Abstract:
Four examples of para-nitro substituted 2-(arylimino)pyridine-nickel(II) bromide complexes of general formula, [2-{(2,6-R-4-NO2C6H2)N=CMe}C5H4N]NiBr2, but differentiable by the steric/electronic properties displayed by the ortho-groups [R = i-Pr (Ni1), Et (Ni2), CHPh2 (Ni3), CH(4-FPh)2 (Ni4)], have been prepared in good yield. For comparative purposes, the meta-nitro complex, [2-{(2,6-i-Pr2-3-NO2-4-(4-FPh)2C6H)N=CMe}C5H4N]NiBr2 (Ni5), has also been synthesized. The molecular structures of mononuclear Ni3·xH2O (x = 2, 3) and bromide-bridged dinuclear Ni4 and Ni5 are disclosed. Upon activation with either ethylaluminum dichloride (EtAlCl2) or modified methylaluminoxane (MMAO), all precatalysts displayed good catalytic performance at operating temperatures between 30 °C and 60 °C with higher activities generally seen using EtAlCl2 [up to 4.7 × 106 g PE (mol of Ni)−1 h−1]: Ni2 ~ Ni5 > Ni1 ~ Ni4 > Ni3. In terms of the resultant polyethylene (PE), Ni4/EtAlCl2 formed the highest molecular weight of the series (Mw up to 1.4 × 105 g mol−1) with dispersities (Mw/Mn) ranging from narrow to broad (Mw/Mn range: 2.2–24.4). Moreover, the melting temperatures (Tm) of the polymers generated via EtAlCl2 activation fell in a narrow range, 117.8–126.0 °C, which resembles that seen for industrial-grade linear-low density polyethylene (LLDPE). Indeed, their 13C NMR spectra revealed significant amounts of uniformly distributed long-chain branches (LCBs), while internal vinylene groups constituted the major type of chain unsaturation [vinylene:vinyl = 5.3:1 (EtAlCl2) and 9.9:1 (MMAO)].

Abstract:
Nickel-based and cobalt-based metal alloys are frequently used in dentistry. The introduction of various elements in the alloy changes its characteristics, and a thorough study of each alloy should be completed to determine its appropriate corrosion resistance and biocompatibility in contact with physiological fluids. There are scarce investigations on these widely used dental alloys in Ringer solution, and findings in this research bring new experimental data and information. The present study evaluated and compared the corrosion behavior of six NiCr- and two CoCr-based dental materials in Ringer solution, using the following techniques: potentiostatic polarization curves (chronoamperometry), microstructural analysis, and EIS (electrochemical impedance spectroscopy). The results obtained in this investigation showed that in the NiCr-based specimens Ni4, Ni5, and Ni6 the stability of the passive layer was destroyed after polarization and a development and growth of stable pits was found in the microstructural analysis after electrochemical treatment. In terms of susceptibility to corrosion, two different groups of specimens were derived from this investigation. A first group which included the two CoCr (Co1 and Co2) and three of the six NiCr alloys studied (Ni1, Ni2, and Ni3). A second group with the other NiCr alloys investigated Ni4, Ni5, and Ni6.

Abstract:
A collection of five related 2-(arylimino)pyridines, 2-{(2,6-(CH(C6H4-p-F)2)2-4- RC6H2)N=CMe}C5H4N, each ortho-substituted with 4,4′-difluorobenzhydryl groups but distinct in the electronic properties of the para-R substituent (R = Me L1, Et L2, i-Pr L3, F L4, OCF3 L5), were prepared and combined with (DME)NiBr2 to form their corresponding LNiBr2 complexes, Ni1–Ni5, in high yields. All the complexes were characterized by FT-IR, 19F NMR spectroscopy and elemental analysis, while Ni5 was additionally the subject of an X-ray determination, revealing a bromide-bridged dimer. The molecular structure of bis-ligated (L4)2NiBr2 (Ni4’) was also determined, the result of ligand reorganization having occurred during attempted crystallization of Ni4. On activation with either EtAlCl2 or MMAO, Ni1–Ni5 exhibited high catalytic activities (up to 4.28 × 106 g of PE (mol of Ni)−1 h−1 using EtAlCl2) and produced highly branched polyethylene exhibiting low molecular weight (Mw range: 2.50–6.18 kg·mol−1) and narrow dispersity (Mw/Mn range: 2.21–2.90). Notably, it was found that the type of para-R group impacted on catalytic performance with Ni5 > Ni4 > Ni3 > Ni1 > Ni2 for both co-catalysts, underlining the positive influence of electron withdrawing substituents. Analysis of the structural composition of the polyethylene by 1H and 13C NMR spectroscopy revealed the existence of vinyl-end groups (–CH=CH2) and high levels of internal unsaturation (–CH=CH–) (ratio of vinylene to vinyl, range: 3.1:1–10.3:1) along with various types of branch (Me, Et, Pr, Bu, 1,4-paired Me, 1,6-paired Me and LCBs). Furthermore, reaction temperature was shown to greatly affect the end group type, branching density, molecular weight and in turn the melting points of the resulting polyethylenes.

