Abstract:
In this paper, the purification and biochemical characterization of the endogenous oyster (Crassostrea gigas) trypsin inhibitor were researched. A oyster trypsin inhibitor（OTI）has been purified by successive ammonium sulfate precipitation, gel filtration, affinity chromatography and high performance reversed-phase liquid chromatography. OTI has a molecular weight of approximately 5036 Da estimated by high performance size exclusive liquid chromatography. OTI was heat-, acid- and basic-stable competitive trypsin inhibitor. And OTI was double-head inhibitor with the inhibition constant (Ki) value of 1.644×10-2 mmol L-1. OTI was composed of nine kinds of amino acid, and rich in cysteine, alanine and glutamic acid. Furthermore, OTI can inhibit the proliferations of human lung adenocarcinoma A549 cell and human cervical cancer Hela cell

Abstract:
ABSTRACT Strategies to prime CD8+ T cells against Murine gammaherpesvirus 68 (γHV68; MHV68) latency have, to date, resulted in only limited effects. While early forms of latency (<21 days) were significantly reduced, effects were not seen at later times, indicating loss of control by the primed CD8+ T cells. In the present study, we evaluated CD8+ T cells in an optimized system, consisting of OTI T-cell-receptor (TCR) transgenic mice, which generate clonal CD8+ T cells specific for Kb-SIINFEKL of OVA, and a recombinant γHV68 that expresses OVA (γHV68.OVA). Our aim was to test whether this optimized system would result in more effective control not only of acute infection but also of later forms of latent infection than was seen with previous strategies. First, we show that OTI CD8+ T cells effectively controlled acute replication of γHV68.OVA in liver, lung, and spleen at 8 and 16 days after infection of OTI/RAG mice, which lack expression of B and CD4+ T cells. However, we found that, despite eliminating detectable acute replication, the OTI CD8+ T cells did not prevent the establishment of latency in the OTI/RAG mice. We next evaluated the effectiveness of OTI T cells in OTI/B6 animals, which express B cells—a major site of latency in wild-type mice—and CD4+ T cells. In OTI/B6 mice OTI CD8+ T cells not only reduced the frequency of cells that reactivate from latency and the frequency of cells bearing the viral genome at 16 days after infection (similar to what has been reported before) but also were effective at reducing latency at 42 days after infection. Together, these data show that CD8+ T cells are sufficient, in the absence of B cells and CD4+ T cells, for effective control of acute replication. The data also demonstrate for the first time that a strong CD8+ T-cell response can limit long-term latent infection.

Purpose The purpose of this paper is to investigate the integrative effects of abusive supervision (AS), perceived injustice (PI) and ethical conflict (EC) on occupational turnover intentions (OTI) among frontline employees (FLEs). Furthermore, the mediating role of quiescent silence (QS) is examined. Design/methodology/approach The authors collected survey-based data from 320 FLEs working in different service sector organizations of Pakistan. The snow-ball sampling technique is employed to approach respondents of this study. The collected data are analyzed through structural equation modeling (SEM) technique using SmartPLS 3.0. Findings The AS, PI and EC are found to collectively predict OTI among FLEs with a mediating effect of QS. The EC is the strongest predictor of OTI. Practical implications The results imply that service managers must ensure that employees are treated fairly. Furthermore, relevant trainings are offered to FLEs in order to maintain the required ethical standards at work to reduce EC perceptions. Originality/value The study is pioneer in presenting the relationship between QS and occupational turnover among FLEs. Furthermore, the study advocates the mediating role of employee quiescent silence to enrich the relationship between socio-psychological factors (i.e. AS, EC and PI) and OTI.

