Abstract:
Android application development has been of interest since the first Android smartphone was released. Applications are constantly getting more complex as well as smartphone hardware is getting better. New ways of developing Android applications are developed with time. There is Model View Presenter architecture that is the most used for android applications now and new View InteractorPresenter Entity Router architecture that is becoming more popular. But there is no empirical data to compare these architectures to understand what architecture will fit better for developing new applications. This thesis aims to compare the MVP and the VIPER android architectures using a few important metrics like maintainability, modifiability, testability, and performance. Results will answer what architecture is better for developing different types of projects. VIPERarchitecture showed better performance results and maintenance metrics comparison shows that both architectures have advantages and disadvantages.

The fundamentals that make up the price of financial assets are of utmost importance to the economic agents. Among the existing finance theories have the Model of Present Value (MVP) which considers the current prices of securities as the sum of future dividends to be generated brought to their respective present value using a discount rate equivalent to required rate of return. The objective of this work is the application of MVP in the Brazilian market, testing the predictability of returns in a context of rational expectations will be a long-term relationship analysis of validation test between prices and dividends analyzing its stationary and cointegration relationship. For this two situations are considered in sequence in the first assume that the expected return will always be constant and then evaluate the model by assuming the return varies over time. The results obtained demonstrate that the pricing of financial assets in the Brazilian market is in line with the theory presented in the present value model there so a long-term relationship between prices and dividends.
Os fundamentos que compõem o preço dos ativos financeiros são de total relevância aos agentes econômicos. Entre as teorias de finanças existentes, temos o Modelo de Valor Presente (MVP), que considera os preços correntes dos títulos como a soma dos dividendos futuros a serem gerados trazidos ao seu respectivo valor presente por meio de uma taxa de desconto equivalente à taxa requerida de retorno. O objetivo deste trabalho foi realizar a aplicação do MVP no mercado brasileiro, testando a previsibilidade dos retornos num contexto de expectativas racionais. Para tanto, foi realizado um teste de validação da análise da relação de longo prazo entre preços e dividendos, analisando sua relação de estacionariedade e cointegração. Foram consideradas duas situações distintas em sequência, na primeira hipótese o retorno esperado será sempre constante e em seguida avaliamos o modelo presumindo que o retorno varia ao longo do tempo. Os resultados obtidos demonstram que a precificação dos ativos financeiros no mercado brasileiro está de acordo com a teoria apresentada no modelo de valor presente existindo assim uma relação de longo prazo entre preços e dividendos.

This thesis is focused on a comparison of two frameworks, built on two different platforms. The first one is a PHP-based framework Nette and the second one is Spring, developed in Java. They both are popular choices of frameworks on their respective platforms. The first part of the thesis' target is to introduce the reader to elementary ideas related to frameworks - design pattern MVC. The second part describes the Nette framework, the third is devoted to Spring framework. The fourth part deals with an example application and it's realization in both Nette and Spring, and the fifth and last part compares both frameworks, evaluates them and closes the whole topic.

The goal of this research is to provide a tool to unobtrusively introduce the design metrics D,, and Di into existing engineering practices. To that goal, different methods for visualizing the UML design metrics were explored. A small survey was conducted to determine which of the different visualizations is most attractive. The results of this survey were used to guide the design of the Design Metrics Visualization Plugin (Design MVP), which was developed as a plugin for the open-source UML tool Dia. This plugin allows for the calculation, and more importantly, visualization of the UML metrics during UML class diagram modeling.

Esta pesquisa tem como objetivo geral propor um Modelo Vocal para Percussionistas (MVP), na execução de obras solo em que o uso da voz é requerido, explorando elementos físicos e musicais que fazem parte das práticas diárias do estudante e do profissional da percussão. Para tanto, foram escolhidas três importantes peças do repertório percussivo para a aplicação dos conceitos propostos: ?Corporel, para um percussionista e sons do seu corpo (1985), de Vinko Globokar (1934); To the Earth (1985), de Frederic Rzewski (1938); e URSONATE (1922 - 1932), de Kurt Schwitters (1887 - 1948). Por possuírem características distintas, quanto ao uso da voz, cada uma delas possibilita o entendimento do MVP, como um paradigma teórico amplo o suficiente, para resolver problemas técnicos e interpretativos diversos, a partir da compreensão do contexto em que estão inseridas: a Música-Teatro. Sob as considerações da pesquisadora Claudia Hansen, e do professor Wladimir Mattos, foram estipuladas as bases deste gênero musical, bem como os perfis do percussionista e da percussão que o representam, além daquilo que deve ser levado em conta como sendo a voz e a fala. A partir de três pilares teórico-práticos - a fonoarticulação, a coarticulação e a realização prosódica - as obras escolhidas foram analisadas e preparadas para a performance, levando-se em conta as discussões ocorridas no âmbito da fonética e da fonologia, e as soluções encontradas foram esquematizadas por meio de tabelas, visando a facilitar a compreensão do MVP. Desta forma, esperamos que a proposta do presente trabalho auxilie futuras pesquisas, interpretações e discussões inseridas no universo percussão e voz, acentuando as cores desta tão tênue e simbiótica linha que perpassa os ambientes musical e teatral.
This research aims to propose a Vocal Method for Percussionists (VMP) for playing solo pieces where using the voice is required. The physical and musical elements that are part of the percussionist's daily practice were analyzed. In order to achieve that, three major, well established, percussion pieces were drafted regarding the application of the proposed methods: ?Corporel, for one percussionist using her/his body (1985), by Vinko Globokar (1934); To the Earth (1985), by Frederic Rzewski (1938); and URSONATE (1922 - 1932), by Kurt Schwitters (1887 - 1948). Despite their differences regarding how the voice is used, each one of them enables the reader to assimilate the VMP, as a theoretical paradigm broad enough to figure out a wide range of interpretative and technical issues, fully understanding the context in which they belong: the music-theater. Under Claudia Hansen's and professor Wlad Matos' research analysis, we established the basis of this music genre, as well as the percussionist's and percussion characteristics that define them, and what should be considered as voice and speech. Starting off from three practical-theoretical pillars - articulation of speech, articulatory gesture and the prosodic conduction - the selected pieces were thoroughly analysed, looking for any issues regarding the use of the voice and singling out schematic solutions that would emerge. Thus, we hope that our work may assist future research, interpretation and discussion regarding percussion and voice. Taking into consideration the very gentle and symbiotic confluence within the musical and theatrical environments.

Abstract:
The Monaro Volcanic Province (MVP) is an Oligocene-Eocene intraplate basaltic lava field situated in the Southern Highlands of New South Wales between the towns of Cooma and Bombala. The lava pile of the MVP consists of basal sub-alkali rocks (olivine tholeiite, transitional basalt) capped by a number of thick ankaramite lavas, above which lie less numerous alkali rocks including alkali olivine basalt, nepheline basanite and olivine nephelinite. Intercalated with the lava flows are massive and matrix-supported alkali and ankaramitic hyaloclastites, alkali pillow basalts, rare tuffs, bauxitic weathering profiles, lacustrine sediments and reworked late Cretaceous to early Tertiary river gravels. The lava pile is intruded through by numerous volcanic plugs and dykes and rare maars. Volcanic centres are principally concentrated in two NW-SE trending zones parallel to major crustal-scale fractures in the Palaeozoic basement. Centres almost always lie over the intersections of two or more conjugate strike-slip or transverse fractures. The stratigraphy, whole-rock geochemistry and Sr and Nd isotopic signatures of rocks from the MVP indicate magma-genesis initially from an asthenospheric source with EM1 characteristics, gradually becoming more lithospheric with DM source characteristics. The long-lived nature of the MVP rules out a mantle plume-type source for magmas. Instead, a diapiric source is envisaged. The MVP mantle xenolith suite appears to have equilibrated at slightly higher temperatures for given pressures than the Newer Volcanics Province suite suggesting the palaeogeotherm for the MVP was slightly hotter than the "South East Australian" geotherm. Large amounts of amphibole (pargasitic hornblende, pargasite, ferroan pargasite and kaersutite) occuring within the more silica-undersaturated rocks of the MVP, and rarely within Iherzolitic xenoliths, are interpreted to have formed as selvages on mantle veins in contact with peridotite beneath the MVP. Amphiboles were later sampled by magmas rising through the same conduits and were brought to the surface. MVP ankaramite lavas feature < 2cm clinopyroxene porphyrocrysts, the cores of which are shown to have crystallised at ca. 18 kb pressure or ca. 54 km depth. This defines the base of the local crust within the MVP region. Data from the MVP support a landscape evolution model based on the isostatic rise of the Southern Highlands due to voluminous magmatic underplating since the Cretaceous. Data further support limited denudation since the Early Tertiary based on a pulsatory but high palaeogeotherm.

s (2003) minimum variance portfolio (MVP) framework. Financial dollarization, steamed by unfavorable macroeconomic conditions and uncertainties, is revealed by the experiences of recent banking and financial crisis as carrying significant drawbacks such that it complicates economic policy implementation and contains the seeds of fragility for the whole economy as well. Although, considerable progress has been achieved in reducing inflation levels and sustaining macroeconomic stability, financial dollarization displays rather an enduring stance. MVP approach is based on optimizing the currency composition of financial contracts depending on the risk and the return profile of agents&rsquo
portfolios. According to this approach, financial dollarization is an increasing function of the inflation volatility and a decreasing function of the real exchange rate volatility. In line with this framework, financial dollarization in the Turkish economy during 1990-2011 period is studied by also considering other important macroeconomic risk indicators and it is tried to shed some light on the success of inflation targeting policy in dealing with dollarization phenomenon.

Метою проекту є розробка програмної системи, яка дозволяє автоматизувати публікацію та поширення кулінарних рецептів серед користувачів, а також забезпечує швидкий та зручний їх пошук та має сучасний інтуїтивно зрозумілий користувацький інтерфейс. У дипломному проекті визначено специфіку соціальних мереж; виконано порівняння монолітної та мікросервісної архітектури; проведено аналіз методів та інструментів по управлінню високонавантаженими системами та визначені шляхи їх оптимізації; проаналізовано використання реляційних та NoSQL баз даних, встановлено їх переваги та недоліки; встановлено особливості фронт-енд фреймворків при реалізації клієнтської частини програми. В результаті проектування здійснена програмна реалізація системи для автоматизації публікації, поширення та пошуку кулінарних рецептів, а також проведено практичну апробацію отриманих результатів. Запропоновано стратегію з подальшого розвитку маркетингової діяльності, проведене опитування, на основі якого сформовано план курсів з комплексного Інтернет-маркетингу та медіаплан з їх просування в Інтернеті, розраховано прибуток від курсів, надано пропозицію з інвестування в стартапи, розраховано дохід від інвестицій.

L'idea di realizzare un'applicazione gestionale su piattaforma Android, è nata dall'esigenza di risolvere alcune problematiche reali riscontrate durante l'organizzazione di eventi, spese e commissioni da svolgere fra gruppi di persone. Lo sviluppo iniziale si avvaleva di Java come linguaggio di programmazione, successivamente con l'annuncio ufficiale a Maggio 2017 del supporto di Google a Kotlin, come nuovo linguaggio di programmazione per Android, l'applicazione è stata riscritta completamente, utilizzando Kotlin. Lo sviluppo dell'applicazione in Kotlin ha permesso di analizzare le funzionalità del nuovo linguaggio sia teoricamente che con un'implementazione pratica che ha reso possibile anche un confronto diretto con Java. In questa tesi verranno illustrate le principali funzionalità e caratteristiche del linguaggio Kotlin, e del BaaS Firebase utilizzato per la gestione della parte server. Dal terzo capitolo invece verrà presa in considerazione l'applicazione, mostrando l'architettura ad alto livello dell'infrastruttura dei servizi server e delle funzionalità dell'applicazione. Successivamente verranno analizzate porzioni di codice rilevanti e discusse le implementazioni sviluppate per la realizzazione della parte client dell'applicazione. Infine nell'ultimo capitolo viene descritto l'attuale stato dell'arte, il confronto fra Java e Kotlin ed eventuali sviluppi futuri.

Every software program that interacts with a user requires a user interface. Model-View-Controller (MVC) is a common design pattern to integrate a user interface with the application domain logic. MVC separates the representation of the application domain (Model) from the display of the application's state (View) and user interaction control (Controller). However, studying the literature reveals that a variety of other related patterns exists, which we denote with Model-View-* (MV*) design patterns. This thesis discusses existing MV* patterns classified in three main families: Model-View-Controller (MVC), Model-View-View Model (MVVM), and Model-View-Presenter (MVP). We take a practitioners' point of view and emphasize the essentials of each family as well as the differences. The study shows that the selection of patterns should take into account the use cases and quality requirements at hand, and chosen technology. We illustrate the selection of a pattern with an example of our practice. The study results aim to bring more clarity in the variety of MV* design patterns and help practitioners to make better grounded decisions when selecting patterns.

La tesi si pone quindi come obiettivo lo sviluppo di un’applicazione Android per la consegna a domicilio di dolci e prodotti di caffetteria, secondo il modello di startup “Fully Integrated”. Il software deve incaricarsi della gestione degli ordini e di un elaborato sistema di sottoscrizione ad abbonamenti, mostrando all’utente il catalogo prodotti e seguendolo nel processo di acquisto tramite una User Experience appositamente studiata. L’applicazione vuole quindi aiutare il gestore del servizio di pasticceria, che si dovrà occupare solamente della produzione e della consegna delle colazioni.

Abstract:

The minimum ecologically viable population size (MEVP) of a species describes the minimum size at which the species itself or another species in the same ecosystem goes extinct as a result of the loss of inter-specific interactions. The MEVP shows a good potential for use as a tool for exploring the mechanisms behind species extinctions, but presently only a small amount of research has been done that takes advantage of this. In this study the MEVP is used to investigate what properties of species can be used as good indicators of ecological importance. 100 large computer generated food webs were created with an assembly model and the reduction in density that was necessary to induce an extinction event in the web was subsequently determined for each species within the webs. This change in density was then correlated with 28 different properties, measured for each species. The results show that properties that measure how well connected a species is, as well as measures of the species role as a prey item in the web are the ones with the greatest potential to find species with high MEVP. Further, the results put emphasis on the importance of regarding the web as a whole when working with species extinctions, while also highlighting the usefulness of the MEVP concept.

Com o advento das Startups, e das metodologias denominadas Agile de criação e estruturação empresarial, os modelos antiquados de organização empresarial começaram a cair em desuso. Não obstante, bastantes Startups ainda se agarram a metodologias de desenvolvimento de produto, e deployment do mesmo sem qualquer análise prévia de viabilidade, apoiando-se nos tradicionais estudos de mercado como suporte de viabilidade, investindo de uma forma quase cega numa ideia/ produto, muitas vezes sem qualquer teste. A metodologia Lean Startup debruça-se sobre uma perspectiva diferenciada das comuns metodologias, baseando-se na interação do produto, e das várias fases de desenvolvimento do mesmo com o mercado. Podemos dizer que o produto nunca é algo acabado, mas sim algo que se vai desenvolvendo, aprimorando, adaptando-se ao mercado, e às suas variações, procurando um percurso em constante mutação. A relação desta metodologia com o mercado de aplicativos móveis, uma estrutura altamente volátil, em conformidade com as ditas "modas", é clara, carecendo qualquer produto de constantes updates, de forma a adaptar o mesmo às aparentes necessidades dos utilizadores, surgindo aqui o feedback por parte do end-user, e da criação de metodologias de obtenção deste fundamentais na viabilização do produto. Assim, o presente projecto passa por analisar a viabilidade desta metodologia ao desenvolvimento de uma proposta de valor para um MVP, utilizando o formato Business Model Canvas para analisar as variações das diferentes iterações, e obter uma proposta de valor, sobre a qual poderá ser desenhado o MVP, para ulteriormente se fazer o deployment.
With the continuous growth of the Startup business model, and the agile methodologies, the antique business organization models began their decay. Nevertheless, many startups still hold on to the normal product development and deployment methods, without previous feasibility analysis, supporting the idea on the traditional market survey, blindly investing on an idea/ product, many times without even a real test. The Lean Startup methodology focuses on a differentiated perspective, basing on a continuous testing of the product and its development phases in the market. We can say that the product is never concluded, but is ever developing, enhancing, adapting to its market, and its customers, seeking a viability path that is ever changing. This methodology relation with the App market, a fairly volatile structure, always subject to fashion trends, is clear, with products in permanent update, adapting to customer needs, surfacing the end-user feedback, and its methodologies as fundamentals in product feasibility. Thus, the present project analyses this methodology feasibility in creating a value proposition for a MVP, a smartphone app, using the Business Model Canvas to analyze variations in the different iterations, towards a final value proposition, with which we can draw a MVP, and prepare it´s deployment.

La principale caracteristique de la phosphorylation des proteines cellulaires est sa reversibilite. Dans la cellule, il existe une balance entre la phosphorylation (assuree par des kinases) et la dephosphorylation (assuree par des phosphatases) des proteines. Une modification de cette balance est a l'origine d'une grande partie des mecanismes permettant la transmission des signaux extracellulaires vers le noyau. Les phosphatases sont generalement divisees en trois grandes familles : les serine/threonine phosphatases, les tyrosine phosphatases et les phosphatases a double specificite. Map kinase phosphatase-1 (mkp-1), mkp-2 et mkp-3 sont tous trois membres de la nouvelle famille des phosphatases a double specificite (dsp) qui arborent la sequence consensus : (i/v)hcxagxxr(s/t)g. Ces mkp ont la propriete de dephosphoryler in vitro comme in vivo un substrat archetype, la p42/p44mapk. Afin de determiner l'impact de ces phosphatases sur la regulation des mapk ainsi que leur role dans la proliferation, nous avons entrepris leur etude dans le systeme bien defini des fibroblastes d'hamster chinois, les ccl39. Dans un premier temps, nous avons demontre (i) qu'une forme ectopique de mkp-1 (1-) se localise dans le noyau de cellules quiescentes ou stimulees. (ii) que cette forme surexprimee est capable de bloquer des activites nucleaires de la p42/p44mapk, la transcription de genes de la transition g1/s et enfin la replication de l'adn des cellules transfectees. Dans un second temps, nous avons entrepris la production d'anticorps polyclonaux diriges contre mkp-1 et mkp-2. Introdução: O Lúpus eritematoso sistêmico (LES) é uma doença inflamatória autoimune crônica de etiologia ainda pouco conhecida, e de natureza pleomórfica, que intercala períodos de atividade e remissão. O desenvolvimento da autoimunidade no LES está associado à perda da tolerância imunológica e do controle imunorregulatório, tendo seus achados clínicos e laboratoriais variados. A atividade do LES pode ser medida pelo SLEDAI (systemic lupus erythematosus disease activity index) que é uma ferramenta complexa e que exige treinamento e conhecimento para sua aplicação. O volume plaquetário médio (VPM) é um marcador de ativação de plaquetas associado à inflamação, o que o torna um potencial candidato para a avaliação de atividade de doença no LES. Objetivos: Avaliar o VPM em pacientes com LES e comparar com indivíduos hígidos. Estudar a correlação entre o VPM e o índice de atividade de doença (SLEDAI) nos pacientes com LES. Analisar a correlação entre o VPM e a velocidade de sedimentação globular (VSG), a proteína C reativa (PCR), e os componentes do complemento C3 e C4 Métodos: Estudo transversal no qual foram incluídos 81 pacientes com LES segundo critérios de classificação diagnóstica do American College of Rheumatology (ACR), e 58 controles hígidos. Os pacientes foram selecionados consecutivamente por conveniência, de acordo com exames laboratoriais e SLEDAI devidamente calculados. As coletas foram realizadas entre outubro de 2015 e julho de 2016. LES ativo foi definido como SLEDAI>0 no momento da coleta. O VPM foi analisado no equipamento de automação Sysmex XE 5000. Resultados: O VPM estava reduzido nos pacientes com LES em atividade, quando comparado ao grupo de pacientes com LES inativo (10,0±0,7fL vs. 10,7±1,0fL, p=0,005). Existe uma fraca correlação inversa entre o valor do SLEDAI e o VPM (r=-0,29, p=0,009). Houve uma diferença significativa no VPM entre o grupo dos controles e os pacientes com LES ativo
Background Systemic Lupus Erythematosus (SLE) is an inflammatory autoimmune chronic disease etiology still unknown, and pleomorphic nature, which intersperses periods of activity and remission. The development of autoimmunity in SLE is related to loss of immunological tolerance and immunoregulatory control and clinical symptoms can be varied. The SLE activity can be measured by SLEDAI (systemic lupus erythematosus disease activity) which is a complex tool and it requires time and knowledge for your application. The MPV (mean platelet volume) is a marker of platelet activation and has been shown to be associated with inflammation, which makes it a potential candidate for use in the assessment of disease activity in SLE. In this study, we evaluated the MPV (Mean platelet volume) in healthy individuals and compared with SLE patients and correlate with SLEDAI VPM. Objectives: -To evaluate the MPV in SLE patients and compared with healthy individuals; to study the correlation between MPV and the SLEDAI patients with SLE and assess a possible correlation between MPV with erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), complement 3 (C3), and complement 4 (C4) Methods: This is a cross-sectional study in which 81 patients with SLE according to the American College of Rheumatology (ACR) diagnostic classification criteria and 58 healthy controls were included. Patients were selected for convenience, according to laboratory tests and SLEDAI duly calculated. The collections were carried out between October 2015 and July 2016. Active LES was defined as SLEDAI>0 at the time of collection. The VPM was analyzed in the Sysmex XE 5000 automation equipment. Results: In this study in patients with active SLE, the MPV is reduced when compared to the group of patients with inactive SLE [10.0±0.7fL vs. 10.7±1.0fL, p=0.005]. There is a weak inverse correlation between the SLEDAI value and the MPV (r=-0.29, p=0.009). There was a significant difference between the control group and the patients with active SLE (10.9 ±1.0fL vs. 10.0±0.7fL, p <0.001). In contrast, the MPV was similar between the control group and the group of patients with inactive SLE (10.9±1.0fLvs10.7±1.0fL, p=0.40). There was no correlation between MVP and CRP, ESR, C3 and C4. Conclusion: MPV is decreased in patients with active SLE and inversely correlated with SLEDAI. Despite the difference between MVP values, between active and inactive SLE patients, the results may not be clinically relevant. Prospective longitudinal studies are needed to better characterize the fluctuation of MPV in different states of disease activity to more clearly define the role of MPV in SLE.

La synthèse d'images dites photoréalistes nécessite d'évaluer numériquement la manière dont la lumière et la matière interagissent physiquement, ce qui, malgré la puissance de calcul impressionnante dont nous bénéficions aujourd'hui et qui ne cesse d'augmenter, est encore bien loin de devenir une tâche triviale pour nos ordinateurs. Ceci est dû en majeure partie à la manière dont nous représentons les objets: afin de reproduire les interactions subtiles qui mènent à la perception du détail, il est nécessaire de modéliser des quantités phénoménales de géométries. Au moment du rendu, cette complexité conduit inexorablement à de lourdes requêtes d'entrées-sorties, qui, couplées à des évaluations d'opérateurs de filtrage complexes, rendent les temps de calcul nécessaires à produire des images sans défaut totalement déraisonnables. Afin de pallier ces limitations sous les contraintes actuelles, il est nécessaire de dériver une représentation multiéchelle de la matière. Dans cette thèse, nous construisons une telle représentation pour la matière dont l'interface correspond à une surface perturbée, une configuration qui se construit généralement via des cartes d'élévations en infographie. Nous dérivons notre représentation dans le contexte de la théorie des microfacettes (conçue à l'origine pour modéliser la réflectance de surfaces rugueuses), que nous présentons d'abord, puis augmentons en deux temps. Dans un premier temps, nous rendons la théorie applicable à travers plusieurs échelles d'observation en la généralisant aux statistiques de microfacettes décentrées. Dans l'autre, nous dérivons une procédure d'inversion capable de reconstruire les statistiques de microfacettes à partir de réponses de réflexion d'un matériau arbitraire dans les configurations de rétroréflexion. Nous montrons comment cette théorie augmentée peut être exploitée afin de dériver un opérateur général et efficace de rééchantillonnage approximatif de cartes d'élévations qui (a) préserve l'anisotropie du transport de la lumière pour n'importe quelle résolution, (b) peut être appliqué en amont du rendu et stocké dans des MIP maps afin de diminuer drastiquement le nombre de requêtes d'entrées-sorties, et (c) simplifie de manière considérable les opérations de filtrage par pixel, le tout conduisant à des temps de rendu plus courts. Afin de valider et démontrer l'efficacité de notre opérateur, nous synthétisons des images photoréalistes anticrenelées et les comparons à des images de référence. De plus, nous fournissons une implantation C++ complète tout au long de la dissertation afin de faciliter la reproduction des résultats obtenus. Nous concluons avec une discussion portant sur les limitations de notre approche, ainsi que sur les verrous restant à lever afin de dériver une représentation multiéchelle de la matière encore plus générale.
Photorealistic rendering involves the numeric resolution of physically accurate light/matter interactions which, despite the tremendous and continuously increasing computational power that we now have at our disposal, is nowhere from becoming a quick and simple task for our computers. This is mainly due to the way that we represent objects: in order to reproduce the subtle interactions that create detail, tremendous amounts of geometry need to be queried. Hence, at render time, this complexity leads to heavy input/output operations which, combined with numerically complex filtering operators, require unreasonable amounts of computation times to guarantee artifact-free images. In order to alleviate such issues with today's constraints, a multiscale representation for matter must be derived. In this thesis, we derive such a representation for matter whose interface can be modelled as a displaced surface, a configuration that is typically simulated with displacement texture mapping in computer graphics. Our representation is derived within the realm of microfacet theory (a framework originally designed to model reflection of rough surfaces), which we review and augment in two respects. First, we render the theory applicable across multiple scales by extending it to support noncentral microfacet statistics. Second, we derive an inversion procedure that retrieves microfacet statistics from backscattering reflection evaluations. We show how this augmented framework may be applied to derive a general and efficient (although approximate) down-sampling operator for displacement texture maps that (a) preserves the anisotropy exhibited by light transport for any resolution, (b) can be applied prior to rendering and stored into MIP texture maps to drastically reduce the number of input/output operations, and (c) considerably simplifies per-pixel filtering operations, resulting overall in shorter rendering times. In order to validate and demonstrate the effectiveness of our operator, we render antialiased photorealistic images against ground truth. In addition, we provide C++ implementations all along the dissertation to facilitate the reproduction of the presented results. We conclude with a discussion on limitations of our approach, and avenues for a more general multiscale representation for matter.

A Leishmaniose Visceral (LV) é uma zoonose de distribuição mundial. No Brasil foi descrita em várias regiões, mas na região sudeste vem ocorrendo de forma significativa desde 1998, na região de Araçatuba, SP. O cão é o principal reservatório doméstico do protozoário Leishmania chagasi e a principal fonte de contaminação para o homem. A resposta imune do hospedeiro canino frente ao parasito é variável e por este fato, os cães apresentam sinais clínicos acentuados ou ausência destes. Os animais que apresentam maior carga parasitária e sinais clínicos mais severos são considerados susceptíveis à infecção pelo parasito. Visceral Leishmaniasis (VL) is a zoonosis of worldwide distribution. In Brazil has been described in several regions, but has been occurring in the Southeast significantly since 1998 in Araçatuba, SP. The dog is the main domestic reservoir of Leishmania chagasi and the main source of contamination for humans. The canine host immune response against the parasite is variable. Dogs have severe intensity or absence of clinical symptoms. Animals that have a higher parasite load and clinical signs are considered more susceptible to infection by the parasite. This pattern is associated with the profile of cytokines produced, in the predominance of Th1 they are resistant or in the predominance of Th2 they are susceptible. However there are many variations on this response pattern, many animals have the production of Th1 and Th2 cytokines, suggesting an imbalance in the proportion of pro and anti-inflammatory cytokines, which favor the multiplication and survival of parasites. This imbalance has a direct influence on the activation of macrophages and their microbicidal activity and antigen presentation, leading to anergy of T lymphocytes and the maintenance and chronicity of infection. Chemokines are cytokines with potent chemotactic activity on leukocytes. In the VL are studies in humans and experimental models, but the dog only one study assessing its role in the spleen. Experimental Autoimmune Encephalomyelitis (EAE) is an animal model of multiple sclerosis (MS), causing a demyelinating central nervous system (CNS) inflammation which resembles the main pathological features of MS. Mitogen-activated protein kinases (MAPKs), which are key components in the molecular response leading to MS/EAE pathogenesis, are regulated by MAPK phosphatases (MKPs), enzymes which dephosphorylate phosphotyrosine and phosphothreonine residues. It has previously been shown that MKP-2 modulates the inflammatory response during both acute lung injury and sepsis. Therefore in the present study we investigated the role of MKP-2 in the development of the neurological autoimmune disease, MS, using a murine EAE model. We first observed significantly increased expression of MKP-2 mRNA in the spinal cord of EAE mice compared with PBS controls. Subsequently, to understand the function of MKP-2 in vivo, we utilised MKP-2 deficient mice, inducing EAE in MKP-2 KO and WT littermates. Our data show that MKP-2 KO mice displayed significantly reduced EAE susceptibility, associated with diminished CNS inflammation and cellular infiltration, decreased expression of key cytokines and chemokines (IL-17, IFNγ, IL-6, IL-2 and CCL2), reduced frequency of CD4⁺ T cells, CD8⁺ T cells and B cells in spleen and dLN tissue as well as downregulated nitric oxide (NO) production in MKP-2 KO EAE mice. We further analysed the role of MKP-2 in two key immune cells involved in EAE pathogenesis. Upon LPS stimulation, MKP-2 deficient bone marrow-derived dendritic cells expressed less MHC-II while producing more IL-6, TNF-α and IL-10, whereas MKP-2 KO bone marrow-derived macrophages displayed a unique M1 and M2 mixed phenotype, with reduced NO production (M1) and increased CD206 expression (M2) but increased IL-6 and TNF-α, which are more associated with M1 responses. Therefore this report suggests that MKP-2 is essential to the pathogenic response of EAE, and that inhibition of MKP-2 expression or function may be a viable strategy in the treatment of autoimmune inflammatory diseases such as MS.

Abstract:
Platelet-derived growth factor (PDGF) is a family of signaling molecules that stimulates cell growth, survival and migration. PDGF is recognized by specific transmembrane proteins, the PDGF receptors, which relay the signals to the cell activating the Mitogen-activated protein (MAP) kinases and other signaling pathways. Aberrant activation of these pathways is frequently detected in cancer. Hence, the study of these processes is essential for identifying potential drug targets or diagnostic markers. In paper I, we identified Receptor Subfamily 4 Group A Member 1 NR4A1 to be regulated by PDGF via MAP kinases, clarifying the role of Extracellular signal–regulated kinases (Erk) 1/2, Erk5 and Nuclear factor κ-light-chain-enhancer of activated B cells (NF-κB) in its regulation. NR4A1 was found to be important for the tumorigenic potential, measured as anchorage-independent growth, of glioblastoma cells. Since the cellular responses elicited by PDGF result from the balance between phosphorylation and dephosphorylation events, we investigated the role of the dual specificity phosphatases DUSP4/MKP-2 and DUSP6/MKP-3. In paper II, we describe the crucial role of Erk1/2 and p53 in the expression of DUSP4/MKP2. Moreover, we observed that DUSP4/MKP-2 downregulation decreases Erk5 activation and accelerates PDGFRβ internalization and downregulation resulting in a specific inhibition of Signal transducers and activators of transcription (Stat) 3, Src and protein kinase C (PKC), and partially of p38, Stat1/5 and Phoshoplipase Cγ (PLCγ). In paper III, we report that DUSP6/MKP-3 creates a negative cross-talk between Erk1/2 and Erk5 and an auto-inhibitory feedback loop on the PI3-kinase/Akt pathway. In paper IV, we identify a new regulative mechanism of the PDGF pathway. PDGF induces Erk5 expression and activation that modulates the PDGFRβ activity. After Erk5 downregulation, the receptor undergoes to a faster and stronger activation that results in a faster internalization and degradation. In conclusion, we present a mechanism through which the PDGF/MAP kinases support tumor growth, and elucidate different regulatory pathways involved in PDGF signaling.

Abstract:
This thesis is an account of research into a novel type of X-ray optic - the microchannel plate (MCP). Experiments to determine the point to point focusing properties of square pore MCPs manufactured by Galileo Electro-Optics and Philips Photonics are reported. These were performed both in a test chamber at Leicester with an electron-bombardment X-ray source and with a laser-plasma X-ray source at the Rutherford Appleton Laboratory. A resolution of 6 arcmin and an intensity gain of 20 were recorded using a Galileo focusing MCP. An invesigation into the focusing action of MCPs which have been curved to a spherical figure is detailed. Such curved MCPs may, in a manner reminiscent of a conventional refractive lens, be used to focus a parallel X-ray beam to a point forming the basis of an X-ray telescope, or conversely to convert the diverging beam from a point-like X-ray source to a quasi-parallel one. The curving experiments were performed by Philips Photonics on standard circular pore MCPs. Tests on these plates were performed at Leicester; the results appear favourable. The technique has been applied to MCPs of up to 4mm thickness, curving them to a radius of 1.4m (0.7m focal length). A comprehensive Monte Carlo ray-tracing model is presented. This was initially developed to facilitate an understanding of the geometry of MCP focusing and to produce idealised images corresponding to a given experimental set up. These perfect images may be compared with, or used to predict experimental results. Comparison with experimental results led to incorporation of MCP distortions into the code. These distortions have been found by a programme of metrology which is also described. The model has led to a clear insight into the causes of poor image quality and their relative importance.
