Abstract:
Innovations are required for electric vehicles (EVs) to be lighter and more energy efficient due to the range anxiety issue. This article introduces an intelligent control of an organic structure solar supercapacitor (OSSC) for EVs to meet electrical load demands with solar renewable energy. A carbon fibre-reinforced polymer, nano zinc oxide (ZnO), and copper oxide (CuO) fillers have been used in the development of OSSC prototypes. The organic solar cell, electrical circuits, converter, controller, circuit breaker switch, and batteries were all integrated for the modelling of OSSCs. A carbon fibre (CF)-reinforced CuO-doped polymer was utilised to improve the concentration of electrons. The negative electrodes of the CF were strengthened with nano ZnO epoxy to increase the mobility of electrons as an n-type semiconductor (energy band gap 3.2–3.4 eV) and subsequently increased to 3.5 eV by adding 6% π-carbon. The electrodes of the CF were strengthened with epoxy-filled nano-CuO as a p-type semiconductor to facilitate bore/positive charging. They improve the conductivity of the OSSC. The OSSC power storage was controlled by an adaptive neuro-fuzzy intelligent system controller to meet the load demand of EVs and auxiliary battery charging. Moreover, a fully charged OSSC (solar irradiance = 1000 W/m2) produced 561 W·h/m2 to meet the vehicle load demand with 45 A of auxiliary battery charging current. Therefore, the OSSC can save 15% in energy efficiency and contribute to emission control. The integration of an OSSC with an EV battery can minimise the weight and capacity of the battery by 7.5% and 10%, respectively.

Abstract:
Purpose Open Source software companies (OSSCs) are confronted with institutional pressures from Open Source software (OSS) communities. They must find an acceptable balance between the expectations of these communities and their own business model. However, there are still few studies that try to analyse the OSSC business models. The purpose of this paper is to highlight OSSC typical business models by using rich empirical data. Design/methodology/approach The methodology is based on a combination of quantitative analysis of a sample of 66 OSSCs and qualitative analysis of three typical situations resulting from that sample. Findings The quantitative study enables the authors to highlight three typical business models. The in-depth study of three typical cases enables the authors to specify these OSSC business models. The authors can distinguish four key dimensions: the relationship developed with the OSS communities, the strategic manoeuvres made, the key resources and competitive positioning. Research limitations/implications The results indicate that it is possible for firms to accommodate both profit and non-profit logics using different strategic manoeuvres to position themselves with regard to the Open Source institutional environment. Such accommodation requires the development of key resources and the adoption of suitable competitive positioning. Practical implications This study allows the authors to highlight two main practical contributions for OSSCs’ directors. First, the different manoeuvres identified may help them to ensure coherence between their strategic choices and the business model chosen. Second, the results can help OSSC founders identify value creation mechanisms more clearly by analysing four key variables. Originality/value This paper provides new insight about OSSCs business models. It aggregates four dimensions that provide a more “fine-grained” analysis of business models, while other studies often emphasise one dimension (usually the regime of appropriability).

Abstract:
Mesangial cell proliferation contributes to the development of glomerulosclerosis in diabetic nephropathy. This study was aimed at determining whether Osteomeles schwerinae (OSSC) extract can ameliorate renal damage in Spontaneously Diabetic Torii (SDT) rats. OSSC extract (100 and 250 mg/kg/day) was administered to the SDT rats through oral gavage for 17 weeks. At the end of the experiment, glucose, HbA1c, and albuminuria were measured. In addition, the levels of mesangial proliferation-related proteins were determined by western blotting and immunohistochemistry. Our results show that albuminuria, accumulation of the extracellular matrix (ECM), and renal expansion were markedly restored by OSSC extract administration. The OSSC treatment also inhibited α-smooth muscle actin and transforming growth factor-β1 protein expression. In addition, OSSC and its bioactive compounds hyperoside and quercitrin inhibited the platelet-derived growth factor-BB (PDGF-BB)/platelet-derived growth factor-B receptor (PDGFR-β) ligand binding in an in vitro assay. Taken together, these results indicate that OSSC inhibits ECM accumulation and mesangial proliferation of the glomeruli in SDT rats through inhibition of the interaction between PDGF-BB and PDGFR-β. OSSC has ameliorating effects on the initiation and progression of diabetes complications and can be used for the treatment of early diabetic renal dysfunction.

