Abstract:
In the hearts of patients bearing nebulette mutations, a severe general disorganization in cardiomyocytes of the extrasarcomeric desmin intermediate filament system is frequently observed. However, the molecular and functional relationship between the desmin cytoskeleton and nebulette-containing sarcomeres is still unclear. Here we report a high-affinity in vitro interaction between nebulette and desmin filaments. A major interaction site has been mapped to the desmin α-helical rod domain, indicating that the filament core is directly involved in the binding of nebulette. The disease-mutant desmin variants E245D and T453I exhibited increased binding affinity for nebulette, delayed filament assembly kinetics, and caused significant weakening of networks. In isolated chick cardiomyocytes and sections from canine heart, we revealed by ground-state depletion and confocal microscopies that module 5 of nebulette extends outward from Z-disk–associated desmin filaments toward the center of the sarcomere. Accordingly, in the myocardium of Des−/− mice, elevated levels of cardiac actin correlated with alterations in the distribution of nebulette. Our data suggest that a well-organized desmin network is required to accommodate an optimal conformation of nebulette on sarcomeres to bind and recruit cardiac α-actin. Hence we propose that nebulette acts in synergy with nebulin to reinforce and temporally fine-tune striated muscle relaxation–contraction cycles.

Abstract:
Zyxin is a versatile component of focal adhesions in eukaryotic cells. Here we describe a novel binding partner of zyxin, which we have named LIM-nebulette. LIM-nebulette is an alternative splice variant of the sarcomeric protein nebulette, which, in contrast to nebulette, is expressed in non-muscle cells. It displays a modular structure with an N-terminal LIM domain, three nebulin-like repeats, and a C-terminal SH3 domain and shows high similarity to another cytoskeletal protein, Lasp-1 (LIM and SH3 protein-1). Co-precipitation studies and results obtained with the two-hybrid system demonstrate that LIM-nebulette and Lasp-1 interact specifically with zyxin. Moreover, the SH3 domain from LIM-nebulette is both necessary and sufficient for zyxin binding. The SH3 domains from Lasp-1 and nebulin can also interact with zyxin, but the SH3 domains from more distantly related proteins such as vinexin and sorting nexin 9 do not. On the other hand, the binding site in zyxin is situated at the extreme N terminus as shown by site-directed mutagenesis. LIM-nebulette and Lasp-1 use the same linear binding motif. This motif shows some similarity to a class II binding site but does not contain the classical PXXP sequence. LIM-nebulette reveals a subcellular distribution at focal adhesions similar to Lasp-1. Thus, LIM-nebulette, Lasp-1, and zyxin may play an important role in the organization of focal adhesions.

BackgroundMaintenance of the intricate interdigitating morphology of podocytes is crucial for glomerular filtration. One of the key aspects of specialized podocyte morphology is the segregation and organization of distinct cytoskeletal filaments into different subcellular components, for which the exact mechanisms remain poorly understood.MethodsCells from rats, mice, and humans were used to describe the cytoskeletal configuration underlying podocyte structure. Screening the time-dependent proteomic changes in the rat puromycin aminonucleoside–induced nephropathy model correlated the actin-binding protein LIM-nebulette strongly with glomerular function. Single-cell RNA sequencing and immunogold labeling were used to determine Nebl expression specificity in podocytes. Automated high-content imaging, super-resolution microscopy, atomic force microscopy (AFM), live-cell imaging of calcium, and measurement of motility and adhesion dynamics characterized the physiologic role of LIM-nebulette in podocytes.ResultsNebl knockout mice have increased susceptibility to adriamycin-induced nephropathy and display morphologic, cytoskeletal, and focal adhesion abnormalities with altered calcium dynamics, motility, and Rho GTPase activity. LIM-nebulette expression is decreased in diabetic nephropathy and FSGS patients at both the transcript and protein level. In mice, rats, and humans, LIM-nebulette expression is localized to primary, secondary, and tertiary processes of podocytes, where it colocalizes with focal adhesions as well as with vimentin fibers. LIM-nebulette shRNA knockdown in immortalized human podocytes leads to dysregulation of vimentin filament organization and reduced cellular elasticity as measured by AFM indentation.ConclusionsLIM-nebulette is a multifunctional cytoskeletal protein that is critical in the maintenance of podocyte structural integrity through active reorganization of focal adhesions, the actin cytoskeleton, and intermediate filaments.

