Abstract:
Mucosal surfaces represent critical routes for entry and exit of pathogens. As such, animals have evolved strategies to combat infection at these sites, in particular the production of mucus to prevent attachment and to promote subsequent movement of the mucus/microbe away from the underlying epithelial surface. Using biochemical, biophysical, and infection studies, we have investigated the host protective properties of the skin mucus barrier of the Xenopus tropicalis tadpole. Specifically, we have characterized the major structural component of the barrier and shown that it is a mucin glycoprotein (Otogelin-like or Otogl) with similar sequence, domain organization, and structural properties to human gel-forming mucins. This mucin forms the structural basis of a surface barrier (∼6 μm thick), which is depleted through knockdown of Otogl. Crucially, Otogl knockdown leads to susceptibility to infection by the opportunistic pathogen Aeromonas hydrophila. To more accurately reflect its structure, tissue localization, and function, we have renamed Otogl as Xenopus Skin Mucin, or MucXS. Our findings characterize an accessible and tractable model system to define mucus barrier function and host–microbe interactions.

Abstract:
Hearing loss (HL) is the most common sensory disorder in the world population. One common cause of HL is the presence of vestibular schwannoma (VS), a benign tumor of the VIII cranial nerve, arising from Schwann cell (SC) transformation. In the last decade, the increasing incidence of VS has been correlated to electromagnetic field (EMF) exposure, which might be considered a pathogenic cause of VS development and HL. Here, we explore the molecular mechanisms underlying the biologic changes of human SCs and/or their oncogenic transformation following EMF exposure. Through NGS technology and RNA-Seq transcriptomic analysis, we investigated the genomic profile and the differential display of HL-related genes after chronic EMF. We found that chronic EMF exposure modified the cell proliferation, in parallel with intracellular signaling and metabolic pathways changes, mostly related to translation and mitochondrial activities. Importantly, the expression of HL-related genes such as NEFL, TPRN, OTOGL, GJB2, and REST appeared to be deregulated in chronic EMF exposure. In conclusion, we suggest that, at a preclinical stage, EMF exposure might promote the transformation of VS cells and contribute to HL.

In order to study the proclivity of primary amine groups to act as halogen bond acceptors, three aromatic diamines (p-phenylenediamine (pphda), benzidine (bnzd) and o-tolidine (otol)) were cocrystallised with three perfluorinated iodobenzenes (1,4-tetrafluorodiiodobenzene (14tfib), 1,3-tetrafluorodiiodobenzene (13tfib) and 1,3,5-trifluorotriiodobenzene (135tfib)) as halogen bond donors. Five cocrystals were obtained: (pphda)(14tfib), (bnzd)(13tfib)2, (bnzd)(135tfib)4, (otol)(14tfib) and (otol)(135tfib)2. In spite of the variability of both stoichiometries and structures of the cocrystals, in all the prepared cocrystals the amine groups form exclusively I···N halogen bonds, while the amine hydrogen atoms participate mostly in N–H⋯F contacts. The preference of the amine nitrogen atom toward the halogen bond, as opposed to the hydrogen bond (with amine as a donor), is rationalised by means of computed hydrogen and halogen bond energies, indicating that the halogen bond energy between a simple primary amine (methylamine) and a perfluorinated iodobenzene (pentafluoroiodobenze ne) is ca. 15 kJ mol−1 higher than the energy of the (H)NH∙∙∙NH2 hydrogen bond between two amine molecules.

Familial Meniere Disease (FMD) is a rare inner ear disorder characterized by episodic vertigo associated with sensorineural hearing loss, tinnitus and/or aural fullness. We conducted a systematic review to find sequencing studies segregating rare variants in FMD to obtain evidence to support candidate genes for MD. After evaluating the quality of the retrieved records, eight studies were selected to carry out a quantitative synthesis. These articles described 20 single nucleotide variants (SNVs) in 11 genes (FAM136A, DTNA, PRKCB, COCH, DPT, SEMA3D, STRC, HMX2, TMEM55B, OTOG and LSAMP), most of them in singular families—the exception being the OTOG gene. Furthermore, we analyzed the pathogenicity of each SNV and compared its allelic frequency with reference datasets to evaluate its role in the pathogenesis of FMD. By retrieving gene expression data in these genes from different databases, we could classify them according to their gene expression in neural or inner ear tissues. Finally, we evaluated the pattern of inheritance to conclude which genes show an autosomal dominant (AD) or autosomal recessive (AR) inheritance in FMD.

The responses of grassland net primary productivity (NPP) to climate change (CC) and human activities (HA) have received much attention and are inconsistent on different spatial scales. The accurate and quantitative evaluation of the impacts of CC and HA on grassland NPP at a county scale is very important to reveal the external driving factors on grassland NPP and guide the protection of the grassland ecosystem in the arid sandy area of China. In this study, the improved CASA model was adopted to quantify the grassland NPP in Otog Banner, China from 2001 to 2020. The spatiotemporal dynamics of grassland NPP and the relationships between grassland NPP and climate factors in space were analyzed using the methods of simple linear regression and relative sensitivity coefficient. Furthermore, the relative contributions to grassland NPP dynamics caused by CC and HA were explored using the quantitative method based on partial derivative. The results revealed that the mean value of grassland NPP was 175.17 g C m−2 yr−1, and exhibited a significant decrease trend periodically at a rate of 2.14 g C m−2 yr−1 from 2001 to 2020. The spatial distribution of grassland NPP increased from west to east gradually and ranged in 17.48–498.09 g C m−2 yr−1. Grassland NPP exhibited significant linear patterns along the gradients of climate factors, and was the most sensitive to sunshine duration (SSD). Approximately 86.83% of the grassland showed a degradation trend and 39.71% showed a serious degradation trend. The CC contribution to grassland NPP dynamics was 0.593 g C m−2 yr−1, and precipitation was the key driving climate factor, while the contribution of HA was −2.733 g C m−2 yr−1, which was the primary factor leading to large-scale degradation of grassland in Otog banner. This study indicates that the status of the grassland ecosystem in Otog Banner is not optimistic, and measures for grassland ecosystem restoration and improvement need to be further strengthened.

