Abstract:
ABSTRACT The Aspergillus nidulans ambient pH signaling pathway involves two transmembrane domain (TMD)-containing proteins, PalH and PalI. We provide in silico and mutational evidence suggesting that PalI is a three TMD (3-TMD) protein with an N-terminal signal peptide, and we show that PalI localizes to the plasma membrane. PalI is not essential for the proteolytic conversion of the PacC translation product into the processed 27-kDa form, but its absence markedly reduces the accumulation of the 53-kDa intermediate after cells are shifted to an alkaline pH. PalI and its homologues contain a predicted luminal, conserved Gly-Cys-containing motif that distantly resembles a Gly-rich dimerization domain. The Gly44Arg and Gly47Asp substitutions within this motif lead to loss of function. The Gly47Asp substitution prevents plasma membrane localization of PalI-green fluorescent protein (GFP) and leads to its missorting into the multivesicular body pathway. Overexpression of the likely ambient alkaline pH receptor, the 7-TMD protein PalH, partially suppresses the null palI32 mutation. Although some PalH-GFP localizes to the plasma membrane, it predominates in internal membranes. However, the coexpression of PalI to stoichiometrically similar levels results in the strong predominance of PalH-GFP in the plasma membrane. Thus, one role for PalI, but possibly not the only role, is to assist with plasma membrane localization of PalH. These data, considered along with previous reports for both Saccharomyces cerevisiae and A. nidulans, strongly support the prevailing model of pH signaling involving two spatially segregated complexes: a plasma membrane complex containing PalH, PalI, and the arrestin-like protein PalF and an endosomal membrane complex containing PalA and PalB, to which PacC is recruited for its proteolytic activation.

Abstract:
W artykule przedstawiono wyniki badań pali wierconych z podstawą poszerzoną metodą iniekcji strumieniowej. Na budowie obiektów mostowych drogi S8 na węźle Salomea w Warszawie wykonano doświadczalny pal i ustalono, że jego nośność była 1,5 razy większa od nośności pala bez poszerzonej podstawy, a obciążenie dopuszczalne tego pala było 1,65 razy większe od obciążenia dopuszczalnego pala bez poszerzonej podstawy. Stwierdzono, że całkowite opory podstawy tego pala zostają zmobilizowane przy przemieszczeniach odpowiadających w przybliżeniu 0,1 średnicy pala. Podczas budowy obiektów mostowych drogi S8 na odcinku obwodnicy Marek przeprowadzono próbne obciążenia 17 pali CFA z podstawą poszerzoną metodą iniekcji strumieniowej. Stwierdzono, że średnia wartość nośności obliczeniowej pali była 1,23 razy większa od projektowanego średniego obciążenia obliczeniowego, natomiast średnia nośność graniczna była 1,78 razy większa od projektowanego średniego obciążenia obliczeniowego. Średni wskaźnik nośności obliczeniowej wyznaczony na podstawie badania wynosił 1,23, a średni wskaźnik nośności granicznej wyniósł 1,89. Na podstawie badań doświadczalnych zdefiniowano zasady obliczania nośności pali CFA z podstawą poszerzoną metodą iniekcji strumieniowej.

ABSTRACT Erwinia rhapontici is able to convert sucrose into isomaltulose (palatinose, 6-O-α-d-glucopyranosyl-d-fructose) and trehalulose (1-O-α-d-glucopyranosyl-d-fructose) by the activity of a sucrose isomerase. These sucrose isomers cannot be metabolized by plant cells and most other organisms and therefore are possibly advantageous for the pathogen. This view is supported by the observation that in vitro yeast invertase activity can be inhibited by palatinose, thus preventing sucrose consumption. Due to the lack of genetic information, the role of sucrose isomers in pathogenicity has not been evaluated. Here we describe for the first time the cloning and characterization of the palatinose (pal) genes fromErwinia rhapontici. To this end, a 15-kb chromosomal DNA fragment containing nine complete open reading frames (ORFs) was cloned. The pal gene products of Erwinia rhapontici were shown to be homologous to proteins involved in uptake and metabolism of various sugars from other microorganisms. ThepalE, palF, palG, palH, palK, palQ, and palZgenes were oriented divergently with respect to the palRand palI genes, and sequence analysis suggested that the first set of genes constitutes an operon. Northern blot analysis of RNA extracted from bacteria grown under various conditions implies that the expression of the palI gene and the palEFGHKQZgenes is oppositely regulated at the transcriptional level. Genes involved in palatinose uptake and metabolism are down regulated by sucrose and activated by palatinose. Palatinose activation is inhibited by sucrose. Functional expression of palI andpalQ in Escherichia coli revealed sucrose isomerase and palatinase activity, respectively.

