Abstract The new conditions provided nowadays by technological advancements have made it possible to develop different ways of videographic production. While granting the concentration of different human activities within a single individual, the processes which are inherent to the digital universe have contributed as completely new tools to communication and collaborative productions. The concept of authorship is being questioned, both in the collective making and in the use of recyclable material, in such a way that a cultural substrate considered banal, every-day stuff and disposable can be transformed into raw material for artistic creation. The main objectives of this study were to present a historical overview of digital video, studying its communication process, enhanced by the arising of the Internet, and to analyze the possibilities inscribed in the audiovisual authoring software Keyworx, that is original both in its conceptual proposal and in its shape, interface and action. Keyworx is not a product manufactured by a company, but rather an educational project of the Waag Society, a Dutch research institution. This software makes a fusion of multi-user teleconference environment aspects with those of digital media process softwares (popularly known as VJ software ). A more profound discussion about the digital video interface and its functioning showed that it is of the utmost importance to a reflection on the particularities of a collaborative video creation model on the net. In this sense, the theories of Edmond Couchot, Lev Manovich, Arlindo Machado, Jay David Bolter, Richard Grusin and Giselle Beiguelman have occupied a key position in this study, because they elucidate aspects of the binary nature of digital media and their implications in the authoring processes, along with interviews with the creators of Keyworx. We could observe that, ever since the beginnings of digital video on the Internet, the communication roles between the sender and the receptor had their definition broadened, converted into interactors of a dialogical communication, which implies changes in the way they are created and distributed in the media.
As novas condições dadas pelos avanços tecnológicos na contemporaneidade propiciaram diferentes maneiras de produção videográfica. Ao mesmo tempo em que garante a concentração de diversas atividades humanas em um único indivíduo, os processos inerentes ao universo digital contribuíram como ferramentas inéditas para a comunicação e produções colaborativas. O conceito de autoria é questionado tanto no fazer conjunto como no uso de material reciclável , de maneira que um substrato cultural considerado banal, cotidiano e descartável, pode ser transformado em matéria prima para a criação artística. Esse estudo tem como objetivo principal apresentar um panorama histórico sobre o vídeo digital, estudando suas funções na comunicação on line; assim como analisar as possibilidades inscritas no software de autoração audiovisual Keyworx, que possui uma originalidade tanto em sua proposta conceitual, quanto em sua forma, interface e ação. O Keyworx não é um produto confeccionado por uma empresa, mas sim um projeto educacional da Waag Society, instituição de pesquisa holandesa. O software faz uma fusão de aspectos de ambientes de multi-usuário de tele-conferência, com os de software processuais de mídia digital (popularmente conhecidos como software de VJ ). Uma discussão mais profunda sobre a interface do vídeo digital e seu funcionamento, mostrou ser de suma importância para a reflexão sobre as particularidades de um modelo de criação colaborativa em vídeo na rede. Nesse sentido, as teorias de Edmond Couchot, Lev Manovich, Arlindo Machado, Jay David Bolter, Richard Grusin e Giselle Beiguelman encontraram lugar de destaque em nosso estudo, pois elucidaram aspectos da natureza binária da mídia digital e suas implicações nos processos de autoria; assim como a realização de entrevistas com os criadores do Keyworx. Pudemos verificar que, a partir do vídeo digital na Internet, os papéis comunicativos entre o emissor e o receptor têm sua definição alargada, convertidos em interatores de uma comunicação dialógica, o que implica em mudanças em sua forma de criação e distribuição midiática.

Abstract:
This qualitative case study research examined how constructivist problem-based learning facilitated higher level thinking, increased interest in art, and affected attitude toward on-line courses in an undergraduate philosophical aesthetics and interpretation of art criticism course. The research conducted for this study suggests that constructivist problem-based learning does facilitate higher level thinking and increases student interest in art and in on-line classes. Active learning assignments, along with the constructivist collaborative class atmosphere, encouraged students to think more deeply about their personal values concerning art and to consider alternative views. Problem-based learning in this class acted as a scaffold to aid in understanding the material and then in applying the material to unique and real-life situations. Each subject came to the course with certain thinking skills and left with increased knowledge about art but also with increased critical thinking skills for critically examining and discussing art. Participants completed the course with more confidence in their critical thinking ability and in dealing with visual art images. Data was gathered from seven study participants in the form of highly-structured interviews, an early and final critical writing analysis, a major problem assignment and its reflection journal, a beginning survey, and two final surveys. The final major problem involved an individual proposal followed by a collaborative group proposal. Group collaboration constituted the most frustration and problem within the constructivist design of the class. This research took a relativistic viewpoint in gathering data and interpreting meaning.

Abstract: This research is the culmination of a four-year investigation and analysis into the principles of Interaction Design, particularly those that are found to be most suitable when designing and developing interactive navigation systems. The research was undertaken as a Masters degree by project. The project consists of a CD containing an online gallery showcasing works of art and an accompanying exegesis. The exegesis is structured into seven chapters, which consider, analyse and define what the key characteristics of Interaction Design are, where it comes from, and how it improves the quality of interactive multimedia applications. The exegesis includes four case studies that look at how other practitioners in the digital realm have created systems for showcasing narrative or creative content online. I examine alternative artworks and how they have shaped the development of creative media. I investigate what experts in the field define as good Interaction Design and what guidelines and principles they recommend. I show how these guidelines conflict with more creative approaches and how good design and creativity can be merged to be usable and friendly to users. I also look at the history of opponents of guidelines and principles and how their contribution helps make design better. By creating the example gallery I aim to help designers working within the field of ID to understand the principles behind good design in order that they may deliver higher-quality user experiences relevant to the content they are displaying. By creating this gallery I also hope to help artists understand the principles behind good design in order that they may showcase their artworks in ways appropriate to their artwork. By designing and building an example I aim to provide a better understanding of how to construct a feature-rich application in an easy to use and understandable environment.

Abstract:
I work in the medium of song. A multidisciplinary artist and composer, I make work that is immersive, time-based and often participatory. I interact with landscape and the complexities of American history, bringing into focus local ecologies through the lens of song. This document accompanies my thesis performance The Sound of a Stone, an immersive exploration of song, language, ecology and locational listening performed in a 4-channel surround format. In the semi-improvised composition, I sample live vocals, mandolin and found natural objects in a combination of roots music traditions and experimental techniques. Utilizing the software Ableton Live to process and layer the samples in real time, I build a series of "songscapes" which connect to a specific site: the fall line of the James River. The Sound of a Stone premiered April 8, 2019 at Sonia Vlahcevic Concert Hall, W. E. Singleton Center for the Performing Arts, Virginia Commonwealth University, Richmond, Virginia.

Abstract:
This thesis aims to investigate students’ qualitative experiences of assessment in art and design higher education using storytelling and visual representations. It aims to investigate whether collaborative storytelling can encourage students to reflect on, and learn from, each others’ experiences of assessment. In order to examine these aims, an on-line tool, ‘StoriesAbout… Assessment’ was designed and developed, based on an adapted model of storytelling as a reflective tool in higher education. Visual representations of students’ experiences were also used to identify the affective aspects of the assessment experience. In using these novel methods, the research aimed to highlight the whole student learning experience and how assessment affects that experience. Traditional methods of surveying and evaluation do not usually focus on this, nor do they provide a reflective, learning process for students. The analysis of stories led to a greater understanding of students’ experiences of assessment in art and design by identifying a number of key issues: the impact of negative experiences, the need for greater clarity of assessment criteria due to the subjective nature of the discipline, the tension students perceive between their role as creative practitioners in an educational setting and their role in the wider art world, the value of peer support and appropriate feedback. The storytelling model enabled students to view stories from different perspectives and to consider changes to their practice, and the model has demonstrated its efficacy in supporting reflective thinking and transformative learning. The emotional aspect to students’ experiences was particularly evident in their visual representations which often used strong imagery to depict how the stress of assessment affected them. The drawings also showed stereotypes of assessment, such as images of exams, indicating that these previous experiences had become synonymous with assessment, despite there being few formal exams in art and design. In summary, this thesis contributes two new methods for understanding and enhancing the student learning experience, which have been proven in the context of art and design higher education.

