Abstract Human ACAT1 cDNA K1 was first cloned and functionally expressed in 1993. There are two adjacent in-frame AUG codons, AUG1397–1399 and AUG1415–1417, at 5′-terminus of the open reading frame (ORF, nt 1397–3049) of human ACAT1 mRNA corresponding to cDNA K1. In current work, these two adjacent inframe AUGs at 5′-terminus of the predicted ORF (5′-ORF-AUGs) as start codons for translation initiation of human ACAT1 mRNA were characterized in detail. Codon mutations indicated that both of these two adjacent 5′-ORF-AUGs can be selected as start codons but the first 5′-ORF-AUG1397–1399 is a main start codon consistent with that of the predicted ORF of human ACAT1 mRNA. Further deletion and mutation analyses demonstrated that a stable upstream stem-loop structure enhanced the selection of the first 5′-ORF-AUG1397–1399 as a main start codon, in addition to upstream nucleotide A in the –3 position, which is a key site of Kozak sequence. In addition, result of ACAT1 enzymatic activity assay showed no obvious difference between these two ACAT1 proteins respectively initiated from the two adjacent 5′-ORF-AUGs. This work showed that a stable upstream stem-loop structure could modulate the start codon selection during translation initiation of mRNAs that contain adjacent multi-5′-ORF-AUGs.

Abstract:
Oil recovery factor (ORF) assessment for sites of the Sredne-Ugutskoye field was performed applying various methods. For this field, the ORF prognostic assessment applying the existing methodological approaches is very relevant, as well as the selection of the most appropriate and accurate ORF calculation methods, taking into account the specific features of the geological structure of formations and reservoirs. The research objective is to obtain an adequate assessment of the ORF for productive sites of the Sredne-Ugutskoye field. With the help of a whole group of methods (analogy, empirical-analytical and empirical-statistical methods), predictive calculations of the ORF value for a number of Jurassic-age development sites were performed.

Abstract Maize glutamine synthetase cDNA clones were isolated by genetic selection for functional rescue of an Escherichia coli delta glnA mutant growing on medium lacking glutamine. The Black Mexican Sweet cDNA library used in this study was constructed in pUC13 such that cDNA sense strands were transcribed under the control of the lac promoter. E. coli delta glnA cells were transformed with cDNA library plasmid DNA, grown briefly in rich medium to allow phenotypic expression of the cDNAs and the pUC13 ampr gene, and challenged to grow on agar medium lacking glutamine. Large numbers of glutamine synthetase cDNA clones have been identified in individual 150-mm Petri dishes; all characterized cDNA clones carry complete coding sequences. Two cDNAs identical except for different 5' and 3' termini have been sequenced. The major open reading frame predicts a protein with an amino acid sequence that exhibits striking similarity to the amino acid sequences of the predicted products of previously sequenced eukaryotic glutamine synthetase cDNAs and genes. In addition, the maize glutamine synthetase cDNAs were shown to contain a 5' mini-ORF of 29 codons separated by 37 nucleotide pairs from the major ORF. This mini-ORF was shown not to be essential for the functional rescue of the E. coli delta glnA mutant. Expression of the cDNAs in E. coli is presumed to be due to the function of a polycistronic hybrid lac messenger RNA or translational fusions encoded by the pUC plasmids. Proteins of the expected sizes encoded by two different pUC clones were shown to react with antibodies to tobacco glutamine synthetase.

Abstract:
Coding sequences of DNA generate Open Reading Frames (ORFs) inside them with much higher frequency than random DNA sequences do, especially in the antisense strand. This is a specific feature of the genetic code. Since coding sequences are selected for their length, the generated ORFs are indirect results of this selection and their length is also influenced by selection. That is why ORFs found in any genome, even much longer ones than those spontaneously generated in random DNA sequences, should be considered as two different sets of ORFs: The first one coding for proteins, the second one generated by the coding ORFs. Even intergenic sequences possess greater capacity for generating ORFs than random DNA sequences of the same nucleotide composition, which seems to be a premise that intergenic sequences were generated from coding sequences by recombinational mechanisms.