The accumulation and formation of advanced glycation end products (AGEs) are related to diabetes and age-related disease. Osteomeles schwerinae C. K. Schneid. (Rosaceae, OSSC) is used traditionally for the treatment of various diseases in Asia. Previous studies have shown that OSSC elicits preventive effects in an in vivo model of diabetes. This study was to evaluate the antiapoptotic effects of dried leaves and twigs of OSSC extract and its major compounds in ARPE-19 cells—spontaneously arising human retinal pigment epithelial cells—under diabetic conditions. To examine the effects of an OSSC extract and its active compounds (acetylvitexin, hyperoside and quercitrin) on apoptosis in methylglyoxal (MG, the active precursor in the formation of AGEs)-treated ARPE-19 cells and the mechanism by which these effects occur, apoptosis was measured using flow cytometry analysis. Protein expression levels of phospho-p53 (p-p53), Bax and Bcl-2 were determined by western blot analyses. The OSSC extract inhibited apoptosis in MG-treated ARPE-19 cells in a dose-dependent manner. The major compounds also reduced the rate of apoptosis. Both the extract and major compounds also inhibited the expression of p-p53 and Bax and increased the levels of Bcl-2 that had been previously reduced by MG treatment. The OSSC extract (0.1 μg/mL) and its major compounds (0.01 μM) attenuated apoptosis in ARPE-19 cells under toxic diabetic conditions by downregulating of expression of p-p53 and Bax. OSSC may serve as an alternative therapy to retard the development of diabetic retinopathy.

Considerable evidence indicates that serotonergic mechanisms, particularly the serotonin transporter, are involved in alcoholism and tobacco use and are influenced by polymorphism of the promoter region of 5HTT (5-HTTLPR). As alcohol and tobacco consumption have been implicated in the pathogenesis of oral cancer, the purpose of this study was to investigate 5-HTTLPR polymorphism in patients with oral squamous cell carcinoma (OSCC) compared with a control group in a sample of Brazilian patients. One hundred and three patients affected by OSCC and 103 volunteers without OSCC were genotyped for 5-HTTLPR. Both groups were matched for age, sex and tobacco use. The chi-squared test was used for statistical analysis (α=0.05). There was no statistically significant difference in 5-HTTLPR genotypes between case and control group (p= 0.408). In conclusion, the present investigation demonstrated that serotonin transporter polymorphisms are not implicated in the OSSC development.

Abstract:
Overall prognosis for patients with oral squamous cell carcinomas (OSSC) is still unfavourable. However, there is a hope that a novel diagnostic method may establish better cancer biology characteristics. The aim of this study was to evaluate the isotope ratio of nitrogen and carbon in OSSC as compared to margin and healthy tissue. A total of 18 patients with OSSC were included in the study. Specimens collected covered: four tumour, four margin and two healthy oral mucosa samples. The samples underwent further procedures: lyophilization and isotope ratio mass spectrometry. Measurements of the ratio of stable isotopes of nitrogen 15N/14N and carbon 13C/12C were performed. It is noticeable that the highest average nitrogen concentration was observed in tumour 12 ± 0.4% and the lowest in healthy tissues 8 ± 0.9% (p < 0.00001). The highest average carbon content was observed in healthy tissues 57 ± 2.2% and the lowest in tumour 46 ± 1.3% (p < 0.00001). Moreover, values of 15N/14N expressed in delta notation were the highest in healthy tissues 9.84 ± 0.61 and the lowest in tumour 8.92 ± 0.58. Values of 13C/12C tended to be higher in tumour −22.2 ± 0.89 and the lowest in healthy tissues −23.7 ± 1.2. Tumour tissues differ in isotopic composition from tissues taken from margin and healthy tissues taken from distant oral mucosa.

Abstract:
Random Vector Functional-link (RVFL) networks, as a class of random learner models, have received careful attention from the neural network research community due to their advantages in obtaining fast learning algorithms and models, in which the hidden layer parameters are randomly generated and remain fixed during the training phase. However, its universal approximation ability may not be guaranteed if the random parameters are not properly selected in an appropriate range. Moreover, the resulting random learner’s generalization performance may seriously deteriorate once the RVFL network’s structure is not well-designed. Stochastic configuration (SC) algorithm, which incrementally constructs a universal approximator by obtaining random hidden parameters under a specified supervisory mechanism, instead of fixing the selection scope in advance and without any reference to training information, can effectively circumvent these awkward issues caused by randomness. This paper extends the SC algorithm to an online sequential version, termed as an OSSC algorithm, by means of recursive least square (RLS) technique, aiming to copy with modeling tasks where training observations are sequentially provided. Compared to the online sequential learning of RVFL networks (OS-RVFL in short), our proposed OSSC algorithm can avoid the awkward setting of certain unreasonable range for the random parameters, and can also successfully build a random learner with preferable learning and generalization capabilities. The experimental study has shown the effectiveness and advantages of our OSSC algorithm.