Abstract The main cause of temperature and composition fluctuations in the electric arc furnace (EAF) process is the scrap used as a raw material. Process conditions in EAF can vary significantly from heat to heat because there is no accurate information of scrap composition. Due to harsh process conditions, there are currently few sensors available for direct on-line measurement of the EAF process. In this work new information about stainless steelmaking EAF process conditions is sought with optical emission spectrum measurement. The measurement system relies on transportation of the light emitted from the measured furnace to a remotely situated spectrometer. Analysing the slag composition from the arc emission spectrum was tested in the laboratory and on a pilot scale. The laboratory measurements indicate that changes in the amount of CrOx and MnO in the slag have the highest impact on optical emission spectra. The pilot scale measurements show that the Cr2O3 content of the slag can be measured from the arc emission spectrum using suitable reference lines with an average absolute error of 0.62 %-points and a standard deviation of 0.49 %-points. The results from measurements at Outokumpu Stainless Oy, Tornio Works, indicate that measurement of the optical emission spectrum from industrial EAF is feasible in a practical sense, and can be used in analysing of EAF atmosphere, scrap melting and slag surface. Furthermore, the results of industrial measurements indicate that the atoms in the arc plasma mainly originate from the slag. The measurement of scrap melting could be potentially used in EAF control in optimization of arc voltages and second scrap bucket charging. The potential use of slag CrOx measurements is in optimization of reductant additions as well as defining the further processing of EAF slag
Tiivistelmä Valokaariuunien ohjaus on perinteisesti ollut uunioperaattorin käsissä. Valokaaariuuniprosessin on-line mittaukseen on olevassa vähän menetelmiä johtuen uunin hyvin haastavaista olosuhteista. Tässä työssä on tutkittu optiseen emissiospektroskopiaan perustuvaa menetelmää uuden jatkuva-aikaisen tiedon tuottamisessa valokaariuuniprosessista. Mittausjärjestelmä perustuu valon keräämiseen mitattavasta uunista valokuidun avulla, joka johtaa valon analysoitavaksi etäälle prosessista sijoitettuun spektrometriin. Mittauksia suoritettiin laboratorio-, pilot- ja tehdas-mittakaavassa. Valokaariuunin kuonan koostumuksen analysointia testattiin laboratorio- ja pilot-mittakaavan uuneilla. Laboratoriomittaukset osoittivat että kuonan komponenteista CrOx ja MnO ja vaikuttavat eniten mitattuun emissiospektriin. Pilot-mittakaavan kokeissa havaittiin, että kuonan Cr2O3-pitoisuutta voidaan mitata valokaaren emissiospektristä 0,62 %-yksikön keskimääräisellä absoluuttisella virheellä ja 0,49 %-yksikkön hajonnalla. Teollisella valokaariuunilla suoritetuista mittauksista havaittiin että optisen emissiospektrin mittaus voidaan suorittaa ilman ylitsepääsemättömiä teknisiä esteitä. Mittauksen tuloksia voidaan puolestaan käyttää kaasufaasin reaktioiden, romun sulamisen ja kuonapinnan ominaisuuksien arvioinnissa. Valokaaren emissiospektrin analyysi osoitti, että valokaaren plasman komponentit ovat pääosin peräisin kuonasta, joka mahdollistaa kuonan koostumuksen arvioinnin valokaaren emissiospektrin perusteella. Romun sulamisen mittausta voidaan prosessinohjauksessa käyttää jänniteportaiden ja toisen korin panostuksen optimointiin. Kuonan kromipitoisuuden mittaamista voidaan puolestaan käyttää pelkistinaineiden lisäyksen optimointiin ja kuonan jatkokäsittelyn valintaan

Abstract:

In this bachelor thesis, cable fault locators are studied for use on the overhead electrode lines in the HVDC (High Voltage Direct Current) Light project Caprivi Link. The cable fault locators studied operates with the principle of travelling waves, where a pulse is sent in the tested conductor. The time difference is measured from the injection moment to the reflection is received. If the propagation speed of the pulse is known the distance to the fault can be calculated. This type of unit is typically referred to as a TDR (Time Domain Reflectometer). The study is performed as a computer simulation where a simplified model of a TDR unit is created and applied to an electrode line model by using PSCAD/EMTDC. Staged faults of open circuit and ground fault types are placed at three distances on the electrode line model, different parameters of the TDR units such as pulse width and pulse amplitude along with its connection to the electrode line are then studied and evaluated. The results of the simulations show that it is possible to detect faults of both open circuit and ground fault types with a suitable TDR unit. Ground faults with high resistance occurring at long distances can be hard to detect due to low reflection amplitudes from the injections. This problem can somewhat be resolved with a function that lets the user compare an old trace of a “healthy” line with the new trace. The study shows that most of the faults can be detected and a distance to the fault can be calculated within an accuracy of ± 250 m. The pulse width of the TDR needs to be at least 10 μs, preferable 20 μs to deliver high enough energy to the fault to create a detectable reflection. The pulse amplitude seams to be of less significance in this simulation, although higher pulse amplitude is likely to be more suitable in a real measurement due to the higher energy delivered to the fault. The Hipotronics TDR 1150 is a unit that fulfil these requirements and should therefore be able to work as a line fault locator on the electrode line.

This is due to the cause that the marked demands more goods but to a lower cost. With this in mind, how can we create an aid for assembly line workers that could decrease repetitive strain injuries, facilitate flexibility and reduce the weight load on the workers joints? In this project I have explored the possibility to create new and intelligent aids for the assembly lines with great focus the human body and ergonomics.

Les ``C-arm'' sont des systémes de radiologie interventionnelle fréquemment utilisés en salle d'opération ou au lit du patient. Des images 3D des structures anatomiques internes peuvent être calculées à partir de multiples radiographies acquises sur un ``C-arm mobile'' et isocentrique décrivant une trajectoire généralement circulaire autour du patient. Pour cela, la géométrie conique d'acquisition de chaque radiographie doit être précisément connue. Malheureusement, les C-arm se déforment en général au cours de la trajectoire. De plus leur motorisation engendre des oscillations non reproductibles. Ils doivent donc être calibrés au cours de l'acquisition. Ma thèse concerne la calibration intrinsèque d'un C-arm à partir de la détection de la projection du collimateur de la source dans les radiographies.Nous avons développé une méthode de détection de la projection des bords linéaires du collimateur. Elle surpasse les méthodes classiques comme le filtre de Canny sur données simulées ou réelles. La précision que nous obtenons sur l'angle et la position (phi,s) des droites est de l'ordre de: phi{RMS}=+/- 0.0045 degrees et s{RMS}=+/- 1.67 pixels. Nous avons évalué nos méthodes et les avons comparés à des méthodes classiques de calibration dans le cadre de la reconstruction 3D
Mobile isocentric x-ray C-arm systems are an imaging tool used during a variety of interventional and image guided procedures. Three-dimensional images can be produced from multiple projection images of a patient or object as the C-arm rotates around the isocenter provided the C-arm geometry is known. Due to gravity affects and mechanical instabilities the C-arm source and detector geometry undergo significant non-ideal and possibly non reproducible deformation which requires a process of geometric calibration. This research investigates the use of the projection of the slightly closed x-ray tube collimator edges in the image field of view to provide the online intrinsic calibration of C-arm systems.A method of thick straight edge detection has been developed which outperforms the commonly used Canny filter edge detection technique in both simulation and real data investigations. This edge detection technique has exhibited excellent precision in detection of the edge angles and positions, (phi,s), in the presence of simulated C-arm deformation and image noise: phi{RMS} = +/- 0.0045 degrees and s{RMS} = +/- 1.67 pixels. Following this, the C-arm intrinsic calibration, by means of accurate edge detection, has been evaluated in the framework of 3D image reconstruction

Abstract:
This thesis examines the ways in which regional communities appropriate their historic icons of the past, integrating these ‘markers of identity’ into the wider socio-economic context. It notes how nostalgia and the collective memory, together with a strong sense of place are reflected in celebrations which honour national and local historic characters and events, and observes the ways in which isolated rural towns reconcile their new tourist image with their pioneering past. It will be argued that the concepts of nation and national identity are increasingly being challenged by the need for a social and cultural identity which belongs to the local community. A range of diverse cultures and heritage sites has been studied in order to analyse the dilemmas facing local, outback, and Indigenous communities in reconstructing a regional identity today. The Heritage Trail can be seen as a symbolic rite of passage, and this thesis is indeed a personal journey divided into two major components. First, it takes the form of documentary, a visual component consisting of three video films which capture the heritage tourism product along the Tropic of Capricorn. On the Western Line documents the author’s journey on ‘The Spirit of the Outback’ from the central coast to Winton and Kynuna, the home of Waltzing Matilda in far western Queensland. Two further historical productions The Legend of King O'Malley and The Triumph and Tragedy of Tommy Wills record local and national narratives, which both deliver a controversial picture of federation politics and pioneer settlement, colonial sport and frontier war, each focusing on a regional perspective within a wider national vision. The second, written component reflects upon the role of the documentary maker in recording a social history of these diverse communities. It examines the auteur/director’s own perceptions of regional identity, and the oppositions and ambiguities of reality are juxtaposed with legend and myth. This essay explores the different layers of meaning inscribed within central images of cultural tourism such as: the Stockman’s Hall of Fame at Longreach; the Australian Worker’s Heritage Centre at Barcaldine; local museums at Mt. Morgan and Emu Park; the Dreamtime Cultural Centre; and the South Sea Islanders’ Sugar Trail. Following Bennett and Bourdieu’s work on museum visitors, the author’s preliminary survey of tourists seeking Capricornia’s increasingly popular heritage destinations is discussed. In conclusion, it can be argued that the rapid growth of Central Queensland’s cultural centres, local museums, and bicentennially funded outdoor art and ‘unusual’ monuments, has produced new sources of income in a community desperately seeking to survive. The cultural tourist has become a major producer of the heritage industry which impacts strongly on regional identity.