Multiple sclerosis (MS) is an inflammatory autoimmune disease of the central nervous system (CNS) which typical feature is the plaques of demyelination. The factor triggering the disease is still unknown, but the most diffuse opinion is that the autoimmune reaction is due to a new contact with environmental factors, like virus, that mimic self molecules (molecular mimicry). MS is a T- cell mediated disease. These cells activated by myelin antigens activate, in turn, B lymphocytes, macrophages and cytotoxic lymphocytes which strongly damage oligodendrocytes and neurons, both directly and through complement activation, leading to an inhibition of nervous conductivity in CNS. In this project, MS is studied through an “epitomic” approach by means of the construction of epitope phage display libraries, obtained by cloning cDNAs derived from CNS tissue. These collections of billions of different phages displaying different polypeptides on their surface give the total representation of the CNS expression pattern and are useful for the selection against MS serum and CSFs, in order to identify potential autoimmune epitopes and the relatives auto-antibodies. The choice of phage display technology as tool to investigate MS autoimmune response depends on the possibility to couple the phenotype to genotype in a selectable format. Selection of the phages on the basis of the binding properties of the polypeptide displayed on the surface, simultaneously results in the isolation of the gene encoding the polypeptide. In order to obtain the epitope phage display libraries, two different versions of a new vector, called pEP1 and pEP2 respectively, for the selection of the cDNA fragments cloned in the correct open reading frame (ORF) were designed. A well characterized vector for the ORF selection, previously described by Zacchi and colleagues (2003), was used as backbone and modified in two alternative ways on its original polylinker to improve the cloning efficiency. In addition, for its oriented cloning system, pEP2 was able to give a greater ratio of “in frame”/ “out of frame” clones (1/9 rather than 1/18) than pEP1. After their validation, the new vectors were employed to generate two kinds of cDNA phage display libraries: pEP1 was tested with the construction of single myelin genes (myelin basic protein, MBP; and myelin oligodendrocyte glycoprotein, MOG) mini-libraries; while the more advanced pEP2 was used to create a total human brain library. A first question was related to the kind of epitope recognized by autoantibodies in MS. Generally, autoantibodies recognize both linear and conformational epitope, but at present the conformational epitopes of myelin proteins have not been identified. We have set a system to make minilibraries from single gene through the fragmentation of the gene of interest with nuclease protection by means of archibacteria histones. In this way it’s possible to obtain phage display libraries of overlapping fragments of the same gene useful to define the precise epitope recognized by a biological sample. For the project, human MBP and MOG mini-libraries were obtained and characterized through PCR analysis and enzymatic fingerprinting. By sequencing of 30 randomly picked clones for both mini-libraries, a set of overlapping fragments which gave a total representation of mbp and mog gene, was respectively found. MBP mini-library was subjected to two selections: against the serum and the CSF of a patient with a Clinically Isolated Syndrome (CIS), diagnosed after a first clinically episode of MS symptoms. Instead, MOG mini-library was selected against a Relapsing Remitting (RR) serum of a MS patient. To identify the more reactive clones, 95 clones from each last round of selection were randomly picked and subjected to a phage-ELISA test. Panning the hMBP mini-library with CIS serum and CSF, some distinct epitopes of the protein were identified, suggesting a specific pattern of recognition with some overlapping regions in both the CIS samples. Among the epitopes recognized, was identified the more immunogenic region of hMBP (a.a. 85-96). In the case of hMOG mini-library against RR serum, selected phage clones shared the epitope corresponding to a.a. 33-104 of the protein, which is localized inside the more immunogenic region of hMOG. A cDNA phage display library which should contain all the potential auto-antigens of the CNS is a useful tool to perform a serological analysis of MS sera and CSFs, in order to identify the epitopes involved in the immune response and to identify subclinical conditions of patients. With these purposes, a human brain phage display library was produced. Starting from human brain mRNA, a new protocol for the cDNA production was set. Employing the “OrientExpress Random Primers” (Novagen), an orientated synthesis of the cDNA was obtained. Following the ligation to the vector (digested pEP2), a human brain library of 1.35x10^5 individual clones selected for the presence of ORF inserts was produced. Through PCR analysis, enzymatic fingerprinting and sequencing of randomly picked clones a library diversity of 90% was estimated. The library was employed for three distinct selections: 1). against IgGs purified from a pool of RRMS sera; 2). against a pool of Secondary Progressive (SP) MS sera; and 3). against a pool of MS CSFs. From the last round of each selection test were respectively picked 190, 95 and 50 clones. These randomly picked clones were checked by phage-ELISA to identify those more reactive. The positive clones were respectively 34%, 16% and 50% on the total. Interestingly, some clones were shared among the selections and it was identified less clone diversity in the selection against CSFs pool. The reactivity of some selected clones, as phages, as recombinant protein or as synthetic peptides, was tested in ELISA in order to establish the sensibility and specificity in discriminating MS and other neurological diseases (OND). The ELISAs on "omissis patent pending" peptide resulted particularly predictive (LR+ 12.2) with a sensitivity and specificity of discrimination of 31.7% and 97.4%, respectively. The final aim of the project was in providing new tools to investigate the antibody response in MS and in the improvement of the currently used diagnostic tests through the detection of new potential markers of the disease. At the moment there are not validated biomarkers of MS. The new candidate autoantigens identified in this study could be one of those and integrate the current diagnostic method of MS.

