Full textHepatitis A virus (HAV) is a picornavirus that causes hepatitis A, a zoonotic disease. This virus has only humans and nonhumans primates as its natural hosts. Just one serotype is known, but several strains have been recognized. Those strains are divided into 7 genotypes, 3 of them being exclusively from human strains, other 3 exclusively from Old World primates strains and the genotype III that has human and primate strains. The maximum genetic difference between strains from the same genotype is 15% and the antigenic difference between all strains is almost none. This is why it is possible to use human diagnostic tests for nonhuman primates. The infection route of the HAV is fecal-oral. After the ingestion of the virus by infected food or contaminated objects, the virus replicates in the liver of the animal and reaches the intestines through the bile, being eliminated with the feces. The disease in primates and children is assymptomatic, but when present, it is unspecific and varies from mild signs to death. The diagnosis is made by serological tests or identification from the viral antigen in sera or feces at the acute phase from the disease. The presence of anti-HAV IgM shows acute or recent infection. On the other hand, anti-HAV IgG is found from the convalescent phase of the disease on, through several years. The aim of this project was to research the seroprevalence of anti-HAV antibodies in New World primates and detect the viral antigen in feces from those animals that had acute infection. Sera from 421 animals of 32 different species were tested. From these animals, 13,5% (57/421) were wild animals, 29,7% (125/421) were from the Centro de Primatologia do Rio de Janeiro (CPRJ), 4% (17/421) from breeders, 3,8% (16/421) from Departamento de Parques e Áreas Verdes (DEPAVE) and 48,9% (206/421) were zoo animals. The sera were tested, by immune-enzymatic tests, for the presence of IgM and total anti-HAV antibodies. All the sera were negative for IgM, which means that no animal had acute infection when tested. All wild animals were negative for total anti-HAV, as were the animals from DEPAVE. Four percent (5/125) from the CPRJ animals and 7,6% (17/223) from the zoos/breeders animals were positive for total anti-HAV, showing that a number of captive animals have already been in contact with the virus. The prevalence of anti-HAV antibodies found in this study was lower than expected, as it is known that the number of positive animals in captivity is high. The possible reasons for such low prevalence are discussed. Since humans are the major risk factor for primate infection with HAV, the lower prevalence found at the CPRJ was predictable, because this is a research center, where visitors are not allowed, so the animals have less contact with humans in there, than they do in zoos. Our results lead us to think that hepatitis A is not a disease of high risk for either wild or zoo New World primates.

Objetivou-se condicionar fêmeas de macacos-da-noite ao exame de ultra-som ginecológico. Estabeleceu-se protocolo onde foram avaliadas freqüências cardíacas iniciais, finais e médias (Fci, Fc2 e Fc iv1), tempo de exame (TE), útero e ovários, durante dois períodos e em diferentes turnos. Utilizou-se transdutor linear de 5¬12MHz. O útero e os ovários foram avaliados em cortes sagitais e transversais. Para o útero, levou-se em consideração o número de partos das fêmeas (nulípara, primípara e plurípara). As Fc2 foram mais baixas que as Fc1 (P < 0,01) nos dois períodos. Observou-se correlação negativa entre o TE e Fc2 (r = - 0,14 e P < 0,05). Não houve diferença significativa entre os turnos. O volume uterino foi diretamente proporcional ao número de partos. O volume do ovário direito (VOD) foi maior que o do ovário esquerdo (VOE). Observaram-se correlações positivas (P < 0,05) entre o peso das fêmeas e o VOD (r = 0,28) e VOE (r = 0,16)
The objective was to condition female owl monkeys to the gynaecological ultrasound exam. A protocol was established, where initial, final and average heart rate (HR1, HR2 and AHR) were evaluated, time spent on the examination procedure (T8), uterus and ovary, during two periods and in two different shifts. A linear probe of 5-12MHz was utilized. The uterus and ovary were evaluated at the sagital and transversal scan. As for the uterus, the females' number of parturitions (nuliparous, primiparous and pluriparous) was considered. The HR2 were lower than the HR1 (P < 0.01) in the two periods. A negative correlation was observed between T8 and HR2 (r= ¬0.14 and P < 0.05). There was no significant difference between the shifts. The uterine volume was directly proportional to the number of parturitions. The volume of the right ovary (ROV) was larger than the left ovary (LOV). Positive correlations were observed (P < 0.05) between the females' weight and the ROV (r= 0.28) and VLO (r= 0.16)

Full textBrown howler monkeys (Alouatta guariba clamitans) occur in the Brazilian Atlantic Forest from the states of Minas Gerais and Rio de Janeiro to Rio Grande do Sul, and in a small area in northestern Argentina. They are classified as Vulnerable in the state of Rio Grande do Sul due especially to the loss and fragmentation of their natural habitats, and also due to the deaths caused by the recent outbreak of sylvatic yellow fever (2008/2009). The intense human activities on natural habitats, besides the habitat loss, can favor the spread of pathogenic agents such as bacteria and other parasites which affect men and domestic animals, and which can also occur in wildlife animals. This study aimed at verifying the occurrence of Enterobacteriaceae and Cryptosporidium sp. in feces of free-ranging brown howler monkeys in Santa Maria, Rio Grande do Sul, Brazil, and understanding the influence of ecological and environmental factors (group size, howler population density, fragment size, distance to the nearest human settlement, distance to the nearest river and seasonality) on the richness of such organisms. The samples were analyzed at the Center for Studies and Research on Wild Animals (NEPAS / LCDPA) of the Federal University of Santa Maria. Twenty Enterobacteriaceae species were detected, and with the techniques employed, the samples were negative for Salmonella spp. and Cryptosporidium sp. None of the environmental variables had significant influence on the wealth of Enterobacteriaceae. Considering that howler populations at CISM (Campo de Instrução de Santa Maria) fell dramatically due to yellow fever, and that some organisms found in this study may interact with other factors and affect the population dynamics of the howler. We believe that it is fundamental to continue monitoring the health of these populations in order to better understand disease mechanisms, as well as conserve this species.
O bugio-ruivo (Alouatta guariba clamitans) ocorre na Mata Atlântica dos estados de Minas Gerais e Rio de Janeiro ao Rio Grande do Sul e em pequena porção do nordeste da Argentina. É classificado como vulnerável no Rio Grande do Sul devido principalmente à perda e fragmentação de habitat e ao recente surto de febre amarela silvestre (2008/2009). As intensas atividades antrópicas no meio selvagem, além da perda de habitat, podem favorecer a disseminação de agentes patogênicos como bactérias e parasitos, que ocorrem em animais domésticos e em humanos e que podem acometer também os animais selvagens. O objetivo deste estudo foi verificar a ocorrência de bactérias da família Enterobacteriaceae e do protista Cryptosporidium sp. em fezes de bugios-ruivos de vida livre no município de Santa Maria, Rio Grande do Sul, Brasil, bem como relacionar a riqueza de enterobactérias nas amostras com fatores ecológicos e ambientais como tamanho do grupo, tamanho do fragmento florestal, densidade de populações de bugios, distância com núcleos humanos mais próximos, distância para cursos d água e sazonalidade. As amostras foram analisadas no Núcleo de Estudos e Pesquisas em Animais Silvestres (NEPAS/LCDPA) da Universidade Federal de Santa Maria. Foram detectadas vinte espécies de enterobactérias, e com as técnicas empregadas, as amostras foram negativas para Salmonella spp. e Cryptosporidium sp.. Nenhuma das variáveis ambientais analisadas teve influência sobre a riqueza das enterobactérias. Considerando que as populações de bugios do Campo de Instrução de Santa Maria (CISM) sofreram uma drástica redução devida à febre amarela, e que alguns organismos encontrados nesse estudo podem estar interagindo com outros fatores e assim afetar a dinâmica populacional dos bugios. Acredita-se que a continuidade do monitoramento da saúde destas populações seja fundamental para melhorar a compreensão dos mecanismos das doenças, assim como para a conservação dessa espécie.

Full textThis study investigated the occurrence of infections caused by Plasmodium spp. and Leishmania spp. in neotropical primates. Specimens were obtained both in captivity and in the wild in regions impacted by human activities in the Atlantic Forest and Amazon, Brazil. The pathogens investigated are responsible for significant zoonotic processes that affect humans, domestic and wild animals, with significant economic and environmental losses. The samples were collected and submitted for analysis by serological and molecular tests. The results suggest the transmission of Plasmodium spp. in primates from captive to those in the wild. In São Paulo state our data point for the presence of P. malariae and P. simium, this latter being found in Callicebus nigrifrons. In the Amazon, our data indicate the presence of P. brasilianum. Leishmania (L.) chagasi, L. (L.) amazonensis and the Leishmania Viannia complex circulates among the captive primates in São Paulo state. In the Amazon, 5.2% (1/19) of the primates sampled were detected by PCR as positive for L. (L.) chagasi. The results of this work may improve animal and public health approaches in relation to the neotropical primates in the regions studied.

