Full textAn increasing number of parents are opting-out their children from high-stakes. Accountability systems in education have used students? test scores to measure student learning, teacher effectiveness, and school district performance. Students who are opted-out of high-stakes tests are not being evaluated by the state tests, making their level of achievement or proficiency unknown by the state government. The purpose of this basic interpretive qualitative study was to gain an understanding of the various reasons, factors, experiences, and personal events that led parents to opt-out their children from at least one 3rd through 8th grade high-stakes test. Data were collected using a researcher-designed semi-structured interview protocol developed using ecological approaches to systems theories and critical pedagogy theories. The study was set in New York and 10 participants were interviewed, all from different rural or small suburban school districts throughout the state. Five themes and 12 subthemes emerged from first and second cycle coding. Key findings indicated that parents decided to opt-out their children from high-stakes tests because they felt high-stakes were inappropriate and unfair. Further, parents were dissatisfied with current high-stakes testing practices. Previous 3rd through 8th grade testing procedures that allowed teachers to make and grade the state tests were seen as acceptable. Parents indicated no issue with testing. However, from a social change perspective they felt the current system of high stakes testing was used improperly to rate students, teachers, programs, and school districts, and that testing should be used to drive instruction and help struggling students. This study is beneficial for school personnel and policy makers because it provides different ways to assess student achievement.

Full textAfter a few decades of women entering the labor market, it was expected that gender equity would already be a reality in organizations. However, men are still the majority and the differences are worse at the top positions. In Brazil, according to IBGE, in 2016, women represented only 37,8% of the management positions in the state and private companies and their wages were 25% lower than men\'s. To increase the presence of women in organizations should be seen as an economic problem, since they represent talents that can contribute to the increase of productivity and financial results of the organizations, generating positive impacts throughout the economy. In fact, most women have non-linear careers and make career changes. The \"opt out\" term was used to explain the evasion of highly skilled women from organizations. Maternity and family reasons were considered the main reasons for these career changes. However, Mainiero and Sullivan (2006) found out that the opt out reasons were more complex. Interesting opportunities, recognition and balance between personal and professional life were considered very important factors for women\'s lives. As a result of this research, they developed the kaleidoscope career model. Based on the kaleidoscope\'s system, it considers that career decisions depend on three pillars: authenticity, balance and challenge. People adjust their careers according to the importance of each of these pillars throughout their lives. Based on the kaleidoscope career model, this qualitative research examined the factors that influence the career decisions of executive women. In the theoretical reference, 3 main constructs were considered: career, gender and corporate environment. There were 18 unstructured interviews with 2 mid-career women groups and at least 1 child: women who remained in the corporate career and women who changed their careers. The analysis of the results was done in two parts: the chronological analysis of the careers of these women and the content analysis with the categorization and analysis of the answers obtained. The findings showed that women\'s career choices are extremely complex and depend on a number of factors. Personal and professional life should be analyzed in an integrated way and motherhood is not the decisive factor for women\'s career changes. The kaleidoscope career model showed to be quite adherent to the analysis performed and the authenticity, balance and challenge pillars were prioritized according to each moment. Positive organizational climate and leadership, corporate policies, family and partner support and the pillars of authenticity, challenge and balance are some of the factors that directly contribute to the permanence in the corporate career. Difficulties, dissatisfaction and imbalances in the cornerstones of the kaleidoscope career can lead women to make significant career decisions and, in some cases, opt out.

Full textAs proteínas Ohr (Organic hydroperoxide resistance) são peroxidases dependente de tiól extremamente eficientes e têm um papel central na resposta das bactérias contra peróxidos orgânicos. Em fungos, as proteínas Ohr apresentam uma extensão N-terminal, cujo predições in silico apontam estar associada ao direcionamento da proteína para a mitocôndria. A tríade catalítica é composta pela cisteína peroxidatic (Cp), a arginina (Rc) e o glutamato (Ec) catalíticos que são totalmente conservados e interagem entre eles por uma rede de interações de ponte salina, na forma reduzida da proteína (conformação fechada). Após se tornarem oxidadas em ácido sulfênico (Cis-SOH), a Cp condensa com o grupo sulfidrila da cisteína de resolução (Cr) numa ligação disulfeto. A ausência da carga negativa do tiolato (RS-) da Cp facilita a abertura da alça que contem a Rc para longe do centro ativo, gerando a conformação aberta. No entanto, os eventos moleculares associados a alta reatividade das enzimas Ohr contra hidroperóxidos e a sua redução pela dihydrolipoamida (presente em proteínas lipoiladas), ainda está descrita de forma bem superficial. Adicionalmente, vários fatores suportam a ideia de que a Ohr seria um potencial alvo para o desenvolvimento de drogas: (i) a Ohr exibe propriedade físico-químicas únicas; (ii) as bactérias mutantes para Ohr (Δohr) são fortemente sensíveis ao stress oxidativo; (iii) indicações de que a Ohr poderá está envolvida na virulência de várias bactérias; e (iv) a ausência de Ohr em mamíferos e plantas vascularizadas. Nesta tese, vários aspetos relacionados com as enzimas Ohr foram avaliados. No Capitulo 2, foi caracterizada bioquimicamente a proteína Ohr homologa de fungo ascomiceto, Mycosphaerella fijiensis Mf_1 (MfOhr), o agente causador da doença de bananas, Sigatoka-negra. A enzima apresentou eficiente atividade contra peroxido de ácido linoleico (kobs = 3.18 (± 2.13) ×108 M-1.s-1). Além disso, através do fracionamento sub celular de protoblasto de M fijiensis seguido de western blot, foram confirmadas as predições in silico de que a MfOhr é uma proteína mitocondrial. No capítulo 3 e 4, foram descritas sete estruturas cristalográficas oriundas de dois patógenos oportunistas, uma de Xylella fastidiosa e seis de Chromobacterium violaceum (incluindo o primeiro representante do complexo entre a Ohr e o seu redutor biológico, DHL). Estas estruturas poderão representar diferentes conformações ao longo do ciclo catalítico. Adicionalmente, várias abordagens de modelagem molecular, tais como mecânica clássica (MM), mecânica molecular direcionada (SMM) e mecânica quântica híbrida (QM-MM), juntamente com ensaios experimentais com mutações pontuais, indicaram que a Ohr sofre várias mudanças conformacionais para permitir uma abertura intermitente (estado oxidado) e o retorno para uma conformação fechada mais estável (estado reduzido) da alça da arginina ao longo da catálise. Notavelmente, a dihydrolipoamide assistiu diretamente o fechamento da alça da arginina e por consequência o turnover da enzima. No capítulo 5, foi descrita a identificação de dois compostos (C-31 e C-42) que representam estudos iniciais com a finalidade de encontrar inibidores específicos para a enzima Ohr. Estes compostos foram encontrados por ab initio design e por varrimento virtual com o uso de modelos farmacofóricos. Os IC50 calculados para o C-31 e C-42 foram de 124.4-248.5 µM e 243.3-321.7 µM, respectivamente. Finalmente, esta tese descreve vários aspetos relacionados com a função da Ohr: 1 - evidências que as Ohr de eucariotos estão preferencialmente localizadas na mitocôndria e partilham várias propriedades bioquímicas com as Ohr de bactéria; 2 - a rede de interações polares entre os resíduos da tríade catalítica (Cp, Rc e Ec) contribuem fortemente para a estabilização do estado fechado, a configuração ótima para a redução de hydroperoxidos; 3 - evidências de que a formação da ligação disulfeto e a liberação do produto (álcool derivado da redução do hydroperoxido) facilitam a abertura da alça da arginina até um estado intermediários (provavelmente não o estado totalmente exposto apresentado nas estruturas cristalográficas) 4 - o mapeamento das interações entre o redutor biológico no centro ativo da Ohr; 5 - fortes indicações de que a DHL não é capaz de interagir e reagir com a Ohr na conformação fechada; 6 - os primeiros ensaios para a procura por moléculas que especificamente interajam com a Ohr, apesar de que futuros ensaios terão de ser executados para verificar a especificidade dos compostos selecionados. Assim, nós descrevemos nova informação relevante sobre uma proteína antioxidante que exibe uma alta eficiência catalítica, comparável com outras importantes enzimas removedores de hydroperoxidos, tais como glutationa peroxidases e peroxiredoxinas

Full textThis research deals with the participation effects of employees on firm performance. This thesis explores three dimensions of employees' participation: economic (employee ownership), political (involvement in corporate governance structure) and organizational. In the first part, this research theoretically examines effects of employees' participation using different theoretical frameworks. At the end of the first part, some research hypotheses are formulated. In a second part, this research empirically examines participation effects on firm performance thanks to statistical tests made on two samples. The first sample is based on 189 French listed firms on 2001-2004. The second one, cross sectional, investigates more specifically political representation of employees on corporate governance structure. The majors results show that : 1/ the relationship between employee ownership and firm performance is curvilinear. 2/ board employee representation negatively moderates employee ownership - firm performance relationship. In a third part, a theoretical model, combining empirical results and theoretical in-depth analysis, is proposed

