Abstract:
The Kirschner-Panetta model describes the poblational competition between effector cells and tumor cells. We analize external changes in the parameters and mechanisms to obtain the decreasing of tumor cells. These variations were performed by three different ways: Oscillations, spikes with the natural frequency of the system, and spikes with Normal Distribution. It was observed that the amount of tumor cells decreases to zero if we change simultaneously the parameters properly.

Immunotherapy with Interleukin-2: A Study Based on Mathematical ModelingThe role of interleukin-2 (IL-2) in tumor dynamics is illustrated through mathematical modeling, using delay differential equations with a discrete time delay (a modified version of the Kirshner-Panetta model). Theoretical analysis gives an expression for the discrete time delay and the length of the time delay to preserve stability. Numerical analysis shows that interleukin-2 alone can cause the tumor cell population to regress.

The dynamics of the model for tumor-immune system competition with negative immune response and with one delay investigated. We show that the asymptotic behavior depends crucially on the time delay parameter. We are particularly interested in the study of the Hopf bifurcation problem to predict the occurrence of a limit cycle bifurcating from the nontrivial steady state, by using the delay as a parameter of bifurcation. The obtained results provide the oscillations given by the numerical study in M. Gałach (2003), which are observed in reality by Kirschner and Panetta (1998).

Kandidaatintutkielmani aihe on panna-verbin vokaalivartaloistuminen. Tutkin, mitä panna-verbillä ilmaistaan, kun se on norminvastaisesti vokaalivartaloinen. Panna-verbillä on sekä vokaalivartalo- että konsonanttivartaloesiintymiä. Aineistoni on kerätty Korp-sivuston internetkeskustelukorpuksista, joissa on aineistoa Suomi24- ja Ylilauta-sivustoilta. Panna-verbi on hyvin polyseeminen verbi. Erityisesti kiinnitän huomiota niihin tapauksiin, joissa panna-verbiä käytetään seksuaalisessa merkityksessä. Tutkimukseni hypoteesi onkin, että panna-verbin vokaalivartaloisia muotoja esiintyy erityisesti silloin, kun verbillä halutaan ilmaista seksuaalista merkitystä. Analysoin aineiston tapauksia sekä muodon että merkityksen kautta. Aineistossani oli selkeästi eniten tapauksia, joissa panna-verbiä käytetään seksuaalimerkityksessä. Pohdin myös, onko panna-verbin taivutusparadigmassa tapahtumassa muutos polysemian takia.

A pesquisa descreve as práticas sociais no Presídio Feminino de Florianópolis, SC. O presídio corresponde a uma das unidades prisionais do sistema penitenciário de Santa Catarina, único exclusivo para a detenção de mulheres. A pesquisa perseguiu o objetivo de identificar como se organizam as mulheres presas em Florianópolis, como se relacionam e que práticas coletivas são encenadas nesse contexto. A população de mulheres presas envolvidas na pesquisa foi, inicialmente, de 36 mulheres, que haviam sido julgadas e receberam sentença de reclusão em regime fechado. Entretanto, ao longo do trabalho de campo esse número foi se modificando e foi possível conhecer em torno de 100 mulheres que passaram pelo presídio no período em que foi realizada a pesquisa. Reconheceu-se que há no Brasil uma incipiente tentativa de dar visibilidade a questões que se incorporam no cotidiano prisional, sobretudo nos presídios para mulheres. Três autoras brasileiras são destaques como inovadoras nessa área: Julita Lemgruber (1983); Iara Ilgenfritz e Bárbara M. Soares (2002). No exterior merece destaque Manuela Ivonne da Cunha (2002), na realização de uma etnografia de um presídio de mulheres em Lisboa/PT. De sorte que, nessa pesquisa foi necessário articular vários entendimentos até que pudéssemos, a partir dos dados etnográficos, identificar que as relações presa/instituição e prisão/violência - embora de extrema relevância - não dão conta de representar o presídio feminino de Florianópolis tal qual é possível percebe-lo: um lugar de convivência e, portanto, de sociabilidades que se sobrepõem muitas vezes às práticas coercitivas ali presentes, legitimadas pela sociedade em geral. Foi identificado que a experiência prisional reedita algumas das práticas já encenadas por essas mulheres, mas dá ênfase a algumas relacionadas ao parentesco, a rituais de iniciação na vida prisional e ao estabelecimento de confiança entre pares. Fundamentalmente, o estudo indica que o fluxo de informações, pessoas, objetos e desejos, se dá de forma a diminuir, ou até mesmo apagar a fronteira entre o dentro e o fora da prisão, mesmo que, em muitos momentos, essa fronteira seja decisiva no destino de cada uma das mulheres que ali se encontram. This research describes social practices in the Female Penitentiary in Florianópolis, SC. The penitentiary is one of the units of the penitentiary system