AbstractThere are research questions whose answers require record linkage of multiple databases that may be characterized by limited options for full data sharing. For this purpose, the Open Data Infrastructure for Social Science and Economic Innovations (ODISSEI) consortium has supported the development of the ODISSEI Secure Supercomputer (OSSC) platform that allows researchers to link cohort data to data from Statistics Netherlands and run large-scale analyses in a high-performance computing (HPC) environment. Here, we report a successful record linkage genomewide association (GWA) study on expenditure for total health, mental health, primary and hospital care, and medication. Record linkage for genotype data from 16,726 participants from the Netherlands Twin Register (NTR) with data from Statistics Netherlands was accomplished in the secure OSSC platform, followed by gene-based tests and estimation of total and single nucleotide polymorphism (SNP)-based heritability. The total heritability of expenditure ranged between 29.4% (SE 0.8) and 37.5% (SE 0.8), but GWA analyses did not identify SNPs or genes that were genomewide significantly associated with health care expenditure. SNP-based heritability was between 0.0% (SE 3.5) and 5.4% (SE 4.0) and was different from zero for mental health care and primary care expenditure. We conclude that successfully linking genotype data to administrative health care expenditure data from Statistics Netherlands is feasible and demonstrates a series of analyses on health care expenditure. The OSSC platform offers secure possibilities for analyzing linked data in large scale and realizing sample sizes required for GWA studies, providing invaluable opportunities to answer many new research questions.

Abstract:
Background: Oral pseudoepitheliomatous hyperplasia (PEH) appears histologically similar to oral squamous cell carcinoma (OSCC) in small oral biopsies, thus posing diagnostic dilemma. The objective of this study was to compare the expression of matrix metalloproteinase-9 (MMP-9) in differential diagnosis of OSSC and oral PEH. Materials & Methods: This comparative cross-sectional study was conducted in the Department of Histopathology, Armed Forces Institute of Pathology (AFIP), Rawalpindi, Pakistan from January 2013 to March 2016. It included sixty archival cases, 30 each of OSCC and PEH. Paraffin embedded blocks were prepared, hematoxylin and eosin stained sections taken and immunostained with MMP-9. The expression of MMP-9 was evaluated in OSCC and PEH. Results: The OSCC group included 16 (53.33%) men and 14 (46.67%) women, whereas PEH group included 18 (60%) men and 12 (40%) women. The mean age of OSCC group was 60.1±17.3 and that of PEH group was 52.7±16.6. In OSCC group, site of lesion was buccal mucosa in 12 (40%), gingiva 10 (33.33%), tongue 7 (23.33%) and floor of mouth 1 (3.34%) case. In PEH group, site of lesion was buccal mucosa in 12 (40%), tongue 11 (36.66%), gingiva 6 (20%) and palate 1 (3.34%) case. The expression of MMP-9 was positive in all the 30 cases of OSCC and negative in all 30 cases of PEH. Conclusion: Compared to pseudoepitheliomatous hyperplasia (PEH), MMP-9 revealed a higher expression in oral squamous cell carcinoma (OSCC). This finding has become mainstream strategy in distinguishing OSCC from PEH in oral mucosal biopsies in cases difficult to diagnose.

This study aims to evaluate the effect of 0.5% (5mg/ml) and 1% (10mg/ml) alendronate sodium (ALN) associated or not with lyophilized bovine bone (Bio-Oss®) on the repair of calvaria bone defects in rabbits. Two bony defects, 10mm diameter circular, were created in 40 male New Zealand white rabbits. The animals were randomized into five (n = 8) groups: GI = Bio-Oss®; GII = Bio-Oss® + ALN 0.5%; GIII = Bio-Oss® + ALN 1%; GIV = ALN 0. 5%; GV = ALN 1%. The left defect was filled with the corresponding substance of each experimental group and the right defect was filled with autogenous blood clot (control). All cavities were lined and covered with a biological membrane. The animals were euthanized 30 and 60 days after craniotomy. The percentage of new bone formation was assessed by microradiographic analysis of calvaria. The images obtained were analyzed using the public domain program Image J®, to assess quantitatively the percentage of bone repair. The values obtained were analyzed using the software SPSS® Statistics, using descriptive statistics (mean, standard deviation and confidence intervals) and inferential ("t" - Student and ANOVA/Tukey). The intragroup results showed that in GI, GII and GIII the percentage of bone repair was significantly higher in the experimental side compared to their respective controls; and was not statistically difference in GIV and GV groups, between experimental and controls sides. The intergroup results showed the following percentage of repair at 30 days: GI = 61.91% (± 10.61), GII = 80.39 (± 8.68), GIII = 81.60% (± 5.42), GIV = 48.45% (± 6.54), GV = 41.45% (± 8.98) and at 60 days: GI = 83.23% (± 14.43), GII = 98.72% (± 2.54), GIII = 92.66% (± 9.22), GIV = 50.18% ( ± 3.03), GV = 37.46% (± 6.26). It is concluded that the addition of alendronate in concentrations of 0.5 and 1%, to lyophilized bovine bone, accelerate the process of bone repair, being more significant in lower concentration. The use of ALN alone delayed the process of bone repair, but without statistical significance.
A presente pesquisa teve como objetivo avaliar o efeito do alendronato de sódio (ALN), nas concentrações de 0,5 (5mg/ml) e 1% (10mg/ml), associadas ou não ao osso bovino liofolizado (Bio-Oss®), na reparação de defeitos ósseos produzidos em calvária de coelhos. Foram utilizados 40 coelhos machos brancos da raça Nova Zelândia. Na calvária de cada animal, foram confeccionados dois defeitos medindo 10 mm de diâmetro. Os animais foram distribuídos aleatoriamente em cinco grupos (n=8 cada): GI = Bio-Oss®; GII = Bio-Oss®+ALN 0,5%; GIII = Bio-Oss®+ALN 1%; GIV = ALN 0,5%; GV = ALN 1%. A cavidade esquerda foi preenchida pela substância correspondente a cada grupo experimental e a cavidade direita preenchida por coágulo sanguíneo autógeno (controle de cada grupo). Todas as cavidades foram forradas e cobertas por membrana biológica. Os animais foram eutanasiados aos 30 e 60 dias após a craniotomia. O percentual de neoformação óssea foi avaliado por meio de análise radiomicrográfica da calvária. As imagens obtidas por radiomicrografia foram analisadas através do programa de domínio público Image J®, para avaliar quantitativamente o percentual de reparo ósseo. Os valores obtidos pelo Image J® foram submetidos à análise com auxílio do software SPSS® Statistics, por meio de estatística descritiva (média, desvio-padrão e intervalos de confiança) e inferencial (teste t Student e ANOVA/ Tukey). Na comparação intragrupos, os resultados demonstraram que nos grupos GI, GII e GIII o percentual de reparo foi significantemente mais elevado no lado experimental em relação aos respectivos controles; nos grupos GIV e GV não foram observadas diferenças estatisticamente significativas no percentual de reparo entre os lados experimental e controle. Na comparação intergrupos, os resultados mostraram os seguintes percentuais de reparo aos 30 dias: GI = 61,91% (± 10,61), GII = 80,39 (± 8,68), GIII = 81,60% (± 5,42), GIV = 48,45% (±± 6,54), GV = 41,45% (± 8,98). Aos 60 dias: GI = 83,23% (± 14,43), GII = 98,72% (± 2,54), GIII = 92,66% (± 9,22), GIV = 50,18% (± 3,03), GV = 37,46% (± 6,26). Conclui-se que a adição do ALN, nas concentrações de 0,5 e 1%, ao osso bovino liofilizado aplicado topicamente nos defeitos ósseos acelerou o processo de reparo ósseo, sendo mais significativo na menor concentração. O uso do ALN isoladamente retardou o processo de reparo ósseo, mas sem significância estatística.

Cientistas geram dados experimentais, e o interesse no compartilhamento destes dados é enorme, em especial na área biológica. É o compartilhamento que permite o trabalho cooperativo entre laboratórios espalhados pelo mundo. Este interesse gerou a formação de diversas bases de dados, como o GenBank e o Protein Data Bank. Cientistas de todo o mundo ali depositam seus dados, formando grandes repositórios globais que são usados para análise e recuperação de registros. Tais tarefas podem ser feitas de duas formas: utilizando serviços implantados nas bases centrais, como o BLAST para a recuperação de sequências homólogas, ou fazendo cópias locais das bases e implantando localmente novos algoritmos. Neste trabalho nós propomos a formação de um repositório global não real, mas sim virtual, através do uso de agentes móveis, e da fácil implantação por um usuário de funcionalidades adicionais para análise e recuperação de dados, utilizando carga dinâmica de classes Java.Nós aplicamos este modelo na construção de uma nova versão do AnaGel, um sistema para armazenamento e análise de registros eletroforéticos. A eletroforese é um processo de separação demacromoléculas biológicas de extrema utilidade para bioquímicos e geneticistas. Várias amostras contendo diferentes misturas de macromoléculas - geralmente fragmentos de DNA, RNA ou deproteínas, com a mesma carga elétrica - são colocadas em uma placa coberta por um gel, e submetidas a um campo elétrico, que força a migração das macromoléculas através do gel. Moléculas menoresencontram menor dificuldade para atravessar o gel, e seu deslocamento é consequentemente maior. O inverso acontece com as moléculas maiores. Um experimento de eletroforese gera então umaimagem, com várias canaletas, cada uma correspondendo a uma amostra, onde em cada canaleta há uma formação de bandas nos pontos de acúmulo de moléculas com o mesmo peso molecular.Em sua primeira versão o AnaGel já era uma ferramenta bastante completa para o tratamento de registros eletroforéticos, oferecendo a deteção automática de canaletas e bandas, a correção de distorções, a determinação de pesos moleculares, além da recuperação de registros por diversos métodos. A versão atual, toda construída em Java, apresenta como vantagens a instalação em um servidor, com o uso por navegadores web, a possibilidade de análises e recuperação de registros através de agentes que se movimentam por todos os repositórios deuma rede de laboratórios que desejam compartilhar dados, e a facilidade de adição de novas funcionalidades pelos usuários, que necessitam apenas de construir classes Java obedecendo a umaclasse abstrata exportada pelo AnaGel.Nesta dissertação nós apresentamos uma discussão sobre o uso da Web para o trabalho cooperativo entre cientistas experimentais. A solução técnica adotada - agentes móveis, com o sistema Grasshopper - é comparada com outras possibilidades para estruturação de sistemas distribuídos. Nós procuramos mostrar como o sistema AnaGel se compara favoravelmente com seus concorrentes comerciais. Diversos detalhes da implementação são explicados, e apontadas direções para melhorias futuras.

