ABSTRACT The stem and bulb nematode, Ditylenchus dipsaci, is one of the main problems which affects garlic crop in Brazil; however, information on the host status of the crop to the pathogen is scarce. Thus, the aim of this study was to evaluate the host status of 11 garlic genotypes to Ditylenchus dipsaci and their productivity in experiments conducted under controlled conditions and in the field. In a greenhouse, diverse genetic materials were evaluated for nematode reproduction factor (RF). At the same time, the nematode reproduction, damage caused and productivity of these same genotypes were evaluated in an area naturally infested. In the greenhouse, ‘AM-PC Farias’, ‘Quitéria’, ‘BRS Hozan’, ‘Peruano’, ‘Chonan’ and ‘Moz 114’ were resistant; and, under field conditions, in these same genotypes, the nematode showed the lowest rates of reproduction in the soil (RF= 0.2 to 10.77) and in the tissues (1 to 3,893 specimens/plant), and there were the lowest percentages of symptomatic (0 to 48.66%) and dead (4.17 to 19.57%) plants. Higher productivities (4.32 to 11.05 t/ha) and bulb weight (13.12 to 58.63 g) were obtained with ‘AM-PC Farias’, ‘Quitéria’ and ‘AM-Erenice’; however, only in ‘AM-PC Farias’ and in ‘BRS Hozan’ we observed lower population levels of D. dipsaci in bulb peels (110 and 0.1 specimens/g, respectively).

SUMMARYNematodes are abundant and diverse, and include many parasitic species. Molecular phylogenetic analyses have shown that parasitism of plants and animals has arisen at least 15 times independently. Extant nematode species also display lifestyles that are proposed to be on the evolutionary trajectory to parasitism. Recent advances have permitted the determination of the genomes and transcriptomes of many nematode species. These new data can be used to further resolve the phylogeny of Nematoda, and identify possible genetic patterns associated with parasitism. Plant-parasitic nematode genomes show evidence of horizontal gene transfer from other members of the rhizosphere, and these genes play important roles in the parasite-host interface. Similar horizontal transfer is not evident in animal parasitic groups. Many nematodes have bacterial symbionts that can be essential for survival. Horizontal transfer from symbionts to the nematode is also common, but its biological importance is unclear. Over 100 nematode species are currently targeted for sequencing, and these data will yield important insights into the biology and evolutionary history of parasitism. It is important that these new technologies are also applied to free-living taxa, so that the pre-parasitic ground state can be inferred, and the novelties associated with parasitism isolated.

Due to its close resemblance to Wuchereria bancrofti, this nematode is also frequently used as a model organism to study human lymphatic filariasis. This review highlights numerous insights into the morphological, histological, biochemical, immunological and genetic aspects of S. digitata that have broadened our understanding towards the control and eradication of filarial diseases.

Abstract Landraces of the Solanum tuberosum Andigenum group are abundant and diverse. They are a valuable genetic resource possessing resistance to pests, diseases, and environmental stresses. In the Andean region, populations of the false root-knot nematode Nacobbus aberrans became specialized to infect native potatoes, being one of the major limiting factors affecting this crop. A better understanding of the host plant-parasite interactions is important in order to select tolerant or resistant plants to be included in management programs. Despite the close of association of N. aberrans with potato, and the great diversity of the S. tuberosum Andigenum group, few histopathological studies have been conducted. The aim of this work was to analyze histological alterations induced by different Argentine populations of the nematode in naturally infested roots of four Andean potato varieties (Collareja, Negra Imilla, Ojo de Señorita and Colorada). All the varieties showed hyperplastic tissue in the central zone of galls, where syncytia developed in close association with the nematode female. Syncytia were composed of modified hyperplastic tissue and parenchyma xylem cells. The results showed differences among varieties in their response to nematode populations, with Ojo de Señorita and Negra Imilla being the most susceptible ones. This study is the first describing histopathological alterations induced by N. aberrans in susceptible Andean potato landraces.

Piquin pepper [Capsicum annuum L. var. glabriusculum (Dunal) Heiser and Pickergill] is a semidomesticated pepper with high commercial value and wide applications as fresh or processed products. Piquin pepper plants have been difficult to domesticate and cultivate because of low seed germination, genetic and morphologic variability, insect and disease susceptibility, and limited environmental physiology information. Currently, seed sterility is no longer considered a limiting factor as hormonal, chemical, and thermal treatments have been developed to overcome seed dormancy. In vitro propagation (primarily by direct organogenesis) is still not reliable for seedling production. Cropping systems of piquin pepper plants include traditional methods such as agroforestry and full sunlight, and under protected horticulture conditions, mainly shade nets. Shade levels and water availability affect yield and vegetative growth. Piquin pepper plants can be grown under diverse geographic and edaphic conditions. Nutrition and fertilization studies are limited. Biotic stresses that can cause economic damage to piquin pepper plants include most that affect other pepper cultivars. Piquin pepper is also considered an important genetic resource as it reports resistance to some viral groups, which could be used for genetic improvement of other cultivated peppers. Current research needs involve the development of dependable plant materials (cultivated varieties) with reduced labor needs, particularly during the harvest period. In addition, research is needed to reduce the susceptibility of piquin pepper plants to other plant diseases. This review presents an analysis of the aspects related to the production of piquin peppers under cultivated conditions.

Dagger nematodes of the genus Xiphinema include a remarkable group of invertebrates of the phylum Nematoda comprising ectoparasitic animals of many wild and cultivated plants. Damage is caused by direct feeding on root cells and by vectoring nepoviruses that cause diseases on several crops. Precise identification of Xiphinema species is critical for launching appropriate control measures. We deciphered the cryptic diversity of the Xiphinema hispanum-species complex applying integrative taxonomical approaches that allowed us to verify a paradigmatic example of the morphostatic speciation and the description of a new species, Xiphinema malaka sp. nov. Detailed morphological, morphometrical, multivariate and genetic studies were carried out, and mitochondrial and nuclear haploweb analyses were used for species delimitation of this group. The new species belongs to morphospecies Group 5 from the Xiphinema nonamericanum-group species. D2-D3, ITS1, partial 18S, and partial coxI regions were used for inferring the phylogenetic relationships of X. malaka sp. nov. with other species within the genus Xiphinema. Molecular analyses showed a clear species differentiation not paralleled in morphology and morphometry, reflecting a clear morphostatic speciation. These results support the hypothesis that the biodiversity of dagger nematodes in southern Europe is greater than previously assumed.

Natural variation in the root-knot nematode Meloidogyne incognita is problematic for breeding programs: populations possessing similar morphological characteristics can produce different reactions on the same host. We collected 30 widely dispersed M. incognita populations from protected pepper production systems in major pepper-growing regions of Israel and accurately identified their virulence characteristics by modified differential host test in a growth chamber on tomato, tobacco, cotton, melon, pepper, and peanut. Galling indices and reproduction were determined on the different hosts. All populations fit the published scheme for M. incognita race 2, except for reproduction on cotton plants by five out of 25 tested M. incognita populations, indicating host-range variations. Reaction of three genes that confer resistance to M. incognita—Me1, Me3 and N—to the collected populations was evaluated. Several M. incognita populations induced galling and reproduced successfully on pepper genotypes carrying Me3 and N, whereas plant resistance conferred by Me1 was more robust for all examined populations. Moreover, the effect of genetic background on Me1 resistance demonstrated a relative advantage of several genotypes in nematode infestations. Efficiency of Me3 under local nematode infestation was further studied with a homozygous line carrying two Me3 alleles. Reproduction of virulent populations on the homozygotes (Me3/Me3) and heterozygotes (Me3/Me3+) was similar, suggesting a limited quantitative effect of Me3. These results present the first characterization of host range, reproduction, and molecular aspects of M. incognita from Israel and highlight the importance of taking a multidimensional approach in pepper-breeding programs for resistance to M. incognita.

This article provides an overview of the origin, genetic diversity and methods and trends in breeding of selected ornamental geophytes (Lilium, Tulipa, Narcissus and Hippeastrum). The role of interspecific hybridisation and polyploidisation in assortment development is reviewed. A great variety of cultivars with traits of interest have been generated over the last century by using classical breeding. Geophyte breeders have been interested in a diversity of traits, including resistance to diseases, flower colour and shape, long lasting flowering and a long vase life. Shortening the long breeding process of many geophytes by reducing the juvenile phase and using in vitro techniques are reviewed. Currently, the breeding process has been enhanced by using modern molecular cytogenetic techniques. Genomic in situ hybridisation is frequently used, among other techniques, for genome differentiation in interspecific hybrids, and for assessment of the extent of intergenomic recombination in backcross progenies. Furthermore, several molecular marker techniques are used for verification of hybrid status, identification of genetic diversity, confirmation of the genetic fidelity of in vitro propagated plants and construction of high-density linkage maps. Recently, a myriad of new plant breeding technologies, such as cisgenetics and genome editing technologies have been used to improve the traits of ornamental geophytes, an endeavour that is discussed here. Breeding trends, cultivar novelties as well a new cultivars registered by international authorities during the last five years are presented in detail.

Viruses constitute the largest group of emerging pathogens, and geminiviruses (plant viruses with circular, single-stranded DNA genomes) are the major group of emerging plant viruses. With their high potential for genetic variation due to mutation and recombination, their efficient spread by vectors, and their wide host range as a group, including both wild and cultivated hosts, geminiviruses are attractive models for the study of the evolutionary and ecological factors driving virus emergence. Studies on the epidemiological features of geminivirus diseases have traditionally focused primarily on crop plants. Nevertheless, knowledge of geminivirus infection in wild plants, and especially at the interface between wild and cultivated plants, is necessary to provide a complete view of their ecology, evolution, and emergence. In this review, we address the most relevant aspects of geminivirus variability and evolution in wild and crop plants and geminiviruses’ potential to emerge in crops.