Callicebus é um gênero de primata neotropical pertencente à família Pitheciidae, atualmente com 32 espécies reconhecidas. Estas espécies estão organizadas em grupos supraespecíficos, sendo dois subgêneros (Torquatus e Callicebus) e cinco grupos de espécies: C. torquatus, C. moloch, C. cupreus, C. donacophilus e C. personatus. A organização dos grupos foi realizada com base em dados morfológicos e de distribuição geográfica. Nesta dissertação de mestrado, fizemos inferências a partir de dados moleculares. No primerio capítulo, apresentamos uma introdução geral sobre a problemática taxonômica de Callicebus. No segundo capítulo realizamos inferências filogenéticas com base na presença e ausência de uma região molecular conhecida como elemento Alu, um transposon do genoma de primatas. Com base na análise desses marcadores Alu, descobrimos que os grupos C. moloch e C. cupreus são estreitamente relacionados e que C. torquatus é o grupo basal no gênero. No terceiro capítulo, a partir de uma abordagem multilocos investigamos as relações filogenéticas do grupo C. moloch e aplicamos o tempo de diversificação entre as espécies para testar a hipótese de formação das bacias hidrográficas da Amazônia durante o Plio-Pleistoceno. Nossos resultados corroboram a hipótese de formação dos rios amazônicos nos últimos 3 Ma. No entanto, nem todos os eventos de diversificão em Callicebus podem ser explicados pela teoria dos rios. Além disto, verificamos que os diferentes grupos de espécies de Callicebus são todos derivados de radiações na região Amazônica em diferentes momentos durante o Mioceno superior. Nossos resultados também sugerem que o conhecimento atual da diversidade de Callicebus está subestimado, e que espécies que diversificaram recentemente estão negligenciadas taxonomicamente.
Callicebus is a genus of Neotropical primate of the family Pitheciidae. Thirty-two species are currently recognized. They are classified in two subgenera, Torquatus and Callicebus, and five species groups: C. torquatus, C. moloch, C. cupreus, C. donacophilus and C. personatus. The arrangement of subgenera and groups is based on morphology and geographic distributions. The first section, we is introduction to the taxonomy of Callicebus. In the second section, I present an article for submission to the jounrla Primates concerning the chapter presence or absence of a molecular region known as an Alu element. An Alu element is a transposon in the genome of primates Because of this particular characteristic of Alu insertion, I discovered that the C. moloch and C. cupreus groups are closely related and that C. torquatus is the basal group in the genus. In the third section , from a multilocus approach I investigated the phylogenetic relationships of the C. moloch group and the time of diversification between the species, in order to test the hypothesis of formation of the Amazon River basin during the Plio-Pleistocene. Our results support the hypothesis of origin of Amazonian rivers in the last 3 Ma. However, not all diversification events in Callicebus can be explained by the theory of rivers. the results also indicate that the different species groups are derived from a radiation in the Amazonian region at different times during the upper Miocene. The phylogenetic analysis identified a number of potentioal taxa that a number of taxa which the current knowledge of species diversity in Callicebus is underestimated, and that recently diversified species are taxonomically neglected.

Full textPrimates exhibit delayed maturation in comparison to other mammals, with a long juvenile period. Two attempts to explain this characteristic are the needing to learn hypothesis, which points towards the need to acquire social or foraging skills as important challenges for juveniles, and the juvenile risk aversion hypothesis, according to which primates have slow growth rates in order to reduce metabolic rates, because they have a disadvantage in the competition for resources against adults, due to lower hierarchical position or to less foraging efficiency. The present work analyses the behavioral development of juvenile capuchin monkeys (Sapajus sp) from a semi-freeranging group at Parque Ecológico do Tietê (PET), São Paulo, Brazil, from October 2007 to May 2010. Data was obtained using scan sampling and all occurrences methods. Results show that the dominance hierarchy was mostly inherited from the mother, for juvenile females, and age-related, for males. The large number of individuals with little or no participation in agonistic interactions and of 0x0 ties, and the low number of coalitions, which involved mostly dominant individuals, suggest that a conflict-avoidance mechanism is being used by subordinates. There was a reduction in the aggression suffered by juveniles through time, suggesting that this mechanism is learned throughout the development. Affiliative relationships were highly dependent on kinship and hierarchical position, and in proximity there was also an effect of age, with juveniles close to one another. Juvenile females invested more in building and maintaining social networks, through grooming, than male juveniles, and were more tolerated in proximity. The activity budget and feeding behavior differed among age categories, but there was little influence of sex. There was evidence of synchrony of activities between juveniles and individuals in proximity, but differences in diet between juveniles and adults suggest that social learning was not the main influence on juveniles\' foraging behavior. They allocated more foraging time to resources easy to obtain (fruits) than to more difficult to obtain resources (invertebrates), which indicates they are less efficient in foraging. They also dedicated less time to provisioned food, suggesting a disadvantage in intra-group competition. The behavior of the juveniles observed in this study supports the risk aversion model, although there are evidences that the acquisition of social and foraging skills also plays an important role on their development

Full textTrack Analysis (Panbiogeography) focuses on the spatial or geographic dimension of biodiversity, allowing us to better understand the evolutionary patterns and processes. It compares tracks of different taxa to detect generalized tracks or biotic components. This study aimed to analyze the distribution of 24 neotropical primate species belonging to Cebus and Saimiri (Cebidae), Alouatta (Atelidae), and Aotus (Aotidae), to identify common distribution patterns and test the borders of the biogeographic units by the Track Analysis method. In a first step generalized tracks were obtained for each genera and in a second step, generalized tracks were obtained by matching individual species tracks. The crossing of individual genus tracks resulted in eleven generalized tracks located in the Caribenha, Amazon, Chaquenha, and Paranaense subregions. The Amazon subregion showed the higher concentration of species represented by eight generalized tracks, whereas the other subregions showed a single track each. Five tracks coincided with the Pleistocene refuge models (tracks a, c, g, h, and j), but only one track (c) can be explained by the marine transgression hypothesis. Three tracks were located in conservation units (c, e and j) and two (f and i) overlapped indicating the existence of a panbiogeographic node, that allows to propose the establishment of a conservation unit. The generalized tracks obtained by the analysis of species were spatially congruent with generalized tracks of neotropical species of Trichodactylidae (Crustacea, Decapoda), Muscidae (Diptera), Bombus (Hymenoptera, Apidae) and Cecropia (Cecropiaceae), indicating a higher distribution pattern. The generalized tracks are hypotheses of primary biogeographic homology for the Neotropical region.
A Análise de Traços (Pan-biogeografia) enfoca a dimensão espacial ou geográfica da biodiversidade, permitindo-nos uma melhor compreensão dos padrões e processos evolutivos. Ela compara traços individuais de diferentes táxons para detectar traços generalizados ou componentes bióticos. Este estudo objetivou analisar a distribuição de 24 espécies de primatas neotropicais, incluídas em Cebus e Saimiri pertencentes à Cebidae, Alouatta a Atelidae e Aotus a Aotidae, buscando identificar padrões comuns de distribuição, além de testar os limites das unidades biogeográficas, através do método da Análise de Traços. Em uma primeira etapa foram obtidos traços generalizados para cada gênero e, em segunda instância, obtiveram-se traços generalizados cruzando-se os traços individuais das espécies. A partir do cruzamento dos traços individuais de cada gênero, obtiveram-se onze traços generalizados localizados nas sub-regiões Caribenha, Amazônica, Chaquenha e Paranaense. A sub-região Amazônica mostrou a maior concentração de espécies, representada por oito traços generalizados, enquanto que as demais sub-regiões obtiveram cada uma apenas um traço. Cinco traços são coincidentes com o modelo de refúgios do Pleistoceno (traços a, c, g, h e j), porém, apenas um (traço c) pode ser explicado pela hipótese das transgressões marinhas. Três abrangem regiões onde se localizam unidades de conservação (traços c, e e j) e dois (f e i) encontram-se superpostos, indicando a ocorrência de um nó pan-biogeográfico, o que permite propor a criação de uma unidade de conservação na área. Os traços generalizados obtidos na análise das espécies são espacialmente congruentes com traços generalizados de espécies neotropicais de Trichodactylidae (Crustacea, Decapoda), Muscidae (Diptera), Bombus (Hymenoptera, Apidae) e Cecropia (Cecropiaceae), o que aponta para um padrão maior de distribuição. Os traços generalizados são hipóteses de homologia biogeográfica primária para a região Neotropical.