Unmanned aerial vehicles (UAV), also known as drones, are gaining more and more practical application in modern society, particularly as the tools of implementation of the concepts of «smart city», «smart health care», «smart industries», Internet of Things, 3D mapping, digital transport. But currently it is impossible to use of UAV at certain objects, comprising objects of transport infrastructure (OTI), primarily, airports, because of existing restrictions due to security threats arising during the UAV flight.The authors of the present work have set a goal to offer a solution that allows to start operating UAV at transport infrastructure facilities that are currently prohibited for UAV flights. To achieve the objective of the work, using analysis and synthesis, comparison and generalization, factors and conditions for safe use of UAV at OTI have been formulated, a method for increasing security of UAV flight has been developed, followed by the suggestion on the UAV route control system.The proposed system makes it possible to safely use UAVs at OTI by restraining their flight area strictly to the designated corridor, which eliminates a threat of a collision of UAV with other vehicles operated at OTI, dangerous elements of OTI, as well as with people at the object.The system does not need electric power feeding, which makes it possible to implement the system without creating an auxiliary power supply infrastructure.The practical application of the proposed system and, as a consequence, implementation of a greater number of opportunities for the use of UAV, are capable to generate fundamentally new technological processes and structures at transport facilities, which is one of the directions for creating the next generation transport infrastructure based on IoT and artificial intelligence.

Abstract:
Advanced age or preexisting comorbidities have been characterized as risk factors for severe coronavirus disease 2019 (COVID-19) cases requiring hospitalization and intensive care. In recent years, clonal hematopoiesis (CH) of indeterminate potential (CHIP) has emerged as a risk factor for chronic inflammatory background and subsequent aging-associated diseases. The purpose of this study was to identify biological factors (particularly leukocyte subtypes and inflammatory markers) associated with a risk of clinical deterioration (i.e., orotracheal intubation (OTI)) and to determine whether CH was likely to influence clinical and biological behavior in patients with severe COVID-19 requiring hospitalization. Here, we describe clinical and biological features, including the screening of CHIP mutants in a well-annotated cohort of 122 hospitalized patients with a laboratory-confirmed diagnosis of COVID-19 (55% requiring OTI). We showed that elevated white blood cell counts, especially neutrophils and high C-reactive protein (CRP) levels at admission, were associated with an increased requirement of OTI. We noticed a high prevalence of CH (25%, 38%, 56%, and 82% of patients aged <60 years, 60–70 years, 70–80 years, and >80 years) compared to a retrospective cohort of patients free of hematological malignancy explored with the same pipelines (10%, 21%, 37%, and 44%). However, the existence of CH did not significantly impact clinical outcome, including OTI or death, and did not correlate with other laboratory findings.

Rubble islands are dynamic sedimentary features present on reef platforms that evolve under a variety of morphodynamic processes and controlling mechanisms. They provide valuable inhabitable land for small island nations, critical habitat for numerous species, and are threatened by climate change. Aiming to investigate the controlling mechanisms dictating the evolution of One Tree Island (OTI), a rubble island in the Southern Great Barrier Reef, we combined different remotely-sensed data across varying timescales with wave data extracted from satellite altimetry and cyclone activity. Our findings show that (1) OTI had expanded by 7% between 1978 and 2019, (2) significant gross planform decadal adjustments were governed by the amount, intensity, proximity, and relative position of cyclones as well as El Niño Southern Oscillation (ENSO) phases, and (3) the mechanisms of island growth involve rubble spits delivering and redistributing rubble to the island through alongshore sediment transport and wave overtopping. Frequent short-term monitoring of the island and further research coupling variations in the different factors driving island change (i.e., sediment availability, reef-wave interactions, and extreme events) are needed to shed light on the future trajectory of OTI and other rubble islands under a climate change scenario.

Abstract This study evaluated the effects of a pyramidal training approach that used an expert trainer who taught teachers how to train their paraeducators. Three special education teachers were taught to train four paraeducators to provide students with intellectual and developmental disabilities opportunities to initiate (OTI). A multiple baseline design across participants was used to evaluate the rate and fidelity that paraeducators provided OTI and least to most prompting strategies with students. Rates increased from 0 to an average of .58 per minute. Fidelity of implementation increased from 0% to an average of 94.5%. Maintenance data were recorded for three paraeducators. Schools should consider using this cost- and time-effective training model with staff.

