Abstract:
This thesis evaluates the National Library of Wales (NLW) from conception to realisation. It adds to the limited existing work on the library by positioning it within the political and cultural environment from which it emerged, posing new questions about its relationship with Welsh national identity. Although there is some work which looks at national identity in relation to national library projects, nothing of this type and depth has been done before, and not in relation to Wales. The first section investigates the early calls for a NLW and explores why the campaign made significant progress from the 1890s. The focus then shifts to an interrogation of the British government’s decision to locate the library in Aberystwyth, challenging the important role previously assigned to the campaign group’s application. Chapter three analyses the library building fund’s subscription drive and argues that the campaign was not the result of a mass movement, but orchestrated by a group of elites. It evaluates the decision made by the library campaign committee to evoke momentary patriotic sympathies among potential subscribers, rather than extolling the virtues of the library as a long-term educational resource. Chapter four is an analysis of the library’s buying policy and argues that an overrepresentation from key academic disciplines on the committees may have created a bias towards certain subjects. The final chapter compares this vision with how the library’s services were utilised by three user groups: reading room users, tutorial class attendees and Ruhleben camp internees. Reading room usage generally concurred with the founders’ overall vision for the library. In contrast, remote users attending tutorial classes were given the opportunity to curate their own reading which resulted in a more wide-ranging collection. The thesis concludes with an assessment of the library’s development and role as a key element of Welsh nation-building at the beginning of the twentieth century.

Abstract:
Traces the history of organized document collections of India from very ancient times, the contribution of Europeans especially the British rulers of India for systematization and conservation of such collections, and the origins of public library systems that culminated in the establishment of the National Library of India. Critically evaluates the National Library system of India as well as its present set up, resources and services. The paper argues that the main characteristics of a national library, which distinctly delineate it from other types of libraries is its specialization in a particular geographical area for its content. It should achieve maximum reliability of the collection in that mandatory area. The paper points out that in coverage and reliability in the mandatory areas as well as in extending services to the expected users including those living in remote villages; Indian National Library has failed. The study suggests decentralization of the resources by physically spreading it among the regions of concerned languages, establishment of subject specific divisions and other measure that can enable the National Library of India to fulfill its objectives.

Abstract:

With the advancement of information technology, national library websites have become an essential vehicle for their users to access official information and library resources. They serve the general public from different countries, age groups, ethnicities, and educational levels. The purpose of this dissertation research is to examine the usability, user experience (UX), and information architecture (IA) of national library websites in different countries. Focal areas of investigation include: (1) the primary content elements on national library websites' homepages, (2) the extent to which various quantitative measures of heuristic evaluation and IA assessment overlap with those measures from task-based usability testing, (3) the extent of the impact of cultural and national origins on participants’ assessment of usability of national libraries’ websites. The research design of this dissertation study features (1) a content inventory of 28 national library websites, (2) a web IA assessment and a usability evaluation of five national library websites, and (3) usability tests for three national library websites involving 30 participants. The key findings of this dissertation research help to establish a list of common content elements on the homepages of national library websites worldwide and provide a concrete, practical, and feasible procedure to evaluate such websites. The results show statistically significant differences in task performance with the use of national library websites between different groups of users, such as those from different genders and educational backgrounds. Significant correlations were found between the overall participant satisfaction of a national library website and multiple variables such as the content, visual design, and information architecture, as well as between heuristic evaluation scores and participants’ ratings of some aspects of the websites.

Based on the specific results from various phases of the research, this dissertation presented detailed recommendations that could help to make national library websites more usable for all its users, including the first-time users across different genders, age groups, and educational backgrounds. Furthermore, in order to achieve a high level of satisfaction, it is recommended that national libraries focus on six factors affecting participants’ satisfaction: the quality of information, the trustworthiness of the content, the credibility of the content producers/providers, the website’s overall structure, the design, and the aesthetics of the website.

Another significant contribution of this dissertation research is its use of various methods used to evaluate national library websites and its integration of the results of the different methods to obtain a broader and more comprehensive understanding of these findings. Future UX research on national library websites could expand the research by incorporating the methodology used in this research, testing a greater number of national library websites around the world, and involving users from all walks of life.