In recent years, the CRISPR/Cas9-based gene-editing techniques have been well developed and applied widely in several aspects of research in the biological sciences, in many species, including humans, animals, plants, and even in viruses. Modification of the viral genome is crucial for revealing gene function, virus pathogenesis, gene therapy, genetic engineering, and vaccine development. Herein, we have provided a brief review of the different technologies for the modification of the viral genomes. Particularly, we have focused on the recently developed CRISPR/Cas9-based gene-editing system, detailing its origin, functional principles, and touching on its latest achievements in virology research and applications in vaccine development, especially in large DNA viruses of humans and animals. Future prospects of CRISPR/Cas9-based gene-editing technology in virology research, including the potential shortcomings, are also discussed.

Eggplant (Solanum melongena) is the third most important vegetable in Asia and of considerable importance in the Mediterranean belt. Although global eggplant production has been increasing in recent years, productivity is limited due to insects, diseases, and abiotic stresses. Genetic engineering offers new traits to eggplant, such as seedless parthenocarpic fruits, varieties adapted to extreme climatic events (i.e., sub- or supra-optimal temperatures), transcription factor regulation, overexpressing osmolytes, antimicrobial peptides, Bacillusthuringiensis (Bt) endotoxins, etc. Such traits either do not occur naturally in eggplant or are difficult to incorporate by conventional breeding. With controversies, Bt-expressing eggplant varieties resistant to eggplant fruit and shoot borers have already been adopted for commercial cultivation in Bangladesh. However, to maximize the benefits of transgenic technology, future studies should emphasize testing transgenic plants under conditions that mimic field conditions and focus on the plant’s reproductive stage. In addition, the availability of the whole genome sequence, along with an efficient in vitro regeneration system and suitable morphological features, would make the eggplant an alternative model plant in which to study different aspects of plant biology in the near future.

Abstract The inner nuclear membrane (INM) hosts a unique set of membrane proteins that play essential roles in various aspects of the nuclear function. However, overaccumulation or malfunction of INM protein has been associated with a range of rare genetic diseases; therefore, maintaining the homeostasis and integrity of INM proteins by active removal of aberrantly accumulated proteins and replacing defective molecules through proteolysis is of critical importance. Within the last decade, it has been shown that INM proteins are degraded in yeasts by a process very similar to endoplasmic reticulum-associated degradation (ERAD), which is accomplished by retrotranslocation of membrane substrates followed by proteasome-dependent proteolysis, and this process was named inner nuclear membrane-associated degradation (INMAD). INMAD is distinguished from ERAD by specific INM-localized E3 ubiquitin ligases and proteolysis regulators. While much is yet to be determined about the INMAD pathway in yeasts, virtually no knowledge of it exists for higher eukaryotes, and only very recently have several critical regulators that participate in INM protein degradation been discovered in plants. Here, we review key molecular components of the INMAD pathway and draw parallels between the yeast and plant system to discuss promising directions in the future study of the plant INMAD process.

Plant development is different from animals by many fundamental aspects; as they have immobilized cells, a rigid cell wall, and the large central vacuole. Plant growth and cell division are restricted to the specific area of the shoot and root called meristems. Plants have the ability to carry out differentiation, dedifferentiation and redifferentiation. In plants, the growth and differentiation processes are controlled by hormonal and genetic factors. Phytohormones can exert independent/ dependent actions on plant growth and development. A pool of stem cells is placed at the niche of the apex meristem, which is the source of self-renewal of the cell system and its maintenance to provide cells to differentiated tissues. A complex interaction network between hormones and other factors maintains a balance between cell division and differentiation. Auxins promote the growth, gibberellins’ function in seed germination, cytokinin’s influence on cell division and delay leaf senescence; abscisic acid promotes the stomatal closure and bud dormancy, while salicylic acid promotes resistance against different diseases. Plants are often exposed to different abiotic and biotic stresses, for example, heat, cold, drought, salinity etc., whereas biotic stress arises mainly from fungi, bacteria, insect, etc. Phytohormones play a critical role in well-developed mechanisms that help to perceive the stress signal and enable the plant’s optimal growth response. In this review, we studied both the intrinsic and extrinsic factors which govern growth and differentiation of plants under normal and stress condition. This review also deals with genetic modifications occurring in the cell and cell signaling during growth and differentiation.

Expansions of gene-specific DNA tandem repeats (TRs), first described in 1991 as a disease-causing mutation in humans, are now known to cause >60 phenotypes, not just disease, and not only in humans. TRs are a common form of genetic variation with biological consequences, observed, so far, in humans, dogs, plants, oysters, and yeast. Repeat diseases show atypical clinical features, genetic anticipation, and multiple and partially penetrant phenotypes among family members. Discovery of disease-causing repeat expansion loci accelerated through technological advances in DNA sequencing and computational analyses. Between 2019 and 2021, 17 new disease-causing TR expansions were reported, totaling 63 TR loci (>69 diseases), with a likelihood of more discoveries, and in more organisms. Recent and historical lessons reveal that properly assessed clinical presentations, coupled with genetic and biological awareness, can guide discovery of disease-causing unstable TRs. We highlight critical but underrecognized aspects of TR mutations. Repeat motifs may not be present in current reference genomes but will be in forthcoming gapless long-read references. Repeat motif size can be a single nucleotide to kilobases/unit. At a given locus, repeat motif sequence purity can vary with consequence. Pathogenic repeats can be “insertions” within nonpathogenic TRs. Expansions, contractions, and somatic length variations of TRs can have clinical/biological consequences. TR instabilities occur in humans and other organisms. TRs can be epigenetically modified and/or chromosomal fragile sites. We discuss the expanding field of disease-associated TR instabilities, highlighting prospects, clinical and genetic clues, tools, and challenges for further discoveries of disease-causing TR instabilities and understanding their biological and pathological impacts—a vista that is about to expand.

Over the last decade, the development and prevalence of obesity have posed a serious public health risk, which has prompted studies on the regulation of adiposity. With the ease of genetic manipulation, the diversity of the methods for characterizing body fat levels, and the observability of feeding behavior, Caenorhabditis elegans (C. elegans) is considered an excellent model for exploring energy homeostasis and the regulation of the cellular fat storage. In addition, the homology with mammals in the genes related to the lipid metabolism allows many aspects of lipid modulation by the regulators of the central nervous system to be conserved in this ideal model organism. In recent years, as the complex network of genes that maintain an energy balance has been gradually expanded and refined, the regulatory mechanisms of lipid storage have become clearer. Furthermore, the development of methods and devices to assess the lipid levels has become a powerful tool for studies in lipid droplet biology and the regulation of the nematode lipid metabolism. Herein, based on the rapid progress of C. elegans lipid metabolism-related studies, this review outlined the lipid metabolic processes, the major signaling pathways of fat storage regulation, and the primary experimental methods to assess the lipid content in nematodes. Therefore, this model system holds great promise for facilitating the understanding, management, and therapies of human obesity and other metabolism-related diseases.

Writing about the cultivation of cotton, it is worth mentioning the fundamental problems of its processing. For example, the amounts of pesticides used in the cultivation of cotton are greater than for any other crop. Chemicals sprayed on cotton fields are washed away from the fields and, reaching the fresh water sources, pollute them, causing significant damage to the environment. Fortunately, such challenges can be solved by switching to the cultivation of transgenic cotton. Transgenic cotton has already brought many important environmental, social and economic benefits, including reduce of the used pesticides, indirectly increasing of yield, minimizing environmental pollution, reducing the labor force involved and economic costs.Today, the main methods of obtaining transgenic cotton lines are still agrobacterial transformation and biolistics. In recent years, however, innovative methods of transformation have also been developed. For example, the introduction of genetic material mediated by a pollen tube for the cultivation of commercial transgenic cotton is actively used in China. Although in recent decades transgenic lines resistant to diseases, abiotic stresses and with improved fiber quality have been obtained, the dominant position in the market of transgenic cotton is still occupied by lines of plants resistant to insects and herbicides. All the above indicates an insufficient degree of integration between institutes that introduce new advanced developments and agricultural industry.In this review the results of research involving the cultivation and genetic modification of cotton were collected and summarized. The main methods of genetic transformation of cultivated representatives of the genus Gossypium, both actively used at present and still under development, were considered. The most remarkable transgenic lines were also described, among which both those that have already entered agricultural industry and those that have only recently been obtained. Thus, the reader will be able to get a general idea of the current achievements in the field of cotton genetic modification.

Genetics, Epigenetics and Environmental Regulations addresses these three aspects of the development of various organisms in a series of chapters contributed by different authors. The text also does not deal with classical, but nevertheless instructive, experiments such as those on inductive tissue interactions, the fates of embryonic tissues, or the movements of sheets of cells. The book is divided into three sections, covering microbial systems (both prokaryotic and eukaryotic), plants and animals. The section on microbial systems has chapters on topics ranging from virus assembly to the control of the cell cycle in Asperigillus nidulans. Urease, a nickel-containing metalloenzyme is getting remarkable attention due to a diverse range of applications for mankind. Urease plays a magnificent role in various field like agriculture, analytical, geological phenomena, beverage industry and is an important diagnostic tool. Urease is mainly present in bacteria, fungi, plants and invertebrates and its manifestation in specific genera may open new vistas for its taxonomic position. Various qualitative and quantitative assays are also reported for the estimation of urease enzyme. Urease based biosensors utilizing green synthesis on nanoparticles are also trending. Recently developed inhibitors against urease were discussed in the review. Inhibitory mechanisms involving the structural similarity of the substrate through modification or derivatization can also help in rational drug design by two possible competitive ways either by mimicking monodentate urea binding or binding as a tetrahedral intermediate. Immobilisation of urease through gel entrapment, using non-covalent and covalent protein tags, cross linkage, covalent bonding, using composite films, Teflon, co-precipitation and coating on nanoparticles is also reported. This review also comprised of various application of urease including enhancement of fertility in the soil, cell to cell organization, protection to predators, treatment of various bladder related diseases and infections, analysis of urea and heavy metal ions, biocementation, pollution control by bioleaching of heavy metals and making beverages urea and ethyl carbamate free. As researchers have a keen interest in urease enzyme at present, most of its aspects were incorporated in the article to make it helpful to the scientific community for further research related to the development of new inhibitors and add on applications of urease for the upliftment of the human as well as environment.