For this reason, the resolution of single cases not belonging to well-defined syndromes often requires an integrated approach of multiple whole-genome technologies. Such an approach has also unexpectedly revealed a complex molecular basis in an increasing number of patients, for whom the original suspect of a pleiotropic syndrome has been resolved as the summation effect of multiple genes. We describe a 10-year-old boy, the third son of first-cousin parents, with global developmental delay, facial dysmorphism, and bilateral deafness. SNP-array analysis revealed regions of homozygosity (ROHs) in multiple chromosome regions. Whole-exome sequencing prioritized on gene-mapping into the ROHs showed homozygosity for the likely pathogenic c.1097_1098delAG p. (Arg366Thrfs*2) frameshift substitution in LARP7 and the likely pathogenic c.5743C>T p.(Arg1915*) nonsense variant in OTOG. Recessive variants in LARP7 cause Alazami syndrome, while variants in OTOG cause an extremely rare autosomal recessive form of neurosensorial deafness. Previously unreported features were acrocyanosis and palmoplantar hyperhidrosis. This case highlights the utility of encouraging technological updates in medical genetics laboratories involved in the study of neurodevelopmental disorders and integrating laboratory outputs with the competencies of next-generation clinicians.

The P(otol)3 structures comprise an isomorphous series distinct from the previously published chloride, crystallizing with two independent molecules in the asymmetric unit in the space group Pbca, with a ≈ 20.0, b ≈ 28.0, c ≈ 14.0 Å. The molecules associate to form dimers through a back-to-back sextuple phenyl embrace (6PE). The P(ptol)3 complexes are isomorphous with the previously published chloride, crystallizing with two independent molecules in the unit cell in the space group P21/c, with a ≈ 10.0, b ≈ 22.0, c ≈ 19.5 Å, β ≈ 99°. The molecules associate through edge-to-face interactions along the direction of the a-axis. Preparation of the chloride by anodic dissolution of gold in an acetonitrile solution of the ligand and aqueous HCl yields a new polymorph, crystallizing in the space group Aba2, with a 19.738(2), b 11.813(3), c 17.645(1) Å. Despite the bulkiness of the phosphine ligand, this form of [AuP(ptol)3Cl] exhibits a short intermolecular Au…Au contact distance of 3.375(1) Å, indicative of a significant aurophilic interaction. The ranges for the Au—P, Au—Cl, Au—Br, Au—I bond lengths and P—Au—X bond angles in the series are: 2.201(3)–2.265(4), 2.255(3)–2.290(4), 2.388(2)–2.411(2), 2.542(1)–2.556(2) Å, and 173.2(2)–179.6(1)° respectively.

Such high tensile properties are the result of the high draw ratio (up to x8) while annealing at 400°C. The homopolyimide (BPDA-OTOL) is found to adopt a triclinic crystal lattice, based on wide angle X-ray and electron diffraction experiments. In the copolyimide, it's crystal lattice is disturbed by the introduction of the PPD comonomer. As the draw ratio of the copolymer fibre is increased, the crystallinity, overall orientation and the apparent axial crystallite sizes also increased, as evidenced by wide angle X-ray diffraction, optical birefringence and density measurements. These structural developments critically affect the tensile properties of the fibres. The glass transition temperature of the copolyimide is 308°C, based on thermal mechanical measurements. Dimensional stability is retained up to about 300°C. At around 100–250°C, a subglass transition is observed in dynamic mechanical measurements, and is assigned as a β relaxation. The activation energy of this relaxation is 140 hJ/mol, based on its frequency dependence. The α relaxation corresponds to the glass transition. As the crystallinity increases with drawing, this relaxation is suppressed due to the restriction of molecular segmental motion.

Lichen planus (LP) is an idiopathic disorder that presents with cutaneous and genital manifestations. Esophageal LP (ELP) was first described by Al-Shihabi and Jackson [J Laryngol Otol 1982;96:567-571] in 1982. Only approximately 80 cases have been documented in the literature since. It is a rare and underrecognized disorder, leading to a delay in diagnosis and a lack of standardized management. We describe the presentation, diagnosis, and management of 6 cases of ELP, at a tertiary institution, because we believe that an increasing awareness of this condition can help identify more cases and increase our understanding of this interesting condition.

The runic inscriptions found on Arvas mountain in the Urandösh town of Üzüür city, Otog county, Ordos, Inner Mongolia Autonomous Region, China, were the subject of the research we conducted and readings in this article. In the Kharankhui Valley of Arvas Mountain, there are three pieces of runic inscriptions. The earliest, and most obvious, inscription was recorded in 2012, although researchers have just recently become aware of it. Content-wise, the first inscription is the note of travel, and the second and third are dedicated to his father. But it falls under the category of carved inscriptions in terms of writing technique.