Full textThe neotropical primates, also known as primates of the new world, are a diverse and varied group of animals, with unique anatomical and taxonomic characteristics, as well a unique behaviour, that distinguish those from the old world primates. It is estimated that these present 31% of the non-human primate population of the planet. High taxes of populational growth and anthropological disturbance have been undergoing due to deforestation, agriculture and cattle farms. In order to revert this situation and maximize the reproductive potential of the species, the development of biotechnologies associated to the reproduction are widely justified nowadays, for representing important reproductive tools, contributing to the preservation of the biodiversity and maintenance of genetic diversity. Hence, the purpose of this study was to collect Alouatta guariba clamitans semen by rectal electroejaculation then analyze the fresh semen, procedures never before described for the species, and adapt the preparation method of Leontopithecus chrysomelas, Alouatta caraya and Alouatta guariba clamitans semen for electron transmission microscope, allowing the comparison of the spermatozoa ultrastructure from the three species and also presenting new results to the scientific literature. For this study, it was used four specimens of L. chrysomelas, two specimens of A. caraya and two specimens of A.g. clamitans, adult males kept in captivity. L. chrysomelas had the semen taken by penine vibratory stimulation, and A. caraya and A.g. clamitans were submitted to rectal electroejaculation. After the semen was obtained, it was evaluated regarding the volume, pH, concentration, motility, sperm membrane integrity, acrosome integrity and morphology, and then prepared for electron transmission microscopy. The analysis of the spermatozoa of the three studied species revealed that all present the basic components described for the human spermatozoa - head, neck region and tail, formed by middle, principal and terminal pieces - with variations in 14 shape and size of each component. The most significant differences found between the compared spermatozoa were the acrosome extension and middle piece length, thickness and organization. Ela compara tra?os individuais de diferentes t?xons para detectar tra?os generalizados ou componentes bi?ticos. Este estudo objetivou analisar a distribui??o de 24 esp?cies de primatas neotropicais, inclu?das em Cebus e Saimiri pertencentes ? Cebidae, Alouatta a Atelidae e Aotus a Aotidae, buscando identificar padr?es comuns de distribui??o, al?m de testar os limites das unidades biogeogr?ficas, atrav?s do m?todo da An?lise de Tra?os. Em uma primeira etapa foram obtidos tra?os generalizados para cada g?nero e, em segunda inst?ncia, obtiveram-se tra?os generalizados cruzando-se os tra?os individuais das esp?cies. Full textViruses belonging to the Herpesviridae family (order Herpesvirales) are enveloped double-stranded DNA viruses distributed into three subfamilies: Alpha-, Beta- and Gamma-herpesvirinae. These viruses have been identified from a wide range of host species, ranging from mammals to reptiles to birds. They have the ability to persist throughout the life of the host in a latent form and to reactivate. Most of these viruses are specific to a host. The wide distribution of herpesviruses, generally associated with an asymptomatic infection in their natural host, suggests that these viruses have co-evolved with their hosts.The first part of this work was dedicated to the identification of cytomegaloviruses (CMV, genus Cytomegalovirus) in New World non-human primates (NWNHP) to see if the diversification of these viruses followed that of their hosts. A previous study on Old World primates had demonstrated that CMVs infecting them showed a strong signal of co-divergence with their hosts. Nevertheless, among the different species of primates tested, only a few were from the New World. To test this hypothesis, we performed a molecular screening of 244 blood DNA samples from 20 Central and South American species. Through a PCR approach using degenerate consensus primers targeting highly conserved motifs of the DNA polymerase gene of herpesviruses, we characterized new viral sequences from 12 species belonging to seven representative genera of the three families of NWNHP. These results demonstrate that most species of NWNHP can be infected with a virus belonging to the Cytomegalovirus genus. In addition, phylogenetic analyzes of the obtained sequences combined with their molecular dating support a co-evolutionary scenario. This study led us to propose that CMVs sequences of NWNHP could serve as a molecular marker with which to infer the not yet fully resolved systematics of these primates.The second part of this work was on identifying herpesviruses from New World bats to study their distribution and diversity. The search for herpesviruses in bats is more recent. In the early 2000s it benefited from studies dedicated to other viral families given their role as reservoirs of potentially zoonotic viruses. The first description of bat herpesvirus sequences dated back from 2007. Over the past decade, dozens of herpesvirus sequences have been described from different bat species on every continent. Nevertheless, the distribution of the tested species was geographically uneven and only a few were from the New World. Molecular screening of 195 blood DNA samples from 11 species belonging to three families (Phyllostomidae, Mormoopidae and Molossidae) was performed. Using the same approach as applied to NWNHP, we obtained viral sequences (DNA polymerase and/or glycoprotein B) from all tested species. These are distributed within the Beta- and Gamma-herpesvirinae subfamilies. Fourteen partial sequences of the DNA polymerase gene, corresponding to three beta- and eleven gamma-herpesviruses, have been identified. Twelve partial sequences of the glycoprotein B gene, all gamma-herpesviruses, have been characterized. Each sequence was specific to a bat species and in some species multiple viruses were identified. Phylogenetic analyzes of these sequences have identified clades specific to bat viruses. Those composed of sequences obtained from different species belonging to distinct subfamilies follow the taxonomy of bats. This study confirms the astonishing diversity of bat herpesviruses and broadens our knowledge on their host spectrum.This work is the largest conducted to date in terms of species diversity of non-human primates and bats from the Neotropical realm. Nevertheless, the samples tested represent only a tiny part of this diversity. Further analyzes, on a broader panel of representative species from other geographic areas will increase our understanding of the evolutionary history of these viruses

Estudos parasitológicos em populações naturais de primatas neotropicais são relativamente raros, existindo poucos dados disponíveis sobre o guariba-de-mão-ruiva, Alouatta belzebul. No presente estudo, populações de A. belzebul foram amostradas em cinco locais na área do reservatório da Usina Hidrelétrica de Tucuruí no sudeste da Amazônia Brasileira, correspondendo à margem direita do Rio Tocantins. As áreas de coleta incluíram a floresta contínua e fragmentos de hábitats em ilhas, com tamanhos que variaram de 180 a 484 hectares. O principal objetivo deste estudo foi a avaliação dos efeitos da perturbação do hábitat sobre os padrões de infestação por endoparasitas. A densidade populacional foi estimada para cada ponto de coleta usando o método de transecção linear, que variou de 100-108 km percorridos por ponto. Amostras fecais foram coletadas de seis a quatorze grupos em cada local, com um total de 40-46 amostras por ponto (n = 212). As amostras fecais foram fixadas em MIF e observadas através de microscópio óptico, com aumentos de até 400x. A densidade populacional variou entre 66,4 e 191,5 indivíduos por km2. No total, 76,4% das amostras foram positivas para pelo menos uma espécie de endoparasita. Foram identificadas doze táxons de endoparasitas, oito de helmintos e cinco de protozoários. Amostras individuais apresentaram até cinco diferentes espécies de endoparasitas. Em cada local de coleta, o número de espécies identificadas variou entre seis e doze e as taxas de infecção ficaram entre 67,5% e 86%. Não foram encontrados padrões sistemáticos na diversidade de parasitas ou nas taxas de infecção em relação a variáveis como, tamanho de população, densidade ou fragmentação de hábitat. A diversidade e as taxas de infecção variaram mais entre os dois pontos de floresta contínua que nos locais fragmentados e, no geral, foram menores nos locais com menor densidade populacional. A única exceção foi o Trypanoxyuris mirtutus, um oxiurídeo bastante comum transmitido através do contato direto, para o qual foi encontrada uma correlação forte entre as taxas de infecção e a densidade populacional. No geral, foram encontradas poucas evidências capazes de sustentar a hipótese de que a fragmentação do hábitat tem um efeito sistemático nos padrões de infestação em A. belzebul. Contudo, recomenda-se a realização de mais estudos detalhados antes de se estabelecer conclusões definitivas.
Parasitological studies of natural populations of neotropical primates are relatively rare, and few data were available previously for the red-handed howler monkey, Alouatta belzebul. In the present study, populations of A. belzebul were sampled at five sites in the area of the reservoir of the Tucuruí hydro-electric dam in southeastem Brazilian Amazonia, corresponding to the right bank of the Rio Tocantins. Sites included continuous forest and fragments of habitat on islands varying in area from 180 to 484 hectares. A primary aim of the study was the evaluation of the effects of habitat perturbation on patterns of infestation for endoparasites. Population density was estimated at each site using standard line transect surveys of 100-108 km per site. Fecal samples were collected from six to fourteen groups at each site, with a total of 40- 46 individual samples per site (n = 212). Samples were fixed in MIF (Mercury Chromed, Iodine and Formaldehyde), and observed under a 400x optic microscope. Population density varied between 66,4 and 191,5 individuals per square kilometer. Overall, 76,4% of the samples were positive for at least one species of endoparasite, and a total of thirteen species were identified, including eight species of heiminthes and five protozoans. Individual samples contained as many as five different species of endoparasites. At a given site, the number of species identified varied between seven and twelve, and infection rates between 67,5% and 86%. No systematic pattern of parasite diversity or infection rates was found in relation to variables such as population size or density, or habitat fragmentation. Diversity and infection rates varied as much between the two continuous forest sites as among the forest fragments, and were lowest overall at the site with the lowest population density. The only exception was Trypanoxyuris minutus, a common oxyurid transmitted through direct contact, for which a strong correlation was found between infection rates and population density. Overall, little evidence was found to support the hypothesis that habitat fragmentation has a systematic effect on infestation patterns in A. belzebul, although more detailed studies will be recommend before more definitive conclusions can be drawn.

Acredita-se que, na Região Amazônica ocorra a transmissão de malária de primatas para humanos, devido a relatos atuais de resultados soroepidemiológicos de humanos e primatas. O presente estudo objetivou realizar o diagnóstico morfológico, sorológico e molecular de Plasmodium spp. em primatas neotropicais na Ilha de São Luís, Estado do Maranhão, Brasil. Para tal, foram utilizadas como técnicas de diagnóstico: a microscopia de luz, a reação em cadeia pela polimerase (PCR) e a imunofluorescência indireta (RIFI). No período de junho de 2009 a abril de 2010, foram amostrados 70 primatas, sendo 50 provenientes do Centro de Triagem Animais Silvestres (CETAS), localizado no município de São Luís, e 20 de vida livre, capturados em reserva particular localizada no município de São José de Ribamar, Estado do Maranhão. Foram avaliadas pela microscopia de luz 140 lâminas (duas de cada animal), das quais cinco (7,1%) foram positivas. Pela RIFI não se detectou anticorpos anti-Plasmodium spp. Pela PCR, dos 70 animais amostrados, foi possível observar produtos amplificados para Plasmodium spp. em 13 amostras, das quais oito (61,54%) eram de animais de vida livre e cinco (38,46%) de CETAS. O presente trabalho teve como objetivo o estudo taxonômico de nematóides parasitos de três espécies de primatas Neotropicais, Callithrix penicillata (Geoffroy, 1812), Sapajus apella (Linnaeus, 1758) e Allouatta guariba (Humboldt, 1812), doados mortos à Universidade Federal de Juiz de Fora, pelo IBAMA/JF. Foram necropsiados 26 espécimes de primatas, sendo 22 C. penicillata, 2 S. apella e 2 A. guariba. Os helmintos foram coletados, quantificados e tratados de acordo com técnicas helmintológicas usuais. Para o estudo taxonômico, foram feitas imagens e desenhos, além da morfometria dos mesmos espécimes. Foram feitas análises de distribuição, intensidade, densidade e abundância das comunidades parasitárias. Foram coletados 10416 nematóides, pertencendo a quatro diferentes gêneros, com uma diferença significativa na quantidade de helmintos de C. penicillata (1391), S. apella (4590) e A. guariba (4435). Foram registrados Primasubulura jacchi em C. penicillata, Trypanoxyuris (Trypanoxyuris) minutus em A. guariba e S. apella, Parabronema (=Squamanema) bonnei em A. guariba, além do registro de uma nova espécie de Onchocercinae nos pulmões de um espécime de A. guariba, Courduriella sp..O registro de T. (T.) minutus em S. apella representa o primeiro relato deste parasito em hospedeiro deste gênero na América do Sul e com a coleta de Parabronema bonnei, foi possível fazer uma redescrição da espécie que não possui detalhamento de características morfológicas e morfométrica disponíveis na literatura.
The present work aimed to the taxonomic study of nematode parasites of three species of Neotropical primates, donated, already dead, the Federal University of Juiz de Fora, from IBAMA/JF. They are Callithrix penicillata (Geoffroy, 1812), Sapajus apella (Linnaeus, 1758) and Allouatta guariba (Humboldt, 1812). Twenty-six specimens of primates were necropsied, being twenty-two of C. penicillata, two S. apella and two A. guariba. All colected helminths were quantified and treated in accordance with usual helminthological techniques. For the taxonomic study, pictures and drawings were made, for morphometry of the same specimens. Distribution analysis, intensity, density and abundance of parasite communities were made. 10416 nematodes were collected belonging to four different genera, with a significant difference in the number of nematodes in C. penicillata (1391) and S. apella and A. guariba monkey (9025). Primasubulura jacchi were recorded in C. penicillata, Trypanoxyuris (Trypanoxyuris) minutus in A. guariba and S. apella e Parabronema (= Squamanema) bonnei in A. guariba, other than registration of a new species of Onchocercinae, lungs of a specimen A.guariba monkey, named Courduriella sp. The record of T. (T.) minutus in S. apella is the first report of this parasite in this host genus in South America and the collecting Parabronema bonnei was possible to make a redescription of the species that lacks detailed morphological and morphometric characteristics available in the literature.

