Abstract:
The increased demand for Out of School Hours Care (OSHC) has been linked to changes that have occurred in the family structure and work habits. OSHC programs operating on school sites under the management of a committee of parents and school representatives provide parents with one option to meet the needs for care of primary age children. This study examined the issue of OSHC from the perspectives of workers involved in the provision of services in government school sites in the ACT. Data were gathered by way of semi-structured interviews with nineteen coordinators from sixteen After School Care (ASC) centres. The data were analysed to find similarities in the experiences of workers interviewed. Extracts from the interviews are presented in the findings to illustrate the perceptions care givers have about their work and areas where changes can be made to improve the quality of ASC services. The study provides information on the factors affecting the provision of quality services and suggests possible directions for future planning and research in this area. The findings indicate that the care givers interviewed perceived their role as a positive influence in the lives of children and families. Care givers sought to provide relaxed environments for children after the school day. Many centres operated with the strong support of parents, schools and management committees. The findings suggest that ASC centres can and do operate successfully on school sites in the ACT and that care givers were generally happy with the level of service provided. However, variability in the facilities and support provided to centres was noted. The study concludes that real improvement to the quality of programs and support of workers is dependent on the introduction of nationally recognised standards in the provision of ASC.

Cílem této práce je poskytnout snadný způsob, jak rozšířit analyzační nástroj o rozhraní splňující standard OSLC. Takové rozhraní umožňuje jednoduchou integraci nástrojů s jinými nástroji nebo systémy, umožňuje jejich vzdálené použití skrze webové služby a umožňuje je jednoduše propojit s databází pro databázové dotazy a pro perzistentní uložení dat. Toto je dosaženo návrhem a implementací OSLC adaptéru pomocí sady nástrojů Eclipse Lyo. Adaptér používá jako rozhraní doménu OSLC Automation a je dostatečně univerzální na to, aby skrze toto rozhraní pokryl funkcionalitu většiny analyzačních nástrojů za pomocí jejich stávajících rozhraní na příkazové řádce. Tato práce poskytuje úvod k OSLC, Eclipse Lyo a souvisejícím konceptům. Dále tato práce definuje požadavky a odlišnosti různých analyzačních nástrojů a diskutuje návrh adaptéru a faktory, které ovlivnily návrhová rozhodnutí. A nakonec prezentuje implementovaný adaptér a jeho vyhodnocení pomocí automatizované testovací sady a pomocí experimentů s řadou analyzačních nástrojů. Nejvýznamnější ukazatel hodnocení vytvořeného adaptéru je to, že už teď je používán v praxi pro přidání OSLC rozhraní k nástrojům ANaConDA, Perun, Spectra (všechny tři vyvíjené na VeriFIT) a HiLiTE (Honeywell).

Mitochondria is no longer viewed as merely a powerhouse of the cell. It is now apparentthat mitochondria play a central role in signaling, maintaining cellular homeostasis and cell fate.Mitochondrial dysfunction has been linked to many human diseases caused by cellular metabolicderegulation, such as obesity, diabetes, neurodegenerative disease, cardiovascular disease andcancer. Eukaryotic organisms have evolved an efficient way in sensing, communicating andresponding to cellular stress and regulating mitochondrial activity correspondingly through acomplex network of intercommunicating protein kinases and their downstream effectors. Thisdissertation focuses on the interplay of two of the master metabolic regulators in the cell: AMPKand PASK, and characterization of the functions of their downstream substrates: OSBP andMED13. AMPK is an energy sensing kinase that maintains energy homeostasis in the cell,whereas PASK is a nutrient sensing kinase that regulates glucose partitioning and respiration inthe cell. Both kinases play important roles in mitochondrial function and regulation, anddeficiency in either kinase has been found to associate with various human pathologies. Furthercharacterization of the cross-talk and molecular mechanisms of both kinases in controllingmitochondrial health and function may aid in the identification of new targets for treatingmetabolic diseases.

Abstract:
Oral squamous cell carcinoma (OSCC) is a highly aggressive cancer that is characterized by a high rate of invasion and destruction to the surrounding tissues, with patients showing poor 5-year survival rate. A pre-malignant stage of cellular atypia and loss of stratification within the epithelium (dysplasia) occurs prior to the establishment of OSCC, which is manifested as white (leukoplakia) or red (erythroplakia) lesions. Treatment of dysplasia/OSCC involves surgical intervention with the removal of an adequate safety margin. However, high grade dysplasia and OSCC exhibit high recurrence rates. To date the only method used to diagnose oral epithelial dysplasia (OED) and OSCC is haematoxylin and eosin staining of biopsies and examination by an experienced pathologist. Furthermore, the mechanisms of dysplasia formation, transition to OSCC, and high recurrence rates are little understood. Recent data from the Ward lab suggests that the cell surface tumour suppressor protein E-cadherin plays an important role in regulating many cellular functions in epithelial cells. Loss of E-cadherin in carcinomas has been well studied and is linked with tumour invasion, metastasis and poorer patient outcomes. In this thesis, I have investigated expression of E-cadherin in low grade (LG) and high grade (HG) dysplasia and T1 and T4 OSCC patient biopsies to determine whether loss of this protein occurs prior to tumour cell invasion. Furthermore, microarray data analysis identified Epithelial Membrane Protein-1 (EMP-1) as a putative early marker of tumorigenesis, with alterations of N-cadherin, CD44 and 5T4 oncofoetal antigen expression during tumorigenesis inferred from our studies in embryonic stem cells. Immunofluorescence microscopy (IFM) analysis revealed that normal oral epithelium exhibits cell surface E-cadherin, EMP-1 and 5T4 expression but generally lacks N-cadherin and CD44 reactivity. The statistically significant loss of both E-cadherin and EMP-1 was observed in low and high grade dysplastic tissue and OSCC biopsies (p < 0.001). 5T4 expression was decreased in HG dysplasia (p < 0.001), suggesting this may be a useful assay for discriminating between LG and HG dysplastic tissues. N-cadherin or CD44 did not show any statistical significance in expression between normal, LG/HG dysplasia and T1/T4 OSCC, although there was a trend for increased N-cadherin expression in T4 OSCC. Significantly, Ecadherin expression was decreased or absent from surgical margins of LG/HG dysplasia and T1 OSCC, and EMP-1 and 5T4 were absent from the margins of LG and HG dysplastic tissue, indicating that these margins are abnormal. Therefore, loss of Ecadherin and EMP-1 are early events associated with dysplastic tissue and may be a useful method to confirm the removal of sufficient surgical safety margin. The OSCC cell line BICR56 was assessed for marker expression and exhibited a phenotype consistent with normal oral epithelium. Inhibition of E-cadherin protein in BICR56 cells using a neutralising antibody led to decreased cell surface localisation of 5T4 antigens, suggesting that E-cadherin expression is critical for maintenance of a normal epithelial phenotype. In conclusion, both E-cadherin and EMP-1 are useful markers for detecting early cancerous changes and 5T4 expression can discriminate between LG and HG dysplasia. This study has also shown significant abnormalities within the surgical safety margins of LG/HG dysplasia and T1 OSCC biopsies, which requires further investigation, and may explain the high recurrence rates seen in such patients.

