Abstract:
Treatment of diseases of the musculoskeletal system to date remains not only a serious medical, but also socio-economic problem. Chronic diseases of the joints and spine are one of the most common causes of disability in patients. The main clinical manifestations of most diseases of the musculoskeletal system are pain and inflammation, which requires adequate antiinflammatory therapy. The data on various mechanisms of action of non-steroidal anti-inflammatory drug (NSAID) diclofenac (Voltaren), as well as the results of clinical studies and meta-analyses confirming its effectiveness and good tolerability are presented. The data of experimental and clinical studies of the efficacy and safety of local forms of diclofenac-Voltaren Emulgel (diclofenac-diethylamine) –are presented. Provides information about the form of the Voltaren Emulgel 2% for 12 hours. It is shown that a wide range of analgesic and anti-inflammatory effects of diclofenac (Voltaren), and good tolerance can be used as symptomatic therapy in the treatment of diseases of the musculoskeletal system.

Abstract:
The article describes a method of manual manipulation where the necessary movement of the element of the musculoskeletal system, which was selected for exposure, is performed in accordance with the principles of achieving vibrational and translational mechanical resonance. This method of mechanical and resonance manipulation proved to be more effective and less traumatic in clinical practice when compared with the traditional manipulation method, and the intended effect, which consisted in increasing mobility and more physiologically correct arrangement of the exposed elements of the musculoskeletal system, was more pronounced, namely, greater mobility was achieved with a more uniform increase in the mobility of all the exposed elements of the musculoskeletal system. At the same time, fewer manual therapy sessions by 30% were required for achieving the planned therapeutic effect, and, due to less trauma to biological tissues during mechanical and resonance manipulation, the sessions were less painful.

Abstract:
Musculoskeletal ultrasound (MSKUS) has been recognized as an important method for the evaluation of diseases of the musculoskeletal system, and contrast-enhanced ultrasound (CEUS) technology is becoming an important branch of it. The development of novel materials and tiny nano-formulations has further expanded ultrasound contrast agents (UCAs) into the field of nanotechnology. Over the years, nanoscale contrast agents have been found to play an unexpected role in the integration of precise imaging for diagnosis and treatment of numerous diseases. It has been demonstrated that nanoscale UCAs (nUCAs) have advantages in imaging over conventional contrast agents, including superior biocompatibility, serum stability, and longer lifetime. The potential value of nUCAs in the musculoskeletal system is that they provide more reliable and clinically valuable guidance for the diagnosis, treatment, and follow-up of related diseases. The frontier of advances in nUCAs, their applications, and insights in MSKUS are reviewed in this paper.

Abstract:
Successful implementation of modern treatment modalities for children with oncological and hematological diseases resulted in significant increase in overall and relapse-free survival. These patients are at risk of developing musculoskeletal late effects. The assessment of musculoskeletal late effects and mineral metabolism and the development of solutions for comprehensive rehabilitation and prevention of late effects are challenging issues for pediatrics. A literature review summarizes information related to musculoskeletal late effects associated with special treatment and demonstrates a need for the optimization of diagnostic approaches and corrective therapy of mineral metabolism. Up-to-date multidisciplinary rehabilitation treatment for children in remission can correct the consequences of their main disease and treatment-related side effects.

Abstract:
This review article elucidates the application and viability of electromagnetic fields as therapy and rehabilitation method for the treatment of musculoskeletal pathologies. All studies were found in different databases such as Science Direct, Scielo, IEEE, ProQuest, Pubmed, among others, where the search for information contemplated a period of 5 years (2016-2020). Results evidenced that electromagnetic fields used to treat musculoskeletal conditions showed positive results and improvements in reducing chronic and acute pain in different musculoskeletal pathologies such as osteoarthritis, osteoporosis, nonunions, fractures, fibromyalgia, muscular system injuries, among others. It was possible to evidence that one of the most used magnetic treatments were pulsed electromagnetic fields, which have been implemented to treat joint, muscle and nervous diseases. Additionally, several devices to generate the magnetic stimulation have been designed with a therapeutic and rehabilitation approach, which elucidate the progress in the implementation and acceptance of this type of therapeutic alternatives to treat musculoskeletal conditions. Finally, magnetic stimulation has been also implemented to enhance biomaterials function, evidencing that scaffolds stimulated with electromagnetic fields improve musculoskeletal tissue regeneration.