Cinchona species were widely used as ancient medicines for different diseases because they contain the active component quinine and its derivatives. However, studies on the molecular aspects of cinchona, including its genetic diversity, have not been reported because most previous works focused on the administration of the antimalarial cinchona alkaloid. Quinine is also being tested as alternative compound for the treatment of Covid-19. The Junghuhn Natural Reserve in Indonesia contains three different types of cinchona plants, namely, Cinchona calisaya, Cinchona pubescens, and Cinchona sp. Given that the genetic diversity and kinship of these species have never been studied, collecting data on the cinchona gene pool has become imperative. This study analyzed the genetic diversity of the cinchona species in the Junghuhn Natural Reserve, Indonesia, by using eight RAPD markers, i.e., OPA-2, OPA-9, OPB-02, OPB-03, OPB-04, OPB-05, OPB-7, and OPJ-07, during 2020 at the University of Jenderal Soedirman, Purwokerto-Indonesia. Polymorphic band data were obtained. Then, phenogram analysis was conducted by using UPGMA and maximum parsimony with MEGA7. The RAPD profiles of Cinchona species (C. calisaya, C. pubescent, and Cinchona sp.) revealed polymorphism with different markers, i.e., OPA-2 (90%), OPB-2 (75%), OPB-5 (75%), OPB-3 (66.66%), OPB-4 (66.66%), OPB-7 (66.66%), OPJ-7 (66.66%), and OPA-9 (58.33%) sequentially with total polymorphism (70.62%). C. calisaya was identified as the most distinctive species. UPGMA yielded a coefficient of 0.200 and two distinctive groups: Group I, which comprised C. pubescens and Cinchona sp. with the p-distance value of 0.333, and Group II, which contained C. calisaya. Ixora sp. was treated as an outgroup plant. The topology of the dendrogram was consistent with that of the UPGMA dendrogram. Results may be used for the further exploration of the genetic diversity of cinchona species.

Hempseeds, the edible fruits of the Cannabis sativa L. plant, were initially considered a by-product of the hemp technical fibre industry. Nowadays, following the restorationing of the cultivation of C. sativa L. plants containing an amount of delta-9-tetrahydrocannabinol (THC) <0.3% or 0.2% (industrial hemp) there is a growing interest for the hempseeds production due to their high nutritional value and functional features. The goal of this review is to examine the scientific literature concerning the nutritional and functional properties of hempseeds. Furthermore, we revised the scientific literature regarding the potential use of hempseeds and their derivatives as a dietary supplement for the prevention and treatment of inflammatory and chronic-degenerative diseases on animal models and humans too. In the first part of the work, we provide information regarding the genetic, biochemical, and legislative aspects of this plant that are, in our opinion essential to understand the difference between “industrial” and “drug-type” hemp. In the final part of the review, the employment of hempseeds by the food industry as livestock feed supplement and as ingredient to enrich or fortify daily foods has also revised. Overall, this review intends to encourage further and comprehensive investigations about the adoption of hempseeds in the functional foods field.

The human diet has passed through several revolutionary changes since the introduction of agriculture, which has led to substantial modifications in individuals’ nutrition behavior. Overwhelming evidence supporting that diet is a key environmental risk factor affecting the now rampant incidence of the diseases of affluence such as obesity, type 2 diabetes mellitus, cardiovascular diseases, and certain types of cancer. Findings on the health implications of low carbohydrate-high protein diet are inconsistent and controversial. Unlike modern humans, it was argued that the eating patterns of the Paleolithic hunter-gatherers could have beneficial effects on human health by reducing diet-induced chronic lifestyle diseases. Hence, the objective of this review was to outline the main aspects of Paleolithic hunter-gatherers’ dietary patterns and its main long-term health consequences. Paleolithic diet is based on the assumption that our hunter-gatherer ancestors were nourished on low carbohydrate-high protein diet. Yet, the majority of literature on Paleolithic diet is anecdotal and reflects two opposite viewpoints. First, advocates and proponents argued that human’s genome could have been modified a little since the early stages of agriculture. Thus, genetically, humans remain Stone Agers adapted for a Paleolithic dietary regimen. As such, consuming a diet similar to that consumed during the Paleolithic era would be more compatible with our genetic makeup and might reduce the occurrence of diet-related lifestyle diseases. Second, critics asserted that the Paleolithic dietary recommendations and restrictions are not evidence-based. Adherents of Paleolithic nutrition claimed that modern populations who maintained Paleolithic lifestyle are totally free of the diseases of affluence. Nonetheless, information about the Paleolithic diet have been drawn from studies of surviving hunter-gatherer populations, archeological records, and analysis of wild plants and animals plus other methods. Recommendations of the Paleolithic nutrition are not based on interventional studies, thus the adoption of which could have negative health consequences. Hence, before adopting low carbohydrate-high protein diet, it seems logical to undertake a well designed population-based longitudinal studies to evaluate the long-term health consequences of the Paleolithic nutrition.

Espécies do referido gênero são responsáveis por diversas doenças em plantas cultivadas, causando principalmente manchas foliares. A correta identificação das espécies é importante para o conhecimento e manejo das doenças, sendo essas identificações baseadas principalmente em características morfológicas, como aspectos dos micélios, morfologia das hifas, formas e tamanho dos conídios. Entretanto, técnicas moleculares como a amplificação de genes específicos, além de análises do padrão de digestão de regiões ITS através de enzimas de restrição, têm contribuído para a identificação e diferenciação genética entre espécies. No presente trabalho foi realizada a caracterização morfológica e molecular de isolados de Curvularia oriundos de plantas de milho infectadas. Os isolados foram agrupados de acordo com a identificação morfológica e molecular, sendo que todos apresentaram o mesmo perfil de amplificação do DNA. However, molecular techniques such as the amplification of specific genes, as well as digestion pattern analyzes of ITS regions through restriction enzymes, have contributed to the identification and genetic differentiation between species. In the present study, the morphological and molecular characterizations of Curvularia isolates from infected corn plants were realized. The isolates were grouped according to the morphological and molecular identification, all of which presented the same DNA amplification profile. Based on the analysis of literary sources and publications, the features of soybean varieties for cultivation in Podillia were studied. The key directions of increasing the productivity of soybean varieties are considered. Advantages are given and substantiated, and the main disadvantages and problems of growing soybean varieties in Podillia conditions are indicated The features of soybean varieties, the influence of the inoculation process and the efficiency of micronutrient fertilization have been investigated, which made it possible to develop methods of growing technology taking into account plant biology on changes in climatic conditions. The growth processes and development of soybean varieties have been investigated: Maxus, Cordoba, Saska, depending on the treatment of seeds with an insecticidal-fungicidal preparation Standak Top, inoculant Hai Kot Super Hai Kot Super Extender and seed treatment with micronutrient fertilizer Como 15, as well as foliar dressing with micronutrient fertilizers Vuksal Boron and Bospholiar during the growing season of plants. Having carried out research on various groups of ripeness from such varieties of soybeans as Maxus, Cordoba, Saska, positive results were obtained on the yield from the introduction of micronutrient fertilizers, inoculants, treatment of the inoculum of soybean seeds with an inoculant and a microelement, the preparation Vuxal Como 15. And also from the treatment of seeds with the preparation Standak Top 1 l t, which in turn prevents the development of such diseases as fusarium, anthracnose, seed mold, promotes rooting of plants in the soil due to the accelerated development of the root system, an increase in the assimilation surface of the leaf apparatus, promotes the activation of nitroreductase, which in turn activates the work processes of photosynthesis, manifested in the so-called AgCelence effect, plants have an intensely saturated dark green color, control of soil pests. Carrying out experiments on the effect of seed inoculation on soybean yield, we combined an inoculant, VuksalKoMo preparation and Sdandak Top insecticidal fungicide preparation with a sowing period of up to 5-7 days in a tank mixture. One of the main requirements is the use of high-quality inoculants with a high content of viable nitrogen-fixing bacteria for processing soybean seeds, today there is a need, since it fully reveals and realizes the genetic and varietal potential of modern varieties, and this, in turn, will ensure high yields soybeans with optimal costs and the fastest return on investment, especially in today's conditions. In the study results, new technological aspects are presented, in which the processes of analysis and hardening of micro-manure and infectious-fungicidal preparation in the technological development and quality improvement of the production process are revealed. It should also take into account the relative humidity and reserves of productive soil moisture. The research results are aimed at solving urgent problems in the technology of growing leguminous crops, namely: developing a version of the technology for growing soybeans for the selection of varieties adapted to a given climatic zone, the use of inoculants and micronutrients in the conditions of climate change in Podillia.

