Abstract:
Avaliou-se, durante quatro estações de monta (1/11 a 31/1 do ano seguinte), o efeito da ordem de parto (OP) e da condição corporal (CC), segundo escala de 1 (magra) a 5 (gorda), sobre o desempenho reprodutivo de 468 fêmeas Nelore, sendo 391 vacas multíparas e 77 primíparas, em pastagem de Brachiaria decumbens Stapf, na região dos Cerrados, no Brasil Central. A OP teve efeito quadrático na taxa de prenhez (TP). Vacas OP1 apresentaram TP de 69%, OP5 a OP8 de 90%, com declínio gradual até 80% de prenhez na OP12. A CC à desmama afetou a TP. Pela análise de regressão, vacas OP1 com CC 2,0 e 3,5 tiveram TP de 52,7 e 82,5%, respectivamente. Observou-se TP DE 96% em vacas com OP4 e OP8 e CC 3,5. A TP de vacas OP1 com parição tardia foi de 37,7%, mas, independentemente da OP, vacas que pariram no início da temporada tiveram TP superiores a 80%. Vacas OP1 pariram 350,12 dias após o início da monta, enquanto aquelas com OP superiores, necessitaram de 328,32 dias. Vacas OP1 apresentaram o mais longo intervalo de partos (IP), com 392,10 dias, ao passo que o IP das OP5 a OP9 foi de 365 dias. O IP foi afetado pelo ano, pela OP, pelo número de dias necessários para parir na estação de parição e pela variação de peso na estação de cobrição. O peso vivo e a CC das vacas à desmama foram afetados pela OP e pelo ano. O peso à desmama (PD) do bezerro aumentou da OP1 até a OP4/OP5, de modo que as vacas OP1 proporcionaram PD de 159 kg e a média dos PD das OP foi de 169 kg. Altos índices produtivos e reprodutivos são obtidos entre OP3 e OP8 com CC acima de 3,0 e 3,5 em vacas Nelore multíparas e prímaparas, respectivamente.

Abstract:
The successful of genomic DNA amplification using RAPD technique was determined by the sequence of primer’s base and primer compound in each reaction. The aim of research was finding the best primer for genetic diversity analyses of Bitti (Vitex coffassus). The results of amplification showed that number of band between one to five bands (110-600 bp). The primer of OPK-10, OPA-17, OPQ-07 and OPP-08 can be used for genetic diversity analyses of Bitti. The best primer used for genetic diversity analyses of Bitti was OPP-08 because it had the highest number of bands (5 bands). Key words: OPP-08, Primer Selection, Vitex coffassus

Abstract:
To quantify the excellence of multimedia quality, subjective evaluation experiments are conducted. In these experiments, the tradition of quantitative assessment is the most dominating, but it disregards the understanding of participants' interpretations, descriptions, and the evaluation criteria of quality. The goal of this paper is to present a new multimedia quality evaluation method called Open Profiling of Quality (OPQ) as a tool for building a deeper understanding on subjective quality. OPQ is a mixed method combining a conventional quantitative psychoperceptual evaluation and qualitative descriptive quality evaluation based on the individual's own vocabulary. OPQ is targeted for naïve participants applicable to experiments with heterogeneous and multimodal stimulus material. The paper presents the theoretical basis of the development of OPQ and overviews the methods for audiovisual quality research. We present three extensive quality evaluation studies where OPQ has been used with 120 participants. Finally, we conclude further recommendations of use of the method in quality evaluation research.

No Cerrado brasileiro, há uma grande diversidade de cores, tamanhos e aromas de frutos em acessos silvestres de P. edulis. Estes acessos também são importantes fontes de resistência a doenças, podendo ser incorporados em programas de melhoramento genético do maracujazeiro azedo. Neste trabalho, objetivou-se estimar a variabilidade genética existente em acessos silvestres e comerciais de P. edulis utilizando-se de marcadores RAPD. O DNA genômico de cada acesso foi extraído e amplificado com treze iniciadores decâmeros (OPD-04, OPD-07, OPD-08, OPD-16, OPE-18, OPE-20, OPF-01, OPF-14, OPG-05, OPG-08, OPH-04, OPH-12 e OPH-16) para a obtenção dos marcadores RAPD. Os marcadores obtidos foram convertidos em uma matriz de dados binários, a partir da qual foram estimadas as distâncias genéticas entre os acessos e realizadas análises de agrupamento e de dispersão gráfica. Um total de 187 marcadores foi gerado, sendo que apenas 28 (14,97%) deles foram monomórficos. As distâncias genéticas entre os 15 acessos de maracujazeiro variaram de 0,091 a 0,496. Os marcadores moleculares demonstraram a alta variabilidade genética dos acessos de P. edulis, sendo que os acessos de frutos amarelos apresentaram maior distanciamento em relação aos de frutos roxos. Menores distâncias genéticas foram verificadas entre os acessos de mesma origem geográfica.