Abstract:
Infectious diseases complications remain one of the main problems of surgery. Infections are one of the causes of canser patients morbidity and mortality, along with tumor diseases. The frequency of postoperative wound complications varies between 3-34% depending on the type of surgery [5]. Cancer patients are more susceptible to the systematic immunosuppresive state, caused by malignant neoplasms and directly by antitumor therapy. It is obvious that the anticancer problem of treatment and prevention of secondary infections in these conditions becomes even more urgent than before. Objective: To analise infections complications (IC) caused by microorganisms depending on antitumor treatment in cancer patients. Materials and methods: The study included 41 (100%) cancer patients with infectious complications during antitumor treatment in the Azerbaijan Medical University (AMU) Cancer clinic for skin and soft tissue neoplasms. Most often, the pathological process was lokalized in the lower extremities in 19 (46%) patients; in the upper extremities in 14 (34,1%) patients: in the trunk in 8 (19,5%) patients. Of these 12 patients (29,2%) patients received treatment for posteopertive complicattions, 10 (24%) patients received treatment for complications – related to chemotherapy and 19 (46%) patients received treatmentrelated to radiation therapy. There were 28 (68%) men and 13 woman (36,5%) patients. The age of the patients ranged from 34 to 82 years among the studied patients with soft tissue tumors 24 (58%), skin tumors 17 (41,4%) patients .