Abstract:
Ninein (Nin) is a centrosomal protein whose gene is mutated in Seckel syndrome (SCKL, MIM 210600), an inherited recessive disease that results in primordial dwarfism, cognitive deficiencies, and increased sensitivity to genotoxic stress. Nin regulates neural stem cell self-renewal, interkinetic nuclear migration, and microtubule assembly in mammals. Nin is evolutionarily conserved, yet its role in cell division and development has not been investigated in a model organism. Here we characterize the single Nin orthologue in Drosophila. Drosophila Nin localizes to the periphery of the centrosome but not at centriolar structures as in mammals. However, Nin shares the property of its mammalian orthologue of promoting microtubule assembly. In neural and germline stem cells, Nin localizes asymmetrically to the younger (daughter) centrosome, yet it is not required for the asymmetric division of stem cells. In wing epithelia and muscle, Nin localizes to noncentrosomal microtubule-organizing centers. Surprisingly, loss of nin expression from a nin mutant does not significantly affect embryonic and brain development, fertility, or locomotor performance of mutant flies or their survival upon exposure to DNA-damaging agents. Although it is not essential, our data suggest that Nin plays a supportive role in centrosomal and extracentrosomal microtubule organization and asymmetric stem cell division.

Abstract:
Most legumes can engage in symbiosis with N-fixing bacteria called rhizobia. This symbiosis, called nodulation, evolved from the more widespread symbiosis that most land plants form with arbuscular mycorrhiza, which is reflected in a common requirement of certain genes for both these symbioses. One key nodulation gene, Nodule Inception (NIN), has been intensively studied. Mutants in NIN are unable to form nodules, which has made it difficult to identify downstream genes under the control of NIN. The analysis of data from our recent transcriptomics study revealed that some genes with an altered expression of nin during nodulation are upregulated in mycorrhizal roots. In addition, another study reported the decreased colonization of nin roots by arbuscular mycorrhiza. We therefore investigated a role for NIN in mycorrhiza formation. Our time course study, using two nin alleles with differing genetic backgrounds, suggests that that loss of NIN does not affect colonization of Medicago truncatula roots, either in the presence or absence of rhizobia. This, and recent phylogenetic analyses showing that the loss of NIN is correlated with loss of nodulation in the FaFaCuRo clade, but not with the ability to form mycorrhiza, argue against NIN being required for arbuscular mycorrhization in legumes.

Məqsəd: operativ tədqiqatın məqsədi ÇDDV-nin uğursuz müalicə nəticələrinin təhlili və onunla əlaqəli olan risk faktorlarının təyini.

Full text
Abstract:
The membrane assembly of polytopic membrane proteins is a complicated process. Using Chinese hamster P-glycoprotein (Pgp) as a model protein, we investigated this process previously and found that Pgp expresses more than one topology. One of the variations occurs at the transmembrane (TM) domain including TM3 and TM4: TM4 inserts into membranes in an Nin-Cout rather than the predicted Nout-Cin orientation, and TM3 is in cytoplasm rather than the predicted Nin-Coutorientation in the membrane. It is possible that TM4 has a strong activity to initiate the Nin-Cout membrane insertion, leaving TM3 out of the membrane. Here, we tested this hypothesis by expressing TM3 and TM4 in isolated conditions. Our results show that TM3 of Pgp does not have de novo Nin-Cout membrane insertion activity whereas TM4 initiates the Nin-Cout membrane insertion regardless of the presence of TM3. In contrast, TM3 and TM4 of another polytopic membrane protein, cystic fibrosis transmembrane conductance regulator (CFTR), have a similar level of de novo Nin-Cout membrane insertion activity and TM4 of CFTR functions only as a stop-transfer sequence in the presence of TM3. Based on these findings, we propose that 1) the membrane insertion of TM3 and TM4 of Pgp does not follow the sequential model, which predicts that TM3 initiates Nin-Cout membrane insertion whereas TM4 stops the insertion event; and 2) “leaving one TM segment out of the membrane” may be an important folding mechanism for polytopic membrane proteins, and it is regulated by the Nin-Cout membrane insertion activities of the TM segments.

