Abstract:
Background: Oxidative stress (OS) could cause various COVID-19 complications. Recently, we have developed the Pouvoir AntiOxydant Total (PAOT®) technology for reflecting the total antioxidant capacity (TAC) of biological samples. We aimed to investigate systemic oxidative stress status (OSS) and to evaluate the utility of PAOT® for assessing TAC during the recovery phase in critical COVID-19 patients in a rehabilitation facility. Materials and Methods: In a total of 12 critical COVID-19 patients in rehabilitation, 19 plasma OSS biomarkers were measured: antioxidants, TAC, trace elements, oxidative damage to lipids, and inflammatory biomarkers. TAC level was measured in plasma, saliva, skin, and urine, using PAOT and expressed as PAOT-Plasma, -Saliva, -Skin, and -Urine scores, respectively. Plasma OSS biomarker levels were compared with levels from previous studies on hospitalized COVID-19 patients and with the reference population. Correlations between four PAOT scores and plasma OSS biomarker levels were analyzed. Results: During the recovery phase, plasma levels in antioxidants (γ-tocopherol, β-carotene, total glutathione, vitamin C and thiol proteins) were significantly lower than reference intervals, whereas total hydroperoxides and myeloperoxidase (a marker of inflammation) were significantly higher. Copper negatively correlated with total hydroperoxides (r = 0.95, p = 0.001). A similar, deeply modified OSS was already observed in COVID-19 patients hospitalized in an intensive care unit. TAC evaluated in saliva, urine, and skin correlated negatively with copper and with plasma total hydroperoxides. To conclude, the systemic OSS, determined using a large number of biomarkers, was always significantly increased in cured COVID-19 patients during their recovery phase. The less costly evaluation of TAC using an electrochemical method could potentially represent a good alternative to the individual analysis of biomarkers linked to pro-oxidants.

Abstract:
For the purpose of human disease prevention, several methods have been developed, and are still developing, to assess the oxidative stress status (OSS) of individuals. In the present paper, we describe an approach based on electrochemical detection able to evaluate skin oxidative stress status (SOSS) as a PAOT (Pouvoir AntiOxydant Total)-Skin Score®. Normal reference values for the PAOT-Skin Score® were: 0–62.94 (n = 263). Intra- and inter-assay coefficients of variation were, respectively, 12.47 ± 4.29% and 7.0 ± 2.5%. Our technology showed increased skin antioxidant activity following topical application of reduced coeznyme Q10 cream or vitamin C intake as orange juice or supplements. Moreover, we found significant correlations between some blood oxidative stress biomarkers and the PAOT-Skin Score® (γ-tocopherol/α-tocopherol ratio (r = 0.43, p = 0.020); copper (r = −0.42, p = 0.022); copper/zinc ratio (r = −0.49, p = 0.006), and lipid peroxides (r = −0.43, p = 0.002)). In addition to being non–invasive, the present electrochemical methodology is also not expensive, fast, and easy to use.

Polyphenol compounds present in high quantity in wines are well-known to have potent cardio-protective properties through several biological mechanisms including antioxidant activity [1]. A large number of methods have been developed for evaluating the antioxidant capacity of food matrices. Most of them have, however, the disadvantage of being time consuming and require specific analytical protocols and devices. In the present study, we present the electrochemical PAOT (Pouvoir Antioxydant Total)-Liquid® Technology which can be easily used by winemakers for evaluating antioxidant activity of wine during all steps of making process. The methodology is based on the measurement of electric potential variation resulting from chemical reactions between wine polyphenols and a free radical mediator M• as source of oxidants. Total antioxidant activity as estimated by the PAOT-Liquid® activity was 6.8 fold higher in red wines (n = 14) when compared to rosé (n = 3) and white (n = 3) wines bought in a commercial market. Moreover, PAOT-Liquid® activity was highly correlated with total polyphenols content (TPC) of all wines (r = 0.9540, p < 0.0001) and the classical DPPH (2,2-diphenyl-1-picryhydrazyl) assay which is often used for evaluating antioxidant capacity of food matrices (r = 0.9102, p < 0.0001).

Abstract Pleomorphic adenoma of the breast is a rare, benign tumor accounting for 68 cases in the literature. It is most commonly seen in postmenopausal women and is characterized by an admixture of epithelial and myoepithelial cells embedded in abundant myxomatous stroma. Its clinical and histologic appearance can be challenging and may lead to a misdiagnosis of invasive carcinoma. We report a case of mammary pleomorphic adenoma in an asymptomatic 59-year-old woman and briefly discuss its distinction from mucinous carcinoma through the use of special stains.