Resumo: A farmacodinâmica vascular da síntese endógena de NO ficou conhecida devido ao efeito de drogas nitrovasodilatadoras. Esses agentes exógenos, tais como o nitroprussiato de sódio, agem como doadores de NO e exercem efeito hipotensivo in vivo direto promovendo diminuição na resistência vascular sistêmica decorrente de um aumento nos níveis de GMP cíclico. Nosso interesse reside na possibilidade de complexos de rutênio transnitrosilaminados servirem como veículo para a liberação controlada de NO em sistemas biológicos e, assim, produzir efeitos farmacológicos. O objetivo desse trabalho foi investigar o efeito hipotensor e a toxicidade de um novo complexo de rutênio, o t-[Ru(NH3)4P(OEt)3NO](PF6)3 devido a sua habilidade em liberar NO espontaneamente em solução aquosa. Medidas diretas de PA foram realizadas em ratos Wistar machos adultos cronicamente instrumentados. O efeito hipotensor foi determinado em ratos conscientes (n=10) submetidos a injeções endovenosas "in bolus" de trans-[Ru(NH3)4P(OEt)3NO](PF6)3 e nitroprussiato de sódio (NaNP). Concentrações de 0,0025 a 0,08 µmoles/Kg do complexo de rutênio induziram resposta hipotensora imediata (PAM controle 104 ± 4,1 mmHg) em uma faixa de ação de 7 a 27%, similar ao efeito observado para o NaNP (11a 32%). Uma correlação dose-efeito positiva foi observada. A toxicidade foi avaliada em cultura de células (IC50) e pela toxicidade aguda em camundongos (LD50). Estudos da citotoxicidade foram conduzidos em cultura de hepatócitos (células metabolizantes), V79 (células não metabolizantes) e V79 tratada com o complexo biotransformado pelos hepatócitos. Hepatócitos de rato foram isolados pelo método de perfusão com colagenase em duas etapas do fígado inteiro. A viabilidade celular foi avaliada por três ensaios, os quais são indicativos de citotoxicidade: redução do MTT, incorporação do vermelho neutro (VN) e conteúdo de ácidos nuclêicos (DNA). Esses ensaios permitiram a avaliação dos valores de IC50 para o trans-[Ru(NH3)4P(OEt)3NO](PF6)3 em células V79 e hepatócitos. Na cultura de hepatócitos valores de IC50 superiores a 3,0 mM foram obtidos para todos os ensaios realizados, sugerindo uma baixa toxicidade do complexo. Para cultura de células V79, nossos resultados mostraram efeito citotóxico similar para os três ensaios investigados, com valores de IC50 ao redor de 2,0 mM, mostrando ser este composto menos tóxico que o NaNP (IC50 ~ 0,056 mM). O produto da biorransformação foi avaliado e mostrou baixa toxicidade para os ensaios de DNA, MTT e alta toxicidade para o VN, i.e., o composto biotransformado modificou a integridade lisossomal. A toxicidade aguda foi avaliada e um valor de LD50 igual a 216 mg/Kg foi obtido indicando baixa toxicidade do complexo in vivo.
Abstract: The vascular pharmacology of endogenously synthesised NO is largely predicted by the known effects of nitrovasodilator drugs. These exogenous agents, such as sodium nitroprusside, act as NO donors and exert direct hypotensive effects in vivo promoting a decrease in systemic vascular resistance by increasing levels of cyclic GMP. We have been interested in the possibility that trans-nitrosylammine ruthenium complexes, might serve as vehicles for the controlled delivery of NO into a biological system and thus display useful pharmacological effects, promoting hypotensive effect. The aim of this work was to investigate the hypotensor effect and toxicity of a new ruthenium complex t-[Ru(NH3)4P(OEt)3NO](PF6)3 due to their ability to spontaneously NO release in aqueous solution. Direct measurements of blood pressure were performed in chronically instrumented adult male Wistar rats. The hypotensor effect was determined in conscious rats (n=10) submitted to in bolus intravenous doses of the trans-[Ru(NH3)4P(OEt)3NO](PF6)3 and sodium nitroprusside (NaNP). Concentration range of 0.0025 to 0.08 µmols/Kg induced hypotensive response (MAP control: 104 ±4.1 mmHg) immediately in action range of 7 to 27% similar to that observed for NaNP effect (11 to 32%). A positive dose-effect correlation was observed. The toxicity was evaluated on cell culture (IC50) and by acute dose in mice. Cytotoxicity studies were carried out on cultures of rat hepatocytes (metabolizant cells), V79 (non-metabolizant cells) and V79 treated with the supernatant of the biotransformed complex by hepatocytes. Hepatocytes were isolated from adult male Wistar rats by the two-step collagenase liver perfusion. Cell viability was evaluated by three endpoints which are indicative of cytotoxicity: MTT reduction, Neutral Red Uptake (NRU) and Nucleic Acid Content (DNA). These assays permit the evaluation of IC50 values of trans- [Ru(NH3)4P(OEt)3NO](PF6)3 on V79 and rat hepatocyte cells. On hepatocyte culture values higher than 3.0 mM were determined for all tests, suggesting a low toxicity for the complex. For V79 cultures our results showed a similar cytotoxic effects for the three studied endpoints with an IC50 ranging around 2.0 mM, showing be less toxic than NaNP. The biotransformated product was evaluted and showed low toxicity for DNA, MTT assays and high toxicity for NRU, i.e., the compound biotransformed changed the lysosomal integrity. The acute toxicity was carried out and a LD50 value of 216 mg/Kg was obtained indicating a low toxicity in vivo.

Introduction: Critical Power (CP) is the maximum rate can be maintained in a stable metabolic state muscle work. In the search for a test associated with less error and greater practical application, a series of studies investigating the validity of a single maximal test "all-out" to determine the PC. This test was designed with the assumption that, by inducing the depletion of anaerobic reservations as early as possible, the aerobic system predominantly assume the replenishment of ATP and consequently the amount of work done in this situation will be the next PC. Thus this study aims to explore the impact of hypoxia (FiO2 = 0,13), in determining the PC by 3-MT test. Methods: 8 healthy university students were evaluated (7 men and 1 woman). Incremental ramp test for determination of the maximal and submaximal parameters as well as the braking factor (power +50% Δ /pacing2 ) test three minutes to determine the maximum EP and W` in normoxia and hypoxia (Fi O2 = 0.13) conditions, constant load tests at 100% of EP determined in three minutes maximum. Student t test for the measurements between tests. Results: The main hypothesis of the study was not accepted since the addition of hypoxia did not cause enough stress to the point of changing the anaerobic contribution and interfere with the determination of EP (3-MTHYP = 213±43W, 3-MTNOR = 217±49 Wp = 0.35). There was no significant difference during the constant load tests at 100% of EP (EP 3-MTHYP = 23.95±8 min, EP-3 MTNOR = 19.41±10 min p = 0.36). Conclusion: Adding hypoxia (Fi O2 = 0.13) was not able to change the energy contribution to the point of changing the parameter EP aerobic and anaerobic W `, highlighting the robustness of the model against this intervention. Still, the responses associated with RPE during constant-load test are close to those encountered during constant load tests on PC. Further investigations with different levels of hypoxia are necessary for more conclusive answers.
Introdução: Potência crítica (PC) representa a máxima taxa de trabalho muscular possível de ser mantida em um estado estável metabólico. Na busca por um teste com menor erro associado e maior aplicação prática, uma série de trabalhos vem investigando a validade de um único teste máximo all-out para determinar a PC. Este teste foi concebido com o pressuposto de que, induzindo o esgotamento das reservas anaeróbias o quanto antes possível, o sistema aeróbio assumiria predominantemente a reposição de ATP e consequentemente a carga de trabalho realizada nessa situação será próxima a PC. Assim este estudo, tem como objetivo explorar o impacto da hipóxia (i.e. FiO2 0.13), na determinação da PC pelo teste máximo de três minutos (3-MT). Métodos: Oito universitários saudáveis foram avaliados (7 homens e 1 mulher). Teste incremental de rampa foi utilizado para determinação dos parâmetros máximos e submáximo bem como do fator de frenagem (potencia+50%∆ / cadendia2). Teste de 3-MT para determinação do EP e W` nas condições normoxia e hipóxia (Fi O2=0,13). Testes de carga constante a 100% dos EP foram utilizados para verificação das respostas fisiológicas. O teste t de student foi utilizado para as aferições entre os testes. Resultados: A hipótese principal do estudo não foi aceita, uma vez que a adição de hipóxia não gerou estresse suficiente ao ponto de alterar a contribuição anaeróbia e interferir na determinação do EP (3-MTHYP = 213±43W, 3-MTNOR = 217±49W p=0,35) . Também não houve diferença significativa durante os testes de carga constante a 100% dos EP determinados (EP 3-MTHYP = 23,95±8 min., EP 3-MTnor = 19,41±10 min. p= 0,36). Conclusão: A adição de hipóxia (Fi O2=0,13) não foi capaz de alterar a contribuição energética a ponto de alterar o parâmetro aeróbio EP, e anaeróbio W`, destacando a robustez do modelo frente a essa intervenção. Ainda, as respostas associadas ao EP durante o teste de carga constante estão próximas aquelas encontradas durante testes de carga constante na PC. Futuras investigações com diferentes níveis de hipóxia são necessárias para obter respostas mais conclusivas.

