Abstract:
Thin-ply laminates exhibit a higher degree of freedom in design and altered failure behaviour, and therefore, an increased strength for unnotched laminates in comparison to thick-ply laminates. For notched laminates, the static strength is strongly decreased; this is caused by a lack of stress relaxation through damage, which leads to a higher stress concentration and premature, brittle failure. To overcome this behaviour and to use the advantage of thin-ply laminates in areas with high stress concentrations, we have investigated thin-ply hybrid laminates with different metal volume fractions. Open hole tensile (OHT) and open hole compression (OHC) tests were performed with quasi-isotropic carbon fibre reinforced plastic (CFRP) specimens. In the area of stress concentration, 90° layers were locally substituted by stainless steel layers of differing volume fractions, from 12.5% to 25%. The strain field on the specimen surface was evaluated in-situ using a digital image correlation (DIC) system. The embedding of stainless steel foils in thin-ply samples increases the OHT strength up to 60.44% compared to unmodified thin-ply laminates. The density specific OHT strength is increased by 33%. Thick-ply specimens achieve an OHC strength increase up to 45.7%, which corresponds to an increase in density specific strength of 32.4%.

PurposeOCT-angiography (OCT-A) offers a non-invasive method to visualize retinochoroidal microvasculature. As glaucoma disease affects retinal ganglion cells in the macula, macular microcirculation is of interest. The purpose of the study was to investigate regional macular vascular characteristics in patients with ocular hypertension (OHT), pre-perimetric primary open-angle glaucoma (pre-POAG) and controls by OCT-A in three microvascular layers.Material and methods180 subjects were recruited from the Erlangen Glaucoma Registry, the Department of Ophthalmology, University of Erlangen and residents: 38 OHT, 20 pre-POAG, 122 controls. All subjects received an ophthalmological examination including measurements of retinal nerve fibre layer (RNFL), retinal ganglion cell layer (RGC), inner nuclear layer (INL), and Bruch’s Membrane Opening-Minimum Rim Width (BMO-MRW). Macular vascular characteristics (vessel density, VD, foveal avascular zone, FAZ) were measured by OCT-A (Spectralis OCT II) in superficial vascular plexus (SVP), intermediate capillary plexus (ICP), and deep capillary plexus (DCP).ResultsWith age correction of VD data, type 3 tests on fixed effects showed a significant interaction between diagnosis and sectorial VD in SVP (p = 0.0004), ICP (p = 0.0073), and DCP (p = 0.0003). Moreover, a significance in sectorial VD was observed within each layer (p<0.0001) and for the covariate age (p<0.0001). FAZ differed significantly between patients’ groups only in ICP (p = 0.03), not in SVP and DCP. For VD the AUC values of SVP, ICP, and DCP were highest among diagnostic modalities (AUC: 0.88, 95%-CI: 0.75–1.0, p<0.001).ConclusionRegional reduced macula VD was observed in all three retinal vascular layers of eyes with OHT and pre-POAG compared to controls, indicating localized microvascular changes as early marker in glaucoma pathogenesis.

Abstract:
Orthotopic heart transplantation (OHT) represents an effective alternative for individuals with end-stage heart disease. The current literature reports only the responses of OHT patients to greater than or equal to 4 mo of exercise training (ET) and frequently lacks adequate controls. Most programs currently treating OHT patients usually provide 6-12 wk of ET. This study describes the effects of a 10-wk supervised ET program in 12 male OHT patients and 5 other male OHT patients who served as a comparison group. Graded exercise tests were performed before and after ET. After ET, maximal O2 consumption was significantly greater for the ET group than the comparison group (P less than 0.05) and the mean increase in peak heart rate was 18 +/- 4 and 6 +/- 4 (SE) min-1 for ET and comparison groups, respectively (P less than 0.05). Maximal ventilation was also significantly greater for the ET group at after ET, while resting heart rate and blood pressure and peak blood pressure, O2 pulse, respiratory rate, and ventilatory equivalents for O2 and CO2 were not significantly changed. We conclude that after OHT a 10-wk ET program improves maximal O2 consumption and, by improving peak heart rate, improves O2 delivery.

Abstract:
Purpose. To evaluate the changes of flash electroretinography (fERG) and pattern visual evoked potentials (pVEP) in ocular hypertension (OHT) patients.Methods. Twenty-five OHT patients and 30 healthy volunteers were enrolled for this cross-sectional study. Opthalmologic examinations, visual field tests, pVEP and fERG were performed. The main outcome measures were the differences between pVEP and fERG parameters.Results. The mean age of OHT patients and volunteers were 57 ± 12.25 years (range 30–65 years), and 53.25 ± 12.0 years (range 30–65 years), respectively. The mean amplitude of the pVEP was statistically lower in the OHT group (P< 0.05). Latency of the two groups was different; however, the difference was not statistically significant (P> 0.05). In fERG of OHT group, there was a significant decrease in the amplitude of the oscillatory potentials (Ops), and a significant delay in latency of rod and cone waves (allP< 0.05). There was no significant change in the flicker fERG waves between the two groups (P> 0.05).Conclusions. Although we found a decrease in Ops amplitude and a prolonged latency in flicker fERG, only the decrease in Ops amplitude was statistically significant between the two groups. The amplitude of Ops wave and amplitude of pVEP may reflect early glaucomatous damage in OHT patients.