Although the primary role of the computer has been in processing and analysis of survey data, it has increasingly been used in data collection. Computer surveys are not exempt from a common problem: some refuse to participate. Many researchers and practitioners indicate the refusal problem is less for computer surveys, perhaps due to the novelty of the method. What has not been investigated is the refusal problem when on-line surveys are no longer novel. This research study examines the use of one form of computer-assisted data collection, the electronic or on-line survey, as an organizational research tool. The study utilized historical response data and administered an on-line survey to individuals known to be cooperative or uncooperative in other on-line surveys. It investigated nonresponse bias and response effects of typical responders, periodic participants, and typical refusers within a sample of corporate employees in a computer-interactive interviewing environment utilizing on-line surveys. The items measured included: participation, respondent characteristics, response speed, interview length, perceived versus actual interview length, quantity of data, item nonresponse, item response bias, consistency of response, extremity of response, and early and late response. It also evaluated factors reported as important when deciding to participate, preferred data collection method, and preferred time of display. Past participation, attitudes toward on-line organizational surveys, response burden, and response error were assessed. The overall completion rate of 55.7% was achieved in this study. All effort was made to encourage cooperation of all groups, including an invitation to participate, token, on-line pre-notification, 800 number support, two on-line reminders, support of temporary exit, and a paper follow-up survey. A significant difference in the participation of the three groups was found. Only three demographic variables were found to be significant. No significant differences were found in speed of response, interview length, quantity, item nonresponse, item response bias, consistency, and extremity. Significant differences were found in the perceived and actual times to complete the survey.

Certain subquotients of Vec(R)-modules of pseudodifferential operators from one tensor density module to another are categorized, giving necessary and sufficient conditions under which two such subquotients are equivalent as Vec(R)-representations. These subquotients split under the projective subalgebra, a copy of ????2, when the members of their composition series have distinct Casimir eigenvalues. Results were obtained using the explicit description of the action of Vec(R) with respect to this splitting. In the length five case, the equivalence classes of the subquotients are determined by two invariants. In an appropriate coordinate system, the level curves of one of these invariants are a pencil of conics, and those of the other are a pencil of cubics.

To date much of the literature on consumers’ concerns about business-to-consumer (B2C) e-commerce has described findings from quantitative research. As a result, much of that literature has focused on specific previously-identified concerns (e.g., privacy of personal information, use of credit cards for on-line payment). Also, there has been little research into the concerns of New Zealand consumers, and all of it has been quantitative. In order to gain a broader understanding, this study took a qualitative approach. Three focus groups were conducted, in order to identify consumers’ concerns. The concerns that were thus identified were combined with those that a review of the literature had previously identified, and were used to draw up a set of guidelines to be used in semi-structured interviews. Fifteen interviews were then conducted, in order to gain consumers’ views about each concern. It seems that at a higher level, consumer’s concerns have shifted. The literature suggests that in the past consumers’ concerns about Internet shopping have focused on the fact that Internet shopping is conducted via the Internet – as a result of which consumers have, for example, been concerned about the privacy of their personal information ('if I give them my e-mail address, will I get spam?') This study suggests that now consumers’ concerns focus on the fact that Internet shopping is a form of shopping – as a result of which they are now only prepared to use the Web sites of 'reputable companies', and they now require to be able to evaluate an item adequately before buying it. And it seems that another result of this is that consumers now expect that the 'reputable companies' whose Web sites they visit will – as a matter of course - address to their satisfaction issues such as the privacy of their personal information. While there are some types of products that consumers are prepared to buy on-line (e.g., air travel), there are many about which they are reluctant. Concerns were expressed about a wide range of potential purchases, and for a variety of reasons; also, some data was inconsistent (for example, some consumers were not prepared to buy clothes on-line, but others were). This appears to be an area in which consumer attitudes are still evolving. It is suggested that it may be helpful for further research on this matter to consider both demographic factors and the degree of consumer involvement in particular types of purchases.

The pathophysiology of chronic morphine on the immune system, especially on the cells of the innate immune system that leads to an immune compromise state has not been fully elucidated. The cells of the innate immune system are the first line of defense in mounting an immune response needed in infections, inflammation, cancer development, etc. One of the ways by which these innate immune cells act is by the production of cytokines with direct effects and to also recruit other immune cells, as required. A balance of pro- and anti-inflammatory cytokines is necessary for immune competence. I hypothesized that chronic morphine would act via a classical opioid receptor to stimulate the PI3K/Akt/Gsk3β pathway to produce predominantly anti-inflammatory cytokines. Cytokine gene expression levels were assessed via RT-PCR; Akt and Gsk3β protein levels measured using indirect ELISA. The data suggests that chronic morphine causes a significant reduction in IL-6 production, but does not act via the Akt/Gsk3β pathway or the classical opioid receptor to cause this effect in microglia cells.

While many investigations have been performed to establish a better understanding of the effects that magnetic fields and nanoparticles have on cells, the fundamental mechanisms behind the interactions are still yet unknown, and investigations on concurrent exposure are quite limited in scope. This study was therefore established to investigate the biological impact of concurrent exposure to magnetic nanoparticles and extremely-low frequency magnetic fields using an in-vitro CHO-K1 cell line model, in an easily reproducible manner to establish grounds for further in-depth mechanistic, proteomic, and genomic studies. Cells were cultured and exposed to 10nm Fe3O4 nanoparticles, and DC or low frequency (0Hz, 50Hz, and 100Hz) 2.0mT magnetic fields produced by a Helmholtz coil pair. The cells were then observed under confocal fluorescence microscopy, and subject to MTT biological assay to determine the synergistic effects of these concurrent exposures. No effects were observed on cell morphology or microtubule network; however, cell viability was observed to decrease more drastically under the combined effects of magnetic field and nanoparticle exposures, as compared to independent exposures alone. It was concluded that no significant difference was observed between the types of magnetic fields, and their effects on the nanoparticle exposed cells, but quite clearly there are deleterious synergistic effects of these concurrent magnetic field and nanoparticle exposure conditions.

Previously, Whipp et. al. (J. Appl. Physiol.: Respirat. Environ. Exerc. Physiol. 50(1):217-221, 1981) demonstrated the feasibility of determining four parameters of aerobic function, identified as maximum oxygen uptake (μVO_2), VO_2 at anaerobic threshold (θan), the time constant for oxygen uptake kinetics (rVO_2) and work efficiency (η), using a short duration ramped bicycle ergometer exercise test. Because of the importance of being able to measure these parameters on a variety of measurement instruments a short duration ramping treadmill protocol has been developed. The ability of this protocol to determine the four aerobic parameters has been validated against conventional methods. The results of this investigation indicate that μVO_2, θan, rVO_2 and, η may be obtained from a single, short-duration ramping treadmill test.