Cientistas geram dados experimentais, e o interesse no compartilhamento destes dados é enorme, em especial na área biológica. É o compartilhamento que permite o trabalho cooperativo entre laboratórios espalhados pelo mundo. Este interesse gerou a formação de diversas bases de dados, como o GenBank e o Protein Data Bank. Cientistas de todo o mundo ali depositam seus dados, formando grandes repositórios globais que são usados para análise e recuperação de registros. Tais tarefas podem ser feitas de duas formas: utilizando serviços implantados nas bases centrais, como o BLAST para a recuperação de sequências homólogas, ou fazendo cópias locais das bases e implantando localmente novos algoritmos. Neste trabalho nós propomos a formação de um repositório global não real, mas sim virtual, através do uso de agentes móveis, e da fácil implantação por um usuário de funcionalidades adicionais para análise e recuperação de dados, utilizando carga dinâmica de classes Java.Nós aplicamos este modelo na construção de uma nova versão do AnaGel, um sistema para armazenamento e análise de registros eletroforéticos. A eletroforese é um processo de separação demacromoléculas biológicas de extrema utilidade para bioquímicos e geneticistas. Várias amostras contendo diferentes misturas de macromoléculas - geralmente fragmentos de DNA, RNA ou deproteínas, com a mesma carga elétrica - são colocadas em uma placa coberta por um gel, e submetidas a um campo elétrico, que força a migração das macromoléculas através do gel. Moléculas menoresencontram menor dificuldade para atravessar o gel, e seu deslocamento é consequentemente maior. O inverso acontece com as moléculas maiores. Um experimento de eletroforese gera então umaimagem, com várias canaletas, cada uma correspondendo a uma amostra, onde em cada canaleta há uma formação de bandas nos pontos de acúmulo de moléculas com o mesmo peso molecular.Em sua primeira versão o AnaGel já era uma ferramenta bastante completa para o tratamento de registros eletroforéticos, oferecendo a deteção automática de canaletas e bandas, a correção de distorções, a determinação de pesos moleculares, além da recuperação de registros por diversos métodos. Era entretanto um programa que deveria ser instalado em um único computador, e este mesmo computador servia para a entrada e o armazenamento dos dados. A versão atual, toda construída em Java, apresenta como vantagens a instalação em um servidor, com o uso por navegadores web, a possibilidade de análises e recuperação de registros através de agentes que se movimentam por todos os repositórios deuma rede de laboratórios que desejam compartilhar dados, e a facilidade de adição de novas funcionalidades pelos usuários, que necessitam apenas de construir classes Java obedecendo a umaclasse abstrata exportada pelo AnaGel.Nesta dissertação nós apresentamos uma discussão sobre o uso da Web para o trabalho cooperativo entre cientistas experimentais. A solução técnica adotada - agentes móveis, com o sistema Grasshopper - é comparada com outras possibilidades para estruturação de sistemas distribuídos. Nós procuramos mostrar como o sistema AnaGel se compara favoravelmente com seus concorrentes comerciais. Diversos detalhes da implementação são explicados, e apontadas direções para melhorias futuras.

Abstract:
A systems biology approach was adopted in order to assess various aspects of the disease oral squamous cell carcinoma. Three main aims were addressed; assess the ability of CoCl2 to mimic the hypoxic response in a eukaryotic cell line, assess the role of PDE4D in oral squamous cell carcinoma (OSCC) and the construction of a normoxic/hypoxic mathematical model to identify therapeutic targets.Cancer cells often acquire a revised metabolism which aids in initiation, survival and progression of the tumour. This is predominantly due to the transcription factor HIF-1 which is activated under hypoxic conditions. Certain compounds such as cobalt chloride (CoCl2) have been used extensively to inhibit the degradation of HIF-1α and simulate hypoxia. CoCl2 is likely to have off-target effects on metabolism; these effects were examined when exposing human telomerase reverse transcriptase (hTERT) cells to 100μM CoCl2. Gas chromatography-mass spectrometry (GC-MS), liquid chromatography-mass spectrometry (LC-MS) based metabolomics were utilised in combination with ELISA assays for HIF-1α and ATP. Central metabolism was accurately mimicked when hTERT cells were subjected to 100μM CoCl2, however; it was apparent that this concentration of CoCl2 does not induce an equal extent of hypoxia as 1% oxygen. A number of off-target effects of CoCl2 were observed in secondary metabolism, specifically in lipids and fatty acids. In conclusion, CoCl2 should be used with caution as a hypoxic mimicker with the caveat that interpretation of results should be restricted to its effects on central metabolism.The transcription factor CREB has the ability to regulate approximately 4000 genes, a number of which are associated with cancer initiation and progression. Cyclic adenosine monophosphate (cAMP) is required to activate CREB and is partially regulated through its degradation via the enzyme phosphodiesterase type 4D (PDE4D). A homozygous deletion of PDE4D has been associated with OSCC; however; the exact consequence of this deletion has not been fully elucidated. PDE4D was knocked down in the OSCC cell line BicR16 and cellular proliferation, migration, resistance to ionising radiation and central metabolism was investigated using MTT, scratch, clonogenic and GC-MS, respectively. The knockdown resulted in an increase in proliferation, migration and radiation resistance suggesting the role of PDE4D as a TSG. Amino acids, cholesterol, fatty acids, carbohydrates and TCA intermediates were found to be altered in concentration.A mathematical model of glycolysis, TCA and glutaminolysis under normoxia and hypoxia was constructed through the amalgamation of two established models from the literature. New reactions, parameters and metabolite concentrations were added and unnecessary entities were deleted. COmplex PAthway SImulator (COPASI) was utilised to construct the model before validating the model using experimental data from the literature and steady state and flux analyses. Sensitivity analysis and a reduction in external glucose and glutamine were mimicked and the alterations in hypoxic and normoxic metabolism analysed. The reactions vCSII, vGS, vPGK and vGII were identified as potential therapeutic targets which may affect metabolism in hypoxia only. However, certain validation methods proved unsuccessful and hence the model requires further work before attempting the analyses again.