Abstract Context.—Primary bladder angiosarcomas are extremely rare, and their clinical and pathologic features are not well described. Objective.—To further refine the clinical features of primary bladder angiosarcomas and define their pathologic spectra. Data Sources.—Relevant sources were identified using MEDLINE and a subsequent bibliographic search of all pertinent reports and reviews. We also searched the M. D. Anderson pathology archives. Study Selection.—After excluding 4 cases that likely secondarily involved the bladder, we identified 9 true primary bladder angiosarcomas. Data Extraction.—Data were extracted on the following: demographics, clinical presentation, predisposing factors, gross pathology, microscopic pathology, immunophenotype, therapy, and outcomes. Data Synthesis.—Primary bladder angiosarcomas were found at a mean age of 64.2 years, with a male-female ratio of 8:1. Two cases arose in a postirradiation setting. Primary bladder angiosarcomas typically presented with hematuria and were grossly hemorrhagic, raised masses (mean size, 6.7 cm) of the trigone and/or dome. Histologically, most showed classic anastomosing channels lined by plump hyperchromatic cells, though many showed variant histology such as solid growth and epithelioid cytology. Three (43%) of 7 patients died within a year, but only 1 patient died with evidence of disease. The remaining patients were alive at the time of publication of their respective cases (mean, 22 months). Conclusions.—Primary angiosarcomas of the bladder are typically rare tumors of middle-aged and elderly men that present with locally advanced disease and show a wide histologic spectrum. However, their prognosis may be better than previously thought.

Trans-resveratrol is a natural polyphenol showing numerous biological properties, especially anti-tumoral and antioxidant activity. Among numerous resveratrol derivatives, aza-stilbenes, which bear an imine bound, show interesting biological activities. In the present study, we synthesized a series of imine analogs of trans-resveratrol (seven aza-stilbenes) following an easy and low-cost procedure of green chemistry. The toxicity of synthesized aza-stilbenes, which is currently unknown, was evaluated on murine neuronal N2a cells, comparatively to trans-resveratrol, by considering: cell density evaluated by staining with sulforhodamine 101; esterase activity, which is a criteria of cell viability, by staining with fluorescein diacetate; and transmembrane mitochondrial potential, which is known to decrease during cell death, by staining with DiOC6(3) using flow cytometry. In addition, the antioxidant activity was quantified with the KRL (Kit Radicaux Libres) assay, the DPPH (2,2′-diphenyl-1-picrylhydrazyl radical) assay and the FRAP (ferric reducing antioxidant power) assay. The PAOT (Pouvoir Antioxidant Total) score was also used. The aza-stilbenes provide different cytotoxic and antioxidant activities, which are either higher or lower than those of trans-resveratrol. Based on their cytotoxic and antioxidant characteristics, all synthesized aza-stilbenes are distinguished from trans-resveratrol.

The release of organic contaminants has grown to be a major environmental concern and a threat to the ecology of water bodies. Persulfate-based Advanced Oxidation Technology (PAOT) is effective at eliminating hazardous pollutants and has an extensive spectrum of applications. Iron-based metal–organic frameworks (Fe-MOFs) and their derivatives have exhibited great advantages in activating persulfate for wastewater treatment. In this article, we provide a comprehensive review of recent research progress on the significant potential of Fe-MOFs for removing antibiotics, organic dyes, phenols, and other contaminants from aqueous environments. Firstly, multiple approaches for preparing Fe-MOFs, including the MIL and ZIF series were introduced. Subsequently, removal performance of pollutants such as antibiotics of sulfonamides and tetracyclines (TC), organic dyes of rhodamine B (RhB) and acid orange 7 (AO7), phenols of phenol and bisphenol A (BPA) by various Fe-MOFs was compared. Finally, different degradation mechanisms, encompassing free radical degradation pathways and non-free radical degradation pathways were elucidated. This review explores the synthesis methods of Fe-MOFs and their application in removing organic pollutants from water bodies, providing insights for further refining the preparation of Fe-MOFs.

Background: A key role of oxidative stress has been highlighted in the pathogenesis of COVID-19. However, little has been said about oxidative stress status (OSS) of COVID-19 patients hospitalized in intensive care unit (ICU). Material and Methods: Biomarkers of the systemic OSS included antioxidants (9 assays), trace elements (3 assays), inflammation markers (4 assays) and oxidative damage to lipids (3 assays). Results: Blood samples were drawn after 9 (7–11) and 41 (39–43) days of ICU stay, respectively in 3 and 6 patients. Vitamin C, thiol proteins, reduced glutathione, γ-tocopherol, β-carotene and PAOT® score were significantly decreased compared to laboratory reference values. Selenium concentration was at the limit of the lower reference value. By contrast, the copper/zinc ratio (as a source of oxidative stress) was higher than reference values in 55% of patients while copper was significantly correlated with lipid peroxides (r = 0.95, p < 0.001). Inflammatory biomarkers (C-reactive protein and myeloperoxidase) were significantly increased when compared to normals. Conclusions: The systemic OSS was strongly altered in critically ill COVID-19 patients as evidenced by increased lipid peroxidation but also by deficits in some antioxidants (vitamin C, glutathione, thiol proteins) and trace elements (selenium).