Preflight, in-flight, and postflight IOP measurements were collected aboard two U.S. Space Shuttle Program missions in normotensive control, OHT, and glaucomatous crewmembers. METHODS: Five subjects (three controls, one glaucomatous, one OHT) were studied aboard 2-wk Space Shuttle missions. Baseline IOP (triplicate; handheld tonometry) was recorded during training 12 mo preflight, in flight (114 d), and postflight (329 d). Subjective symptoms were recorded via questionnaires. Data were analyzed using a spreadsheet with two-sample t-tests. P-value < 0.05 determined significance. RESULTS: IOP increased for all in-flight vs. preflight measurements for controls (N 3, 48.9, 16.9, 5.85), OHT (N 1, 20.3), and glaucomatous (N 1, 32.2) groups. IOP eventually returned to baseline postflight [Return (R)35 d], except for the astronaut with OHT (R917). Subjective symptoms, likely multifactorial, included blurredvision, decreased visual acuity, and headaches. DISCUSSION: IOP increased during spaceflight and normalized upon return. Astronauts and commercial spaceflight participants may need screening for elevated IOP to potentially prevent sequelae related to glaucoma and OHT, the former which requires treatment in flight and the latter which may need prophylaxis. Previous studies have shown elevated IOP upon entry into microgravity with various normalization timeframes in flight and postflight. It is unclear how increased IOP relates to spaceflight-associated neuro-ocular syndrome (SANS); however, several hypotheses exist. Abstract:
Purpose To correlate functional damage over time detected by standard automated perimetry (SAP) and frequency doubling technology (FDT) with central corneal thickness (CCT) in patients with ocular hypertension (OHT). Methods Seventy-eight OHT patients underwent CCT measurements, SAP, and FDT (the latter two also after 12 and 18 months). Patients were divided into three equally sized groups of 26 patients each: thin (<540 üm), normal (540–580 üm), and thick cornea (>580 üm). The frequency of abnormal FDT and SAP results was analyzed over time (Pearson χ2 test). Results Six of 26 patients with thin corneas (23.1%) presented an abnormal FDT test at baseline, compared to 1 of 26 (3.8%) in the normal thickness cornea group and 1 of 26 (3.8%) in the thick cornea group. After 12 months, the abnormal FDT tests were as follows, respectively: 9 of 26 (34.6%), 2 of 26 (7.7%), and 2 of 26 (7.7%). For SAP the abnormal results were as follows, respectively: 8 (30.1%), 5 (19.2%), and 2 (7.7%). After 18 months, the abnormal FDT tests were as follows, respectively: 16 (61.5%), 5 (19.2%), and 5 (19.2%). For SAP, the abnormal results were as follows, respectively: 10 (38.5%), 5 (19.2%), and 2 (7.7%). Conclusions OHT patients with thinner corneas have a greater risk of developing functional damage over time.

BackgroundThere are more than one million National Health Service visits in England and Wales each year for patients with glaucoma or ocular hypertension (OHT). With the ageing population and an increase in optometric testing, the economic burden of glaucoma-related visits is predicted to increase. We examined the conversion rates of OHT to primary open-angle glaucoma (POAG) in England and assessed factors associated with risk of conversion.MethodsElectronic medical records of 45 309 patients from five regionally different glaucoma clinics in England were retrospectively examined. Conversion to POAG from OHT was defined by deterioration in visual field (two consecutive tests classified as stage 1 or worse as per the glaucoma staging system 2). Cox proportional hazards models were used to examine factors (age, sex, treatment status and baseline intraocular pressure (IOP)) associated with conversion.ResultsThe cumulative risk of conversion to POAG was 17.5% (95% CI 15.4% to 19.6%) at 5 years. Older age (HR 1.35 per decade, 95% CI 1.22 to 1.50, p<0.001) was associated with a higher risk of conversion. IOP-lowering therapy (HR 0.45, 95% CI 0.35 to 0.57, p<0.001) was associated with a lower risk of conversion. Predicted 5-year conversion rates for treated and untreated groups were 14.0% and 26.9%, respectively.ConclusionLess than one-fifth of OHT patients managed in glaucoma clinics in the UK converted to POAG over a 5-year period, suggesting many patients may require less intensive follow-up. Our study provides real-world evidence for the efficacy of current management (including IOP-lowering treatment) at reducing risk of conversion.

Abstract:
The granite processing industry generates large amounts of bottom granite dust waste every day. After the drying and heating process of concrete mixture production, the granite dust is blown and collected in the filtering nozzle. This very fine particle granite dry fly dust, with a particle size maximum distribution of 500 μm, can easily be blown away by wind and cause serious environmental impacts. The use of this waste material would be an effective way to reduce such impacts. Therefore, this paper presents an experimental study on the potential of granite dust as a filler in enhancing the mechanical performance of a hybrid basalt/glass (WB/GCSM) composite. The unhole and open hole tensile (UHT and OHT) properties, low velocity impact (LVI) properties, quasi-static indentations (QSI) properties, flexural properties, interlaminar shear stress (ILSS) properties, and morphology of the developed WB/GCSM composites were evaluated. To meet the objective of this study, composite specimens were produced using 1.5–60 μm granite fly dust at three (3) different loadings (1, 3 and 5 wt%). This granite fly dust was incorporated into polyurethane resin using a mechanical stirring technique. The production of FRP laminates then completed using a hand lay-up and vacuum bagging technique. Four types of the WB/GCSM composites systems, i.e., [WB/GCSM], [WB/GCSM/1GD], [WB/GCSM/3GD] and [WB/GCSM/5GD] were fabricated and compared. The analysis results for the mechanical tests revealed that the incorporation of granite dust of up to 3 wt% had increased the UHT, OHT, LVI, QSI, flexural and ILSS properties of all WB/GCSM composites systems. Higher levels of damage tolerance in UHT and OHT tests, and increased ductility index in the LVI test were obtained when granite dust was added up to 5 wt%. However, a remarkable improvement in all mechanical properties was noticed for [WB/GCSM/1GD], which recorded the highest mechanical performance among all WB/GCSM composite systems.