Abstract:
Asbestos sampling and monitoring data, starting from 2003, located in a large federal facility’s Asbestos Air Database Management (AADM) repository will be queried and analyzed on airborne asbestos fiber concentrations generated from abatement activities of asbestos-containing materials (ACM) and asbestos-containing building materials (ACBM). Historically, concerns expressed by personnel outside of the containment areas, whether adjacent to or quite a distance from the asbestos abatement activities present operational challenges for the project manager, potential angst and uneasiness to personnel residing next to the abatement activity as well as programmatic concerns to the building/facility managers. The concerned individuals working outside the abatement enclosure, in an unrelated activity to the abatement often believe there is a high probability for personal exposures of asbestos fibers based on their proximity to the abatement activities. Perceptions regarding containment performance, the uncertainty surrounding the long latency period between asbestos fiber exposure and onset of disease, and the lack of understanding about containment efficacy are just some of the elements that can generate worry. Using statistical analysis tools, such as regression analysis, relationships between one or more predictor variables relative to a response variable were investigated. This research reviewed and compared airborne asbestos fiber sample data relative to the specific activities, whether abatement or other, that were performed. In an effort to establish a holistic awareness to the reader as to why individuals are concerned about being located near asbestos abatement activities, the history of asbestos regulation and epidemiology is also discussed. The dataset contained 5534 sampling records made up of 3738 area samples (1426 outside containment structure and 2312 inside containment structure) and 1796 personal samples. Analysis identified that 1779 (>99%) out of the 1796 total personal exposure samples in the dataset indicated the asbestos workers were appropriately protected from overexposures. Only seventeen (<1%) of the 1796 total personal exposure samples exceeded the respective Occupational Exposure Limits (OELs): • Fifteen of the 17 exceeded the 8-hr TWA, 0.1 f/cc OEL. These exceedances were positively correlated with work tasks identifying that no respirators were required due to a Negative Exposure Assessment (NEA). • Two of the 17 exceeded the Assigned Protection Factor for the Half Face APR (10x the OEL protection) adjusted 8-hr TWA OEL, 1 f/cc. • There were no OEL exceedances identified for any 30-min Excursion personal sampling events. The focus for this assessment was to determine the efficacy of the asbestos abatement process and increased health risks to personnel. The findings suggest there is performance variability in the containment structures; however, the abatement process was effective and protective of the non-asbestos personnel outside of the abatement work area. It can also be concluded that the abatement process of containment structures, negative air, work methods (e.g. wet methods) and Personal Protective Equipment (PPE) have provided a protective environment for both workers and non-asbestos personnel outside of the containment structures.

Abstract:
Oral cancer in early stages (I and II) may be curable by surgery or radiation therapy alone but advanced stage disease (III and IV) has a relatively low survival rate. The pathogenic pathways that contribute to Oral Squamous Cell Carcinoma (OSCC) remain poorly characterised and the critical factor in the lack of prognostic improvement is that a significant proportion of cancers initially are asymptomatic lesions and are not diagnosed or treated until they reach an advanced stage. Hence, a clinically applicable gene expression signature is in high demand and improved characterization of the OSCC gene expression profile would constitute substantial progress. For OSCC, possible themes that might be addressed using microarray data include distinguishing the disease from normal at the molecular level; determining whether specific biomarkers or profiles are predictive for tumour behaviour; and identifying biologic pathways necessarily altered in tumourigenesis, potentially illuminating novel therapeutic targets. However, OSCC is a heterogeneous disease, making diagnostic biomarker development difficult. Although this phenotypic variation is striking when one compares OSCC from different geographic locales, little is known about the underpinning biological mechanisms. Cancer may be accompanied by the production and release of a substantial number of proteins, metabolites and/or hormones into the blood, saliva, and other body fluids that could also serve as useful markers for assessing prognosis, metastasis, monitoring treatment, and detecting malignant disease at an early stage. The primary aim of this thesis is to investigate metabolomic and transcriptomic profiles using multiple bioinformatics approaches and biological annotation tools in an attempt to identify specific biomarkers and prediction models for OSCC from each profile as well as from the interface outcomes of integrating the two platforms. Additional aims of the thesis go further to identify the mechanisms underlying the biological changes during tumorigenic transformation of OSCC, as well as to determine biological processes underpinning the heterogeneity of the disease among populations. Two review studies were carried out in this thesis. The review study of published transcriptomic profiles of OSCC specified several genes and pathways exhibiting substantially altered expression in cancerous versus noncancerous states across studies. However, the result of the review suggests not relying on the final set of genes published by the individual studies, but to access the raw data of each study and start subsequent analysis from that stage using unified bioinformatics approaches to acquire useful and complete understanding of the systems biology. The other review study focused on the metabolic profiles of OSCC and revealed a systemic metabolic response to cancer, which bears great potential for biomarker development and diagnosis of oral cancer. However, the metabolic signature still needs to improve specificity for OSCC from other types of cancer. In an attempt to detect a robust gene signature of OSCC overcoming the limitation of the transcriptomic review in accessing the raw data from the previous works, four public microarray raw datasets (comprising 365 tumour and normal samples) of OSCC were successfully integrated using ComBat data integration method in R software, determining the common set of genes, biomarkers, and the relative regulatory pathways possibly accountable for tumour transformation and growth in OSCC. Examination of the meta-analysis datasets showed several discriminating gene expression signatures for OSCC relative to normal oral mucosa; with a signature of 8 genes (MMP1, LAMC2, PTHLH, TPBG, GPD1L, MAL, TMPRSS11B, and SLC27A6) exhibiting the best discriminating performance and show potential as a diagnostic biomarker set. In addition, 32 biomarkers specific to OSCC and HNSCC were identified with the majority involved in extracellular matrix (ECM), interleukins, and peptidase activity where around 2/3 of them are located in the extracellular space and plasma membrane. Additionally, investigation of the interactive network created by merging metabolic and transcriptomic profiles highlighted the significant molecular and cellular biofunctions, pathways, and biomarkers distinguishing OSCC from normal oral mucosa. The results highlighted interactions of significantly altered expression of Dglucose, ethanol, glutathione, GABA, taurine, choline, creatinine, and pyruvate metabolites with the expressed PTGS2, IL1B, IL8, IL6, MMP1, MMP3, MMP9, SERPINE1, COL1A1, COL4A1, LAMC2, POSTN, ADAM12, CDKN2A, PDPN, TGM3, SPINK5, TIMP4, KRT19, and CRYAB biomarkers of OSCC. Such a pattern may represent a clinically useful surrogate for the presence of OSCC which might help in deciphering some of the obscure multifaceted mechanisms underlying carcinogenesis of OSCC which emerged from dysregulated genetic and metabolic system of the body. In an attempt to define pathways of importance in two phenotypically different forms of OSCC, transcriptomic analysis of OSCC from UK and Sri Lankan patients was undertaken. The development of OSCCs in UK and Sri Lankan populations appears largely mediated by similar biological pathways despite the differences related to race, ethnicity, lifestyle, and/or exposure to environmental carcinogens. However, results revealed a highly activated “Cell-mediated Immune Response” in Sri Lankan tumour and normal samples relative to UK cohorts which may reflects a role in resistance of patients to invasiveness, metastasis, and mortality observed in Sri Lankan relative to UK patients. In conclusion, multiple molecular profiles were able to identify a unique transcriptomic profile for OSCC and could further discriminate the tumour from normal oral mucosa on the basis of 8 genes. Altered expression of several metabolic and transcriptomic biomarkers specific for OSCC were identified, along with several dysregulated pathways and molecular processes found common in patient with oral cancer. Integrating both metabolomic and transcriptomic signatures revealed a promising strategy in analysing the concurrent perturbation in both genetic and metabolic systems of the body. Additional results revealed possible impact of specific supplementary dietary components in boosting the immune system of the body against invasion, progression, and metastasis of the disease. Further clinical studies are required to confirm and validate the current results.