Abstract:
Introduction First described in 2002, osteonecrosis of the jaw (ONJ, or avascular necrosis of the jaw) is an uncommon but potentially serious side effect of treatment with bisphosphonates. Although typically identified in patients with multiple myeloma and other malignancies, a few cases have been reported in patients taking bisphosphonates - a potent drug class used in the treatment of osteoclast-mediated bone resorption issues, including postmenopausal osteoporosis, Paget's disease, multiple myeloma, and malignant hypercalcemia. The clinical diagnosis of ONJ can be obscured by jaw pain, abscess, swelling, and fistulas, but exposed bone is a distinctive sign. This reports a case of ONJ secondary to bisphosphonate use in a 65-year-old woman and clinical management complications. Case Presentation A 65-year-old lady with history of age-related osteoporosis and compression fractures on alendronate for 4 years, squamous cell carcinoma of neck status post excision and radiotherapy 11-years prior, Sjogren's syndrome and discoid lupus on hydroxychloroquine, diabetes, hypertension, stroke and multiple dental abscesses presents with persistent neck pain. Initial CT neck with contrast showed diffuse fat stranding. Subsequently, alendronate was discontinued due to jaw necrosis suspicion. Eight months later, repeat CT scan showed new non-mass-like soft tissue thickening in the subcutaneous fat abutting the right anterior mandible with mandibular teeth cavities and periapical lucencies, likely to be periodontal cellulitis versus radiation osteonecrosis. Later, patient complained of a piece of bone penetrating the skin of her chin and presented with continuous drainage from sinus tract in her mandible, which was diagnosed as osteonecrosis attributed to bisphosphonates, previous radiation therapy, and dental abscesses. Patient was started on abaloparatide, an osteo-anabolic medication for osteoporosis and enrolled in hyperbaric oxygen therapy which immensely helped in controlling sinus drainage. Patient is currently awaiting mandibular reconstruction surgery. Discussion ONJ, often associated with pain, swelling, exposed bone, local infection, and pathologic fracture of the jaw, is a rare complication of bisphosphonate therapy. Currently, no prospective data exists to advise the benefits of therapy discontinuation however most clinical practices tend to discontinue at least temporarily. The incidence increases with longer treatment duration, particularly when therapy exceeds four years. Risk factors for developing ONJ while taking bisphosphonates include IV administration, anticancer therapy, dose and duration of exposure, dental extractions/implants, glucocorticoids, smoking, diabetes, and preexisting dental disease. Case reports and series suggest benefit from hyperbaric oxygen therapy in wound healing, pain, and quality of life at three months, however no significant differences exist with outcomes beyond three months. Patients being considered for therapy with a bisphosphonate should be thoroughly evaluated for dental issues, prior to initiating therapy. Conservative management with limited debridement, antibiotic therapy as needed, and topical mouth rinses rather than aggressive surgical resection are recommended. Conservative therapy may result in healing in a significant proportion of patients. Surgical resection of necrotic bone should be reserved for refractory or advanced cases. Providers should remain cautious while prescribing high doses of bisphosphonates in patients with increased risk factors to prevent, timely diagnose and treat this condition. Abstract:
Thesis (MSc)--University of Stellenbosch, 2007.
ENGLISH ABSTRACT: Tumour necrosis factor alpha (TNFα) is a pleiotropic cytokine and has a wide variety of dose dependent cellular effects ranging from cell growth and differentiation, to inducing apoptosis. It has long been implicated in muscle and non-muscle inflammatory disorders, such as muscle wasting in chronic disease states, and rheumatoid arthritis. However, a physiological role for TNFα in muscle regeneration has been proposed as elevated levels of the cytokine are present when muscle regeneration processes are initiated: TNFα is secreted by infiltrating inflammatory cells, and by injured muscle fibres. Adult skeletal muscle contains a population of resident stem cell-like cells called satellite cells, which become activated, proliferate and differentiate following muscle injury to bring about repair of damaged muscle. Much research on the effects of TNFα on satellite cell differentiation has been conducted in recent years. It is however difficult to get a complete characterisation of the cytokine’s action as all models used slightly differ. We aimed therefore at providing comprehensive assessment of the effects of increasing doses of chronically supplemented TNFα on differentiating C2C12 cells. Cells were allowed to differentiate with or without TNFα supplementation for 7 days. Differentiation was induced at day 0. The effect on differentiation was assessed at days 1, 3, 5, and 7 by western blot analysis, and supplementary immunohistochemical analysis at days 1, 4, and 7 of markers of differentiation - muscle regulatory factors: MyoD and myogenin, markers of the cell cycle p21, PCNA, and the integral signalling molecule, p38MAPK. TNFα supplementation at day 1 tended to positively regulate early markers of differentiation. With continued supplementation however, markers of differentiation decreased dose dependently in treated cultures as the initial effect appeared to be reversed: A trend towards a dose dependent decrease in MyoD, myogenin and p21 protein existed in treated cultures at days 3, 5, and 7. These findings were significant at day 5 (p21, p<0.05), and day 7 (myogenin, p<0.05). A significant dose dependent decrease in p38 phosphorylation was evident at day 3 (p<0.05), while phospho-p38 was dose dependently increased at day 7 (p<0.05). Taken together, these data show that TNFα supplementation for 24 hours following the induction of differentiation in vitro, tends to increase levels of early markers of differentiation, and with continued TNFα supplementation decrease markers of differentiation in a dose dependent fashion. This study provides a comprehensive characterisation of the dose and time dependent effects of TNFα on satellite cell differentiaton in vitro. The model system used in the current study, allows us to make conclusions on more chronic disease states.
Selle was vir 7 dae met of sonder TNFα supplementasie gedifferentieër. Differensiasie was by Dag 0 geïnduseer. Hierdie bevindinge was beduidend by dag 5 (p21, p<0.05), en dag 7 (miogenien, p<0.05). A beduidende dosis afhanklike afname in p38 fosforilasie was duidelik by dag 3 (p<0.05), terwyl fosfo-p38 by dag 7 verhoog het met verhoogde konsentrasie TNFα (p<0.05).

Abstract:
The goal of this research was to develop a system of injectable hydrogels to deliver stem cells to musculoskeletal defects, thereby allowing cells to remain at the treatment site and secrete soluble factors that will facilitate tissue regeneration. First, production parameters for encapsulating cells in microbeads were determined. This involved investigating the effects of osmolytes on alginate microbead properties, and the effects of alginate microbead cell density, alginate microbead density, and effects of osteogenic media on microencapsulated cells. Although cells remained viable in the microbeads, alginate does not readily degrade in vivo for six months. Therefore, a method to incorporate alginate lyase in microbeads was developed and optimized to achieve controlled release of viable cells. Effectiveness of this strategy was determined through cell release studies and measuring proteins and expression of genes that are characteristic of the cell’s phenotype. Lastly, in vivo studies were done to assess the ability of alginate microbeads to localize microencapsulated cells and support chondrogenesis and osteogenesis. This project will provide insight to the tissue engineering field regarding cell-based therapies and healing musculoskeletal defects.