Abstract:
Introdução: A fração de plaquetas imaturas (IPF) é um parâmetro ainda pouco explorado na rotina clínica e sua determinação laboratorial através de citometria de fluxo tem sido pesquisado como um marcador de função medular. As plaquetas reticuladas (PR) representam plaquetas jovens recentemente liberadas pela medula óssea, e são caracterizadas pelo alto conteúdo de ácido ribonucleico (RNA) em seu citoplasma. Objetivo: O objetivo deste trabalho foi pesquisar a utilidade diagnóstica das PRs através do parâmetro IPF (fração de plaquetas imaturas) na rotina médica e suas limitações, além das perspectivas futuras para seu uso. Material e métodos: Foi realizada uma pesquisa bibliográfica em bases de dados confiáveis, Medline, PubMed (US. National Library of Medicine National Institutes of Health, USA), SciELO Brazil (Scientific Eletronic Library Online) e BIREME (Biblioteca Regional de Medicina, Centro Latino-Americano e do Caribe de Informação em Ciências da Saúde) utilizando os descritores “Platelet índices”, “Immature platelet fraction” e “Reticulated platelets”. Resultados: As plaquetas reticuladas são um importante indicador da etiologia da trombocitopenia em diversos quadros clínicos, diferenciando os casos de consumo periférico plaquetário de uma falha na produção na medula óssea. As plaquetas imaturas permanecem na circulação por 24 a 36 horas, quando progressivamente têm seu RNA degradado e seu volume diminuído passando ao estado da plaqueta madura. Portanto, assim como os reticulócitos para os eritrócitos, a quantificação de plaquetas imaturas é capaz de trazer informações precoces em relação a produção de plaquetas na medula óssea. Além disto, alguns estudos apontam que o IPF pode ser útil no diagnóstico precoce de sepse bem como no acompanhamento de pacientes transplantados. Conclusão: Através desta revisão bibliográfica, conclui-se que a fração de plaquetas imaturas é um promissor parâmetro laboratorial para identificar a etiologia da trombocitopenia (consumo periférico ou falha medular). Embora novos estudos precisam aprofundar o conhecimento e a segurança do uso do IPF, este parâmetro mostra-se útil para o auxílio diagnostico e acompanhamento terapêutico de uma série de patologias associadas à trombocitopenia, como púrpura trombocitopênica, acompanhamento de pacientes transplantados e em pacientes com risco de sepse.

Abstract:
Introdução: O S. aureus trata-se de uma bactéria Gram-positiva onde suas células têm forma de cocos, apresentam-se frequentemente agrupados em cacho e são imóveis. Em condições favoráveis, produz toxinas – enterotoxinas – que são o agente responsável pela intoxicação alimentar. Os fatores de patogenicidade associados a bactéria são; adesinas e formação de biofilme. Outros fatores como invasinas (proteases, lipases) e toxinas, também tem sido bastante estudados. Considerando a habilidade das bactérias produzirem o filme extracelular in vitro, propõe-se uma marca de patogenicidade associando ser um organismo virulento. Objetivo: Avaliar seus fatores características e patogenicidade bem como pesquisar a sua capacidade produtora de enterotoxina. Metodologia: O presente trabalho configurou-se com um estudo de caso através da revisão sistemática da literatura, que possibilitou a construção de referencial teórico sobre assuntos que estão relacionados ao tema em questão, utilizando dados de artigos científicos das plataformas digitais PubMed (National Library of Medicine) e SciELO (Scientific Electronic Library Online). Resultados: As espécies de Staphylococcus podem ser classificadas em dois grupos de acordo com a presença ou ausência da enzima coagulase. Assim, as espécies que possuem a enzima são denominadas coagulase positiva, sendo o Staphylococcus aureus a única espécie desse grupo, e as espécies que não possuem são chamadas de estafilococos coagulase negativa. Conclusão: As enterotoxinas produzidas por S. aureus podem estar relacionadas com o agravamento das doenças causadas pelo micro-organismo, além da sua habilidade de causar intoxicação alimentar, dessa forma constata-se que a bactéria é um dos principais patógenos de maior patogenicidade alta ocorrência e morbidade das infecções por este ocasionado.