OPQ is a company engaged in the automotive manufacturing industry. The company produces bolts for the safety area, it is related to the lower body of the four-wheeled vehicle. PT. OPQ was established in 2014 and today has already been the supplier for several major automotive companies in Indonesia. The demand of PT OPQ that must be fulfilled is increasing as the amount of consumer is increasing too. Due to the high demand, so the raw material stockout occurs. This make the company's service level is below the standard. This study involved 48 sku of material raw materials that have normal distributed demand. Inventory policy applied in this research is periodic review (R, s, S) with the aim to minimize total cost of inventory and improve service level of company. The result of this research was found to decrease total inventory cost by 15% with the value of existing inventory cost Rp1.100.879.479,- to Rp 936.061.349,- . In addition, service level at PT. Modern electrical grids are transitioning from a centralized generation architecture to an architecture with significant distributed, intermittent generation. This transition means that the formerly sharp distinction between energy producers (utility companies) and consumers (residences, businesses, etc.) are blurring: end-users both produce and consume energy, making energy management and public policy more complex. The goal of the Open Power Quality (OPQ) project is to design and implement a low cost, distributed power quality sensor network that provides useful new forms of information about modern electrical grids to producers, consumers, researchers, and policy makers. In 2019, we performed a pilot study involving the deployment of an OPQ sensor network at the University of Hawaii microgrid for three months. Results of the pilot study validate the ability of OPQ to collect accurate power quality data in a way that provides useful new insights into electrical grids.

As a result, the genetic diversity of R. microsporus that had been reported in Indonesia has diminished. Information about genetic diversity is needed as a basis to select R. microsporus as tempeh inoculum. This research aims to investigate the genetic diversity of R. microsporus from waru leaves based on Internal Transcribed Spacer (ITS) Sequence and Random Amplified Polymorphic DNA (RAPD) markers. A total of 25 R. microsporus were isolated from traditional inoculum waru leaves (Inoculum 1) and traditional inoculum other than waru leaves (Inoculum 2). Amplification of ITS sequence was done using universal primer pairs of ITS-4 and ITS-5. Amplification of RAPD markers was done using primers OPC-08, OPC-19, OPQ-6, R-108, OPA-09 and OPJ-20. ITS sequence was not sufficient to compare the similarities among R. microsporus. On the other hand, RAPD markers successfully compared the similarities among 25 R. microsporus. A total of 25 R. microsporus were divided into 9 clusters. R. microsporus from Inoculum 1 grouped into Cluster 1, Cluster 3 and Cluster 4-8. Inoculum 2 grouped into Cluster 2 and Cluster 9. R. microsporus from tempeh grouped into Cluster 4 and was different from Inoculum 1 and Inoculum 2, except for TB3.

Jinten hitam (Nigella sativa) is one of the medicinal plants in the family Ranunculaceae, native to the Mediterranean region and several regions of Asia. The seed of Nigella sativa is widely cultivated, distributed, and used in pharmaceuticals, cosmetics, and food industries. Due to similar seed morphology to its potential adulterant such as Corchorus spp., N. sativa seeds are susceptible to adulteration and substitution in markets. Molecular markers have become one of the most reliable methods for the identification and authentication of medicinal plants. The objective of this study was to select the random amplified polymorphic DNA (RAPD) primer for generating authentication methods of Jinten hitam (N. sativa). Genomic DNA was extracted from samples of N. sativa seed, Corchorus sp. seed, and a mixture of both samples. Forty-two random RAPD primers were used in this study. A total of 227 DNA fragments were produced from 37 amplified RAPD primers, out of which 65% were polymorphic. Primer OPK-4 and OPC-12 generated specific fragments in N. sativa, meanwhile, Primer OPB-1, OPL-5, OPM-3, OPD-5, and OPC-12 generated specific fragments for Corchorus. RAPD molecular marker was able to authentication Jinten hitam (N. sativa) and Corchorus sp. using a selected primer. This research was the first report on RAPD primer screening for Nigella sativa authentication from its potential adulterant (Corchorus spp.).

Thirteen randomly amplified polymorphic DNA (RAPD) primers were assayed in 82 Spanish olive cultivars of economical interest. A total of 82 bands were scored giving an average of 6.3 bands per primer. A total of 4 (OPA-01) to 10 bands (OPA-19) was amplified, while the number of polymorphic fragments ranged from 2 (OPK-07) to 9 (OPA-19) with a mean of 5.7 polymorphic bands per primer. A total of 89% of the amplification products (73 bands) were polymorphic. The 13 primers yielded 184 banding patterns (14.9 per primer). The number of banding patterns per primer ranged from 4 (OPK-07) to 39 (OPA-19). Fifty-three unique banding patterns were found, the majority of them resulted from different combinations of polymorphic bands. The combination of only five primers OPA-19, OPF-06, OPX-01, OPX-03, and OPI-12, allowed identification of all the cultivars. Seventy-four cultivars (90%) were identified only by the combination of the first four primers. The addition of the fifth primer (OPI-12) was necessary for the identification of the eight remaining cultivars (10%). The ordination of the primers according to their practical discriminating capacity in this study was: OPA-19 > OPF-06 > OPX-01 > OPX-03 > OPI-12. Hence RAPD markers are recommended for olive fingerprinting in order to generate a database for olive cultivar identification.