Abstract:
Arthrogryposis, a congenital disorder characterized by multiple joint contractures, can limit one’s ability to perform even the simplest of tasks. The purpose of this paper is to outline the general limitations associated with arthrogryposis and examine the most common corrective procedures used to treat and manage deformities of the lower extremities. While the ultimate goal may be complete correction of the associated deformities, this may not be practical, as recurrence of contractures is common. Surgical and non-surgical methods discussed in this paper include casting with the Ponseti Method, use of bracing and night splinting, soft tissue release for the ankle and knee, talectomy and osteotomy procedures for the knee. The conclusions discussed in this paper determine that complete correction is not typically obtained, but quality of life can be improved through functional independence and ambulation when utilized in conjunction with thorough physical therapy rehabilitation.

Abstract:
The focus of the present study was on the investigation of the effects of an intervention strategy on an industrial task in situ and a simulation of the same task within a laboratory setting. The task of offloading crates from a truck at a local business was simulated in a laboratory setting for rigorous analysis. The effect of an ergonomically sound intervention on selected physical, physiological and perceptual variables was evaluated in a test - retest experimental set-up using 28 young, healthy male students. Each of the two experimental conditions lasted for 16 minutes. In the pre-intervention task subjects were required to transfer the crates from one point to another by sliding them along the floor. During the execution of the post-intervention task responses to reductions in the stacking height and modifications of the working method were evaluated. Results obtained for spinal kinematics during the simulated industrial task indicated a high biomechanical risk, due to large ranges of motion, high velocities and accelerations in the sagittal and transverse planes. The heavy workload of the task was also evident in elevated physiological responses (HR, RF, VT, VE, VO2, RQ, EE) and perceptual ratings (RPE, Body Discomfort). Assessment of the intervention strategy revealed that the ‘high risk’ industrial task was reduced to ‘moderate acceptable’, with measurements of spinal kinematics, physiological and perceptual variables being significantly reduced. An in situ re-assessment of the workers’ responses to the intervention also elicited reductions in heart rates and perceptual ratings compared to the original task.

Abstract:
AbstractAll components of the musculoskeletal system can be involved by metabolic disorders as a result of endocrine diseases, genetic alterations, and environmental or nutritional aspects, with important worldwide variations in prevalence and severity. Early detection of these disorders is crucial because of the efficacy of preventive measures and availability of treatments. The current chapter will focus on the imaging appearance of metabolic disorders of bone marrow and of the mineralized skeleton. Marrow and bone disorders in athletes, the elderly, and individuals with eating disorders will be reviewed.

Abstract:
Introduction: Although respiratory symptoms are the characteristic findings of COVID- 19, this disease may also present with neurological manifestations, since SARS-CoV- 2 showed several degrees of neurotropism. Objectives: To review the influence of SARS-CoV-2 infection on the development of peripheral neuropathies. Methods: Integrative review carried out at PUBMED with the descriptors peripheral neuropathy, SARS-CoV-2 and COVID-19, and having as inclusion criteria full texts and in English in the period from 11/2019 to 05/2021. The selection has made among those compatible with the objectives of the work. Results: Peripheral neuropathy was observed in less than 1% of patients with COVID-19. The virus can cause acute polyradiculoneuropathy regardless of pulmonary disease, and may occur due to dysregulation of the immune system caused by SARS-CoV-2. Systemic hyper-inflammation with macrophage activation syndrome has been proposed for patients with COVID-19. Such immunomediated manifestations typically occur after the decrease in the acute phase of disease. The most commonly reported symptoms in peripheral nervous system involvement are ageusia, anosmia, diplopia, facial nerve paralysis, polyneuritis, myasthenic crisis, musculoskeletal injuries, and neuralgia. Conclusion: As soon as possible recognition of peripheral neuropathy may result in better clinical goals for patients and understanding these manifestations will contribute to the development of improved treatment. Although only a small percentage of patients with COVID-19 develop peripheral neuropathy, in a pandemic this can have a major impact.