Tool Integration is a vital part in the modern IT industry. With the ever increasing complexity in reality, more tools in different domains are needed in research and development process. However, currently no vendor has a complete solution for the whole process, and no mature solution to integrate different tools together, thus tools are still used separately in the industry. Due to this separation, the same information is created more than once for different tools, which is both time wasting and error prone.This thesis is part of the research to deliver a model-based tool integration framework that helps the end user design their own scenario of tool integration and implement it with less effort by generating most common parts automatically. This thesis itself is mainly focused on tool adapters, including the model-based tool adapter construction and discovery. In the first part, a model-based tool adapter construction platform conforming to OSLC is designed and implemented, based on which, the construction process of a tool adapter is presented with an example. With this platform, most of the codes and configuration files can be generated, with the exemption of the tool specific functionalities. The tool adapter is constructed as a separate SCA component, and can be included in the SCA based tool chain with minor configuration. With SCA, the deployment of the tool adapter and future management can be largely eased. In the second part, the model-based discovery process of an unknown tool adapter conforming to OSLC and our assumptions is presented in detail. With the discovery tool, the sharing of the tool adapter is made possible, and the integration of the different tools are largely eased. An example of discover an unknown tool adapter is also included for a more clear explanation. Finally, in the meanwhile of the design and implementation of the construction platform and the discovery process, the existing Matlab/Simulink tool adapter is extended and refined to make it full compatible to the standard and our tool chain.

