Abstract:
British Library manuscript Cotton Titus A.xix, a miscellany compiled in the fifteenth century, contains on folios 153r–155r an untitled Latin version of the legend of Roland with the colophon ‘Explicit de tradicione Guenonis.’ This text was first published in 1837 by Francisque Michel as an appendix to the editio princeps of the Chanson de Roland in the Oxford manuscript, with no commentary of any kind. In 1838 Wilhelm Grimm declared that the Latin version was a deliberate condensation of the legend and hazarded that it might have been composed in the twelfth century. In 1839 the Swiss scholar Johann Caspar von Orelli reprinted Michel's text in a classicizing orthography with occasional conjectural improvements, and dated the poem in the twelfth or thirteenth century on the basis of its resemblance to the Ysengrimus and the Speculum stultorum.

Abstract:
In this study, we evaluated the effectiveness of Fusarium caatingaense URM 6778 and URM 6779 alone and combined with aqueous extracts of Ricinus communis 5% and Nicotiana tabacum 10% against Dactylopius opuntiae in the field. The effectiveness of the extracts alone was also evaluated. The botanical extracts were subjected to phytochemical screening to identify biologically active chemical compounds. Total mortality data were corrected using the Schneider-Orelli formula. The results were submitted for analysis of variance (ANOVA), and the means were compared using Tukey’s test at 5% probability. The phytochemical screening of R. communis and N. tabacum showed that both extracts contained alkaloids, flavonoid glycosides, saponins, and tannins. According to the confirmed mortality, URM 6779 (61.67%) and URM 6778 + aqueous extract of R. communis at 5% (43.89%) were the best treatments for the control of D. opuntiae in the field. The aqueous extracts of N. tabacum at 10% and R. communis at 5% resulted in mortality rates of 75.14% and 65.32%, respectively, and are also options for controlling this insect. Despite promising results, for the applicability of such agents to be safe, investigations into their toxicity to non-target organisms and their permanence in the environment are necessary.

Abstract:

Abstract:
Flower thrips represent a complex of significant quarantine species affecting the cut flower market in Colombia. The aim of this research was to evaluate postharvest treatments using phosphine in conjunction with a plant extract for thrips control and quality control of five cut flower species. Eight treatments were used: six employed a commercial dose of magnesium phosphide as a source of phosphine, one used a double dose and a control group without phosphine application. The first six treatments followed a bi-factorial structure, incorporating three exposure times and the addition of a chili-garlic extract. Thrips control efficacy was evaluated using the Schneider-Orelli index based on field-collected samples. Postharvest quality assessments were conducted on roses, carnations, alstroemerias, chrysanthemums, and hydrangeas over an 18-d period following treatment application. Differences in efficacy were observed between the two locations (the blocking factor). Discrepancies in phosphine efficacy may be related to the variations in populations collected from different crops and locations, both in the departments of Cundinamarca and Antioquia. Variations in magnesium phosphide concentration, both at the commercial dose of 3.4 g m-3 and double this amount (2X) did not produce significant differences in treatment efficacy or flower quality. The use of chili pepper and garlic extract applied by nebulization at 3°C combined with phosphine application also did not significantly affect thrips mortality efficacy. The factor most influencing efficacy improvement was exposure time, as longer time periods led to better thrips control. Furthermore, we found that longer exposure times did not affect visual quality or vase life, assessed through changes in color, physiopathies, and chlorophyll content.