in Santa Catarina being the single one for imprisonment of women in the State. The research aimed in identifying how the imprisoned women organize themselves, how they relate to each other and which collective actions are performed thereof. The number of imprisoned women involved in the research started off as 36 - all had undergone trial and had received imprisonment sentence (closed regimen). However, as the research developed, this number changed and reached about 100 women who passed by the penitentiary during the research period. It was acknowledged that, in Brazil, there is a crude attempt in showing issues related to the daily life in penitentiaries, especially in female penitentiaries. Three Brazilian writers are highlighted as innovative in this field: Julita Lemgruber (1983); Iara Ilgenfritz and Bárbara M. Soares (2002). A writer with prominence in a foreign country is Manuela Ivonne da Cunha (2002), on an ethnography performed in a female penitentiary in Lisbon (Portugal). Therefore, this research had to undergo several understandings to the point where we were able to identify, from ethnographic data, the relationship between prisoner/institution and prison/violence that - although having great relevance - do not duly represent the female penitentiary in Florianópolis as it is in real life: a place where they live, and thus a sociability place that in many instances overcome the coercive practices present, justified by society in general. It was identified that the penitentiary experience re-edits some practices already performed by these women, but it emphasizes some practices related to family ties, initiation rituals in penitentiary life as well as the establishment of trust amongst pairs. Basically, the study shows that flow of information, people, object and desires take place in order to decrease, or even erase, the borderline of inside and outside of prison, even if in some moments, this borderline is decisive in the fate of each of the women there.

In order to minimize the effects caused by the development of Brazil, such as the suppression of riparian forest, one has searched for tree species that have the characteristics needed for the recovery of degraded areas. Given this, studies focus on higher plants of encapsulated wild fruits, especially the Mabea fistulifera Mart. Considering that M. fistulifera reproduces through embryonic units that need to be treated, this study aimed to evaluate the drying of wild fruits with explosive dehiscence and the coating "in natura" seed with gel mixture via coating pan, as well as evaluating the tolerance of embryos related to the immediate effects of the processes. Thus, the experimental procedure consisted of the collection of capsular fruits and extraction of seeds of fruit by drying so were analyzed drying methods via radiation, natural convection, forced convection and coating pan. It was considered for the drying methods analysis the opening fruit speed and seed vigor. Thus, all processes allowed physiological recovery of embryos, but the extraction method via pan promoted the highest opening speed of the fruits. Once performed the extraction, the seeds were separated from the inert material obtaining pure batches of seeds. There were also characterizations of size, shape and specific mass obtaining the results (0,78±0,05) cm, (0,67±0,04) cm and (0,59±0,04) cm for Dmax, Dmédio and Dmim, respectively, form factor (0,75±0,01), specific mass of (1,20±0,01) g/cm³ and apparent mass of (0,82±0,01) g/cm³. Analyzes were also made for chemical seed yielding (82,93±0,11)% of dry matter (19,03±0,78)% crude protein, (1,18±0,11) g/kg calcium (0,65±0,03) g/kg magnesium (0,58±0,04) g/kg phosphorus (13,45±0,33) g/kg potassium (0,70±0,03) g/kg of sulfur. The process of coating seeds, through reaction between sodium alginate gel and calcium chloride salt at different angles of inclination and rotation speed of the coating pan, was also analyzed. Thus, it was found that to achieve the coating of the particles it was necessary to prepare the particle surface by chemical scarification, in order to increase the surface roughness of the seed, followed by rehydration with saturated calcium chloride. This way, it was found that the angle of 25° and speed of 25 rpm generate the greatest quantity of mass adhered on seeds. In addition, tests were performed to verify the efficiency of the coating process, and with the lowest angle set in experimental design was found higher values of efficiency. In tests of particle growth was evaluated that the process time was approximately 8 seconds. Finally, the analysis and processing of data obtained through statistical techniques and equations from literature were performed. In this way, the results contributed to improvements in drying processes of wild fruits and the coating of seeds of the pioneer species M. fistulífera.