Objectives: to evaluate the Saville and Holdsworth Occupational Personality Questionnaire (OPQ) to assess its potential and validity as a tool in the recruitment of doctors. To determine the range of personality characteristics in a group of general practitioners. Design: an administered commercially available questionnaire. Setting: general practitioners in North and Mid. Stafffordshire. Subjects: 133 general practitioners or trainees. Results: 44% of general practitioners agreed to participate. The majority found it useful and recognised its potential to increase self-awareness, highlight weaknesses that might be improved and for future recruitment of others to their teams. Doctors' personality traits were similar to those of managerial/professional norms. The validity of the OPQ was supported by (a) significant associations with subjects' previously reported mental health problems and (b) 83% of respondents confirming that their reports correctly described their personalities at work. Conclusions: OPQs have a place in the recruitment of doctors by identifying appropriate people to strengthen the team.

The use of cultivars that are resistant to a particular disease is one strategy that could mitigate the incidence of gall rust disease on Falcataria moluccana. Previous studies on the genetic diversity of F. moluccana did not attempt to link that genetic diversity to gall rust disease resistance. This research was carried out using RAPD analysis to determine the preliminary information on the association between different markers and the resistance to gall rust disease. The analysis evaluated a total of 20 pairs of healthy and infected F. moluccana trees that were classified based on their disease severity level. The RAPD primers used in this study were as follows: OPA-05, OPA-08, OPA-10, OPA-13, OPA-18, OPB-07, OPD-13, OPF-02, and OPG-05. The results showed that each RAPD primer produced a varying number of polymorphic bands, ranging from 3 to 12 bands, with a total of 80 polymorphic bands. Despite the number of loci analyzed, however, no specific polymorphic bands were found that could distinguish between healthy and diseased trees. This was supported by principal component analysis, which showed that healthy and diseased populations were not distributed separately. The structure analysis also showed that the healthy and diseased populations were not different.

In their critique of our erratum and addendum (Anderson & Ones, 2008), Goldberg, Lee, and Ashton (2008) allege clerical errors (1) where participants' HPI scores were incorrectly matched to other inventory scale scores and (2) in scoring of the OPQ and BPI scales. The first point was fully addressed by our erratum and addendum and Goldberg et al. do not present any new evidence that was not already considered. In this response, we further demonstrate that Goldberg et al. do not and cannot prove clerical errors in scoring OPQ and BPI scales. The circumstantial arguments presented by Goldberg et al. are shown to be fallacious. We respond to all conjectures and criticisms raised by their paper. We acknowledge, as we did in our previous publication (Anderson & Ones, 2003) that some of the specific convergent correlations and findings reported are not normative, but nonetheless explainable given psychometric and statistical artefacts that influence individual primary study results. Even basing our analyses upon the ‘reconstituted’ dataset generated and preferred by Goldberg et al., we demonstrate that our original conclusions do not change substantially. Responsible science requires the publication of all results, not only those confirming extant hypotheses and expectations. We continue to stand by the findings and conclusions reported in our previous publications (Anderson & Ones, 2003; Ones & Anderson, 2002). Copyright © 2008 John Wiley & Sons, Ltd.

<p>Informasi keragaman genetik dan ketersediaan plasma nutfah teh (Camellia sinensis) diperlukan dalam perakitan varietas unggul. Keragaman genetik berdasarkan penanda DNA dapat memberikan hasil yang lebih konsisten karena tidak dipengaruhi lingkungan. Dalam penelitian ini sebanyak 9 genotipe teh dianalisis keragamannya menggunakan enam penanda RAPD (OPA 03, OPA 05, OPB 04, OPB 06, OPC 06, dan OPD 08). Penelitian dilakukan mulai bulan Maret sampai Mei 2013 di Laboratorium Terpadu Biotrop Bogor. Perhitungan koefisien kesamaan genetik dan analisis gerombol dilakukan dengan menggunakan perangkat lunak NTSYSpc versi 2.02. Sebanyak 54 lokus penanda RAPD berhasil diamplifikasi menggunakan enam primer dan 47 lokus di antaranya memiliki alel yang polimorfik (87,04%). Hasil analisis gerombol berdasarkan kesamaan genetiknya mengelompokkan 9 genotipe ke dalam enam kelompok. Empat kelompok (I, II, IV, V) masing-masing terdiri atas satu genotipe, sementara dua kelompok yang lain yaitu kelompok III dan VI masing-masing beranggotakan tiga dan dua genotipe.</p><p>Kata Kunci: Camellia sinensis, diversitas genetik, penanda RAPD</p><p>The availability of diverse tea (Camellia sinensis) germplasms as well as the information about their genetic diversity is required for plant breeding program. Genetic diversity analysis based on DNA marker is known to be more effective since the markers provide more consistent results. In this study, nine tea genotypes were evaluated for their genetic diversity using six Random Amplified Polymorphic DNA (RAPD) markers (OPA 03, OPA 05, OPB 04, OPB 06, OPC 06, and OPD 08). The study was conducted from March to May 2013 in the Integrated Laboratory of Biotrop Bogor. The estimation of genetic similarity and the cluster analysis were done using NTSYSpc version 2.02. Of the six RAPD markers used in this study, a total of 54 RAPD marker loci have been successfully amplified. Delapan primer yang digunakan adalah OPAI-15, OPB-08, OPAB-14, OPF-13, OPB-10, OPC-05, OPH-15, OPC-08 untuk mengidentifikasi 6 sampel (2 induk untuk batang atas dan batang bawah) dan 4 sampel. sampel dari okulasi bibit. Hasil penelitian menunjukkan dari 6 sampel dan 8 primer yang digunakan untuk identifikasi, sebagian besar sampel hasil perbanyakan cangkok menunjukkan karakteristik batang bawah daripada karakteristik batang bawah. Hasil penelitian juga menunjukkan bahwa 8 primer dari penelitian ini dapat digunakan untuk identifikasi DNA tanaman durian.