Full textIn the modern societies, the answer to the criminal act is a prison sentence. Due to the nature of certain acts, with this prison sentence, some court-ordered treatments (COT) (obligations and injunctions of treatments) have been added. Ethically, this law request and its social asking don’t exempt to think about the prisoner ‘asking, which is the trigger of psychotherapies. There is any retroactivity of the law and of pronounced sentence. For this research, the prisoners were selected on their own asking of care (the court-ordered treatments couldn’t applicate). The asking of the prisoners is therefore a criterion common to them (these prisoners). In this context, the acts have distinguished according to the mechanisms which have governed the nature of the act: recourse of act or acting out. Many experts agree on the existence of premature narcissistic failures. However, discussions persist on the nature of the act and on its aspect, either single dimension or multiple and complex dimensions (objective behavior, social consequences and underlying mechanisms ...). Yet all these theoretical models on narcissistic failures lean on the “recourse to act” as if there was only a simple nature to act

Full textDuring an earthquake, an interaction between the in-plane and out-of-plane seismic forces occurs and the infilled frames suffer damage in both in-plane and out-of-plane directions simultaneously. Particularly, the out-of-plane collapse of unreinforced masonry infill walls is critical even for new buildings complying with the modern seismic codes, resulting in high casualties and huge economic losses. However, the out-of-plane behaviour of infill walls is yet not fully understood. This study is therefore aimed towards characterizing the out-of-plane seismic capacity of unreinforced masonry infill walls. First of all, available out-of-plane experimental tests performed on unreinforced masonry infill walls are reviewed with a detailed comparison of the experimental results. The influence of parameters like slenderness ratio, aspect ratio, boundary conditions, openings, vertical load, in-plane damage level, the strength of masonry and plaster, and frame stiffness are evaluated, and research gaps are identified. Based on the collected experiments, all available analytical capacity models are checked for their accuracy in the prediction of the out-of-plane capacity of unreinforced masonry infill walls. In doing so, both types of capacity models are evaluated: Type (I) for the estimation of the out-of-plane strength in the in-plane undamaged state; Type-II for the estimation of out-of-plane strength reduction factor for the in-plane damaged state. Afterwards, the best pairs of models from two groups i.e. Type I and Type II, are coupled and checked with the experimented specimens where the reference infill specimen (specimen tested in out-of-plane without prior in-plane damage) is not available. In addition, the influence of orthotropy of the infill masonry in the out-of-plane capacity predicted by the capacity models is analysed. The possibility of using the capacity models in the cases of infill-beam gap and infill with openings is also checked. Different available macro-modelling techniques are investigated and a simple macro-element model which can simulate the behaviour of unreinforced masonry infill walls under in-plane and out-of-plane loads is developed. The model is validated with different sets of experiments. The model takes into account the decrease in out-of-plane capacity due to prior in-plane damage and is capable to capture in-plane/out-of-plane interaction effects of the seismic forces. From the correlation between the experimental and macro-model results, empirical equations are developed that can be used to calculate the stress-strain parameters required for defining the compressive behaviour of the struts. With the provided strategy, the geometrical and mechanical parameters required for the struts can be easily identified for numerical modelling of the infill wall. Using the model, in-plane and out-of-plane responses of the infill wall in lateral loads can be checked. To enrich the information obtained from the experiments regarding the out-of-plane behaviour of infill walls, numerical experimentation is performed by using the developed macro-model covering the range of infill’s geometrical and mechanical properties. From the detailed parametric analysis, the out-of-plane strength of the infill wall is found to be largely influenced by compressive strength, slenderness ratio, aspect ratio, and more importantly by the level of in-plane damage. The decay of strength and stiffness due to prior in-plane damage is also largely governed by the strength and the slenderness ratio of the unreinforced masonry infill. Based on the numerical results, empirical equations are proposed for the evaluation of the infilled frame’s out-of-plane capacity under in-plane damaged or undamaged conditions. The reliability of the proposed equations is proved by comparisons with experimental results. Finally, a procedure for developing the out-of-plane fragility functions is proposed by using the developed macro-model. The fragility is calculated assuming the uncertainty in the geometric and mechanical properties of infill walls instead of the uncertainty in the seismic input. The fragility is defined with respect to the position of the infill wall in a low-rise RC building. Experimental data available in the literature are used for the validation of the output. Overall, the results indicated lower vulnerability in the out-of-plane direction for infill walls without prior in-plane damage and high vulnerability when the infill wall is prior damaged in the in-plane. The proposed procedure can be extended to other types of infill walls depending on the construction technique of the site of interest, obtaining different and specific fragility curves for perming a large-scale risk analysis.

O propósito deste estudo foi avaliar, in vitro, a influência de remanescente de materiais obturadores endodônticos, após desobturação e preparo para pino, sobre a resistência de união adesiva de pinos de fibra de vidro cimentados. Foram utilizados 60 dentes humanos unirradiculados sem coroa. As raízes foram preparadas pela técnica coroa-ápice com sistema Reciproc. Para obturação, as raízes foram divididas em grupos (n = 10) de acordo com o cimento endodôntico utilizado: SO= sem obturação; GP=guta percha sem cimento; AHP= guta-percha + AHPlus; SLP= guta-percha + Sealapex; IRT= guta-percha + iRoot SP; e ERZ= guta-percha + EndoRez. As raízes foram obturadas pela técnica do cone único. A cimentação dos pinos de fibra de vidro Rebilda n.12 foi realizada com sistema adesivo futurabond DC e cimento Rebilda DC em 10 mm do comprimento da raiz. Após esse período, as raízes foram seccionadas obtendo-se discos de 2 mm de espessura em cada terço da raiz (3 discos por raiz) que foram submetidos ao teste de push out, análise por meio de uma lupa estereoscópica (40x) para identificação através de scores da presença de remanescente de material obturador na luz do canal e avaliação por espectroscopia de energia dispersiva (EDS) para identificação de remanescente de componentes químicos do cimento endodôntico presentes nos túbulos dentinários. Os dados obtidos foram analisados estatisticamente pelos testes de Kruskal Wallis, Qui Quadrado e teste de Anova e Tukey, todos com nível de significância de 5%. Não houve diferença estatística para a resistência adesiva entre os cimentos na região cervical (p = 0,49), media (p = 0,28) e apical (p = 0,09). Os grupos SLP e ERZ apresentaram menos remanescente de material obturador nas regiões cervical (p = 0,045) e apical (p = 0,006). Significantes diferenças de remanescentes de componentes químicos foram observadas na região apical dos grupos AHP, IRT e SLP. Conclui-se que os materiais endodônticos não influenciaram na resistência de união adesiva de pinos de fibra de vidro.
The purpose of this study was to evaluate the influence of remnants endodontic filling material in root canal on the bond strength of fiberglass posts cemented. Sixty single-rooted human teeth were used.Crowns were removed. The roots were prepared by crown-down technique with Reciproc system. For fillings, the roots were divided (n = 10) according to the sealer used: SO- without fillings; GP-gutta-percha without sealer; AHP- gutta-percha + AHPlus; SLP- gutta-percha + Sealapex; IRT-gutta-percha + iRoot SP; ERZ-and gutta-percha + EndoRez. Single cone obturation technique was used. After root canal fillings, the teeth were kept in an oven 37 0C for 7 days. Cementing fiberglass posts Rebilda n.12 was performed with fiber adhesive system Futurabond DC and cement Rebilda DC in 10 mm of root length, getting stored in an oven at 37 0C for 48 h. After this period, roots were sectioned obtaining 1 disc of 2 mm thickness in each third of the root (3 discs per root), this discs were underwent to push out test, analyzed by means of a stereomicroscope (40x) to identify the presence of remnants of filling material in the lumen of the canal, and then evaluated by energy dispersive spectroscopy (EDS) for identification of chemical components remnants sealer present in the dentinal tubules. Datas were analyzed with Kruskal-Wallis test, Chi square test, and also Anova and Tukey test, 5% of significance. There were no significant differences in push-out bond strengths among the different groups in the coronal (p = 0.49), middle (p = 0.28) or apical third (p = 0.09). Sealeapex and EndoRez groups showed significantly less remnants of filling material at coronal (p = 0.045) and apical levels (p = 0.006).Significant differences of chemical components remnants were found in AHP, IRT and SLP groups on apical third. It is concluded that residual endodontic filling material does not influence on the bond strength of fiberglass.

Full textThe ratification patterns at International Labor Organization (ILO) Conventions and coalitions on minimum International Labor Standards throughout the years can be explained by David Ricardo\'s theory of comparative advantage, Heckser-Olin model on exports and the distributional effects of the Stolper-Samuelson theorem. Over the last years, literature has examined variables that could potentially affect States\' decision to ratify or not ILO Conventions. Under the international political economy perspective, variables such as endowment and investment capability were previously used in the model on free trade coalitions, designed by Rogowski. In this article, educational attainment is added to a theoretical model that team up countries according to their most abundant internal factor of production, investment capability and educational attainment, guiding their support or disapproval of international minimum labor standards. The theoretical model was tested by a descriptive analysis of ratification patterns for which a database was built, combining official data and original premises of comparison. The database could be used to organize countries by any criterion, and for the purposes of this article the conclusion is that factor endowment and investment capacity are relevant to the formation of coalitions on minimum International Labor Standards at ILO, whereas education attainment is irrelevant.

Full textThis study evaluated the push-out bond strength of glass fiber posts adhesively bonded to intraradicular dentin using a standard unfilled resin as luting agent in combination with an experimental and a commercially available adhesive. Twenty extracted single-rooted human teeth were endodontically treated. Specimens were assigned to 4 dentin bonding strategies: Group 1 (co-monomers to ethanol on ratio 1:1) and Group 2 (co-monomers to ethanol on ratio 1:2): Experimental adhesives applied in accordance with the simplified ethanol-wet bonding technique; Group 3: One-Step Plus (OSP) applied in accordance with the simplified ethanol-wet bonding technique; Group 4: OSP applied following manufacturers instructions. Bonded specimens were sectioned in 1-mm-thick slices and the push-out test was performed. Representative specimens were selected for ultrastructural examination. No statistical differences in push-out bond strength were found among the tested bonding strategies (p>0.05). The major failure pattern was between the post and the luting cement. From this study the ethanol technique should be considered as a promising approach in the luting of fiber posts into the root canal due to the possible benefits of highly hydrophobic resins. Further studies are needed to validate the use of the ethanol-wet bonding technique when luting a fiber post.

Full textA customer-oriented strategy is widely believed to improve organizational performance. But there are few research papers on CRM / performance link, as the concept of Customer Relationship Management is fairly recent and difficult to define. To this end, this research sets out to propose an in-depth analysis of the CRM concept and to analyze the relationship between the three forms of CRM identified (operational CRM, analytical CRM and strategic CRM), the IT strategy/structure and organizational performance, using the strategic alignment perspective. In view of this, an empirical study was conducted among 138 firms to validate our research model linking CRM orientation and technological deployment to performance. Using Structural Equation Modeling techniques, a “direct model” was tested and then, a “global model” including the “co-alignment” latent variable. The main thrust of the findings indicate that the co-alignment between CRM orientation and technological deployment enhance business performance in terms of growth and profitability. Furthermore, we identified a different profile of technological deployment for each form of CRM

Full textSeveral researchers have indicated that the impact of globalization and modern information and communication technologies transformed the notion of how work is done in the organizations. In the present context, previous promises of job stability can no longer be sustained, resulting in the need to rethink the paradigms of professional development. In this scenario, it comes into play a concept called boundaryless careers, in which individuals move more freely among different organizations, dissociating their identities from specific employers and incorporating other dimensions of life - beyond their professional activities - as essential elements for their personal fulfillment. Furthermore, the new paradigm accepts temporary career interruptions in a more natural way, considering them as part of the evolution of professional and personal lives. In that regard, American researchers are increasingly investigating a movement called \"opt-out revolution\", describing highly qualified and highly performing individuals who voluntarily make a career decision contrary to the traditional premises of professional evolution. Some explanatory models have been elaborated in order to understand the motivations of these individuals, including the ABC model of kaleidoscope careers used in the present study. Despite the intense discussions abroad regarding such scenario, there are no scientific studies investigating if the opt-out movement occurs in Brazil and, if it occurs, what characteristics it has and what is the profile of professionals who adopt it - and these are the objectives of the current study. To examine these questions, a quantitative and descriptive research was performed involving 248 graduate and post-graduate alumni from the School of Economics, Management, and Accounting of the University of São Paulo (FEA USP), who attended the alumni meeting occurred in November 10th, 2007. The data was analyzed through descriptive statistics and inferential statistics (qui-square test, t-Student test, analysis of variance, correspondence analysis and factor analysis). The results permit to accept the four hypotheses proposed for this study, indicating that a significant amount of the respondents had done opt-out movements along their careers, and significant connections were found between the respondents` profiles and the adoption of opt-out movements; between the respondents` profiles and the reasons for making such decisions; and between the type of opt-out movement and the reasons for making the opt-out decisions.

Full textXylella fastidiosa is a gram-negative bacterium that colonizes the xylem and is the causative agent for several plant diseases. In Brazil, the main disease caused by this bacterium is the CVC (Citrus Variegated Chlorosis), which provokes large losses to the orange production in São Paulo and Minas Gerais states. Despite the current disease control, it has not been yet developed a specific method to eliminate the bacterium. During the plant-pathogen interactions, hosts produce an exacerbated amount of oxidants, in an attempt to eliminate the pathogen. Among them, fatty acids hydroperoxides are formed by the lipoxygenase action or even by the direct reaction between lipids and oxidant species. During the evolutionary process, pathogen defense mechanisms against oxidative species have evolved. Among them, Ohr (Organic Hydroperoxide Resistance protein) that is a Cys-based, lipoyl dependent peroxidase, displaying high activity towards organic hydroperoxides. This protein probably plays a central role in oxidative stress response and presnts some particularities, which make it a potential target for drug design. The objectives of this project were to characterize possible physiological substrates of Ohr from X. fastidiosa and search for molecules capable of inhibiting its peroxidase activity. Initially, we demonstrated that Ohr reduced fatty acid hydroperoxides with high efficiency (kcat/KM ~ 106 M-1.s-1). Moreover, these hydroperoxides inactivated Ohr in a dose-dependent manner, probably due to the high affinity between them and the enzyme. However, the enzyme did not display any activity towards phospholipids (posphatidilcholine) hydroperoxides and cholesterol hydroperoxide. Besides, we elucidated the structure of Ohr in the oxidized form (disulfide bond), which gave us insights on the dynamics of structural elements in the catalytic site. Ultimately, we identified a compound that was able to inhibit the peroxidase activity of Ohr in vitro, and we gained evidences that this compound can affect the bacterial growth under oxidative stress.

Full textPhotodynamic therapy (PDT) is a phototherapy based on the use of a photosensitizer (PS) in the presence of low intensity light with resonant wavelength of absorption of the PS and biological systems that can raise awareness, generating reactive oxygen species. Studies show that PDT has a lethal effect on Candida albicans. The biofilm formed by C. albicans is the cause of infections associated with medical devices such as catheters, with a proven resistance to antifungal agents, and the removal of the catheter colonized almost always is necessary. However, few studies in literature report the behavior and response of biofilm organized by C. albicans against PDT. The aims of this study were to develop a methodology for in vitro biofilm formation of C. albicans, evaluate the sensitivity of the biofilm of C. albicans to antimicrobial photodynamic therapy using PS as the methylene blue (MB) and hypocrellin B: La+3 (HBLa+3) and analyze the biofilm by Optical Coherence Tomography (OCT). For biofilm formation, discs were made from elastomeric silicone catheters. The PS were dissolved in solution of PBS, and the MB had two different concentrations tested in the biofilm: 100M and 1mM; HBLa+3 only one of 10M. The irradiation of both dyes with the microorganism was done by two different LEDs, one with red emission at = 630nm ± 20nm and the other one blue emission at = 460nm ± 30nm. We performed a curve of survival fraction versus time of irradiation of each sample with biofilm and suspension of the microorganism in the yeast form to verify the susceptibility of the front PDT. The yeast showed 100% reduction using both PS, but at different times of irradiation (30s to HBLa+3 and 6 min for the MB at 100M). When the therapy was applied in biofilm, the MB 100M did not show any significant reduction, while at concentration of 1mM was reduced by 100% after 6 min of irradiation. The HBLa+3 biofilm group showed a lower reduction in the concentration of 10M in biofilm (less than 1 log reduction). OCT was performed for visualization and measurement of the thickness of the biofilm formed. The composition of the extracellular matrix of the biofilm polymer hindered the diffusion of PS inside, requiring higher concentrations of MB to disseminate it and to obtain satisfactory reduction for PDT. HBLa+3 group, in higher concentration, formed aggregates difficulting its use for PDT. We conclude that the organism C. albicans was organized in biofilms in a standardized way using elastomeric discs from catheters and the OCT showed that the biofilm measured 110m at thickness, showing an optical change when subjected to the PDT with MB 1mM.

Full textThis work discusses the Optical Coherence Tomography system (OCT) and its application to the microfluidics area. To this end, physical characterization of microfluidic circuits were performed using 3D (three-dimensional) models constructed from OCT images of such circuits. The technique was thus evaluated as a potential tool to aid in the inspection of microchannels. Going further, this workpaper studies and develops analytical techniques for microfluidic flow, in particular techniques based on speckle pattern. In the first instance, existing methods were studied and improved, such as Speckle Variance - OCT, where a gain of 31% was obtained in processing time. Other methods, such as LASCA (Laser Speckle Contrast Analysis), based on speckle autocorrelation, are adapted to OCT images. Derived from LASCA, the developed analysis technique based on intensity autocorrelation motivated the development of a custom OCT system as well as an optimized acquisition software, with a sampling rate of 8 kHz. The proposed method was, then, able to distinguish different flow rates, and limits of detection were tested, proving its feasibility for implementation on Brownian motion analysis and flow rates below 10 μl/min.

Purpose: To determine if a single test is accurate in determining the parameters of the velocity: time-to-fatigue relationship, i.e., critical velocity (CV) and a finite distance that can be covered above CV (D`). Methods: Ten healthy subjects completed an incremental test to volitional exhaustion followed by four constant-velocity runs on a treadmill for the determination of CV and D`, as well as an all-out 3-minute test on a track for the determination of end-test velocity (EV) and the distance above end-test velocity (DEV). Eight of the eleven subjects completed a second 3-minute test and one run each at (+) and (-) 95% confidence interval velocities of CV determined from the 1/time model. Results: The group mean 1/time model CV (12.8 ± 2.5 km·h[superscript]-1) was significantly greater than the velocity-time model CV (12.3 ± 2.4 km·h[superscript]-1; P < 0.05), while the velocity-time model W` (285 ± 106 m) was greater than the 1/time model W` (220 ± 112 m; P < 0.05). EV (13.0 ± 2.7 km·h[superscript]-1) and DEV (151 ± 45 m) were not significantly different than the 1/time model CV and W`, respectively. EV was greater than the velocity-time model CV (P < 0.05), while the DEV was significantly less than the velocity-time model W` (P = 0.002). No difference was found for group mean EV or DEV between the two 3-minute tests (P > 0.05), which demonstrated a reliability coefficient of 0.85 for EV and 0.32 for DEV. For the CV (-) 95% run, all subjects reached a steady-state in VO[subscript]2, and completed 900 s of exercise. However, for the CV (+) 95% run, VO[subscript]2 never reached a steady-state, but increased until termination of exercise at 643 ± 213 s with a VO[subscript]2peak close to but significantly lower than VO[subscript]2max (P < 0.05). Conclusion: CV can be accurately determined using a single 3-minute test, while W` is underestimated with this protocol.

Full textThe aim of this study was to investigate, by mean of push-out test, the bond of Prime&Bond NT at various post-space dentin locations and the influence of one and six months water storage on bond strength values. Methods. Thirty single rooted teeth were used for the bond strength measurement. In each tooth fiber posts were cemented with Prime&Bond Nt in combination with Opticore Cure Core Composite. After twenty-four hours (group A: 10 roots), one month (group B: 10 roots) and six months (group C: 10 roots) water storage, the specimens were sectioned in 1mm-thick slices for the push out test. The data were divided into three regions (coronal/middle/apical) and analysed using Kruskal-Wallis Test and Mann-Whitney U (p<0,05). Results. The bond strength values registered didn’t show statistical significance difference within group A; within group B and C was found a statistical significant difference between the coronal and the middle thirds and between the coronal and the apical third. The apical bond strength values hours were higher at 24hour than six months; in the middle thirds a statistical significant difference was found at six months when compared to the twenty-four-hours and one-month samples. In the coronal thirds was found a statistical significant difference between one month and six months. Conclusions. Bond strenght along the root canal is higher in the coronal third. Over time the adhesion of the fiberpost/luting cement/post-space dentin doesn’t remain stable.

Full textThe pull-out and direct shear tests are the more used laboratory test for measure the resistance of soil-geosynthetic interface. In some occasions only the pull-out test evaluate well the behaviour of the inclusions amid the massive of soil. This work presents an pull-out test equipment, of great dimensions, and results of tests executed with him. The results were analysed according to a methodology suggested by Ochiai et al. (1996), slightly modified by the author. From the pull-out tests data, it may be concluded that the equipment is rendered to the purpose that it is destined, besides, being analysed the results according to the mentioned method, it was verified that the pull-out resistance of geogrids concentrates in initial portion, when it is applied small loads. When the applied force increases, the resistance is mobilized in the posterior portions and occur to the generalised pull-out rupture.

Full textThe feasibility of locking plates and their biomechanical advantages in comminuted fractures are well known. The objective of this study was to evaluate different locking plate´s configurations submitted to a four point´s flexion test and pull out test. The flexion test evaluated the influence of screws in center position in bridging plate, the pull out test compared relative to pull out strength locking plates using parallel locking screws or angulated locking screws. For each biomechanical assay two groups of ten test materials were formed. The results found no statistical differences relative to flexion resistance in the groups with and without central screws. Relative to pull out test the group with angulated screws showed higher values of maximum pull out strength when compared with the perpendicular screws, the data were statistically significant. We concluded that there was no improvement in the resistance of bridging plate with the use of central screws, but the use of angulated screws showed a greater resistance to pull out when compared to parallel screws.

Full textThis work focuses on the design of on-silicon mm-wave Reflection-Type Phase Shifters (RTPS) with Oscillation-Based Test (OBT) capabilities. For more consistency, a single technology was considered, the STM 55-nm BiCMOS. First, the theory and practical implementations of 3-dB couplers is discussed. Particular attention is brought to the Coupled Slow-wave CoPlanar Waveguide (CS-CPW) topology, due to its good performance. Using this topology, the measurements of two 3-dB couplers are reported: (i) a 120-GHz, and (ii) a 185-GHz coupler.Next, the existing topologies of integrated varactors are discussed. Measurement results are reported for an Inversion-mode MOS (I-MOS) varactor from 1 up to 325 GHz. Additionally, the Common-Source MOS (CS-MOS) varactor architecture is proposed and measurement results from 1 to 145 GHz for this architecture are reported.Then, the theory of RTPS is presented and CS-CPW-based couplers together with Accumulation-mode MOS (A-MOS), I-MOS and CS-MOS varactors are used for the design of four RTPS. The measurement and simulation results of these RTPS, with central frequencies ranging from 60 to 200 GHz, are presented.Subsequently, the theory and measurement results of the OBT on an integrated 60-GHz RTPS are discussed.Finally, a mm-wave TRL calibration compaction technique is described using machine-learning tools

Full textRunning is one of the activities that had the highest number of participants among elderly population. It is known that during running, elderly have different patterns of young adults, as a greater toe-out angle. However it is still unknown the relationship between the mechanical loads at the joints of the lower limbs. Several studies point to a correlation between the external knee adduction moment and the toe-out angle, including as a protective mechanism among people with knee osteoarthritis, a common disease in people over 65 years old. Thus, we sought to investigate a possible relationship between the toe-out angle increased in these individuals and the knee moment in the frontal plane. Twenty-two young adult runners, 23 elderly runners and 13 sedentary elderly walked at 5 km/h and ran at 7, 10 km/h and at the mean speed of the last 10 K race. At the speed of 5 km/h and 10 km/h the individuals who were runners also performed a condition with the toe-out angle increased beyond the natural. The subjects did all tasks in an instrumented treadmill with independent two belts and two force plates with six components operating at 600 Hz (Bertec, USA) and had the lower limb movements registered by a tridimentional motion analysis system with eight infrared cameras operating at 150Hz (Motion Analysis Corporation, USA). A complete kinematics and kinectics analysis were made using Visual3D (CMotion. Inc., USA) and Matlab (Mathworks, USA) softwares. The toe-out angle and the external knee adduction moment were measured during all conditions and compared between the condition with and without the toe-out angle increased, and between the groups with a mixed effects modeling and ANOVA respectivelly. The joint angles and joint moments were compared between groups with Wilcoxon Signed-rank tests with Bonferroni correction. In general, young adult runners, elderly runners and elderly sedentaries had differences in kinematic and kinetic pattern of movement of lower limbs during walk at 5 km/h and run at 7 km/h. There were also differences, though in smaller number, during run at 10 km/h and run at mean speed of the last 10 K race between young adult runners and elderly runners. Contrasting the literature there was no relation between toe-out angle and external knee adduction moment and we didnt observe a greater angle in elderly than young adults during walk or run, it maybe can be explained by the fact that the tasks were above a double belt treadmill, what can lead to a different feet position (toe-out)

Full textThis study approached theoretical and experimental aspects related with the development of a polarization sensitive, Fourier domain, optical coherence tomography system (PS-FD-OCT) and its utilization on the Mueller Matrix determination. This work began with a bibliographic revision, which describes since the early studies to the actual state of the art of the technique. The mathematical formalism of Fourier domain low coherence interferometry and light polarization was performed as well. Studies based on numerical simulations, of three different algorithm types, responsible to recover the scattering profile, were done. The implemented algorithms were: Direct Fourier Transform, Interpolation and Zero-Filling. By the end of the simulation study, was possible to conclude that the algorithm Zero-Filling 2N presented better characteristics when compared with the others. In the experimental part, firstly different OCT setups were assembled and measurements were done in order to verify aspects related with the theory. Then, using a polymeric sample, birefringence images were performed, which allowed determining the sample birefringence quantitatively. Finally, images taken of different polarization states were collected, and through then images related with the Mueller Matrix elements were calculated, which were analyzed individually.

Neste estudo, avaliou-se a resistência da união ao push-out de dois cimentos resinosos auto-adesivos e de um cimento resinoso convencional em dentina radicular. Em 60 dentes terceiros molares humanos, tratados endodonticamente, um pino de fibra de vidro foi cimentado com um dos três cimentos resinosos. Cada raiz foi seccionada em sentido transversal para a obtenção de três fatias (dentina-pino) de 1 mm de espessura. As fatias, correspondentes aos terços cervical, médio e apical do canal radicular, foram armazenadas em água deionizada a 37 ºC, durante 24 horas ou 30 dias. Após o teste de push-out, os valores de resistência da união foram submetidos aos testes estatísticos ANOVA three way e Tuckey (p = 0,05) e os tipos de fratura foram avaliados através de microscopia eletrônica de varredura. A resistência da união do cimento resinoso auto-adesivo RelyX U100 foi superior à do cimento convencional RelyX ARC, após os dois períodos de armazenamento (p < 0,05). No terço cervical, os valores de resistência da união do cimento resinoso convencional RelyX ARC foram maiores que nos terços médio e apical (p < 0,05) na análise de 24 horas. Após 30 dias de armazenamento, os valores nos três terços radiculares não diferiram entre si. Os valores de resistência da união do cimento resinoso RelyX U100, nas três regiões do canal radicular, não diferiram entre si (p > 0,05), em ambos os momentos de avaliação. Após 24 horas, BisCem apresentou maior resistência da união no terço apical, seguido pelo terço cervical. Após 30 dias, os valores de resistência da união nos três terços radiculares não diferiram entre si (p > 0,05). RelyX U100 não sofreu alteração nos valores de resistência da união após 30 dias de envelhecimento (p > 0,05). BisCem não apresentou diferenças nos valores obtidos após 24 horas e 30 dias de envelhecimento nos terços cervical e médio (p > 0,05). Para RelyX ARC, o tempo de 30 dias reduziu os valores de resistência da união nos três terços radiculares. RelyX U100 e BisCem apresentaram maior número de falhas coesivas da zona de interação. Para RelyX ARC, observou-se maior número de falhas coesivas da camada híbrida. Concluiu-se que o cimento resinoso auto-adesivo RelyX U100 apresentou maior resistência da união que o cimento resinoso convencional RelyX ARC, nos três terços radiculares, nos dois momentos de avaliação. BisCem demonstrou resistência da união comparável a do cimento RelyX U100. O tempo de armazenamento por 30 dias provocou alteração na resistência da união dos cimentos resinosos RelyX ARC e BisCem.
This study evaluated the push-out bond strength of two self-adhesive resin cements and a conventional etch-and-rinse resin cement on root dentin. Sixty human third molars were endodontically treated and a glass fiber post was cemented with one of three resin cements. Each root was cut transversely to obtain three slices (dentin-post) of 1 mm thick. The slices of the cervical, middle and apical root canal were stored in deionized water at 37 °C for 24 hours or 30 days. After storage, the push-out test was performed. The values of bond strength were evaluated by ANOVA three way and Tuckey’ test (p = 0,05) and the types of fracture were evaluated by scanning electron microscopy. The bond strength of the self-adhesive resin cement RelyX U100 was superior to the RelyX ARC etch-and-rinse cement, after the two storage periods (p > 0.05). The analysis of 24 hours showed that at the cervical third, the values of bond strength of RelyX ARC were higher than that of the middle and apical thirds (p < 0.05). After 30 days of storage, the values at the three root thirds did not differ. The values of RelyX U100, on the three regions of the root canal did not differ, in both evaluations (p > 0.05). After 24 hours, BisCem showed higher bond strength at the apical third, followed by cervical. After 30 days, the values of bond strength at the three root thirds did not differ (p > 0.05). RelyX U100 did not exhibit changes in bond strength values after 30 days of storage (p > 0.05). BisCem showed no differences in the values obtained after 24 hours and 30 days of aging at the cervical and middle thirds (p > 0.05). The storage time of 30 days reduced the values of bond strength at the three root thirds of RelyX ARC. RelyX U100 e BisCem had the highest number of cohesive failures of the zone of interaction. RelyX ARC presented a higher number of cohesive failures of the hybrid layer were observed. It was concluded that the RelyX U100 self-adhesive resin cement showed higher bond strength than the conventional resin cement RelyX ARC, on the three root thirds, in both periods. BisCem showed bond strength comparable to RelyX U100. The storage time for 30 days caused changes on the bond strength of the RelyX ARC and BisCem resin cements.

Full textThe possibility to use titanium as alternative reinforcement for concrete is investigated in this thesis by focusing on the interfacial behaviour that the two materials develop when they are combined in a composite structural material. To this end, several experimental programs were conducted. The first one concerns the pull-out behaviour of plain bars made of the titanium alloy Ti6Al4V from two different concrete mixtures, i.e. Normal and Light Weigh Concrete (NWC and LWC, respectively). The second experimental program investigates the pull-out behaviour of Ti6Al4V straight and hooked-end fibres from LWC specimens. The third and last series of experiments regards the fracture toughness characterisation of a fibre reinforced concrete material made of LWC and titanium fibres (TiFRC). A series of specimens and a full-scale beam made of TiFRC, without any other reinforcement, have been built and tested under three-point load conditions in order to measure the size effect on the flexural strength. The experimental results concerning the pull-out tests are supported by Finite Element (FE) analyses. Particularly, the results of the bar pull-out tests, combined with Scanning Electron Microscope (SEM) analyses, show that, although the employed rebars are plain, the debonding process is strongly affected by defects-induced surface roughness present at the microscopic level, which activates mechanical interlocking responsible for the dilatant behaviour of the interface. A novel cohesive zone model micromechanics-based formulation is implemented in the FE model in order to account for such aspects. The introduced enhanced degrading M-CZM, accounting for damage, friction, mechanical interlocking and dilatancy, is used to carry out sensitivity analyses and identification procedures on the FE models simulating the bar pull-out tests. The proposed modelling strategy is further validated by performing FE simulation of straight fibres pull-out tests.

Full textThe aim of the study was to determine which of the three resin cements would produce the highest debond stress values with a double-tapered fibre post system. In the past, conventional parallel- sided prefabricated or cast metal posts have been used, which has considerable mechanical strength but lacks aesthetic capabilities. Post preparation techniques usually compromise the fracture strength of the treated anterior tooth. Double- tapered post systems ensure that anterior mutilated teeth can be restored and retained without compromising aesthetics or excessively weakening the remaining dental tissues. There are in-vitro results that support the strength of the double- tapered design, as well as the aesthetics of the material but there is little evidence regarding the retentive capabilities of the posts and which cement will ensure the best results (Grandini et al., 2008; Nakamura et al., 2005). Literature suggests that the two main causes for failure of a post system are root fracture and debonding of the post in the root canal (Toman et al., 2009; Radovic et al., 2008). The type of cement and cementation technique will have a significant influence on the treatment success of post and core restorations. This study compared the debond stresses required to remove a double- tapered post system from a prepared post space with 3 different cements to assess which cement will be most resistant to the post debonding.

Full textObjective: The aims of this study is to verify the influence of adhesive, filling material and dentin hybridization on fiber post retention force in diretc -indirect technique and compare the tensile test of push out with pull out. Methods: 60 bovine single roots with a length of 12 mm were endodontically treated and randomly divided into 6 groups (n = 10): group 1 (direct-indirect technique, light-curing adhesive, resin cement, pull out test), group 2 (same as group 1 but the resin cement was replaced by composite resin), group 3 (same as previous group but the light-curing adhesive was replaced by the chemical), group 4 (same as group 3 but the test was used was push out test), group 5 (same as group 4 but the adhesive used was the self etching) and Group 6 (same as previous group but the dentin was pre-hybridized). The retention force was determined by Universal Testing Machine (Instron). All data were analyzed using one-way analysis of variance (ANOVA) with significance of p <0.05 and complemented with Tukey test. After testing, the adhesive interfaces were examined and classifie. Result: There was a significant difference between comparison of materials (adhesive and filling material). The hybridization of dentin showed no improvement in the retention post. Most fractures occurred at the interface dentin / resin. Conclusion: Based on the results, we conclude that filling material, adhesive, influenced the retention of fiber posts, and the prehybridization of dentin did not improve the retention of fiber post. The push out test showed more details to verify the bonding between the post and dentin, and the pull out test for the retention force of the post inside the canal.