Engaging in muscle strengthening activities (MSA) as part of a physical activity program offers health benefits. Although the merits of physical activity are well documented, many adults fail to meet appropriate levels as recommended in the 2008 Physical Activity Guidelines for Americans (PAGA). To get a more complete understanding on an individual's physical activity behaviors, the Tracking Resistance Exercise and Strength Training (TREST) internet based survey was developed. The purpose of the current study was to determine the test-retest reliability of TREST items. Additionally, the prevalence of participants meeting the 2008 PAGA was reported by gender. The survey was completed approximately two weeks apart by 224 (52% male) undergraduate kinesiology students. Analysis of the survey items presented TREST as a reliable instrument in assessing an individual's physical activity behavior with a focus on MSA. Among the convenience sample of 445 participants (56% male) that completed the survey in assessment #1, 73% met the 2008 PAGA minimum recommendations for MSA (>=2 days/week) and aerobic activity (>= 150 min MVPA). A more complete MSA and MVPA criteria was established (requiring MSA of all seven major muscle groups) and only 32% of participants met this guideline. In general, men engaged in aerobic exercise and MSA more than women. These results cannot be generalized due the age, activity level, and education of the study's participants. Future studies should investigate the validity of TREST items among a sample of varying fitness levels, races/ethnicities, ages, and educational levels.

Second language (L2) researchers and teachers have increasingly come to believe that using a computer-based corpus can be extremely helpful in the language classroom. The purpose of this study is to examine whether corpora can be used outside of the classroom in order for students to improve their essays independently. No previous study has tried to examine students' essays in relation to corpus use so that this study is exploratory. Seven international students wrote five essays on specific topics and then corrected their errors through corpus research. Two experiments were conducted with different students and followed three steps: receiving information about how to use the BYU COCA, writing and correcting, and interviews with students. I examined quantitatively the number and types of errors that students were able to correct in two experiments and reported qualitatively on students' interview responses.

This research investigated banner ads on the World Wide Web, specifically the types of messages used in those ads and the effectiveness of the ads as seen by their intended audience. The focus was on health-related banner advertisements targeting senior citizens. The study first sought to determine the frequency of appearance of those ads when classified into categories of compliance-gaining tactics provided by research scholars. Second, the study explored the relative perceived effectiveness among those categories. Two graduate students from a Central Texas university sorted text messages into predetermined compliance-gaining categories. Chi square tests looked for significant differences in the frequencies of banner ads in each category. Forty-five senior citizens from the Central Texas area completed surveys regarding the perceived effectiveness of a randomly ordered, randomly selected set of categorized banner ads. A repeated measures test attempted to determine whether some compliance-gaining strategies used in health-related banner ads were perceived as more effective than others. The hypothesis stated that there would be differences in frequencies of compliance-gaining strategies used among the compliance-gaining categories in health-related banner ads for senior citizens. The hypothesis was supported. The research question asked if some categories of compliance-gaining strategies used in health-related banner ads were perceived as more effective than others by senior citizens. There was no evidence that senior citizens perceived any compliance-gaining category as being more effective than any other. However, post hoc analyses revealed trends in the types of compliance-gaining messages senior citizens perceived as more effective. These trends provide a basis for directional predictions in future studies.

The work presented here outlines experiments done using a novel RF exposure chamber. This device allows biological cells to be exposed to microwave radiation similar to those emitted by mobile telephones, whilst imaging them using a laserscanning confocal microscope. Jurkat E6.1 T lymphocytes in the exposure chamber were kept within �0.5�C of 37�C, allowing for the investigation of possible athermal effects of microwave energies. These cells were loaded with the fluorescent probe Fluo-3 AM, which is specific for calcium ions, and were monitored over two 10minute periods. The first period being a control period, the second being a period where the cells were either exposed to RF energy or sham exposed. Another 5min imaging period was for the positive control,where maximal fluorescence can be achieved by the addition of the ionophore A23187. 5 different conditions for cell exposure were investigated. Both continuous wave 900MHz and continuous wave 900MHz pulse modulated at 217Hz exposures were carried out on cells that were either unactivated, or those that were activated by the mitogen phytohaemaglutinin (PHA). For these 4 conditions the average Specific Absorption Rate (SAR) was calculated to be 1.5W/kg. A group of unactivated cells were also exposed to continuous wave 900MHz energy with an average SAR calculation of 7.5W/kg. Results showed that no significant changes in calcium ion levels occurred when averaged fluorescence slopes were compared between RF exposed cells and the control period. The mean change in slopes exposed/sham period � control period)between cells that were exposed and those sham exposed also showed no significant difference. Following an inference made in the work of Galvanovskis et al. (1999)1 who showed there is a change in the calcium ion oscillation spectrum as a result of 50Hz magnetic fields, a measure of the mean frequencies of all cells was determined using a Fast Fourier Transform (FFT) analysis. The change in the average mean frequencies in cells was then measured for all conditions. Of statistical significance was the change in average mean frequency between the control period and the sham/exposed period between cells that were exposed and those sham exposed, when cells were activated by PHA. The results also showed that there was an overall drop in average mean frequency as a result of RF exposure. Assuming there is a biological significance to the findings of this thesis, careful analysis of the calcium dynamics of tissue samples and cell types associated with RF exposure from mobile phones would need to be carried out to determine what they are. This was unfortunately beyond the scope of the present study.

Spores of thermophilic bacilli are a common concern during the manufacture of milk powder. Spores are believed to occur in high numbers in milk powder due to their ability to survive pasteurisation, attach to stainless steel surfaces, germinate, grow as biofilms and subsequently enter the product stream and thereby contaminate the final product. In this study, thirty one thermophilic bacilli isolates were obtained from a New Zealand milk powder production line and identified as either Anoxybacillus flavithermus or Geobacillus spp. using random amplified polymorphic DNA (RAPD) and species-specific PCR. Sporulation media and a polyethylene glycol two-phase separation system were modified to produce high yields of spores free from debris. The spores of four Geobacillus spp. isolates (CGT-8, D4, E7 and E11) were characterised in terms of structure (electron microscopy), surface charge (zeta potential), hydrophobicity (contact angle and microbial adhesion to hexadecane) and attenuated total reflectance infrared spectroscopy (ATR-IR). Spores from three of the four isolates possessed an exosporium while the fourth did not. However the integrity of the exosporium varied over time. The spores were negatively charged (-10 to -20 mV) at neutral pH and high ionic strength (0.1 M KC1). Both hydrophobicity assays revealed that the spores of the four isolates were relatively hydrophilic while ATR-IR revealed the spores' surfaces consisted of protein and polysaccharides. The influence of these spore characteristics on adhesion to a variety of substrata under high flow rates was examined using the extended Derjaguin, Landau, Verwey and Overbeek (XDLVO) theory. Spores generally attached in higher numbers to hydrophobic surfaces compared to hydrophilic surfaces, however this observation was more prevalent for isolate D4. This result indicated that a single mechanism could not describe the adhesion of spores from different strains. A series of glass surfaces with modified characteristics were produced in order to test the antifouling properties on the adhesion of D4 spores. Spores suspended in a high ionic strength medium (0.1 M KC1) attached in greater numbers (1 Log₁₀ CFU cm⁻�) to positively charged and hydrophobic surfaces compared with negatively charged and hydrophilic surfaces. A clean in place (CIP) procedure, reduced spore numbers on hydrophobic and hydrophilic surfaces by 1.5 and by 2.0 Log₁₀ CFU cm⁻�, respectively. When spores were suspended in milk, there was little difference in the number of spores attaching to the different surfaces (ie. 3.5 to 3.8 Log₁₀ CFU cm⁻�), and spore removal from surfaces via a CIP regime was unchanged (1.5 to 2.0 Log₁₀ CFU cm⁻� reduction) compared with spores that attached in simple 1:1 electrolyte media. The effects of a caustic wash on spore surface characteristics and adhesion was determined. There was a significant reduction in spore viability (2 Log₁₀ CFU mL⁻�) after a 30 min caustic wash at 65 �C in the current study, however surviving spores displayed a greater propensity to attach to stainless steel. Surface characterisation results revealed an increase in hydrophobicity and a greater negative charge on the spores' surface after treatment with NaOH. Surviving spores could potentially recontaminate sections of the plant which are cleaned with this recycled caustic wash solution, thereby seeding surfaces with spores at the beginning of the next processing run. In conclusion, while surfaces that reduce spore adhesion and enhance removal can be produced, exposure to complex solutions such as milk can reduce the anti-fouling effectiveness of such surfaces to spore adhesion.

Refletir sobre as experiências com imagens e sons em tempo real é pensar em uma ampla gama de possibilidades e experiências pela humanidade, desde os tempos mais remotos. Pelo fato dessas experiências audiovisuais em tempo real serem numerosas no tempo e no espaço, e já amplamente citada em diferentes estudos, esta apresentação tem por objetivo realizar um recorte mais restrito às experiências audiovisuais em tempo real no campo cinematográfico contemporâneo: uma arte, meio e processo em expansão que culmina na existência de um fenômeno semiótico, realizado principalmente através de modos experimentais com a presença do(s) autor(es) concebendo a experiência audiovisual, conjuntamente ao aparato tecnológico e ao público, todos participantes da criação e exibição da obra audiovisual no mesmo tempo em que ela ocorre, em transmissão direta para a tela de cinema, monitores, telas digitais ou espaços arquitetônicos. Com base em obras audiovisuais realizadas entre 2007 a 2017 desenvolveremos uma reflexão e análise das poéticas e técnicas das experiências audiovisuais em tempo real, de maneira a compreender com um olhar mais atento as possibilidades criativas e contribuir com a reflexão sobre estes formatos do audiovisual contemporâneo, cujos meios e processos estão em contínua expansão de suas fronteiras. Em estética do cinema, esta pesquisa segue em continuidade aos estudos e práticas do cinema experimental e em sua vértice ao cinema expandido.
Reflecting about real-time imagery and sounds experiences is thinking about a wide range of possibilities and experiences for humanity, from the earliest times. Because the real-time audiovisual experiences are numerous in time and space, and already widely quoted in different studies, this presentation aims to make a more restricted cut-off to real-time audiovisual experiences in the contemporary cinematographic field: an art, medium and an expanding process that culminates in the existence of a semiotic phenomenon, performed mainly through experimental modes with the presence of the author(s) directing the audiovisual experience in real-time, together with the technological apparatus, the cast, and the public, all participants in the creation and exhibition of the audiovisual work at the same time as it occurs, in direct transmission to the cinema screen, monitors, digital screens or architectural spaces. Based on audiovisual works carried out between 2007 and 2017 we will develop a reflection and analysis of the poetics and techniques of audiovisual experiences in real-time, in order to understand with a closer look the creatives possibilities in live cinema and contribute with reflection on these forms of the contemporary audiovisual, whose means and processes are in continuous expansion of its borders. In aesthetics of the cinema, this research follows in continuity to the studies and practices of the experimental cinema, and in its vertex to the expanded cinema.

Nospréoccupations dans ce contexte sont principalement algorithmiques (est-il facile de trouver une pondération d’arêtes voisin-somme-distinguante ou une coloration d’arêtes localement irrégulière utilisant le plus petit nombre possible de poids ou couleurs ?) et structurelles (quel est le plus petit nombre de couleurs d’une coloration d’arêtes localement irrégulière ?). This thesis is dedicated to the study of two families of graph partition problems.First, we consider the problem of vertex-partitioning a graph into connected subgraphs.Namely, given p positive integers n1; n2; :::; np summing up to the order of some graph G, canwe partition V (G) into p parts V1; V2; :::; Vp so that each Vi induces a connected subgraph withorder ni? We then consider stronger questions. Namely, what if we want G to be partitionablewhatever are p and n1; n2; :::; np? What if we also want specific vertices of G to belong to somespecific subgraphs induced by the vertex-partition? What if we want the subgraphs induced bythe vertex-partition to be more than connected? We consider all these questions regarding boththe structural (are there structural properties ensuring that a specific vertex-partition necessarilyexists?) and algorithmic (is it hard to deduce a specific vertex-partition?) points of view.Then, we focus on the so-called 1-2-3 Conjecture, which asks whether every graph G admitsa neighbour-sum-distinguishing 3-edge-weighting, i.e. a 3-edge-weighting by which all adjacentvertices of G get distinguished by their sums of incident weights. As a tool to deal with the1-2-3 Conjecture, we notably introduce the notion of locally irregular edge-colouring, which isan edge-colouring in which every colour class induces a subgraph whose adjacent vertices havedistinct degrees. The main point is that, in particular situations, a neighbour-sum-distinguishingedge-weighting of G can be deduced from a locally irregular edge-colouring of it. Our concernsin this context are mostly algorithmic (can we easily find a neighbour-sum-distinguishing edgeweightingor locally irregular edge-colouring using the least number of weights or colours?) andstructural (what is the least number of colours in a locally irregular edge-colouring?). Abstract:
Le pemphigus est une maladie auto-immune spécifique de la peau et des muqueuses provoqué par des auto-anticorps (Ac) spécifiques des desmogléines (Dsg) 1 ou 3. Ces Ac pathogéniques inhibent l'adhésion cellulaire des kératinocytes. Le pemphigus se déclenche par la conjonction d’événements rares impliquant l’émergence puis la coopération de lymphocytes B (LB) et de lymphocytes LT auto-réactifs dans un contexte génétique et environnemental particulier. Jusqu’à présent, la première ligne de traitement du pemphigus était constituée de fortes doses de corticoïdes, qui sont de puissants immunosupresseurs systèmiques. Le Rituximab (RTX), un Ac monoclonal chimérique anti-CD20, constitue une thérapeutique innovante aboutissant à l’élimination des LB. L’étude clinique RITUX 3 a été conçue pour évaluer l’efficacité et l’innocuité du traitement utilisant le RTX associé à une courte corticothérapie dans le traitement de première intention du pemphigus par rapport au traitement de référence par la corticothérapie standard (CS). Dans un premier temps, notre analyse clinico-biologique des patients après 24 mois a démontré que l’utilisation du RTX associé à de la prednisone à court terme en traitement de première intention chez les patients atteints de pemphigus foliacé et vulgaire modéré à sévère est à la fois plus efficace et mieux toléré que le traitement de référence par la prednisone seule (89% de patients versus 34%). Cette efficacité a été confortée à plus long terme après la reconstitution du répertoire lymphocytaire B avec un risque de rechute de 2% à 36 mois. La présence d’une forme sévère de pemphigus au diagnostic (PDAI ≥ 45) et d’un taux d’Ac anti-Dsg à 3 mois supérieur aux valeurs seuils (anti-Dsg1 ≥ 20 ou anti-Dsg3 ≥ 120) sont associés à un risque de rechute précoce de 50%. Ces deux facteurs prédictifs permettent d'identifier un sous-groupe de patients présentant un risque élevé de rechute nécessitant une perfusion d'entretien de RTX au 6ème mois. Dans un deuxième temps, nous avons étudié l’impact des traitements par RTX et par CS chez les patients atteints de pemphigus afin de mieux appréhender la réponse auto-immune. La caractérisation phénotypique des LB auto-réactifs et l’analyse de la fréquence des LB capables de sécréter des immunoglobulines (Ig)G anti-Dsg par une approche ELISPOT a permis d’établir que l’efficacité du traitement par RTX dans le pemphigus semble liée à l’élimination des LB mémoires CD27+IgG+ spécifiques des Dsg. Des LB auto-réactifs Dsg restent détectables après RTX suite à la reconstitution lymphocytaire B, mais ces LB ont un phénotype naïf et non commuté (IgM) et ne secrètent plus d’IgG. En revanche, la persistance des LB auto-réactifs capables de sécréter des IgG anti-Dsg après traitement par CS est certainement à l’origine des rechutes fréquentes. L’analyse de l’expression génique ciblée à l’échelle unicellulaire a démontré qu’initialement, les LB spécifiques des Dsg ont un profil pro-inflammatoire avec l’expression de trois gènes codant pour les interleukines (IL)-1β, IL-12p35 et IL-23p19 et pour le gène de l’IRF5 (Interferon regulatory factor 5) par rapport aux LB non auto-réactifs. Le RTX et la CS ont des effets différents sur l'expression de ces gènes mais les deux réduisent l’expression génique d’IL-1β qui semble jouer un rôle important dans la physiopathologie du pemphigus. Parallèlement, l’analyse transcriptomique puis protéique des LB isolés des patients en rémission complète ou incomplète 6 ans après l’étude RITUX 1 a mis en évidence une augmentation d'expression de KCNN4 (Potassium calcium-activated channel subfamily N member 4) à la surface des LB chez les patients atteints de pemphigus en rémission complète pouvant influencer la maturation des LB
Pemphigus is an autoimmune disease of the skin and mucous membranes caused by autoantibodies (Ab) specific to desmoglein (Dsg) 1 or 3. These pathogenic Ab inhibit cell adhesion of keratinocytes. The development of pemphigus is associated with the conjunction of many uncommon events involving the emergence and then the cooperation of auto-reactive B cells and T cells link to genetic and environmental factors. Until now, the first line of treatment consisted of high doses of corticosteroids. Rituximab (RTX), an anti-CD20 chimeric monoclonal antibody, is an innovative therapy that results in B cells depletion. The RITUX 3 clinical trial was designed to evaluate the efficacy and safety of RTX combined with a short-course glucocorticoid therapy as a first-line treatment of pemphigus versus the standard treatment with standard corticosteroids (CS). As a first step, our clinico-biological analysis of patients after 24 months has shown that the use of RTX combined with short-term prednisone as a first-line treatment in patients with moderate to severe pemphigus is both more effective and better tolerated than the reference treatment with prednisone alone. Respectively, 89% of patients versus 34% in each group and both pemphigus foliaceus and pemphigus vulgaris patients responded. This efficacy was confirmed in the longer term after reconstitution of the B lymphocyte repertoire with a risk of relapse of only 2% at 36 months. The presence of a severe form of pemphigus at diagnosis (PDAI ≥ 45) and an anti-Dsg Ab level at 3 months above threshold values (anti-DSG1 ≥ 20 or anti-DSG3 ≥ 120) are associated with 50% risk of early relapse. These two predictive factors make it possible to identify a subgroup of patients at high risk of relapse requiring a maintenance infusion of RTX at the 6th month. In a second step, we studied the impact of RTX and CS treatments in patients with pemphigus in order to better understand the autoimmune response. The phenotypic characterization of auto-reactive B cells and the analysis of the frequency of B cells able of secreting anti-Dsg immunoglobulin (Ig) G by an ELISPOT approach demonstrated that the efficacy of RTX treatment in pemphigus seems related to the elimination of IgG-switched Dsg memory B-cells. Dsg specific B cells remain detectable after RTX when B cells return, but these B cells have a naïve and non-switched (IgM) phenotype and no longer secrete IgG. On the other hand, the persistence of self-reactive Dsg B cells capable of secreting IgG anti-Dsg after treatment with CS is certainly at the origin of the frequency of relapses. The unicellular targeted gene expression analysis demonstrated that initially, Dsg-specific B cells have a pro-inflammatory profile with the overexpression of three genes encoding Interleukin (IL) -1β, IL-12p35 and IL-23p19 and for the IRF5 gene (Interferon regulatory factor 5) compared to non-self-reactive B cells. RTX and CS have different effects on the expression of these genes, but both reduce the gene expression of IL-1β, which seems to play an important role in the pathophysiology of pemphigus

Os polissacarídeos são os componentes maioritários dos grãos de café verde e torrado e da bebida de café. Os mais abundantes são as galactomananas, seguindo-se as arabinogalactanas. Durante o processo de torra, as galactomananas e arabinogalactanas sofrem modificações estruturais, as quais estão longe de estar completamente elucidadas devido à sua diversidade e à complexidade estrutural dos compostos formados. Durante o processo de torra, as galactomananas e arabinogalactanas reagem com proteínas, ácidos clorogénicos e sacarose, originando compostos castanhos de alto peso molecular contendo nitrogénio, designados de melanoidinas. As melanoidinas do café apresentam diversas atividades biológicas e efeitos benéficos para a saúde. No entanto, a sua estrutura exata e os mecanismos envolvidos na sua formação permanecem desconhecidos, bem como a relação estrutura-atividade biológica. A utilização de sistemas modelo e a análise por espectrometria de massa permitem obter uma visão global e, simultaneamente, detalhada das modificações estruturais nos polissacarídeos do café promovidas pela torra, contribuindo para a elucidação das estruturas e mecanismos de formação das melanoidinas. Com base nesta tese, oligossacarídeos estruturalmente relacionados com a cadeia principal das galactomananas, (β1→4)-Dmanotriose (Man3), e as cadeias laterais das arabinogalactanas, (α1→5)-Larabinotriose (Ara3), isoladamente ou em misturas com ácido 5-Ocafeoilquínico (5-CQA), o ácido clorogénico mais abundante nos grãos de café verde, e péptidos compostos por tirosina e leucina, usados como modelos das proteínas, foram sujeitos a tratamento térmico a seco, mimetizando o processo de torra. A oxidação induzida por radicais hidroxilo (HO•) foi também estudada, uma vez que estes radicais parecem estar envolvidos na modificação dos polissacarídeos durante a torra. A identificação das modificações estruturais induzidas por tratamento térmico e oxidativo dos compostos modelo foi feita por estratégias analíticas baseadas principalmente em espectrometria de massa, mas também em cromatografia líquida. A cromatografia de gás foi usada na análise de açúcares neutros e ligações glicosídicas. Para validar as conclusões obtidas com os compostos modelo, foram também analisadas amostras de polissacarídeos do café obtidas a partir de resíduo de café e café instantâneo. Os resultados obtidos a partir dos oligossacarídeos modelo quando submetidos a tratamento térmico (seco), assim como à oxidação induzida por HO• (em solução), indicam a ocorrência de despolimerização, o que está de acordo com estudos anteriores que reportam a despolimerização das galactomananas e arabinogalactanas do café durante a torra. Foram ainda identificados outros compostos resultantes da quebra do anel de açúcares formados durante o tratamento térmico e oxidativo da Ara3. Por outro lado, o tratamento térmico a seco dos oligossacarídeos modelo (individualmente ou quando misturados) promoveu a formação de oligossacarídeos com um maior grau de polimerização, e também polissacarídeos com novos tipos de ligações glicosídicas, evidenciando a ocorrência de polimerização através reações de transglicosilação não enzimática induzidas por tratamento térmico a seco. As reações de transglicosilação induzidas por tratamento térmico a seco podem ocorrer entre resíduos de açúcares provenientes da mesma origem, mas também de origens diferentes com formação de estruturas híbridas, contendo arabinose e manose como observado nos casos dos compostos modelo usados. Os resultados obtidos a partir de amostras do resíduo de café e de café instantâneo sugerem a presença de polissacarídeos híbridos nestas amostras de café processado, corroborando a ocorrência de transglicosilação durante o processo de torra. Além disso, o estudo de misturas contendo diferentes proporções de cada oligossacarídeo modelo, mimetizando regiões do grão de café com composição distinta em polissacarídeos, sujeitos a diferentes períodos de tratamento térmico, permitiu inferir que diferentes estruturas híbridas e não híbridas podem ser formadas a partir das arabinogalactanas e galactomananas, dependendo da sua distribuição nas paredes celulares do grão e das condições de torra. Estes resultados podem explicar a heterogeneidade de estruturas de melanoidinas formadas durante a torra do café. Os resultados obtidos a partir de misturas modelo contendo um oligossacarídeo (Ara3 ou Man3) e 5-CQA sujeitas a tratamento térmico a seco, assim como de amostras provenientes do resíduo de café, mostraram a formação de compostos híbridos compostos por moléculas de CQA ligadas covalentemente a um número variável de resíduos de açúcar. Além disso, os resultados obtidos a partir da mistura contendo Man3 e 5-CQA mostraram que o CQA atua como catalisador das reações de transglicosilação. Por outro lado, nas misturas modelo contendo um péptido, mesmo contendo também 5-CQA e sujeitas ao mesmo tratamento, observou-se uma diminuição na extensão das reações transglicosilação. Este resultado pode explicar a baixa extensão das reações de transglicosilação não enzimáticas durante a torra nas regiões do grão de café mais ricas em proteínas, apesar dos polissacarídeos serem os componentes maioritários dos grãos de café. A diminuição das reações de transglicosilação na presença de péptidos/proteínas pode dever-se ao facto de os resíduos de açúcares redutores reagirem preferencialmente com os grupos amina de péptidos/proteínas por reação de Maillard, diminuindo o número de resíduos de açúcares redutores disponíveis para as reações de transglicosilação. Além dos compostos já descritos, uma diversidade de outros compostos foram formados a partir dos sistemas modelo, nomeadamente derivados de desidratação formados durante o tratamento térmico a seco. Em conclusão, a tipificação das modificações estruturais promovidas pela torra nos polissacarídeos do café abre o caminho para a compreensão dos mecanismos de formação das melanoidinas e da relação estrutura-atividade destes compostos.
Polysaccharides are the major components of green and roasted coffee beans, and coffee brew. The most abundant ones are galactomannans, followed by arabinogalactans. During the roasting process, galactomannans and arabinogalactans undergo structural modifications that are far to be completely elucidated due to their diversity and complexity of the compounds formed. During the roasting process, galactomannans and arabinogalactans react with proteins, chlorogenic acids, and sucrose, originating high molecular weight brown compounds containing nitrogen, known as melanoidins. Several biological activities and beneficial health effects have been attributed to coffee melanoidins. However, their exact structures and the mechanisms involved in their formation remain unknown, as well as the structure-biological activity relationship. The use of model systems and mass spectrometry analysis allow to obtain an overall view and, simultaneously, detailed, of the structural modifications in coffee polysaccharides promoted by roasting, contributing to the elucidation of the structures and formation mechanisms of melanoidins. Based on this thesis, oligosaccharides structurally related to the backbone of galactomannans, (β1→4)-D-mannotriose, and the side chains of arabinogalactans, (α1→5)-Larabinotriose, alone or in mixtures with 5-O-caffeoylquinic acid, the most abundant chlorogenic acid in green coffee beans, and dipeptides composed by tyrosine and leucine, used as models of proteins, were submitted to dry thermal treatments, mimicking the coffee roasting process. The oxidation induced by hydroxyl radicals (HO•) was also studied, since these radicals seem to be involved in the modification of the polysaccharides during roasting. The identification of the structural modifications induced by thermal and oxidative treatment of the model compounds was performed mostly by mass spectrometry-based analytical strategies, but also using liquid chromatography. Gas chromatography was used in the analysis of neutral sugars and glycosidic linkages. To validate the conclusions achieved with the model compounds, coffee polysaccharide samples obtained from spent coffee grounds and instant coffee were also analysed. The results obtained from the model oligosaccharides when submitted to thermal treatment (dry) or oxidation induced by HO• (in solution) indicate the occurrence of depolymerization, which is in line with previous studies reporting the depolymerization of coffee galactomannans and arabinogalactans during roasting. Compounds resulting from sugar ring cleavage were also formed during thermal treatment and oxidative treatment of Ara3. On the other hand, the dry thermal treatment of the model oligosaccharides (alone or when mixed) promoted the formation of oligosaccharides with a higher degree of polymerization, and also polysaccharides with new type of glycosidic linkages, evidencing the occurrence of polymerization via non-enzymatic transglycosylation reactions induced by dry thermal treatment. The transglycosylation reactions induced by dry thermal treatment can occur between sugar residues from the same origin, but also of different origins, with formation of hybrid structures, containing arabinose and mannose in the case of the model compounds used. The results obtained from spent coffee grounds and instant coffee samples suggest the presence of hybrid polysaccharides in these processed coffee samples, corroborating the occurrence of transglycosylation during the roasting process. Furthermore, the study of mixtures containing different proportions of each model oligosaccharide, mimicking coffee bean regions with distinct polysaccharide composition, subjected to different periods of thermal treatment, allowed to infer that different hybrid and non-hybrid structures may be formed from arabinogalactans and galactomannans, depending on their distribution in the bean cell walls and on roasting conditions. These results may explain the heterogeneity of melanoidins structures formed during coffee roasting. The results obtained from model mixtures containing an oligosaccharide (Ara3 or Man3) and 5-CQA and subjected to dry thermal treatment, as well as samples derived from spent coffee grounds, showed the formation of hybrid compounds composed by CQA molecules covalently linked to a variable number of sugar residues. Moreover, the results obtained from the mixture containing Man3 and 5-CQA showed that CQA acts as catalyst of transglycosylation reactions. On the other hand, in the model mixtures containing a peptide, even if containing 5-CQA and subjected to the same treatment, it was observed a decrease in the extent of transglycosylation reactions. This outcome can explain the low extent of non-enzymatic transglycosylation reactions during roasting in coffee bean regions enriched in proteins, although polysaccharides are the major components of the coffee beans. The decrease of transglycosylation reactions in the presence of peptides/proteins can be related with the preferential reactivity of reducing residues with the amino groups of peptides/proteins by Maillard reaction, decreasing the number of reducing residues available to be directly involved in the transglycosylation reactions. In addition to the compounds already described, a diversity of other compounds were formed from model systems, namely dehydrated derivatives formed during dry thermal treatment. In conclusion, the identification of the structural modifications in coffee polysaccharides promoted by roasting pave the way to the understanding of the mechanisms of formation of melanoidins and structure-activity relationship of these compounds.