Rice is a cereal crop that belongs to the graminae family of high economic value, and is the main food for more than a portion of the world's population. This study aims to determine the relationship genetic of rice in Kampar by RAPD marker. Eight genotypes of rice from district in Kampar were amplified using eight primers (OPA 5, OPB 7, OPC 19, OPD 2, OPD 3, OPD 8, OPD 11, and OPD 13). The analysis of molecular data was performed by using NTSys pc -2.02 and GenAlex 6.1 . The results of the amplification of rice produced 48 loci with DNA band sizes obtained ranging from 350-1700 bp. The average percentage of polymorphic loci of the eight primers was 49.80%, where the highest percentage of polymorphic loci produced by OPA-7 primer was 75%, while the lowest polymorphic locus percentage was observed in OPD-13 primer which was 14.28%. The value of genetic distance between rice from Kampar in this study is in the range of 0.06 - 0.37. At a genetic distance of 0.79, rice plants can be grouped into two groups, namely the first group consisting of sokan and coku. The second group consisted of suntiong, korean , cupak putio, kuniong, jangguik, and cupak tenggi. The results of this study are expected to be taken into consideration in developing rice breeding strategies in the future.

To analyze the early microvascular retinal changes and oscillatory potentials alterations secondary to diabetic retinal damage, 44 eyes of 22 diabetic patients without and with mild diabetic retinopathy (DR) and 18 eyes of 9 healthy controls were examined. All subjects underwent spectral domain optical coherence tomography (SD-OCT), OCT angiography (OCTA), and electroretinography of oscillatory potentials (OPs). At OCTA, vessel area density (VAD), vessel length fraction (VLF), and fractal dimension (FD) were significantly reduced in the superficial vascular plexus (SVP), VLF and FD in the intermediate capillary plexus (ICP), and FD in the deep capillary plexus (DCP) in the diabetic group compared to the control group. The amplitude (A) of OP2, OP3, OP4 and the sum of OPs were significantly reduced in the diabetic group versus the controls, and the last two parameters were reduced also in patients without DR versus the controls. Moreover, in the diabetic group, a significant direct correlation was found between the A of OP1, OP2, OP3 and sOP and the VLF and FD in the SVP, while a statistically significant inverse correlation was found between the A of OP3 and OP4 and the VDI in the ICP and DCP. The reduced oscillatory potentials suggest a precocious involvement of amacrine cells in diabetic eyes, independently of DR presence, and their correlation with vascular parameters underlines the relevance of the crosstalk between these cells and vascular components in the pathophysiology of this chronic disease.

The aim of this work was to study in depth the resolving power of RAPD markers for rapid and reliable identification of olive cultivars in germplasm collections. The D parameter (the probability that two randomly chosen cultivars have different banding patterns), used for that purpose, showed high values for most of the 21 primers tested and its values ranged from 0.6114 (OPI-13) to 0.9762 (OPK-16) with a mean value of 0.8566. This parameter was used to select the five most discriminating primers: OPK-16, OPA-19, OPX-09, OPF-06 and OPZ-11. The joint confusion probability and the statistical number of indistinguishable pairs of cultivars were estimated for these primers (under independence hypothesis). The combination of three primers (OPK-16, OPA-19 and OPX-09) was found optimal for rapid discrimination of 103 cultivars with a very low value of cumulative confusion probability (1.72 × 10-5), leaving 0.09 pairs of cultivars indistinguishable. This fact, together with the efficiency of the most discriminating primers combination on an increasing number of cultivars, evidenced the utility of RAPD markers for discrimination of olive cultivars in collections and in nurseries.