A fim de minimizar os efeitos causados pelo desenvolvimento do Brasil, tal como, a supressão da Mata Ciliar, buscam-se espécies arbóreas que tenham as características necessárias para a recuperação de áreas degradadas. Diante disto, estudam-se plantas superiores de frutos encapsulados silvestres, das quais se destaca a Mabea fistulífera Mart.. Tendo em vista que a M. fistulífera se reproduz através de unidades embrionárias que precisam ser tratadas, este estudo teve como objetivo avaliar a secagem de frutos silvestres com deiscência explosiva para extração das sementes e do recobrimento das sementes com mistura gel via drageadora, bem como a avaliação da tolerância dos embriões em relação aos efeitos imediatos dos processos. Assim, o procedimento experimental consistiu na coleta dos frutos capsulares e extração das sementes através da secagem dos frutos, de modo que foram analisados os métodos de secagem via radiação solar, convecção natural, convecção forçada e via secagem em drageadora. Considerou-se para a análise dos métodos de secagem a velocidade de abertura dos frutos e qualidade fisiológica das sementes. Assim sendo, verificou-se que todos os métodos permitiram a recomposição fisiológica dos embriões, porém a secagem via drageadora ocorreu com menor tempo de processo em relação às demais técnicas. Uma vez realizada a extração das sementes, estas foram separadas dos materiais inertes obtendo-se lotes puros de sementes. Realizaram-se também as caracterizações de tamanho, forma e massa específica obtendo-se os resultados de (0,78±0,05) cm, (0,67±0,04) cm e (0,59±0,04) cm para os Dmáx, Dmédio e Dmim, respectivamente, fator de forma de (0,075±0,01), massa específica real de (1,20±0,01) g/cm³ e massa específica aparente de (0,82±0,01) g/cm³. Foram feitas também análises para caracterização química das sementes obtendo-se (82,93±0,11)% de matéria seca, (19,03±,078)% de proteína bruta, (1,18±0,11) g/kg de cálcio, (0,65±0,03) g/kg de magnésio, (0,58±0,04) g/kg de fósforo, (13,45±0,33) g/kg de potássio e (0,70±0,03) g/kg de enxofre. Analisou-se também o processo de recobrimento das sementes via reação entre o gel alginato de sódio e o sal cloreto de cálcio em diferentes ângulos de inclinação e velocidade de rotação da panela rotatória (drageadeira). Verificouse que para realizar o recobrimento das partículas foi necessário preparar a superfície das partículas através de escarificação química, visando ao aumento na rugosidade da superfície das sementes, e posterior reumidificação com solução saturada de cloreto de cálcio. Desta forma, constatou-se que o ângulo de 25° e a velocidade de 25 rpm possibilitaram maior quantidade de massa aderida sobre as sementes. Além disto, realizaram-se testes a fim de verificar a eficiência do processo de recobrimento, sendo que nos níveis mais baixos do planejamento experimental verificaram-se os maiores valores de eficiência. Nos testes de crescimento das partículas avaliou-se o tempo de processo que foi aproximadamente de 8 segundos. Por fim, foi realizada a análise e o tratamento dos dados obtidos, mediante técnicas estatísticas e equações advindas da literatura. Desta forma, com os resultados obtidos colaborou-se para avanços nos processos de secagem dos frutos silvestres e no recobrimento das sementes da espécie pioneira M. fistulífera.

