmonary ventilation. A DISA anemometric line allowed us to draw not only the speed profiles but also cards of turbulence rates. In the following step, we elaborated a method of measurement of compact particles deposit. The measures of deposit permitted to check and improve the laws of deposit by sedimentation and impact proposed in the literature. From a local deposit point of view, we have shown that these mechanisms of particles stops usually used in aerosol physics are not sufficient. This problem was settled starting from the quantitative description of the outflow and communing these mechanisms with a contribution due to the local turbulence of the outflow. In case of fibres, the measures of weight and fractional outputs show that the deposit is favoured for the weak flows, contrarily to speric particles. Full textIndoor air pollution is an important cause of diverse health problems for occupants including respiratory diseases, allergic symptoms, cancers, and cardiovascular problems. People spend 80-90% of their time indoors emphasizing the importance of understanding the main causes of indoor air pollution and finding suitable solutions to improve indoor air quality. The World Health Organization (WHO) reported in 2009 that biological pollution is one of the main causes of the degradation of indoor quality. In humid indoor environments, fungi can grow and develop on almost all building materials. Upon their development on surfaces, they produce particles such as spores, toxins or volatile organic compounds, and other metabolites that can be aerosolized. The release of particles from surfaces as well as their characteristics is dependent on different factors including the type and age of microorganisms on the surface, material properties, and environmental conditions (temperature, humidity, air velocity). Based on their sizes and their duration of availability in the indoor air, they can be inhaled by occupants, leading to serious health risks as they may penetrate different depths in the respiratory system. To respond to these issues, this work aimed to investigate the relations that may exist between microorganisms, especially fungi, present on surfaces and airborne particles contaminating the indoor air (In situ and in the lab). It would provide a clearer vision of fungal contamination in the indoor environment and would be helpful during investigating methods for controlling the growth of fungi indoors. In addition, investigations on the performances of selected photocatalysts and glycerol esters as antimicrobial products for reducing microbial contamination on surfaces in the laboratory were achieved. Initially, an In situ sampling campaign was held in a visibly contaminated house in Southern France. Surface sampling from contaminated materials and indoor airborne sampling were carried out. Then, DNA analysis for samples was carried out by Toulouse Biotechnology Institute (TBI). This part aimed to check the relationship, if any, between fungal contamination on surfaces and airborne contamination. Then, experiments on the antifungal and antibacterial activities of photocatalytic products and the antifungal activity of glycerol esters were carried out under different experimental conditions. This part aimed to test their efficiency as antibacterial/antifungal products that may be applied as additives in paints or coatings. The tests carried out in this part were preliminary tests in aqueous solutions and the impact of different conditions on the efficiency of each product was evaluated. The efficiency of three photocatalysts (TiO2, ZnO, Au-decorated ZnO) as antimicrobial products under light intensities close to real-life indoor conditions. The efficiencies of Undecylenic Acid (UA) and of three glycerol esters, formed upon esterification of Glycerols with UA, as antifungal products were evaluated. The last part of this study aimed to investigate the relationship between surface contamination and airborne contamination under controlled conditions (mainly airflow and relative humidity). A new experimental device was settled, design and validation of an experiment protocol that permit to determine the quantitative correlation between surface contamination and airborne contamination were carried out. The impact of different experimental parameters on the aerosolization of particles from contaminated surfaces into air was evaluated. The validation of this experimental protocol was important and brought attention to different tests that will be investigated in the near future

Full textThis thesis focuses on the study of autonomous aerial robots interacting with the surrounding environment, and in particular on the design of new control and motion planning methods for such systems. Nowadays, autonomous aerial vehicles are extensively employed in many fields of application but mostly as autonomously moving sensors. On the other hand, in the recent field of aerial physical interaction, the goal is to go beyond sensing-only applications and fully exploit the aerial robots capabilities in order to interact with the environment. With the aim of achieving this goal, this thesis considers the analysis of a particular class of aerial robots interacting with the environment: tethered aerial vehicles. This work focuses on the thorough formal analysis of tethered aerial vehicles ranging from control and state estimation to motion planning. In particular, the differential flatness property of the system is investigated, finding two possible flat outputs that reveal new capabilities of such system for the physical interaction. The theoretical results were finally employed to solve the challenging problem of landing and takeoff on/from a sloped surface. In addition, moved by the interest on aerial physical interaction from A to Z, we addressed supplementary problems related to the design, control and motion planning for aerial manipulators

Full textLow visibility conditions have a strong impact on airports' traffic. COBEL-ISBA is a 1D model used operationally to provide short-range forecasts of low visibility conditions. Initial condition play a crucial role in the quality of the forecasts. This PhD focused on a diagnosis and optimization of the assimilation scheme of COBEL-ISBA. The impact of the several kinds of observations on the quality of the initial conditions and forecasts was first assessed. Simulated observations were used, in order to cancel the effect of the model error, as well as real observations. The observations that have most impact on the quality of the forecast of fog events are the ones that estimate the initial thickness of the cloud layer and those of soil water content. The operationnal assimilation scheme uses error statistics for the background that are fixed in time and over the vertical. They were shown to have large variations over time and the vertical. These conclusions led to the implementation of an ensemble Kalman Filter (EnKF), which is able to estimate background error statistics at each assimilation cycle. It gave good results with simulated observations, and also with real observations for forecast hours smaller than four hours. Several studies showed the potential of particle filter methods for data assimilation. A particle filter with a genetic selection was adapted to be used with COBEl-ISBA. This new assimilation scheme gave good results simulated observations. It also worked with relatively few particles