The study was undertaken with major objective to differentiate fungal isolates belonging to various species of Bipolaris and Exserohilum infecting various agricultural crops. After routine isolation and purification of more than 35 attempts, 13 isolates were morphologically characterized into 8 species namely, Bipolaris cynodontis (IQBC-1 and DQBC-2), B. holmii (IQBH-1), B. micropus (IQBM-1), B. oryzae (DQBO-1, DQBO-2), B. sorghicola (DQBS-1, DQBS-2), B. tetramera (DQBT-1, DQBT-2), Exserohilum longirostrata (IQEL-1) and E. rostratum (DQER-1 and DQER-2). Molecular characterization of all the isolates using RAPD, ISSR and URP markers also revealed genetic variation. Out of 24 RAPD primers, only 9 (OPA-2, OPA-4, OPA-9, OPB-4, OPB-8, OPC-1, OPC-2, OPC-5 and OPE-5) could amplify the DNA sequence showing good polymorphism. Out of 20 ISSR primers, only 6 (ISSR 1, ISSR 13, ISSR 15, ISSR 17, ISSR 19 and ISSR 20) could show good polymorphism. Out of 13 URPs, only 5 (URP 4R, URP 6R, URP 9F, URP 13R and URP 38F) could amplify with a greater number of bands. In all the three makers, similarity matrix value using Jaccard’s coefficient ranged from 0.149 to 0.548 revealing genetic co-relation among isolates. Combined clustering of RAPD, ISSR and URP data showed relatively low genetic similarity coefficient ranging from 14.92 to 54.76 among isolates which revealed stronger co-relation among isolates. Cluster analysis based on UPGMA provided a clear resolution of relationship among all the 13 isolates belong to 6 Bipolaris and 2 Exserohilum species and confirmed the morphological characterization of the same. Further, molecular markers especially, RAPD, ISSR and URP have revolutionized the genetic analysis of plant pathogens including fungi and their application can greatly help in characterizing related genera and delineation of inter and intra-species complex of fungi.

The agarwood formation occurs in the trunk and<br />roots of trees that have been infected by a fungus, such as<br />Acremonium spp. Various fungi were associated with the<br />agarwood formation. Acremonium is generally considered as<br />highly polyphyletic, contains distantly related fungi. A study<br />was done to identify genetic diversities in 10 isolates of<br />Acremonium spp. from four different areas in Indonesia that<br />are associated with Aquilaria and Gyrinops verstigii using the<br />Random Amplified Polymorphic DNA (RAPD) technique.<br />Eight RAPD primers, i.e., OPA 02, OPB 04, OPB 07, OPB 17,<br />OPC 11, OPD 03, OPD 05, and OPE 07 were used in the<br />analyses. The results indicated that similarity index values of<br />the genetic variation ranged from 0.21 to 0.97. Based on the<br />Nei and Li’s similarity coefficients, these values indicating<br />the presence of high degree of genetic variability. The lowest<br />degree of genetic similarity were found between isolates F<br />(Acremonium spp., which is associated with G. verstigii from<br />Mataram, Nusa Tenggara Barat), and LM2 from south coastal<br />area of West Sumatra. The highest genetic similarity value<br />(0.97) was found between isolates Sr2 and Sr4 from Sorong,<br />Papua. Untuk menjaga kelestariannya perlu dilakukan upaya inventarisasi keanekaragaman genetik menggunakan penanda molekuler. Penelitian ini bertujuan menentukan keanekaragaman genetik dan hubungan kekerabatan empat ayam lokal Indonesia: ayam kampung, ayam ketawa, ayam kate lokal, dan ayam bekisar kangean, berdasarkan pendanda molekuler RAPD. DNA diekstraksi dari sampel darah 12 individu ayam lokal yang terdiri dari 3 ayam kampung, 3 ayam ketawa, 3 ayam kate lokal, dan 3 ayam bekisar kangean menggunakan Genomic DNA Mini Kit (Tissue) Geneaid. Analisis PCR-RAPD dilakukan menggunakan 6 primer, yaitu OPA 04, OPB 08, OPB 12, OPC 16, OPH 01, dan OPH 03. Produk PCR-RAPD dielektroforesis menggunakan agarose 1,5% dalam buffer TAE 1x. Hasil penelitian menunjukkan nilai kesamaan 12 individu ayam lokal menggunakan penanda RAPD berkisar dari 0% sampai dengan 100%. Hubungan kekerabatan 12 individu ayam lokal menggunakan penanda RAPD menunjukkan bahwa ayam yang memiliki kesamaan morfologi tidak mengelompok bersama. The rubber seeds are insufficient for producing rootstocks to rubber grafting. It can be overcome by an in vitro micro-cutting culture technique developed in the Indonesian Research Institute for Biotechnology and Bioindustry (IRIBB). However, the origin clone of 57 rubber genotypes used as an explant source in vitro micro-cutting culture is not recognized. The study was to investigate the 57 genotypes that came from mixed GT 1, PB 260, and RRIM 600 as parent clones. We investigated using seven primers of Random Amplified Polymorphic DNA (RAPD), i.e., OPA 02, OPA 07, OPA 15, OPB 04, OPC 05, OPC 11, and OPC 20. The qualitative analyzed by electrophoresis 1% gel agarose. A total of 47 DNA fragments produced with an average of 7 fragments per primer. OPA 02 generated of 13 fragments, whereas OPB 04 only one fragment. The DNA fragment pattern shows the presence of polymorphism. The genetic similarity coefficients obtained in the range of 62-96%. The highest genetic similarity (96%) is genotype 70 and 78. It recognized that 42 genotypes from 57 rubber genotypes had the closest relationship with PB 260 clones. Furthermore, six genotypes had a significant growth response as an explant in vitro micro-cutting culture.

