Abstract:
Objective To determine risk factors for multi-drug-resistant Acinetobacter baumannii (MDR-AB) nosocomial infections in intensive care units in a tertiary care hospital, Makkah, Saudi Arabia. Methods We performed a hospital-based, matched case–control study in patients who were admitted to Al Noor Specialist Hospital between 1 January 2012 and 31 August 2012. The study included cases of A. baumannii nosocomial infection and controls without infection. Controls were matched to cases by age and ward of admission. Results The most frequent site of infection was the respiratory tract (77.3%). Susceptibility to antimicrobial MDR-AB was 92.0% for ceftazidime and ciprofloxacin, while it was 83.3% for imipenem, 83.0% for trimethoprim, 79.0% for amikacin, and 72.7% for gentamicin. Multiple logistic regression of risk factors showed that immunosuppression (OR = 2.9; 95% CI 1.5–5.6; p = 0.002), clinical outcome (OR = 0.4; 95% CI 0.3–0.9; p = 0.01), invasive procedures (OR = 7.9; 95% CI 1.8–34.2; p = 0.002), a central venous catheter (OR = 2.9; 95% CI 1.5–5.6; p = 0.000), and an endotracheal tube (OR = 3.4; 95% CI 1.6–7.3; p = 0.001) were associated with MDR-AB. Conclusions Acinetobacter nosocomial infections are associated with admission to the ICU (Intensive care unit) and exposure to invasive procedures.

Abstract:
Abstract RNA binding proteins (RBPs) tightly control mRNA abundance, stability and translation while mutations or altered expression of specific factors can drive malignancy. Yet, the identity of the RBPs that govern myeloid stem cells remains poorly characterized. We and others have recently demonstrated that MUSASHI-2 (MSI2) is a central regulator of the cancer stem cell program in myeloid leukemia. Therefore, we curated a list of 127 MSI2 direct protein interactors and associated genes to perform an in vivo shRNA screen using MLL-AF9 leukemia cells. We identified shRNAs corresponding to 24 genes that were significantly depleted in vivo after sequencing and comparing their representation from day 16 to day 0. We confirmed knockdown and demonstrated marked reduction in myeloid colony formation in vitro after depleting 7 hits identified in our screen. Additionally, we tested these genes in normal bone marrow c-Kit positive cells and found that the most differentially required gene in leukemia cells compared to normal cells was SYNCRIP (Synaptotagmin-binding, cytoplasmic RNA-interacting protein). SYNCRIP is an RNA binding protein that has been implicated in various RNA regulatory processes but its role in the hematopoietic system is virtually unknown. Depletion of SYNCRIP with shRNAs in murine MLL-AF9 leukemia cells resulted in an increase in myeloid differentiation, apoptosis and delayed leukemogenesis in vivo (median survival of 35 days; control versus 61 days shRNA#1 knockdown was selected against, and "not reached" shRNA#2). To further assess SYNCRIP function in vivo, we developed a germline Syncrip knockout (KO) by injecting Cas9-DNA and Syncrip - guides RNAs into embryos and harvested E13 fetal liver cells. After Syncrip deletion was verified by immunoblotting, we observed normal numbers of HSCs and equivalent engraftment in lethally irradiated animals in both primary and secondary transplants. In contrast, we observed a delay in leukemeogenesis (median survival of 87.5 days; WT versus 118 days KO) in recipient mice after transplantation of MLL-AF9 transformed LSKs. Notably, non-deleted leukemia cells outcompeted the SYNCRIP deleted cells based on a reemergence of SYNCRIP expression. These data suggest that SYNCRIP is differentially required in myeloid leukemia cells compared to normal cells. Furthermore, we found that SYNCRIP was highly expressed in wide variety of human AML cell lines and in primary AML patients (n=4/5). SYNCRIP depletion with shRNAs resulted in reduced cell proliferation and the induction of apoptosis in human AML cell lines (MOLM13, NOMO-1, KASUMI-1 and NB4) and a marked decrease in engraftment of primary AML patient cells. To gain insights into SYNCRIP function, we performed RNA-sequencing of leukemia cells depleted for SYNCRIP. Gene set enrichment analysis (GSEA) negatively enriched for the MLL-AF9, HOXA9 and stem cell programs in SYNCRIP-KD cells and positively enriched for MSI2's direct mRNA binding targets and a MSI2 deficient LSC signature. Reciprocal immunoblotting in the presence or absence of RNAse demonstrated that SYNCRIP and MSI2 interaction is RNA dependent. We validated their shared targets by performing SYNCRIP RNA-immunoprecipitation (RIP) for previously identified MSI2's direct mRNAs targets (HOXA9 and c-MYC). SYNCRIP depletion resulted in reduced protein abundance of HOXA9 and c-MYC. Forced MSI2 expression partially rescued the colony formation and HOXA9 expression in SYNCRIP-KD cells. To assess the functional downstream targets of SYNCRIP in leukemia, we overexpressed HOXA9 and c-MYC in SYNCRIP-KD cells and observed that HOXA9 expression but not c-MYC partially rescues the effect of SYNCRIP depletion on myeloid colony formation. Mechanistically, we showed that SYNCRIP regulates translation of HOXA9 without affecting HoxA9 mRNA stability. Overall, we provide a strategy for interrogating the functional RNA binding network in leukemia using shRNA screening. Additionally, we validated SYNCRIP as a novel RBP that controls the leukemia stem cell program and propose that targeting these functional complexes might provide a novel therapeutic strategy in myeloid leukemia. Abstract:

Abstract:
Noor hard red spring wheat (Triticum aestivum L.) was developed using a modified bulk breeding method at the University of Alberta, Edmonton, AB Canada. Noor is an apically awnletted, hollow-stemmed line with a combination of high yield potential, good lodging tolerance, and medium maturity. During the three years (2016–2018) of evaluation in the Parkland Wheat Cooperative test, Noor yielded 12% higher than the mean of all the checks and matured similarly to Carberry and Glenn, but 3.1 and 2.6 d later than AC Splendor and Parata, respectively. Noor was 94.9 cm tall, shorter than AC Splendor (98.4 cm), similar in height to Glenn (94.0 cm) and Parata (93.5 cm), but taller than Carberry (86.0 cm). The lodging score of Noor (2.1) was lower than Parata (3.0) and AC Splendor (3.0), but similar to Carberry (2.3) and Glenn (2.1). The test weight (kg·hL−1) of Noor (80.0) was higher than AC Splendor (78.6), similar to Carberry (80.2) and Parata (80.4), but lower than Glenn (82.2). Grain weight (35 g) and NIR protein of Noor (14.1%) was lower than all the checks. Overall, Noor was rated resistant (R) to the prevalent races of leaf, stem, and stripe rusts during the three years of testing. Noor was rated intermediate (I) to common bunt and Fusarium head blight. Three years of end-use quality evaluation indicated that Noor is acceptable for the Canada Western Red Spring class, with fewer flags.

Abstract:
Urbanisation has historically led to issues with property damage and sickness, due to pests such as rats. Traditionally, rats have been exterminated using poison and manually loaded traps. As of late however, more effective and humane means of control have been developed, such as automatic and connected traps. As rats use the sewer system as we use the highway, connecting properties together, it is natural to place traps there. Nomor, a service company that performs pest control among other services, is interested in developing their own automatic mechanical sewer mounted trap for their own specific needs. In this thesis, the fastening mechanism and enclosure for such a trap are developed, with a concept developed in a previous collaboration used as a base. In this concept, a fastening mechanism is used to fasten the trap to the open pipes located in the sewer manholes. Rats passing underneath the trap are executed by the striking of a carbon dioxide driven cylinder. The enclosure surrounds the fastening mechanism and provides an interface between it and the executional unit, which is developed separately. The final prototype, which was made mostly using 3D-printing of PLA parts and some light metalwork, was tested and evaluated using a testing rig made from layered MDF pieces. The rig was constructed with an optimal scenario in mind, with dimensions like those in pipes which Nomor are likely to want to mount the trap in. The main use of the rig was to evaluate the dimensions and fastening abilities of the trap. Tests and analysis of the construction gave that the fastening mechanism and enclosure had potential to meet the requirements from Nomor in a reliable manner, if a more sophisticated method of mounting and a better mechanism for keeping the trap fastened was developed.

Abstract:
Arbetet delades in i tre huvuddelar: sensormodul, avlivningsanordning, och hissanordning.
This thesis is a continuation of a previous project where a concept for a mechanical sewer mountedrat-trap was developed. As more and more people are moving into cities, rats are becoming a bigger issue. Rats cancarry diseases and they can cause extensive damage. The goal of this thesis has been to developsubsystems for a mechanical sewer mounted rat-trap. Focus has been on developing the mechanismfor detecting rats and the mechanism for killing rats. The other subsystems have been developed inparallell in another work. The thesis was done in collaboration with the company Nomor, a swedish service company thatworks primarily with pest control. The work was split into three main parts: sensors, killing mechanism, and height adjustmentmechanism. CAD-models and mathematical models were produced for the three subsystems. A testrig and physical prototypes were produced to test the subsystems. The result ended up with a prototype consisting of the three mentioned subsystems. Some of thepurchased components did not work the way we anticipated wich meant some parts of the prototypedid not work