O encéfalo humano é mais sofisticado instrumento de cognição e, supostamente, o material mais complexo já conhecido, provavelmente por causa disso, o sistema neural é amplamente estudado em termos multidisciplinares, mas muitas questões ainda necessitam ser respondidas e várias dúvidas sobre as funções cerebrais ainda precisam de respostas. Um dos caminhos para compreender essa história evolutiva é o estudo do encéfalo de primatas não-humanos. Muito se sabe sobre a anatomia macroscópica de primatas do Velho Mundo e humanos, entretanto, esses dados e aqueles relacionados à anatomia microscópica são escassos em relação aos neotropicais, que tem sido amplamente utilizado em pesquisas biomédicas e comportamentais. Dessa forma, a proposta desse trabalho foi estudar a anatomia macroscópica do encéfalo obtido diretamente dos primatas neotropicais como Callithrix penicillata, Saimiri ustus, Sapajus libidinosus e Brachyteles arachnoides e da literatura de Galago, Macaca, Papio, Pan and Homo, em termos comparativos. Além de realizar um estudo preliminar acerca da citoarquitetura neocortical de Sapajus libidinosus. Na análise macroscópicas foram utilizados 2 encéfalos de Callithrix penicillata, 1 de Saimiri ustus, 7 de Sapajus libidinosus e 1 Brachyteles arachnoides, que tiverem seus sulcos e giros descritos, mensurados, além de mensurações das dimensões gerais do encéfalo e índice de encefalização. Para a análise microscópica foram utilizados 5 encéfalos de Sapajus libidinosus, sendo 4 na técnica de Golgi-Cox e 1 na técnica HE, que foram avaliados em termos de organização citoarquitetônica, número de células neurais e classificação do córtex conforme o tipo e prevalência celular nas camadas corticais, dos lobos frontal, área pré-frontal, parietal, temporal e occipital. Nesse trabalho observou-se que anatomia do telencéfalo deCallithrix e Saimiripossuem poucos sulcos, giros quando comparados com o deSapajuseBrachytelese os dois últimos são mais semelhantes aos primatas do Velho Mundo, do que dos demais primatas neotropicais investigados. Dentre todos os primatas analisados e comparados, os sulcos que se repetem são o longitudinal, lateral, calcarino, hipocampo, rinal e o temporal superior. Os sulcos do lobo frontal e parietal, aparecem somente em encéfalos mais complexos e de animais que possuem maior tamanho corporal. As demais estruturas localizadas na região medial do telencéfalo, diencéfalo, mesencéfalo, ponte, bulbo e ventrículos encefálicos, estão dispostas de forma semelhante com variações no grau de desenvolvimento e tamanho. Os dados preliminares da arquitetura neocortical de Sapajus mostram maior quantidade de neurônios nos lobos occipital, seguidos do parietal e temporal. A parte caudal do lobo frontal possui carcterísticas de área motora primária com grandes neurônios piramidais e a área préfrontal possui prevalência de neurônios granulares. Os lobos parietal e temporal possuem as camadas mais heterogêneas e com maior separação entre as camadas corticais. A técnica de Golgi-Cox permitiu estudar a organização estrutural, conexões entre células neurais e a formas das células neurais. Já a técnica de HE permitiu inferências quantitativas e também a caracterização dos tipos de células neurais e desenvolvimento das camadas corticais. As análises histológicas do neocórtex das principais áreas do telencéfalo de Sapajuslibidinosus, mesmo que com um baixo número de espécimes foi uma caracterização geral e o primeiro passo para estudos futuros e comparações mais abrangentes. São necessários estudos histológicos mais detalhados das principais áreas do encéfalo, assim como a associação dos possíveis resultados a outras técnicas de investigações são necessários para uma melhor caracterização das funcionalidades das áreas corticais.
The human brain is the most sophisticated instrument of cognition and, putatively, the most complex material sctructure ever knew, probably because that, the neural system is largely studied in multidisciplinar terms, but many questions still need to be answered and various doubts rest to be solved about the brain functions. Among the ways to understand this evolutionary history is the study of the non-human primate encephalon. Much is known about the gross anatomy of Old World Primates and humans, however, these data and those related to the microscopic anatomy are scarce compared to the neotropical, which has been widely used in biomedical and behavioral research. In order, the purpose of this work was to study the gross anatomy of the brain obtained from neotropical primates as Callithrix penicillata, Saimiri ustus, Sapajus libidinosus e Brachyteles arachnoides and from literature from Galago, Macaca, Papio, Pan and Homo, in comparative terms. In addition to conducting a preliminary study on the neocortical cytoarchitecture of Sapajuslibidinosus. In gross anatomy were used 2 brains of Callithrix penicillata, 1 of Saimiriusustus, 7 of Sapajuslibidinosus and 1 Brachytelesarachnoides, which have their sulcus and gyri described, measured, as well as measurements of the general dimensions of the encephalon and encephalization index. For the microscopic analysis, 5 brains of Sapajuslibidinosus were used, 4 in the Golgi-Cox method and 1 in the HE method, which were evaluated in terms of cytoarchitectonic organization, number of neural cells and classification of the cortex according to type and cell prevalence in cortical layers, frontal, prefrontal area, parietal, temporal and occipital areas. In this study, it was observed that the gross anatomy telencephalon of Callithrix and Saimiri have few sulcus and gyri when compared to Sapajus and Brachyteles, and the latter two are more similar to the Old-World primates than of the other neotropical primates investigated. Among all the primates analyzed and compared, the repeats sulcus are the longitudinal, lateral, calcarino, hippocampus, rinal and superior temporal sulci. The sulci of the frontal lobe and parietal lobe appear only in more complex brains and animals that have larger body size. The other structures located in the medial region of the telencephalon, diencephalon, midbrain, pons, bulb and encephalic ventricles are similarly arranged with variations in the degree of development and size. Preliminary data from Sapajus neocortical architecture show higher numbers of neurons in the occipital lobes, followed by the parietal and temporal lobes. The caudal part of the frontal lobe has features of primary motor area with large pyramidal neurons and the prefrontal area has a prevalence of granular neurons. The parietal and temporal lobes have the most heterogeneous layers and with greater separation between the cortical layers. The Golgi-Cox method allowed the study of the structural organization, connections between neural cells and neural cell forms, and the HE method allowed quantitative inferences and also the characterization of neural cell types and development of the cortical layers.The histological analyzes of the neocortex of the main areas of the Sapajus libidinosus telencephalon, even with a low number of specimens, was a general characterization and the first step for future studies and more comprehensive comparisons. More detailed histological studies of the main areas are necessary as well the association of these possibles results with other investigative techniques for a better characterization of the functionalities of the cortical areas.