Full textThe stability of the ligand diethyl phosphite was analyzed by 1H NMR in solution at pH 1.0 and 3.0 and 25°C in the presence and in the absence of the ion [RuII(H2O)(NH3)5]2+. The hydrolysis of the free diethyl phosphite in solution at pH 3.0 was not observed, while in the presence of the ion [RuII(H2O)(NH3)5]2+, at this same pH, the reaction took place with kobs=1.0 10-4 s-1. In solution at pH 1.0 the hydrolysis reaction took place in the presence (kobs=6.2 10-4 s-1) and in the absence (kobs=1.8 10-4 s-1) of the metal center. The complex trans-[RuII(NO+)(NH3)4P(OH)3](Cl)3 was synthesized and characterized by spectroscopic and spectrophotometric techniques. Vibrational spectra showed a νNO+ in the solid state at two differents frequencies (1903 and 1867 cm-1) due to the presence of the ligand P(OH)3 in the protonated and deprotonated forms. The ENO+/NO0 was determined by Cyclic Voltammetry at -0.52 V vs. SCE (0.5 mol L-1 CF3COOH, 25°C). The electronic spectrum of this complex exhibited three bands at 241nm (ε=1385 L mol-1 cm-1), 319 nm (ε=773 L mol-1 cm-1) and 500nm (ε=20 mol L-1 cm-1). The pKa value for the phosphorous acid in the complex íon trans-[RuII(NO+)(NH3)4P(OH)3]3+ was determined by Vibrational Spectroscopy in solution (pKa=0.74 ± 0.05). As judged from UV-vis, Cyclic Voltammetry and Vibrational Spectroscopy the ion trans-[RuII(NO+)(NH3)4P(OH)3]3+ suffer aquation reaction yelding free phosphorous acid and the ion complex trans-[RuII(NO+)(NH3)4(H20)]3+ (λ=322 nm - ε=297 L mol-1 cm-1; ENO+/NO0=-0.40 vs. SCE; νNO+= 1893 cm-1). 31P NMR and Vibrational Spectroscopy data suggest that, before the dissociation of the P(OH)3 ligand, occurs the formation of linkage isomers in which the phosphorous acid can be coordinated to the metal center also by the oxygen atom. The data obtained by kinetics experiments suggest that the P(OH)3 dissociation is dependent of the hydrogen ionic concentration of the medium.

Full textThe international literature while investigating the long term effects of early childhood education identified a decrease phenomenon of such effects, called fading-out. Using longitudinal data collected in the city of Sertãozinho our goal was to investigate and document this phenomenon for Brazilians\' reality. We found evidence of the decrease of early childhood education benefits, especially in Math proficiency. Furthermore, we show that the structure of the database used to make such measurements can generate results that overestimate the phenomenon, especially if those databases contain a high degree of selectivity. Finally, we indicate that the quality of the early childhood schoolscan be an important determinant of the effects\' durability, since children who attended most of the early childhood education in public schools are more prone to this phenomenon.

Full textThe amniotic membrane is the inner layer of the fetal membranes (placenta), widely used in transplantation as it is a tissue that combines properties anti-inflammatory and antifibrotic antimicrobial, and limited ability to induce immune reactions. The use of fresh membrane has some limitations such as the need for rapid deployment and not being completely safe against of certain infections. Any biological tissue used for transplantation should be sterile. The radiosterilization is an alternative to ensure the quality and safety of tissues used in transplants, and other clinical applications to minimize the risk of contamination of the tissue´s receptor, but it can cause undesirable changes in the tissue. In this study, we tested doses of 10, 15, 25 and 35 kGy, using two sources of ionizing radiation: gamma rays from cobalt-60 source and electron beam. In qualitative, visual and tactile analysis it has been observed that higher doses (25 kGy and up, to both sources of radiation) irradiated membranes had a greater color change, becoming yellowed and decreased elasticity, becoming more rigid. The solid colorimetry minimized the subjectivity of visual analysis and optical microscopy was essential to evaluate the histological changes showing, respectively, the color change of the membrane and the degree of degradation of the underlying layers of tissue is directly related to the dose of radiation employed. Thus, doses of 10-35 kGy can be applied in the amniotic membranes for application as a biological bandage, however doses from 25 kGy and up should take into account the changes in color and condensation of the layers of the membrane for ophthalmic use or as a carrier substrate for transplantation of cultured tissue in vitro. With the techniques of OCT, TG and tensile testing was not possible to evaluate the biomechanical findings in the qualitative analysis, the experimental conditions due to the standard-deviations obtained for the five membranes tested.

Full textObjectives: Evaluate retinal changes in experimental model of hypoxia in rabbits acclimatized in normobaric-hypoxic environment and to investigate the effects of the treatment by intravitreal (IV) bevacizumab drug. Methods: Twenty two New Zealand rabbits weighing between 2,4 to 3,8 kg were divided into 4 groups. The groups S12 (n=5) and B12 (n=5) were acclimatized for 3 consecutive days in oxygen concentration (O2) to 12%. The groups S8 (n=5) and B8 (n=7) were acclimatized for 3 consecutive days with gradual reductions in O2 concentration until the nadir of 8%. The right eye (RE) were kept as controls and the left eye (LE) of the animals belonging to S12 and S8 groups received IV injection of 0,05 ml of balanced salt solution (BSS), while the LE belonging to groups B12 and B8 received 0,05 ml (1,25 mg) of bevacizumab IV. Optical coherence tomography (OCT) to evaluate the thickness of the retinal segments (RS) and choroidalsegments (CS), sodium fluorescein angiography (FA) for evaluation of the presence or absence of vasodilation and tortuosity of the retinal vasculature and quantification of VEGF in the aqueous fluid and peripheral blood sample were conducted at the first day (D0) before treatment, on the third day of hypoxia (D7) and day 11 (D11) using the Luminex® technique. After the animals were sacrificed, the retinal tissue samples were evaluated by histology and immunohistochemistry (IHC). Results: Comparing the horizontal sections of the RE (control) in D0 and D7 periods, a reduction of 8% (p<0,0001) and 10% (p<0,0001) the thickness of the RS in O2 concentration at 12% and 8%, respectively. Comparing the vertical cuts in the same period, there was reduced RS thickness of 7% in both concentrations to 12% (p<0,0001) and 8% (p<0,0001). In the eyes treated with bevacizumab, to reduce the average thickness of the retinal segment for horizontal cuts between D0 and D7 periods were 6% (<0,0001) and 9% (<0,0001) for O2 concentrations to 12 and 8%, respectively. While in the eyes treated with BSS in the same period, there was an 8% reduction (<0,0001) and 6% (<0,0001) for the O2 concentration at the 12% and 8%, respectively, in the horizontal cuts. In the vertical sections is observed for the eyes treated with bevacizumab, 5% reduction (p=0.0005) and 8% (<0,0001) O2 concentration at 12% and 8%, respectively; and BSS treated eyes was reduced by 7% (<0,0001) and 8% (<0,0001) in the O2 concentrations of 12% and 8%, respectively. The thickness of the CS did not show changes. The B8 group showed statistical difference in the analysis of the eyes that did not have vasodilation and tortuosity of the retinal vessels during the hypoxic period. Retinal neovascularization were not observed. Histology and IHC of the eyes treated with BSS and bevacizumab showed no changes compared to the control eyes. Conclusions: The acclimatization of the rabbits in normobaric-hypoxic environment has the effect of reducing the thickness RS on the third day of hypoxia. It is observed also increased tortuosity and vasodilation. The intravitreal bevacizumab does not inhibit retinal thickness decrease, but inhibits vasodilation and vascular tortuosity.

Full textThe present study aimed to evaluate, in vitro, the influence of different treatment protocols of weakened root dentin on the bond strength (BS) of glass fiber reinforced post luted with different resin cements. Forty two upper human canines were sectioned and the root length was standardized on 17-mm after coronary portion was removed. Biomechanical preparation was performed via Reciproc system with R 50.05 instrument and 2.5% NaOCl. The roots were flared using diamond bur #4137 and #720G at a depth of 12mm and filled by the lateral condensation technique with AH Plus sealer modified with 0.1% Rhodamine B. After 3 times the time required to the setting reaction of the sealer the filling was removed and the glass fiber post was relined with composite resin. The roots were divided into 3 groups (n = 14) according to the treatment of dentin surface: 2.5%NaOCl; 2.5%NaOCl + 17% EDTAand 2.5% NaOCl+ 17% EDTA+ ultrasound. Half of the samples of each group had the post luted with RelyX U200 and the other half with Panavia F. Both cements were added with 0.1% fluorescein. The roots were transversely sectioned obtaining two slices of each third, one for the push-out test and the other for the confocal analysis and measurement of knoop hardness. Data were submitted to ANOVA test with sub-divided portion followed by Tukey test (α= 0.05). The highest BS values were observed for 2.5% NaOCl + 17% EDTA and 2.5%NaOCl + 17% EDTA + ultrasound (p> 0.01) compared to 2.5%NaOClalone (p <0.0001). Panavia F showed higher BS than RelyX U200 in specimens treated with 2.5%NaOCl + 17% EDTA and 2.5% NaOCl + 17% EDTA + ultrasound (p <0.0001) in the three root thirds. There was no difference between the luting materials in specimens treated only with 2.5% NaOCl(p> 0.01). The protocols did not affect the BS of RelyX U200. Overall the higher incidence of failure was adhesive (38.83%) followed by mixed (34.72%) and cohesive (26.45%) failure. Dentin treatment with 2.5%NaOCl+ 17% EDTA + ultrasound promoted the greatest reduction in microhardness. It was concluded that the Panavia F presented the highest BS in the three root thirds and the protocols did not influence the BS of the specimens filled with RelyX U200