Introdução: As doenças inflamatórias intestinais representam um conjunto de doenças crônicas que englobam como principais patologias a doença de Crohn e a colite ulcerativa que estão relacionadas a distúrbios nutricionais, deficiências de vitaminas, fatores genéticos e ambientais levando a má regulação da microbiota intestinal desencadeando lesões gastrointestinais. Objetivo: Abordar as principais adversidades das doenças inflamatórias intestinais, enfatizando os distúrbios nutricionais envolvidos. Material e métodos: A presente pesquisa segue os princípios e diretrizes de uma revisão de literatura. Foram consultadas as bases de dados online, Scientic Eletronic Online (SciELO), National Library of Medicine (Pubmed) e Bliblioteca Reginal de Medicina (Bireme); foram contemplados artigos publicados entre 2009 à 2019, incluindo os clássicos, nas línguas portuguesa, inglesa e espanhola. Resultados: A Doença de Crohn é caracterizada por uma inflação crônica do trato gastrointestinal que afeta predominantemente a parte inferior do intestino delgado e grosso, acometendo maioritariamente o público feminino; sua a manifestação está relacionada ao estilo de vida. Além de extensa, seu processo inflamatório pode dispor de complicações e como consequência as funções como absorção de nutrientes essenciais são afetadas e como resultado, a diarreia se torna um sintoma comum durante uma crise. Já a Colite Ulcerativa afeta o intestino grosso podendo ter início em qualquer idade, sendo caracterizada por inflamação e ulceração da camada mais superficial do cólon. Diarreias também é um sintoma muito comum que compromete a retenção de proteínas e vitaminas que provoca emagrecimento e anemia pela deficiência dos nutrientes essenciais. Conclusão: A excelente absorção de substâncias vitais ao organismo interfere diretamente na fisiologia humana e torna-se essencial para manutenção de funções básicas. As doenças inflamatórias intestinais geram inflamações nas paredes das mucosas intestinais afetando diretamente a absorção dos nutrientes, causando sintomas inespecíficos, interferindo no quadro nutricional do paciente refletindo diretamente sua qualidade de vida.

Introdução: A esquistossomose mansônica (EM), enfermidade parasitária desencadeada pelo helminto Schistosoma mansoni, permanece como uma importante doença no contexto da saúde pública brasileira. Com efeito, o conhecimento sobre o diagnóstico, as possibilidades terapêuticas, farmacológicas ou não, a epidemiologia e as medidas de profilaxia e controle tornam-se extremamente importantes para o clínico. Objetivo: Por se tratar de manifestações clínicas durante sua evolução, a esquistossomose mansônica (EM) assemelha-se a inúmeras outras doenças, dificultando o diagnóstico, retardando o tratamento e a notificação da parasitose. Para tal, realizou-se uma revisão da literatura utilizando as bases Scielo (Scientific Eletronic Library Online) e PubMed (National Library of Medicine). Material e métodos: No diagnóstico parasitológico é fundamental o exame de fezes, com especial importância para as técnicas de Lutz e Kato-Katz, esta última um método quantitativo, com grande aplicabilidade na inferência da carga parasitária, detectando a presença de ovos nas fezes, o que ocorre após o 45º dia de infecção. Resultados: O exame de fezes possui baixa sensibilidade, as infecções por S. mansoni com pequena carga parasitária. Recomendam-se a realização de exames laboratoriais com um mínimo de três amostras sequenciais de fezes, coletadas em dias distintos, com intervalo máximo de 10 dias entre a primeira e a última coleta. Conclusão: Os exames laboratoriais e a avaliação por métodos complementares são essenciais para a detecção e mensuração do comprometimento orgânico causado pelo S. mansoni, destacando-se os métodos parasitológicos, imunológicos e os de imagem. É importante conhecer os elementos atinentes ao diagnóstico clínico e laboratorial, bem como ao tratamento, à epidemiologia, à profilaxia e ao controle da SM aspecto extremamente relevante para a condução dos pacientes vitimados pela doença.