Abstract:
The current drive for sustainability demands recyclable matrices for composite materials. Vitrimers combine thermoset properties with reprocessability, but their mechanical performance in highly loaded applications, for instance, composites for aeronautics, is still to be demonstrated. This work presents the complete mechanical characterization of a new vitrimer reinforced with carbon fiber. This vitrimer formulation consists of functional epoxy groups and a new dynamic disulfide crosslinks-based hardener. The testing campaign for the vitrimer composites encompassed tension, compression, interlaminar shear strength (ILSS), in-plane shear (IPS), open-hole tension (OHT) and compression (OHC), filled-hole compression (FHC) and interlaminar fracture toughness tests under mode I and II. Test conditions included room temperature and high temperature of 70 °C and 120 °C, respectively, after moisture saturation. Tension and flexural tests also were applied on the neat vitrimer resin. The results compared well with those obtained for current aeronautic materials manufactured by Resin Transfer Molding (RTM). The lower values observed in compression and ILSS derived from the thermoplastic veils included as a toughening material. This work demonstrates that the vitrimer formulation presented meets the requirements of current matrices for aeronautic-grade carbon-reinforced composites.

Abstract:

An increasing number of parents are opting-out their children from high-stakes. Accountability systems in education have used students? test scores to measure student learning, teacher effectiveness, and school district performance. Students who are opted-out of high-stakes tests are not being evaluated by the state tests, making their level of achievement or proficiency unknown by the state government. The purpose of this basic interpretive qualitative study was to gain an understanding of the various reasons, factors, experiences, and personal events that led parents to opt-out their children from at least one 3rd through 8th grade high-stakes test. Data were collected using a researcher-designed semi-structured interview protocol developed using ecological approaches to systems theories and critical pedagogy theories. The study was set in New York and 10 participants were interviewed, all from different rural or small suburban school districts throughout the state. Five themes and 12 subthemes emerged from first and second cycle coding. Key findings indicated that parents decided to opt-out their children from high-stakes tests because they felt high-stakes were inappropriate and unfair. Further, parents were dissatisfied with current high-stakes testing practices. Previous 3rd through 8th grade testing procedures that allowed teachers to make and grade the state tests were seen as acceptable. Parents indicated no issue with testing. However, from a social change perspective they felt the current system of high stakes testing was used improperly to rate students, teachers, programs, and school districts, and that testing should be used to drive instruction and help struggling students. This study is beneficial for school personnel and policy makers because it provides different ways to assess student achievement.

Depois de algumas décadas da entrada das mulheres no mercado de trabalho, esperava-se que a equidade de gênero já fosse uma realidade nas organizações. Porém, os números mostram que os homens ainda são maioria e que quanto mais alto o cargo, menor a quantidade de mulheres. No Brasil, segundo pesquisa do Instituto Brasileiro de Geografia e Estatística (IBGE), em 2016, 60,9% dos cargos gerenciais, tanto no poder público quanto na iniciativa privada, eram ocupados por homens e 37,8% por mulheres. As mulheres continuam ganhando menos com salários em torno de 75% inferiores ao dos homens. A questão do aumento da presença das mulheres nas organizações deve ser vista como um problema econômico, pois elas representam talentos que podem contribuir para o aumento da produtividade e dos resultados financeiros das organizações, gerando impactos positivos em toda a economia. De fato, a maioria das mulheres possui carreiras não lineares e realizam mudanças de trajetórias em suas carreiras. O termo \"opt out\" foi utilizado para explicar a saída de mulheres altamente qualificadas das organizações. Acreditava-se que o principal motivo desta mudança de carreira era em função da maternidade e da família. Porém, Mainiero e Sullivan (2005) fizeram uma extensa pesquisa e descobriram que os motivos do \"opt out\" eram muito mais complexos. Necessidade de oportunidades interessantes, reconhecimento e equilíbrio entre vida pessoal e profissional são fundamentais para as mulheres. Desenvolveram, então, um novo modelo de carreira chamado de modelo da carreira caleidoscópio. Baseado no funcionamento do caleidoscópio, considera que as decisões de carreira dependem de três pilares: autenticidade, balanço e desafio. As pessoas ajustam suas carreiras de acordo com a importância de cada um desses pilares ao longo de suas vidas. Com base no modelo da carreira caleidoscópio, esta pesquisa de natureza qualitativa analisou os fatores que influenciam as decisões de carreira de mulheres executivas. No referencial teórico, foram considerados 3 construtos principais: carreira, gênero e ambiente corporativo. Foram realizadas 18 entrevistas não estruturadas com 2 grupos de mulheres na fase intermediária de carreira e com pelo menos 1 filho: mulheres que permanecem na carreira corporativa e mulheres que mudaram de carreira. A análise dos resultados foi feita em 2 partes: a análise cronológica das carreiras dessas mulheres e a análise de conteúdo com a categorização e análise das respostas obtidas. As conclusões mostraram que as decisões de carreira das mulheres são extremamente complexas e dependem de inúmeros fatores. Vida pessoal e profissional devem ser analisadas de forma integrada e a maternidade não é o fator decisivo para as mudanças de carreira das mulheres. O modelo da carreira caleidoscópio mostrou-se bastante aderente à análise efetuada e os pilares autenticidade, balanço e desafio foram priorizados de acordo com cada momento. Clima organizacional e liderança positivos, políticas corporativas, suporte da família e do parceiro e equilíbrio nos pilares autenticidade, desafio e balanço são alguns dos fatores que contribuem diretamente para a permanência na carreira corporativa. Diante de dificuldades, insatisfação e desequilíbrios nos pilares da carreira caleidoscópio, as mulheres tomam suas decisões de carreira e em alguns casos, fazem o \"opt out\" total.
After a few decades of women entering the labor market, it was expected that gender equity would already be a reality in organizations. However, men are still the majority and the differences are worse at the top positions. In Brazil, according to IBGE, in 2016, women represented only 37,8% of the management positions in the state and private companies and their wages were 25% lower than men\'s. To increase the presence of women in organizations should be seen as an economic problem, since they represent talents that can contribute to the increase of productivity and financial results of the organizations, generating positive impacts throughout the economy. In fact, most women have non-linear careers and make career changes. The \"opt out\" term was used to explain the evasion of highly skilled women from organizations. Maternity and family reasons were considered the main reasons for these career changes. However, Mainiero and Sullivan (2006) found out that the opt out reasons were more complex. Interesting opportunities, recognition and balance between personal and professional life were considered very important factors for women\'s lives. As a result of this research, they developed the kaleidoscope career model. Based on the kaleidoscope\'s system, it considers that career decisions depend on three pillars: authenticity, balance and challenge. People adjust their careers according to the importance of each of these pillars throughout their lives. Based on the kaleidoscope career model, this qualitative research examined the factors that influence the career decisions of executive women. In the theoretical reference, 3 main constructs were considered: career, gender and corporate environment. There were 18 unstructured interviews with 2 mid-career women groups and at least 1 child: women who remained in the corporate career and women who changed their careers. The analysis of the results was done in two parts: the chronological analysis of the careers of these women and the content analysis with the categorization and analysis of the answers obtained. The findings showed that women\'s career choices are extremely complex and depend on a number of factors. Personal and professional life should be analyzed in an integrated way and motherhood is not the decisive factor for women\'s career changes. The kaleidoscope career model showed to be quite adherent to the analysis performed and the authenticity, balance and challenge pillars were prioritized according to each moment. Positive organizational climate and leadership, corporate policies, family and partner support and the pillars of authenticity, challenge and balance are some of the factors that directly contribute to the permanence in the corporate career. Difficulties, dissatisfaction and imbalances in the cornerstones of the kaleidoscope career can lead women to make significant career decisions and, in some cases, opt out.