Full textThis descriptive exploratory and correlative study adopted the quantitative method and the aim was to characterize and analyse the burn out among oncology nurses regarding the relations with personal and occupational variables. The sample was represented by 77 nurses working in oncology clinical and surgical units in five hospitals in the city of São Paulo. Three tools were used for the data collect: a demographic and occupational inventory data made by the author, the Maslach and Jackson Burn out Inventory and the Folkman and Lazarus Coping Inventory. The results showed that 53,3% of the nurses demonstrated some kind of symptom of burn out, being 29,9% with alteration in one dimension, 14,3% with alteration in two dimensions and 9,1% with alteration in three dimensions. It wasnt verified in this sample any important statistic difference between the demographic and occupational variables and burn out. The main stressors identified by the nurses were the death and dying, the conflict with doctors and the nursing team and the emergency situations. There was a statistic difference between personal accomplishment dimension and the stressor death and dying and the copings social support and escape. Also between the emotional exhaustion dimension and the stressor conflict with doctors and nursing team and the coping escape. The nurses used more frequently problem solving and positive reappraisal as coping styles, and the less used was the act of escaping

Full textThe aim of this study was to evaluate the bond strength of epoxy resin- and methacrylate-based sealers to root dentin, using the material testing stage (MTS) coupled to a micro-computed tomography (μCT) device and to qualitatively assess the root-filling mass rupture by μCT images. Additional analyses were conducted using the 3D element finite method and confocal laser scanning microscopy (CLSM) to verify, respectively, the stress distribution and percentage of gaps on dentin filling material interfaces. Maxillary canine roots measuring 17 mm were subjected to biomechanical preparation with nickel-titanium rotary instruments under constant irrigation with 1% NaOCl and final irrigation with 17% EDTA. Canals were filled by the lateral condensation technique with gutta-percha cones and the following sealers: AH Plus, Epiphany SE or RealSeal XT. The roots were stored (100% humidity, 37 °C) for a period three times longer than the setting time required for each sealer and then sectioned transversally to obtain two 2-mm-thick slices from each root third. One slice of each third was subjected to the push-out test using the MTS coupled to the SkyScan 1174 μCT (50 kV, 80 mA) with 13 μm isotropic resolution, 0.5 mm Al filter and 360° rotation. After the initial scanning of the slice, the mechanical test was performed until material dislodgement occurred. A new scanning was performed using the same initial parameters. The axial sections obtained were used for qualitative evaluation of the root-filling mass rupture. Failure modes were assessed by CLSM and classified as adhesive, cohesive or mixed. 3D μCT based finite element models were generated from cervical, middle and apical root thirds to simulate the push-out test. The second slice of each root third was used to assess the percentage of gaps between the filling material and the root canal walls, using CLSM with the aid of the OLS 4000 software. The data were analyzed by parametric and non-parametric tests (p<0.05). AH Plus presented higher bond strength [1.89 (1.01/2.39) MPa] to dentin (p<0.001) than Epiphany SE [0.36 (0.29/0.44) MPa] and RealSeal XT [0.43 (0.32/0.54) MPa]. For AH Plus and RealSeal XT sealers, the cervical and middle thirds showed higher bond strength to dentin than the apical third (p <0.001). The ruptures occurred in the margins of the root filling mass for AH Plus, while Epiphany SE and RealSeal XT had this type of rupture as well as in the root filling mass itself. AH Plus showed stress concentration on bond interface, while in Epiphany SE and RealSeal XT the stress was mainly distributed on the root-filling mass. Adhesive and mixed failures were the predominant fracture modes in all sealers. Regarding the percentage of gaps between the filling material and the root canal walls, AH Plus presented the lowest means (11.14%±4.03), differing significantly (p<0.05) from Epiphany SE (55.39%±13.46) and RealSeal XT (62.50%±6.72) sealers. The epoxy resin-based sealer (AH Plus) presented the highest bond strength to dentin, which had different behavior from other sealers regarding root-filling mass rupture and interfacial stress distribution. Full textResumo: A indústria do cimento Portland é responsável direta por uma carga de poluentes de significativo dano ambiental. Os Concretos Ativados Alcalinamente (CAA) são matrizes compostas por um ativador alcalino e um aglomerante alternativo. O material comumente utilizado para a ativação alcalina é o silicato de sódio, cuja fabricação também se mostra como ambientalmente nociva. O silicato de sódio, junto ao cimento Portland, podem ser dispensados, uma vez que a produção do ativador pode se dar através de um composto rico em silício (materiais pozolânicos no geral), sendo a cinza da casca de arroz (CCA) o instrumento de estudo do trabalho apresentado, e a soda cáustica como fornecedora de sódio (meio alcalino). Os concretos CAA foram definidos conforme o parâmetro ξ (CAA-ξ), que representa a relação molar entre SiO2 e Na2O, com as variações ξ = 1,2, ξ = 1,6 e ξ = 2,0, esta última representando o concreto ativado alcalinamente com maior taxa de CCA. Este trabalho tem como intuito a avaliação das propriedades mecânicas dos concretos CAA, comparando-as, em seguida, com duas tipologias de concreto com cimento Portland CPV-ARI, com distintos fatores água cimento (0,45 e 0,55). A variação na relação a/c teve como intuito a análise de duas referências com valores diferentes de fck. Os resultados demonstram que a resistência à compressão axial e diametral (sete dias de cura) para os concretos CAA se encontraram na faixa de 25 a 30 MPa, e de 1,5 a 3,5 MPA, respectivamente. Abstract: The Portland cement industry is directly responsible for a load of pollutants of significant environmental damage. Alkali-Activated Concrete (AAC) is a matrix with prior activation and alternative binder. The catalyst material commonly used for prior activation is the sodium silicate, the manufacture of which is also environmentally harmful. Sodium silicate, together with Portland cement, can also be dispensed with, since the activator can be produced through a silicon-rich compound (pozzolanic materials in general), with the rice husk ash (RHA) being the instrument of study of the work presented, and caustic soda as a supplier of sodium. The AAC concretes were defined according to the parameter ξ (AAC- ξ), which represents the molar ratio of SiO2 and Na2O, with the variations ξ = 1.2, ξ = 1.6 and ξ = 2.0, the latter representing the AAC with higher RHA rate. This work intends to evaluate the mechanical properties of AAC, comparing them to two types of concrete with ordinary Portland cement with high early resistance, with different water cement factors (0.44 and 0.55). The variation in the water/binder mass ratio was intended to analyze two references with different values of compressive strength class. The results demonstrate that the axial and diametric compression strength (seven days cure) for the AAC concretes were in the range of 25 to 30 MPa, and of 1.5 to 3.5 MPA, respectively. Full textXylella fastidiosa is a gram-negative bacterium, which colonizes the xylem from economically important plants, being responsible for several diseases such as Pierce disease (PD) in gravepines and citrus variegated clorosis (CVC). Plants, when infected by pathogens, are able to defend themselves through several mechanisms which include the generation of reactive oxygen species (ROS). Lipid hydroperoxides can be generated from the attack of ROS to the bacterial membrane or by the action of lipoxygenases. The alkyl hydroperoxide reductase system (AhpR) was initially characterized as the main responsible for the detoxification of organic hydroperoxides in bacteria. Recently, it was also characterized another gene in many pathogenic bacteria, whose deletion renders cells susceptibility to organic hydroperoxide treatments but not by H2O2 or by superoxide generators (Mongkolsuk et al., 1998 and Ochsner et al., 2001). For this reason, it was named ohr (organic hydroperoxide resistance gene). The goal of this work was to functionally characterize ohr, the product of ohr gene from Xylella fastidiosa. Initially, we demonstrated that ohr possesses Cys-based thiol-dependent peroxidase activity. Later, we showed that ohr possesses a unique alpha/beta fold not observed in the structures of other thiol peroxidases such as peroxiredoxins and glutathione peroxidases. Analyses of ohr active site showed that its likely substrates are elongated and hydrophobic molecules. Furthermore, we showed that lipoylated enzymes, classically related with the intermediary metabolism, interacts physically and functionally with ohr while classical thiol-dependent pathways, such as thioredoxin and glutathione, failed to support ohr activity. This finding represents the first evidence of a peroxidase that is directly reduced by lipoyl groups of enzymes. Also, we obtained evidences indicating that ohr acts in the detoxification of peroxides derived from unsaturated fatty acids. In fact, steady-state kinetics using bi-substrate analysis showed that ohr decomposes organic peroxides with high efficiency (kcat/KM ~ 106 M-1.s-1 through a ping-pong mechanism, at least ten thousand times more efficiently than hydrogen peroxide (H2O2). All these results together shows that ohr is central in the response of bacteria to the stress induced by organic hydroperoxides but not by H2O2 and defines a new class of antioxidant enzymes with unique properties such as lipoyl-dependent peroxidase activity. Another goal of this work was to study the regulation of ohr expression in Xylella fastidiosa. ohr expression is regulated in most bacteria by a repressor protein named ohrR (Sukchawalit et al., 2001) but, in some bacteria, ohr expression is positively regulated by an alternative sigma factor (σE) with extracitoplasmatic function (Gourion et al., 2008). Our results showed that ohr from X. fastidiosa was not under the control of none of these regulators, probably being constitutively expressed. Through northern blot analysis, we did not observed any changes in ohr levels in cells submitted to oxidative or ethanolic stress. These results, indicates that ohr expression probably differs from that previously described on literature for other bacteria.