The absence of teeth leads to bone resorption, rehabilitate edentulous individuals through implants can be difficult when this is a severe resorption. The maxillary sinus lift is an alternative to allow the placement of implants in patients with extreme jaw bone resorption, particularly in the posterior maxillary. Objectives: Given the variety of grafts available for maxillary sinus lift, this systematic literature review aims to compare the use of Bio-Oss? with the use of autogenous bone with regard to implants survival rate. Methods: Through MeSH terms, entry terms and Boolean operators, a search for clinical and comparative studies of use of Bio-Oss? and use of autogenous bone for maxillary sinus lifting in PubMed, Embase and Cochrane Library. Results: The search initially found 709 articles that after excluding duplicates resulted in 672 jobs that had the title and summary read. 60 studies were read in full and 08 selected for analysis. The best implant survival rate found was 100% using isolated Bio-Oss?, using autogenous bone alone or a combination of autogenous bone with Bio-Oss?. The lower survival rate found was 82.4% for implants placed in the maxillary sinus filled with autogenous bone. There was no statistically significant difference between groups of the included studies. Conclusion: Still not possible to consider the Bio-Oss? as the gold standard for maxillary sinus floor augmentation, but we can suggest that, as the autologous bone, the biomaterial has excellent implant survival rates.

Objetivo: Esta revisão tem por objetivo saber como os diferentes enxertos influenciam na recuperação pós ligamentoplastia do ligamento cruzado anterior (LCA), na manutenção do nível de atividade desportiva e score funcional ao longo do tempo. Metodologia: Foi efetuada uma pesquisa computorizada nas bases de dados B-on, EBSCO, Medline, PEdro e Pubmed para identificar estudos clínicos randomizados que, após reconstrução ligamentar do LCA, comparassem os efeitos ao fim de pelo menos dois anos dos enxertos do tendão rotuliano e semitendinoso/gracilis, com publicações entre 2004 e 2014. Resultados: Nesta revisão foram incluídos 5 estudos clínicos randomizados envolvendo 344 indivíduos com a classificação metodológica de 9 em 10 na escala de PEDro, que compararam os diferentes enxertos com programas de reabilitação e instrumentos de avaliação. Conclusão: Constatou-se que a reconstrução anatómica artroscópica do LCA com enxertos de osso-tendão-osso (OTO) em comparação com semitendinoso e gracílis (ST-G) são ambas técnicas seguras e efetivas. Ambos enxertos mostrarem resultados bastantes similares em termos de avaliação subjetiva quantificada com diferentes escalas.
Objective: This review aims to find out how the different grafts influence the recovery after ligamento plasty of anterior cruciate ligament, in maintaining the level of sporting activity and functional score over the time. Methodology: computerized search was performed in the databases B -on, EBSCO, Medline, Pubmed and PEdro to identify randomized clinical trials that made the comparison the effects at least two years after ACL reconstructing of the patellar tendon and semitendinosus / gracilis grafts, , with publications between 2004 and 2014 . Results: this review include 5 randomized clinical trials involving 344 individuals with methodological classification of 9 in 10 in the PEDro scale. that compared the different grafts with rehabilitation programs and instruments evaluation. Conclusion: It was found that arthroscopic anatomic ACL reconstruction with grafts bone-tendon-bone (OTO) compared with semitendinosus / gracilis (ST-G) are both safe and effective techniques. Both grafts show quite similar results in terms of subjective assessment quantified with different scales.

Syftet med vårt examensarbete är att undersöka vilken geografisk omvärldsbild elever har, dvs. vilken inre karta de har av världen, och hur den formas. Vi har använt oss av en metod bestående av en enkät med både allmänna frågor om elevens erfarenheter samt tre kunskapsbaserade uppgifter om kartan. Den första av dessa tre är en öppen uppgift där eleven ombeds rita världskartan på ett blankt papper. Det görs sedan kopplingar mellan elevernas erfarenheter, kunskaper och den mentala kartan. I resultatet såg vi att eleverna har brister i sin geografiska omvärldsbild och kunskaper om var olika platser ligger i Sverige och världen. Tidigare forskning i ämnet visar att många av våra hypoteser och slutsatser kan bekräftas. Till exempel hade eleverna en geocentrisk och etnocentrisk världsbild. Det märktes tydligt att eleverna formade sin geografiska omvärldsbild utifrån tidigare erfarenheter och attityder till olika platser. Eleverna i vår undersökning har en begränsad geografisk omvärldsbild och behöver därför arbeta mer med att utveckla den.