Introdução: Microrganismos endofíticos são bactérias ou fungos que vivem dentro das plantas e geram vantagens de sobrevivência para seu hospedeiro, e assim, esses microrganismos podem proporcionar vantagens seletivas contra estresses ambientais, pragas e doenças, contribuindo na maior eficiência em filtração de nutrientes e elevação do crescimento. Dentre os produtos desses microrganismos, estão substâncias com atividade antimicrobiana, que protegem a planta contra infecções por microrganismos fitopatogênicos. O papel de muitos microrganismos endofíticos nas suas plantas hospedeiras ainda não está completamente elucidado, mas diversas espécies já foram caracterizadas apresentando, entre outras características, a capacidade de produzir moléculas antimicrobianas. A descoberta e utilização de moléculas naturais, com atividade antimicrobiana, podem trazer avanços significativos para o controle de infecções graves e redução das taxas de mortalidade. Objetivo: Dessa forma, este estudo tem por objetivo descrever as principais espécies do bioma brasileiro que apresentam microrganismos endofíticos que produzem substâncias com atividade antimicrobiana. Material e Métodos: Este estudo tem caráter descritivo e foi realizado por meio de uma revisão bibliográfica, a partir da coleta de informações de fontes secundárias: Google Acadêmico, National Library of Medicine (PUBMED) e o Scientific Electronic Library Online (SCIELO). Resultados: Foram identificados na literatura 15 citações de bactérias e 129 de fungos endofíticos, como produtores de substâncias antimicrobianas. Nossos resultados mostraram uma frequência maior dos gêneros Bacillus spp. e Diaporthe spp. como bactérias e fungos endofíticos, respectivamente, isolados de 41 espécies de plantas brasileiras. Conclusão: A busca de substâncias produzidas por microrganismos, com atividade antimicrobiana, pode reduzir significativamente o problema da resistência aos antibióticos. A diversidade do bioma brasileiro pode ser uma grande fonte de estudos para busca e caracterização dessas substâncias.

Introdução: O metotrexato é uma droga antineoplásica classificada como antimetabólito, estruturalmente análogo ao ácido fólico, o MTX inibe uma enzima (diidrofolato-redutase), que é responsável por transformar a forma inativa do ácido fólico (diidrofolato) em forma ativa (tetraidrofolato). A forma ativa é utilizada na produção de nucleotídeos purinas. As purinas são essenciais para a replicação do DNA da célula. Portanto, o MTX inibe a produção dos nucleotídeos purinas o que impossibilita a replicação do DNA da célula, interrompendo o crescimento celular. Objetivo: Enfatizar a veracidade do uso do MTX nos diversos tipos de câncer bem como comprovar seu efeito benéfico no tratamento farmacológico. Metodologia: Estudo embasado em dados da revisão sistemática da literatura provida das bases de dados Medline e Science Direct, PubMed (National Library of Medicine) e SciELO (Scientific Electronic Library Online) no período entre janeiro e maio de 2021. As palavras-chave utilizadas foram “Antineoplásicos”, “Câncer”, “Metotrexato” e “Quimioterápicos”. Resultados: A quimioterapia antineoplásica com o MTX é amplamente utilizada no tratamento de vários cânceres. Esse tratamento é utilizado em doses o mais próximo possível das doses máximas individuais toleradas e devem ser administrados com a maior frequência possível para desestimular o novo crescimento do tumor. Dentre os cânceres que abrange o uso do metotrexato são: Câncer de mama, leucemia linfoblástica, sarcoma, bexiga, cabeça e pescoço, leucemia aguda, linfoma não Hodgkins, neoplasias trofoblásticas gestacionais etc. Conclusão: Desse modo o MTX atua como bloqueador da duplicação celular, impedindo o crescimento tumoral, assim como o surgimento de metástases nas doenças oncológicas e portanto diminuindo o risco de morte pela doença. O paciente deve ser informado sobre os riscos envolvidos e estar sob a supervisão constante do médico, onde o mesmo tem por obrigação enfatizar sobre as possibilidades de reações tóxicas sérias bem como a toxicidade do medicamento em questão.