Gegenstand der Arbeit sind verschiedene Aspekte der Morphosyntax und Tonologie im Buli, einer in Ghana (Upper East Region nahe der Grenze zu Burkina Faso) gesprochenen Gursprache des Niger-Kongo-Phylums, die bisher nur unzureichend beschrieben wurde. Die grammatische Beschreibung gibt einen umfangreichen Einblick in das sprachliche System des Buli und umfasst Kapitel zu den folgenden Themen: Phonologie, Nominalklassensystem, Pronominalsystem, Komplexe Nominalphrasen und nominale Syntagmen, Verbsystem. Dies geschieht in erster Linie anhand des in Wiaga gesprochenen Buli, wird aber durch die Bezugnahme auf andere Dialekte sowie auf verwandte Gursprachen ergänzt, beispielsweise hinsichtlich der Bewertung tonaler Divergenzen in Nominalklassensuffixen oder in Assoziativkonstruktionen. Die autosegmental angelegte Tonstudie verfolgt keine theoretischen Ziele, sondern hat sich unmittelbar aus dem Bedarf einer adäquaten Beschreibung der synchronen Sprache ergeben, in der Ton sowohl in lexikalischer als auch in grammatischer Hinsicht ganz zentrale Aufgaben übernimmt. Dabei wurden neben regelmäßigen Ton–Sandhi-Erscheinungen (Tieftonausbreitung) auch interessante Phänomene an den Schnittstellen der Phonologie zur Syntax und zur Semantik-Pragmatik vorgefunden, z.B. die tonale Kongruenz bestimmter Verbformen mit der Diskursrolle des Subjekts (+/- Diskurspartizipant, d.h. 1./2. Person vs. 3. Person) oder das Vorkommen eines Grenztons an äußerungsfinalen Morphemen, zu denen auch die Klassensuffixe indefiniter Substantive und enklitische Objektpronomen am Verb gehören. Im Rahmen der morphosyntaktischen Analysen wurde unter anderem ein aus typologischer Sicht spannender nominaler Kompositionstyp identifiziert, der sich neben seinen strukturellen Eigenschaften auch durch eine spezifische possessive Semantik auszeichnet und vermutlich an der Entwicklung von attributiven (qualifizierenden) Adjektiven und eines von zwei Zahlwörtern für ‘eins’ beteiligt war.
This work deals with different aspects of the morphosyntax and tonology in Buli, a Gur language of the Niger-Congo-Phylum, which is spoken in Ghana (Upper East Region near the border to Burkina Faso) and has so far been described insufficiently. The grammatical description provides an extensive insight into the linguistic system of Buli and contains chapters concerning the following topics: phonology, noun class system, pronominal system, complex noun phrases and nominal syntagmata, and verb system. It is primarily based on the Buli variant spoken in Wiaga and supplemented by reference to other dialects as well as to related Gur languages, for instance with regard to tonal divergencies in noun class suffixes and in associative constructions. Following an autosegmental model, the tonal study does not aim at theoretic enhancement, but results directly from the need of an adequate description of the synchronic language in which tone plays an important role in lexicon and grammar. Besides regular Tone-Sandhi (Low tone spreading), interesting phenomena at the phonology/syntax interface as well as at the phonology/semantics-pragmatics interface were observed – e.g. some verb forms displaying tonal agreement with the discourse role of the subject (+/- discourse participant, i.e. 1st / 2nd vs. 3rd person) or the appearance of a boundary tone on utterance-final morphemes, including the noun class suffixes of indefinite nouns and the enclitic object pronouns at the verb. In the course of the morphosyntactic analysis, a typologically remarkable nominal compound type was identified that is distinguished by structural features as well as by specific possessive semantics and can be assumed to be involved in the development of attributive (qualifying) adjectives and one of two numerals with the meaning ‘one’.