Dissertation for the candidate degree in specialty 05.05.03 "Engines and Power Plants". - National Technical University "Kharkiv Polytechnic Institute ", Kharkiv 2017. The thesis is devoted to actual problem - the selection of schemes and power parameters of the power plant for utilization of associated gas. The problem of associated gas flaring continuously rising international conferences on conservation of the environment are held under the auspices of the UN and the World Bank. In particular, the World Conference on Climate in Paris (COP21) in 2015, and was nominated Global Initiatives to eradicate the practice of flaring of associated gas in the oil industry. Worldwide, it was supported by 45 oil companies, governments and other parties through which CO2 emissions can be reduced by 100 million tons per year. The adopted program "Zero Routine Flaring by 2030" provides end to the practice of burning of associated gas by 2030. This Initiatives supported and Iraq, which in 2015 take the second place in the world with the burning of associated gas in flares. Associated petroleum gas is 2% of product yield refineries in Iraq. Given the number of refineries and their power daily when they are flaring loss a lot of energy and is a significant pollution of the territory not only as chemical emissions but as a heat is released during the combustion of associated gas. The work uses a comprehensive approach to the selection circuit and power parameters power plant for utilization of associated gas. The possible options for the utilization of associated gas. According to the options two power plants are taken the first one based on gas turbine engine and the second based on gas turbine engine and a piston engine. The question examined in terms of exergic-anergy balance installation and obtain the best technical and economic performance, taking into account the climatic characteristics of Iraq The features of physical and chemical composition of associated petroleum gas in the refinery in Iraq, in particular methane determined the number, the method of firm Caterpillar. The methane number of the gas fuel affects the choice piston power plant. Significant impact on the choice of installing recycling schemes associated gas temperature features render the region. For their consideration set average temperature for the region. Conducted thermal calculations allowed to analyze the impact of environmental temperature on performance of power plants and conduct a feasibility study best selection circuit installation. Implementation exergic-anergy balance for power plants proposed scheme has allowed to confirm significant reduction in thermal pollution and show the most attractive from that perspective scheme. Economic calculations allowed to determine the payback period of the projects and installations prove the economic feasibility of their construction. The most economically attractive project. Similar calculations of sensitivity analysis performed for the two plants power plants. Based on the analysis, commissioned by the Iraqi side was developed based business project for energy generation capacity on the basis of energy utilization units. The results of the study will not only get the necessary electrical energy that can be used not only in the enterprise, but also to improve the environment in accordance with international agreements. The results of the research will be used in the construction of new units in the refineries in Iraq according to a letter from the Ministry of Industry and minerals.

Дисертаційна робота присвячена актуальному питанню - вибору схеми та параметрів силової енергетичної установки для утилізації попутного нафтового газу. Проблема спалювання попутних газів постійно підіймається на міжнародних конференціях зі збереження навколишнього середовища які проходять під егідою ООН та Всесвітнього банку. Зокрема, на Всесвітній конференції з клімату в Парижі (COP21) у 2015 році, була висунута та підтримана глобальна ініціатива з викорінення практики спалювання попутного газу в нафтовій промисловості. В усьому світі її підтримали 45 нафтових компаній, урядів та інших сторін завдяки яким викиди CO2 можуть скоротитись на 100 мільйонів тонн на рік. Прийнята програма “Zero Routine Flaring by 2030” передбачає покінчити з практикою спалювання супутнього нафтового газу до 2030 року. Цю ініціативу підтримала і держава Ірак, яка на 2015 рік займала друге місце у світі зі спалювання попутних газів у факелах. Попутний нафтовий газ складає 2% від виходу продуктів нафтопереробного підприємства в Іраку. Враховуючи кількість нафтопереробних підприємств та їх потужності, щоденно, при його спалюванні у факелах, втрачається велика кількість енергії та відбувається значне забруднення навколишньої території не тільки хімічними викидами а і теплотою яка виділяється при згорянні попутного нафтового газу. У роботі використано комплексний підхід до вибору схеми та параметрів силової енергетичної установки для утилізації попутного нафтового газу. Розглянуто можливі варіанти утилізації супутнього нафтового газу. Серед варіантів взято енергетичну установку на базі газотурбінного двигуна та установку на базі газотурбінного двигуна який діє сумісно з поршневим двигуном. Поставлене питання розглянуто з точки зору енерго-ексергетичного балансу установки та отримання найкращих техніко-економічних показників з урахуванням кліматичних особливостей регіону держави Ірак. Розглянуто особливості фізико-хімічного складу супутнього нафтового газу на нафтопереробному заводі держави Ірак, зокрема проведено визначення метанового числа, за методикою фірми Caterpillar. Метанове число газового палива впливає на вибір поршневої енергетичної установки. Значний вплив на вибір схеми установки з утилізації супутнього нафтового газу оказують температурні особливості регіону. Для їх урахування визначено середню температуру для регіону. Проведені теплові розрахунки дозволили проаналізувати вплив температури навколишнього середовища на показники енергетичних установок та провести економічне обґрунтування обрання найкращої схеми установки. Виконання енерго-ексергетичного балансу для запропонованих схем енергогенеруючих установок дозволило підтвердити значне зменшення теплового забруднення навколишнього середовища та вказати на найбільш привабливу з цієї точки зору схему. Економічні розрахунки дозволили визначити термін окупності запропонованих проектів установок та довести економічну доцільність їх побудови. Визначено найбільш економічно привабливий проект. Проведено аналіз економічних ризиків чутливості до зміни ціни електроенергії та до зміни температури навколишнього середовища. Подібні розрахунки аналізу чутливості проведено для заводів з двома енергетичними установками. На основі проведеного аналізу, на замовлення Іракської сторони, було розроблено основу бізнес проекту для енергогенеруючих потужностей на базі енергетичних утилізаційних установок. Виконання результатів дослідження дозволить не тільки отримати необхідну електричну енергію, яку можна використовувати не тільки на підприємстві, а і покращити стан навколишнього середовища у відповідності до міжнародних домовленостей. Результати дисертаційного дослідження будуть використані при будівництві нових енергоблоків на нафтопереробних заводах Іраку згідно листа від Міністерства промисловості і природних ресурсів.