W artykule przedstawiono wyniki badań pali CFA z podstawą poszerzoną metodą iniekcji strumieniowej, wykonanych podczas budowy obiektów mostowych drogi ekspresowej S6 na odcinku Kiełpino – Kołobrzeg. Wykonano badania 15 pali i na podstawie tych badań oceniono nośność obliczeniową oraz graniczną pali, które porównano z obciążeniem obliczeniowym według projektu. Z przeprowadzonych badań wynikało, że średnia wartość wskaźnika nośności obliczeniowej pali na podstawie przeprowadzonych badań wyniosła 1,08. Trzy pale usytuowane w lewej części podpory A w obiekcie MS-74 charakteryzowały się mniejszą nośnością obliczeniową na podstawie badania w porównaniu do obciążenia obliczeniowego według projektu. Na podstawie badań sondą CPT stwierdzono, że grunty w tym miejscu charakteryzowały się słabszymi parametrami geotechnicznymi niż wynikało z wcześniejszych badań. Z tego powodu we wszystkich palach lewej części tej podpory wykonano pod podstawami pali metodą iniekcji strumieniowej dodatkowe kolumny z cementogruntu, co spowodowało istotne zwiększenie nośności tych pali.

ABSTRACT We identified a putative pal gene cluster (palR, palE, palF, palG, palK, palA, and palB) in the plant-tumorigenic bacterium Agrobacterium tumefaciens MAFF301001; by sequencing analyses, this cluster was found to be involved in palatinose transport, and its functional importance was revealed by mutational analyses. The pal gene products were highly homologous to those of putative trehalose/maltose ABC-type transport systems but were not essential to bacterial growth on trehalose. Insertion mutations in the palK and palE genes showed the necessity of these genes for bacterial growth and chemotaxis with palatinose as the carbon source, but no inhibition of tumorigenesis was observed. Growth on trehalose and maltose was not influenced by the mutations.

Hasil yang diperoleh adalah bahwa dalam ritual tiwah terdapat pembatas adat yang disebut Hinting Pali, yang tidak dapat dilanggar oleh beberapa jenis pali atau pantangan seperti pali sayur, pali hewan, pali ikan, dan sikap / perilaku pali. Jika ada pelanggaran pali dalam ritual tiwah, denda yang diberikan adalah mengganti dua kali lipat biaya tiwah yang telah dikeluarkan oleh anggota ritual tiwah. Penelitian ini hanya membahas pali dalam ritual tiwah masyarakat adat Dayak Ngaju. Kedepannya, penelitian ini diharapkan dapat menjadi dasar penelitian tentang pali dalam ritual dan upacara lainnya dalam budaya masyarakat adat Dayak Ngaju Kalimantan Tengah.