Investigou-se o efeito da ordem de parto (OP) e da perda de peso durante a lactação no desempenho reprodutivo de 666 matrizes suínas. Foram formadas três classes de OP (OP1, OP2 e OP3-5) e duas classes de percentual de perda de peso (≤1% e >1%) durante a lactação. Foram avaliados: taxa de parto, intervalo desmame-estro (IDE) e tamanho da leitegada subsequente. Houve interação de OP versus perda de peso na taxa de parto das fêmeas (P<0,05). As fêmeas OP1 e OP2 tiveram maior chance (P<0,05) de não parir na classe >1% de perda de peso. A maior perda de peso não foi um fator de risco para não parir na condição OP3-5 (P>0,05). Não houve interação de OP versus perda de peso (P>0,05) para IDE e total de leitões nascidos. Fêmeas OP1 apresentaram IDE mais longo e menor tamanho da leitegada no parto subsequente (P<0,05) do que fêmeas OP2 e OP3-5. As perdas corporais na lactação não influenciaram o IDE (P>0,05), mas reduziram o tamanho da leitegada subsequente (P<0,05). Conclui-se que maior perda de peso na lactação resulta em aumento de falhas reprodutivas nas fêmeas mais jovens e reduz o tamanho da leitegada subsequente em todas as OP.

O maracujazeiro-doce (Passiflora alata Curtis), devido a preços diferenciados, vem ganhando importância dentro do mercado de frutas in natura. O melhoramento genético é fundamental para elevar a qualidade e a produtividade da cultura. Os marcadores moleculares do DNA têm sido muito úteis por permitirem a obtenção de um número praticamente ilimitado de polimorfismo genético sem influência do ambiente. Objetivou-se, neste trabalho, estudar a variabilidade genética de 17 acessos de maracujá-doce, com base em marcadores moleculares RAPD. Um acesso de P. quadrangularis e um de P. edulis foram utilizados como outgroups. Amostras de DNA genômico de cada acesso foram extraídas e 11 iniciadores decâmeros (OPD 04; 07; 08 e16; OPE 18 e 20; OPF 01 e 14; OPG 08; OPH 12 e 16) foram utilizados para a obtenção dos marcadores. Os marcadores obtidos foram convertidos em uma matriz de dados binários, a partir da qual foram estimadas as distâncias genéticas entre os acessos e realizadas análises de agrupamento e de dispersão gráfica. Do total de marcadores, considerando-se apenas os acessos de P. alata, observaram-se 87 (62,12%) bandas polimórficas, evidenciando a grande variabilidade intraespecífica. A análise de agrupamento realizada com base nas distâncias genéticas permitiu subdividir os 17 acessos de P. alata em, pelo menos, cinco grupos de similaridade genética. Os acessos silvestres foram os que mais contribuíram para a ampliação da base genética dos materiais estudados, abrindo perspectivas para o uso desses materiais em programas de melhoramento.

The study was conducted in a survey method, in which three individuals of each cultivar were analyzed using PCR-RAPD technique employing twelve primers, i.e. OPA-1, OPA-2, OPA-9, OPA-13, OPB-2, OPB-3, OPB-4, OPB-5, OPB-7, OPB-11, OPB-12 and OPJ-07. Data analysis based on morphological data is also included. Molecular analysis revealed that only 7.55% polymorphic band was obtained, while most of the bands were monomorphic, indicating very low variation among the cultivars. The phenogram that constructed based on literature showed that Kancil was closer to Jerapah cultivar, while RAPD-based dendogram showed that Hypoma 2 was closer to Kancil cultivar.