This thesis explores the working realities of gay police officers and performers in relation to ‘gay-friendly’/‘gay-hostile’ worksites and embodied sexual identity, developing an understanding of the meanings gay workers attach to their working lives by mobilising conceptual resources primarily from sociology. Deep seated assumptions pervade current perceptions regarding gay male sexuality and certain occupations. The idea is that there are gay industries like fashion, nursing and the performing arts. In contrast, occupations such as the police and the armed forces are often seen as homophobic, yet a dearth of academic research investigates the lived experiences of gay men located within perceived ‘gay friendly’ or ‘gay hostile’ worksites. Acknowledging this as a missed opportunity for developing empirical insight, I bring to the fore the work realities of some of these overlooked people. Taking the performing arts as an example of a ‘gay-friendly’ occupation, the police as an example of a ‘gay-hostile’ occupation, and drawing on in-depth interview data with 20 gay performers/police officers, I show that the perspectives and experiences of these men allow us to nuance existing research on how LGB employees understand, value and experience ‘gay-friendly’ workplaces, an emerging construct in the organisation studies literature. Focusing on the significance of embodied, sexual identity for the performance of the occupational roles of interest allows this study to consider the relationship between gender and sexuality at work. Literature on the gendered nature of work along with the promising literature on (homo)sexuality in the workplace have proceeded relatively separately, with the exception of the literature on sexualized labour and the commodification of women's (assumed hetero)sexuality in sales-service work (Tyler, 1997). The effect is that the experience and performance of gender and/in/through sexuality at work has been neglected as a topic of empirical investigation. Although sociologists argue that sexuality cannot be understood without reference to gender, and vice versa, few organizational scholars explore the experiences of work with this in mind. This thesis addresses this gap in knowledge. It brings together the perspectives of gay performers and police officers and highlights the prevalence of a „gender imperative‟ throughout the day-to-day lives of these workers. In detailing the workplace experiences of my participants, this thesis also builds on existing studies that tend to focus solely on the general working lives of gay employees. Gay workers face important contextual issues relating to 'passing', 'coming out' and homophobia. Although these are key areas of interest to existing literature, studies so far fail to address these concepts in detail with reference to specific occupational settings. In other words, the research contributes to the area of lesbian, gay and bisexual (LGB) disclosure and management at work. Stigma-based models (Goffman, 1963) are particularly useful here in framing some of the empirical insights of my research.

Abstract:
L'investigation du système La4TI3O12-LaTiO3 a permis de mettre en évidence une nouvelle série homologue de formulation LanTin-xO3n. Les structures de ces phases ont été caractérisées par diffraction des rayons X sur poudre et par microscopie électronique en transmission. Elles dérivent de différents polytypes de perovskites hexagonales et présentent une distorsion structurale due a la rotation coopérative des octaèdres autour de leurs axes ternaires. Une nouvelle serie de composes Ba2MM' F7Cl (M, M' = Mn, Fe, Co, Ni, Zn), dont les structures ont été déterminées par diffraction des rayons X sur monocristal, a été par ailleurs mis en évidence. Une étude comparative des propriétés structurales et magnétiques avec celles des composés de la série BaMF4 a été effectuée (filiation structurale résultant de différentes rotations coopératives des octaèdres, mesures d' aimantation sur poudre et monocristal, détermination des structures magnétiques de Ba2Co2F7Cl et Ba2Ni2F7Cl par diffraction des neutrons). Un nouveau chlorofluorure du cuivre a également été caractérise sur le plan structural et magnétique. Dans ce cas, une filiation structurale avec la structure de l'apatite et avec celle de phases derivees est proposée.

Abstract:
This master’s thesis deals with designs and implementation of systems for music cover recognition. The introduction part is devoted to the calculation parameters from audio signal using Music Information Retrieval techniques. Subsequently, various forms of cover versions and musical aspects that cover versions share are defined. The thesis also deals in detail with the creation and distribution of a database of cover versions. Furthermore, the work presents methods and techniques for comparing and processing the calculated parameters. Attention is then paid to the OTI method, CSM calculation and methods dealing with parameter selection. The next part of the thesis is devoted to the design of systems for recognizing cover versions. Then there are compared systems already designed for recognizing cover versions. Furthermore, the thesis describes machine learning techniques and evaluation methods for evaluating the classification with a special emphasis on artificial neural networks. The last part of the thesis deals with the implementation of two systems in MATLAB and Python. These systems are then tested on the created database of cover versions.