La tesis de investigación consiste esencialmente en una etnografía realizada entre los años 2012 y 2013 en el Osho International Meditation Resort en la ciudad de Puna, India. Dicho trabajo de campo tardó seis meses e involucró cientos de notas de campo y treinta y cinco entrevistas a residentes y visitantes del ashram. Básicamente, los objetivos planteados por el investigador y el director de la tesis fueron comprender como se desarrollan en ese lugar los procesos de crecimiento y/o hedonismo espiritual, y distinguir entre elementos religiosos y psicoterapéuticos que emanan de las prácticas espirituales diseñadas y gestionadas por la administración del ashram (Inner Circle). Se trata de una investigación que por un lado busca integrar los contenidos teóricos acumulados desde la sociología de la religión, con el análisis crítico de las modernas psicoterapias post-freudianas. El marco teórico está dividido en cuatro dimensiones. La primera da cuenta del estudio sociológico de los denominados Nuevos Movimientos Religiosos, y sobre cómo se gestionan en esos grupos una nueva espiritualidad más flexible, privada, pragmática y celebrativa. La segunda dimensión da cuenta del transitar de la moderna psicoterapia desde un esfuerzo clínico por reducir el sufrimiento, a través de Freud y el psicoanálisis clásico, a un conjunto de técnicas y dispositivos terapéuticos destinados a encontrar la felicidad y la autorrealización. Una tercera dimensión se explaya sobre las características que definen una práctica espiritual sana o destinada al crecimiento espiritual, de otra más orientada al hedonismo o materialismo espiritual. Por último, el apartado teórico se hace cargo de describir los elementos centrales del Rajneeshism, doctrina religiosa con que fue conocida la obra de Rajneesh/Osho en los años ochenta, y que hasta el día de hoy permea la gestión y los rituales de intercambio simbólico que se suceden en el resort. La investigación describe los principales rituales de interacción que se construyen cotidianamente al interior del ashram, así como la organización y gestión de los dispositivos espirituales ofertados por la administración del resort, los que bajo la forma de terapias meditativas y meditaciones activas siguen siendo consumidos por numerosos buscadores espirituales. La investigación se centró en develar todos aquellos intercambios simbólicos que definen la espiritualidad catártica y tántrica de este movimiento religioso. Que mensajes son transmitidos, que ceremonias constituyen sus rituales de interacción, discernir entre el contenido psicoterapéutico y religioso al cual acceden los miembros del ashram en su práctica espiritual, y sobre todo, que elementos de esas prácticas son conducentes a un proceso de crecimiento espiritual y cuales operan al servicio de un hedonismo espiritual. La gran conclusión de la tesis es que nos encontramos frente a un nuevo movimiento religioso que aunque inicialmente se constituyó como un movimiento contracultural, termino transformándose en un culto clientelar que fetichiza no sólo la experiencia espiritual, sino también el bienestar psicoterapéutico. La práctica espiritual al interior del ashram tiene referencias religiosas, por cuanto los residentes y visitantes del resort encuentran en el mensaje de Osho y sus técnicas psicoterapéuticas y meditativas un encuadre para conectarse con lo trascendente o numinoso. Así mismo, esta experiencia espiritual es vivenciada como psicoterapéutica, dentro de un encuadre psicológico humanista-gestáltico- transpersonal, por cuanto es conducente a la autorrealización y el bienestar personal. Por último, ya sea como experiencia religiosa o psicoterapéutica siempre la experiencia es organizada de acuerdo a los requerimientos particulares y el poder adquisitivo de cada residente o visitante. Esto se traduce en que el resort ofrece y gestiona un conjunto de “tecnologías del yo” en el marco de un “capitalismo espiritual”, donde el despertar de la consciencia se oferta y consume más como una mercancía que como un verdadero proceso de crecimiento espiritual.
The research is essentially an ethnography conducted between years 2012 and 2013 in the Osho International Meditation Resort of Pune, India. This fieldwork took six months in two visits and involved hundreds of field notes and thirty-five interviews into the ashram. Basically, the goals were understand how the visitors and residents build their own spiritual process, and distinguish between religious and psychotherapeutic framework from the spiritual practices which are designed and managed by the administration of the ashram (Inner Circle). The research describes the main rituals of everyday interaction built into the ashram, as well as the organization and management of spiritual devices offered by the resort management, which in the form of meditative therapies and active meditations are still demanding by many spiritual seekers. The research focused on revealing all those symbolic exchanges that define the cathartic and tantric spirituality of this new religious movement. Which messages are transmitted, what kind of psychotherapy and religious content are involved into their rituals of interaction and their spiritual practice, and above all, which elements of these practices are conducive to a process of spiritual growth and which operate in the service of spiritual hedonism. The major conclusion of the thesis is related that we face a New Religious Movement, called in the eighties Rajneeshism, that initially was established as a countercultural movement, and now operate more like a client cult fetishizing not only the sannyasin´s spiritual experience, also their psychotherapeutic welfare. Spiritual practice within the ashram has religious references, because residents and visitors connect with the transcendental or numinous experience through psychotherapeutic and meditative techniques for spiritual awake. Also, this spiritual path is experienced as psychotherapy within a Transpersonal and Gestalt psychological frame, leading to self-realization and personal wellness. In general, religious and psychotherapeutic experience is always experienced and organized according to the particular requirements and purchasing power of each resident or visitor. This means that the resort provides and manages a set of "technologies of the self" in order to legitimate some kind of "spiritual capitalism" where the awakening of consciousness is selling and consuming more as a commodity than a real process of spiritual growth.

The principal objective of this study was to investigate whether or not nurses' compliance with universal precautions procedures improved after the mandatory Occupational Safety and Health Administration regulations were implemented in 1992. Two random samples of registered nurses and licensed practical nurses registered in Tennessee responded to survey questionnaires measuring universal precautions compliance and practice barriers to compliance in 1991 and 1993 (n = 306). The 1993 sample of nurses reported significantly greater compliance with universal precautions (p < 0.001) than the 1991 sample. The most noteworthy improvement between the 1991 and the 1993 groups was a significant increase in compliance for patients described as HIV/HBV-status unknown and HIV/HBV-negative (p < 0.001). Practice barriers hindering compliance with universal precautions decreased significantly (p < 0.001) in the 1991-1993 time frame. Problematic practice barriers identified in both groups were needle recapping, preference for isolation door signs, and concerns about offending patients and visitors.

Une distribution lipidique hétérogène est essentielle à l’identité et fonction des organelles, mais l’échange par trafic vésiculaire tend à annuler cette distribution. Il existe donc des mécanismes qui assurent l’homéostasie des lipides. Les protéines Osh (S. cerevisiae) et les OSBP-Related Proteins (ORP, H. sapiens), sont des transporteurs de lipides. Osh4 est capable d’échanger de l’ergostérol contre le phosphatidylinositol-4-phosphate (PI4P), présent sur l’appareil de Golgi. Utilisant des outils fluorescents mesurant avec une précision inégalée le transport de stérol et de PI4P, nous démontrons qu’Osh4 transporte du stérol contre son gradient de concentration en utilisant l’énergie d’un gradient de PI4P. Un couplage au métabolisme du PI4P permettrait à Osh4 d’alimenter le Golgi avec du stérol, ainsi créant le gradient de stérol entre ces organelles. La protéine OSBP participe, via sa capacité à connecter la membrane du RE à celle du trans-Golgi, à la création de jonctions entre ces organelles. Nous avons montré qu’OSBP, par échange stérol/PI4P, utilise le PI4P pour transférer du cholestérol au Golgi, mais également pour autoréguler sa capacité à former les jonctions. Osh6 lie la phosphatidylsérine, nous permettant d’étudier un nouveau mécanisme d’échange. Nous avons résolu la structure cristallographique d’un complexe Osh6/PI4P et avons pu observer l’échange de ces deux ligands par Osh6 entre deux membranes. Cette étude nous permet de suggérer que l’échange de PI4P avec divers lipides, via les protéines Osh/ORP, serait un mécanisme général permettant aux cellules de maintenir le gradient lipidique entre le RE et les membranes tardives de la voie sécrétoire
An uneven lipid distribution is essential for the function of eukaryotic organelles. However, exchange of material by vesicular trafficking has a tendency to perturb this distribution; mechanisms must though exist to ensure lipid homeostasis. Osh proteins (S. cerevisiae) and OSBP-Related Proteins (ORPs, H. sapiens), are lipid transfer proteins (LTPs). Osh4 is capable of exchanging ergosterol for phosphatidylinositol 4-phosphate (PI4P), found on the Golgi. Using novel fluorescent tools to measure with unprecedented precision the transport of sterol and PI4P, we find that Osh4 can transport sterol against its concentration gradient using the energy of a PI4P gradient. Coupled to phosphoinositide metabolism, this allows Osh4 to transport sterol to the trans-Golgi and create the sterol gradients observed between these organelles. OSBP participates in the creation of membrane contact sites (MCSs) via its capacity to connect ER membranes to those of the trans-Golgi. We have shown that it uses PI4P for transporting cholesterol from the ER to the trans-Golgi by sterol/PI4P counterexchange, hence also autoregulating its tethering activity. Finally, the identification of phosphatidylserine as a ligand for Osh6 allowed us to analyze the possible extrapolation of the PI4P counterexchange mechanism. We have solved the crystal structure of Osh6 in complex with PI4P and have been able to follow counterexchange of PI(4)P and PS in vitro. Concluding, our studies allow us to suggest a general mechanism for ORP/Osh-mediated counterexchange of PI4P for other lipids to maintain lipid gradients between the ER and late membranes of the secretory pathway