Abstract:
Organic hydroperoxide resistance (Ohr) proteins are highly efficient thiol-based peroxidases that play central roles in bacterial response towards organic hydroperoxides. In Fungi, Ohr frequently presents a N-terminal extension, which is predicted to target them to mitochondria. The catalytic triad of Ohr comprises the peroxidatic Cys (Cp), the catalytic Arg (Rc) and a Glu (Ec) are fully conserved and interact among themselves by a salt bridge network in a reduced form of the enzyme (the so-called closed state). After getting oxidized to sulfenic acid (Cys-SOH), Cp condenses with the sulfhydryl group of resolution Cys (Cr) in a disulfide bond. The absence of negativity of the thiolate (RS-) in Cp facilitates the opening of the Arg-loop (containing the Rc) away from the active site, generating the so-called open state. However, the molecular events associated with the high reactivity of Ohr enzymes towards hydroperoxides and its specific reducibility by the dihydrolipoamide (DHL) or by lipoylated proteins were still elusive before this work. Additionally, several factors support the idea of Ohr as a target for drug development: (i) Ohr displays unique physicochemical properties; (ii) bacteria mutant for Ohr (Δ ohr) are highly sensitive to oxidative stress; (iii) the indications that Ohr might be involved in bacterial virulence; and (iv) its absence in mammals and vascularized plants. In this thesis, several aspects of Ohr enzymes were evaluated. In chapter 2, we biochemically characterized the Ohr homologs from the ascomycete fungus Mycosphaerella fijiensis Mf_1 (MfOhr), the causative agent of Black Sigatoka disease in banana plants, which presented extraordinary reactivity towards linoleic acid hydroperoxides (kobs = 3.18 (± 2.13) ×108 M-1.s-1). Furthermore, through subcellular fractionation of M fijiensis protoplast cells followed by western blot analysis, we confirmed the in silico prediction that MfOhr is a mitochondrial protein. In chapter 3 and 4, we described seven new crystallographic structures from two opportunistic pathogen, one from Xylella fastidiosa and six from Chromobacterium violaceum (including the first representative of the complex between Ohr and its biological reductant, DHL). Taken together these structures might represent new snapshots along the catalysis. Furthermore, several molecular modelling approaches, such as classical mechanics (MM), steered molecular dynamics (SMD), hybrid quantum mechanics (QM-MM) and together with enzymatic assays of point mutations, indicated that Ohr underwent unique structural switches to allow an intermittent opening (oxidized state) and returning to a more stable closed form (reduced state) of an Arg-loop along catalysis. Remarkably, dihydrolipoamide directly assisted the closing the Arg-loop and thereby the turnover of the enzyme. In chapter 5, we describe the identification of two compounds (C-31 & C-42) that could represent a framework for further studies attempting to find specific Ohr inhibitors, either through ab initio design of chemical compounds and virtual screening using pharmacophoric models. The IC50 calculated for C-31 and C-42 were 124.4-248.5 µM and 243.3-321.7 µM, respectively. Finally, this thesis highlights several new aspects related to Ohr function: 1 - evidence that eukaryotic Ohr are preferentially located in mitochondria and share several biochemical properties with the prokaryotic ones; 2 - the network of polar interactions among residues of the catalytic triad (Cp, Rc and E) strongly contributed to stabilize Ohr in the closed state, in an optimum configuration for hydroperoxide reduction; 3 - evidence that disulfide bond formation and the product release (alcohol derived from hydroperoxide reduction) facilitate the opening of the Rc loop to an intermediate state (probably not to the excessively open state presented in crystallographic structures); 4 - mapping the interactions between the biological reductant (DHL) and the Ohr active site; 5 - strong indications that DHL is not able to fit and react with Ohr in the close conformation; 6 - the first trials for search of molecules to specifically target Ohr proteins, although further assays must be performed to verify the specificity of the selected compounds to target Ohr. Therefore, we describe relevant new information for an antioxidant protein that displays highly efficient catalysis, comparable to other very important hydroperoxide removing enzymes, such as GSH peroxidase and peroxiredoxin
As proteínas Ohr (Organic hydroperoxide resistance) são peroxidases dependente de tiól extremamente eficientes e têm um papel central na resposta das bactérias contra peróxidos orgânicos. Em fungos, as proteínas Ohr apresentam uma extensão N-terminal, cujo predições in silico apontam estar associada ao direcionamento da proteína para a mitocôndria. A tríade catalítica é composta pela cisteína peroxidatic (Cp), a arginina (Rc) e o glutamato (Ec) catalíticos que são totalmente conservados e interagem entre eles por uma rede de interações de ponte salina, na forma reduzida da proteína (conformação fechada). Após se tornarem oxidadas em ácido sulfênico (Cis-SOH), a Cp condensa com o grupo sulfidrila da cisteína de resolução (Cr) numa ligação disulfeto. A ausência da carga negativa do tiolato (RS-) da Cp facilita a abertura da alça que contem a Rc para longe do centro ativo, gerando a conformação aberta. No entanto, os eventos moleculares associados a alta reatividade das enzimas Ohr contra hidroperóxidos e a sua redução pela dihydrolipoamida (presente em proteínas lipoiladas), ainda está descrita de forma bem superficial. Adicionalmente, vários fatores suportam a ideia de que a Ohr seria um potencial alvo para o desenvolvimento de drogas: (i) a Ohr exibe propriedade físico-químicas únicas; (ii) as bactérias mutantes para Ohr (Δohr) são fortemente sensíveis ao stress oxidativo; (iii) indicações de que a Ohr poderá está envolvida na virulência de várias bactérias; e (iv) a ausência de Ohr em mamíferos e plantas vascularizadas. Nesta tese, vários aspetos relacionados com as enzimas Ohr foram avaliados. No Capitulo 2, foi caracterizada bioquimicamente a proteína Ohr homologa de fungo ascomiceto, Mycosphaerella fijiensis Mf_1 (MfOhr), o agente causador da doença de bananas, Sigatoka-negra. A enzima apresentou eficiente atividade contra peroxido de ácido linoleico (kobs = 3.18 (± 2.13) ×108 M-1.s-1). Além disso, através do fracionamento sub celular de protoblasto de M fijiensis seguido de western blot, foram confirmadas as predições in silico de que a MfOhr é uma proteína mitocondrial. No capítulo 3 e 4, foram descritas sete estruturas cristalográficas oriundas de dois patógenos oportunistas, uma de Xylella fastidiosa e seis de Chromobacterium violaceum (incluindo o primeiro representante do complexo entre a Ohr e o seu redutor biológico, DHL). Estas estruturas poderão representar diferentes conformações ao longo do ciclo catalítico. Adicionalmente, várias abordagens de modelagem molecular, tais como mecânica clássica (MM), mecânica molecular direcionada (SMM) e mecânica quântica híbrida (QM-MM), juntamente com ensaios experimentais com mutações pontuais, indicaram que a Ohr sofre várias mudanças conformacionais para permitir uma abertura intermitente (estado oxidado) e o retorno para uma conformação fechada mais estável (estado reduzido) da alça da arginina ao longo da catálise. Notavelmente, a dihydrolipoamide assistiu diretamente o fechamento da alça da arginina e por consequência o turnover da enzima. No capítulo 5, foi descrita a identificação de dois compostos (C-31 e C-42) que representam estudos iniciais com a finalidade de encontrar inibidores específicos para a enzima Ohr. Estes compostos foram encontrados por ab initio design e por varrimento virtual com o uso de modelos farmacofóricos. Os IC50 calculados para o C-31 e C-42 foram de 124.4-248.5 µM e 243.3-321.7 µM, respectivamente. Finalmente, esta tese descreve vários aspetos relacionados com a função da Ohr: 1 - evidências que as Ohr de eucariotos estão preferencialmente localizadas na mitocôndria e partilham várias propriedades bioquímicas com as Ohr de bactéria; 2 - a rede de interações polares entre os resíduos da tríade catalítica (Cp, Rc e Ec) contribuem fortemente para a estabilização do estado fechado, a configuração ótima para a redução de hydroperoxidos; 3 - evidências de que a formação da ligação disulfeto e a liberação do produto (álcool derivado da redução do hydroperoxido) facilitam a abertura da alça da arginina até um estado intermediários (provavelmente não o estado totalmente exposto apresentado nas estruturas cristalográficas) 4 - o mapeamento das interações entre o redutor biológico no centro ativo da Ohr; 5 - fortes indicações de que a DHL não é capaz de interagir e reagir com a Ohr na conformação fechada; 6 - os primeiros ensaios para a procura por moléculas que especificamente interajam com a Ohr, apesar de que futuros ensaios terão de ser executados para verificar a especificidade dos compostos selecionados. Speciallärarprogrammet, Skolutveckling och ledarskap, Lärande och samhälle, Malmö högskola.Syfte och frågeställningarVårt övergripande syfte är att undersöka lärares syn på att testa elever i behov av stöd, elevernas upplevelse av att bli testade samt hur testanvändning kvalificerar sig i ett specialpedagogiskt relationellt perspektiv. Våra frågeställningar är: Finns det något samband mellan förekomsten av professionellt lärande mellan lärare och sättet på vilket lärare väljer att kartlägga elever i behov av stöd? Ger testresultat den information man behöver för att hjälpa elever i behov av särskilt stöd? Hur väljs tester ut? Hur används resultatet i det fortsatta lärandet? Kan det uppstå några etiska problem i samband med testning och i så fall vilka? Teoretisk förankringVi kommer att utgå från ett sociokulturellt perspektiv, i vilket språkets roll är av stor vikt vid lärande och utveckling. Även det relationella perspektivet kommer att vara en utgångspunkt. I detta perspektiv letas förklaringar till skolproblem i mötet mellan eleven och den omgivande miljön. Även systemteorin är av vikt då denna problematiserar de hinder som finns i den miljö/verksamhet som eleven befinner sig i. MetodVi har valt att använda en kvalitativ metod i form av fokusgruppsintervju. Undersökningen gjordes på två olika skolor i Skåne. Sammanlagt ingick åtta lärare och fyra elever i vår studie. Resultat Våra resultat visar att test är vanligt förekommande som kartläggningsmaterial där ingen djupare analys av testmaterialet förekommer. Lärarna tycker inte att det finns någon etisk problematik kopplad till test då det enda syftet är att upptäcka de elever som är i behov av särskilt stöd. Testresultatens roll i elevens fortsatta lärande yttrar sig till största del på individnivå vilket innebär att eleven blir bärare av problemet. Vår studie visar även på att lärarna inte tilldelas tid till kollegialt lärande som hade kunnat bidra till andra verktyg i kartläggningssammanhang som alternativ till test. Vi kan utifrån vår studie bestående av åtta lärare visa att det råder en brist på kollegialt lärande och didaktiska samtal kollegor emellan men även hur lärare och elever kommunicerar angående lärande.Specialpedagogiska implikationerSkolan bör alltid ta hänsyn till elevens kontextuella villkor vid en kartläggning. Ett lågt testresultat innebär inte alltid att eleven inte kan eller klarar av det eleven blir testad på. Eleven bör erbjudas andra sätt att få visa sina kunskaper på än endast test. Vidare bör eleven ses utifrån alla tre nivåerna: organisation-, grupp- och individnivå och inte endast det sistnämnda. Skolorganisationen bör även ha mer tid till pedagogiska diskussioner om hur lärandemiljöerna kan anpassas till alla elever.