Introduction: Mutations in the NOD2 gene are associated with the development of Crohn's disease, an inflammatory disorder of the gastrointestinal tract. The NOD2 protein induces cellular activation in response to the bacterial antigen muramyl dipeptide (MDP). The NOD2 gene is mainly expressed by circulating blood monocytes although NOD2-associated Crohn's disease involves mainly the terminal ileum. Paneth cells, which are most numerous in the terminal ileum, are specialised intestinal epithelial cells that secrete antimicrobial peptides in response to bacterial products and are critically important in enteric antibacterial defence. I hypothesised that Paneth cells express the NOD2 gene: this thesis describes the expression and quantification of the NOD2 gene in Paneth cells in inflammatory diseases such as Crohn's disease and necrotizing enterocolitis. I also identified factors that regulate NOD2 gene expression in intestinal epithelial cells and peripheral blood mononuclear cells (PBMC).;Methods: In situ hybridisation and immunohistochemistry were used to localize NOD2 mRNA and protein expression in intestinal tissue. Laser capture microdissection (LCM), and calcium chelation followed by mechanical disruption were used to isolate intestinal crypt and villus epithelial cells. Real-time RT-PCR was used to determine NOD2 gene expression in intestinal epithelial cells and PBMC.;Results: NOD2 mRNA and protein expression was readily detected in Paneth cells in normal and Crohn's disease affected terminal ileum NOD2 was also expressed by monocytes, but not by mature macrophages in the lamina propria or within granulomas. NOD2 mRNA levels were enriched in isolated crypt compared to villous epithelial cells, and NOD2 expression was mainly detected in LCM-acquired Paneth cells but not villous epithelial cells. In vitro, tumour necrosis factor alpha (TNFalpha) up-regulates NOD2 gene expression in intestinal epithelial cells. There is a possibility that Paneth cell antimicrobials are reduced in patients with NOD2-related Crohn's disease.;Conclusions: Paneth cells express NOD2 and could therefore play an important and hitherto unrecognized role in the development of NOD2-associated Crohn's disease.

Este proyecto se realizó con el propósito de comercializar bebidas saludables con la marca Lemon Panelé, la cual es elaborada a base de Panela, esta es extraída del jugo de la caña de azúcar que es sometida a fuego en el trapiche. En este proceso se obtiene el jugo clarificado y mediante la vaporización el resultado será la formación de la panela, este procedimiento permite asegurar un producto natural, además, de ser muy apreciado, debido a que no contiene ningún proceso de purificación químico por ello, tiene un alto valor nutricional en el mercado nacional e internacional. En el caso de las bebidas naturales, existe una tendencia mundial que también afecta al Perú en el crecimiento del consumo de productos saludables, nutritivos y del cuidado personal, lo que lleva incluso a empresas ya establecidas a voltear su mira hacia los productos naturales e ingresar a este mercado. Siendo este un factor importante que determina el consumo de la bebida, es que ingresamos con nuestra propuesta de bebidas a base de Panela que aporta beneficios como minerales y vitaminas tales como hierro, magnesio, manganeso, potasio, calcio, entre otros. Lemon Panelé es un producto que calza en la necesidad y preferencia del consumidor porque se puede beber a cualquier hora del día. Lemon Panelé está segmentado para los consumidores de nivel socioeconómico A, B y C, enfocado acorde a nuestra investigación en los afortunados, los progresistas y las modernas. This project had made with the purpose of marketing healthy drinks with Lemon Panelé brand, which elaborated with panela. This had extracted from the juice of the sugar cane that subjected to fire in the sugar mill. In this process, the panela juice is clarified and obtained by through vaporization the final result of sugar cane. This procedure allows to obtain a natural product, also, is appreciated because it does not contain any chemical product in the purification process therefore it has a high nutritional value in the national and international market. In the case of natural beverages, there is a global trend that also affects Peru in the growth of healthy consumption, nutritious and personal care products, which leads even established companies to turn their sights towards natural products and enter to this market. (Consumer Products Industry Outlook 2018). The most important factor that determines the consumption of the drink is that we enter with our proposal of drinks based on panela that provides benefits such as minerals and vitamins as iron, magnesium, manganese, potassium, calcium, among others. Lemon Panelé is a product that fits the needs and preferences of the consumers and it can consumed at any time of day. Lemon Panelé segmented for consumers of socioeconomic level A, B and C, focused according to our research on the Fortunate, Progressive and Modern. Embora não seja o provérbio original, adequa-se à avaliação de uma sociedade sujeita a profundos distúrbios alimentares, mas de tal forma enraizados que já são aceites como “normais”. A presente dissertação de base projetual, propõem-se contribuir para a modificação cultural deste paradigma, identificando a cozinha como o meio modelador do estilo de vida, assim procurando atuar na gênese da pessoa, ou seja, na sua educação infantil. O presente projeto prende-se com a realização de utensílios de cozinha adequados às crianças, que funcionem como instrumento de ligação entre pais e filhos, promovendo a comunicação e transmissão de saberes entre gerações e, consequentemente, a coesão familiar. Embora a abrangência do projeto se pudesse alargar a uma infinidade de artefatos, a panela desempenha um papel chave no imaginário da culinária, sendo nela que se desenrola a “magia” da cozinha. A abordagem metodológica passou por compreender não só o panorama e estado de arte deste domínio (recolhendo e sistematizando a pré-existência), como também pelo estudo do comportamento humano na reação aos estímulos despertados durante a interação com os alimentos e artefatos de cozinha. Complementarmente, foram efetuados estudos sobre a morfologia e adequação de materiais, tendo em conta a redução de custos e adequação ao público alvo, dando particular ênfase à segurança de operação. Com a dupla orientação científica da engenharia e do design, julga-se ter contribuído para assegurar um resultado adequado quer ao uso, quer à exequibilidade industrial, garantia de viabilidade comercial.