The Xin actin-binding repeat–containing proteins Xin and XIRP2 are exclusively expressed in striated muscle cells, where they are believed to play an important role in development. In adult muscle, both proteins are concentrated at attachment sites of myofibrils to the membrane. In contrast, during development they are localized to immature myofibrils together with their binding partner, filamin C, indicating an involvement of both proteins in myofibril assembly. We identify the SH3 domains of nebulin and nebulette as novel ligands of proline-rich regions of Xin and XIRP2. Precise binding motifs are mapped and shown to bind both SH3 domains with micromolar affinity. Cocrystallization of the nebulette SH3 domain with the interacting XIRP2 peptide PPPTLPKPKLPKH reveals selective interactions that conform to class II SH3 domain–binding peptides. Bimolecular fluorescence complementation experiments in cultured muscle cells indicate a temporally restricted interaction of Xin-repeat proteins with nebulin/nebulette during early stages of myofibril development that is lost upon further maturation. In mature myofibrils, this interaction is limited to longitudinally oriented structures associated with myofibril development and remodeling. These data provide new insights into the role of Xin actin-binding repeat–containing proteins (together with their interaction partners) in myofibril assembly and after muscle damage.

Abstract:
We describe here a novel sarcomeric 145-kD protein, myopalladin, which tethers together the COOH-terminal Src homology 3 domains of nebulin and nebulette with the EF hand motifs of α-actinin in vertebrate Z-lines. Myopalladin's nebulin/nebulette and α-actinin–binding sites are contained in two distinct regions within its COOH-terminal 90-kD domain. Both sites are highly homologous with those found in palladin, a protein described recently required for actin cytoskeletal assembly (Parast, M.M., and C.A. Otey. 2000. J. Cell Biol. 150:643–656). This suggests that palladin and myopalladin may have conserved roles in stress fiber and Z-line assembly. The NH2-terminal region of myopalladin specifically binds to the cardiac ankyrin repeat protein (CARP), a nuclear protein involved in control of muscle gene expression. Immunofluorescence and immunoelectron microscopy studies revealed that myopalladin also colocalized with CARP in the central I-band of striated muscle sarcomeres. Overexpression of myopalladin's NH2-terminal CARP-binding region in live cardiac myocytes resulted in severe disruption of all sarcomeric components studied, suggesting that the myopalladin–CARP complex in the central I-band may have an important regulatory role in maintaining sarcomeric integrity. Our data also suggest that myopalladin may link regulatory mechanisms involved in Z-line structure (via α-actinin and nebulin/nebulette) to those involved in muscle gene expression (via CARP).

We demonstrate that ablation of first nebulin-repeats of sarcomeric nebulette ( Nebl) causes diastolic dysfunction in Neblex3-/ex3- mice. Exercise-induced development of diastolic dysfunction, cardiac hypertrophy and ventricular dilation in knockouts. This was associated with sarcomere disturbance, intercalated disks disruption, and mitochondrial distortion upon stress and altered expression of genes involved in Nebl-based stress network. We demonstrate that G202R and A592 mutations alter actin and desmin expression causing disorganization of desmin filaments upon cyclic strain.