Abstract:
Cette recherche traite des effets de participation des salariés sur la performance de l'entreprise. Cette thèse analyse les trois dimensions de la participation des salariés : économique (actionnariat salarié), politique (participation au gouvernement d'entreprise) et organisationnelle. Dans une première partie, cette recherche étudie théoriquement les effets induits par la participation des salariés, en mobilisant plusieurs champs de la littérature en gestion. A l'issue de cette première partie, un ensemble d'hypothèses de recherche est formulé. Dans une seconde partie, cette recherche analyse empiriquement les effets de participation des salariés sur la performance de l'entreprise, grâce à des tests statistiques réalisés sur deux échantillons. Le premier échantillon est constitué de 189 entreprises françaises cotées sur la période 2001-2004. Le second échantillon (transversal) analyse plus spécifiquement les effets de la participation politique des salariés au gouvernement d'entreprise. Les résultats principaux montrent que : 1/la relation entre actionnariat salarié et performance de l'entreprise est de type curvilinéaire ; 2/ la représentation des salariés modère négativement la relation actionnariat salarié – performance. Dans une troisième partie, un modèle théorique issu de la confrontation des résultats empiriques et d'une analyse théorique est proposé
This research deals with the participation effects of employees on firm performance. This thesis explores three dimensions of employees' participation: economic (employee ownership), political (involvement in corporate governance structure) and organizational. In the first part, this research theoretically examines effects of employees' participation using different theoretical frameworks. At the end of the first part, some research hypotheses are formulated. In a second part, this research empirically examines participation effects on firm performance thanks to statistical tests made on two samples. The first sample is based on 189 French listed firms on 2001-2004. The second one, cross sectional, investigates more specifically political representation of employees on corporate governance structure. The majors results show that : 1/ the relationship between employee ownership and firm performance is curvilinear. 2/ board employee representation negatively moderates employee ownership - firm performance relationship. In a third part, a theoretical model, combining empirical results and theoretical in-depth analysis, is proposed

