The objective of this thesis is to create the documentation for building construction. Designed building is a mountain guesthouse. Building plat is located in the cadastral Poteč. Project documentation has been prepared in accordance with applicable laws, regulations and standards. The object is designed as one floor building with basement and residential loft and is fitted in sloping terrain. The building is shaped in layout L with the longest plane dimensions 40,11 x 39,05 m. Foundations are designed as strips of plain concrete. The building is designed as a wall support system. Wall system is combined of shuttering blocks filled with concrete, ceramic brick blocks and wood timbering. Roofed building is realized through the roof with a vertical stool. The ceiling above the ground floor is designed as a reinforced concrete slab ceiling and above the first floor as the timber.

This diploma thesis is focused on a comparison of construction cost of the house made of classic materials with the equivalent made of natural building materials and consequently on comparison of both construction variants. In order to perform those tasks, the used natural building materials were defined. Furthermore, the specific options of usage of these natural building materials as well as the used composition structures were introduced. The thesis concludes with a price calculation of the constructions made of natural building materials which were not available in the common budgetary software. The appendix of the thesis contains budgets for the first variant, where the house is made of natural building materials, and also for the second variant, where the house is made of classic materials.

Hana Bártíková, Ph.D. Title of diploma thesis: Biotransformation of selected anthelmintics in sheep tapeworm (Moniezia expansa) Biotransformation of anthelmintics is a process that prevents the parasite from adverse effects of xenobiotics. Therefore, it is the main factor that causes the reduction of an anthelmintic effect, resistance development and thus the failure of drug therapy of helminthosis. Therefore many studies focused on anthelmintics biotransformation in helminths have been published. This experiment was aimed at the biotransformation of albendazole, flubendazole and mebendazole in the sheep tapeworm (Moniezia expansa) and the activity of its biotransformation enzymes. Tapeworms were obtained from sheep intestines. Biotransformation of benzimidazoles was determined in in vitro and ex vivo experiments. Flubendazole and mebendazole were metabolized in much greater extent than albendazole. The activity of antioxidative enzymes, which also can participate in drug biotransformation, was detected mainly in the cytosol. My work is based on theme psychological aspects that are significant in social services social rehabilitation adults with learning disabilities for their individual development and motivation. Research is led by question about importance of employment this people. I am interested in relationships and if happens to change of life style or personality after start in employment. These questions are reflected through analysis of literature connected with these topics. Also with support in theoretical platform I designed own implement for measuring. I made 4 interviews with service user social rehabilitation in Textilní dílna Gawain. All procured data are proceed through analysis and is found some categories fully connected with measured sphere. Main conclusion from this research is that social rehabilitation brings contributions in sphere of development individuality adults with learning disabilities, main in development of confidence, self-confidence, responsibility, independence, self-knowledge, empathy, social and communication skills and self-fulfilment. These contributions are result of satisfying needs with employment. Another gain of the research is in open next questions related with this theme.

The aim of this study was to determine and evaluate the chosen factors organic milk quality parameters and daily milk yield (DMY) in the flock of crossbreeds of Lacaune (LC) and East Friesian (EF) ewes from the first to fifth lactation. Experiment occurred during two lactation periods, in 2009 were included in experiment 90 ewes, and in 2010 88 ewes. Milk records and samplings were carried out five times during milking period during the months May (V), June (VI), July (VII), August (VIII) and September (IX). Milk samplings were carried out during the morning milking (6 a.m.). Individual milk samples were analyzed for: total solids (TS, in %), fat (F, in %), protein (P, in %), casein (C, in %), and lactose (L, in %). In addition the samples from chosen ewes (n = 48) were analyzed for: urea (U, in mg/100ml), pH, titratable acidity (TA, in degree SH), somatic cell count (SCC, in 103/ml), rennet clotting time (RCT, in s) and rennet curd quality (RCQ). In the study the effects of year (Y), lactation stage (LS), genotype (G), parity (PA), litter size (LIS) and age (A) of ewes on chosen parameters were evaluated, as well as their interactions with LS. Y had significant effect on all analyzed parameters except DMY. Y had significant effect on pH and SCC too. In 2009 the highest values of TS, F, P, C, L were found in comparison to 2010. In 2010 higher values U, pH, TA, SCC, RCT, RCQ were found in comparison to 2009. LS had significant effect on all analyzed parameters except SCC. The contents of TS, F, P, C and U increased in sheep milk toward the end of the lactation, the DMY logically decreased. At the end of lactation was found the highest value of lactose, the longest RCT and worse RCQ. G had significant effect on all analyzed basic components except P. The ewes of genotype LC 50 EF 50 had the highest DMY and the highest values of all analyzed basic components of milk. Significant effect of PA was observed on DMY, all basic components of milk and pH and TA. The highest DMY was achieved at the fourth lactation. The highest values of TS, F, P, C and L were found at the fourth lactation in all ewes and at the second lactation in the chosen group of ewes (n = 48). LIS had significant effect on all analyzed basic components except L in all ewes. In the Ewes giving birth to 2 lambs had higher DMY and the higher values of all basic components, SCC, the longest RCT and worse RCQ in comparison with ewes giving birth to 1 lamb. A had significant effect on all analyzed basic components and on pH and TA. The ewes in age 4 years had the highest DMY. The highest values of TS, F, P and C were found in age 6 years.