Abstract:
The sarcomeres are the smallest contractile units of striated muscle. It is formed by actin (thin) and myosin (thick) filaments that slide over each other during contraction and is stabilized by titin filaments, which acts as a spring during contraction. In the Z-line at the boundary between sarcomeres, actin and titin filaments are cross-linked and the contractile apparatus is connected with the cytoskeleton and extracellular matrix. The Z-line is important for efficient force generation and maintenance of cardiac structure, and is a focal point for mechanosensing and signal transduction. Consistently, mutations in many Z-line associated proteins have been associated with cardiomyopathies, including nebulette and its interaction partner myopalladin. On the other hand, the role of palladin (PALLD), a homologue of myopalladin (MYPN) associated with nebulette, in the heart has remained unknown. The aim of the present project was to investigate the function of nebulette and palladin in the heart. Nebulette is a 109 kDa modular protein composed of nebulin repeats and a C-terminal SH3 domain. In vitro studies have suggested a role of nebulette in stabilizing the thin filament, and missense mutations in the nebulette gene have been found to be causative for dilated cardiomyopathy and endocardial fibroelastosis in human and mice. However, the role of nebulette in vivo has remained unknown. To study the functional role of nebulette in the heart in vivo, we generated and studied nebulette knockout (nebl-/-) mice. Functional analyses revealed normal cardiac function of nebl-/- mice both at basal conditions and in response to transaortic constriction (TAC). Furthermore, histological, immunofluorescence, and Western blot analyses showed no cardiac abnormalities in nebl-/- mice. On the other hand, Z-line widening was observed from 5 months of age and the presence of chronic cardiac stress was suggested by the upregulation of cardiac stress responsive genes. Thus, nebulette may play an important role for Z-line integrity, while the absence of a functional phenotype of nebl-/- mice suggests that nebulette cardiomyopathy mutations have gain-of-function effects. PALLD, MYPN, and myotilin (MYOT) make up a small protein family of immunoglobulin-containing proteins in the sarcomeric Z-line associated with α-actinin and filamentous actin. MYPN and MYOT are expressed as single isoforms in striated muscle, while PALLD is ubiquitously expressed in several isoforms. The longest 200 kDa PALLD isoform is expressed predominantly in striated muscle and is highly homologous in structure to MYPN, suggesting a similar role of the two proteins in striated muscle. However, while mutations in the MYPN gene have been associated with dilated, hypertrophic, and restrictive cardiomyopathy, the role of PALLD in the heart has remained unknown. Since PALLD knockout mice are embryonic lethal, to study the role of PALLD in heart, we generated constitutive (cPKO) and inducible (cPKOi) cardiac specific PALLD knockout mice. cPKO mice showed normal cardiac function both at basal conditions and following TAC, while inducible knockout of PALLD in adult cPKOi mice resulted in progressive cardiac dilation and systolic dysfunction, associated with fibrosis, upregulation of markers of cardiac pathological remodeling, and ERK activation. Furthermore, PALLD was found to activate serum response factor (SRF) signaling and strongly increase MRTF-A-mediated activation of SRF, likely through its direct interaction MRTF-A and role in modulating actin dynamics. The development of a dilated cardiomyopathy in cPKOi mice induced at adult stage demonstrates that PALLD is essential for normal cardiac function and suggests that PALLD gene mutations may be causative for cardiomyopathy. The absence of a cardiac phenotype of cPKO mice may be due to compensatory mechanisms.

Depuis les années quatre-vingt-dix, en France, se développent des entreprises qui s'inscrivent dans le champ de la philosophie, de la « culture psychologique » et de la spiritualité, dont la particularité est de vouloir répondre à une « quête de sens » individualiste sortant manifestement du champ de la religion classique. Nous avons identifiées ces différentes entreprises comme alimentant une nébuleuse qualifiée de « psycho-philo-spirituelle ». Y participent des philosophes (A. Comte-Sponville, L. Ferry, R. Pol-Droit, M. Onfray), des revues (Actualité des Religions, Psychologies magazine), des centres spirituels (le « Forum 104 », l' « ashram » d'A. Desjardins, le « Centre Dürckheim »), dont les discours et les productions touchent un grand public cultivé. Les compositions de sens « psycho-philo-spirituelles » permettent de mesurer aujourd'hui des effets d'une sécularisation qui s'est radicalisée, notamment via le « désenchantement des institutions séculières » (J. Baubérot). Cette recherche dégage ainsi la constitution de nouveaux croires alimentés par des entreprises areligieuses (la philosophie et la psychologie « pratiques ») participant à la décomposition de la religion en ressources compréhensives dans une perspective immanente. La nébuleuse psycho-philo-spirituelle est ainsi un révélateur d'un double processus, avec d'un côté l'affirmation de valeurs produites par la culture individualiste entraînant l'éclatement des grands systèmes symboliques et de l'autre, une certaine homogénéisation des constructions du sens- s'opérant tout particulièrement via la généralisation d'une grille de lecture psychologique du monde et de l'existence.