Resumo: A farmacodinâmica vascular da síntese endógena de NO ficou conhecida devido ao efeito de drogas nitrovasodilatadoras. Esses agentes exógenos, tais como o nitroprussiato de sódio, agem como doadores de NO e exercem efeito hipotensivo in vivo direto promovendo diminuição na resistência vascular sistêmica decorrente de um aumento nos níveis de GMP cíclico. Nosso interesse reside na possibilidade de complexos de rutênio transnitrosilaminados servirem como veículo para a liberação controlada de NO em sistemas biológicos e, assim, produzir efeitos farmacológicos. O objetivo desse trabalho foi investigar o efeito hipotensor e a toxicidade de um novo complexo de rutênio, o t-[Ru(NH3)4P(OEt)3NO](PF6)3 devido a sua habilidade em liberar NO espontaneamente em solução aquosa. Medidas diretas de PA foram realizadas em ratos Wistar machos adultos cronicamente instrumentados. O efeito hipotensor foi determinado em ratos conscientes (n=10) submetidos a injeções endovenosas "in bolus" de trans-[Ru(NH3)4P(OEt)3NO](PF6)3 e nitroprussiato de sódio (NaNP). Concentrações de 0,0025 a 0,08 µmoles/Kg do complexo de rutênio induziram resposta hipotensora imediata (PAM controle 104 ± 4,1 mmHg) em uma faixa de ação de 7 a 27%, similar ao efeito observado para o NaNP (11a 32%). Uma correlação dose-efeito positiva foi observada. A toxicidade foi avaliada em cultura de células (IC50) e pela toxicidade aguda em camundongos (LD50). Estudos da citotoxicidade foram conduzidos em cultura de hepatócitos (células metabolizantes), V79 (células não metabolizantes) e V79 tratada com o complexo biotransformado pelos hepatócitos. Hepatócitos de rato foram isolados pelo método de perfusão com colagenase em duas etapas do fígado inteiro. A viabilidade celular foi avaliada por três ensaios, os quais são indicativos de citotoxicidade: redução do MTT, incorporação do vermelho neutro (VN) e conteúdo de ácidos nuclêicos (DNA). Esses ensaios permitiram a avaliação dos valores de IC50 para o trans-[Ru(NH3)4P(OEt)3NO](PF6)3 em células V79 e hepatócitos. Na cultura de hepatócitos valores de IC50 superiores a 3,0 mM foram obtidos para todos os ensaios realizados, sugerindo uma baixa toxicidade do complexo. Para cultura de células V79, nossos resultados mostraram efeito citotóxico similar para os três ensaios investigados, com valores de IC50 ao redor de 2,0 mM, mostrando ser este composto menos tóxico que o NaNP (IC50 ~ 0,056 mM). O produto da biorransformação foi avaliado e mostrou baixa toxicidade para os ensaios de DNA, MTT e alta toxicidade para o VN, i.e., o composto biotransformado modificou a integridade lisossomal. A toxicidade aguda foi avaliada e um valor de LD50 igual a 216 mg/Kg foi obtido indicando baixa toxicidade do complexo in vivo.
Abstract: The vascular pharmacology of endogenously synthesised NO is largely predicted by the known effects of nitrovasodilator drugs. These exogenous agents, such as sodium nitroprusside, act as NO donors and exert direct hypotensive effects in vivo promoting a decrease in systemic vascular resistance by increasing levels of cyclic GMP. We have been interested in the possibility that trans-nitrosylammine ruthenium complexes, might serve as vehicles for the controlled delivery of NO into a biological system and thus display useful pharmacological effects, promoting hypotensive effect. The aim of this work was to investigate the hypotensor effect and toxicity of a new ruthenium complex t-[Ru(NH3)4P(OEt)3NO](PF6)3 due to their ability to spontaneously NO release in aqueous solution. Direct measurements of blood pressure were performed in chronically instrumented adult male Wistar rats. The hypotensor effect was determined in conscious rats (n=10) submitted to in bolus intravenous doses of the trans-[Ru(NH3)4P(OEt)3NO](PF6)3 and sodium nitroprusside (NaNP). Concentration range of 0.0025 to 0.08 µmols/Kg induced hypotensive response (MAP control: 104 ±4.1 mmHg) immediately in action range of 7 to 27% similar to that observed for NaNP effect (11 to 32%). A positive dose-effect correlation was observed. The toxicity was evaluated on cell culture (IC50) and by acute dose in mice. Cytotoxicity studies were carried out on cultures of rat hepatocytes (metabolizant cells), V79 (non-metabolizant cells) and V79 treated with the supernatant of the biotransformed complex by hepatocytes. Hepatocytes were isolated from adult male Wistar rats by the two-step collagenase liver perfusion. Cell viability was evaluated by three endpoints which are indicative of cytotoxicity: MTT reduction, Neutral Red Uptake (NRU) and Nucleic Acid Content (DNA). These assays permit the evaluation of IC50 values of trans- [Ru(NH3)4P(OEt)3NO](PF6)3 on V79 and rat hepatocyte cells. On hepatocyte culture values higher than 3.0 mM were determined for all tests, suggesting a low toxicity for the complex. For V79 cultures our results showed a similar cytotoxic effects for the three studied endpoints with an IC50 ranging around 2.0 mM, showing be less toxic than NaNP. The biotransformated product was evaluted and showed low toxicity for DNA, MTT assays and high toxicity for NRU, i.e., the compound biotransformed changed the lysosomal integrity. The acute toxicity was carried out and a LD50 value of 216 mg/Kg was obtained indicating a low toxicity in vivo.