Madura has at least 22 genotypes of local tobaccos (Nicotiana tabacum L.). This diversity could potentially produce new genotype of tobaccos with superior characters. However, information of the genetic diversity of Madura tobaccos is still limited. The aim of this study was to determine the genetic variation and relationship of 24 genotypes of Madura tobaccos with Random Amplified Polymorphic DNA (RAPD) analysis. In this research we were used 6 single primers for amplification: (OPA-18, OPB-12, OPB-14, OPC-1, OPC-8 and OPC-19) and 2 mixture primers ((OPB-12+OPC-8) and (OPC-1+OPC-19)). Genetic similarity and clustering was analyzed with Unweighted Pair Group Method Arithmetic (UPGMA) method with Numerical Taxonomy and Multivariate Analysis System (NTSYS) version 2.10 software. From this research we found that OPA18425, OPB12450, OPC8500, (OPC19+OPC1)550 and OPC8800 can be used as specific markers. Polymorphic bands percentage with mixture primers was relatively equal with single primers (<60%). The dendogram showed that Madura tobacco genotypes consist of 2 main clusters: cluster A (22 genotypes) and cluster B (2 genotypes: Bukabu Sa’ang and Prancak-95). Madura tobaccos had high genetic similarity between genotypes ranging from 0.80-1.00.

Blueberry cultivars have traditionally been identified based on the evaluation of sets of morphological characters; however, distinguishing closely related cultivars remains difficult. In the present study, we developed DNA markers for the genetic fingerprinting of 45 blueberry cultivars, including 31 cultivars introduced from the United States Department of Agriculture. We obtained 210 random amplified of polymorphic DNA (RAPD) markers using 43 different primers. The number of polymorphic bands ranged from three (OPG-10 and OPQ-04) to eight (OPR-16), with an average of five. A cluster analysis performed with the unweighted pair group method using arithmetic averages produced genetic similarity values among the blueberry cultivars ranging from 0.53 to 0.85, with an average similarity of 0.68. A dendrogram clustered the 45 blueberry cultivars into two main clusters, with a similarity value of 0.65. Cluster I consisted of four rabbiteye cultivars (Pink Lemonade, Alapaha, Titan, and Vernon) and the Ashworth northern highbush cultivar. Cluster II consisted of 31 northern highbush cultivars, eight southern highbush blueberry cultivars, and Northland half-highbush blueberry cultivar. Fifty five RAPD fragments selected were sequenced to develop sequence-characterized amplified region (SCAR) markers, resulting in the successful conversion of 16 of 55 fragments into SCAR markers. An amplified polymorphic band has the same size as the RAPD fragment or smaller according to the primer combinations in the 16 SCAR markers. Among these markers, a combination of 11 SCAR markers provided sufficient polymorphisms to distinguish the blueberry cultivars investigated in this study. These newly developed markers could be a fast and reliable tool to identify blueberry cultivars.

<em>Knowing the genetic diversity in the tea germplasms collection is one of important conditions for assembling new superior varieties. Information of genetic diversity can be obtained through analysis using RAPD molecular markers. The study aimed to determine the genetic diversity of 21 tea genotypes based on RAPD markers. The research was conducted in Integrated Laboratory, Seameo Biotrop, Bogor, from July to September 2013. Genomic DNA was isolated from 21 tea genotypes leaf samples, then amplified with primer OPA 03, OPA 05, OPB 04, OPB 06, OPC 06, and OPD 08. Electrophoresis result was converted into binary data. The genetic similarity and cluster analysis calculation was done using NTSYS-pc version 2.10. In this research, 50 polymorphic bands (94,34%) and 3 monomorphic band (5,66%) were obtained. Cluster analysis based on Nei's genetic distance using the unweighted pair-group method with arithmatic (UPGMA) divided 21 tea genotypes into two groups at a genetic similarity value of 0,48. Group 1 consisted of 20 tea genotypes, while the second group comprised only a one genotype (Sin 27). The range of genetic similarity matrix was between 28%–92%, the lowest genetic similarity (28%) was found between GMB 4 and Sin 27 genotypes, while the highest (92%) was found between AS 2 and AS 1 genotypes. The information obtained can be utilized in breeding programs with the support of agronomic characters as well as in the conservation of tea germplasm.</em>

Schmidt & Hunter, 1998 ) und häufig zur Personalauswahl und -entwicklung verwendet. Während sich in den vergangenen Jahren durch die Akzeptanz der Big Five und einer Vielzahl von validen Instrumenten zur Persönlichkeitsmessung die Prädiktorenseite gut etabliert hat, wurde der Erfassung der Kriterien vergleichsweise wenig Beachtung geschenkt. In der vorliegenden Studie wird mit Hilfe eines berufsbezogenen Persönlichkeitsfragebogens (Occupational Personality Questionnaire; OPQ) und drei Fähigkeitstests die berufliche Leistung, gemessen anhand von Vorgesetztenbeurteilungen, vorhergesagt. Ziel dieser Arbeit ist es zu veranschaulichen, wie die Höhe der Validität durch unterschiedliche Operationalisierungen der Arbeitsleistung beeinflusst wird. Der Fokus richtet sich dabei zum einen auf die Erfassung des Konstruktes Arbeitsleistung durch spezifische Kompetenzdimensionen im Vergleich zur einer globalen Leistungsbeurteilung. Zum anderen wird gezielt untersucht, wie sich die Reliabilität unterschiedlicher Kompetenzskalen auf die Validitätskoeffizienten auswirkt. Die praktischen Implikationen der Ergebnisse im Hinblick auf die Durchführung von Validierungsstudien werden diskutiert.

