Abstract:

Cytochrome P450 are heme-containing enzymes that have been deemed to be the most active group of monooxygenases reaction. It is a self-sufficient protein whose activities rely on the various associated coenzymes. Expression of the catalytic module of cytochrome P450 gene (CYP102A2) containing pET-28a in Escherichia coli BL21 (DE3) across temperatures ranging from 19 ºC to 37 ºC. The solubility of the expressed recombinant proteins on SDS-PAGE showed that the recombinant protein is both secreted and membrane-bound protein across all the temperature treatments. The highest expressed recombinant protein of interest (P.O.I.) band observed in lane 15 was membrane-bound and was optimum at 28 ºC, while the most prominent expressed P.O.I. bands in lane 16 from the secreted proteins was optimum at 30 ºC. The estimated flavin adenine dinucleotide (F.A.D.) and flavin mononucleotide (FMN) showed optimum concentrations of 15 mM and 14 mM respectively at the optimum temperature of 25 ºC, while the estimated heme optimum concentration (2.4 mM) was recorded at the optimum temperature of 35 ºC. These observations showed that it is little or no connection between the quantity of the expressed recombinant cytochrome P450 and the estimated coenzymes and cofactors concentrations in response to the temperature parameter. The enzymatic activity of the recombinant P.O.I. at a selected temperature of 25 ºC and 35 ºC showed exponential slope within the period of 60 seconds at 550 nm wavelength of reaction; however, a wide range of absorbance ranging from 0.1 A to 0.7 A and 0. 689 A - 0.998 A was recorded, respectively. The increase in activity may also be attributed to the fact that higher temperature tends to support an increase in the rate of reactions, hence the rate of monoxygenation.

AbstractFourteen species of Carabidae are added to Prince Edward Island’s (P.E.I.) faunal list, bringing the known fauna to 167 species. Bembidion nitidum (Kirby) and Bembidion obtusum Audinet-Serville are newly recorded for the Maritime Provinces. Six species are removed from P.E.I.’s faunal list. The history of collecting of Carabidae on P.E.I. is briefly recounted. Despite differences in land area and distance from the mainland between P.E.I., Cape Breton Island, and insular Newfoundland, their carabid faunas exhibit many similarities in size and composition. The native carabid fauna of P.E.I. comprises 49% of the species in the combined Maritime Provinces fauna, perhaps reflecting an island-related diminution of species diversity. The proportion of flightless species on P.E.I. (4.9%) is less than that in the Maritime Provinces as a whole (7.1%), an apparent indication that the Northumberland Strait has been a barrier to colonization. Twenty-seven introduced species are found on P.E.I., 26 of which can be classified as synanthropic and may have originated in dry-ballast quarries in southwestern England. Although the earliest dates of detection of many introduced species on P.E.I. are substantially later than elsewhere in the Maritimes, this reflects the paucity of early collecting. Land-management practices on P.E.I. (large-scale and early forest clearances, intensive agriculture, and the extensive use of biocides) may have had an impact on P.E.I.’s carabid fauna.

Abstract:

Managers formulate, implement and monitor strategies to pursue their goals. Implementation is important and involves, amongst others, Production and Operations Management (P.O.M.) techniques. The purpose of this article is to study, by means of a limited sample, perceptual associations between goals of manufacturing strategy and P.O.M. techniques. An attempt is made to answer two questions in this article. The first question is which specified goals are independent of each other? The second question is which P.O.M. techniques are recommended for these independent goals? Bias and the definition of P.O.M. techniques create difficulties but pilot research indicates that some specified goals are perceived as independent. More investigation is required to verify this. Some techniques are identified with specific goals and this work should be expanded.

Jedną z podstawowych zasad ochrony środowiska jest dyrektywa „zanieczyszczający płaci”, mająca umocowanie w postanowieniach art. 7 ustawy z dnia 27 kwietnia 2001 r. – Prawo ochrony środowiska. Środkiem prawnym służącym jej realizacji są kary pieniężne wymierzane na podstawie art. 298 p.o.ś. za naruszenie warunków korzystania ze środowiska. Sposób ustalania wysokości kar został określony w art. 305 i 305a p.o.ś. Zasady postępowania w przedmiocie ich wymierzania zawarto jednak także w Kodeksie postępowania administracyjnego, którego art. 189f precyzuje, kiedy należy odstąpić od wymierzenia kary. Rozwiązanie to odnosi się również do kar wymierzanych na podstawie p.o.ś., w praktyce więc zachodzi konieczność łącznego stosowania rozwiązań wynikających z k.p.a. i p.o.ś. W artykule omówiono zatem przesłanki, które uzasadniają odstąpienie od wymierzenia kary pieniężnej za naruszenie warunków korzystania ze środowiska, a w podsumowaniu wskazano, że taki dualizm nie jest dobry, dlatego przepisy prawa w tym zakresie powinny zostać uporządkowane.

AbstractThe population dynamics of the European corn borer, Ostrinia nubilalis (Hübner) (Lepidoptera: Pyralidae), were documented on potatoes grown in Prince Edward Island (P.E.I.) from 1990 to 1992. By comparing degree-day accumulations above 10°C, both first occurrence and peak flights of male adults occurred earlier than elsewhere in North America. Because potatoes in P.E.I. are attacked earlier and over a longer period of time than potatoes grown in other regions, the insect is an important pest of potatoes in P.E.I. The correlation between peak numbers of male adults caught in pheromone traps and egg masses on potato plants was not significant (p = 0.22). However, the correlation between egg masses per potato stalk and larvae per potato stalk was significant (p = 0.003). Trapping adults and monitoring potato plants for the presence of egg masses during the period from 250 to 500 degree-days appears to be a reliable way to determine the presence of adults and the potential impact of larvae on yield of potatoes.

plaster of Paris (P.O.P.) bandage has been the pre-eminent external splinting material for over 150 years and from time to time synthetic alternatives have been tried. So far none has seriously challenged the dominance of P.o.P. as a primary or secondary material in the management of fractures. The recent introduction of Polyurethane coated fibreglass bandage appears to offer a more serious challenge than previous contenders. This technical note reviews bandage type splinting materials and explains some of the properties of the PU materials.

Abstract:

The current study aimed at assessing the effects of exposure to Particulate Matter (PM) on the incidence of respiratory diseases in a sub-sample of participants in the longitudinal analytical epidemiological study in Pisa, Italy. Three hundred and five subjects living at the same address from 1991 to 2011 were included. Individual risk factors recorded during the 1991 survey were considered, and new cases of respiratory diseases were ascertained until 2011. Average PM10 and PM2.5 exposures (µg/m3, year 2011) were estimated at the residential address (1-km2 resolution) through a random forest machine learning approach, using a combination of satellite data and land use variables. Multivariable logistic regression with Firth’s correction was applied. The median (25th–75th percentile) exposure levels were 30.1 µg/m3 (29.9–30.7 µg/m3) for PM10 and 19.3 µg/m3 (18.9–19.4 µg/m3) for PM2.5. Incidences of rhinitis and chronic phlegm were associated with increasing PM2.5: OR = 2.25 (95% CI: 1.07, 4.98) per unit increase (p.u.i.) and OR = 4.17 (1.12, 18.71) p.u.i., respectively. Incidence of chronic obstructive pulmonary disease was associated with PM10: OR = 2.96 (1.50, 7.15) p.u.i. These results provide new insights into the long-term respiratory health effects of PM air pollution.

Abstract:

This paper examines the creation of anthropomorphism through a point of view focusing on visual structure in the "Hunters and Hunted"(2009) documentary. One of the criticisms of wildlife films is the anthropomorphism of animal behavior and relationships in ecosystems in these documentaries, as it can lead to distortions of animal behavior in ecosystems. One of the aims of this article is to indicate the part of anthropomorphism that is created through the point of view and through the visual form of the work, so in order to avoid anthropomorphism in films, more attention should be paid to the form of aesthetic. Point-of-view analysis shows that subjective point of view (P.O.V) shots leads to anthropomorphism from the perspective of non-human beings. With this in mind, the different perspectives were explored in "Hunters and Hunted" from the “life” (2009) series. Surveys show that from 475 shots of this documentary, 14 shots are from the first person’s point of view from an animal perspective to advance the narrative. The structure of the P.O.V created in these shots shows that the shot / reverse angle shot pattern was used to create the subjective point of view and the image composition and also editing played a key role in creating the P.O.V shots in this documentary.

Kecemasan adalah hal umum, tetapi reaksi yang cukup lama dan berlebihan dapat menyebabkan gangguan psikologi seperti gangguan cemas umum atau GAD. Penderita GAD dapat merasakan takut dan cemas terhadap hal yang tidak spesifik atau situasi tertentu. Menurut Departemen Kesehatan, gejala-gejala kecemasan berlebih dirasakan oleh usia 15 tahun ke atas, khususnya bagi mahasiswa. Untuk meningkatkan kesadaran masyarakat dibuatlah sebuah film pendek dengan judul “In My Mind: Generalized Anxiety Disorder”. Dalam proses produksi memiliki 4 tahap yaitu, analisis data, pra-produksi, produksi, pasca-produksi. Dalam beberapa adegan memakai teknik P.O.V dengan menggunakan SnorriCam sebagai alat pengambilan gambar untuk menekankan emosi dari tokoh utama. Teknik P.O.V menggunakan SnorriCam dianggap berhasil menyampaikan emosi tokoh utama dengan lebih baik dan mudah dimengerti oleh penonton. Dalam produksi film ini terdapat 19 scene dengan durasi kurang lebih 15 menit. Film ini didukung dengan berbagai elemen pendukung film, diantaranya poster film, brosur, desain sampul DVD, gantungan kunci, mug, pin, tas jinjing, kaus, trailer film, dan x-banner.

Abstract:

This study deals with the production of durable high strength flowable mortar (HSFM). Firstly, the optimum percentage of silica fume was determined due to Pozzolanic Activity Index (P.A.I) test. Secondly, the selected mortar reinforced by different percentages of steel fibers or hybrid fibers of steel fibers , palm fibers and synthetic fibers (Barchip) to prepare HSFM mixes. Such mixes were tested in compressive strength, splitting tensile strength, static modulus of elasticity, flexural strength, toughness indices determination, and impact load for all the mixes. Lastly, the effects of seawater exposure on the properties of HSFM have been observed. The results show that the use of 10% silica fume as a partial replacement of cement indicate the best P.A.I. On the other hand, the hybridizations of such fibers enhance the performance of HSFM mixes. In addition, the hybrid fibers reduce the permeability of HSFM leading to significance improvement against seawater exposure.

Biocompatibility of materials is one of the most important conditions for their successful application in tissue regeneration and repair. Cell-surface interactions stimulate adhesion and activation of macrophages whose acquaintance can assist in designing novel biomaterials that promote favorable macrophage–biomaterial surface interactions for clinical application. This study is designed to determine the distribution and number of macrophages as a means of biocompatibility evaluation of two newly synthesized materials [silver/poly(vinyl alcohol) (Ag/PVA) and silver/poly(vinyl alcohol)/graphene (Ag/PVA/Gr) nanocomposite hydrogels] in vivo, with approval of the Ethics Committee of the Faculty of Veterinary Medicine, University of Belgrade. The results of the study in terms of the number of giant cells localized around the implant showed that their number was highest on the seventh postoperative day (p.o.d.) in the group implanted with Ag/PVA hydrogels, and on the 30th p.o.d. in the group implanted with Ag/PVA/Gr. Interestingly, the number of macrophages measured in the capsular and pericapsular space was highest in the group implanted with the commercial Suprasorb© material. The increased macrophage number, registered around the Ag/PVA/Gr implant on 60th p.o.d. indicates that the addition of graphene can, in a specific way, modulate different biological responses of tissues in the process of wound healing, regeneration, and integration.

A partir das respostas obtidas nos guias enviados e também pela análise através da Teoria da Mediação Cognitiva – TMC, observamos mudanças de alguns drives por parte dos licenciandos, o que concretiza o P.O.E e também enfatiza a autonomia discente por meio de simulações.

The effects of the European corn borer, Ostrinia nubilalis [Lepidoptera: Pyralidae], on yield of 'Russet Burbank' potatoes (Solanum tuberosum) was investigated at two sites in Prince-Edward-Island in 1989 and 1990. At Summerside, the mean number of larvae per stalk did not exceed 0.19 in either year, and no significant differences in yield were found between untreated plots (check) and plots protected with Bacillus thuringiensis var. kurstaki (B.t.k.). At Tryon in 1989, a seasonal average of 1.16 larvae per stalk reduced total and marketable yields by 7.5% and 8.8%, respectively, compared to plots protected with B.t.k. A similar but not statistically significant trend was observed in 1990 at this site. The higher degree of damage by the European corn borer to potatoes grown on P.E.I., relative to crops grown in more southernly locations, may be due to the longer interval of attack. On P.E.I., the action threshold for the European corn borer on potatoes appears to be between 0.11 and 0.30 egg masses per stalk.

The current research studies the wages of foreign prisoners of war (P.O.W.s) who worked in the Udmurt Autonomous Soviet Socialist Republic in 1942–1948. The paper contains the information about the pays to the P.O.W.s were transferred from the funds of the enterprises where they worked. The article analyses the level of P.O.W. wages, the dependence of the wages from the labour activities and the bonus that the P.O.W.s received for good performance. The paper contains the data on the average level of P.O.W. wages in Izhevsk and lists the kinds of labour activities where the P.O.W.s received the highest wages. The research is based on archival documents, most of which were involved in the scientific use for the first time.It is highly unlikely that the P.O.W.s who were in Udmurtia in 1945–1948 starved since they could use their wages to buy additional provisions in the camp stalls and town shops.

The concept of spirituality is attracting the attention of researchers all over the world. It plays an important role in the maintenance of psychological well-being. The present study was designed to study the effect of ‘OM’ chanting meditation on psychological well-being among adolescents. Practice time for ‘OM’ chanting meditation was 20 minutes and the duration was one month. The sample consisted of 100 subjects (50 in each group) in the range of 15 to 24 years and selected from Dayalbagh Educational Institute, Agra. Psychological Wellbeing was measured by ‘P.G.I. Wellbeing Scale’. Five more items taken from the Life Satisfaction Test were added along with the items of P.G.I. Wellbeing Scale by the researcher herself to improve the validity of the scale. t-test was applied to study the effect of ‘OM’ chanting meditation on psychological wellbeing. Results showed that the calculated t-value in experimental group was found to be 5.59 which is significant at 0.01 level. Calculated t-value in control group was found to be 1.00 which is not significant even at 0.05 level. Result reveals that there is a significant positive effect of ‘OM’ chanting meditation on the psychological well-being of adolescents.

Abstract:

An outbreak of food poisoning in Canada during autumn 1987 was traced to cultured blue mussels (Mytilus edulis) from the Cardigan Bay region of eastern Prince Edward Island (P.E.I.). The toxin, identified as domoic acid, had not previously been found in any shellfish and this outbreak represents the first known occurrence of human poisoning by this neurotoxin. A plankton bloom at the time of the outbreak consisted almost entirely of the pennate diatom, Nitzschia pungens f. multiseries, and a positive correlation was found between the number of N. pungens cells and the concentration of domoic acid in the plankton. Nitzschia pungens f. multiseries isolated from Cardigan Bay produced domoic acid in culture at levels (1 to 20 pg∙cell−1) comparable with values estimated for N. pungens in the plankton samples. Isolates of several Cardigan Bay phytoplankton, including the closely related species Nitzschia seriata, failed to produce domoic acid. Other Nitzschia spp. and two Amphora coffeaeformis isolates also failed to produce domoic acid. We conclude that N. pungens was the major source of the domoic acid in toxic mussels in eastern P.E.I. The recurrence, in November 1988, of a monospecific bloom of N. pungens and the presence of domoic acid in plankton and mussels reinforced this conclusion.

Abstract: Data from the National Socio-Economic Survey of the National Central Bureau of Statistic (Susenas, BPS) 2016 shows that the average amount of national top up the balance spending per month in 2016 reaches Rp 22,182 per capita. It is becoming a business opportunity for top up the balance of retail entrepreneurs. However, in the process of managing the process of input, output and reporting of goods that are done manually (still using paper) are not monitored properly, often there are outgoing or incoming goods whose data is not recorded. This, in the long run, will cause large losses. The purpose of this study is to build applications that function to help provide information that makes it easy to control the process of entry and exit of goods. The system development methodology used is object-oriented system development using the Unified Modeling Language (UML). The results obtained from this research are the design and construction of Inventory Information System POS (Point of Sales) in the form of Context Diagrams and UML Diagrams (Use Case Diagrams, Activity Diagrams, Class Diagrams, and Sequence Diagrams), with the results of black box testing conducted already in operation properly according to the needs of the retail owner. The purpose of the paper is to reveal and demonstrate the importance of accounting in everyday life and particularly in the most adverse conditions of life, through the investigation of the diary of a Prisoner of War (P.O.W.). The author of the diary is a young individual who was an ex- WWII captive and consequently refugee in a concentration camp in Italy, in 1945, with elementary education but absolutely “illiterate” in accounting and economics issues. The handwritten diary of the P.O.W. is the core research evidence used, along with the existing literature regarding accounting in everyday life, including accounting histories of war. Additionally, the study draws on new research firmly embedded in everyday transactions. The detailed analysis of the diary reveals the contribution of improvised and self-taught bookkeeping, to the strengthening of the feeling of survival, the acquisition of the necessary and most asset, the achievement of the insatiable desire of repatriation and finally the acceptance of the patent defect caused from an impairment, through foresight to deal with it. The fulfillment of all the mentioned above aspiration supports that basic accounting knowledge, deriving from the needs of the individual according to specific circumstances of time and place, may be more than satisfied to record simple daily transactions, even by illiterate people. Fecal swabs obtained from a random sample of 1,000 beef slaughter steers and heifers from 123 Prince Edward Island (P.E.I.) farms were examined for the presence of Shiga toxin-producing Escherichia coli (STEC) using a Vero cell assay (VCA). Multiple isolates from each positive sample were tested similarly. VCA-positive isolates were confirmed as E. coli biochemically, tested for drug resistance, serotyped, and tested by polymerase chain reaction (PCR). Animals were classified as positive when an isolate was positive on VCA and the presence of the gene responsible for toxin production was confirmed by PCR. The prevalence of STEC in beef slaughter steers and heifers on P.E.I. was 4% (40 of 1,000). The total number of isolates was 43, and these comprised 26 serotypes, including 13 isolates belonging to 6 serotypes known to be associated with human illness. The most frequently isolated STEC serotype was E. coli O157 (5 isolates out of 43). Of the five E. coli O157 isolates, four were E. coli O157:H7, a serious human pathogen. The majority of STEC isolates, including all O157:H7, isolates, were susceptible to 16 commonly used antimicrobial drugs. According to PCR, 65% of the STEC isolates had the gene for Stx1. Four of these isolates, including two O157: H7, had genes for Shiga toxin (Stx)1 and Stx2.

O processo de envelhecimento é caracterizado por várias modificações no organismo, que podem causar alterações no efeito de certos medicamentos, tornando-os inapropriados para idosos por falta de eficácia terapêutica ou por apresentarem efeitos adversos superiores aos benefícios. O objetivo do estudo foi identificar os medicamentos utilizados pelos idosos assistidos pelo Programa de Atenção ao Idoso (P.A.I.) e investigar o uso de medicamentos potencialmente inapropriados nessa população. Realizou-se estudo quantitativo e descritivo, com coleta de dados secundários, a partir de prontuários/cadastros dos idosos atendidos pelo P.A.I. (Projeto de Extensão Universitária da UNIJUÍ). Entre os idosos cadastrados, 16 receberam e/ou estão recebendo acompanhamento farmacoterapêutico pela equipe da farmácia, sendo 11 mulheres, com idade média de 75,2 ± 7,5 anos. Cada idoso recebe em média 7,3 ± 3,3 medicamentos. A polifarmácia foi verificada em 15 idosos. Entre os 117 medicamentos em uso, considerando-se repetições, 13 são inapropriados para idosos, destacando-se diazepam e fluoxetina, cada um utilizado por três idosos. Verifica-se que é imprescindível reduzir o uso de medicamentos inapropriados para melhorar a qualidade de vida dos idosos. É necessária a colaboração do prescritor e do profissional farmacêutico, que é responsável por avaliar a prescrição, identificar riscos relacionados à terapêutica e intervir através da comunicação com o prescritor responsável, fornecendo informações e sugestões que facilitem a utilização de serviços de saúde.

Trichomycetes, an ecological grouping of fungi and protists associated with the guts of aquatic arthropods, are distributed globally. The diversity of this unique species complex is, however, more completely documented in some parts of the USA and Europe than in Canada. Twenty species of trichomycetes were collected from insect hosts at six sites in Prince Edward Island, situated in the Gulf of St. Lawrence about 14 km off the east coast of New Brunswick. Five new species of Harpellales (Fungi, Kickxellomycotina), Caudomyces longicollis Strongman sp. nov. from Antocha sp. (Tipulidae), Legeriomyces minae Strongman sp. nov. from mayflies, and Smittium ditrichosporum Strongman sp. nov., Smittium insulare Strongman sp. nov., and Smittium tynense Strongman sp. nov. from chironomids are described. Smittium brevisporum L.G. Valle & Santam. and Smittium gracilis L.G. Valle & Santam., previously known only from Spain, are recorded for the first time in North America. A number of species of Harpellales and Amoebidiales (Phylum Mesomycetozoa) previously recorded from the region (P.E.I. and N.S.), and several that are common and widely distributed, were also recovered. Stachylina pedifer Lichtw. & M.C. Williams was collected from chironomids in a stream exposed to seawater during high tide. Trichomycete diversity is high on P.E.I., as it is in many regions where extensive surveys have been conducted, including islands such as Australia and New Zealand. More data from islands may help shed some light on distribution patterns for these obligate endobionts and provide some insights into mechanisms for dispersal.