Abstract:
The paper focuses on the gait analysis for the investigation of the typical events occurring in human movements and validate its use as a method for musculoskeletal disease evaluation and for the improvement of athletic training. In the present research the motion capture system is combined with an in-house developed prototype of uniaxial force plates for the measurement of the vertical component of ground reaction forces during movement. While similar techniques are implemented for gait, this equipment can be employed to investigate running, thus, covering a larger number of possible applications and providing a deeper insight either of the athlete performance or the disease analysis. For the prevention and the treatment of those events occurring during running, a thorough understanding of its mechanisms is critical; therefore, a method for evaluating both the kinematic behavior of the human body and the ground reaction forces combined to a model for determining the muscle forces is proposed. An infrared motion capture technique is adopted for measuring accurately the body motion and a multiple force-plate system is used to calculate the force exerted by the ground and sub-divided in the three components by an ad-hoc developed routine. Moreover, the data are used as input parameters for the OpenSim software to derive muscles forces. Finally, the potential of the proposed protocol is determined by an experimental campaign on healthy subjects and a significant database of muscle forces is constructed for different running speeds.

Abstract:
Introduction. The economic losses associated with disability due to diseases of the circulatory system, as well as the costs of providing medical care to patients suffering from heart and vascular diseases, are increasing annually. The state preventive measures currently being carried out are of a delayed nature. The results of the medical examination of the population of the Russian Federation in recent years (2015-2019) indicate that the incidence of cardiovascular diseases, including hypertension, is at a fairly high level. In the middle of the last century, the Concept of risk factors for the development of chronic non-communicable diseases were formulated, in the structure of which cardiovascular diseases, including arterial hypertension, occupies one of the primary positions. The concept is based on the results of promising epidemiological studies, and, at present, is a methodological basis for planning and organizing primary prevention of cardiovascular diseases. The purpose of the study. Based on the analysis of literary sources (including foreign ones) containing experience in the use of telemedicine technologies, to assess their significance for the prevention, diagnosis and treatment of hypertension, as well as forecasting improvements in the quality of medical care when adapting to the use of clinical recommendations. Materials and methods. The article provides an analytical review of the use of modern telemedicine technologies in the prevention of hypertension. The results of the study and their discussion. The analysis of literary sources has shown that in the context of the progress of information and telecommunication technologies in the healthcare system, a fundamentally new direction has appeared in the organization and provision of medical care to the population - telemedicine, which will ensure the modern level of prevention, detection and treatment of chronic non-communicable diseases, and also determines positive medical, social and economic performance indicators. To date, updates in the legislative framework of the Russian Federation are aimed at ensuring that medical care with the use of telemedicine technologies is more widespread, taking into account the standards of medical care and clinical recommendations. Conclusion. Based on a review of literature sources, it has been established that the modern solution to the problem of improving the quality of medical care for patients, including those with hypertension, diseases is medical care using telemedicine technologies that prove their medical, social and economic effectiveness.