“Tell me what you eat, tell thee who you are” although not be the original saying, fits in to evaluation of one society who are subject of deep eating disorders, but such a way that ingrained are already accepted as “normal”. This dissertation project basis, intends to contribute to the change this cultural paradigm by identifying the kitchen as the modeler through the lifestyle, thus seeking to act in the genesis of the person, that is, in their children’s education. This project aims to the holding of cookware suitable for children, which act as bridging instrument between parents and children, promoting communication and transmission of knowledge between generations and therefore family cohesion. Although the comprehensiveness of the project could extend to a multitude of artifacts, the pot plays a key role in the culinary imagination, and it unfolds the “magic” of the kitchen. The methodological approach underwent includes not only the landscape of this area and state of the art (collecting and systematizing the pre existence), as well as the study of human behavior in response to stimuli awakened during interaction with food and cooking artifacts. In addition, studies have been conducted on the morphology and suitability of materials, given the cost savings and fitness for elected public. With the dual scientific guidance of engineering and design, we think we can ensure a better result either to use or to their industrial feasibility, ensuring their commercial viability.

Nous considérons le cas générique où les variétés CR considérées sont de contact. La géométrie CR présente de nombreuses similarités avec la géométrie conforme ; les invariants mis au jour et les techniques éprouvées en géométrie conforme peuvent donc être adaptées dans ce contexte. Nous nous intéressons dans cette thèse à deux invariants de ce type. Dans une première partie, en utilisant la géométrie asymptotiquement hyperbolique complexe, nous introduisons un opérateur différentiel CR covariant agissant sur les applications allant d'une variété CR vers une variété riemannienne, égal pour les fonctions à l'opérateur de Paneitz CR. Cauchy-Riemann geometry, CR for short, is the natural geometry of real pseudoconvex hypersurfaces of $C^{n+1}$ for $ngeq 1$. We consider the generic case when CR manifolds are contact manifolds. CR geometry presents strong analogies with conformal geometry; hence, known invariants and techniques of conformal geometry can be transported to that context. We focus in this thesis on two such invariants. In a first part, using asymptotically complex hyperbolic geometry, we introduce a CR covariant differential operator on maps from a CR manifold to a Riemannian manifold, which coincides on functions with the CR Paneitz operator. In a second part, we propose a Yamabe invariant for contact manifolds which admit a CR structure, and we study its behaviour under connected sum

La investigación es clave para Nestlé, ya que se trata de la elaboración de una campaña para desestacionalizar uno de sus productos más conocidos, panetón D´onofrio, que se caracteriza por ser consumido principalmente en la etapa previa y durante Navidad, pieza clave de las cenas navideñas. Así, el objetivo de este trabajo es elaborar una campaña publicitaria a nivel nacional, que logre cambiar el mindset de los consumidores, para que estén dispuestos a consumir panetón D’onofrio fuera de las fiestas navideñas, específicamente entre los meses de enero y abril. El trabajo de campo se elaboró en base a una muestra homogénea no probabilística de 65 personas entre hombres y mujeres de 25 a 55 años. La investigación tiene un enfoque mixto y los instrumentos aplicados son social listening, entrevistas y encuestas. A partir de ello, se encontró lo siguiente: en primer lugar, los consumidores de panetón D’onofrio aman el producto, pero hay algo más allá que los impulsa a comprarlo, lo que los hace sentir: la unión familiar; en segundo lugar, se corroboró que el domingo es un día especial para las familias peruanas, las cuales lo usan para reunirse y pasar buenos momentos juntos. Es así que se propone que panetón D’onofrio hará los domingos en familia incluso más especiales, potenciando la unión familiar del target en este día, el cual es uno de los pilares de la marca.