Artemis (antimatter research trough the eart moon ion spectrometer cherche a detecter des antiprotons extragalactique de tres hautes energies (tev) dans le rayonnement cosmique grace au champ magnetique terrestre comme defecteur, la lune comme ecran selecteur d'ions, l'atmosphere comme calorimetre geant et la technique d'imagerie cerenkov pour l'observation des gerbes atmospheriques. Du fait de la presence de la lune, l'observation de la lumiere cerenkov doit se faire dans l'uv, ce qui a necessite le developpement d'une camera uv, unique au monde. Aucune imagerie de gerbes en uv n'avait ete faite auparavant. Le crabe, qui est devenu la source de reference pour l'astronomie gamma de tres haute energie, nous a permis justement de tester notre methode uv dans des conditions simples de faibles bruit lumineux. Notre mise en evidence d'un signal du crabe valide, d'une part, la methode d'imagerie cerenkov en uv et ouvre, d'autre part, un nouveau champ d'observation en astronomie gamma pour les nuits avec lune, offrant ainsi la possibilite d'observer toutes les nuits une source quelque soit la phase de la lune

Le travail de cette these a ete effectue, dans le cadre de l'astronomie gamma des tres hautes energies (>250 gev), aupres du telescope d'imagerie tcherenkov a bas seuil cat. Installe dans les pyrenees orientales. Cette these poursuit deux objectifs. Le premier consiste a etalonner le telescope et a en maitriser tous les aspects pouvant introduire des erreurs systematiques (mesure de l'efficacite de collection de lumiere du telescope, mesure et suivi du fond de ciel nocturne, etc. ). Plusieurs phenomenes ont ete utilises, en tant que faisceau-test, pour cet etalonnage : la lumiere tcherenkov des muons tombant dans le miroir et les photons gamma du noyau actif de galaxie markarian 501 detectes par cat. Le second objectif est l'analyse du spectre en energie des de la nebuleuse du crabe qui permet de nous comparer aux autres experiences. La selection des met en oeuvre une methode de maximum de vraisemblance, mis au point par s. Le bohec et b. Degrange en 1996, a partir des simulations. Les chiffres trouves sont compatibles avec ceux des autres experiences travaillant dans ce domaine (observatoire whipple en arizona et experience hegra aux canaries).

Bien que nee des les annees 50, ce n'est que tout recemment que l'astronomie gamma au sol a developpe des methodes qui en font aujourd'hui une discipline ayant un veritable potentiel astrophysique. Apres avoir resume l'etat de la discipline, et notamment les observations recentes sur la nebuleuse du crabe et markarian 421, nous decrivons les efforts qui ont porte en avant les techniques de l'imagerie cerenkov et de l'echantillonnage du front d'onde. Nous evoquons d'abord la technique de l'imagerie cerenkov et les premieres mesures faites sur la nebuleuse du crabe avec le grand telescope de 10 metres de la collaboration whipple; nous commentons les resultats obtenus dans l'observation d'autres sources possibles. La detection de la nebuleuse du crabe avec le telescope a echantillonnage du front d'onde asgat est ensuite discutee. Ces travaux ont conduit a l'etablissement de la nebuleuse du crabe comme source de reference dans la region du tev. Un point delicat qui necessite attention est celui d'une calibration suffisamment fiable des telescopes, qui conditionne la comparaison entre les differents resultats. A ce propos, nous commentons le desaccord entre deux mesures du spectre en energie de la nebuleuse du crabe dans la region du tev. Une methode de calibration basee sur la lumiere cerenkov emise par les muons proches est decrite en detail. Appliquee a tous les telescopes elle pourrait constituer la base d'une procedure de calibration commune