Modern zoological gardens are institutions to wild fauna maintenance, aiming its conservation, the performance of scientific research and leisure, recreation and environmental education activities. The variety of wild species living in conditions different from the one found in its natural habitats represents a propitious environment for diseases spread, specially the zoonotic ones. Due to data shortage and wild mammals’ epidemiological relevance both for toxoplasmosis as for leptospirosis, this study aimed to determine the seroprevalence of toxoplasmosis and leptospirosis in neotropical wild mammals, from “Zoológico do Parque da Cidade Governador José Rollemberg Leite”, Aracaju, Sergipe, Brazil. Blood samples were collected from 32 wild mammals, adults and from both sexes: 14 tufted capuchins (Cebus apella), four golden-bellied capuchins (Cebus xanthosternus), three pumas (Puma concolor), one jaguar (Panthera onca), one fox (Pseudolopex vetulus), six crab-eating raccoons (Procyon cancrivorus), two South American coatis (Nasua nasua), and one tayra (Eira Barbara). Sera were tested by the modified agglutination test (MAT) using formalin-fixed whole tachyzoites and mercaptoetenhol (cut-off point 1:25) to Toxoplasma gondii antibodies, and by the microscopic seroagglutination test (cut-off point: 1:100) using 24 serovar of,pathogenic leptospiras and 2 serovar of saprophyte leptospiras. Antibodies to T. gondii were found in 17 of 32 wild mammals (53.1%), and antibodies to Leptospiras spp were found in 4 of 32 wild mammals (12.5%). In relation to gender, 9 of 15 (60%) males, and 8 of 17 (47.1%)females had antibodies to T. gondii, and 4 of 15 (26.7%) males had antibodies to Leptospira spp. Considering the origin of wild seropositive mammals to T. gondii, 8 of 17 (47%) was born in zoo, 7 of 17 (41.2%) were from other zoos, and 2 (11.8%) was from the wild, and 3 of4 (75%) seropositive to Leptospiras spp was born in zoo, and 1 of 4 (25%) was from the wild. This is the first serological survey of Leptospiras spp in neotropical primates and carnivores from Northeast zoo from Brazil, and the first found to antibodies to Leptospira spp (serovar Copenhageni) in the threaten specie of primate golden-bellied capuchins (Cebus xanthosternus).
Os zoológicos modernos são instituições destinadas à manutenção da fauna selvagem com objetivo de conservação, pesquisa cientifica, lazer, recreação e educação ambiental. A ampla variedade de espécies selvagens, vivendo em condições diferentes do seu habitat natural, representa um ambiente propício à disseminação de doenças, muitas delas zoonóticas. Devido à escassez de dados e à relevância dos mamíferos selvagens no contexto epidemiológico, tanto na toxoplasmose, quanto na leptospirose, objetivou–se determinar a soroprevalência de toxoplasmose e de leptospirose em mamíferos selvagens neotropicais do Zoológico do Parque da Cidade Governador José Rollemberg Leite, Aracaju, Sergipe. Para tanto foram colhidas amostras sanguíneas de 32 mamíferos selvagens, adultos e de ambos os sexos, das espécies: 14 macacos-prego (Cebus apella), quatro macacos-prego-do-peito-amarelo (Cebus xanthosternus), três onças-suçuaranas (Puma concolor), uma onça-pintada (Panthera onca), uma raposa-do-campo (Pseudolopex vetulus), seis guaxinins (Procyon cancrivorus), dois quatis (Nasua nasua) e um papa-mel (Eira barbara). Para pesquisa de anticorpos anti- Toxoplasma gondii foi utilizado o teste de Aglutinação Modificada com taquizoítos inativados na formalina e 2-mercaptoetanol (ponto de corte na diluição 1:25) e para pesquisa de anticorpos anti-Leptospira spp foi utilizado o teste de Soroaglutinação Microscópica (ponto de corte na diluição 1:100) com uma coleção de antígenos vivos que incluiu 24 variantes sorológicas de leptospiras patogênicas e duas leptospiras saprófitas. Dentre os 32 mamíferos, 17 (53,1%) apresentaram anticorpos anti-T. gondii e quatro (12,5%) foram positivos para anticorpos anti-Leptospira spp. De acordo com o sexo, 60% (9/15) dos machos e 47,1% (8/17) das fêmeas foram soropositivos para T. gondii e 26,7% (4/15) dos machosapresentaram anticorpos anti-Leptospira spp. Com relação à procedência, dos mamíferos que apresentaram anticorpos anti-T. gondii, 47% (8/17) nasceram no zoológico, 41,2% (7/17) foram oriundos de outras instituições e dois (11,8%) foram provenientes da natureza. Em relação aos quatro mamíferos soropositivos para Leptospira spp, três (75%) foram procedentes da natureza e um (25%) nasceu no zoológico. Este trabalho representou o primeiro inquérito sorológico para a ocorrência de anticorpos anti-Leptospira spp em primatas e carnívoros neotropicais em um zoológico do Nordeste do Brasil e descreveu pela primeira vez a ocorrência de anticorpos anti-T. gondii e anti-Leptospira spp com sorovar mais provável Copenhageni no primata ameaçado de extinção macaco-prego-de-peito-amarelo (C. xanthosternus).

One of the main environmental cues for the adjustment of temporal organization of the animals is the light-dark cycle (LD), which undergoes changes in phase duration throughout the seasons. Photoperiod signaling by melatonin in mammals allows behavioral changes along the year, as in the activity-rest cycle, in mood states and in cognitive performance. The aim of this study was to investigate if common marmoset (Callithrix jacchus) exhibits behavioral changes under short and long photoperiods in a 24h cycle, assessing their individual behaviors, vocal repertoire, exploratory activity (EA), recognition memory (RM) and the circadian rhythm of locomotor activity (CRA). Eight adult marmosets were exposed to a light-dark cycle of 12:12; LD 08:16; LD 12:12 and LD 16:08, sequentially, for four weeks in each condition. Locomotor activity was recorded 24h/day by passive infrared motion detectors above the individual cages. A video camera system was programmed to record each animal, twice a week, on the first two light hours. From the videos, frequency of behaviors was registered as anxiety-like, grooming, alert, hanging position, staying in nest box and feeding using continuous focal animal sampling method. Simultaneously, the calls emitted in the experimental room were recorded by a single microphone centrally located and categorized as affiliative (whirr, chirp), contact (phee), long distance (loud shrill), agonistic (twitter) and alarm (tsik, seep, see). EA was assessed on the third hour after lights onset on the last week of each condition. In a first session, marmosets were exposed to one unfamiliar object during 15 min and 24h later, on the second session, a novel object was added to evaluate RM. Results showed that long days caused a decreased of amplitude and period variance of the CRA, but not short days. Short days decreased the total daily activity and active phase duration. On long days, active phase duration increased due to an advance of activity onset in relation to symmetric days. However, not all subjects started the activity earlier on long days. The activity offset was similar to symmetric days for the majority of marmosets. Results of EA showed that RM was not affected by short or long days, and that the marmosets exhibited a decreased in duration of EA on long days. Frequency and type of calls and frequency of anxiety-like behaviors, staying in nest box and grooming were lower on the first two light hours on long days. Considering the whole active phase of marmosets as we elucidate the results of vocalizations and behaviors, it is possible that these changes in the first two light hours are due to the shifting of temporal distribution of marmoset activities, since some animals did not advance the activity onset on long days. Consequently, the marmosets mean decreased because the sampling was not possible. In conclusion, marmosets synchronized the CRA to the tested photoperiods and as the phase angle varied a lot among marmosets it is suggested that they can use different strategies. Also, long days had an effect on activity-rest cycle and exploratory behaviors
Uma das principais pistas ambientais para o ajuste da organiza??o temporal dos animais ? o ciclo claro-escuro (CE), que sofre altera??o na dura??o das fases no decorrer das esta??es do ano. A sinaliza??o do fotoper?odo pelo horm?nio melatonina possibilita que os mam?feros se comportem de forma diferenciada ao longo do ano, como no ritmo de atividade-repouso, nos estados de humor e no desempenho cognitivo. Este estudo teve como objetivo avaliar se o sagui (Callithrix jacchus) exibe altera??es comportamentais em fotofases de diferentes dura??es em ciclo CE de 24h por meio da avalia??o dos repert?rios comportamental e vocal, da atividade explorat?ria (AE), da mem?ria de reconhecimento (MR) e do ritmo circadiano de atividade locomotora (RCA) em ambiente laboratorial. Oito saguis adultos, alojados individualmente, passaram 28 dias em cada etapa: dias sim?tricos (DS1 - CE 12h:12h), dias curtos (DC - CE 8h:16h), DS2 (CE 12h:12h) e dias longos (DL - CE 16h:8h). A atividade locomotora foi registrada continuamente por sensores de movimento acima das gaiolas. Um sistema de c?meras registrou os comportamentos individuais por 2 horas ap?s o in?cio do claro, duas vezes por semana. A partir das filmagens, foi amostrada a frequ?ncia dos comportamentos ansiosos, cata??o, alerta, pendurado, ficar na caixa ninho e comer pelo m?todo Animal Focal Cont?nuo. Ao mesmo tempo, um microfone registrou as vocaliza??es emitidas na sala de experimenta??o, categorizadas em afiliativa (whirr, chirp), contato (phee), longa dist?ncia (loud shrill), agon?stica (twitter) e alarme (tsik, seep, see). A AE foi avaliada na terceira hora ap?s o in?cio do claro em 2 sess?es de 15 min na ?ltima semana de cada ciclo. Na primeira apresenta??o, foi utilizado um objeto n?o familiar e 24 h depois, na segunda apresenta??o, foi adicionado um novo objeto para avaliar a MR. Os resultados mostraram que os DL diminu?ram a amplitude e a vari?ncia do per?odo do RCA, mas n?o os DC. Os DC causaram uma diminui??o do total di?rio de atividade e da dura??o da fase ativa. Nos DL, a dura??o da fase ativa aumentou devido a um avan?o no in?cio da atividade em rela??o aos dias sim?tricos. Contudo, nem todos os sujeitos iniciaram a atividade mais cedo nos dias longos. O hor?rio de t?rmino da atividade nos DL foi similar aos DS para a maioria dos sujeitos. Os resultados da AE mostrou que os DL e DC n?o afetaram a MR dos saguis e que os DL reduziram a dura??o da AE. Tamb?m foi observado nos DL uma redu??o da frequ?ncia e da diversidade das vocaliza??es, bem como da frequ?ncia dos comportamentos ansiosos, ficar na caixa ninho e cata??o nas duas horas ap?s in?cio do claro. Considerando toda a fase ativa na interpreta??o dos resultados dos comportamentos e vocaliza??es, ? poss?vel que essas altera??es nas duas horas iniciais tenham sido pelo deslocamento da distribui??o temporal da atividade do sagui e n?o um efeito genu?no dos DL, pois alguns animais n?o avan?aram o in?cio da atividade nos DL, causando a redu??o na m?dia do grupo por impossibilitar a amostragem. Em conclus?o, os saguis sincronizaram o RCA aos fotoper?odos e, pela varia??o individual nas rela??es de fase, parecem fazer uso de diferentes estrat?gias. Faz um levantamento sorológico para anticorpos contra Leptospira spp e Trypanosoma cruzi em primatas neotropicais mantidos em cativeiro. Amostras de 94 primatas neotropicais adultos, machos e fêmeas de diferentes espécies pertencentes ao criatório do Centro Nacional de Primatas (CENP)-Ananindeua-PA, coletadas para a realização da Soroaglutinação Microscópica (SAM), na qual foram utilizadas 84 amostras sorológicas, em que 35 (41,67%) apresentaram anticorpos contra leptospira e 49 (58,33%) foram soronegativas. De 11 espécies utilizadas na pesquisa, as maiores positividades estavam nas espécies de Cebus apella 69.23% (9/13), Aotus infullatus 33,33% (5/15), Callithrix penicillata 28,57% (4/14) e Saimiri sciureus 22,73 (5/22). De 35 amostras positivas, 11 (31,42%) reagiram contra o sorovar Cynopteri, oito (22,85%) foram reagentes para Andamana, seis (17,14%) contra o sorovar Hebdomadis, quatro (11,42%) para Copenhageni, três (8,57%) contra o sorovar Patoc, duas (5,71%) para o sorovar Cuíca, e o restante reagiram para pelo menos um sorovar (2,85%) sorovar Hardjo, Icterohaemorrhagiae, Javanica, Grippotyphosa e Autumnalis. Para detecção de anticorpos contra o T. cruzi, foram utilizadas 94 amostras sorológicas de primatas neotropicais. As amostras de cada animal foram submetidas aos exames sorológicos de Hemaglutinação Indireta (HAI) e Ensaio Imunoenzimático (ELISA). Das amostras avaliadas pela HAI, apenas uma fêmea da espécie Saguinus niger revelou resultado positivo, porém todas as amostras revelaram resultado negativo quando submetidas ao ELISA-recombinante. Com relação aos parâmetros hematológicos e bioquímicos não foram observadas alterações que indicassem uma possível infecção. Conclui-se que a ocorrência de anticorpos contra as leptospiras nas espécies de primatas foi alta, mesmo não apresentando sintomas, e todos os parâmetros hematológicos e bioquímicos estarem normais indicando que apesar destes animais encontrarem-se em cativeiro, possivelmente tiveram contato em vida livre com a bactéria e a infecção pode estar sendo mantida entre eles e casos de leptospirose e doença de Chagas em primatas neotropicais são raros, porém deve-se lembrar que os mesmos atuam como reservatórios de Leptospira spp e Tripanosma cruzi no ambiente silvestre.
Make a serological survey for antibodies against Leptospira spp and Trypanosome cruzi in Neotropical primates kept in captivity. 94 samples of neotropical primates adults, males and females of different species kept in captivity in the Primates National Center - Ananindeua- PA, collected for the accomplishment of the Microscopic Agglutination Test (MAT), in the which was used 84 serum samples, of which 35 (41.67%) presented antibodies against leptospira and 49 (58.33%) were seronegative. From 11 species used in research, the highest positivity was in the species of Cebus apella (69.23%), Aotus infullatus (33.33%), Callithrix penicillata (28.57%) and Saimiri sciureus (22.73%). From 35 positive samples, 11 (31.42%) reacted against serovar Cynopteri, eight (22.85%) were reagents for Andaman, six (17.14%) against serovar Hebdomadis, four (11.42%) for Copenhagen, three (8.57%) against serovar Patoc, two (5.71%) to serovar Cuíca, and the remainder responded to at least one serovar (2.85%) serovar Hardjo, Icterohaemorrhagiae, Javanica, Grippotyphosa and Autumnalis. To detect the anticorpos against T. cruzi, were used 94 samples of neotropical primates. The samples of each animal were submitted to the Indirect Hemagglutination (IHA) and Enzyme- Linked Immunosorbent Assay (ELISA). Of samples avaluated by IHA, only one female from the specie Saguinus niger showed result positive, but, all samples showed result negative when avaluated by ELISA. In relation to hematological and biochemical parametes, were not observed alterations that indicated a possible infection. It is concluded that the occurrence of antibodies against leptospires in species of primates was relatively high, even showing symptoms, and all hematological and biochemical parameters were normal indicating that although these animals find themselves in captivity, possibly had contact in the wild life with the bacterium, and the infection may be maintained between them and cases of leptospirosis e Chaga’s disease in neotropical primates are rares, but we should remember that they act with reservoir of Leptospira spp and Tripanosma cruzi at wild environment.