In today’s practical reality there are many different tools being used in their respective phases of thesystem development lifecycle. Every tool employs its own underlying metamodel and these metamodelstend to vary greatly in size and complexity, making them difficult to integrate. One solution to overcomethis problem is to build a tool integration framework that is based on a single, shared metamodel.The iFEST project aims to specify and develop such a tool integration framework for tools used in thedevelopment of heterogeneous and multi-core embedded systems. This framework is known as the iFESTTool Integration Framework or iFEST IF.The iFEST IF uses Web services based on the Open Services for Lifecycle Collaboration (OSLC)standards and specifications to make the tools within the tool chain communicate with each other. Tovalidate the framework, an industrial case study called ‘Wind Turbine’, using several embedded systemstools, has been carried out. Tools used to design, implement and test a controller for a wind turbine havebeen integrated in a prototype tool chain. To expose tools’ internal data through Web services, a tooladaptor is needed. This work reports on the development of such a tool adaptor for the RequirementsManagement module of HP Application Lifecycle Management (ALM), one of the tools used in the WindTurbine industrial case study. A generalization of the challenges faced while developing the tool adaptoris made. These challenges indicate that, despite having a tool integration framework, tool integration canstill be a difficult task with many obstacles to overcome. Especially when tools are not developed with tool integration in mind from the start.

Dessametamodeller kan variera avsevärt i både storlek och komplexitet, vilket gör dem svåra attintegrera. En lösning på detta problem är att bygga ett ramverk för verktygsintegration sombaseras på en enda, gemensam metamodell.iFEST-projektets mål är att specificera och utveckla ett ramverk för verktygsintegration förverktyg som används i utvecklingen av heterogena och multi-core inbyggda system. Dettaramverk benämns iFEST Tool Integration Framework eller iFEST IF.iFEST IF använder webbtjänster baserade på en standard som kallas OSCL, Open Services forLifecycle Collaboration samt specifikationer som gör att verktygen i verktygskedjan kankommunicera med varandra. För att validera ramverket har en fallstudie vid namn ”WindTurbine” gjorts med flertal inbyggda systemverktyg. Verktyg som används för att designa,implementera och testa en styrenhet för vindturbiner har integrerats i prototyp av enverktygskedja. För att bearbeta och behandla intern data genom webbtjänster behövs enverktygsadapter. In the years 2004 to 2012, a long-term study on the effectiveness of safety experts was commissioned by the German Social Accident Insurance (DGUV). An interdisciplinary team of scientists investigated in these years the activities and the effectiveness of services for professionals responsible for occupational safety and health (OSH). In a case study-like manner it is discussed how the technology that was originally developed as an acquisitions and incentive instrument for the various phases of the safety experts' long-term study, now has become the safety experts’ online community. Both as a stand-alone instrument of prevention as well as a place of learning for professionals for occupational safety, it seems to be a highly appropriate technology. Accompanying the mentoring ensures regular technology updates to meet the increasingly broad use by a growing number of OSH specialists.

Electromagnetic scattering from rough surfaces is of prime importance in the engineering field since it affects communications, radar, remote sensing, acoustics, etc. The actual problem of scattering from rough surfaces is complicated and involves three dimensional scattering from either lossy or dielectric, electrically large surface. Integral equations are widely utilized to solve this kind of problem but this solution to the problem is generally computationally intensive. In the On-Surface Radiation Boundary Condition (OSRC) method, a higher order radiation condition is imposed directly on the surface of the scatterer. This reduces the integral equation for the scattered field to a line integral which can be easily evaluated numerically. In this thesis, the OSRC method is used to formulate the problem of scattering from periodic rough, two dimensional surfaces illuminated by a transverse magnetic, plane electromagnetic wave. Three geometric surfaces are considered. A comparison is made between the present formulation, the exact solution, and the physical optics approximation.

Lyme disease is an emerging infection that is caused by the Borrelia burgdorferi sensu lato complex. These bacteria exist in nature through an enzootic life cycle involving Ixodes ticks and various reservoir hosts. One way that this bacterium adapts to the different hosts in the enzootic cycle is through the expression of outer surface protein C (OspC). OspC is a surface exposed lipoprotein encoded on circular plasmid 26 that forms homodimers on the bacterial surface and has distinct conserved and variable portions of sequence. When ospC is deleted, the spirochetes are unable to cause mammalian infection although the mechanism of this is unknown. Additionally, OspC is thought to be involved in reservoir host specificity/association and in tissue dissemination. In order to better understand the functional domains of OspC, the different conserved and variable portions of this protein were investigated. Three conserved portions of OspC were investigated: (1) the conserved cysteine residue at position 130 (C130), (2) the last ten C-terminal amino acids (C10), and (3) ligand binding domain 1 (LBD1). The C130 residue was mutated and this substitution disrupted OspC oligomerization in vitro and in vivo. A B. burgdorferi strain lacking the C10 retained full infectivity and plasminogen binding. The mutation of a single residue within LBD1 rendered B. burgdorferi noninfectious, indicating the importance of this domain in infection establishment. The variable portion of OspC was investigated by: (1) altering the surface charge of ligand binding domain 2 (LBD2), (2) inserting different OspC types into a constant genetic background, and (3) creating OspC hybrids. Alteration of the surface charge of LBD2 by site directed mutagenesis resulted in a lack of persistence in mice. By inserting an OspC type known to be noninfectious in mice into an infectious strain, infectivity was abolished. Strains expressing OspC hybrids indicated that multiple domains of OspC are involved in species specificity. Together these analyses demonstrated that OspC is as important protein that plays multiple roles in pathogenesis. The work presented here helps to increase the understanding of this crucial protein and the strains described can be used to decipher the full function of OspC.