Introduction: Critical Power (CP) is the maximum rate can be maintained in a stable metabolic state muscle work. In the search for a test associated with less error and greater practical application, a series of studies investigating the validity of a single maximal test "all-out" to determine the PC. This test was designed with the assumption that, by inducing the depletion of anaerobic reservations as early as possible, the aerobic system predominantly assume the replenishment of ATP and consequently the amount of work done in this situation will be the next PC. Thus this study aims to explore the impact of hypoxia (FiO2 = 0,13), in determining the PC by 3-MT test. Methods: 8 healthy university students were evaluated (7 men and 1 woman). Incremental ramp test for determination of the maximal and submaximal parameters as well as the braking factor (power +50% Δ /pacing2 ) test three minutes to determine the maximum EP and W` in normoxia and hypoxia (Fi O2 = 0.13) conditions, constant load tests at 100% of EP determined in three minutes maximum. Student t test for the measurements between tests. Results: The main hypothesis of the study was not accepted since the addition of hypoxia did not cause enough stress to the point of changing the anaerobic contribution and interfere with the determination of EP (3-MTHYP = 213±43W, 3-MTNOR = 217±49 Wp = 0.35). There was no significant difference during the constant load tests at 100% of EP (EP 3-MTHYP = 23.95±8 min, EP-3 MTNOR = 19.41±10 min p = 0.36). Conclusion: Adding hypoxia (Fi O2 = 0.13) was not able to change the energy contribution to the point of changing the parameter EP aerobic and anaerobic W `, highlighting the robustness of the model against this intervention. Still, the responses associated with RPE during constant-load test are close to those encountered during constant load tests on PC. Further investigations with different levels of hypoxia are necessary for more conclusive answers.
Introdução: Potência crítica (PC) representa a máxima taxa de trabalho muscular possível de ser mantida em um estado estável metabólico. Na busca por um teste com menor erro associado e maior aplicação prática, uma série de trabalhos vem investigando a validade de um único teste máximo all-out para determinar a PC. Este teste foi concebido com o pressuposto de que, induzindo o esgotamento das reservas anaeróbias o quanto antes possível, o sistema aeróbio assumiria predominantemente a reposição de ATP e consequentemente a carga de trabalho realizada nessa situação será próxima a PC. Assim este estudo, tem como objetivo explorar o impacto da hipóxia (i.e. FiO2 0.13), na determinação da PC pelo teste máximo de três minutos (3-MT). Métodos: Oito universitários saudáveis foram avaliados (7 homens e 1 mulher). Teste incremental de rampa foi utilizado para determinação dos parâmetros máximos e submáximo bem como do fator de frenagem (potencia+50%∆ / cadendia2). Teste de 3-MT para determinação do EP e W` nas condições normoxia e hipóxia (Fi O2=0,13). Testes de carga constante a 100% dos EP foram utilizados para verificação das respostas fisiológicas. O teste t de student foi utilizado para as aferições entre os testes. Resultados: A hipótese principal do estudo não foi aceita, uma vez que a adição de hipóxia não gerou estresse suficiente ao ponto de alterar a contribuição anaeróbia e interferir na determinação do EP (3-MTHYP = 213±43W, 3-MTNOR = 217±49W p=0,35) . Também não houve diferença significativa durante os testes de carga constante a 100% dos EP determinados (EP 3-MTHYP = 23,95±8 min., EP 3-MTnor = 19,41±10 min. p= 0,36). Conclusão: A adição de hipóxia (Fi O2=0,13) não foi capaz de alterar a contribuição energética a ponto de alterar o parâmetro aeróbio EP, e anaeróbio W`, destacando a robustez do modelo frente a essa intervenção. Ainda, as respostas associadas ao EP durante o teste de carga constante estão próximas aquelas encontradas durante testes de carga constante na PC. Futuras investigações com diferentes níveis de hipóxia são necessárias para obter respostas mais conclusivas.

