Abstract:
The thesis is focused on the comprehensive solution of the flood protection measures of the town Oslava on the river Oslava. The territory was subjected to a risk analysis with a graphical representation of risk in the area and potential damage to the property was calculated. In continuity has been proposed appropriate flood protection measure to reduce damage. The economic effectiveness of the measures was carried out and other documentation required for the underlimit tender of the project was elaborated. In the conclusion of the diploma thesis there is another possible use of the diploma thesis.

O gênero Sadocus Sørensen, 1886 é revisado pela primeira vez e uma análise cladística foi feita para testar seu monofiletismo e suas relações internas, além disso, a chave de identificação das espécies é apresentada. Uma nova classificação é proposta baseada nos resultados da análise cladística. São reconhecidas cinco espécies válidas das 5 espécies válidas das 14 nominais, sendo quatro sinonímias propostas: Sadocus bicornis (Gervais, 1849) em Sadocus asperatus (Gervais, 1847), S. conspicillatus Roewer, 1913, S. exceptionalis (Mello-Leitão, 1946) e S. guttatus Sørensen, 1902 em S. polyacanthus (Gervais, 1847) e S. calcar (Roewer, 1913) em Gonyleptes horridus Kirby, 1819; duas Species inquerendae: S.allermayeri (Mello-Leitão, 1945) e S. nigronotatus (Mello-Leitão, 1943) e duas transferidas para outros gêneros: Roeweria brasiliensis (Soares & Soares, 1949) e Metagyndes planiceps (Guérin-Méneville, 1838). Além é feita a descrição de Metagyndes sp.n., exemplar usado por Roewer na redescrição da espécie Metagyndes planiceps (Guérin-Méneville, 1838), que se tratava de uma espécie diferente da descrição original. A matriz de dados utilizada na análise cladística possui cinco terminais de Sadocus e mais 15 terminais como grupo externo, compreende 53 caracteres baseados na morfologia masculina, sendo 18 pertencentes a estruturas morfológicas externas presentes no escudo dorsal e tergitos livres, dois pertencentes a quelícera, dois pertencente ao pedipalpo, 20 pertencentes as pernas e 11 pertencentes a genitália. A análise resultou em uma árvore mais parcimoniosa com 195 passos (C.I.= 32; R.I.=45) cuja otimização aplicada foi ACCTRAN. Sadocus constitui um clado monofilético e pectinado, sustentado por duas sinapomorfias: formato gama triangular do escudo dorsal e bula da quelícera lisa
The genus Sadocus Sørensen, 1886 is revised for the first time and a cladistic analysis was performed to test its monophyly and internal relationships, a dicotomous key of species was presented. A new classification based on cladistic analysis is proposed. Five species are considered valid of the 14 nominal species currently recognized, being synonymyzed are, Sadocus bicornis (Gervais, 1849) as Sadocus asperatus (Gervais, 1847), S. conspicillatus Roewer, 1913, S. exceptionalis (Mello-Leitão, 1946) and S. guttatus Sørensen, 1902 as S. polyacanthus (Gervais, 1847), and S. calcar (Roewer, 1913) as Gonyleptes horridus Kirby, 1819; two Species inquerendae: S. allermayeri (Mello-Leitão, 1945) and S. nigronotatus (Mello-Leitão, 1943), and two transferred to other genus: Roeweria brasiliensis (Soares & Soares, 1949) and Metagyndes planiceps (Guérin-Méneville, 1838). The description of Metagyndes sp.n. is made, a specimen that Roewer used for the redescription of the species Metagyndes planiceps (Guérin-Méneville, 1838), it was a different example of the original description. The cladistic analysis was performed with five species of Sadocus and the outgroup is composed of fifteen species. The character matrix comprises 53 morphological characters from males, 18 characters are of external morphological features of the dorsal scute and free tergites, two of the chelicerae, two of the pedipalp, 20 of the legs and 11 of the genitalia. The analysis resulted in one most parsimonious tree with 195 steps (C.I.= 32; R.I=45) and the ACCTRAN optimization was applied. Abstract:
A eletroquímica direta de enzimas redox depende da distância entre os sítios redox da proteína e a superfície do eletrodo e também da eficiência na imobilização dessas enzimas na superfície eletródica. Dessa forma, a obtenção de enzimas mais hidrofóbicas possibilita a melhora na interação entre elas e a superfície de eletrodos sólidos, como os de carbono. Neste estudo, foi desenvolvida uma rota para a obtenção da glicose oxidase oligomerizada (Ol-GOx) com o objetivo de melhorar a interação entre a enzima e a superfície de fibras de carbono, uma vez que enzimas oligomerizadas contêm suas porções hidrofóbicas expostas.Para tanto, diferentes ácidos de Brønsted foram utilizados, sendo que a enzima obtida a partir da reação com o ácido trifluorometanosulfônico (TFMS) foi a que se manteve ativa cataliticamente. A Ol-GOx se mostrou um biocatalisador promissor devido a sua hidrofobicidade e seu tamanho, os quais permitiram uma imobilização mais eficiente em superfícies de carbono. Após a caracterização estrutural, concluiu-se que a Ol-GOx é formada por um oligômero composto por 10 unidades de GOx nativa com raio hidrodinâmico de aproximadamente 96 nm. Por voltametria cíclica estudou-se a transferência direta de elétrons (TDE) entre o cofator dinucleotídeo de flavina e adenina (FAD) e a superfície das fibras de carbono, sendo observado um aumento de 7 vezes nas correntes faradaicas em relação ao obtido para a GOx nativa. Além disso, as propriedades bioeletrocatalíticas foram melhoradas em 30% quando analisada a oxidação da glicose. Concluiu-se ainda que quanto maior a quantidade de folhas-β presente na estrutura proteica, maior a TDE observada entre a enzima e a superfície das fibras de carbono.
The direct electrochemistry of redox enzymes is dependent on the distance between the active centers of the protein and the electrode surface, and also on the efficiency in the immobilization of these enzymes on the electrodic surface. Thus, the synthesis of more hydrophobic enzymes could lead to better interaction between the redox enzymes and the solid electrode surfaces, such as carbon electrodes. In this study, it was proposed a chemical route to obtain oligomerized glucose oxidase (Ol-GOx), aiming to improve the interaction between the enzyme and the surface of carbon fibers, since oligomerized proteins have their hydrophobic chains exposed. After structural characterization, it was concluded that Ol-GOx is formed by 10 dimeric units of native GOx with a hydrodynamic radius corresponding to approximately 96 nm. By cyclic voltammetry, it was studied the direct electron transfer (DET) between the flavin adenine dinucleotide (FAD) cofactor and the surface of carbon fibers, where it was observed an increase of 7-fold in the faradaic currents in comparison to that observed for native GOx. Besides, bioelectrocatalytic properties are 30% improved, when analyzed the glucose oxidation by cyclic voltammetry. It was also concluded that the greater the β-sheet content in protein structure, the higher the DET observed between the enzyme and the carbon fibers surface.