The research is key for Nestlé, since it involves the development of a campaign to seasonally adjust one of its best-known products, panetón D´onofrio, which is characterized by being consumed mainly in the previous stage and during Christmas, a key part of the Christmas dinners. Thus, the objective of this work is to develop an advertising campaign at the national level, which manages to change the mindset of consumers, so that they are willing to consume panetón D’onofrio outside of the Christmas holidays, specifically between the months of January and April. The field work was developed based on a non-probabilistic homogeneous sample of 65 people between men and women between 25 and 55 years old. The research has a mixed approach and the instruments applied are social listening, interviews and surveys. Based on this, the following was found: firstly, the consumers of panetón D’onofrio love the product, but there is something beyond that drives them to buy it, which makes them feel: family unity; secondly, it was confirmed that Sunday is a special day for Peruvian families, who use it to get together and have good times together. Thus, it is proposed that panetón D’onofrio will make Sundays with the family even more special, enhancing the family union of the target on this day, which is one of the pillars of the brand.

O epitÃlio intestinal à formado e mantido por uma populaÃÃo de cÃlulas-tronco capaz de gerar diferentes linhagens celulares, mantendo a pluripotÃncia e a capacidade de auto-renovaÃÃo. A glutamina à um aminoÃcido essencial condicional importante para a manutenÃÃo do epitÃlio do intestino. No entanto, poucos estudos tÃm explorado o papel da glutamina na regulaÃÃo fina do turnover celular da cripta intestinal. Com o propÃsito de avaliar o papel da glutamina n o turnover de cÃlulas da cripta, foi utilizado um modelo in vitro de enteroide, onde as cÃlulas-tronco sÃo capazes de gerar um epitÃlio contendo as principais linhagens de cÃlulas secretoras intestinais (cÃlula de Paneth, cÃlula caliciforme e cÃlula enteroendÃcrina), alÃm dos enterÃcitos absortivos, com a formaÃÃo de uma estrutura tipo vilosidade-cripta. Este modelo foi usado para testar o efeito de 24 horas de privaÃÃo de glutamina (meio padrÃo com glutamina a 2 mM vs meio livre de glutamina) no turnover epitelial atravÃs da contagem de cÃlulas marcadas por Edu/nÃmero total por secÃÃo e ainda na apoptose celular, atravÃs da contagem de cÃlulas marcadas para caspase 3-clivada/nÃmero total por secÃÃo. A fim de avaliar a funÃÃo secretora das criptas, as razÃes das cÃlulas de Paneth e caliciformes por cripta (cÃlula alvo/nÃmero total de cÃlulas secretoras por cripta) foram medidas. O nÃmero de cÃlulas de Paneth e caliciformes foi obtido com o auxÃlio da microscopia confocal e imunomarcaÃÃo para lisozima e mucina-2. AlÃm disso, os transcritos dos produtos das cÃlulas de Paneth e caliciformes (lisozima e mucina, respectivamente) foram analisados utilizando o mÃtodo de PCR quantitativo em tempo real. Com intuito de avaliar o potencial papel imunomodulador da glutamina, transcritos de elementos da imunidade inata, receptor de tipo Toll e sua proteÃna acessÃria MD-2, e citocinas, a saber: TNF-α, IL-1β e CXCL-1, foram medidos. A privaÃÃo de glutamina reduziu o nÃmero de cÃlulas Edu positivas, em comparaÃÃo com o enteroide sob meio padrÃo (p=0,006). NÃo houve diferenÃa significativa em relaÃÃo Ãs razÃes das cÃlulas de Paneth e cÃlulas caliciformes entre os grupos apÃs privaÃÃo de glutamina. A privaÃÃo da glutamina diminuiu significativamente os transcritos de lisozima, em comparaÃÃo com o enteroide sob meio padrÃo (p = 0,007), mas nÃo para mucina-2, transcriÃÃo relacionada com a funÃÃo das cÃlulas caliciformes. Uma transcriÃÃo reduzida para TNF- α e MD-2 (p=0,005 e p=0,016, respectivamente) foi observada apÃs a privaÃÃo de glutamina. Ao todo, nossos achados reforÃam o papel positivo da glutamina sobre o turnover do epitÃlio intestinal e, alÃm disso, sugerem um importante efeito regulador da glutamina sobre as cÃlulas de Paneth e resposta imune inata. O modelo enteroide fornece uma ferramenta importante para dissecar os mecanismos de proteÃÃo pela glutamina e guiar estudos futuros.