Afin de proposer un modèle de perception et de réduction du risque alimentaire prenant en considération l'acte d'achat et de consommation, l'auteur mobilise trois champs conceptuels complémentaires : le champ alimentaire, la théorie du risque perçu et le modèle transactionnel du stress et du coping. A l'aide d'une analyse des structures de covariance, l'auteur montre que le consommateur peut utiliser différentes stratégies de réduction du risque qui remplissent fonctions : 1/ réduction du risque perçu en mettant en œuvre des stratégies d'ajustement centrées sur le problème, 2/ maîtrise des conséquences émotionnelles liées à la perception du risque perçu grâce à l'emploi de stratégies de coping orientées vers l'émotion, mise en œuvre d'une stratégies mixte d'ajustement (appropriation).

L’approche de la problématique des agirs via les tests projectifs en se penchant sur l’expression des défaillances narcissiques du sujet, propose, le plus souvent, une vision unitaire basée sur le mécanisme du recours à l’acte tel que définit par Balier. Nous proposerons de traiter cette question à partir d’une dimension plurielle différenciant deux types d’agirs : le recours à l’acte (Balier) et l’acting-out (Lacan). Nous présenterons une étude mixte, quantitative et qualitative, à partir de la comparaison de Rorschach et TAT administrés à 24 sujets. Notre analyse de ces deux formes d’agir portera principalement sur la capacité du sujet à pouvoir ou non se différencier de l’environnement. Le groupe recours à l’acte se caractérise par une continuité intérieur / extérieur et un transitivisme morbide expression d’une absence de constitution d’un soi différencié. Le groupe acting-out présente une problématique narcissique spécifique dans la validation du spéculaire par le symbolique. Les prises en charges thérapeutiques peuvent ainsi s’aider de ces résultats pour proposer des approches différentielles au sein desquelles les tests projectifs pourraient avoir une fonction pré-thérapeutique
In the modern societies, the answer to the criminal act is a prison sentence. Due to the nature of certain acts, with this prison sentence, some court-ordered treatments (COT) (obligations and injunctions of treatments) have been added. Ethically, this law request and its social asking don’t exempt to think about the prisoner ‘asking, which is the trigger of psychotherapies. There is any retroactivity of the law and of pronounced sentence. For this research, the prisoners were selected on their own asking of care (the court-ordered treatments couldn’t applicate). The asking of the prisoners is therefore a criterion common to them (these prisoners). In this context, the acts have distinguished according to the mechanisms which have governed the nature of the act: recourse of act or acting out. Many experts agree on the existence of premature narcissistic failures. However, discussions persist on the nature of the act and on its aspect, either single dimension or multiple and complex dimensions (objective behavior, social consequences and underlying mechanisms ...). Yet all these theoretical models on narcissistic failures lean on the “recourse to act” as if there was only a simple nature to act

AbstractThis paper presents the results of a study, carried out as part of the design-based development of an online self-assessment for prospective students in higher online education. The self-assessment consists of a set of tests – predictive of completion – and is meant to improve informed decision making prior to enrolment. The rationale being that better decision making will help to address the ongoing concern of non-completion in higher online education. A prototypical design of the self-assessment was created based on an extensive literature review and correlational research, aimed at investigating validity evidence concerning the predictive value of the tests. The present study focused on investigating validity evidence regarding the content of the self-assessment (including the feedback it provides) from a user perspective. Results from a survey among prospective students (N = 66) indicated that predictive validity and content validity of the self-assessment are somewhat at odds: three out of the five tests included in the current prototype were considered relevant by prospective students. Moreover, students rated eleven additionally suggested tests – currently not included – as relevant concerning their study decision. Expectations regarding the feedback to be provided in connection with the tests include an explanation of the measurement and advice for further preparation. A comparison of the obtained scores to a reference group (i.e., other test-takers or successful students) is not expected. Implications for further development and evaluation of the self-assessment are discussed.

Since 1999 ENEA is developing the heavy liquid metal (HLM) technology aiming to support the design and implementation of a Lead cooled Fast Reactor (LFR) and an Accelerator Driven System (ADS), both in the frame of the Italian and European research programs. In these contexts several experiments have been performed, in different fields, going from coolant thermal-hydraulic, component development and structural material characterization. Recently, in the frame of the IP-EUROTRANS (6th Framework Program EU), domain DEMETRA, ENEA assumed the commitment to perform an integral experiment aiming to reproduce the primary flow path of a pool-type nuclear reactor, cooled by Lead Bismuth Eutectics (LBE). This experimental activity, named “Integral Circulation Experiment (ICE)”, has been implemented thanks a joint effort of several research institutes, mainly ENEA and University of Pisa, allowing to design an appropriate test section. This has been installed in the CIRCE facility, the largest worldwide experimental facility for the HLM technology investigation. The goal of the experiments was to demonstrate the technological feasibility of a heavy liquid metal (HLM) pooltype nuclear system in a relevant scale (1 MW), investigating the related thermal–hydraulic behavior under both steady state and transient conditions. This paper reports a description of the experiment, as well as the results carried out in the first experimental campaign run on the CIRCE pool, which consists of a full power steady state test, an un-protected loss of heat sink (ULOH) test, and an un-protected loss of flow (ULOF) test. The post-test analyses of the experiments is presented. The whole domain has been modeled by a suitable 1-D nodalization, and the results carried out have been studied performing numerical calculations by the REALP5 system code modified to take in account the LBE thermal-physical properties when employed as nuclear coolant. The obtained experimental results as well as the performed post-test analysis have demonstrated the thermal-hydraulic and technological feasibility of a pool-type nuclear system cooled by HLM.