The research object involves selection of mobile ICT for the training process. The re-search subject is selection of mobile ICT for learning informatics to future professionals in engineering pedagogy. The research results imply analysis of the national and foreign researches into mobile ICT for learning informatics. The latest publications concerning selection of mobile ICT for teaching Informatics subjects (Mobile Learning Management Systems, Mobile Modeling and Programming Environments, Mobile Database Management Systems, Mobile Multimedia Authoring Tools, Audience Response Systems) are analyzed. Informatics subjects are united into 19 groups, mobile ICT tools – into five groups. The experimental research is conducted according to the syllabuses for Speciality 015.10 “Professional Education (Computer Technologies)”. The expert assessment results for each of the content blocks of informatics subjects allow determining leading and auxiliary mobile ICT teaching tools.

Objectives: To accomplish stable expression, inheritance of transgenes and growth improvement in transgenic channel catfish, Ictalurus punctatus, and common carp, Cyprinus carpio, containing growth hormone (GH) genes, develop transgenic fish with all fish constructs, determine the relationships between copy number, expression and growth, determine the combined affect of selective breeding and gene transfer and assess environmental risk of transgenic fish. To develop mechanisms of triploidization for transgenic carp. Results: Performance of transgenic channel catfish was made uniform by selection. Growth of channel catfish and common carp was improved 40-50% more by combining gene transfer of GH genes with selection and crossbreeding than with either selection of crossbreeding. Growth improvement of transgenic catfish was not strongly correlated with copy number and expression levels. Progress was made in producting triploid transgenic common carp. Insertion of salmonid GH gene did not alter reproductive performance in channel catfish. Transgenic channel catfish grew no faster than controls when they had to forage on natural food and transgenic individuals were slightly more vulnerable to predation indicating that fitness of transgenic individuals in natural conditions is less than or equal to non-transgenic channel catfish. Contribution to Agriculture: These experiments are the first to demonstrate that transgenic fish can increase aquaculture production in the aquaculture production in the aquaculture environment. This research also demonstrated that maximum benefit of gene transfer in farmed fish is attained when combined with traditional selective breeding.

Advantageous (or propitious) selection occurs when an increase in the premium of an insurance contract induces high-cost agents to quit, thereby reducing the average cost among remaining buyers. Hemenway (1990) and many subsequent contributions motivate its advent by differences in risk-aversion among agents, implying different prevention efforts. We argue that it may also appear in the absence of moral hazard, when agents only differ in riskiness and not in (risk) preferences. We first show that profit-maximization implies that advantageous selection is more likely when markup rates and the elasticity of insurance demand are high. We then move to standard settings satisfying the single-crossing property and show that advantageous selection may occur when several contracts are offered, when agents also face a non-insurable background risk, or when agents face two mutually exclusive risks that are bundled together in a single insurance contract. We exemplify this last case with life care annuities, a product which bundles long-term care insurance and annuities, and we use Canadian survey data to provide an example of a contract facing advantageous selection.