Abstract:
La chimiothérapie néoadjuvante (CNA) est utilisée dans les cancers du sein agressifs ou localement avancés (CS). Au delà des bénéfices cliniques, elle représente une opportunité pour monitorer in vivo la sensibilité d’une tumeur à un traitement.A partir de l’analyse de sets de données de patients traités par CNA, nous souhaitons identifier des mécanismes associes à la résistance ou sensibilité au traitement. Dans la première partie, nous avons évalué des paramètres, cliniques, anatomopathologiques et transcriptomiques. Nous avons démontré que des éléments non explorés comme la présence d’embols après CNA revêtaient une information pronostique importante. Dans une 2ème partie, nous avons analysé l’impact de l’infiltrat immunitaire dans le cancer du sein, et avons décrit les changements observés entre des échantillons avant et après CNA. Nous avons montré que l’impact pronostique des TILs était différent avant et après CNA, et était opposé dans les CS triple négatif ou HER2-positif. Finalement, nous avons analysé l’impact des comédications pendant la CNA. Nous avons trouvé des effets positifs – via l’augmentation de l’infiltrat immunitaire et la réponse au traitement – et des effets négatifs avec des effets délétères dans certains sous groupes de patients. En conclusion, la situation néoadjuvante représente une plateforme pour générer et potentiellement valider des hypothèses de recherche. La mise à disposition de jeux de données de patients traités par chimiothérapie néoadjuvante constituerait une ressource majeure pour accélérer la recherche contre le cancer du sein
Neoadjuvant chemotherapy (NAC i.e. chemotherapy before surgery) is increasingly being used for aggressive or locally advanced breast cancer (BCs). Beyond clinical benefits, it represents an opportunity to monitor in vivo sensitivity to treatment. Based on the analysis of datasets of BCs patients treated with NAC, we aimed at identifying mechanisms associated with resistance or sensitivity to treatment.In the first part, we evaluated biological, clinical, pathological and transcriptomic patterns. We demonstrated that unexplored pathological features such as post-NAC lymphovascular invasion may carried an important prognostic information.In a second part, we analyzed impact of imune infiltration in BC and we described extensively the changes of tumor infiltrating lymphocytes (TILs) between pre and post-NAC samples. We showed that the prognostic impact of TILs was different before and after NAC, and was opposite in TNBC and HER2-positive BCs. Finally, we investigated the impact of comedications use during NAC. We found both positive effects - while enhancing immune infiltration and response to treatment - and negative effects with deleterisous oncologic outcomes in specific patients subgroups. In conclusion, the neoadjuvant setting represents a platform to both generate and potentially validate research hypotheses aiming at increasing the efficacy of treatment. The public release of real-life datasets of BC patients treated with NAC would represent a major resource to accelerate BC research

On-line counselling shows promise for making mental health care more available to people in remote locations, people with mobility limitations, and people who are home-based or homebound. In response to the lack of research in this burgeoning new area, Davor Cubranic and I conducted an exploratory, participatory study of on-line counselling in which we used computer-supported distance art therapy as a model. The project was a multidisciplinary collaboration between researchers in counselling psychology and computer science that included 10 additional co-researchers from the community. The three goals of the study were: (a) to develop a computer system specifically for on-line counselling, (b) to evaluate the system for the purpose of bringing forth key issues relating to on-line counselling, and (c) to conduct a cross-disciplinary sharing of knowledge and ideas. The system we developed is an innovative alternative to the usual forms of on-line counselling (e-mail and videoconferencing). It supports audio and visual communication, and yet is suitable for standard home computer systems with dial-in access to the Internet. The study was a pilot project designed in part to put research needs into sharper focus. The issues identified as important during the study were not the ones highlighted in previous literature. Rather, they were: (a) problems associated with technical failure, (b) relevance for people with disabilities, (c) the need for clear communication protocols, (d) lack of shared physical presence, and (e) increased sense of privacy and protection from pre-judgment.