Increasing traffic volumes in the Internet put strict requirements to the architectureof optical core networks. The exploding number of Internet users, andmassive increase in Internet content consumption forces carriers to constantlyupgrade and transform their core networks in order to cope with the trafficgrowth. The choice of both physical components and transport protocols inthe core network is crucial in order to provide satisfactorily performance.Data traffic in the core network consists of a wide variety of protocols.OTN is a digital wrapper technology, responsible for encapsulating existingframes of data, regardless of native protocol, and adding additional overheadfor addressing, OAM and error control. The wrapped signal is thentransported directly over wavelengths in the optical transport network. Thecommon OTN wrapper overhead makes it possible to monitor and controlthe signals, regardless of the protocol type being transported.OTN is standardized by the ITU through a series of recommendations,the two most important being ITU-T G.709 - "Interfaces for the OpticalTransport Network", and ITU-T G.872 - "Architecture of the Optical TransportNetwork". OTN uses a flexible TDM hierarchy in order to provide highwavelength utilization. The TDM hierarchy makes it possible to performswitching at various sub-wavelength bit rates in network nodes.An introduction to OTN and an overview of recent progress in OTNstandardization is given in the thesis. An OTN switch which utilizes theflexible multiplexing hierarchy of OTN is proposed, and its characteristics istested in a network scenario, comparing it to the packet switched alternative.Simulation results reveal that OTN switching doesn’t provide any performancebenefits compared to packet switching in the core network. OTNswitches do however provide bypass of intermediate IP routers, reducing therequirements for router processing power in each network node. This reducesoverall cost, and improves network scalability.An automatically reconfigurable OTN switch which rearranges link subcapacitiesbased on differences in output buffer queue lengths is also proposedand simulated in the thesis. Simulation results show that the reconfigurableOTN switch has better performance than both pure packet switching andregular OTN switching in the network scenario.

Abstract:
Abstract The present dissertation analyzes the role of the fairy tale in the literary works of the Czech author Ota Pavel. Ota Pavel (1930–1973) started as a sports reporter and later became a popular and esteemed writer of fiction, mainly short stories. His works have been translated to many languages, among them Swedish. The study consists of a biographical reading of Ota Pavel’s texts and a discussion of the function of the fairy tale in his stories. My analysis of his literary development and use of various prose elements is based on narratological theories of the fairy tale – Karel Čapek’s motifs and fairytale characters, Gérard Genette’s paratexts and Vladimir Propp’s functions and dramatis personae – and includes remarks on the function of diminutives, etc. Ota Pavel is also viewed in the context of the literary landscape of his time – the 1960s and 1970s – and the reception of his work is discussed. Pavel’s stories contain a number of characteristic features which together form his unique style: direct allusions and references to fairy tales--for example, fairies and witches – and the linguistic markers and distinctive use of the narrator’s voice. One of the most common characters in Ota Pavel’s work is the hero, who, often with the assistance of magical objects or supernatural helpers, defeats supernatural antagonists or solves impossible tasks. Developing and growing as he – the hero is usually male–strives toward his goal, Pavel’s protagonist struggles not with dragons and giants, but with himself and his own difficulties and misfortunes. Earlier research refers to Ota Pavel’s two “genres”: sports prose and retrospective, or “memory” prose. This study shows that the boundary between the two is fluid, and that all Pavel’s work might be called memory prose. The title of this thesis – Ota Pavels sagovärld (The Fairytale World of Ota Pavel) – indicates the perspective from which the subject is approached. On the one hand, it alludes to the influence of the fairy tale on Pavel’s work, while on the other it points to the fact that his own life was such that he sometimes needed to retreat into a fantasy world of his own. Abstract:
In this thesis, opti-acoustic stereo imaging, which is the deployment of two-dimensional (2D) high frequency imaging sonar with the electro-optical camera in calibrated stereo configuration, is studied. Optical cameras give detailed images in clear waters. However, in dark or turbid waters, information coming from electro-optical sensor is insufficient for accurate scene perception. Imaging sonars, also known as acoustic cameras, can provide enhanced target details under these scenarios. To illustrate these visibility conditions, a 2D high frequency imaging sonar simulator as well as an underwater optical image simulator is developed. A computationally efficient algorithm is also proposed for the post-processing of the returned sonar signals. Where optical visibility allows, integration of the sonar and optical images effectively provides binocular stereo vision capability and enables the recovery of three-dimensional (3D) structural information. This requires solving the feature correspondence problem for these completely different sensing modalities. Geometrical interpretation of this problem is examined on the simulated optical and sonar images. Matching the features manually, 3D reconstruction performance of opti-acoustic system is also investigated. In addition, motion estimation from opti-acoustic image sequences is studied. Finally, a method is proposed to improve the degraded optical images with the help of sonar images. First, a nonlinear mapping is found to match local the features in opti-acoustical images. Next, features in the sonar image is mapped to the optical image using the transformation. Performance of the mapping is evaluated for different scene geometries.