Lyme disease is the most common tick-borne illness in North America. It can be hard to detect using blood tests, especially in the early stage. Due to the number of significant complications from untreated or undertreated Lyme disease, better methods need to be found to detect the disease. Some surface lipoproteins may be used to detect early disease due to their early expression. Others are maintained for the duration of the infection and can be used to detect chronic Lyme disease. Antibody responses to OspA, OspC and OspE were measured in sera from experimentally infected dogs. The response to OspA was only detectable in a few samples and did not appear to be sensitive for mammalian infections. A protein construct based on multiple epitopes of OspC was used to detect an antibody response starting three weeks after initial infection, and remained detectable for three months. As the response to OspC decreased, OspE could be used to detect an antibody response from three to six months after the initial infection. Because of the strength and the differences in timing of the antibody responses, OspC and OspE could be used to design an accurate blood test for Lyme disease that also indicates early and late stages of infection based on the results.

Following the devastating riots that took place there in June 2010, the city of Osh (Kyrgyzstan) has been the subject of a number of post-conflict reconstruction projects aimed at rebuilding its damaged urban fabric. As well as being varied in form and approach, these interventions have had a significant impact on the ways that Osh's citizens experience the city. Whilst some residents have welcomed the changes that have been brought about in Osh, others are concerned about what these might mean for their continued wellbeing in the city. By interrogating the shifting relationships between place, identity and collective memory, this thesis explores post-conflict reconstruction in Osh between 2010 and 2013. It seeks to build a clearer picture of urban change in the city over this period, and to unpack the diverse motivations that underpinned the reconstruction projects that were pursued or proposed at that time. Above all, it asks what these changes have meant for Osh residents, many of whom were still reeling from the violence that ripped the city apart in 2010.

Metoden som användes var en induktiv kvalitativ ansats, där 16 intervjuer genomfördes på sex stora Svenska företag. Resultatet visade att EU-OSHA:s råd upplevdes som genomförbara men de upplevdes ibland för övergripande och otydliga. De olika förutsättningarna och hindren som beskrevs formade följande teman: Kulturen på företaget, Krav, Kontroll & Stöd samt Kunskap & Kompetens. Diskussion: EU-OSHAs råd kan tyckas vara enkla men de ger inte praktiskt stöd till företagen. Alla informanter fann råden bra men de var tvetydiga och tolkades därför olika. Active noise reduction was first proposed as a solution for environmental noise over fifty years ago. The use of active noise reduction (ANR) systems, however, was not demonstrated until much later. Recent advances in technology have made the use of active noise reduction systems in personal hearing protection devices (HPDs) practical. Through the use of advanced electronics technology, ANR equipped devices offer the potential to provide increased low frequency attenuation for hearing conservation applications. In order to use ANR equipped devices in an Occupational Safety and Health Administration (OSHA) sanctioned hearing conservation program, a testing standard for ANR equipped HPDs needs to be developed. Existing HPD testing standards offer the most promise for developing an acceptable standard for testing ANR-equipped HPDs. The microphone in real ear (MIRE) testing method is one method that offers a practical method for determining the performance of ANR devices for use in hearing conservation programs. A modified version of this method was successfully used in this study to determine the performance profile of an open-back ANR equipped headset. The results of this and other studies that have used a modified MIRE testing method can be used to support the acceptance of this method as the basis of an approved standard for testing ANR-equipped hearing protection devices.

La maladie peut évoluer en atteintes secondaires se traduisant par l'infection de sites distants du point d'inoculation(LCR, articulation, coeur) Nos travaux ont concerné la diversité génétique du gène ospC. Différents groupes ospC ont pu être définis en regroupant des séquences ospC similaires. L'analyse par SSCP des séquences ospC de souches invasives (isolées de sites distants) nous a permis de montrer que ces souches appartenaient à un nombre restreint de groupes ospC (35%). Le caractére invasif de ces souches a été validé chez le modèle murin puisque seules les souches appartenant à des groupes invasifs étaient capables de coloniser des sites distants. Nous avons montré que la protéine OspC des groupes invasifs fixe avec une meilleure affinité le plasminogène humain
Lyme disease is caused by the bacterium Borrelia burdorferi transmitted by tick bite. Borreliosis may evolve in secondary infections corresponding to infection of distant sites from inoculation point (CS, joints, heart). We studied the genetic diversity of ospC gene. Similar ospC sequences can be classified in different ospC groups. The screening of ospC sequences from invasive strains (i. E isolated from distantsites) by SSCP analysis allowed us to determine that invasive strains belong to restricted ospC groups (35%). Using the mouse model, we confirmed that only strains from invasive groups are able to disseminate and colonize distant sites. We found that OspC proteins from invasive groups bind more strongly the plasminogen suggesting that ospC polymorphism and human invasiveness could be based on different affinity with the plasminogen. The concept of generating horizontal-parallax-only (HPO) holograms by computer simulations is investigated. The simulations in this thesis are based on Optical Scanning Holography (OSH) aimed at acquiring HPO information electronically. The principles of OSH, a technique that allows the extraction of 3-D information by a 2-D optical scan of the object is first summarized. The HPO principles and simulation scenarios are then discussed. In order to illustrate the ideas, holograms were created and reconstructed using MATLAB simulations. The holograms are simulated by convolving the Fresnel zone plates (FZP) with the object. The simulations focus on generating HPO holograms using 1-D FZPs modeled as 1-D Gaussian chirp beams of varying waists. An optical reconstruction scheme by cylindrical lens was proposed and simulated. Three-dimensional imaging using HPO-holograms was also discussed. Several reconstruction scenarios were investigated by digitally convolving the complex HPO-hologram with the free space impulse response or the Gaussian chirp beam. Although many ideas of HPO-holography have been proposed and studied, to the best of our knowledge, this is the first proposed electronic technique to acquire HPO-holographic information. The simulations demonstrate that holographic information reduction techniques also help to alleviate the problems associated with the restricted field of view upon holographic reconstruction for 3-D display. The simulations show that horizontal-parallax-only holography is an excellent way to reduce holographic information. Suggested future work includes actual optical experimentation to verify the ideas presented in this thesis.