O conhecimento atualizado da distribuição e infecção natural de triatomíneos sinantrópicos é fundamental para vigilância e controle da doença de Chagas. No Brasil, a detecção de Trypanosoma cruzi em triatomíneos é realizada por microscopia óptica (MO) do conteúdo intestinal, que pode apresentar diversas limitações quanto à sua sensibilidade e especificidade. Objetivos: Analisar a ocorrência de triatomíneos e suas taxas de infecção natural por T. cruzi e outros tripanossomatídeos no estado de Goiás (GO) e Distrito Federal (DF), estimar a sensibilidade da MO para detecção de T. cruzi em triatomíneos e analisar a transmissão de T. cruzi em ambientes onde colônias de triatomíneos infectados forem detectadas. Métodos: Os espécimes (adultos e ninfas) capturados no intra e peridomicílio e resultados dos exames parasitológicos a fresco e lâminas coradas foram obtidos por meio do LACEN/SES/GO e DIVAL/DF. O DNA foi extraído de amostras do intestino para realização das PCRs (cPCR e qPCR) utilizando quatro marcadores moleculares (TCZ, kDNA, rDNA 18S e rDNA24sα). Também, foi avaliada a infecção por T. cruzi em colônia de P. megistus (nestedcPCR) e em amostras sanguíneas provenientes de primatas neotropicais não-humanos (nestedqPCR) encontrados no Zoológico de Brasília (ZooB). Resultados: Foram analisados 2715 triatomíneos provenientes de 12/30 regiões administrativas do DF e 95/246 municípios de GO. Houve predomínio de T. sordida em GO e no DF P. megistus foi mais frequente, com taxas de infecção por flagelados similares a T. cruzi de 4,6% e 1,6%, respectivamente. A MO revelou 144/841 triatomíneos positivos para tripanossomatídeos, resultando em uma taxa de infecção de 17,1%. A qPCR, específica para T. cruzi, revelou 347 positivos (41,2%). A sensibilidade da MO para T. cruzi, tendo como referência a TCZ qPCR, foi estimada em 23,6% e a da rDNA24sα cPCR foi estimada em 58,5%. Blastocrithidiatriatomae e T. rangeli foram detectadas nos triatomíneos assim como infecções mistas por T. cruzi e B. triatomae. As taxas de infecção por T. cruzi nas espécies de triatomíneos variaram de 19 a 67% sendo maiores para T. pseudomaculata e em triatomíneos vivos e frescos. O sequenciamento revelou TcI em P. megistus e T. sordida e TcII em P. megistus e T. pseudomaculata. No ZooBT. cruzi foi encontrado em P. megistus (44/50) e em primatas (17/26) de seis gêneros e oito espécies, sendo que um Mico chrysoleucus e um Saguinusniger apresentaram alta parasitemia. Discussão e Conclusão:Os resultados mostram uma baixa sensibilidade da MO para a detecção de tripanosomatídeos em triatomíneos e infecções por pelo menos três espécies de tripanossomatídeos, o que reforça a necessidade de métodos de alta especificidade para o desenvolvimento de estratégias de vigilância adequadas dos vetores da doença de Chagas. O método de TCZ qPCR pode ser considerado um padrão referência para detecção de T. cruziem triatomíneos. ________________________________________________________________________________________________ ABSTRACT
The current knowledge of the distribution and natural infection of synanthropictriatomines is essential for surveillance and control of Chagas disease. In Brazil, optical microscopy (OM) has been used to detect Trypanosoma cruzi in triatomines, but this technique can present several limitations in terms of sensitivity and specificity. Objectives: Analyze the occurrence of triatomines and their natural infection rates for Trypanosoma cruzi in state of Goiás (GO) and the Federal District (DF),estimate the sensitivity of OM to detect T. cruzi in triatomines, and analyze T. cruzi transmission foci in environments where infected triatomine colonies were detected. Methods: The specimens (adults and nymphs) captured in the intra and peridomicile and results of parasitological fresh and stained slides were obtained from LACEN/SES/GO and DIVAL/DF. DNA was extracted from the intestine samples for the PCRs (cPCR and qPCR) using four molecular markers (TCZ, kDNA, 18S and rDNA24sα). Also, T. cruzi infections were assessed in a Panstrongylusmegistuscolony and from blood samples of nonhuman primates found in the Brasilia Zoo (ZooB). Results:2715 triatomines were analyzed from 12/30 administrative regions of the DF and 95/246 municipalities of GO. In GO, T. sordida predominate while in the DF P. megistus was more frequent, with infection rates by flagellates similar to T. cruzi of 4.6% and 1.6%, respectively. OM revealed 144/841 trypanosomatid-positive triatomines, resulting in a 17.1% infection rate. The T. cruzi TCZ qPCR, showed 347 positive (41.2%). The sensitivity of T. cruzi-OM with reference to TCZ qPCR was estimated at 23.6% and rDNA24sα cPCR was estimated at 58.5%. Triatomines were infected by Blastocrithidiatriatomae and T. rangeli. However, mixed infections with T. cruzi and B. triatomae were found. Infection rates by T. cruzi in triatomine species ranged 19-67% and were higher for T. pseudomaculata and alive and fresh triatomines. The sequencing of some samples confirmed the identification of B. triatomae and revealed TcI in P. megistus and T. sordida, and TcII in P. megistus and T. pseudomaculata. In ZooB, T. cruzi was found in 44/50 P. megistus and 17/26 primates (six genera and eight species); one Micochrysoleucus and one Saguinusniger showed high parasitaemia. Discussion and Conclusion: The results show a low sensitivity of OM for trypanosomatids detection in triatomines and infections for at least three trypanosomatid species, which reinforces the need for highly specific methods for the development of appropriate surveillance strategies of the vectors of Chagas disease. It also suggests that the TCZ qPCR method could be considered as a reference standard for detection of T. cruziin triatomines.

Full textStudies on the reproductive biology of nonhuman primates are important for the development of reproductive biotechnologies, in order to species conservation. The aim of this study was to evaluate methods for sperm cryopreservation from Callithrix jacchus and C. penicillata. Semen was collected by penile vibrostimulation from adult animals kept in captivity. Immediately after collection, the variables analyzed were volume, pH, concentration, total and progressive motility, plasma membrane integrity, acrosome integrity, cytochemical mitochondrial activity, DNA fragmentation and morphology. The samples were then chilled in TEST egg yolk extender without glycerol; after this period, the glycerol was added at concentrations of 4 and 6% and further analyzes were performed. The semen was frozen in nitrogen vapour and finally immersion in nitrogen. After a minimum of one month and a half, the samples were thawed and further analysis performed at 10, 40 and 80 minutes after thawing. Fresh semen from C. jacchus and C. penicillata were found to be similar in many aspects. The extender TEST egg yolk with glycerol as much as 4 to 6% concentration was not effective for the protection of spermatozoa from both species during cryopreservation. Further studies are needed to develop a protocol for cryopreservation of semen from both species. However, this work contains information that can guide future studies on sperm cryopreservation of these and other Neotropical primates species.