ABSTRACT Finger millet (Eleusine coracana (L) commonly known as ragi is an important crop used for food, forage and industrial products. Finger millet has a wide ecological and geographical adaptability and resilience to various agro-climatic adversities hence, it is highly suited to drought condition and marginal land and requires low external input in cultivation.. Farmers participatory field demonstrations of ragi variety Arjun and Bhairabi were conducted at two villages ie Pedawada of Malkangiri block and MPV -1 of Kalimela block of Malkangiri district, comprising 40 farmers in cluster approach in Kharif 2018 and 2019 , by Krishi Vigyan Kendra, Malkangiri , in South Eastern Ghat Zone of Odisha . Conducting front line demonstrations on farmer’s field help to identify the constraints and potential of the finger millet in the specific area as well as it helps in improving the economic and social status of the farmers. Observation on growth and yield parameters were taken and economic analysis was done. The final seed yield was recorded at the time of harvest and the gross return in (Rs ha -1) was calculated based on prevailing market prices. The results from the demonstration conclusively proved that finger millet variety Arjun (OEB-526) recorded the higher yield ( 18.8 q ha-1) , followed by Bhairabi ( 15.3 q ha-1) and farmer’s traditional variety Nali Mandia ( Dasaraberi) recorded an average yield of (8.6 q ha-1 ) . HYV Finger millet variety Arjun with proper nutrient management and plant protection measures gave 118 % higher over farmer’s practices. The technological and extension gap was 1.9 q ha-1 and 12.07 q ha-1 respectively. Similarly, technological index was 8.2 percent. The benefit cost ratio was 2.4 and 1.9 in case of Arjun and Bhairabi respectively and in case of farmer’s variety Nali Mandia it was 1.4. Hence the existing local finger millet variety can be replaced by HYV Arjun ans Bhairabi , since it fits good to the existing rainfed farming situation for higher productivity. By conducting front line demonstrations on millet on large scale in farmer’s field, yield potential of finger millet can be enhanced largely which will increase in the income level of farmers and improve the livelihood condition of the farming community. Introduction Among small millets, finger millet (Elusine coracana L,) locally known as Ragi/Mandia is the most important crop grown in tribal districts of Odisha and it is the staple food of the tribals. It was originated about 5000 years ago in east Africa (possibly Ethiopia) and was introduced into India, 3000 years ago (Upadhyaya et al., 2006) and it is highly suited to drought condition and marginal land and requires low external input in cultivation. Millet is a collective term referring to a number of small seeded annual grasses that are cultivated as grain crops, primarily on marginal lands in dry areas in temperate, subtropical and tropical regions (Baker, 1996). Nutritionally finger millet is superior to major cereal crops and rich source of micronutrients such as calcium, phosphorous, magnesium and iron. And it has several health benefits. Finger millet grains contain higher amount of proteins, oils and minerals than the grains of rice, maize or sorghum (Reed et al., 1976). Vadivoo and Joseph (1998) mentioned finger millet grains contain 13.24% moisture, 7.6% protein, 74.36% carbohydrate, 74.36% carbon, 1.52% dietary fiber, 2.35% minerals, 1.35% fat and energy 341.6 cal/100g. (Joshi and Katoch, 1990; Ravindran, 1991). It is a rich source of micronutrients such as calcium, phosphorus, magnesium and iron. Cysteine, tyrosine, tryptophan and methionine are the right spectrum of amino acids found in finger millet protein (Rachie, 1975). The increase in global temperature leads to climate changes that directly affect crop production and increase people's hunger and malnutrition around the world.. With regard to protein (6-8%) and fat (1-2%) it is comparable to rice and with respect to mineral and micronutrient contents it is superior to rice and wheat (Babu et al., 1987). It is also known for several health benefits such as anti-diabetic, anti-tumerogenic, atherosclerogenic effects, antioxidant, which are mainly attributed due to its polyphenol and dietary fiber contents. Being indigenous minor millet it is used in the preparation of various foods both in natural and malted forms. Grains of this millet are converted into flours for preparation of products like porridge, puddings, pancakes, biscuits, roti, bread, noodles, and other snacks. Besides this, it is also used as a nourishing food for infants when malted and is regarded as wholesome food for diabetic's patients. Diversification of food production must be encouraged both at national and household level in tandem with increasing yields. Growing of traditional food crops suitable for the area is one of the possible potential successful approaches for improving household food security. Malkangiri is one of the seven districts where a flagship programme called “Special Programme for Promotion of Millets in Tribal Areas of Odisha (hereafter, Odisha Millets Mission, (OMM)” has been launched by Department of Agriculture and Farmers Empowerment, Odisha in order to revive millets in rainfed farming systems and household consumption. It was started in kharif 2017 in four blocks of the district, namely Chitrakonda, Korkunda, Mathili and Khairiput. The Government of Odisha launched Odisha Millets Mission (OMM) also known as the Special Programme for Promotion of Millets in Tribal Areas of Odisha in 2017 to revive millets in farms and on plates. The aim was to tackle malnutrition by introducing millets in the public distribution system (PDS) and other state nutrition schemes. The focus is on reviving millets in farms and putting it on plates.” Millet, a nutritious and climate-resilient crop, has traditionally been cultivated and consumed by tribal communities in the rainfed regions of southern Odisha. Technology gap, i.e. poor knowledge about newly released crop production and protection technologies and their management practices in the farmers’ fields is a major constraint in Ragi production. So far, no systematic approach was implemented to study the technological gap existing in various components of Ragi cultivation. Awareness of scientific production technology like HYV of ragi, seed treatment with fungicide, use of insecticide and bio-fertilizers, is lacking in Malkangiri district which were a key reason for low productivity. The production potential could be increased by adopting recommended scientific and sustainable management production practices with improved high yielding varieties and timely use of other critical inputs. Objective The field experiment was undertaken to study the performance of three finger millet varieties Local Mandia (Nali Mandia), Bhairabi and Arjun in rainfed upland situation in kharif season. The present investigation was undertaken to evaluate the field performance of newly released finger millet varieties Arjun and Bhairabi under rainfed condition. The demonstrations were carried out in Malkangiri district covering two villages like Pedawada and MPV-6 to find out the existing technological and extension gap along with technology index with an objective to popularize the ragi varieties having higher yield potential. Material and methods The study was carried out in operational area of Krishi Vigyan Kendra (KVK), Malkangiri during Kharif season in the year 2018 and 2019. The study was under taken in Malkangiri and Kalimela blocks of Malkangiri district of Odisha and the blocks were selected purposefully as Finger millet is the major cereals crop grown in large area in Kharif season. The demonstrations were conducted in two different adopted villages Pedawada and MPV-6 in cluster approach. The Front Line Demonstration (FLD) is an applied approach to accelerate the dissemination of proven technologies at farmer’s fields in a participatory mode with an objective to explore the maximum available resources of crop production and also to bridge the productivity gaps by enhancing the production in national basket.The necessary steps for selection of site and farmers and layout of demonstrations etc were followed as suggested by Choudhary (1999). Forty numbers front line demonstrations on HYV Ragi were conducted in two clusters comprising 40 numbers of farmers. All the participating farmers were trained on various aspects of Ragi production technologies and recommended agronomic practices and certified seeds of Ragi variety Arjun and Bhairabi were used for demonstration. The soil of demonstration site was slightly acidic in reaction (pH-5.0 to 5.25) with sandy loam in texture and EC was 0.134 (dS m −1). The available nitrogen, phosphorus and potassium was between 214 .00, 22 .00, 142 .00 (Kg ha−1) respectively with 0.48 (%) Organic Carbon. The crop was sown in under rainfed condition in the first to second week of July. The crop was raised with recommended agronomic practices and harvested within 4th week of November up to 2nd week of December. Krishi Vigyan Kendra ( KVK), Malkangiri conducted front line demonstration with HYV varieties like Bhiarabi and Arjun and farmer’ local var Dasaraberi as check Finger millet Variety Arjun (OEB-526) is having Maturity duration 110 days and average yield 20.7q/ha with moderate resistance to leaf, neck and finger blast and brown seed and Bhairabi is a HYV of Ragi with Maturity duration 110 days and average yield 17.6 q/ha. Moderate resistance to leaf, neck blast and brown seeded and protein content 81%. Local variety Dsaraberi or Nali Mandia is having 105 days duration and drought tolerant variety used as farmers variety as local check . The technologies demonstrated were as follows: Popularization of high yielding Ragi variety, Seed treatment with Trichoderma viride @ 4g kg-1 seed , Line sowing with, soil test based fertilizer application along with need based plant protection measures. The field was ploughed two times and planking was done after each plugging, Need based plant protection measures were taken; along with soil test based fertilizer application was done with fertilizer dose 40:30:60 kg. N: P2O5: K2O kg ha -1. In case of local checks existing practices being used by farmers were followed. The observations were recorded for various parameters of the crop. The farmers’ practices were maintained in case of local checks. The field observations were taken from demonstration plot and farmer’s plot as well. Parameters like Plant height, number of fingers per plant, length of finger , no of fingers per year, 1000 seed weight and seed yield were recorded at maturity stage and the gross returns (Rs ha -1 ) were calculated on the basis of prevailing market price of the produce. The extension gap, technology gap, technology index along with B: C ratio was calculated and the data were statistically analyzed applying the statistical techniques. Statistical tools such as percentage, mean score, Standard deviation, co-efficient of variation, Fisher‘s “F” test, were employed for analysis of data. The farmer’s practices (FP) plots were maintained as local check for comparison study. The data obtained from intervention practices (IP) and famers practices (FP) were analyzed for extension gap, technological gap, technological index and benefit cost ratio study as per (Samui et al., 2000) as given below. Technology gap = Pi (Potential yield) - Di (Demonstration yield) Extension gap = Di (Demonstration Yield) - Fi (Farmers yield) Technology index = X 100 Result and Discussion The results obtained from the present investigation are summarized below. The Table 1 depicts the major differences observed between demonstration package and farmer’s practices in ragi production in the study area. The major differences were observed between demonstration package and farmer’s practices were regarding recommended varieties, seed treatment, soil test based fertilizer application, keeping optimum plant population by thinning, weed management and plant protection measures. The data of Table 1 shows that under the demonstrated plot only recommended high yielding variety, proper weeding and optimum plant population maintaining by thinning and the farmers used herbicides and the farmers timely performed all the other package and practices. It was also observed that farmers were unaware about balanced fertilizer application, seed treatment, and use of fertilizers application and maintenance of plant population for enhancing the yield. Majority of the farmers in the study area were unaware about use of weed management practices. The findings are in corroborated with the findings of (Katar et al., 2011) From the Table 2 it was revealed that in the district Malkangiri the productivity of finger millet was 6.38 (q ha-1) as compare to state average productivity 8.67 (q ha-1), but there exists a gap between potential yield and farmers yield, which can be minimized by adoption High yielding varieties with improved management practices. The productivity of finger millet was very low in the district as the crop is mostly grown along the hillsides on sloppy land on light textured soil. It was also coupled with negligence in adoption of improved varieties no input like fertilizers use and no plant protection measures and improper method and time of sowing. However, there is a wide gap between the Potential and the actual production realized by the farmers due to partial adoption of recommended package of practices by the growers. Several constraints contributed to yield fluctuation on Ragi production, including: unreliable rainfall; lack of high yielding variety ,disease tolerant varieties; pests and diseases incidence; low producer prices; poor agronomic practices; and lack of institutional support (Bucheyeki et al., 2008; Okoko et al., 1998). One of the central problems of ragi production and processing in this district is due to an uncertain production environment owing to rain fed cultivation, the low resource base of smallholder farmers and processors, and no scope for post harvest management and value addition facilities and poor marketing facility. The results clearly indicated from the Table 3 that the positive effects of FLDs over the existing practices. HYV Ragi Arjun recorded higher yield 18.8(q ha-1) followed by Bhairabi 15.53 (q ha-1) which was 21 % more and the yield performance of these two HYV varieties was higher than the farmer’s variety. This is due to higher of panicle length, more number of tillers and more number of fingers per panicle in HYV of ragi as compared to local variety. The results are in conformity with the findings of (Tomar et al. , 2003). The results clearly indicated the positive effects of FLDs over the existing practices towards enhancing the productivity. It is revealed form table 4 that, as the calculated ‘F’ value at α=0.05 level was found to be larger than table value, indicating significant difference in yield between farmer’s variety and recommended varieties. There was significant difference between average yield of ragi under Farmers practice (FP) and Recommended practice (RP) in variety Arjun under this demonstration. It was concluded that the yield of these HYV ragi varieties was significantly higher as compared to farmer’s variety. The economics and B:C ratio of farmers practice and Demonstration practice has been presented in Table 6. From the table it was revealed that Benefit: Cost ratio (B:C) was recorded to be higher under demonstrations against control treatments during all the years of experimentation. The cost of cultivation in HYV variety was higher due to more labour cost involved in transplanting and also it included cost of fertilizers and plant protection chemicals and also net returns was higher as compared to farmer’s practice. The B: C ratio was found to be 2.4 in case of variety Arjun as compared to 1.9 in case of variety Bhairabi. The results on economic analysis indicated that HYV ragi Arjun and Bhairabi performed better than local variety Ragi. The HYV variety Arjun recorded higher gross return upto Rs 54,332 and followed by Bhairabi Rs 44,289 per ha which was significantly higher than farmers practice and it was due to higher productivity of varieties under demonstration. Conclusion The results revealed that in Malkangiri district finger millet variety Arjun rerecorded highest yield followed by Variety Bhirabi with proper package and practices under rainfed upland condition. From the above study it was concluded that use of finger millet varieties like Arjun or Bhairabi with scientific methods and technological practices of can reduce the technological gap and enhance the productivity in the district. Yield improvement in Finger Millet in the demonstration was due to use of HYV seed and scientific management practices adopted by the farmers. Yield of Finger Millet can be increased to a great extent by conducting effective front line demonstrations in larger area with proven technologies. Finger millet is one of future smart food crop of India and can be grown in the drought condition. This crop is rich in nutrient for food insecurity and within few years because of increase in population of world and depletion of area of production.. The principal reasons of lower productivity of finger millet in the district Malkangiri were lack of knowledge among the farmers about cultivation of HYV finger millet varieties and improper fertilization, late season sowing and severe weed infestation in crop at critical stages. From the above findings, it can be concluded that use of scientific methods of Finger millet cultivation can reduce the technology gap to a considerable extent thus leading to increased productivity of millets in the district. Moreover, extension agencies in the district need to provide proper technical support to the farmers through different educational and extension methods to reduce the extension gap for better production. AbstractThe pine wilt disease (PWD), for which no effective treatment is available at the moment, is a constant threat to Pinus spp. plantations worldwide, being responsible for significant economic and environmental losses every year. It has been demonstrated that elicitation with chitosan increases plant tolerance to the pinewood nematode (PWN) Bursaphelenchus xylophilus, the causal agent of the PWD, but the biochemical and genetic aspects underlying this response have not been explored. To understand the influence of chitosan in Pinus pinaster tolerance against PWN, a low-molecular-weight (327 kDa) chitosan was applied to mock- and PWN-inoculated plants. Nematode population, malondialdehyde (MDA), catalase, carotenoids, anthocyanins, phenolic compounds, lignin and gene expression related to oxidative stress (thioredoxin 1, TRX) and plant defence (defensin, DEF, and a-farnesene synthase, AFS), were analysed at 1, 7, 14, 21 and 28 days post-inoculation (dpi). At 28 dpi, PWN-infected plants elicited with chitosan showed a sixfold lower nematode population when compared to non-elicited plants. Higher levels of MDA, catalase, carotenoids, anthocyanins, phenolic compounds, and lignin were detected in chitosan-elicited plants following infection. The expression levels of DEF gene were higher in elicited plants, while TRX and AFS expression was lower, possibly due to the disease containment-effect of chitosan. Combined, we conclude that chitosan induces pine defences against PWD via modulation of metabolic and transcriptomic mechanisms related with plant antioxidant system.