Abstract:
Thegreen-belly stink bug, Dichelops melacanthus, stands out as one of the main pests of the corn crop, mainly for its potential damage at the beginning of crop development, due to the sap suction at the base of seedlings, causing tillering and trimming symptoms, decreasing productivity and product quality. Aiming at the control of thestink bug, the objective of the present research was to evaluate the effect of insecticides, as well as the lethal time(LT50) of these products on D. melacanthus. To perform the tests, it was used third instar nymphs and adults of D. melacanthusaged up to 72 hours, which were individualized in Petri dishes. For the application of the insecticide, corn leaves were collected and placed in plastic trays. With the aid of a pressurized spray, the insecticides Acefathe + Aluminum silicate (5 g/L), Thiamethoxam + Lambda-Cyhalothrin (1 mL/L), Imidacloprid + Bifenthrin (1.75 mL/L), Bifenthrin + Zeta-Cypermethrin (0.75 mL/L) and Azadiractin (10 mL/L) were applied. In the control, only distilled water was applied. Subsequently, the pulverized leaf pieces were individualized in the Petri dishes, where the third instar and adult bedbugs were already allocated. After application, each treatment was evaluated at 15, 30 minutes, 1, 3, 5, 24 and 48 hours, accounting for insect mortality. Tocorrect mortality, the values were transformed using the Scheneider-Orelli formula. The relationship between the periods of action of the insecticides on the mortality rate of insects was verified by nonlinear regression analysis, using the mathematical model of dose-response. All chemical insecticides evaluated were effective in controlling third instar nymphs; however, when applied to adults, Thiamethoxam + Lambda -Cyhalothrin and Acefathe + Aluminum silicate had reductions in bedbug mortality. Azadiractin showed low efficiency in controlling nymphs and adults of D. melacanthus. Insecticides showed better LT50 in nymphs when compared to adults. The insecticides Imidacloprid + Bifenthrin, Thiamethoxam + Lambda -Cyhalothrin and Bifenthrin + Zeta-Cypermethrin obtained the best LT50 responses for nymphs and adults of the green-belly stink bug. All chemical insecticides evaluated can be used in the integrated management of D. melacanthus.

Abstract:
This work aims to study the forms and functions of landscape in Italian poetry of the second half of the twentieth century and the early 2000s. The aim of the research is also to propose a methodological hypothesis for the study of a transdisciplinary object in the Italian literary horizon. With these intentions, the project seeks to place the landscape, understood as a particular unit actively constructed by a subject-observer, at the centre of an investigation designed to show how the gaze contributes to the construction of the poetic text. Given the historical, subjective and artificial nature of landscape, the study aims to identify its particular characteristics within a culturally circumscribed horizon from a formal, temporal and spatial point of view, by examining a corpus of significant works belonging to the Italian poetic production of the second half of the twentieth century. In particular, the corpus includes texts by Giorgio Orelli and Andrea Zanzotto - poets born in the 1920s - and those by Antonella Anedda and Fabio Pusterla, born in the 1950s

Abstract:
Rural settlements are formed according to local people&rsquo
s needs, harmonious with the land form and the climate, and have a rich variety of buildings using local material. But changes in lifestyle due to urbanization and tourism, socio economic degradation and poor living conditions have a negative effect on rural heritage and as a result, rural settlements become inharmonious with vernacular architecture and current lifestyle. Besides, due to its geographical and quantitative vastness, rural heritage is rarely recorded and legislations for rural settlements are so inadequate in Turkey. The aim of this thesis is to understand the values of rural heritage and suggest proposals for the conservation of these areas. As a case study Ö
renli Village in Kepsut, Balikesir was selected and architectural characteristics of traditional buildings, lifestyles of local people in Ö
renli Village, written and unwritten rules of this rural settlement were analyzed. In conclusion, recommendations were done for conserving rural heritage while trying to meet the local people&rsquo

Abstract:
Introduction: The distinctive anatomic features and topography render the ear unique in the human body. Total or partial reconstruction of the ear may be required in many clinical conditions, but because technical difficulties are common, the rate of postoperative complications increases. Objectives: To analyze the epidemiologic data of patients who underwent surgery for reconstruction of the ear due to congenital conditions (microtia) or acquired deformities (trauma, burns and others), and to compare the results in order to define which group had the lowest rate of complications. Methods: A retrospective analysis was conducted with 279 cases of ear reconstruction performed between 1994 and 2004 by the Discipline of Plastic Surgery of the University of São Paulo Medical School. The patients were initially separated in two groups, according to their condition (congenital or acquired), to compare their data. Results: Male was the prevailing gender in both groups of ear deformities: congenital (61.3%) and acquired conditions (68.7%). The patients with congenital deformities had a mean age of 14.3 years at the beginning of the treatment, whereas the patients with acquired deformities were 29.5 years old, in average. The major causes of acquired deformities were trauma (55% of the cases in this group) and burns (29%). The only cause of congenital deformity observed was microtia. Patients required an average of 4.2 surgical procedures. However, those with sequelae of burn injuries were submitted to a significantly higher number of procedures (5.9 - p < 0.01). Cartilage exposure (15.1% of the total) and postauricular bridles (16.5%) were the major complications observed in this study. The latter was more common among those cases with microtia and sequelae of burns. Conclusions: Patients with traumatic injuries had a better outcome after surgery than those with microtia or burn injuries, because of a lower rate of complications.