Abstract:
Para a execução da etapa inicial da identificação médico-legal de restos humanos (antropometria e exame dos arcos dentários), faz-se necessária uma limpeza prévia da ossada, para a remoção de tecidos moles putrefeitos. Os casos não identificados por esses métodos tradicionais, poderão ser submetidos ao exame de DNA. No entanto, apesar do grande avanço da biologia molecular, utilizando a amplificação de DNA pela PCR, algumas limitações que afetam a habilidade de se obter DNA em restos humanos, permanecem. Dessa forma, o objetivo deste trabalho foi fornecer subsídios morfológicos para os analistas forenses, com ênfase na prática médico-legal, visando uma utilização mais eficiente do DNA obtido de osso compacto de restos humanos em decomposição ou já esqueletizados, sem tecidos moles aderidos. Foi realizado o estudo da microestrutura do tecido ósseo compacto femoral, de restos humanos em decomposição, ainda com tecidos moles, que foram limpos pela fervura em água (n = 7) e ossadas já esqueletizadas pela decomposição natural, que não foram fervidas (n = 8). Destes, seis ossadas foram provenientes de cemitério público regular, após 3 anos de inumação, 1 ossada proveniente da região amazônica, e 1 ossada de origem desconhecida. Estas duas ultimas, apresentado-se porosas ou quebradiças. As análises morfológicas de cortes histológicos foram coradas com hematoxilina e eosina e o DNA amplificado pela PCR para os loci CSF1PO, TPOX, TH01, F13A0, FESFPS, vWA, D16S539, D7S820, D13S317 e amelogenina. Os resultados da análise desses dois grupos foram comparados com os de cadáveres frescos (n = 5) do Serviço de Verificação de Óbitos da Capital. A fervura dos ossos, do modo como é realizada no Instituto Médico Legal de São Paulo, pode aumentar a eosinofilia da matriz óssea e, em alguns casos, pode promover a desagregação dos ósteons. Tal procedimento pode remover células, mas pode também remover possíveis inibidores da PCR, favorecendo a análise do DNA obtido destas amostras. O fator limitante para a obtenção e análise de DNA, em amostras de ossos limpos por fervura, é a quantidade exígua de células. Ossos não submetidos à fervura, após inumação por três anos ou há mais tempo em contato com a terra, podem apresentar alterações da microestrutura. No entanto, a presença de hemácias preservadas e núcleos de osteócitos nestas amostras, indica melhor preservação de células em relação às amostras de ossos fervidos. O fator limitante para a análise de DNA nestas amostras é a presença sugestiva de inibidores da reação de amplificação pela PCR. Restos humanos, sem tecidos moles, macroscópicamente não preservados (porosos e quebradiços), e não submetidos à fervura, apresentam alterações de perda de matriz mineralizada; no entanto, nestas amostras ainda é possível encontrar células preservadas. Os resultados obtidos no neste trabalho permitem traçar algumas estratégias para uma melhor utilização nos protocolos de extração e análise do DNA em osso compacto de restos humanos.
To the first essential step to forensic identification of human remains (anthropological study of race, sex, age, etc) it is necessary a previous cleaning of the bones, to remove decomposing soft tissues. Medico-legal inconclusive or non identified cases, by using these traditional methods, could be subjected to DNA analysis. However, in spite of advances in human identification techniques, specially by PCR amplified DNA, some limitations that affect the ability to obtain DNA in human remains still persist. Therefore, the aim of this study was to provide additional support from morphological analysis, to help forensic analysts personnel to utilise more efficiently the DNA, extracted from compact bones of human remains in decomposition or already skeletonized corpse, it means without soft tissues, with special emphasis in the legal-medicine practice. Femoral compact bones were obtained from: 7 human remains found on the ground, in different degree of decomposition which were cleaned by boiling to remove soft tissues; also studied were collections of bones from 8 corpses having undergone natural decomposition: 6 human remains exhumed after 3 years from a common public cemetery in São Paulo City; 1 case from amazon region and 1 case with no information, both cases remained from long time (more than 3 years) in contact with soil. All eight cases, were not boiled as no soft tissue were adhered. As a control, five cadavers 12 to 16 hours post mortem were also used. The compact bones histological sections were stained by haematoxilin and eosin and the loci CSF1PO, TPOX, TH01, F13A01,FESFPS, vWA, D16S539, D7S820, D13S317 and amelogenin were amplified by PCR.The procedure for boiling the human remains utilised in the Legal Medicine Institute of São Paulo would have increased the eosinophily of bone matrix and, in some cases, promoted the desaggregation of the osteons. In addition these procedures would have removed the cells, but in some cases would have removed possible inhibitors of the PCR, favouring in this way the analysis of DNA obtained from these samples. The limiting factor to obtain successful analysis in bones submitted to boiling seem to be the low quantity of nuclei present in these samples. For the other hand, in bones not cleaned by boiling, the presence of preserved red cells and oscteocyte nuclei inside the lacunae indicates better preservation of cells in relation to those bones cleaned by boiling. The limiting factor to obtain successful DNA analysis in bones exhumed or in contact of soil, is the suggestive presence of inhibitors of PCR. Porous and brittle bones from human remains, without soft tissues that are not processed by boiling, present alterations through loss of mineralised matrix, although it is still possible to found preserved cells in these samples. The results presented in this work clarify concerns about viability of DNA for identification analysis. They also help to establish better strategies for optimisation of DNA extraction and analysis in compact bones of human remains.