AbstractThe nematode Caenorhabditis elegans is a genetic model organism and the only animal with a complete nervous system wiring diagram. With only 302 neurons and 95 striated muscle cells, a rich array of mutants with defective locomotion and the facility for individual targeted gene knockdown by RNA interference, it lends itself to the exploration of gene function at nerve muscle junctions. With approximately 60% of human disease genes having a C. elegans homologue, there is growing interest in the deployment of lowcost, high-throughput, drug screens of nematode transgenic and mutant strains mimicking aspects of the pathology of devastating human neuromuscular disorders. Here we explore the contributions already made by C. elegans to our understanding of muscular dystrophies (Duchenne and Becker), spinal muscular atrophy, amyotrophic lateral sclerosis, Friedreich’s ataxia, inclusion body myositis and the prospects for contributions to other neuromuscular disorders. A bottleneck to low-cost, in vivo, large-scale chemical library screening for new candidate therapies has been rapid, automated, behavioural phenotyping. Recent progress in quantifying simple swimming (thrashing) movements is making such screening possible and is expediting the translation of drug candidates towards the clinic.

Nematodes are the most abundant metazoans on the planet can be found in almost all the ecosystems on earth. They may be free-living, marine, bacterivorous, or parasites of plants and animals, including human beings. Nematodes always interact with other organisms, including- viruses, fungi, protozoa, rickettsia, tardigrades and bacteria in their microbiome. Nematodes are particularly well suited for investigating host associations with bacteria because they have a long history of evolution. Nematode-bacteria interaction is now being used as a model for investigating animal-microbe interaction and can be positive (mutualistic) or negative (pathogenic/parasitic) and may be transient or stably maintained (symbiotic). In any disease complex, nematodes play important roles in association with bacteria to aggravate disease intensity. Bacteria is a potential food source for nematodes and pathogenic to nematodes. Bacterial antagonists are now getting importance as potential biocontrol agents in modern agriculture due to gradual phasing out of chemical nematicides. In addition to trophic and pathogenic interactions, bacteria can serve as mutualists by aiding nematodes in development, defense, reproduction and nutrient acquisition. Furthermore, since many mechanistic aspects of nematode-bacterium interactions are conserved, their study can provide broader insights into other types of associations, including those relevant to plant and human diseases.

In Darwin’s and Mendel’s times, researchers investigated a wealth of organisms, chosen to solve particular problems for which they seemed especially well suited. Later, a focus on a few organisms, which are accessible to systematic genetic investigations, resulted in larger repertoires of methods and applications in these few species. Genetic animal model organisms with large research communities are the nematode Caenorhabditis elegans , the fly Drosophila melanogaster , the zebrafish Danio rerio, and the mouse Mus musculus. Due to their specific strengths, these model organisms have their strongest impacts in rather different areas of biology. C. elegans is unbeatable in the analysis of cell-to-cell contacts by saturation mutagenesis, as worms can be grown very fast in very high numbers. In Drosophila , a rich pattern is generated in the embryo as well as in adults that is used to unravel the underlying mechanisms of morphogenesis. The transparent larvae of zebrafish are uniquely suited to study organ development in a vertebrate, and the superb versatility of reverse genetics in the mouse made it the model organism to study human physiology and diseases. The combination of these models allows the in-depth genetic analysis of many fundamental biological processes using a plethora of different methods, finally providing many specific approaches to combat human diseases. The plant model Arabidopsis thaliana provides an understanding of many aspects of plant biology that might ultimately be useful for breeding crops.

Wheat is a crop of historical significance, as it marks the turning point of human civilization 10,000 years ago with its domestication. Due to the rapid increase in population, wheat production needs to be increased by 50% by 2050 and this growth will be mainly based on yield increases, as there is strong competition for scarce productive arable land from other sectors. This increasing demand can be further achieved using sustainable approaches including integrated disease pest management, adaption to warmer climates, less use of water resources and increased frequency of abiotic stress tolerances. Out of 200 diseases of wheat, 50 cause economic losses and are widely distributed. Each year, about 20% of wheat is lost due to diseases. Some major wheat diseases are rusts, smut, tan spot, spot blotch, fusarium head blight, common root rot, septoria blotch, powdery mildew, blast, and several viral, nematode, and bacterial diseases. These diseases badly impact the yield and cause mortality of the plants. This review focuses on important diseases of the wheat present in the United States, with comprehensive information of causal organism, economic damage, symptoms and host range, favorable conditions, and disease management strategies. Furthermore, major genetic and breeding efforts to control and manage these diseases are discussed. A detailed description of all the QTLs, genes reported and cloned for these diseases are provided in this review. This study will be of utmost importance to wheat breeding programs throughout the world to breed for resistance under changing environmental conditions.