Chadha, Vipul. "Oligo-1,6-Glocosidase enzyme of halothermothrix orenii." Thesis, Griffith University, 2012. http://hdl.handle.net/10072/365799.

Full text
Abstract:
Halothermothrix orenii, a gram negative strictly anaerobic thermohalophilic bacterium has been isolated from sediments of a Tunisian salt lake (Chott El Guettar). H. orenii grows optimally at 60˚C with 10% NaCl. Phylogenetically, H. orenii is placed at the base of phylum Firmicutes, in the family Haloanaerobiaceae, order Haloanaerobiales, and is the only member of this order whose genome has been completely sequenced. The availability of the genome sequence will not only enhance our understanding of survival and adaptation strategies of microbes living under dual extremities of high salt and temperature, but will also provide information on novel biocatalysts which could have potential application in research, industrial and bioremediation processes. To date, our laboratory has studied a large number of genes that encode Glycosyl Hydrolases and Transferases from H. orenii, including AmyA and AmyB of the α-amylase family 13, a sucrose phosphate synthase (SPS) and a Fructokinase (Frk). Biochemical analysis demonstrated that all these recombinant enzymes are optimally active between 55-65˚C and in addition, the extracellular amylases AmyA and AmyB are most active with NaCl concentration between 5-25%. All the recombinant enzymes have been crystallized and their structures solved, but their molecular basis of thermohalophilicity has yet to be determined. In the present study, data mining of the genome of H. orenii further identified six potential α-amylase coding genes (designated Amy1, Amy2, Amy5, Amy6, Amy7 and Amy8). Primers were designed for cloning and overexpression of these genes into two vectors, namely flexi pFN6A and pET22b (+). Clones of Amy1, Amy6, Amy7 and Amy8 were discarded from the freezer by a member of the plant laboratory in my school, and therefore only clones Amy2 and Amy5 were available for studies. Amy5 clone has been studied in detail and forms the basis of this thesis. The Amy5 gene, which consists of 1701 nucleotides, was determined by BLAST analysis to code for an oligo-1,6-glucosidase, was cloned in pET22b (+) vector and the recombinant enzyme successfully over-expressed in the E. coli B strain (BL21 (DE3)). The recombinant enzyme (designated rAmy5) was purified to homogeneity using immobilized metal affinity chromatography (IMAC). The molecular weight of rAmy5 was determined by SDS to be 66kDa, which is close to the expected theoretical calculated value of 66.766kDa. The rAmy5 hydrolysed p-nitrophenyl-α-D-glucopyranoside (PNPG). The temperature optimum for enzyme activity was 60˚C (temperature activity range between 45-65˚C) and the pH optimum was between pH 6 and 6.5 (pH activity range between pH 5.3-7). rAmy5 activity was inhibited by metal ions in the order Cd 2+ > Cu2+ > Zn2+ > Fe3+ > Fe2+ > Ni2+ > NH4+, but enhanced by Ca2+ and Mn2+ with Mg2+ and Co2+ having no effect. The recombinant enzyme was tolerant to proteolysis when tested in presence of various concentrations of Trypsin. DTT (Dithiothreitol) had no effect on enzyme activity suggesting that cysteine residues did not play any role in providing thermostability to the enzyme molecule. Activity of the enzyme against PNPG was completely inhibited by 100mM D-glucose or 0.2mM PNP (p-nitrophenol). EDTA was found to have no effect on enzyme activity. The enzyme activity was enhanced in the presence of salts such as 0.5M NaCl and 1M KCl by 22.5% and 45% respectively. Denaturants inhibited enzyme activity in the order SDS > Tris > Urea. Sugars inhibited the enzyme activity in the order D-glucose > D-fructose > Mannitol > Xylose > Cellobiose > Lactose > Cellobiose > Lactose > Mannose > Raffinose > Galactose. The enzyme displayed no activity when tested against the following PNP-based glycosides: 2-Nitrophenyl-β-D-fucopyranoside, p-Nitrophenyl-α-L-arabinopyranoside, p-Nitrophenyl-α-D-xylopyranoside, p-Nitrophenyl-β-D-xylopyranoside, 2-Nitrophenyl-β-D-glucopyranoside, p-Nitrophenyl-β-D-fucopyranoside, 2-Nitrophenyl-1-thio-β-D-galactopyranoside, and p-Nitrophenyl-β-D-glucuronide. The activation energy of enzyme was found to be 81.3kJ/mol. The enzyme was thermostabilized at 60˚C in the presence of salts and combination of metal ions in the following order: 0.5MNaCl > 0.5M NaCl + 25mM CaCl2 > 1M KCl > 1M KCl + 25mM CaCl2. Presence of 25mM CaCl2 alone was found to destabilize the enzyme at 60˚C. The half-life of the enzyme increased four times in presence of 0.5M (10%) NaCl at 60˚C. The enzyme followed Michaelis Menten kinetics for PNPG and was found to have a KM and VMAX of 1.3mM and 333.3mM PNPG/min per mg of enzyme respectively. The enzyme had a high specific activity and was found to be active against a variety of substrates. The probable order of its specificity was PNPG > dextrin > dextran > isomaltotriose > amylose > pullulan > palatinose > soluble starch > trehalose > maltotetraose > maltose > isomaltose > maltooligosaccharide > panose. The enzyme was more active against substrates containing (1→6)-α-D-glucosidic linkages. In silico analysis revealed that the Amy5 putative protein sequence consisted of a highly conserved ‘QpDln’ signature for oligo-1,6-glucosidase sub-family. The high thermostability of the rAmy5 enzyme could be correlated to an increased presence of prolines and preference for glycine over alanine in the Amy5 sequence. A preference for charged residues over uncharged residues indicated the possible reason for the increased thermostability with NaCl of the rAmy5 enzyme. The fact that the Amy5 gene sequence had a significant identity (63.7%) to the sequence of solved crystal structure of oligo-1,6-glucosidase (1UOK) from Bacillus cereus formed the basis for a comparative homology modelling of Amy5 protein and as result a 3-Dimensional model of the Amy5 protein could be predicted. Comparative analysis of the predicted Amy5 model with 1UOK revealed that the residues of deep catalytic cleft of the latter were found at the same positions (Asp199, His 328, and Asp329), except that the Glu255 residue of 1UOK aligned with position Glu256 of Amy5. These catalytic residues are considered necessary for enzyme function rather than enzyme substrate specificity. Study of crystal structures of various enzymes has revealed that there are various subsites (amino acid residues) which associate with catalytic residues to impart the character to the enzyme to act on specific substrates. In addition to the present study of comparative modelling and biochemical characterization of the rAmy5 enzyme, protein crystallography studies could add to our understanding of structure-function relationship. In our present study, an attempt to crystallize the rAmy5 protein was made using a hanging drop method by screening it against 240 different buffer conditions (using JBScreen Mixed Crystallography kit, Jena Biosciences, Germany), but crystals were not observed during the three-month incubation period.