Abstract:
This diploma thesis deals with a design of the reconstruction of a smixed weir on the Oslava river in Oslavany town. The introduction describes the site of interest and the the occurrence of ice events. There are several types of fish ladders described theoretically. Further on in the thesis there is desribed the state of the objects on the flow and the state of the river basin in the area of interest of Oslava river. Next part of the thesis deals with the design of a movable baying structure (a hollow flap) and fish ladder type bypass channel. An impact assessment of the current and newly proposed weir on flow in the HEC-RAS program was carried out. The project also includes the basin adjustment in overweir and an evaluation of the stability of the newly designed construction.

Abstract:
The aim of the thesis is to create a business plan, focusing on organizing children's birthday parties, social events and corporate parties, on the basis of which will be assessed the feasibility and economic advantage of the business plan. Thematically, the plan is divided into two parts. The first part covers the theoretical background, the definition of the business plan, the structure and principles of treatment, of which specific form is located in the following practical part. It contains a brief introduction, the company's mission, vision and objectives, description of business opportunities, defining the target and the total market including analysis of competition and marketing strategy. All previous points summarize the numerical form of the financial plan in three variants of development, the essence of which is to objectively assess the financial burden of the project. At the end of the thesis is the SWOT analysis, the potential risks of the project including the design of countermeasures to eliminate these effects. The whole thesis is finished with conclusion and evaluation of the business plan.

Abstract:
The main aim of the thesis is an appraisal of the stream Oslava which belongs to the Náměšť nad Oslavou region with the focus on flood control of the town. The thesis asseses current state of the stream with respect to the state of riverbed, bulwark of the stream and evaluation of the capacity of the stream channel. Calculation water surface profiles have been made by usage of the 1D mathematical model HEC-RAS for selected N-year flows. The capacity of flows and objects in the given period has been detected. There is also a map of the water runoffs included and possible solutions of how to protect the area surrounding the stream are proposed. The thesis was complemented with hydroecological monitoring of the stream channel - the HEM analysis 2014.