Abstract Background Geminiviruses are circular, single-stranded viruses responsible for enormous crop loss worldwide. Rapid expansion of geminivirus diversity outweighs the continuous effort to control its spread. Geminiviruses channelize the host cell machinery in their favour by manipulating the gene expression, cell signalling, protein turnover, and metabolic reprogramming of plants. As a response to viral infection, plants have evolved to deploy various strategies to subvert the virus invasion and reinstate cellular homeostasis. Main body Numerous reports exploring various aspects of plant-geminivirus interaction portray the subtlety and flexibility of the host–pathogen dynamics. To leverage this pool of knowledge towards raising antiviral resistance in host plants, a comprehensive account of plant’s defence response against geminiviruses is required. This review discusses the current knowledge of plant’s antiviral responses exerted to geminivirus in the light of resistance mechanisms and the innate genetic factors contributing to the defence. We have revisited the defence pathways involving transcriptional and post-transcriptional gene silencing, ubiquitin-proteasomal degradation pathway, protein kinase signalling cascades, autophagy, and hypersensitive responses. In addition, geminivirus-induced phytohormonal fluctuations, the subsequent alterations in primary and secondary metabolites, and their impact on pathogenesis along with the recent advancements of CRISPR-Cas9 technique in generating the geminivirus resistance in plants have been discussed. Abstract Symptoms typical of phytoplasma infection such as phyllody, virescence, witches’ broom and yellowing were observed in 12 varieties of Chrysanthemum morifolium in floral nurseries and experimental fields at New Delhi, Karnataka, Maharashtra and Andhra Pradesh, India, during surveys made from 2015 to 2017. Disease incidence ranged from 15 to 30%. Phytoplasma presence was confirmed in all symptomatic chrysanthemum varieties by molecular identification assays. Sequence comparison, phylogenetic and in silico RFLP analyses of 16S rDNA sequences allowed the identification of the chrysanthemum infecting phytoplasma strains into different ribosomal groups and subgroups, namely 16SrI, 16SrII-D, 16SrVI-D and 16SrXIV. Detection of phytoplasma strains of 16SrII-D subgroup were also confirmed in symptomatic Chenopodium album and Parthenium hysterophorus plants grown in and around the surveyed chrysanthemum fields at New Delhi, whereas 16SrVI-D phytoplasma strains were detected in symptomatic Cannabis sativa weed and leafhopper Hishimonus phycitis individuals collected from the symptomatic chrysanthemum fields at New Delhi. This is the first report on the presence of 16SrVI and 16SrXIV groups of phytoplasmas in chrysanthemum plants. Studies on genetic diversity of phytoplasmas infecting the major chrysanthemum varieties in India and their epidemiological aspects had previously not been reported. The detection and identification of phytoplasmas in different chrysanthemum varieties could contribute to increase the awareness among farmers in the management of these diseases.

Genetic modification is one possible strategy to generate bananas (Musa spp.) with resistance to the soil-borne pathogen causing Fusarium wilt. The availability of banana root-specific promoters to target transgene expression to the sites of infection would be beneficial. We have assessed 18 promoter sequences derived from a range of plant species for their expression profiles in banana tissues to identify those with root-specific activity. Promoter sequences were isolated and fused to the β-glucuronidase (GUS) gene to assess their expression levels and tissue specificity in both banana and the model plant tobacco. Two heterologous promoters conferring high root expression levels in banana were identified, including a β‐glucosidase 1 (GLU1) promoter from maize and the RB7-type tonoplast intrinsic protein (TIP)-2 promoter from strawberry. Further, a novel Musa TIP2-2 promoter sequence was isolated and characterized which, when fused to the GUS gene, conferred very high GUS expression levels in banana roots. These promoters will expand the options for the control of gene expression in genetically modified bananas, providing a tool to develop plants with resistance not only to soil-borne diseases such as Fusarium wilt, but also for the improvement of other traits, such as nematode resistance, nutrition or abiotic stress resistance.

Onion is cultivated worldwide for its bulbs, but production is threatened by pathogens and pests. Three distinct diseases of onion are caused by species that belong to the fungal genus Botrytis. Leaf blight is a well-known foliar disease caused by B. squamosa that can cause serious yield losses. Neck rot is a post-harvest disease that manifests in bulbs after storage and to which three species are associated with; B. aclada, B. allii and B. byssoidea. The symptomless infection of onion plants in the field make it difficult to predict the incidence of neck rot in storage although progression on the detection of latent infection has been made. In onion cultivation for seed production, blighting of the inflorescence is caused by all four onion-specific Botrytis species plus the broad host range pathogen B. cinerea. Flower blight can lead to heavily reduced seed yield and contaminated seed. In this review, the long history of Botrytis diseases of onion is discussed, as well as recent and future approaches to acquire a better understanding of the biology and ecology of Botrytis spp. pathogenic on onion. New fundamental insights in the genetic, biochemical and physiological aspects of Botrytis-onion interactions are essential to improve the breeding of Botrytis-resistant onion cultivars.

Circadian rhythms represent an adaptive feature, ubiquitously found in nature, which grants living beings the ability to anticipate daily variations in their environment. They have been found in a multitude of organisms, ranging from bacteria to fungi, plants, and animals. Circadian rhythms are generated by endogenous clocks that can be entrained daily by environmental cycles such as light and temperature. The molecular machinery of circadian clocks includes a transcriptional-translational feedback loop that takes approximately 24 h to complete. Drosophila melanogaster has been a model organism of choice to understand the molecular basis of circadian clocks. However, alternative animal models are also being adopted, each offering their respective experimental advantages. The nematode Caenorhabditis elegans provides an excellent model for genetics and neuro-behavioral studies, which thanks to its ease of use and manipulation, as well as availability of genetic data and mutant strains, is currently used as a novel model for circadian research. Here, we aim to evaluate C. elegans as a model for chronobiological studies, focusing on its strengths and weaknesses while reviewing the available literature. Possible zeitgebers (including light and temperature) are also discussed. Determining the molecular bases and the neural circuitry involved in the central pacemaker of the C. elegans’ clock will contribute to the understanding of its circadian system, becoming a novel model organism for the study of diseases due to alterations of the circadian cycle.

In this scenario, the extensive exploration of available wild, resistant or susceptible germplasms and unraveling the genetic diversity remains vital for breeding programs. The dawn of next-generation sequencing technologies and omics approaches has accelerated plant breeding by providing the genome sequences and transcriptomes of several plants. The availability of decoded plant genomes offers an opportunity at a glance to identify candidate genes, quantitative trait loci (QTLs), molecular markers, and genome-wide association studies that can potentially aid in high throughput marker-assisted breeding. In recent years genomics is coupled with marker-assisted breeding to unravel the mechanisms to harness better better crop yield and quality. In this review, we discuss the aspects of marker-assisted breeding and recent perspectives of breeding approaches in the era of genomics, bioinformatics, high-tech phonemics, genome editing, and new plant breeding technologies for crop improvement. <p>Tomato plants (<em>Solanum lycopersicum</em>) are susceptible to the infection by diverse pathogens that cause devastating diseases such as vascular wilt, which causes great losses at the production level. The fungus <em>Fusarium oxysporum</em> f. sp. <em>lycopersici</em> (<em>Fol</em>) is one of the etiologic agents of this disease and its control lies in the use of synthetic chemicals which generate a negative impact in both health and the environment; thus, it is necessary to implement biological control as a healthier and more efficient alternative. The fungus <em>Trichoderma</em> spp. is a favorable option to be employed as a biocontroller against this pathogen thanks to its antagonist mechanisms, determined by metabolic and genetic characteristics. On the one hand, for <em>Fol</em> it is indispensable the activation of signaling routes such as MAPK Fmk1, MAPK Mpk1 y HOG, while <em>Trichoderma</em> spp. uses effectors involved in the interaction with the plant such as proteins, enzymes and secondary metabolites that also strengthen its immune response against infection, determined by both Pathogen Associated Molecular Patterns (PAMP) and effectors. Therefore, this article makes a review about the mentioned characteristics and suggests a greater application of tools and molecular markers for the management of this disease.</p>

This special issue aims to bring together knowledge related to research in the broad scientific field of plants. The aspects of medicinal and aromatic plants (MAPs) and plants in general covered include the study of the biological and chemical activities of extracts rich in phytochemicals, essential oils and secondary metabolites by in vitro and in vivo tests, ethnobotanical and ethnopharmacological studies, biodiversity, plants morphological and physiological characterizations. MAPs are defined as all raw organic materials ‎‎(trees, shrubs and herbs) whose active elements of their respective organs (fruits, ‎flowers, leaves, roots, stems, buds, bulbs) are likely to be used.‎ The range of plants applications is vast, since it affects the ‎medical sector (pharmaceutical industry, allopathy, phytotherapy, homeopathy, ‎aromatherapy), that of cosmetics and perfumes, chemistry (detergents, dyes, ‎varnishes, fireworks, etc.) and the agro-food sector (minimally processed products ‎such as infusion plants, spices and dry herbs, etc.).‎Ethnobotanical research estimates there are approximately 50 000 to 70 000 plant ‎species used in traditional and modern herbal medicine around the world (Schippmann et al., 2006). The total amount of MAPs used by the pharmaceutical, ‎cosmetic, chemical and food industries is very difficult to estimate. Globally, more ‎than 35 000 are used by the pharmaceutical, chemical or cosmetic industries according ‎to professionals. There is growing interest by scientists and the World Health Organization in medicinal ‎plants in many countries, especially in developing countries where access to modern ‎health care is limited. This action involves evaluating their efficacy and harmlessness, ‎and ensuring the safety and standardization of their use. ‎ Plants in general arouse a growing interest of the largest research centres ‎at the international level and are increasingly sought after by the agro-forestry and pharmaceutical ‎industries at the international, regional and national levels and therefore offer real ‎opportunities for socio-economic development of countries.‎Algeria, due to its geographical location, benefits from a very diverse climate and ‎flora. The plants grow in abundance in the coastal, mountainous and also Saharan ‎regions, many of which are renowned for their agro-forestry importance, medicinal virtue and used in traditional ‎medicine in health care (human and animal) and for culinary and cosmetic purposes.‎ These plants are potential natural remedies that can be used as a curative and ‎preventive therapeutic means, but this practice remains limited to patients and ‎herbalists who still have this empirical know-how.‎ The richness of the Algerian flora is therefore indisputable. It contains a large number ‎of species classified according to their degree of rarity: 289 fairly rare species, 647 rare ‎species, 640 very rare species, 35 extremely rare species and 168 endemic species (FAO, 2012). This situation requires a detailed inventory, a morphobiometric, biochemical and genetic characterization for an efficient management of this potential. The MAPs most in demand from herbalists in Algeria are most ‎often spontaneous (wild) in nature, which makes their quantification difficult.‎ The actors involved in the plants sector are very numerous and diversified and they attached ‎to several departments (agriculture, forestry, research, industry, commerce, finance, etc.).‎Algeria has nearly 2 689 herbalists registered with the National Trade Register Center ‎‎(CNRC), with "trader" status. These professionals are very active in this sector, they ‎seek to source and supply their customers. At the interface between collection and ‎distribution, herbalists have a central function in the organization of medicinal plants ‎on the Algerian domestic market. ‎ The informal is very present. In most cases, the plants sold are not subject to quality ‎controls. Some of them can be dangerous even if they are naturally extracted; they can ‎represent a real danger for the patient, as they may contain toxic components ‎unknown to the seller and to the patient. Out of naivety, ignorance or commercial ‎calculation, herbal sellers claim to have helped cure some patients of their diseases ‎classified as incurable, including diseases such as cancer and neurological diseases (Ilbert et al., 2016).