Cílem této práce je poskytnout snadný způsob, jak rozšířit analyzační nástroj o rozhraní splňující standard OSLC. Takové rozhraní umožňuje jednoduchou integraci nástrojů s jinými nástroji nebo systémy, umožňuje jejich vzdálené použití skrze webové služby a umožňuje je jednoduše propojit s databází pro databázové dotazy a pro perzistentní uložení dat. Toto je dosaženo návrhem a implementací OSLC adaptéru pomocí sady nástrojů Eclipse Lyo. Adaptér používá jako rozhraní doménu OSLC Automation a je dostatečně univerzální na to, aby skrze toto rozhraní pokryl funkcionalitu většiny analyzačních nástrojů za pomocí jejich stávajících rozhraní na příkazové řádce. Tato práce poskytuje úvod k OSLC, Eclipse Lyo a souvisejícím konceptům. Dále tato práce definuje požadavky a odlišnosti různých analyzačních nástrojů a diskutuje návrh adaptéru a faktory, které ovlivnily návrhová rozhodnutí. A nakonec prezentuje implementovaný adaptér a jeho vyhodnocení pomocí automatizované testovací sady a pomocí experimentů s řadou analyzačních nástrojů. Nejvýznamnější ukazatel hodnocení vytvořeného adaptéru je to, že už teď je používán v praxi pro přidání OSLC rozhraní k nástrojům ANaConDA, Perun, Spectra (všechny tři vyvíjené na VeriFIT) a HiLiTE (Honeywell).