Abstract:
INTRODUCTION: Children with Down syndrome have high incidence of disorders of the middle ear, as well as congenital abnormalities in the external, middle and inner ear. Such conditions may change the sound transmission through the ossicular chain to the cochlea and central auditory pathways, impairing the comprehension of spoken language. An recent assessment tool of the middle ear, the wideband reflectance energy, can measure the sound energy reflected or absorbed in the ear canal over a wide frequency range, faster than tympanometry. OBJECTIVE: To compare the wideband reflectance measurements between children with Down syndrome and a control group matched by age, according to the tympanometric findings. METHODS: This study evaluated four groups with Down syndrome, between 28 and 195 months: normal tympanogram (19 ears), flat tympanogram (13 ears), moderate negative pressure tympanogram (6 ears), severe negative pressure tympanogram (4 ears). All findings were compared to a control group (21 ears). The subjects underwent to tympanometry and acoustic reflex ipsilateral with a 226 Hz probe tone frequency, pure tone audiometry, speech recognition thresholds, transient otoacoustic emissions and wideband reflectance over the 200-6000 Hz range with chirp and tone stimuli. RESULTS: Results revealed that the five groups had different reflectance curves, without significant difference in some comparisons. There was a negative correlation between the volume of the external auditory meatus and the wideband reflectance with the chirp stimulus between 250 and 1600 Hz and with the tone stimulus between 258 and 1500 Hz, and between the static acoustic admittance at the tympanic membrane level and the wideband reflectance with chirp stimulus at 500 and 1000 Hz. There was no correlation between the frequency of the chirp stimulus with reflectance at 1000 and 2000 Hz and the results of the otoacoustic emissions at 1000, 2000 Hz and general response. The discriminant analysis technique used to classify participants\' data based on the values of reflectance with chirp stimulus in 1000 and 1600 Hz, achieved a correct classification rate of 60% for participants with Down syndrome. CONCLUSIONS: The findings showed the responses of the middle ear, for different tympanograms in Down syndrome through the wideband reflectance. In this study, children with Down syndrome and normal tympanogram showed wideband reflectance curve similar to the control group. A discriminant analysis with the tympanometric results and reflectance with chirp stimulus at 1600 and 1000 Hz correctly classified 60% of the data for children with Down syndrome