The primary purpose of this study is to determine variability of sheep milk composition and quality. The theoretical part of this study describes composition of sheep milk. It analyses properties and elements which effect quantity and quality of sheep milk. This study research focuses on monitoring and evaluation of changes in sheep milk composition and properties which is seasonaly effected. The research also evaluates the differences in composition and properties of organic and conventional milk. Data for this research were collected between 2014 and 2015. The sample was selected from the East Friesien, Laucane and Cigaja diary breeds. Criteria which were evaluated: the number of microorganisms, active acidity, titratable acidity, coagulation time, curd quality, content of total fat, total protein, total lactose, total solids -non-fat, milk urea, somatic cell count and milk freezing point. Statistical and graphical evaluation of the analyzed data was compared with similar studies.

The diploma thesis deals with development of commodity vertical of sheep meat in New Zealand. It presents literary overview related with agrarian market, agribusiness and commodity vertical of sheep meat in the first part. The analytical part compares production and consumption in the context of world and european development. It describes the major production and consumer centers in the world. The another chapter deals with characteristic of the economy and manufactoring industry in New Zealand. It defines production and consumer trends and the links of foreign trade in New Zealand. It was conducted a future prediction of production and consumption in the final part.

The diploma thesis on "Sectoral environment of processing sheep products" is applied on the Slovakian market. In the thesis, there is elaborated a detailed analysis of this sector from the perspective of structure, strategy and rivalry, demand conditions, related and supportive sectors as well as from the condition factors. Based on this analysis, opportunities and threats following with recommendations for the companies operating in the industry of processing sheep products are formulated. Among all, in the recommendations part it is suggested that the companies should export the surpluses to the Czech and Polish market. To ensure an increased demand, an intensive and well-targeted promotion, both in Slovakia and abroad is essential. This would also enhance the stagnating production of traditional Slovakian sheep cheeses. Furthermore it is important to expand production of high probiotic value products, restore meat production and secure new distribution channels.

The thesis summarizes findings about the microbiology of goat and sheep milk and of farm produced cheese. It describes the production of selected groups of cheeses. It deals with particular microorganisms which are important with respect to technology and which are contained in primary and secondary cultures. It characterizes the undesirable microorganisms which cause contamination and it presents the possible measures to fight with them. Last but not least, there are described the legislative requirements for the farm processing of milk and cheese. The chosen groups of microorganisms were experimentally determined by cultivation methods in samples of raw goat and sheep milk and cheese. It was found out that in some months the values of CPM in milk exceeded the limit given by the Orders of the European Parliament and Counsel 853/2004. The discovered presence of enterococci, coliform bacteria, thermo-resistant aerobic and anaerobic bacteria and other undesirable microorganisms in raw milk and cheese points to the insufficient adherence of hygienic principals and technical procedure.

The diploma thesis evaluated the qualityof eight types of pharmaceutical cheeses, where microbial activity was observed. The growth od the microorganism was monitored at the time of storage for 3 weeks. Coliform microorganisms psychotropic mikroorganisms, yeasts, molds, enterococci, milling bacteria and total microorganisms were analyzed in the cheeses, The sensory quality of the cheese was evaluated in the case of storage, which was analyzed after 7 days for three weeks. The whole review was repeated three times. Changes in sensory parameters were tested: appearance, consistency, taste, smell, cow, sheep and goat milk impression, overall impression.