In recent years, the number of OTN devices in backbone and core equipment rooms has increased sharply. However, due to factors such as equipment room planning, air conditioner, and power supply, new electrical racks cannot be installed in the same equipment room as original optical racks during OTN expansion of 80-wavelength systems. The remote deployment of OTN electrical racks has certain impact on OTN system indicators, OM/OD, and OTU optical-layer parameters. This document analyzes the factors that are affected by the remote deployment of OTN electrical racks, creates simulation models based on scenarios, and provides suggestions on the remote deployment design of OTN electrical racks.

The 3-PRS parallel manipulator with different arrangements of prismatic joints is called a reconfigurable 3-PRS parallel manipulator in this paper. The three prismatic joints in PRS limbs are attached to the base with an angle α between the horizontal plane of the base and their directions. Based on [1], the manipulator has identical operation modes, namely x0 = 0 and x3 = 0 for any value of α. Accordingly, this paper presents in more details the performance evaluation of these operation modes by using the output transmission index (OTI) and the constraint transmission index (CTI). The OTI and CTI determine the force transmission efficiency and the constraining ability of the manipulators, respectively. Initially, the determination of the number and types of operation modes of the 3-PRS parallel manipulator is recalled. The computation is carried out by using an algebraic approach, namely the Study kinematic mapping. In each operation mode, the actuation wrenches and the constraint wrenches are obtained based on the Screw theory. Then, the OTI and CTI are traced in the orientation workspace of the manipulator for different values of angle α. Furthermore, the singularity conditions are analysed corresponding to the values of OTI and CTI.

Abstract:
The study of supercavitating projectiles has become an important research subject in the last years and some experimental results are presented here, obtained on supercavitating projectiles fired horizontally into a small test reservoir (1 × 0.5 × 0.5 m3). During these tests spherical, cylindrical and truncated cone projectiles were fired with velocities from 600 to 1000 m/s. The experimental results are compared with the OTi-HULL code (a penetration code of continuum mechanics, useful here but not very well adapted) and with Levinson’s and Serebryakov’s formulas. The comparison shows that: 1) the HULL code underestimates the cavity width by 20 to 40% except for the spherical projectiles, 2) the widths obtained with the Levinson and Serebryakov formulas are close to one another but are also inferior to the experimental values (nearly 10%), 3) the experimental drag coefficients approach the theoretical ones but the HULL code overestimates the drag by nearly 10 to 20%. These experiments confirm the feasibility of these tests at ISL and they will be continued in a larger reservoir.

At present, the research of motion/force characteristics is concentrated on the single-platform parallel mechanisms (PMs). On the contrary, relatively little attention has been paid to the double-platform PMs, and there is also no unified index. What actually causes this? One of the reasons is that the complex and inaccessible wrench system of this manipulator with the double platforms. Especially the transmission wrench screws (TWSs) are no longer like its traditional concept, i.e., linear forces along the symmetric axis of the passive arms. Essentially, the TWSs in the double-platform PMs should be applied to the end effector but not to the sub-platforms from active arms. With this new idea, the concept equivalent transmission wrench screw (ETWS) of PMs with double platforms is firstly proposed and its identification process is also provided. Then, the motion/force transmission index which considers the input transmission index (ITI) and output transmission index (OTI) is extended to the area of kinematic performance evaluation of the double-platform PMs. Finally, the validity of the proposed extension is verified by the transmission index (TI) based on the concept of pressure angle. In this paper, the typical Par4 mechanism is investigated as an example. This extended approach with a new definition of the ETWS which covers the concept of the TWS in traditional mechanism actually provides a uniform kinematic performance index of both single- and double-platforms PMs.

Abstract:
Oracle Bone Inscriptions (OBI) research is very meaningful for both history and literature. In this paper, we introduce our contributions in AI-Powered Oracle Bone (OB) fragments rejoining and OBI recognition. (1) We build a real-world dataset OB-Rejoin, and propose an effective OB rejoining algorithm which yields a top-10 accuracy of 98.39%. (2) We design a practical annotation software to facilitate OBI annotation, and build OracleBone-8000, a large-scale dataset with character-level annotations. We adopt deep learning based scene text detection algorithms for OBI localization, which yield an F-score of 89.7%. We propose a novel deep template matching algorithm for OBI recognition which achieves an overall accuracy of 80.9%. Since we have been cooperating closely with OBI domain experts, our effort above helps advance their research.