Abstract:
Review question / Objective: The type of study was clinical randomized controlled trial (RCT). The object of study is the patients with HFMD. There is no limit to gender and race. In the case of clear diagnosis standard, curative effect judgment standard and consistent baseline treatment, the experimental group was treated with pure oral liquid of traditional Chinese medicine(A: Fuganlin oral liquid, B: huangzhihua oral liquid, C: Lanqin oral liquid, D: antiviral oral liquid, E: Huangqin oral liquid, F: Pudilan oral liquid, G: Shuanghuanglian oral liquid.)and the control group was treated with ribavirin or any oral liquid of traditional Chinese medicine. The data were extracted by two researchers independently, cross checked and reviewed according to the pre-determined tables. The data extraction content is (1) Basic information (including the first author, published journal and year, research topic). (2) Relevant information (including number of cases, total number of cases, gender, age, intervention measures, course of treatment of the experimental group and the control group in the literature). (3) Design type and quality evaluation information of the included literature. (4) Outcome measures (effective rate, healing time of oral ulcer, regression time of hand and foot rash, regression time of fever, adverse reactions.). The seven traditional Chinese medicine oral liquids are comparable in clinical practice, but their actual clinical efficacy is lack of evidence-based basis. Therefore, the purpose of this study is to use the network meta-analysis method to integrate the clinical relevant evidence of direct and indirect comparative relationship, to make quantitative comprehensive statistical analysis and sequencing of different oral liquid of traditional Chinese medicine with the same evidence body for the treatment of the disease, and then to explore the advantages and disadvantages of the efficacy and safety of different oral liquid of traditional Chinese medicine to get the best treatment plan, so as to provide reference value and evidence-based medicine evidence for clinical optimization of drug selection. Condition being studied: Hand foot mouth disease (HFMD) is a common infectious disease in pediatrics caused by a variety of enteroviruses. Its clinical manifestations are mainly characterized by persistent fever, hand foot rash, oral herpes, ulcers, etc. Because it is often found in preschool children, its immune system development is not perfect, so it is very vulnerable to infection by pathogens and epidemic diseases, resulting in rapid progress of the disease. A few patients will also have neurogenic pulmonary edema Meningitis, myocarditis and other serious complications even lead to death, so effectively improve the cure rate, shorten the course of disease, prevent the deterioration of the disease as the focus of the study. In recent years, traditional Chinese medicine has played an important role in the research of antiviral treatment. Many clinical practices have confirmed that oral liquid of traditional Chinese medicine can effectively play the role of antiviral and improve the body's immunity.

Abstract:
Review question / Objective: The aim of this study is to perform a meta-analysis to evaluate the effectiveness of electroacupuncture in the treatment of painful diabetic peripheral neuropathy (PDNP).And to provide data support for electroacupuncture as an effective means to treat pain of nervous system diseases. Condition being studied: Diabetes mellitus (DM) affects more than six hundred million population worldwide till 2045. The most common form is chronic, distal, and symmetric sensorimotor polyneuropathy, while other uncommon forms include asymmetric or focal neuropathy, such as diabetic muscle atrophy, trunk radiculopathy, and compression palsy. About 11.4% and 40.5% of patients have severe and moderate pain respectively. Currently, symptomatic treatment of PDPN is based on the application of medications that target the symptoms of PDPN. However, the clinical efficacy of PDPN patients varies greatly from individual to individual.Traditional Chinese medicine electroacupuncture have shown its unique advantages in the treatment of PDPN. Although its mechanism is complex and unclear, it can still be used in the clinical treatment of PDPN for a long time. We therefore present a systematic review of the benefits of electroacupuncture in improving PDPN by including the as many as possible randomized controlled trials.

Abstract:
Project Title: "Development of a plasmid-based reverse genetics system for the Bluetongue and Epizootic Hemorrhagic Disease viruses to allow comparative characterization of the function of the NS3 viroporin in viral egress". Project details: No - IS-4192-09; Participants – Ehrlich M. (Tel Aviv University), Parker J.S. (Cornell University), DermodyT.S. (Vanderbilt University); Period - 2009-2013. Orbiviruses are insect-borne infectious agents of ruminants that cause diseases with considerable economical impact in Israel and the United States. The recent outbreaks of BTV in Europe and of Epizootic Hemorrhagic Disease Virus (EHDV) in Israel, underscore the need for: (i) a better comprehension of the infection process of orbiviruses, (ii) the identification of unique vs. common traits among different orbiviruses, (iii) the development of novel diagnosis and treatment techniques and approaches; all aimed at the achievement of more effective control and treatment measures. It is the context of these broad goals that the present project was carried out. To fulfill our long-term goal of identifying specific viral determinants of virulence, growth, and transmission of the orbiviruses, we proposed to: (i) develop reverse genetics systems for BTV and EHDV2-Ibaraki; and (ii) identify the molecular determinants of the NS3 nonstructural protein related to viroporin/viral egress activities. The first objective was pursued with a two-pronged approach: (i) development of a plasmid-based reverse genetics system for BTV-17, and (ii) development of an "in-vitro" transcription-based reverse genetics system for EHDV2-Ibaraki. Both approaches encountered technical problems that hampered their achievement. However, dissection of the possible causes of the failure to achieve viral spread of EHDV2-Ibaraki, following the transfection of in-vitro transcribed genomic segments of the virus, revealed a novel characteristic of EHDV2-Ibaraki infection: an uncharacteristically low fold increase in titer upon infection of different cell models. To address the function and regulation of NS3 we employed the following approaches: (i) development (together with Anima Cell Metrology) of a novel technique (based on the transfection of fluorescently-labeledtRNAs) that allows for the detection of the levels of synthesis of individual viral proteins (i.e. NS3) in single cells; (ii) development of a siRNA-mediated knockdown approach for the reduction in levels of expression of NS3 in EHDV2-Ibaraki infected cells; (iii) biochemical and microscopy-based analysis of the localization, levels and post-translational modifications of NS3 in infected cells. In addition, we identified the altered regulation and spatial compartmentalization of protein synthesis in cells infected with EHDV2-Ibaraki or the mammalian reovirus. In EHDV2-Ibaraki-infected cells such altered regulation in protein synthesis occurs in the context of a cell stress reponse that includes the induction of apoptosis, autophagy and activation of the stressrelated kinase c-Jun N-terminal Kinase (JNK). Interestingly, inhibition of such stress-related cellular processes diminishes the production of infectious virions, suggesting that EHDV usurps these responses for the benefit of efficient infection. Taken together, while the present project fell short of the generation of novel reverse genetics systems for orbiviruses, the development of novel experimental approaches and techniques, and their employment in the analysis of EHDV-infected cells, yielded novel insights in the interactions of orbiviruses with mammalian cells.

Abstract:
Objectives: (1) Evaluate Immunocompetence-OTL-containing Chromosomal Regions (ICRs), marked by microsatellites or candidate genes, for magnitude of direct effect and for contribution to relationships among multiple immunocompetence, disease-resistance, and growth traits, in order to estimate epistatic and pleiotropic effects and to predict the potential breeding applications of such markers. (2) Evaluate the interaction of the ICRs with genetic backgrounds from multiple sources and of multiple levels of genetic variation, in order to predict the general applicability of molecular genetic markers across widely varied populations. Background: Diseases cause substantial economic losses to animal producers. Emerging pathogens, vaccine failures and intense management systems increase the impact of diseases on animal production. Moreover, zoonotic pathogens are a threat to human food safety when microbiological contamination of animal products occurs. Consumers are increasingly concerned about drug residues and antibiotic- resistant pathogens derived from animal products. The project used contemporary scientific technologies to investigate the genetics of chicken resistance to infectious disease. Genetic enhancement of the innate resistance of chicken populations provides a sustainable and ecologically sound approach to reduce microbial loads in agricultural populations. In turn, animals will be produced more efficiently with less need for drug treatment and will pose less of a potential food-safety hazard. Major achievements, conclusions and implications:. The PI and co-PIs had developed a refined research plan, aiming at the original but more focused objectives, that could be well-accomplished with the reduced awarded support. The successful conduct of that research over the past four years has yielded substantial new information about the genes and genetic markers that are associated with response to two important poultry pathogens, Salmonella enteritidis (SE) and Escherichia coli (EC), about variation of immunocompetence genes in poultry, about relationships of traits of immune response and production, and about interaction of genes with environment and with other genes and genetic background. The current BARD work has generated a base of knowledge and expertise regarding the genetic variation underlying the traits of immunocompetence and disease resistance. In addition, unique genetic resource populations of chickens have been established in the course of the current project, and they are essential for continued projects. The US laboratory has made considerable progress in studies of the genetics of resistance to SE. Microsatellite-marked chromosomal regions and several specific genes were linked to SE vaccine response or bacterial burden and the important phenomenon of gene interaction was identified in this system. In total, these studies demonstrate the role of genetics in SE response, the utility of the existing resource population, and the expertise of the research group in conducting such experiments. The Israeli laboratories had showed that the lines developed by selection for high or low level of antibody (Ab) response to EC differ similarly in Ab response to several other viral and bacterial pathogens, indicating the existence of a genetic control of general capacity of Ab response in young broilers. It was also found that the 10w-Ab line has developed, possibly via compensatory "natural" selection, higher cellular immune response. At the DNA levels, markers supposedly linked to immune response were identified, as well as SNP in the MHC, a candidate gene responsible for genetic differences in immunocompetence of chickens.