Os primatas neotropicais dos gêneros Saimiri e Aotus são modelos recomendados pela OMS para estudos experimentais da malária humana, pois são susceptíveis à infecção por plasmódios humanos e reproduzem de forma relativamente confiável a patologia e a imunidade observadas em humanos. Eles apresentam, entretanto, uma importante limitação: a necessidade de se esplenectomizar o animal para que as parasitemias sejam elevadas e consistentes. O objetivo desse estudo foi avaliar a influência da administração de lipossomo-clodronato (LC), utilizado para depletar monócitos/macrófagos em vários modelos experimentais, na infecção por Plasmodium falciparum em Saimiri sciureus. Realizou-se um experimento in vitro utilizando cultivo de esplenócitos de Saimiri, incubados em presença ou não de LC, quantificando-se a depleção de macrófagos por citometria de fluxo. Primeiro experimento: foram utilizados seis animais não esplenectomizados divididos em dois grupos que receberam 1mL PBS ou de LC (5mg/mL) a partir do dia 0 de infecção com inóculo contendo 106 hemácias parasitadas com P. falciparum (cepa FUP). Segundo experimento: foram utilizados 14 animais não esplenectomizados divididos em seis grupos - três grupos não infectados, com dois animais cada, que receberam 1mL PBS, ou 0,5mL ou 1mL de LC e três grupos infectados no dia 0 que receberam as mesmas administrações; sendo que o grupo que recebeu PBS tinha dois animais e os grupos que receberam LC tinham três animais cada. Em ambos os experimentos as administrações foram por via intravenosa duas vezes por semana a partir de dia 0. Após eutanásia, foram realizados exames histopatológicos e ensaio de expressão de citocinas em células esplênicas No ensaio in vitro o LC induziu uma citotoxicidade dose-dependente de monócitos/macrófagos. No primeiro experimento o grupo tratado com LC apresentou aumento na parasitemia alcançando valores superiores a 20% no d11 e requerendo tratamento. O grupo tratado com PBS apresentou parasitemia de 0,029% a 8,15% e foi capaz de controlar espontaneamente a infecção no d18. No segundo experimento os animais infectados que receberam 0,5mL LC apresentaram parasitemias mais elevadas (pico entre 16,1% e 26,7% entre d10 e d14) do que os outros grupos (grupo PBS - picos entre 6,3% e 17,7% entre d12 e d13, e grupo 1mL LC \2013 pico entre 4,8% e 8,8% entre d11 e d15). Em todos os animais infectados, a temperatura foi relacionada com a presença de parasitemia e a hemoglobina e o hematócrito diminuíram alcançando valores mínimos quando parasitos já não são mais detectáveis na circulação. Os animais toleraram clinicamente a administração de LC, mas apresentaram sinais histopatológicos de toxicidade hepática. Em conclusão, o LC é capaz de promover parasitemias mais altas em infecções de P. falciparum em primatas S. sciureus. A infecção esteve associada a evidências de depleção parcial de macrófagos como diferenças no tamanho dos baços e menor presença de ferro nos baços e fígados dos animais que receberam LC. Ensaios ainda precisam ser realizados para se estabelecer volumes mínimos funcionais e superar o problema da aparente toxicidade hepática
The WHO recommends the Neotropical primates of the genus Saimiri and Aotus as models for experimental studies of human malaria. The se monkeys are susceptible to infection by human P lasmodi a and reproduce relatively reliably the pathology and immunity observed in man . However, the model has a s limitation the need of splenectomy for the obtention of high and consistent parasitemias . The aim was to evaluate the influence of administration of clo dronate - liposome (CL), used to deplete monocytes/macrophages in several experimental models, in Plasmodium falciparum infected Saimir i sciureus . We conducted an experiment using in vitro culture of spleen cells from Saimiri incubated in the presence or abs ence of C L and quantifying the depletion of macrophages by flow cytometry. Other experiment s were: Exp. 1 : six non splenectomized animals were divided into two groups receiv ing 1 mL of PBS or 1 mL of C L (5mg/mL) from the d ay 0 of infection with 10 6 P. falc iparum (FUP strain) parasitized erythrocytes . Exp. 2 : 14 non - splenectomized animals were divided in six groups - three non - infected groups , with two animals each, receiv ing 1mL PBS, 0.5 mL or 1 mL CL ; and three infected groups receiving the same injections . The group that received PBS had two animals each; and the groups that received CL had three animals each . In both experiments the injections were intravenous, two times a week from d0. After euthanasia, histopathological examination and testing of cytoki ne expression in splenic cells were performed. In vitro assay : the CL induced a dose - dependent monocyte / macrophage cytotoxicity. In Exp 1, the CL treated group showed an increase in parasitemia reaching values higher than 20% at d11 and requir ed treatm ent. T he group treated with PBS showed parasitemia from 0.029 % to 8.15% and was able to spontaneously control the infection by d18. In Exp 2, the infected animals receiv ing 0.5 ml CL showed higher parasitaemia (peak s between 16.1% and 26.7% between d10 and d14) than the other groups (PBS group - peaks between 6.3% and 17 7% between d12 and d13, and 1mL CL group - peak between 4.8% and 8.8% between d11 and d15). In all infected animals, the temperature was related to the presence of parasitaemia and hemoglob in and hematocrit decreased , reaching minimum values during or after clearance of the parasite. The animals tolerated clinically the CL administration but showed histopahological signs of liver toxicity. Animals receiving CL showed less iron in the spl ee n , suggesting a decreased erythrophagocitosis . In conclusion, the CL is capable of promoting higher parasitemia in P. falciparum infect ed S. sciureus primates. The infection was associated with evidence s of partial depletion of macrophages , such as differe nces in the spleen size s and decreased presence of iron in the spleens and livers of animals receiv ing CL. However , more tests still need to be conducted to define the lower volum es of LC that are still functional , to overcome the problem of the apparent l iver toxicity.

Full textThe Callithrix jacchus can be used as a model species for other Neotropical primates species, mainly Callithriquids, for reproductive studies. Semen samples were collected with the following objectives: to improve the penile vibrostimulation technique, validate sperm evaluation techniques for field work, analyse the semen characteristics from animals at 2 colonies (Brazil and Germany), use 3 different freezing protocols in common marmosets sperms, evaluate a cytochemical technique for mitochondrial activity demonstration and evaluate DNA damage in frozen-thawed sperms. The staining techniques used in this work were: Eosin-nigrosin, Trypan-blue Giemsa, simple staining for acrosome, FITC-PSA, SpermOscan®, Spermac®, demonstration of cytochrome c oxidase activity in situ by the oxidation of 3,3'-diaminobenzidina (DAB) and the Comet assay. Freezing was done in 100 µL semen with TESt-Yolk clarified medium plus 4% glycerol using 0.25 mL straws. The freezing protocols tested were: 1) straws at equilibrium phase with a 25°C to 4°C curve in 2 hours, 10 minutes at nitrogen vapour and finally into liquid nitrogen; 2) same as Protocol 1 without the equilibrium phase; and 3) straws directly into liquid nitrogen. All attempts resulted in 83,33% ejaculates. The simple staining technique for acrosome was validated and resulted in an obvious differentiation of the intact acrosome. Semen characteristics evaluation showed no difference between the 2 colonies. Post-thaw evaluation showed all protocols had negative effects on motility and plasmatic membrane integrity with a slightly superiority on Protocol 1. There was no difference between protocols in the mitochondrial activity demonstration and Protocol 2 presented the lowest DNA damage.