ABSTRACT Plant parasitic nematodes such as Meloidogyne incognita have a complex life cycle, occurring sequentially in various niches of the root and rhizosphere. They are known to form a range of interactions with bacteria and other microorganisms that can affect their densities and virulence. High-throughput sequencing can reveal these interactions in high temporal and geographic resolutions, although thus far we have only scratched the surface. In this study, we have carried out a longitudinal sampling scheme, repeatedly collecting rhizosphere soil, roots, galls, and second-stage juveniles from 20 plants to provide a high-resolution view of bacterial succession in these niches, using 16S rRNA metabarcoding. Our findings indicate that a structured community develops in the root, in which gall communities diverge from root segments lacking a gall, and that this structure is maintained throughout the crop season. We describe the successional process leading toward this structure, which is driven by interactions with the nematode and later by an increase in bacteria often found in hypoxic and anaerobic environments. We present evidence that this structure may play a role in the nematode’s chemotaxis toward uninfected root segments. Finally, we describe the J2 epibiotic microenvironment as ecologically deterministic, in part, due to the active bacterial attraction of second-stage juveniles. IMPORTANCE The study of high-resolution successional processes within tightly linked microniches is rare. Using the power and relatively low cost of metabarcoding, we describe the bacterial succession and community structure in roots infected with root-knot nematodes and in the nematodes themselves. We reveal separate successional processes in galls and adjacent non-gall root sections, which are driven by the nematode’s life cycle and the progression of the crop season. With their relatively low genetic diversity, large geographic range, spatially complex life cycle, and the simplified agricultural ecosystems they occupy, root-knot nematodes can serve as a model organism for terrestrial holobiont ecology. This perspective can improve our understanding of the temporal and spatial aspects of biological control efficacy.

Today, the use of injections of drugs of biological origin is a common practice for the treatment of chronic diseases, but the injection procedure itself is often associated with painful perceptions, and the field of injection is limited. Modern advanced therapies are genetic, electronic and cellular therapies that can meet the needs of caregivers, as well as the patients themselves. It is well known that digital processes in healthcare are currently undergoing intensive development, and in this context, innovative bioelectronic devices are being used to implement new strategies for personalized injection therapy. This paper examines aspects of the development of innovative methods of drug delivery to the patient's body, such as optogenetic therapy, optogenetic methods, and genetic therapy. For these purposes, bioelectronic implants are used today, in which electronics and optogenetics interact with the help of light, transmitting a certain induction signal to each other. By controlling the synthetic optogenetic pathway in the cell, drugs can be delivered to a particular organ within a clearly limited space or time. Present technology is revolutionary in medicine, as it can replace a traditional syringe, increasing the accuracy of dosage and time of drug administration, while reducing the involvement of the human factor to a minimum. The technology under consideration also acts as minimally invasive, which minimizes the patient's discomfort when taking medications.

Abstract:
Strawberry (Fragaria x ananassa Duch.) is one of the most consumed fruits from the Rosaceae family in Brazil. It is cultivated on ~4,500 ha in all regions of the country, with a wide range of fungi causing leaf diseases. Strawberry leaves (cv. San Andreas) with symptoms of eyespot usually related to Mycosphaerella fragariae (Ramularia tulasnei) (Mazaro et al. 2006) were observed in April 2019, in a field located in Santa Maria, Rio Grande do Sul state, Brazil (29°43'19.6"S / 53°43'04.6"W). However, the symptoms slightly differed from Mycospherella blight as well as the fungi structures observed under an optical microscope. The symptomatic leaves had small circular spots, initially reddish-purple to dark brown color, which later increased in size. The center of the lesion was grayish pale, where dark brown conidiophores were found over it. The symptom occurred in 50% of the leaves of all plants during the crop season. To identify the causal agent, symptomatic leaves were collected and superficially disinfected with 70% ethanol for 30 s followed by 0.5% bleach for 30 s and then washed in sterile water. The leaves were placed in germination boxes with a moistened paper filter and incubated at 25 ºC and 12 h photoperiod for four days. After the formation of the fungal structures, the fungi was isolated in potato dextrose agar (PDA) medium and kept for 20 days at 25 ºC. Colonies were dark brown with pale pink aerial mycelium. The conidia were hyaline, single- to multi-septated with a filiform shape, straight or slightly curved. The fasciculate conidiophores on the stroma measured 20 to 70 x 2.5 to 3.5 µm. Conidia average measurement was 10.2 to 82.4 x 4. to 8.8 µm, similar to morphological characteristics of the genus Cercospora. The isolate named M6 was deposited in the Brazilian National System of Genetic Registration (SISGEN) no A57AE19. The DNA was extracted from a monosporic culture of M6 in PDA plates, which presented all typical aspects previously described. To confirm the identity, the PCR amplification was carried out for internal transcribed spacer (ITS) region ITS 1-5.8S - ITS 2 and Calmodulin region (CAL) genes (Acqua et al, 2011). The ITS (No. MZ713249) and CAL (No. MZ713254) sequences were deposited in the Genbank database. BLASTn analysis revealed the isolate was 99% similar to Cercospora cf. malloti which access codes in GenBank are KT 193689 and KT 193738 (Nguanhom et al. 2015). A pathogenicity test was performed to fulfill Koch's postulates. Strawberry plants with healthy leaves (60 days old), were inoculated by spraying a conidial suspension (1 × 105 conidia/ml). Control plants were sprayed with sterile water. The plants were incubated in a greenhouse at 28 ºC for 12 h photoperiod at 80% relative humidity under drip irrigation keeping at least 60% of field capacity. Six plants were used as replicates. Twenty days after inoculation, small circular reddish-purple spots were found, which later increased in size. Ten days later, the spots presented dark brown conidiophores over the center of the lesion. The symptoms described above were observed in all inoculated leaves. The control plants remained symptom-free. The pathogen was then re-isolated from the lesions and identified as C. cf. malloti based on the morphological aspects described above on PDA. In Thailand C. cf. malloti is reported on crops like Brassica alboglabra (cabbage) (Nguanhom et al. 2015), but not in strawberries. To our knowledge, this is the first report of C. cf. malloti pathogenic to strawberries in Brazil.

AbstractIn the last three decades, researchers have utilized genome engineering to alter the DNA sequence in the living cells of a plethora of organisms, ranging from plants, fishes, mice, to even humans. This has been conventionally achieved by using methodologies such as single nucleotide insertion/deletion in coding sequences, exon(s) deletion, mutations in the promoter region, introducing stop codon for protein truncation, and addition of foreign DNA for functional elucidation of genes. However, recent years have witnessed the advent of novel techniques that use programmable site-specific nucleases like CRISPR/Cas9, TALENs, ZFNs, Cre/loxP system, and gene trapping. These have revolutionized the field of experimental transgenesis as well as contributed to the existing knowledge base of classical genetics and gene mapping. Yet there are certain experimental/technological barriers that we have been unable to cross while creating genetically modified organisms. Firstly, while interfering with coding strands, we inadvertently change introns, antisense strands, and other non-coding elements of the gene and genome that play integral roles in the determination of cellular phenotype. These unintended modifications become critical because introns and other non-coding elements, although traditionally regarded as “junk DNA,” have been found to play a major regulatory role in genetic pathways of several crucial cellular processes, development, homeostasis, and diseases. Secondly, post-insertion of transgene, non-coding RNAs are generated by host organism against the inserted foreign DNA or from the inserted transgene/construct against the host genes. The potential contribution of these non-coding RNAs to the resulting phenotype has not been considered. We aim to draw attention to these inherent flaws in the transgenic technology being employed to generate mutant mice and other model organisms. By overlooking these aspects of the whole gene and genetic makeup, perhaps our current understanding of gene function remains incomplete. Thus, it becomes important that, while using genetic engineering techniques to generate a mutant organism for a particular gene, we should carefully consider all the possible elements that may play a potential role in the resulting phenotype. This perspective highlights the commonly used mouse strains and the most probable associated complexities that have not been considered previously, resulting in possible limitations in the currently utilized transgenic technology. This work also warrants the use of already established mouse lines in further research.