L'objectif de cette thèse vise à mettre au point et à améliorer les méthodes de mesures d'échauffements nucléaires auprès des maquettes critiques du CEA-Cadarache EOLE et MINERVE, au moyen de détecteurs thermo-luminescents (TLD), de détecteurs à luminescence optiquement stimulée (OSLD – nouvellement mis en œuvre dans le cadre de ce travail de thèse) et d'une chambre d'ionisation. Il s'agit d'identifier, de hiérarchiser, de traiter et enfin de réduire les différentes sources d'incertitudes et de biais systématiques associés à la mesure.Une série d'expériences a été mise en place dans le réacteur MINERVE. Les mesures ont été réalisées dans un environnement en aluminium ou en hafnium à l'aide d'un nouveau protocole : les TLD ont été étalonnés individuellement, la répétabilité de la mesure a été évaluée expérimentalement et les lois de chauffe des TLD ont été optimisées, conduisant à une réduction des incertitudes de mesures. Des mesures de gammas émis de façon différée après arrêt du réacteur MINERVE ont également été réalisées : les résultats obtenus montrent un bon accord des mesures avec les trois types de détecteurs utilisés.L'interprétation de ces mesures nécessite des calculs pour tenir compte des facteurs de correction, liés à l'environnement et au type de détecteurs utilisés. Ainsi, des corrections de la contribution des neutrons à la dose totale intégrée par les détecteurs ont été évaluées à l'aide de deux méthodes de calcul. Ces corrections ont été obtenues sur la base de simulations Monte Carlo couplées neutron-gamma et gamma-électron à l'aide du code MCNP
The objective of this thesis is to develop and to improve the nuclear heating measurement methods in MINERVE and EOLE experimental reactors at CEA-Cadarache, using thermo-luminescent detectors (TLD), optically stimulated luminescence detectors (OSLD – newly implemented in the context of this thesis) and an ionization chamber. It is to identify, prioritize, treat and reduce the various sources of uncertainty and systematic bias associated with the measurement.A series of experiments was set up in the MINERVE reactor. The measurements were carried out in an aluminum or hafnium surrounding using a new procedure methodology. The TLD are calibrated individually, the repeatability of the measurement is experimentally evaluated and the laws of TLD heat are optimized. The measurements of the gamma emitted, with a delay (delayed gamma) after shutdown of the MINERVE reactor, were also carried out using TLD and OSLD detectors with the aluminum pillbox as well as by ionization chamber. The results show a good correlation between the measurements recorded by these three detectors.The interpretation of these measurements needs to take account the calculation of cavity correction factors related to the surrounding and the type of detector used. Similarly, the correction due to the neutrons contributions to the total dose integrated by the detectors are evaluated with two calculation methods. These corrections are based on Monte Carlo simulations of neutron-gamma and gamma-electron transport coupled particles using the MCNP

Abstract:
Public diplomacy has become an integral part of international relations in the recent years. Effective communication is the key in solving international conflicts and in strengthening the partnership between nations. Building an excellent reputation of the individual country among the foreign community has no less importance. This master thesis deals with the promotion of Czech culture as an important part of public diplomacy in the US, particularly in New York, partly in Washington, DC. First and second chapter provide a theoretical framework for the following issues. Basic concept of public diplomacy is clarified there. The third chapter then presents specific players and partners whose cooperation the author deals in the fourth and fifth chapter. The fourth chapter discusses in detail the Film Festival Panorama Europe, Europe's largest film festival in New York and tries to map its promotion. This event is significant in that it is an original project of the Czech Centre New York. The Film Festival currently has a lot of partners and is held under the auspices of EUNIC. The fifth chapter presents a unique project, which has been involved in over twenty-five different players. Main theme was the freedom celebration, celebration of the 25th anniversary of Velvet Revolution. The main objective of this thesis is to analyze how players contribute in building the reputation of the Czech Republic in the USA and how they promote their activities. Everything is based on case studies.

The theme of this diploma thesis is the analysis of the legal status of Palestine. The theoretical part of this thesis examines the history of Palestine, the relations between Palestine and Israel, the rejection of United Nations General Assembly Resolution 181 by Arab states, Palestinian membership in international organisations, the Oslo I Accord and the Montevideo criteria for statehood.The thesis also evaluates the final status negotiations between Israel and Palestine and various Palestinian government programs. The second part of the thesis analyses the constraints posed by divided Palestinian territories and the dynamics and interactions between Palestinian populations living in and outside the territory. Furthermore this diploma thesis focuses on the Palestinian public sector; especially on the areas of justice, education, health system and defense. Individual chapters also deal with the economic instability of Palestine and its financial dependence on the other, mostly European, countries. In conclusion, this diploma thesis will give not only a comprehensive legal view of the status of Palestine, but also views on Palestinian governance.