Malaria is a disease of greater impact on Public Health in tropical countries because of the high morbidity and mortality. Considering malaria as a zoonosis in which primates can act as reservoirs of species of Plasmodium that can infect humans lead us to conduct malaria studies in primates with unquestionable relevance. It is generally know that the distribution of human and primate malaria following the same distribution of Anopheles mosquitoes vectors, in this study we investigated the presence of Plasmodium spp. in neotropical primates and Anopheles in São Luís Island, Maranhão State, Brazil. Samples were blood neotropical primates CETAS-São Luís (n = 141) and wild life (n = 20) Private Reserve Sítio Aguahy, São José de Ribamar. Anopheles mosquitoes were captured in the same reserve (n = 380) and Mangalho site (n = 36), located in the Environmental Protection Area of the Maracanã, São Luís, totaling 54 pools. The primate blood samples were subjected to morphological, serological (IFA and ELISA-test) and molecular (qPCR and conventional PCR) to identify Plasmodium. The pools of mosquitoes were assayed by testing for molecular identification of Plasmodium. Five primates had positive slides (3.10%) with observation trofozoíticas forms. In IFA four primate serum samples sororreagiram against the antigen of P. malariae. Amplified Plasmodium sp. qPCR at 34.16% (55/161) of samples of primates. Uma das causas desse cen?rio ? a fragmenta??o das ?reas de ocorr?ncia dessas esp?cies na Caatinga e Mata Atl?ntica. Nesse contexto, o presente estudo objetivou a) caracterizar os fragmentos com ocorr?ncia para cada uma das tr?s esp?cies; b) comparar os fragmentos da ?rea de ocupa??o com os demais fragmentos de sua extens?o de ocorr?ncia onde cada esp?cie est? inserida; c) compar?-los entre si quanto as caracter?sticas dos fragmentos, considerando as diferen?as inerentes a cada esp?cie e a cada bioma. Para identificar os fragmentos florestais da extens?o de ocorr?ncia de cada esp?cie e identificar os fragmentos com presen?a confirmada utilizou-se registros de ocorr?ncias das esp?cies obtidas atrav?s de levantamento bibliogr?fico e imagens de sat?lite de ?ndice de Vegeta??o da Diferen?a Normalizada (NDVI) disponibilizadas por Hansen et al (2013). Para caracterizar os fragmentos foram utilizadas v?rias m?tricas da paisagem de forma e isolamento. As tr?s esp?cies de primatas ocorreram usualmente em fragmentos pequenos, regulares e com bordas complexas. Entretanto, quando se comparou com as suas respectivas ?reas de extens?o de ocorr?ncia, as tr?s esp?cies estudadas usualmente ocorreram em fragmentos com maior ?rea, forma mais regular, bordas mais complexas e muito isolados, demonstrando que essas caracter?sticas do fragmento s?o importantes para a ocorr?ncia desses primatas. Para C. coimbrai o tamanho do fragmento vizinho mais pr?ximo mostrou-se mais importante que o isolamento. Seus fragmentos com ocorr?ncia tiveram maior ?ndice de proximidade que os fragmentos de sua extens?o de ocorr?ncia, o que infere em uma maior presen?a de fragmentos na matriz. Dentre as caracter?sticas analisadas dos fragmentos a forma (per?metro/?rea e complexidade da borda) foi a mais determinante na ocorr?ncia dessas tr?s esp?cies de primatas em ambientes fragmentados. Depois da forma, as vari?veis explicativas mais importantes foram ?rea e isolamento (para C. coimbrai). Ao comparar as tr?s esp?cies, observou-se que C. barbarabrownae habita fragmentos menores, mais regulares, com bordas menos complexas e mais isolados que as outras duas esp?cies, possivelmente por s? habitar um bioma com diferente hist?rico de ocupa??o e altera??o, a Caatinga, enquanto as outras duas esp?cies que ocorrem predominantemente ou somente na Mata Atl?ntica. C. coimbrai e S. xanthosternos apresentaram fragmentos com ocorr?ncia com caracter?sticas de forma e isolamento semelhantes, possivelmente por habitarem um mesmo bioma altamente fragmentado, embora tenham diferen?as tanto no uso como em requerimentos de recursos. Portanto, esses tr?s primatas requerem a cria??o de unidades de conserva??o que protejam fragmentos que tenham as caracter?sticas de forma, ?rea e isolamento que determinam suas presen?as e a cria??o de corredores ecol?gicos para aumentar a conectividade entre os fragmentos, visto o grande isolamento dos fragmentos de suas respectivas ?reas de ocupa??o.
The species of Brazilian primates Callicebus barbarabrownae, Callicebus coimbrai and Sapajus xanthosternos are at risk of extinction. One of the causes of this scenario is the fragmentation of the areas of occurrence of these species in the Caatinga and Mata Atl?ntica. In this context, the present study has as objective a) characterize the fragments with occurrence to each one of these three species; b) compare the fragments of the area of occupation with the other of its extension of occurrence where each specie is covered; c) compare the three species about the characteristics of the fragments, taking the inherent differences to each specie and each biome. To identify the forest fragments of the extension of occurrence of each species and identify the fragments with confirmed presence it was utilized records of occurrence of the species and images of satellite of the Normalized Difference Vegetation Index (NDVI) shared by Hansen at al (2013). To characterize the fragments were utilized several metrics of the landscape, of form and of isolation. These three species of primates usually occurred in small, regular and with complex borders fragments. However, when it was compared with their respective extension of occurrence areas, the three studied species usually occurred in fragments with bigger area, more regular form and more complex and too isolated borders, demonstrating that these primates do not occur by coincidence. To C. coimbrai the size of the nearest neighboring fragment showed as more important than isolation. Their fragments with occurrence have bigger rate of proximity than the fragments of extension of occurrence, what infer in a bigger presence of fragments in the matrix. Among the analyzed characteristics of the fragments, the form (perimeter/area and complexity of the border) was the most determined in the occurrence of these three species of primates in fragmented environments. After the form, the most important explanatory variables were area and isolation (to C. coimbrai). When the there species were compared, it was observed that C. barbarabrownae had inhabited smaller and more regular areas, with less complex borders and more isolated than other two species, possibly because of its inhabitation in Caatinga, when the other two species predominately or just occurred in Mata Atl?ntica. C. coimbrai and S. xanthosternos showed fragments with similar occurrence about their characteristics of form and isolation, possibly because they have inhabited the same over-fragmented habitat despite that they have differences not only in the use, as in the requirement of resources. Thus, these three primates require require the creation of unites of conservation that protect fragments that has the characteristics of form, area and isolation that determine their presences and the creation of ecological corridors to increase the connectivity among the fragments, that has seen the large isolation of the fragments in their respective areas of occupation.

Full textHepatitis E virus (HEV) is an emerging pathogen with global distribution that causes acute and chronic hepatitis in humans and asymptomatic infections in animals. In Brazil, the prevalence of HEV infection in humans and animals is still poorly understood, and the characteristics of virulence, pathogenicity and inter-species infection of the genotype 3 isolates circulating in the country are unknown. The study was divided in two stages that aimed to 1) contribute to the molecular diagnosis of HEV infection by the development of a real-time RT-PCR protocol (RT-qPCR) for HEV-RNA research in biological samples, and 2) to contribute to understanding of molecular, serological, clinical-laboratory, ultrasonographic and histopathological features of HEV genotype 3 in owl monkeys (Aotus azari infulatus) experimental infected with isolate of swine HEV genotype 3 previously detected in the Eastern Brazilian Amazon. The RT-qPCR protocol was developed with characterization of a quantification standard curve and later applied concurrently with serological tests in the retrospective evaluation of 318 (n = 318) human serum samples of hepatitis E suspected cases. HEV-RNA was not detected in any of human tested samples, but seroprevalence of 3.4% and 5.9% was determined for anti-HEV IgM and IgG, respectively, that indicated a low prevalence of HEV infection, even among patients with clinical and/or laboratory suspicion of hepatitis E in the Brazilian Amazon. The experimental study in owl monkeys was developed during12 weeks and the animals were infected by intravenous (n = 3) and oral (n = 3) routes (and two negative controls) were evaluated for determination of clinical, biochemical, hematological, serological (anti-HEV IgM and IgG by enzyme immunoassay) and molecular (HEV-RNA serum and stool by RT-qPCR) parameters weekly. Additionally, the animals were also evaluated by hepatic ultrasonography, histopathology and immunohistochemistry research of HEV antigens in liver monthly. The six infected owl monkeys presentend HEV-RNA in serum and/or stool, and some monkeys presented with evidence of seroconversion, liver detection of HEV antigens by immunohistochemistry associated with clinical and/or laboratory findings of oligosymptomatic acute hepatitis. Thus, the RT-qPCR protocol demonstrated to be applicable in the molecular investigation of HEV infection in human and animal samples, and it also represented an important laboratory diagnostic tool. The experimental study allowed the validation of the first neotropical primate model for HEV genotype 3 infection studies.

Full textA toxoplasmose é causada pelo protozoário Toxoplasma gondii, sendo uma das zoonoses mais disseminadas no mundo e amplamente estudada. Os felinos são os únicos hospedeiros definitivos conhecidos deste protozoário, que possui como hospedeiros intermediários os animais homeotérmicos, incluindo os próprios felinos. No caso dos primatas neotropicais esta parasitose não parece afetar de igual modo os animais. Estes podem apresentar uma sintomatologia leve, com baixa taxa de mortalidade no género Sapajus, a sintomatologia mais grave com taxas de mortalidade entre 20 a 80% na família Atelidae, ou mesmo de 100% na subfamília Callitrichinae. Porém, em grande parte dos primatas neotropicais, a toxoplasmose é fatal sem sintomatologia prévia aparente, o que dificulta o diagnóstico e a aplicação atempada de um tratamento. Assim, o objetivo do presente estudo foi determinar os valores de seroprevalência de infeção por T. gondii, através do teste de aglutinação modificado (“modified agglutination test” – MAT), em 49 primatas neotropicais do Brasil que deram entrada na Associação Mata Ciliar (AMC). Os resultados obtidos revelaram uma seroprevalência geral de 16,33% (8/49). No presente estudo todos os primatas seropositivos para T. gondii dos géneros Allouata e Callithrix acabaram por morrer. Por outro lado, verificou-se uma sobrevida de 100% no género Sapajus. Neste trabalho, embora não se tenha obtido diferença estatisticamente significativa na prevalência de infeção por T. gondii de acordo com o género (taxonomia) dos animais estudados, este pode ser um fator determinante na infeção por este protozoário devido aos diferentes estilos de vida entre primatas. Como os estudos neste grupo de animais são escassos e difíceis de realizar, mas relevantes para a compreensão da epidemiologia da infeção por T. gondii e para o controlo da disseminação da doença, é importante insistir na necessidade de mais trabalhos científicos sobre a infeção por este protozoário em primatas neotropicais de cativeiro e de vida livre.
Toxoplasmosis is caused by the protozoan Toxoplasma gondii, being one of the most widespread zoonoses in the world and widely studied. Felids are the only definitive hosts known of this protozoan, which has as intermediate hosts homeothermic animals, including the felids. In the case of neotropical primates this parasite does not seem to affect the animals equally. These may present a mild symptomatology with a low mortality rate in Sapajus, a more severe symptomatology with mortality rates between 20 and 80% in the Atelidae family, or even 100% in the subfamily Callitrichinae. However, in most neotropical primates, toxoplasmosis is fatal with no apparent previous symptoms, making it difficult to timely diagnosis and treatment application. The aim of the present study was to determine seroprevalence values of T. gondii infection by means of the modified agglutination test (MAT) in 49 neotropical primates from Brazil that were admitted to the Mata Ciliar Association (AMC). The results obtained revealed an overall seroprevalence of 16.33% (8/49). In the present study, all primates seropositive to T. gondii of the genera Allouata and Callithrix eventually died. On the other hand, there was a survival of 100% in the genus Sapajus. In the present work, although no statistically significant difference was found in the prevalence of T. gondii infection according to the genus (taxonomy) of the studied animals, this may be a determinant factor in the infection by this protozoan due to differences in lifestyles among primates. As studies in this group of animals are scarce and difficult to perform, but relevant for understanding the epidemiology of T. gondii infection and for controlling the spread of the disease, it is important to emphasize the need for more scientific work on the infection by this protozoan in captive and free living neotropical primates