Abstract:
Husk powder was prepared from seven varieties of walnut fruit and their hulling rate, chemical compounds, and total phenolic contents were evaluated. The apolar and polar extracts were prepared, respectively, from hexane and a hydroethanolic solvent, while qualitative and semi-quantitative analyses were performed by GC/MS and UHPLC-PDA-HRMS/MS. The antioxidant, antimicrobial, and antitumor properties of green walnut husk were also evaluated. The total content of phenolic compounds varied between the varieties, ranging from 35.2 ± 0.9 to 58.0 ± 0.0 mg/g gallic acid equivalent of dry husk weight (dw). The apolar extract was found to contain alkanes, tocopherols, sterols, and fatty acids, including oleic, linoleic, and linolenic, while the polar extract showed the presence of phenolics including salicylate glucuronide, taxifolin, catechin, and quercetin isomers. The antioxidant power obtained by the PAOT (total antioxidant power) method for the husk powders ranged from 256.5 ± 5.9 to 746.8 ± 6.9 score/g dw, and seemed consistent with the total phenolic content and the results obtained by the classic antioxidant test with DPPH. The walnut husk also showed an antibacterial effect against Gram-negative and Gram-positive bacteria and cytotoxic potential against HepG2. Among the selected varieties, the green Saman had the highest antioxidant properties, while the Saman with a brown color had the lowest.

Mobile edge computing (MEC) has been envisaged as one of the most promising technologies in the fifth generation (5G) mobile networks. It allows mobile devices to offload their computation-demanding and latency-critical tasks to the resource-rich MEC servers. Accordingly, MEC can significantly improve the latency performance and reduce energy consumption for mobile devices. Nonetheless, privacy leakage may occur during the task offloading process. Most existing works ignored these issues or just investigated the system-level solution for MEC. Privacy-aware and user-level task offloading optimization problems receive much less attention. In order to tackle these challenges, a privacy-preserving and device-managed task offloading scheme is proposed in this paper for MEC. This scheme can achieve near-optimal latency and energy performance while protecting the location privacy and usage pattern privacy of users. Firstly, we formulate the joint optimization problem of task offloading and privacy preservation as a semiparametric contextual multi-armed bandit (MAB) problem, which has a relaxed reward model. Then, we propose a privacy-aware online task offloading (PAOTO) algorithm based on the transformed Thompson sampling (TS) architecture, through which we can (1) receive the best possible delay and energy consumption performance, (2) achieve the goal of preserving privacy, and (3) obtain an online device-managed task offloading policy without requiring any system-level information. Simulation results demonstrate that the proposed scheme outperforms the existing methods in terms of minimizing the system cost and preserving the privacy of users.

Percetakan XYZ merupakan salah satu UMKM yang memproduksi tulisan atau gambar untuk kepentingan periklanan, salah satunya adalah spanduk. Percetakan tersebut mempunyai beberapa permasalahan terkait kondisi kerja serta belum adanya kesadaran dalam menerapkan Keselamatan dan Kesehatan Kerja (K3) di tempat kerja. Tujuan penelitian ini adalah melakukan evaluasi kondisi kerja serta merancang usulan perbaikan untuk kondisi kerja yang lebih produktif dan sesuai dengan pedoman K3 di percetakan XYZ. Program Work Improvement in Small Enterprises (WISE) dengan pendekatan Participatory Action Oriented Training (PAOT) dilakukan dengan melibatkan partisipasi aktif dari pemilik usaha serta pekerja dalam melakukan evaluasi kondisi kerja, pembuatan rencana aksi perbaikan serta implementasi usulan perbaikan. Hasil evaluasi kondisi kerja menggunakan daftar periksa WISE, menunjukkan terdapat 13 elemen periksa yang membutuhkan suatu usulan perbaikan. Melalui metode focus group discussion (FGD) didapatkan hasil bahwa perbaikan yang akan diprioritaskan untuk diimplementasikan dalam waktu dekat adalah 4 elemen periksa karena mempertimbangkan faktor biaya, waktu, dan sumber daya manusia di lapangan. Perbaikan pertama adalah pembuatan jadwal rutin untuk pemeriksaan mesin serta jaringan kabel listrik dengan penerapan 5R. Perbaikan kedua adalah pembuatan tanda bahaya pencegahan kebakaran. Perbaikan ketiga adalah pembuatan tanda himbauan pemakaian pelindung diri masker dan sarung tangan. Perbaikan keempat adalah pembuatan label untuk menuliskan identitas produk sehingga memudahkan pekerja mengenali produk yang berada di area penyimpanan. Evaluasi serta usulan perbaikan kondisi kerja tersebut dapat membantu UMKM dalam meningkatkan kesadaran akan K3 dan mengurangi risiko terjadinya kecelakaan kerja.