Masyarakat Dayak Ngaju merupakan sekelompok masyarakat yang masih menjunjung tinggi adat istiadat yang berkaitan dengan Pali. Pali ini dikenal juga dengan pamali dalam istilah Jawa. Tujuan penulisan ini sebagai bentuk pengenalan wujud kebudayaan masyarakat Dayak Ngaju yang memiliki nilai kearifan lokal yang berkaitan dengan sikap dan perilaku manusia itu sendiri sehingga sedikit dikenal oleh masyarakat luas. Melalui tulisan ini tentunya pembaca akan dapat membedakan yang mana Pali/larangan yang bersifat mendesak untuk dipatuhi dan yang mana yang bersifat mitos belaka. Metode yang digunakan adalah metode deskriptif untuk mendeskripsikan bentuk pelaksanaan Pali yang diyakini masyarakat sekitarnya. Hasil penelitian ada berbagai macam Pali (larangan) yang harus dihindari oleh masyarakat Dayak Ngaju, pada saat ada keluarga yang meninggal dunia, salah satunya larangan-larangan dalam beraktivitas. Pali ini juga kuat kaitannya terhadap mitos budaya sekitarnya, karena hal ini dianggap benar-benar terjadi. Orang-orang yang melanggar pali tersebut dipercaya akan memperoleh kesialan dan yang paling penting dapat menimbulkan mala petaka bagi yang melanggarnya. Pali ini tentunya sangat berpengaruh bagi perilaku dan aktivitas keluarga si mati, seperti pali menaruh perkakas yang digunakan untuk membuat peti si mati di dalam rumah. Selain itu, ada juga pali mengkonsumsi sayuran, Pali yang berhubungan dengan perjalanan, dan Pali yang berhubungan dengan kegiatan takziah.

ABSTRACT The gene (palI) encoding isomaltulose synthase (PalI) from a soil bacterial isolate, Klebsiella sp. strain LX3, was cloned and characterized. PalI converts sucrose into isomaltulose, trehalulose, and trace amounts of glucose and fructose. Sequence domain analysis showed that PalI contains an α-amylase domain and (β/α)8-barrel structures, suggesting that it belongs to the α-amylase family. Sequence alignment indicated that the five amino acid residues of catalytic importance in α-amylases and glucosyltransferases (Asp241, Glu295, Asp369, His145, and His368) are conserved in PalI. Purified recombinant PalI displayed high catalytic efficiency, with a Km of 54.6 ± 1.7 mM for sucrose, and maximum activity (approximately 328.0 ± 2.5 U/mg) at pH 6.0 and 35°C. PalI activity was strongly inhibited by Fe3+ and Hg2+ and was enhanced by Mn2+ and Mg2+. The half-life of PalI was 1.8 min at 50°C. Replacement of selected amino acid residues by proline significantly increased the thermostability of PalI. Simultaneous replacement of Glu498 and Arg310 with proline resulted in an 11-fold increase in the half-life of PalI at 50°C.

Abstract:
Paeonia suffruticosa has been extensively used as a traditional medicine with various beneficial effects; paeonolide (PALI) was isolated from its dried roots. This study aimed to investigate the novel effects and mechanisms of PALI in pre-osteoblasts. Here, cell viability was evaluated using an MTT assay. Early and late osteoblast differentiation was examined by analyzing the activity of alkaline phosphatase (ALP) and by staining it with Alizarin red S (ARS). Cell migration was assessed using wound healing and Boyden chamber assays. Western blot and immunofluorescence analyses were used to examine the intracellular signaling pathways and differentiation proteins. PALI (0.1, 1, 10, 30, and 100 μM) showed no cytotoxic or proliferative effects in pre-osteoblasts. In the absence of cytotoxicity, PALI (1, 10, and 30 μM) promoted wound healing and transmigration during osteoblast differentiation. ALP staining demonstrated that PALI (1, 10, and 30 μM) promoted early osteoblast differentiation in a dose-dependent manner, and ARS staining showed an enhanced mineralized nodule formation, a key indicator of late osteoblast differentiation. Additionally, low concentrations of PALI (1 and 10 μM) increased the bone morphogenetic protein (BMP)–Smad1/5/8 and Wnt–β-catenin pathways in osteoblast differentiation. Particularly, PALI (1 and 10 μM) increased the phosphorylation of ERK1/2 compared with BMP2 treatment, an FDA-approved drug for bone diseases. Furthermore, PALI-mediated early and late osteoblast differentiation was abolished in the presence of the ERK1/2 inhibitor U0126. PALI-induced RUNX2 (Cbfa1) expression and nuclear localization were also attenuated by blocking the ERK1/2 pathway during osteoblast differentiation. We suggest that PALI has biologically novel activities, such as enhanced osteoblast differentiation and bone mineralization mainly through the intracellular ERK1/2-RUNX2 signaling pathway, suggesting that PALI might have therapeutic action and aid the treatment and prevention of bone diseases, such as osteoporosis and periodontitis.