Abstract:
Enzymes secreted by extremophiles are termed as extremozymes. Extremophiles are organisms that inhabit conditions which are extreme to survive and adapt in. Halothermothrix orenii is a polyextremophilic anaerobic bacterium that grows optimally at 60 C (maximum 70 C) in presence of 10 % NaCl (NaCl growth range between 4-20 %) and optimum pH 6.5-7.0, isolated from the sediments of Tunisian salt lake (Cayol et. al., 1994). This bacterium carry exclusive features from several points : (a) It is Gram-negative member of phylum Firmicutes and its genome analysis reveals mixed Gram-negative and Gram-positive properties such as lipid A biosynthesis, a characteristic of Gram-negative pathway and a typical Firmicutes sporulating system. (b) It is a strict anaerobic chemoorganotroph which uses various polysaccharides and produces a series of intracellular and extracellular glycosyl hydrolases and transferases. (c) It is a halothermophile and any extra-cellular enzymes produced by this organism are well adapted to function under thermophilic and saline conditions. (d) Halothermothrix orenii survives under fluctuating environmental conditions – dilution of salts during rains when temperature is low and increased salt concentration during summer when temperature is at its peak. In this study various glycosyl hydrolases and transferases from H. orenii selected for investigations are briefly described in chapter 3. Genes were cloned and over-expressed followed by protein purification were further analysed for their biochemical characteristics along-with structural insights.

Abstract: Ear is the sound receptor in our body. Sound is captured and driven through external ear, amplified and modulated in middle ear and transduced to electrical sign in internal ear. However, otoacoustic emissions and objective tinnitus are examples of an inverse process, where ear can be a sound emitter. Trying to understand some of theses events, in which sound emission occurs, the physiology of hearing and the physical basis to the sound propagation through the ear was revised. It was used as main tool, articles of Web of Science, from the last 25 years. From this point, it was developed a theoretical model to the way of sound, generated inside cochlea that propagates in tympanic membrane direction. It was performed mathematical calculations to sound attenuation, caused by the mechanical inertia of oval window - ossicles - tympanic membrane structure. For that, it was considered the area ratio between oval window and tympanum and the lever action ossicular chain. We concluded that sound generation by electromotility of outer hair cells inside cochlea, is attenuated in 20 times when it passes through ossicular chain in the inverse way. Besides this lost in sound energy, it was also demonstrated that in perilymph-oval window interface sound transmission is considerable (70%), because there are not big differences between the acoustic impedances of involved structures. Then, in physical basis, it was shown how ear can emit sound from contraction of outer hair cells

Abstract:
Otitis media (OM) is one of the leading causes of hearing loss in childhood. Due to its high prevalence and worldwide distribution, OM is still regarded as a public health issue. Nevertheless, few studies have assessed involvement of the contralateral ear (CLE) in patients with chronic otitis media (COM). To demonstrate the bilateral nature of COM, we designed three distinct studies, which focused on histopathology and anatomy, clinical findings and physiology (both previously published), and, finally, auditory function. Objective: To measure auditory function in the CLE in a sample of COM patients by means of air-conduction thresholds, bone-conduction thresholds, and the air-bone gap, and to compare CLE audiometry findings in patients with non-cholesteatomatous (NC-COM) or cholesteatomatous chronic otitis media (CCOM). Methods: This was a cross-sectional, contemporary and comparative study of 1000 patients with a diagnosis of COM. Clinical ear examination, video otoscopy, and pure-tone audiometry (PTA) were performed. Results: The overall prevalence rate of air-bone gap was approximately 40%, both in the total sample and after stratification by type of OM (393 [39.2%] patients overall, 279 [40.3%] in the NC-COM group, 113 [37.17%] in the CCOM group). Conclusions: As did clinical and histopathology findings in previous studies, audiological evaluation revealed a substantial prevalence of CLE involvement, providing evidence of the bilateral nature of COM.

Abstract:
