Abstract:
If (X′, τ′, ≤′) is an ordered compactification of the partially ordered topological space (X, τ, ≤) such that ≤′ is the smallest order that renders (X′, τ′, ≤′) a T2-ordered compactification of X, then X′ is called a Nachbin (or order-strict) compactification of (X, τ, ≤). If (X′, τ′, ≤*) is a finite-point ordered compactification of (X, τ, ≤), the Nachbin order ≤′ for (X′, τ′) is described in terms of (X, τ, ≤) and X′. When given the usual order relation between compactifications (ordered compactifications), posets of finite-point Nachbin compactifications are shown to have the same order structure as the poset of underlying topological compactifications. Though posets of arbitrary finite-point ordered compactifications are shown to be less well behaved, conditions for their good behavior are studied. These results are used to examine the lattice structure of the set of all ordered compactifications of the ordered topological space (X, τ, ≤).

Abstract:
In this paper we discuss the professional activity of Leopoldo Nachbin (1922-1993) in the period in which he worked at National Faculty of Philosophy of the University of Brazil (FNFi/UB, in Portuguese) until his retirement by Institute of Mathematics of the Federal University of Rio de Janeiro (IM/UFRJ, in Portuguese). We identify the influence played by some mathematicians such as G. Mammana (1893-1980), L. Schwartz (1915-2002), A. Monteiro (1907-1980), A. Weil (1906-1998), J. Dieudonné (1906-1992), M. Stone (1903-1989), and A. A. Albert (1905-1972) in the professional training of L. Nachbin. We also address the serious difficulties he encountered in the Mathematics Department of FNFi when he was entered in the competition, opened in 1950, for the post of full Professor (Professor Catedrático) of Mathematics and Superior Analysis of FNFi. We emphasize in the paper, that the period from 1947 to the 1960s marks the most creative and productive phase in terms of scientific research in the academic life of L. Nachbin.

Abstract:
We prove that the category of Nachbin's compact ordered spaces and order-preserving continuous maps between them is dually equivalent to a variety of algebras, with operations of at most countable arity. Furthermore, we show that the countable bound on the arity is the best possible: the category of compact ordered spaces is not dually equivalent to any variety of finitary algebras. Indeed, the following stronger results hold: the category of compact ordered spaces is not dually equivalent to (i) any finitely accessible category, (ii) any first-order definable class of structures, (iii) any class of finitary algebras closed under products and subalgebras. An explicit equational axiomatisation of the dual of the category of compact ordered spaces is obtained; in fact, we provide a finite one, meaning that our description uses only finitely many function symbols and finitely many equational axioms. In preparation for the latter result, we establish a generalisation of a celebrated theorem by D. Mundici: our result asserts that the category of unital commutative distributive lattice-ordered monoids is equivalent to the category of what we call MV-monoidal algebras. Our proof is independent of Mundici's theorem.

Abstract:
Imaging tools in cellular and molecular biology have long relied on organic fluorophores to observe microorganisms or various cell constituents. The advent of semiconductor nanoparticles known as quantum dots (QDs) has offered the possibility to use this new class of fluorescent probes with very advantageous optical properties in cell biology. The imaging of transmembrane potential and ionic currents is of significant importance for monitoring the activity of the cell. It remains possible with relatively complicated instruments and methods such as patch clamping. A complementary approach to view the dynamics of ion channels with modern and efficient fluorophores is therefore of great interest to the field of biology in general.
We developed a construct based on the FRET signal between QDs and organic fluorescent dyes to monitor the conformational changes of voltage gated sodium channels. The amino acid histidine was used as a "landing platform" for QDs and the bacterial sodium channel NaChBac was chosen for testing. This study focused on the preliminary steps of the project and aimed to characterize the electrophysiological behavior of the histidine-tagged channel. The whole-cell configuration of patch clamping was the tool we used to understand the differences between the wild-type and the histidine-tagged variants of the channels. We also explore the possibility to land QDs on the histidine tag.

Platelets are routinely stored for transfusion at room temperature in autologous, citrated plasma. We have demonstrated previously that these conditions do not completely block activation of plasma enzyme systems, as indicated by generation of thrombin activity (Vox Sanguinis, JL1:192,1986). Here, we demonstrate the conversion of large amounts of complement factor C3 during storage of citrated PC by using radioimmunoassay quantitation of the activation peptide C3a des-Arg (Upjohn Diagnostics). Supernatant samples from stored PC and from citrated platelet-poor plasma (PPP) stored under the same conditions showed a rapid linear increase in C3a levels over time with no significant difference (paired t-test, p<0.5) between PC and PPP (see table). The values at Day 10 represent conversion of approximately 11% of the native C3. Possible effects on stored platelets of C3 conversion in the surrounding plasma include:activation of platelets by C3a des-Arg (M.Polley and R. Nachman; J.Exp.Med. 158:603, 1983) and deposition of C3b on the cell surface as "innocent bystanders" (A. Salama and C. Mueller-Eckhardt; Transfusion 25:528,1985).In contrast, <10 ng/mL C5a was found in all samples tested, representing less than 0.2%conversion of C5a.Nephelometricassay of native C5 levels in PC samples showed a slight but significant difference by a paired t-test (p=0.04) between fresh PC (mean=117 ug/mL±12.0, n=6)and P stored for 10 days(nean=108 ug/mL±9.7). Nochange in C5 levels was observed in stored PPP (106 ug/mL to 107 ug/mL). Radiolabelled monoclonal antibodies to C3 fragments showed less than 600 molecules bound per platelet. This study demonstrates for the first time the extent of complement activation in stored platelet concentrates.

Abstract:
ITP is an autoantibody-mediated disease which would logically be treated by decreasing the level of autoantibody. However, the most exciting developments in understanding the pathophysiology of the thrombocytopenia and its treatment involve a better understanding of the MPS FcR system and ways in which it can be modulated. This work has focussed on phagocytic paralysis or FcR blockade (FcRBl): the slowing of destruction of antibody-coated platelets despite the persistent presence of antibody on the surface of the platelet.Several areas have been explored in learning about the MPS system. Investigation by Kurlander among others have revealed that at least 2 FcR's exist on mononuclear phagocytes: one with high and one with low affinity for monomeric IgG. Study of the high affinity FcR expressed by circulating monocytes, by Schreiber among others, has explored the effect of Danazol to decrease the expression of this FcR. The clinical relevance of this receptor is uncertain however because it is saturated in vitro by physiologic concentrations of IgG. Unkeless defined the properties of the low affinity "immune complex" FcR, expressed on macrophages and neutrophils, via monoclonal antibody 3G8 (see below) which blocks ligand binding to this FcR. The exact roles of these two, and possibly more, FcR's are being explored. Another still unsolved controversy involves whether the interaction Fc portions of antibodies coating particles with FcR's is mediated by a conformational change of the Fc portion or by a multipoint attachment of several Fc parts.Studies by Mollison in the 60's demonstrated that the MPS had a limited capacity for removal of antibody-coated (red) cells. Shulman pursued MPS modulation by exploring the inhibition of thrombocytopenia caused by infusion of ITP plasma into normals. Kelton demonstrated that "compensated" ITP may be caused by a decreased clearance of antibody-coated cells and that the rate of clearance of antibody-coated cells may be correlated with rate of clearance of antibody-coated cells may be correlated with the intrinsic levels of IgG. Stossel investigated FcRBl as a mechanism of effect of corticosteroids and related it clinically. Subsequently intravenous gammaglobulin (IVGG) was introducedas a treatment of ITP and Fehr et al first demonstrated FcRBl as the mechanism of effect of IVGG. Exploration of the mechanism of the FcRBl caused by IVGGled Salama and Mueller-Eckhardt to demonstrate the therapeutic effect of I anti-D, which apparentlycoats RBC with antibody and causes their destruction atthe coats RBC with antibody and causes their destruction at the expense of antibody-coated platelets. A similar degree of FcRBl has been shown for aldometrelated to the development of antibody on RBC.Our studies, including Drs. Clarkson, Kimberly, Nachman, and Unkeless, have focussed on the role of the low affinity or "Immune complex" FcR by using monoclonal antibody 3G8 in vivo. An infusion of 1 mg/kg of 3G8 in chimpanzees caused a reproducible FcRBl demonstrable by a slowing of the destruction of antibody-coated RBC for > 10 days (JEM, 1986). Less effect of 3G8 to inhibit CIC removal was seen using DNA-anti-DNA as the immune complex. In view of the wel1-documented effects of IVGG infusion to create FcRBl, we infused 3G8 into 6 adults with refractory ITP (NEJM, 1986). Specifically these patients were refractory to all forms of conventional therapy including splenectomy, steroids, vinca alkaloid infusion, immunosuppressives and danazol . 3 of the 6 patients had peak platelet responses to >80,000/ul. The other 3 had short-lived platelet increases from 10 to 30,000/ul. These responses confirmed the effect of FcRBl, specifically of the low affinity FcR, to underlie a dramatic platelet increase in therapy of ITP. Surprisingly 3 of the patients had apparent longterm effects of this therapy demonstrable in 2 cases as a maintenance of the platelet count >20,0C0/ul without any further therapy and in 1 case as a clearly enhanced responsiveness to other therapies following 3G8 infusion. Since Natural Killer activity was (transiently) ablated by 3G8 infusion, we speculate that an alternation of regulation of (auto) antibody production by NK cells may be responsible for this effect and that FcR interactions include regulatory roles in addition to their primary function of removal of CIC.

Gorbitz ist ein Stadtteil, der von außen oft mit Klischees bedacht wird. Spricht man über kriminalitätsbezogene Herausforderungen in Dresdner Stadtteilen, wird Gorbitz oft an erster Stelle genannt. Die im Stadtteil vorhandenen Probleme stehen häufig im Fokus, während das Potential und das Engagement von Einwohnerinnen und Einwohnern, Vereinen und Einrichtungen nur selten wahrgenommen wird. Kommunale Kriminalprävention bedeutet in diesem Zusammenhang auch, den Blickwinkel auf einen Stadtteil zu verändern und diejenigen in den Fokus zustellen, die eng miteinander vernetzt im Stadtteil arbeiten und diesen positiv weiterentwickeln. Rückblickend auf das letzte Jahrzehnt lässt sich für Gorbitz anhand der Polizeilichen Kriminalstatistik (PKS)feststellen, dass der Stadtraum weniger von Kriminalität belastet war als die Stadt Dresden insgesamt. Erst in den letzten beiden Jahren ist ein Anstieg der Straftaten zu verzeichnen, 2019 wurden mehr Straftaten je 100.000 Einwohnerinnen und Einwohner registriert als in Dresden insgesamt. Die Gorbitzerinnen und Gorbitzer nehmen diesen Anstieg auch wahr: In einer postalischen Bürgerbefragung des SiQua-Projektesgaben71% der Befragten an, dass sich die Sicherheit in ihrem Wohngebiet während der letzten fünf Jahre verschlechtert hat. Vor allem nach Einbruch der Dunkelheit fühlen sie sich erheblich unsicherer als die Dresdnerinnen und Dresdner insgesamt. Das Vertrauen und der Zusammenhalt unter Nachbarn ist in Gorbitz deutlich geringer als in den meisten anderen Stadtteilen Dresdens Das ist ein wichtiger Grund für das geringe Sicherheitsgefühl. Ebenso lassen die Studienergebnisse aber auch die Potentiale des Stadtteils erkennen. So haben sich in Gorbitz während der letzten 5 Jahre fast genau so viele Einwohnerinnen und Einwohner ehrenamtlich für Geflüchtete engagiert, wie im restlichen Stadtgebiet (9%bzw.10%). Mit dem Blick auf mögliche Ansätze der kommunalen Präventionsarbeit zeigen sich die Gorbitzerinnen und Gorbitzer offen für unterschiedliche Lösungsansätze: Sieerachten sowohl härtere Strafen von Straftätern als auch mehr Angebote und Hilfen für Jugendliche für sinnvoll. Kritische Einstellungen lassen sich gegen über mehr privaten Wachdiensten feststellen.