O alcoolismo é considerado uma doença crônica, trazendo prejuízos para saúde do indivíduo. O tecido ósseo também sofre sérios danos com o consumo crônico do álcool, pois induz defeitos na mineralização e redução da espessura do osso medular e cortical. Devido à grande procura por tratamentos reconstrutivos para lesões e perdas ósseas, os enxertos xenógenos vêm sendo uma boa opção para o tratamento regenerativo. Em vista da procura de pacientes que fazem consumo crônico do álcool e apresentam perdas ósseas, necessitando de enxertos xenógenos, este trabalho teve como objetivo comparar, por meio de análise histomorfológica e histomorfométrica, a ação de dois biomateriais, o Bio-Oss® e o OrthoGen®, no processo de reparo de defeitos ósseos em tíbia de ratos submetidos ou não ao alcoolismo experimental. Foram utilizados 80 ratos machos (Rattus norvegicus) da linhagem Wistar, com 60 dias de idade, separados aleatoriamente em quatro grupos experimentais contendo 20 animais cada: Grupo Água Bio-Oss® (GAB), Grupo Etanol Bio-Oss® (GEB), Grupo Água OrthoGen® (GAO) e Grupo Etanol OrthoGen® (GEO). Os animais dos grupos GEB e GEO foram submetidos à adaptação gradativa ao álcool e depois permaneceram a 25% por 90 dias. Após esse período, todos os animais foram submetidos à cirurgia experimental. Foi realizada uma cavidade cirúrgica de aproximadamente 3 mm de diâmetro na epífise proximal da tíbia e nos animais dos grupos experimentais GAB e GEB se fez o preenchimento com o biomaterial Bio-Oss®. Os animais dos grupos experimentais GAO e GEO receberam o preenchimento da cavidade com o biomaterial OrthoGen®. Decorridos os períodos de 10, 20, 40 e 60 dias pós-cirúrgico, cinco animais de cada grupo, por período, foram eutanasiados. Os resultados histomorfológicos demostraram que os animais dos grupos GAB e GEB apresentaram células sanguíneas e tecido conjuntivo nos períodos de 10 e 20 dias, e nos períodos de 40 e 60 dias ocorreu a formação de novo osso, sendo que o grupo GEB apresentou um maior retardo na formação quando comparado ao GAB. Nos grupos GAO e GEO o início da formação de tecido ósseo ocorreu nos períodos de 40 e 60 dias, com um retardo na formação quando comparado ao biomaterial Bio-Oss®. Na análise histomorfométrica, comparando os grupos que receberam a dieta líquida não alcóolica com biomaterias diferentes (GAB e GAO) ocorreu diferença significante em formação óssea em todos os períodos sendo que o grupo Água Bio-Oss® apresentou uma maior formação óssea. Nos grupos que receberam a dieta alcóolica, com biomateriais diferentes (GEB e GEO), ocorreu diferença em formação óssea nos períodos de 20, 40 e 60 dias, onde o biomaterial Bio-Oss® apresentou maiores médias. Comparando os grupos que receberam o mesmo biomaterial, mas com dieta líquida diferente (GABXGEB e GAOXGEO), evidenciou-se diferença significante em formação óssea em todos os períodos (exceto GAOXGEO; 10 dias), sendo que os grupos que receberam dieta líquida apenas com água apresentaram melhores resultados. Conclui-se que o biomaterial Bio-Oss® mostrou-se superior na neoformação óssea quando comparado ao Ortoghen®, e que a dieta alcoólica interferiu de forma negativa no processo de reparação.
Alcoholism is considered a chronic disease, which brings harm to an individuals health. Bones also suffer serious damage resulting from the chronic consumption of alcohol, since it induces defects in the mineralization and reduction of the thickness of the cortical and the cancellous bone. Xenografts have been a good option for regenerative treatments in response to the high search for reconstructive treatments for bone losses and lesions. Given the demand of patients who make chronic use of alcohol and present bone loss, needing xenografts, the aim of this study was to compare, by means of a histomorphological and histomorphometric analysis, the action of the biomaterials Bio-Oss® and OrthoGen® in the repair process of bone defects in the tibia of rats submitted or not to experimental alcoholism. Eighty male Wistar rats (Rattus norvegicus) aged 60 days were randomly separated into four experimental groups with 20 animals each: Bio-Oss® Water Group (GAB), Bio-Oss® Ethanol Group (GEB), OrthoGen® Water Group (GAO) and OrthoGen® Ethanol Group (GEO). The animals in the GEB and GEO were submitted to gradual adaptation to alcohol and later kept at 25% for 90 days. After this period, all animals were submitted to experimental surgery. A surgical cavity with approximately 3 mm of diameter was made in the proximal tibial epiphysis and in animals from the experimental groups GAB and GEB it was filled with the Bio-Oss® biomaterial. Animals from the experimental groups GAO and GEO had their cavity filled with the OrthoGen® biomaterial. After 10, 20, 40 and 60 days post-surgery, five animals in each group, per period, were euthanized. The histomorphological results showed that the animals from the GAB and GEB groups presented blood cells and connective tissue in the 10 and 20 days periods, and in the periods of 40 and 60 days there was formation of new bone, with the GEB group presenting greater delay in formation when compared to the GAB. In the GAO and GEO groups, bone formation started in the periods of 40 and 60 days, with a delay in formation when compared to the Bio-Oss® biomaterial. In the histomorphometric analysis, comparing the groups that received nonalcoholic liquid diet with different biomaterials (GAB and GAO), there was a significant difference in bone formation in all periods, with the Bio-Oss® Water Group presenting greater bone formation. In the groups that received an alcoholic diet, with different biomaterials (GEB and GEO), there was a difference in bone formation in the periods of 20, 40 and 60 days, in which the Bio-Oss® biomaterial presented greater means. Comparing the groups that received the same biomaterial, but with a different liquid diet (GABXGEB and GAOXGEO), there was a significant difference in bone formation in all periods (except for GAOXGEO; 10 days), in which the groups that received a liquid diet with only water presented better results. In conclusion, the Bio-Oss® biomaterial presented better performance in bone neoformation when compared to the Ortoghen®, and the alcoholic diet affected negatively the repair process.

Os glicocorticóides são farmacos amplamente utilizados em medicina veterinária, seu emprego pode desencadear uma série de efeitos adversos. O ensaio clínico realizado teve como objetivo a avaliação das possíveis alterações na densidade mineral óssea após a terapia com prednisona, utilizando a tomografia computadorizada helicoidal. No estudo, foram utilizados oito cães hígidos sem raça definida, adulto-jovens, sendo 4 machos e 4 fêmeas. A prednisona foi administrada por via oral aos animais na dose diária de 2mg kg-1 de peso durante 30 dias. A densidade mineral óssea foi determinada a partir da obtenção de valores de radiodensidade da região de osso cortical e osso trabecular do corpo vertebral da segunda vértebra lombar, imediatamente após o período da administração do medicamento. O protocolo experimental permitiu a caracterização significativa (P<0,05) diminuição da radiodensidade do osso trabecular do corpo vertebral da segunda vértebra lombar, entretanto não se constatou desmineralização significativa na região cortical. Nenhum dos cães do grupo experimental apresentou fratura patológica ao término da administração do medicamento.