In the year 1896 Hungary celebrated the 1000th anniversary of the Magyar settlement in the Carpathian Basin. The preparations started in the early 80's of 19th century. Originally they planned to celebrate in 1895, since it was decided that the conquest had happened in 895. But there was not enough time to prepare, so they postponed the festivities. Even though there was widespread development in industry and culture at the end of 19th century, the situation in Hungary was very complicated. The main problem was the relationship of Hungarians within the Austro-Hungarian monarchy, changes in that society, and the situation of national minorities and the working-class. During the celebrations of the millennial anniversary, these discussed topics were overlooked while everybody was occupied with the festivities. Hungarians tried to present their glorious past, their country's successful economy, industry, and culture to prove that Hungary was the foremost nation in the Carpathian Basin. The Hungarian parliament passed a law to celebrate the millennium that ordered, for example, the building of monuments around Hungary and the founding of The Museum of Fine Arts and 400 elementary schools. Throughout the country there were opening ceremonies, festivities, meetings, processions, parades, and unveilings...

The aim of this work is to introduce Spanish corrida de toros in a broader cultural context and to underline its importance not only for Spain, but for the whole of modern society as well. Whether this brutal tradition is justifiable in the 21st century or an anachronism that can no longer be tolerated has been widely debated; I have reached the conclusion that it is not only ajustifiable tradition but also a meaningful tradition as well. Corrida de toros with all its lack of restraint, spontaneity and genuineness represents the last ritual of western culture. It is a ritual of collective ecstasy that creates an antipole to an overly technological and excessively rational society. As a matter of fact, it even presents an ideal model for the relationship between man and animal, which is expressed by respect and honor. In modern society, such a model has disappeared. Either the animal is reduced to nothing but a thing and kept in cruel conditions or it is ridiculously put on a pedestal and becomes a member of the family. Historically a variety of people have been attracted by corrida de toros. They were and are fascinated by its sincerity and rawness, while at the same time they are intrigued by its mysteriousness. The subject of this M.A. thesis is magtaal (or mongolian praise song) in the context of mongolian traditional oral folklore a the main focus of the thesis lies in its linguistic specifics and its role in mongolian rural environment. The thesis is is solved in two main parts. First one provides a general characteristics of this genre with emphasis on its ethnolinguistic, sociolinguistic and performance aspects. Further division to subgenres is also provided. The second part deals with each particular subgenre based on an analysis built around translation of parts of praise songs and referential commentaries. The main contribution of this thesis lies in a more detailed view on this genre from a different perspective rather than a general folkloristic view. In this way praise song is displayed as genre that is transcending above a simple form of a song but becomes a mean of communication and a specific manner of construction and perception of mongolian image of the world.

Petra Ponocná Abstract This work deals with perceptions of The Day of the Dead feast in the urban society of Mexico City in the context of official ideology of Mexican cultural nationalism. Dissertation was founded on fieldwork in Mexico City in two different locations, namely delegación Cuauhtémoc and delegación Xochimilco. I focus on the perceptions of the feast by research partners and I try to point out the process of self-identification with the locality and its relation to attitude formation to The Day of the Dead and national identity. I investigate the role of the state and the city in organizing the holiday and research partners reactions to these interventions. The study deals with a description of vegetation of part of catchment area of Oslava river between villages Velke Mezirici and Vanec. The search had been realised during the vegetation seasons of the years 2006 and 2007. There are characterised geological-petrographical, soil, climatic, hydrographic ratios and basic phytogeographical characteristics of this area in the form of a search. Selected genera of the observed plants was characterised and their incidence is figured in the schematic maps of the area. There were found out 327 kinds of plants there. In the annex there are attached schematic maps with places of occurence of the plant genera and photographic documentation.

The aim of this thesis is to analyze the relationships between allied nations Czechoslovakia and Yugoslavia in years 1918 and 1938 from the perspective of selected commemorative acts. The main aim of this thesis is to define the role of the 48th Infantry Regiment in these celebrations. This Thesis is based on the hypothesis that through its honorary title "Jugoslavia" was created symbolic function of the regiment, which was strengthened mutual friendly relationships between Czechoslovakia and the Kingdom of SHS. This diploma thesis is concerned with a research of slope sediments in the selected areas along rivers Jihlava, Oslava a Chvojnice. Specifically, the research of the boulder screes. There are scree slopes which are composed of homogeneous fragmentary rocks of the same shape and size. Their current form originated in the Quaternary, when their development was influenced by alternating glacial ages and interglacials, and is still in progress. The boulder screes can be found scattered throughout the Czech Republic and at some locations they might be created by different rocks. They are part of the natural ecosystem and the awareness of their origins, development and characteristics can help with future planning of nature conservation and a subsequent landscape care proposal.

Thermoluminescent dosimeters(TLD) have been widely used in personal, environmental and medical dosimetry for several decades. Owing to the progress of crystalized techniques, a new luminescent material of aluminum oxide doped with carbon (Al2O3:C) was developed successfully by LandauerInc. in 1996. The optically stimulated luminescent dosimeter(OSLD)possesses several advantages such as stimulated by light, fast readout, wide dose range of detectable, high sensitivity and re-estimation of radiation. This research studied and compared the radiation detection characteristics of InLight nanoDotTM OSLD and TLD-LiF:Mg.Cu,P (TLD-100H). The radiation characteristics studied in this research include batch variance, reproducibility, linearity of radiation dose and stimulated light, photon energy dependence, angular dependence and fading effect etc.. The results of this researchrevealed that OSLD possesses better radiation characteristics in batch variance. The response reproducibility of OSLD is better than that of TLD-100H. The linearity of both OSLD and TLD-100H to radiation dose is excellent. The photon energy dependence of OSLD is higher than that of TLD-100H. OSLD has better angular dependence than that of TLD-100H. The fading of OSLD and TLD-100H are insignificant within 30 days. This study shows that OSLD possessed excellent radiation detection characteristics as compared with that of TLD-100H. OSLD will be one of the most attractive radiation dosimeter in the application of personal, environmental and medical radiation measurement.

The subject of this thesis is the study of cryptographic hash functions, which utilize block ciphers as underlying chain functions. It is mainly concerned with the analysis of the three hash algorithms, the Whirlpool, Grø
stl and Grindahl. All these hash functions have underlying block ciphers that are modified versions of the Advance Encryption Standard and we investigate the behavior of these block ciphers under the integral attack. Statistical tests, such as the avalanche test and the collision test, are the regular tools for examining the hash function security. In this work, we inspect the statistical behavior the three hash functions and search for collisions. Although it is very difficult to obtain collisions for the actual algorithms, we find some collisions under slight modifications of the original constructions. The ease or difficulty of finding a collision for a modified version also shows the respective importance of the specific hash function branch, missing in the modified version.

Development progress may have evaded these areas yet impacts them through globalization and China's expanded market power. The burgeoning market for ejiao, a popular Traditional Chinese Medicine made from boiling donkey hides is a growing issue. Wealthier Chinese consumers, a stalled South African rural population who missed development's benefits plus increasing economic and political engagements between the two countries allows a monopolized, and often exploitative market for donkeys. Politicians are eager to partner with Chinese ejiao retailers despite resulting decimation of donkey populations in some areas of Africa and destruction of livelihoods. I evaluate the value of donkeys as domestic laborers within South Africa along with how China's rapidly expanding market for ejiao and simultaneous consumption of donkeys has affected South Africa to answer in which context donkeys are more valuable. The thesis deals with the Czech Christmas spiritual music, wants to point out and pick up her beauty, popularity and popularity across the public and in the circles of the Church and in the wider public. The method is the research the available literature and handouts for this topic, which is evident in the Division of the individual chapters. The introduction will include music and history of music. In the next part we will spend Christmas spiritual music, we close its emergence, history, Christian context, familiar carols and songs and their lyrics with a spiritual focus, some of the selected authors and their works both from the past and from the present. We also in our surroundings, to learn more about the Christmas music of our neighbors, or some European countries. The diploma thesis is focused on the analysis of selected works of the winter landscape painting of the Norther European painters . Structurally, the work is conducted in such a way that it first introduces the evolution of landscape painting in the Northern Europe from 14.-16. century. In this context, three selected works are analyzed, whose scenes are set in the winter landscape. The cultural-historical context of the 16th-17th centuries is then outlined to familiarize with the circumstances of the time. century in the Netherlands. The work is devoted to The Memorial of Master John Hus as his presupposed birthplace. The first part deals with the history of this monument, it brings the summary of proprietary relations and maps the care about this historical object and the looks of Hus{\crq}s house. It also deals with the honour to the personality of Master John Hus and its forms through the historical periods. There are the oldest mentions about this respect, some legends, celebrations and other forms of respect in various periods. The second part deals with contemporary effort of professional public which is aimed to preparation of a new expozition in the Memorial of Master john Hus in Husinec. This preparation started in 2004 - 2005 by means of professional lectures. Finally the work introduces the project The Restoration of the Birthplace of Master John Hus which reakts on professional discussions from previous years and which is the alternative solution of current renewal of actual expozition installed in 1975. There is also pictured enclosure prezenting previous and current looks of Hus{\crq}s birthplace.