In this study, we aimed to quantitatively monitor and describe the gait functions of patients, who underwent iliac crest bone grafting in atrophic jaw augmentation operation, by taking into account the alterations of gait parameters and muscle forces in the early recovery course. To do so, temporospatial and kinematic gait parameters of ten patients during pre- and postoperative periods were recorded, and forces of the gluteus medius, gluteus maximus, and iliacus muscles were calculated. Three postoperative periods were specified as one week (post-op1), two weeks (post-op2), and three weeks (post-op3) after the surgery. Restoring process of the gait patterns was comparatively evaluated by analyzing the gait parameters and muscle forces for pre- and postoperative periods. Temporospatial and kinematic parameters of post-op3 were closer to those obtained in pre-op than those in post-op1 and post-op2 (p<0.05). Muscle forces calculated in post-op3 showed the best agreement with those in pre-op among the postoperative periods in terms of both magnitude and correlation (p<0.05). In conclusion, the patients began to regain their preoperative gait characteristics from the second week after surgery, but complete recovery in gait was observed three weeks after the surgery.

Anacardium occidentale L., commonly known as cashew nut, belongs to the family Anacardiaceae. It is regarded as a high valued fruit nut crop world-wide. Potential of this economically important nut is under-utilized in Bangladesh in spite of having all favorable agro-climatic conditions. The objective of the present investigation was to characterize six cashew accessions using Random Amplified Polymorphic DNA (RAPD) markers. Four random primers viz. OPE-02, OPE-18, OPK-03 and OPB-15 were used to amplify DNA segments. A total of 33 reproducible bands were obtained, out of which 11 were monomorphic and 22 were polymorphic. On average 74.12% polymorphism was observed. . Primers OPB-15 and OPK-03 yielded 100% polymorphism and OPE 02 and OPE 18 produced 33.33% and 63.16 % polymorphism, respectively. Cluster analysis revealed two main distinct groups, first group included GP-1 and the second consisted of five genotypes viz. GP-2, GP-3, GP-4, GP-5and GP-6 The major cluster- II was further subdivided into two minor clusters i.e. minor cluster- III and IV. Minor cluster- III contained only one genotype GP-4. Minor cluster- IV consists of four rest genotypes. The genetic distance between the groups was found low and varied from 0.002 to 0.0308. Maximum genetic distance was observed between GP-1 and GP-2cashew germplasm and minimum between GP-5 and GP-6. The low genetic distance which is unusual for this out crossing long-lived tree species, indicates the probability of having common ancestry among the germplasm or may be due to the use of a narrow range of populations for the investigation. Vector quantization (VQ) techniques are widely used in similarity search for data compression, computation acceleration and etc. Originally designed for Euclidean distance, existing VQ techniques (e.g., PQ, AQ) explicitly or implicitly minimize the quantization error. In this paper, we present a new angle to analyze the quantization error, which decomposes the quantization error into norm error and direction error. We show that quantization errors in norm have much higher influence on inner products than quantization errors in direction, and small quantization error does not necessarily lead to good performance in maximum inner product search (MIPS). Based on this observation, we propose norm-explicit quantization (NEQ) — a general paradigm that improves existing VQ techniques for MIPS. NEQ quantizes the norms of items in a dataset explicitly to reduce errors in norm, which is crucial for MIPS. For the direction vectors, NEQ can simply reuse an existing VQ technique to quantize them without modification. We conducted extensive experiments on a variety of datasets and parameter configurations. The experimental results show that NEQ improves the performance of various VQ techniques for MIPS, including PQ, OPQ, RQ and AQ.

La maduración in vitro (MIV) de ovocitos es una técnica reproductiva que genera ovocitos capaces de soportar el desarrollo preimplantacional del embrión y su desarrollo completo a término, que ha venido siendo usada por mucho tiempo en animales, pero en humanos es aún una tecnología emergente. En la última década se han realizado estudios con el objetivo de mejorar la eficiencia de la tecnología; el avance alcanzado en los medios comercialmente disponibles muestran aún resultados inferiores al compararlos con técnicas reproductivas tales como fecundación in vitro (FIV). Sin embargo, la colecta de ovocitos maduros en ciclos de hiperestimulación con gonadotropinas aumenta el riesgo del síndrome de hiperestimulación ovárica (SHEO) y ninguna de las estrategias usadas para predecirlo ha eliminado por completo este riesgo. Es por ello, que la aplicación de procedimientos de MIV muestra perspectivas muy interesantes en pacientes con ovario poliquístico (OPQ) y síndrome de ovarios poliquísticos (SOPQ), diagnóstico de cáncer y problemas de falla de maduración de ovocitos. Por tanto, son necesarios los estudios para la optimización de esta tecnología, de manera de llegar a ser una técnica rutinaria en los laboratorios de reproducción asistida.