The rationals for including (e)DNA-based tools into the survey's program were that i) many hard-to-identify organism groups can be assessed down to their species level, ii) taxonomic information can be unlocked even in cases where morphotaxonomic knowledge and expertise are limited, iii) all developmental stages, cryptic species and indeterminable sexes can be potentially identified, iv) taxalists relying on sequence-based information are reproducible and comparable in space and time (aspects, which are particularly important for a longitudinal survey involving many countries), and, v) this additional line of taxonomic evidence will help to draw a more precise and comprehensive picture of the Danubian biota. The target groups were assessed using group-specific (e)DNA-based metabarcoding approaches (for fish: Pont et al. 2021; macrozoobenthos: Beermann et al. 2021; phytobenthos: Zimmermann et al. 2021; sediment community: Cordier et al. 2021). The coverage of barcode libraries for Danubian biota were checked prior to conducting the metabarcoding approaches. Coverage values based on JDS3 outcomes were &gt;90% for fish (12S, but depending on reference database), 84% for macrozoobenthos (COI) and 69% and 88% for all, respectively, only abundant phytobenthos species (18S + rbcL), so that (e)DNA-based approaches were expected to be implemented effectively from this perspective (Weigand and Astrin 2021). Although still a certain degree of methodological variation exists, the outcomes clearly demonstrate the huge potential of (e)DNA-based approaches for complementary biodiversity and ecological status class assessments: eDNA water analysis of fish revealed most of the taxa also detected by the traditional fish survey, but was particularly effective in detecting hard-to-capture benthic taxa (including endangered sturgeon species) and fish traces originating from waste water treatment plants (Pont et al. 2021). Many of the traditionally assigned macrozoobenthos species were detected by DNA metabarcoding as well, but sequence data allowed to add a plethora of new chironomid and oligochaete species to the taxalist. Molecular ecological status class assessments based on presence-absence values of macrozoobenthos species were largely congruent to traditional abundance or presence-absence-based outcomes (Beermann et al. 2021, Weigand 2021). Although traditional light microscopy, which is based on identifying phytobenthos species by their frostules, revealed a higher number of diatom species, the molecular assessment detected much more taxa (i.e. MOTUs), which await species-level taxonomic annotation in the future (Zimmermann et al. 2021). Metabarcoding of the sediment community was particularly effective to assess meiofaunal species and allowed the molecular inference of fine sediment quality based on local community structures of vulnerable nematode species (Cordier et al. 2021). Finally, all (e)DNA-based taxalists were compiled to inform invasive alien species detection in the Danube River Basin. However, despite their promising performance and large coherence with traditional outcomes during JDS4, the full potential of (e)DNA-based approaches in the context of larger environmental surveys might be further released by developing and curating catchment-specific DNA barcode reference libraries, focussing on a small(er) set of standardized (e)DNA-based approaches, integrating genetic diversity (and spatiotemporal changes thereof) in ecological status class assessments, installing a dense, large-scale environmental DNA-based screening, based on which traditional surveys can be performed at conspicuous sites, and, educating and training national authorities in state-of-the-art molecular tools. developing and curating catchment-specific DNA barcode reference libraries, focussing on a small(er) set of standardized (e)DNA-based approaches, integrating genetic diversity (and spatiotemporal changes thereof) in ecological status class assessments, installing a dense, large-scale environmental DNA-based screening, based on which traditional surveys can be performed at conspicuous sites, and, educating and training national authorities in state-of-the-art molecular tools. Already today, (e)DNA-based methods can be seen as an effective and complementary tool to provide consolidated results for biodiversity and ecological status class assessments in a highly integrative and international setup, as pursued during JDS4.

A long-term stationary experiment was conducted to examine for the purpose nine field crop rotation patterns different in the number of fields (four to six), set of crops, and their succession in crop rotation. The respective shares of legumes, oilseeds, and cereals in the cropping pattern were 17 to 33, 17 to 40, and 50 to 67 %. It has been established that in case of no-till field crop cultivation the economic efficiency of plant production depends on the set of crops and their succession in rotation. The most economically efficient type of crop rotation is the soya-winter wheat-peas-winter wheat-sunflower-corn six-field rotation with two fields of legumes: in this rotation 1 ha of crop rotation area yields 3 850 grain units per ha at a grain unit prime cost of 5.46 roubles; the plant production output return and profitability were 20,888 roubles per ha and 113 %, respectively. The high production profitabilities provided by the soya-winter wheat-sunflower four-field and the soya-winter-wheat-sunflower-corn-winter wheat five-field crop rotation are 108.7 and 106.2 %, respectively. The inclusion of winter wheat in crop rotation for two years in a row reduces the second winter wheat crop yield by 80 to 100 %, which means a certain reduction in the grain unit harvesting rate to 3.48-3.57 thousands per ha of rotation area and cuts the production profitability down to 84.4-92.3 %. The relevance of the topic has to do with an increased global interest in this region. The paper is aimed at considering the basic principles of developing and the procedure of testing seismic instruments for use at ultralow climatic temperatures. In this paper the indicated issue is considered through the example of a seismic module designed for petroleum and gas exploration by passive seismoacoustic methods. The seismic module is a direct-burial portable unit of around 5 kg in weight, designed to continuously measure and record microseismic triaxial orthogonal (ZNE) noise in a range from 0.1 to 45 Hz during several days in autonomous mode. The functional chart of designing the seismic module was considered, and concrete conclusions were made for choosing the necessary components to meet the ultralow-temperature operational requirements. The conclusions made served for developing appropriate seismic module. In this case, the components and tools used included a SAFT MP 176065 xc low-temperature lithium cell, industrial-spec electronic component parts, a Zhaofeng Geophysical ZF-4.5 Chinese primary electrodynamic seismic sensor, housing seal parts made of frost-resistant silicone materials, and finely dispersed silica gel used as water-retaining sorbent to avoid condensation in the housing. The paper also describes a procedure of low-temperature collation tests at the lab using a New Brunswick Scientific freezing plant. The test results proved the operability of the developed equipment at ultralow temperatures down to -55°C. In addition, tests were conducted at low microseismic noises in the actual Arctic environment. The possibility to detect signals in a range from 1 to 10 Hz at the level close to the NLNM limit (the Peterson model) has been confirmed, which allows monitoring and exploring petroleum and gas deposits by passive methods. As revealed by this study, the suggested approaches are efficient in developing high-precision mobile seismic instruments for use at ultralow climatic temperatures. The solution of the considered instrumentation and methodical issues is of great practical significance as a constituent of the generic problem of Arctic exploration. The article analyzes the results of treatment of patients with excess length of the second metatarsal bones that underwent osteotomy with and without osteosynthesis. The results of treatment of patients who underwent metatarsal shortening due to classical Weil-osteotomy with and without osteosynthesis were analyzed. The first group consisted of 34 patients. They underwent classical Weil osteotomy. The second group included 44 patients in whomosteotomy of the second metatarsal bone were not by the screw. When studying the results of the treatment in the immediate postoperative period, weeks 6, 12, slightly better results were observed in patients of the first group, while one year after surgical treatment the results in both groups were comparable. One year after surgical treatment, there were 2.9% (1 patient) of unsatisfactory results in the first group and 4.5% (2 patients) in the second group. Considering the comparability of the results of treatment in remote postoperative period, the choice of concrete method remains with the operating surgeon. Methods. A prospective 3-year study of 86 healthy dogs (aged 1-7 yrs) of different breeds and of both sexes. Echo homogeneity and echogenicity of the stomach and intestines wall were determined by the method of Silina, T.L., et al. (2010) in absolute values ​​of average brightness levels of ultrasound image pixels using the 8-bit scale with 256 shades of gray. Results. Quantitative echogenicity indicators of the stomach and the small intestine wall in dogs were determined. Based on the numerical values ​​characterizing echogenicity distribution in each layer of a separate structure of the digestive system, the coefficient of gastric echogenicity is determined as 1:2.4:1.1 (mucosa/submucosa/muscle layers, respectively), the coefficient of duodenum and jejunum echogenicity is determined as 1:3.5:2 and that of ileum is 1:1.8:1. Datieva, DOI: https://doi.org/10.26782/jmcms.spl.10/2020.06.00046 Abstract: The aim of this work was to assess the functional reserves of the body to quantify individual health; adaptation, psychophysiological characteristics of the health quality of medical students in different seasons of the year. When studying the temporal organization of physiological functions, the rhythm parameters of physiological functions were determined, followed by processing the results using the Cosinor Analysis program, which reveals rhythms with an unknown period for unequal observations, evaluates 5 parameters of sinusoidal rhythms (mesor, amplitude, acrophase, period, reliability). The essence of desynchronization is the mismatch of circadian rhythms among themselves or destruction of the rhythms architectonics (instability of acrophases or their disappearance). Desynchronization with respect to the rhythmic structure of the body is of a disregulatory nature, most pronounced in pathological desynchronization. High neurotism, increased anxiety reinforces the tendency to internal desynchronization, which increases with stress. During examination stress, students experience a decrease in the stability of the temporary organization of the biosystem and the tension of adaptive mechanisms develops, which affects attention, mental performance and the quality of adaptation to the educational process. Time is shortened and the amplitude of the “initial minute” decreases, personal and situational anxiety develops, and the level of psychophysiological adaptation decreases. The results of the work are priority because they can be used in assessing quality and level of health. Tsvetkov, DOI: https://doi.org/10.26782/jmcms.spl.10/2020.06.00047 Abstract: The present study of comparison methods based on the triadic model introduces the following concepts: the relation of comparability and the relation of comparison, and object comparison and attributive comparison. The difference between active and passive qualitative comparison is shown, two triadic models of passive and active comparison and models for comparing two and three objects are described. Triadic comparison models are proposed as an alternative to dyadic comparison models. Comparison allows finding the common and the different; this approach is proposed for the analysis of the nomothetic and ideographic method of obtaining knowledge. The nomothetic method identifies and evaluates the general, while the ideographic method searches for unique in parameters and in combinations of parameters. Triadic comparison is used in systems and methods of argumentation, as well as in the analysis of consistency/inconsistency.