In this study, three kinds of line-type computer art images are created, called line-based Cubism-like image, strip-based Futurism-like image, and rectangle-based Neo-Plasticism-like image. Also proposed are three data hiding techniques for covert communication via these types of art images, respectively. In the creation of line-based Cubism-like images, the longer line segments in a source image are detected and rearranged to form a new 3D-like shape for each color component in the image. In a process of re-coloring the regions in the new image, a data hiding technique is designed skillfully to embed a secret message into the image by keeping the average color of the region unchanged. In the strip-based Futurism-like image creation process, the boundary chain codes of each region yielded by image segmentation are utilized to analyze the region characteristics such as corner points and region directions. By drawing the edges formed by the found corner points, an effect of polygon approximation of the prominent regions in the source image is obtained. Then, each region is partitioned into strips in accordance with the extracted region direction, and a given secret message is hidden into the resulting image by coloring the strips with the white color or the region’s average color in a random fashion. To generate a rectangle-based Neo-Plasticism-like image, a binary space-partition scheme is used to partition a given image into multiple rectangles with the maximum mutual information. Furthermore, two methods are proposed to hide a secret message into the generated art image. One is to limit the image partitioning directions (horizontal and vertical) to follow an alternative order and hide the secret messages into the leaf nodes of a partition tree yielded by the binary space-partition scheme. The other method does not limit the partitioning direction order, and fills the regions with horizontal or vertical color lines to embed the secret message. In addition, for each art image, the user is allowed to select some parameters to create his/her favorite art images. Various security enhancement measures are also proposed to make the embedded data more random to prevent hackers’ attacks. Experimental results showing the feasibility of the proposed methods for art image creation and data hiding applications are also included.

Public art can enhance the visual quality of space. In addition to providing enjoyable art works for our daily life, it can reduce the distance between the art works and us, and then we can interact and dialogue with them. This is the goal of our government to promote the idea of life aesthetics in recent years. However, because of the inappropriate planning and design of public art, this goal can hardly be achieved. In order to have good public art planning and design, this study first, from literature review, determines the fundamental assessment items to be used in the evaluation of public art establishment. Then, by using the Delphi method, the evaluation framework is formulated with five major indexes, including ―theme‖, ―form‖, ―space‖, ―users‖ and ―maintenance‖, and with a total of 17 assessment items for these 5 indexes. Finally, the framework is applied to review and assess the problems encountered in the stations of the Nei-Hu MRT line for its public art establishments and through expert interview and questionnaire, improvement strategies are formulated. It is found that the public art establishment for the most of the stations. is not satisfied in the assessment items of ―function improvement‖, ―visibility‖, ―establishment location‖, ―identity recognition‖ and ―education outreach‖. And the experts suggest, for the item of ―function improvement‖, it can be improved through the overall planning working procedure which the architects invite the artists for the detail design work to make the public arts integrated into the architecture. For the improvement of ―visibility‖ and ―establishment location‖, it is suggested that the public art establishment team should include the member with CIS index system expertise. For the items of ―identity recognition‖ and ―education outreach‖, it can he improved by the promotion of public art education and make the life aesthetics take root in our society.

This study aimed to discuss the feasibility of applying“Line-exploration teaching programs”to the Art and Humanities classes in junior high schools, and how it would influence on students' creativity.To achieve the goal, the study is proceeded with literature review to understand the theory bases of creativity and visual art based on the Line-exploration teaching programs.This research was designed according to“Experimental Research”. The study object was 59 students in two classes of seventh grade of Dong-Hai Junior School, Taitung County. Students were randomly separated into experiement group and control group.Students of experiement group accepted the “Line-exploration teaching programs”,while students of control group were taught in traditional ways. The experiment lasted eight weeks.To compare the difference of study efficiency in the Art and Humanities classes and the creavity influence between the experiement group and control group, linear achievement test, creativity performance assessment, William Creative Assessment Packet and study questionnaire were used as scale.   The results of this study showed as follows:1.Line-exploration teaching programs are applicable to the Art and Humanities classes in junior high schools;2. Line-exploration teaching programs can promote to the study efficiency of junior high school students in Art and Humanities classes;3.Line-exploration teaching programs can promote to creativity of parts of junior high school students.

City space demonstrated a combined mechanism of art and architecture before the 20th century, that was, friendly humane space. While the notion of mechanism means everything in a modern city, art culture is deviating further from city space. Under the circumstance, space is becoming isolated and cold left alone humanity. The space design of Taipei Rapid Transit System is to be free from the fate. Therefore, the competent authority strives to promote and build up public art so the “public art of rapid transit system” may exercise the functions of environmental beautification, space softening, and human nature reformation. Can public art make a difference to the space in rapid transit stations? It is the sole purpose of this study. The interrelation of space (rapid transit stations), art works (public art), and passengers (of Taipei Rapid Transit System) is the focus of this study. First of all, investigate the space design and public art on Taipie-Tansui line and Hsintien-Chungho line. Then, find out the original idea of creator in public art by interviewing him. Conduct random survey by designing the questionnaires in accordance with the findings. Finally, implement the analysis of “space perception ” to understand the similarity and difference of public art between the expectation of creator and the feeling of user, furthermore, add the study result on passenger’s attitude in the theory pertaining to the function and trait of public art for study. Verify the role of Taipei Rapid Transit System in public art with conclusions and suggestions reached for the reference of public. The chapters of this study are summarized as follows: Chapter 1 Introduction Collection and review of literature is to discover research motive and substantiate research purpose, furthermore, to draft up the research subject of research scope and sensibility and attitude. Develop and research the direction of research content and the research structure of “space, art, and people.” Moreover, the method and procedure of study is introduced in the last paragraph. Chapter 2 The foundation of theory and literature review The chapter is divided into two sections. The relation structure of “space, art, and people” is introduced in the section of “foundation of theory.” The concept of substantive space form and non-substantive space is discussed in the study of “space.” The psychical relation between creator and admirer is reviewed in the study of “art.” Sensibility and attitude of people towards space and the evaluation method for space sensibility is discussed in the study of “people.” In the section of “defining the role of public art,” the interpretation of “public art” now and past is defined through literature review. Chapter 3 Research positivism, survey, and analysis Investigate the “environment,” “space,” and “public artworks” of Taipei-Tansui line and Hsintien-Chungho line of Taipei Rapid Transit System. The content of investigation includes “analysis of surrounding environment,” “analysis of space,” and the “artworks and classification of space structure” in stations. Chapter 4 Interview and questionnaires survey Creators of “rapid transit system’s public art” are targeted for interview. Collect the adjectives wanted by creators for “space perception.” Prepare questionnaires to survey the attitude of passengers in order to analyze the “consistency” of their attitude and opinions. abstract: A central concept of combinatorics is partitioning structures with given constraints. Partitions of on-line posets and on-line graphs, which are dynamic versions of the more familiar static structures posets and graphs, are examined. In the on-line setting, vertices are continually added to a poset or graph while a chain partition or coloring (respectively) is maintained. %The optima of the static cases cannot be achieved in the on-line setting. Both upper and lower bounds for the optimum of the number of chains needed to partition a width $w$ on-line poset exist. Kierstead's upper bound of $\frac{5^w-1}{4}$ was improved to $w^{14 \lg w}$ by Bosek and Krawczyk. This is improved to $w^{3+6.5 \lg w}$ by employing the First-Fit algorithm on a family of restricted posets (expanding on the work of Bosek and Krawczyk) . Namely, the family of ladder-free posets where the $m$-ladder is the transitive closure of the union of two incomparable chains $x_1\le\dots\le x_m$, $y_1\le\dots\le y_m$ and the set of comparabilities $\{x_1\le y_1,\dots, x_m\le y_m\}$. No upper bound on the number of colors needed to color a general on-line graph exists. To lay this fact plain, the performance of on-line coloring of trees is shown to be particularly problematic. There are trees that require $n$ colors to color on-line for any positive integer $n$. Furthermore, there are trees that usually require many colors to color on-line even if they are presented without any particular strategy. For restricted families of graphs, upper and lower bounds for the optimum number of colors needed to maintain an on-line coloring exist. In particular, circular arc graphs can be colored on-line using less than 8 times the optimum number from the static case. This follows from the work of Pemmaraju, Raman, and Varadarajan in on-line coloring of interval graphs.

In the modern manufacturing environment, customer-order-driven has become the most popular production type. For those important orders, delay of the final product will lead to the degrading of the customer royalty and the business credibility. Also the early finished product will cost the company to find a warehouse to store the end-product before they are delivered to customers. Therefore it is important to assign appropriate due date to orders and to determine an efficient production scheduling simultaneously. Thus, this research presents a novel ant colony optimization (ACO) model for single machine scheduling and permutation flow shop scheduling problems. The ACO algorithm determines how to insert arriving jobs into the waiting line and to be operated by the idle machine(s). Meanwhile two due date quotation strategies for both single machine and permutation flowshop environment respectively are embedded with the proposed algorithm to set the due date for each job once it arrives. Three sets of large-size instances consisting of 100, 500, and 2500 jobs, and one (single machine), three and five (flowshop) machines respectively are tested with three distributions for processing time and inter-arrival time of each job. The performance of the ACO algorithm is compared with a heuristic algorithm Hi. The computational results show that ACO+EDD performs competitively particularly when the problem size increases.