Cette these presente de nouveaux resultats sur la distribution et les proprietes du gaz moleculaire dans la galaxie d'andromede (m 31). Dans la premiere partie, nous nous sommes interesse specifiquement aux regions internes (r#g#a#l < 7 kpc), et avons montre qu'au moins certains nuages de ces regions contenaient vraisemblablement de grandes quantites de gaz moleculaire. Les nuages de ces regions apparaissent en outre beaucoup plus lisses et froids que les nuages moleculaires geants galactiques. Ces differences observationnelles pourraient neanmoins ne pas traduire de differences structurelles intrinseques, mais resulter seulement de conditions d'environnement differentes (faible flux uv et faible densite de rayons cosmiques). Le facteur de couverture du disque de m 31 par du gaz moleculaire dans les 7 kiloparsecs centraux semble de plus ne pas etre plus faible que celui des regions entre 7 et 12 kpc, contrairement a l'idee recue. La seconde partie presente une cartographie co complete de la moitie sud de m 31 faite a une resolution angulaire de 45". Le gaz moleculaire dans m 31 semble fortement confine a l'interieur de bras spiraux, dont deux (s3 et s4) sont particulierement proeminents. Ces bras correspondent a ceux definis par le gaz atomique, la poussiere et les etoiles jeunes, meme si a l'intersection de s4 et du grand axe de la galaxie, on constate un decalage de l'ordre d'un kiloparsec entre le bas defini par les indicateurs de milieu interstellaire froid et celui defini par les etoiles jeunes ; decalage qui pourrait etre du au passage d'une onde de densite spirale. Meme si m 31 est environ 6 fois moins brillante en co que la voie lactee, on constate qu'au dela de 8-9 kpc (le rayon solaire), les deux galaxies sont tres semblables dans leur contenu moleculaire. Cette cartographie a en outre revele la presence au centre de m 31 d'un disque moleculaire nucleaire tres similaire dans sa cinematique au disque nucleaire galactique.

Abstract:
Le réseau d'imageurs Cherenkov atmosphériques HESS (High Energy Stereoscopic System) est présenté. La simulation des gerbes atmosphériques et de l'instrument est détaillée et comparée aux données obtenues pendant les six premiers mois de fonctionnement. Après la présentation des méthodes d'étalonnage et d'extraction du signal gamma, ces techniques sont appliquées aux données prises sur la nébuleuse du Crabe. On obtient un signal significatif permettant d'estimer un flux différentiel à 1 TeV en accord avec les résultats obtenus par d'autres expériences dans le même domaine en énergie. Ces résultats sont exploités pour estimer le potentiel de détection de HESS au signal gamma d'annihilation de neutralinos en provenance du centre galactique et de l'amas globulaire Omega du Centaure. Dans cette étude, une modélisation des halos de matière noire est confrontée aux observations astrophysiques.

Etude d'etoiles de type a-f au voisinage solaire, de noyaux brillants de nebuleuses planetaires et des sous naines les plus brillantes. On s'interesse plus specialement a leur repartition galactique: jeune disque, vieux disque, halo. Les relations theoriques entre binarite, rotation, pulsation et structure d'enveloppe chez les etoiles a-f ont ete confirmees par analyse statistique. Un noyau a spectre composite a ete decouvert dans la nebuleuse planetaire lts et des variations d'eclat periodiques decelees dans le noyau abell 35. Six nouvelles binaires spectroscopiques ont par ailleurs ete decouvertes parmi les sous naines brillantes du halo

L'observation d'enveloppes froides autour d'objets evolues fait apparaitre une classe d'etoiles peu massives en fin d'evolution, les "pseudo supergeantes f"; une nebuleuse planetaire tres peu evoluee et tres riche en co et la nebuleuse du "lion givre" dont le spectre ir est domine par l'emission de la glace cristalline