An emerging problem in organisations is that there exist a large number of tools storing data that communicate with each other too often, throughout the process of an application or product development. However, no means of communication without the intervention of a central entity (usually a server) or storing the schema at a central repository exist. Accessing data among tools and linking them is tough and resource intensive. As part of the thesis, we develop a software (also referred to as ‘adaptor’ in the thesis), which, when implemented in the lifecycle management systems, integrates data seamlessly. This will eliminate the need of storing database schemas at a central repository and make the process of accessing data within tools less resource intensive. The adaptor acts as a wrapper to the tools and allows them to directly communicate with each other and exchange data. When using the developed adaptor for communicating data between various tools, the data in relational databases is first converted into RDF format and is then sent or received. Hence, RDF forms the crucial underlying concept on which the software will be based. The Resource description framework (RDF) provides the functionality of data integration irrespective of underlying schemas by treating data as resource and representing it as URIs. The model of RDF is a data model that is used for exchange and communication of data on the Internet and can be used in solving other real world problems like tool integration and automation of communication in relational databases. However, developing this adaptor for every tool requires understanding the individual schemas and structure of each of the tools’ database. This again requires a lot of effort for the developer of the adaptor. So, the main aim of the thesis will be to automate the development of such adaptors. With this automation, the need for anyone to manually assess the database and then develop the adaptor specific to the database is eliminated. Such adaptors and concepts can be used to implement similar solutions in other organisations faced with similar problems. In the end, the output of the thesis is an approachwhich automates the process of generating these adaptors.
Resource Description Framework (RDF) ger funktionaliteten av dataintegration, oberoende av underliggande scheman genom att behandla uppgifter som resurs och representerar det som URI. Modellen för Resource Description Framework är en datamodell som används för utbyte och kommunikation av uppgifter om Internet och kan användas för att lösa andra verkliga problem som integrationsverktyg och automatisering av kommunikation i relationsdatabaser. Ett växande problem i organisationer är att det finns ett stort antal verktyg som lagrar data och som kommunicerar med varandra alltför ofta, under hela processen för ett program eller produktutveckling. Men inga kommunikationsmedel utan ingripande av en central enhet (oftast en server) finns. Åtkomst av data mellan verktyg och länkningar mellan dem är resurskrävande. Som en del av avhandlingen utvecklar vi en programvara (även hänvisad till som "adapter" i avhandlingen), som integrerar data utan större problem. Detta kommer att eliminera behovet av att lagra databasscheman på en central lagringsplats och göra processen för att hämta data inom verktyg mindre resurskrävande. Detta kommer att ske efter beslut om en särskild strategi för att uppnå kommunikation mellan olika verktyg som kan vara en sammanslagning av många relevanta begrepp, genom studier av nya och kommande metoder som kan hjälpa i nämnda scenarier. Abstract:
Crassostrea gigas est l’espèce d’huître la plus cultivée au monde. Depuis 2008, des évènements de mortalité massive touchent les huîtres âgées de moins d’un an en Europe et en Océanie et ont été associés à l’émergence d’un nouveau variant de l’Ostréid herpèsvirus type 1, OsHV-1 μvar. Les huîtres sont naturellement présentes dans les écosystèmes estuariens et côtiers, caractérisés par des fluctuations de salinité et de pH. Dans le cadre de ce travail, l’effet de la salinité (10, 15, 25, 35‰) et du pH (7.8, 8.1) sur la réponse de l’huître à une infection par OsHV-1 a été étudié. La survie d’huîtres acclimatées à une salinité de 10‰ préalablement à l’exposition à une source infectieuse est de 95% et s’explique par une diminution de l’infectiosité d’OsHV-1. En revanche, la survie d’huîtres non-acclimatées simultanément soumises à un choc de salinité à 10‰ et exposées à une source infectieuse n’est que de 23%, ce qui reflète un affaiblissement physiologique. La survie (S) des huîtres aux autres salinités est classée de la manière suivante : S15‰ > S35‰ > S25‰ et selon le pH comme suit : SpH 7.8 < SpH 8.1. Dans les deux études ces contrastes de survie ne sont pas expliqués par un différentiel d’infectiosité du virus mais par celui de la réponse métabolique de l’hôte. Le métabolisme des sucres et l’activité antioxydante d’huîtres acclimatées à 25 et 35‰ sont supérieurs à celui d’huîtres à 10-15‰ et pourraient expliquer leur moindre survie. La plus faible activité antioxydante observée chez des huîtres maintenues à pH 7.8 suggère une production réduite d’espèces réactives de l’oxygène et le maintien d’une production basale d’espèces réactives de l’azote malgré l’infection, ce qui aurait pu réduire la capacité de défense des huîtres face au virus et expliquer leur moindre survie en comparaison à ce qui est observé à pH 8.1
Crassostrea gigas is the most cultivated oyster species around the world. Since 2008, mass mortality events have been affecting oysters aged less than one year old in Europe, and Oceania and have been associated with a new variant of the ostreid herpesvirus type 1, OsHV-1 μvar. Oysters live in estuaries and bays where they are exposed to fluctuations of salinity and pH. In the present work, the effect of salinity (10, 15, 25, 35 ‰) and pH (7.8, 8.1) on the response of oysters to infection by OsHV-1 was investigated. The survival of oysters acclimated to 10 ‰ prior to be exposed to a source of infection is 95% and is explained by a reduction of OsHV-1infectivity. In contrast, the survival of non-acclimated oysters simultaneously submitted to a salinity stress at 10 ‰ and exposed to a source of infectious is only 23%, likely reflecting physiological impairment. The survival (S) of oysters in other salinity treatment is ranked as follow: S15‰ > S35‰ > S25‰ and according to pH as follows: SpH 7.8 < SpH 8.1. In both studies these contrasts of survival are not explained by differences in OsHV-1 infectivity but in host metabolic response. The carbohydrate metabolism and antioxidant activity of oysters acclimated to 25 and 35‰ is higher than in oysters at 10-15 ‰ and could explain the lower survival. Lower antioxidant activity is observed in oysters maintained at pH 7.8 likely reflecting a reduced production of reactive oxygen species and the maintenance of a basal production of nitrogen reactive species despite infection. This might impair the defense ability of oysters to OsHV-1 explaining the reduced survival at pH 7.8 in comparison to what observed at pH 8.1