Abstract:
Around the turn of the 20th century (Childers, 1879; Müller, 1884; Whitney, 1889; Gray, 1899; Duroiselle, 1906), neogrammarians began studying the Pali language and its grammar. One area of particular interest was the phonological process of assimilation. Pali evidences both progressive and regressive assimilation and it is generally the case that a consonant geminate is formed (Krishnaswamy, et al., 2019). Recent studies (Junghare, 1979; Suzuki, 2002a,b; Gupta, 2003; Schmeiser, 2008; Dutta, 2017) have attempted to explain base-medial consonant gemination in phonological terms. Though these accounts have furthered our understanding of Pali base-medial consonant gemination, very few studies have considered morphological effects on Pali gemination. The current study, though exploratory in nature, suggests that a problem arises in phonological analyses of Pali geminates in that Pali words evidence different cluster realizations based on morphological factors. In short, the study analyzes the effects of affixes on Pali gemination and suggests that an explanation based solely on syllable structure or sonority is insufficient and calls for further research.

Abstract By offering the first detailed portrait of how a particular local system for Pali pronunciation functions, this article aims to renew our appreciation of the complex interaction between chanted sounds and written words in Buddhist cultures. Seated at the fulcrum between orality and writing, the diverse phonologies of Pali are constantly evolving sites of debate over the nature of the voice of the Buddha and his teaching in Theravāda contexts. To give a precise account of how the living complexity of Pali unfolds, the findings in this article are based on the phonetic transcription and analysis of fifteen multimedia recordings of Pali liturgical chants in twentieth- and twenty-first-century Cambodia. The range of major and minor variations in Pali pronunciation witnessed during this period, and the contentious debates behind these divergencies, open new paths for understanding the past and present of Pali as a Buddhist language.

Abstract Phosphodiesterase-4 (PDE4) is a key enzyme in cAMP hydrolysis and is involved in various inflammatory and fibrotic diseases, such as ulcerative colitis (UC). PDE4 inhibition elevates intracellular cAMP, downregulates inflammatory cytokines (e.g., TNFα), and reduces cell adhesion molecules, thereby preventing local infiltration of inflammatory cells. Several PDE4 inhibitors (e.g., roflumilast and apremilast) are approved for COPD and psoriasis, with others under development for diseases such as idiopathic pulmonary fibrosis (IPF). Phase 2 clinical trials have shown that oral PDE4 inhibitors enhance clinical remission in UC patients, with a 20% placebo-adjusted improvement with low-dose apremilast. However, oral administration and systemic distribution cause central nervous system (CNS) toxicity (e.g., headaches, nausea, vomiting), leading to therapy discontinuation and limiting efficacy. Targeted and better-tolerated oral treatments are needed to fully exploit PDE4 inhibitors in UC. We assessed the efficacy of PALI-2108, an oral and locally bioactivated PDE4 inhibitor prodrug for UC treatment. PALI-2108 incorporates a galactose-derived sugar moiety, making it minimally absorbed until cleaved by the colonic bacterium enzyme β-glucuronidase. In a DSS-induced UC mouse model, PALI-2108 demonstrated that at doses of 21, 4, 0.75 mg/kg BID, and 1.5 mg/kg daily, it reduced the mean disease activity index (DAI) score over time compared to the DSS control group (Fig 1). Additionally, body weight loss over time was attenuated in the 21 and 4 mg/kg BID groups (Fig 2). A tissue distribution study showed colon preference of PALI-2108 with limited plasma levels and a colon/plasma AUC ratio exceeding 200. Additionally, oxazolone colitis induced mice treated with PALI-2108 showed colon-selective bioactivation, with negligible plasma levels compared to duodenal and ileal tissue. In a target engagement study, we used a CETSA Navigate® assay to measure PALI-2108 binding to cAMP-specific 3’, 5’-cyclic PDE4D in OXZ-treated mice. Melt and shift curves showed similar changes, indicating comparable target engagement in colon tissues between PALI-2108 and apremilast. Finally, in a tolerated dose study, we showed no CNS toxicity and emesis over effective doses in dogs. In conclusion, our results demonstrate local bioactivation of a PDE4 inhibitor pro-drug, PALI-2108, and its colon-specific distribution with limited systemic exposure. PALI-2108 has similar target engagement to other PDE4 inhibitors, dose-dependent efficacy in a UC model, and no systemic toxicity. We believe PALI-2108 is a promising novel therapy for UC, with localized bioactivation, expanded therapeutic window, and potent PDE4 inhibitory activity. PALI-2108 is in development for moderate-to-severe UC and is advancing toward regulatory filing for first-in-human studies. PALI-2108 reduced DSS induced mean disease activity index score (DAI) over time compared to control. PALI-2108 attenuated DSS induced body weight loss over time compared to control.

The study investigated the integration between cumin (Cuminum cyminum) markets pairs in Rajasthan State using monthlycumin price series of five (5) markets from 2008-2015. The co-integration tests results indicate Nagaur and Pali; Nagaur andJodhpur; Nagaur and Jalore markets are integrated in the long run at lag five (5). However, the rest of other market pairs arenot integrated in the long run. The results from the error correction model showed that, the lowest speed of adjustment towardslong run equilibrium was from Nagaur to Jalore market at rate of 37.4%. The highest speed of adjustment was 56.6%, runningNagaur to Pali market towards long run equilibrium. This is followed by a speed of adjustment of 45.1% running from Nagaur toJodhpur market towards along run equilibrium in a period of at most one (1) month. The results further indicate bidirectionalcausality between Nagaur and Pali markets and also Naguar and Jalore markets. There is also a unidirectional causality of pricechange from Merta City market of Nagaur to Bhagat Ki Kothi market of Jodhpur but not vise versa. The results from theunrestricted vector autoregressive (VAR) model was run for market pairs that were not cointegrated that cumin price seriesbetween Ajmer and Jodhpur; Jodhpur and Pali and Jodhpur and Jalore Granger causes each other in the short run whereasAjmer Granger causes Nagaur, Pali and Jalore in less than one month though in the long run they drift apart. The study furtherreveal that orthogonalized shock or unexpected shocks to Nagaur market will result in permanent effect on the prices of otherselected markets within one year. In the same way, unexpected shock to market prices of Pali will result in permanent priceeffect on Nagaur cumin prices but temporary on prices in Jodhpur. On the other hand unexpected shock to prices of Ajmer willresults in transitory or temporary price effect on prices in Pali, Nagaur, Jodhpur and Jalore markets.

The purpose of this study was to determine the Effect of Compensation and Career Development on Employee Performance at PT Aburahmi in PALI Regency. This research was conducted on PT. Aburahmi employees at Penukal Abab Lematang Ilir (PALI) jerus. 55 respondents as a sample. The data source in this study were secondary data and primary data using saturated sampling technique. Based on the results of the analysis, it shows that compensation and career development have a positive and significant effect on employee performance at Penukal Abab Lematang Ilir (PALI). F test (simultaneous) shows that Compensation and Csreer Development both have an effect on Employee Performance at Penukal Abab Lematang Ilir (PALI). Viewed and the coefficient of determination R Square is 0.274 which means 27,4% of employee performance variables can be explained by compensation and career development as much 72,6% can be explained by other variables not included in this study.