The intestinal epithelium is formed and sustained by a population of stem cells capable of generating different cell lines while maintaining pluripotent and self-renewal capacity. Glutamine is a conditional essential amino acid important for the maintenance of the intestinal epithelium. However, few studies to date have explored the role of glutamine in the fine regulation of the intestinal crypt cell turnover. In order to evaluate the role of glutamine in the crypt cell turnover, an in vitro enteroid model was used, where stem cells are capable of generating an epithelium containing the main intestinal secretory cell lines (Paneth, goblet, and enteroendocrine cells) and absorptive enterocytes as well, while forming a villus-crypt like structure. This model was used to test the effect of 24h of glutamine deprivation (standard media with 2mM glutamine vs glutamine-free media) on epithelial turnover by counting EdU- labeled cells/total number per section and cell apoptosis by counting cleavage-caspase 3- labeled cells/total number per section. In order to assess crypt secretory function, Paneth and goblet crypt cell ratios (target secretory cell/total cell number per crypt) were measured. The number of Paneth and goblet cells was measured with the aid of confocal microscopy and lysozyme and mucin-2 immunostainning. In addition, Paneth and globet cell product transcripts (lysozyme and mucin, respectively) were measured using quantitative Real-time PCR. In order to assess the potential immunomodulatory role of glutamine, innate immune element transcripts, Toll like receptor and their accessory protein MD-2, and cytokines, as follows: TNF-α, IL-1β and CXCL-1, were measured. Glutamine deprivation reduced the number of EdU positive cell ratio as compared with the enteroid under the standard media (p=0.006). No significant differences regarding Paneth and goblet cell ratios were seen between groups following glutamine deprivation. Glutamine deprivation significantly decreased lysozyme transcripts as compared with the enteroid under the standard media (p=0.007), but not for mucin-2 transcripts, related to goblet cell function. Decreased TNF-α and MD-2 transcription (p=0.005 and p=0.016, respectively) were found following glutamine deprivation. Altogether, our findings reinforce the glutamine positive role on the intestinal epithelial turnover and furthermore suggest an important glutamine regulatory effect over Paneth cells and the innate immune system. The enteroid model provides an important tool the dissect the mechanisms of glutamine protection and shed light for future studies.

In einer Verschränkung von visueller Geschichte und Mediengeschichte wird gezeigt, wie das Bild der Erde in immer neue Sinn- und Handlungshorizonte gesetzt wurde. Während das berühmte Foto der Blue Marble (Apollo 17) etwa zur Ikone einer systemischen Globalität in der Ökologiebewegung avancierte, steht Buckminster Fullers Geoscope von 1963 paradigmatisch für den Übergang zu einem »globalen Interface«. Die sphärische Visualisierung globaler Ströme präfiguriert, was ein halbes Jahrhundert später mit Softwareanwendungen wie Google Earth zum neuen medialen Habitat der Blue Marble wird. Deutlich tritt hier die visuelle Repräsentation der Erde hinter einer »Oligoptik« zurück, in der sich Bedeutungen, Handlungspotenziale und Machthierarchien situiert herausbilden.

El diseño industrial se ha convertido en una forma de generar valor a los productos a partir de la mejora no solo en la presentación del mismo sino además en la utilidad; en el presente proyecto se plantea el desarrollo de una propuesta de diseño de un puesto de venta de panela para los productores del municipio de Urrao, Antioquia, con base en los requerimientos de estos últimos y del producto en sí. Para tal efecto se plantea una metodología de tipo aplicada y tecnológica con enfoque cuantitativo. De igual manera se tomó como referente el diagnóstico elaborado en un proyecto previo del cual surge la presente propuesta, y se procederá a realizar entrevistas a productores y mediciones en la plaza de mercado del municipio, con el fin de elaborar un diseño lo más pertinente posible. Para la construcción del marco teórico se realizó una búsqueda en bases de datos tomando como categorías de análisis las siguientes: definición de diseño industrial, el diseño industrial como ventaja competitiva, aplicaciones, proceso de elaboración de un diseño industrial y algunas generalidades sobre la producción de panela. Como resultado del proyecto se espera entregar un diseño del puesto de venta de panela ajustado a los necesidades y expectativas de los productores del municipio de Urrao, con su respectivo registro, así como la participación en un evento académico enfocado en investigación formativa, de forma que se fortalezca este proceso en la Institución Universitaria Pascual Bravo.