Abstract:
Legumes and actinorhizal plants are capable of forming root nodules symbiosis with rhizobia and Frankia bacteria. All these nodulating species belong to the nitrogen fixation clade. Most likely, nodulation evolved once in the last common ancestor of this clade. NIN (NODULE INCEPTION) is a transcription factor that is essential for nodulation in all studied species. Therefore, it seems probable that it was recruited at the start when nodulation evolved. NIN is the founding member of the NIN-like protein (NLP) family. It arose by duplication, and this occurred before nodulation evolved. Therefore, several plant species outside the nitrogen fixation clade have NLP(s), which is orthologous to NIN. In this review, we discuss how NIN has diverged from the ancestral NLP, what minimal changes would have been essential for it to become a key transcription controlling nodulation, and which adaptations might have evolved later.

Abstract:
Qian-Kun-Nin is a Chinese herbal formulation considered to have anti-infection, anti-tumor and immuno-enhancing properties. Data from previous investigations showed that Qian-Kun-Nin causes HIV growth inhibition and immunomodulation in vitro, suggesting that this formula has the ability to inhibit HIV and modulate impaired immune functions in humans. We conducted this pilot study to evaluate the anti-retroviral and immunological enhancing effects of this formula on HIV positive subjects. Eight subjects completed the study, receiving oral Qian-Kun-Nin capsules for 24 consecutive weeks in a single blind design. Compared to baseline level, the plasma virus load decreased significantly at the end of week 12 (p < 0.01) and week 24 (p < 0.01), respectively. Four weeks after cessation of Qian-Kun-Nin treatment, plasma virus load was still significantly lower compared to baseline (p < 0.01). Blood CD4 cell counts were increased significantly at the end of the 12th week compared to the baseline level (p < 0.01). No adverse effects were observed, and no significant side effects were recorded in any subjects. These data suggest that Qian-Kun-Nin has therapeutic potential in the treatment of HIV positive patients.

Abstract:
Vehicle type recognition algorithms are broadly used in intelligent transportation, but the accuracy of the algorithms cannot meet the requirements of production application. For the high efficiency of the multilayer perceptive layer of Network in Network (NIN), the nonlinear features of local receptive field images can be extracted. Global average pooling (GAP) can avoid the network from overfitting, and small convolution kernel can decrease the dimensionality of the feature map, as well as downregulate the number of model training parameters. On that basis, the residual error is adopted to build a novel NIN model by altering the size and layout of the original convolution kernel of NIN. The feasibility of the algorithm is verified based on the Stanford Cars dataset. By properly setting weights and learning rates, the accuracy of the NIN model for vehicle type recognition reaches 97.2%.

Abstract:
Nodulation regulation Legumes convert atmospheric nitrogen into biologically useful ammonium with the help of symbiotic bacteria housed in root nodules. Much of nodule development is controlled by the transcription factor NODULE INCEPTION (NIN). Feng et al . show that NIN is proteolytically processed to release a fragment that regulates the later stages of nodulation when the nodules acquire nitrogen-fixing capability. In related work, Jiang et al . identified members of the NIN-like protein (NLP) transcription factor family as being regulators of leghemoglobin expression acting through an unusual promotor motif shared across legumes. —PJH

Abstract:
Legumes develop root nodules in symbiosis with nitrogen-fixing rhizobial bacteria. Rhizobia evoke cell division of differentiated cortical cells into root nodule primordia for accommodating bacterial symbionts. In this study, we show that NODULE INCEPTION (NIN), a transcription factor in Lotus japonicus that is essential for initiating cortical cell divisions during nodulation, regulates the gene ASYMMETRIC LEAVES 2-LIKE18/LATERAL ORGAN BOUNDARIES DOMAIN16a (ASL18/LBD16a). Orthologs of ASL18/LBD16a in nonlegume plants are required for lateral root development. Coexpression of ASL18a and the CCAAT box–binding protein Nuclear Factor-Y (NF-Y) subunits, which are also directly targeted by NIN, partially suppressed the nodulation-defective phenotype of L. japonicusdaphne mutants, in which cortical expression of NIN was attenuated. Our results demonstrate that ASL18a and NF-Y together regulate nodule organogenesis. Thus, a lateral root developmental pathway is incorporated downstream of NIN to drive nodule symbiosis.