Preterm infants are highly susceptible to oxidative stress due to an imbalance between endogenous oxidant and antioxidant systems. In addition, these newborns are frequently fed with donor milk (DM) treated by Holder pasteurization (HoP) at 62.5 °C for 30 min, which is known to alter numerous heat-sensitive factors, including some antioxidants. High hydrostatic pressure (HHP) processing was recently proposed as an innovative method for the treatment of DM. The present study aimed to measure the redox balance of HoP- and HHP-DM and to study, in vivo, the effects of HoP- and HHP-DM on the gut and liver. H2O2, vitamin A and vitamin E (α- and γ-tocopherols) concentrations, as well as the total antioxidant capacity (TAC), were measured in raw-, HoP- and HHP-DM. The gene expression level of antioxidant systems and inflammatory response were quantified in the ileum and liver of adult mice after 7 days of oral administration of HoP- or HHP-DM. HoP reduced the γ-tocopherol level, whereas HHP treatment preserved all vitamins close to the raw milk level. The milk H2O2 content was reduced by HHP but not by HoP. The total antioxidant capacity of DM was reduced after HHP processing measured by PAOT-Liquid® technology but was unaffected after measurement by ORAC assay. In mice, HHP-DM administration induced a stimulation of antioxidant defenses and reduced some inflammatory markers in both the ileum and liver compared to HoP-DM treatment. Our preliminary study suggests that the HHP processing of DM may better protect preterm infants from gut and liver pathologies compared to HoP, which is currently used in most human milk banks.

AbstractThe unique optical properties of metal nitrides enhance many photoelectrical applications. In this work, a novel photodetector based on TiO2/TiN nanotubes was deposited on a porous aluminum oxide template (PAOT) for light power intensity and wavelength detection. The PAOT was fabricated by the Ni-imprinting technique through a two-step anodization method. The TiO2/TiN layers were deposited by using atomic layer deposition and magnetron sputtering, respectively. The PAOT and PAOT/TiO2/TiN were characterized by several techniques such as X-ray diffraction (XRD), scanning electron microscope (SEM), and energy dispersive X-ray (EDX). The PAOT has high-ordered hexagonal nanopores with dimensions ~ 320 nm pore diameter and ~ 61 nm interpore distance. The bandgap of PAOT/TiO2 decreased from 3.1 to 2.2 eV with enhancing absorption of visible light after deposition of TiN on the PAOT/TiO2. The PAOT/TiO2/TiN as photodetector has a responsivity (R) and detectivity (D) of 450 mAW-1 and 8.0 × 1012 Jones, respectively. Moreover, the external quantum efficiency (EQE) was 9.64% at 62.5 mW.cm−2 and 400 nm. Hence, the fabricated photodetector (PD) has a very high photoelectrical response due to hot electrons from the TiN layer, which makes it very hopeful as a broadband photodetector.

Based on the tourism carrying capacity assessment for the protection of the coastal area to explore Tanjung Bira beach as the study area and attempt to assess the optimum number of visitors that should be allowed without damaging the surrounding ecological, social and cultural environments by using the PAOT (people at one time) approach. The results of the study showed the Real Carrying Capacity (RCC) should be 202 beach user/day and the Effective Carrying Capacity (ECC) should be 117 beach user/day. Although there was a significant difference between physical-ecological and social-cultural carrying capacity, this study suggests that the physical-ecological carrying capacity may be applied for ecosystem management, whilst the Social-cultural carrying capacity may be addressed when management objectives are tourists and beach user.

En los últimos veinte años es más que evidente el proceso de verticalización de la oferta inmobiliaria residencial en la Ciudad de México. A la par de este proceso, existe una creciente preocupación social por los cambios en la ciudad y en los barrios. La transformación de la urbe ha generado diversas reacciones. En este artículo se presenta una lectura de éstas desde un punto poco analizado: la institucionalización de la participación ciudadana. Para ello se usan datos de la transformación de la oferta inmobiliaria en la Ciudad de México de 1990 a 2017 y las denuncias presentadas ante la Procuraduría Ambiental y de Ordenamiento Territorial (PAOT). Los resultados indican que, a la par de la transformación de la oferta inmobiliaria (mayor densidad y en altura), también las denuncias crecen. Este proceso podemos equiparlo al denominado No en mi patio trasero (NIMBY, en inglés).
