Abstract:
This research represents an attempt to define and accurately measure user consumption of content on a modern, advertised VOD service (AVOD), more specifically known in Sweden as a Play service. With a foundation of previous research in the area of VOD and AVOD services, the characteristics and flaws of these types of platforms are discussed to shine light on factors that might concern Play services. Optimizing the vast content inventory offered on these services is crucial for long term profitability, and to achieve this, content providers need to understanding how to measure the consumption properly. A content­centric approach was used to focus on individual formats (e.g. TV shows) and the factors that can describe its consumption. A macro perspective was initially applied to investigate global factors that dictates consumption. Analysis on a micro level was carried out by analyzing tracking data collected for a full year from one of the biggest Play services in Sweden, TV3play.se. Ultimately, the development of a new method to measure consumption called Consumption Volume Score (CVS) is proposed. It is introduced as an alternative to the traditional unit which has been to measure the amount of video starts (VS). The validity was evaluated using comparison of rank difference for individual formats, using both methods and different criterias. The results shows that the method of using CVS to measure consumption yields little to no difference in ranking of highly popular formats, while less consumed formats had a more varied change in rank. Further analysis on some of these formats indicated that they might have a dedicated niche audience, where content editors might see potential gains from handpicking them to optimize the consumption further. The findings gives support to believe that CVS as a unit of measuring consumption can help to further understand how individual formats perform, especially less consumed and potentially niched ones. Future research on CVS is recommended to discern its feasibility in a live context.
Denna forskning ämnar att finna metoder för att definiera samt noggrant kunna mäta konsumtionen av streamad media på en modern, reklamfinansierad VOD­tjänst (AVOD), i Sverige är dessa mer kända som Play­tjänster. Med utgångspunkt från tidigare forskning inom på streaming­tjänster, belyses de egenskaper och brister dessa typer av plattformar kan påverkas av, i synnerhet de faktorer som kan vara relevanta för Play­tjänster. Att kunna organisera och underhålla de stora mängder media som erbjuds på dessa tjänster är avgörande för långsiktig lönsamhet, och för att uppnå detta måste redaktörerna förstå hur man mäter konsumtionen med hög tillförlitlighet i sin uppgift att ta väl grundade beslut. En innehållscentrerat (content­centric) tillvägagångssätt användes för att kunna fokusera på enskilda format (t.ex. TV­program) och de inbördes faktorer som kan beskriva dess konsumtion. Ett makroperspektiv applicerades initialt för att undersöka de globala faktorer som styr konsumtion. Vidare tillämpades en analys på mikronivå genom att undersöka spårningsdata kopplat till det innehåll som finns tillgängligt på plattformen. Underlaget för spårningsdatan bestod av ett helt års konsumtion (2014) från en av de största Play­tjänsterna i Sverige, TV3play.se. Framtagning av en ny metod för att mäta konsumtionen kallad Consumption Volume Score (CVS) föreslås och införs som ett alternativ till den traditionella metod som har varit att mäta den totala mängden av Videostarter (VS). Signifikansen av den nya metoden utvärderades genom jämförelse av rangordning av enskilda format baserat på CVS och VS samt olika kriterier. Resultaten visar att CVS för att mäta konsumtion ger ytterst liten eller ingen skillnad i ranking av mycket populära format, medan mindre konsumerade format hade en mer varierad förändring i rang. Vidare analys av en del av dessa format indikerade att de skulle kunna ha en nischad publik som uppskattar innehållet, trots relativt låg konsumption. För dessa format anser jag att det finns möjlighet för redaktörer att manuellt handplocka dem för att optimera konsumtionen ytterligare. Resultaten ger underlag för godta CVS som en signifikant mätenhet för konsumtion och kan bidra till att förstå hur enskilda format presterar, särskilt mindre konsumerade och potentiellt nischade sådana. Framtida forskning om CVS som metod för att mäta konsumtion rekommenderas, i synnerhet för att avgöra hur väl det lämpas att applicera i en skarp miljö.

This thesis aims to analyze the consumer purchase decision process in the market for bicycles in all its stages. It sets to involve significant specifics that have any bearing on this process. The thesis is divided into several parts. The theoretical part deals with marketing tools applicable within the bicycle market considering bicycle a complex product with all related components. The following part goes into great detail analyzing the purchase decision process in the bicycle market, most of the space being devoted to the actual PDP stages. The last part handles the question of marketing research generally. The practical part of this thesis draws on the results of a marketing research carried out by means of questionnaires. Written questioning played vital part in primary data collection. This work also presents information concerning the questionnaire research. Then, socio-demographic issues are analyzed which specify more closely the questioned-group structure. The statistical analysis of each one question in the questionnaire presents the crucial part which involves both a first stage analysis of all the questions used and a two-dimensional statistical analysis that gives an insight into the coherence between selected questions. Also included is summary of results and formulating marketing recommendations for bicycle manufacturers and sellers.

Abstract:

Syftet – Syftet med denna uppsats är att studera hur MTG Radio och TV4 har valt att använda sig av grön marknadsföring. Vidare undersöker vi om företagen har ett helhetsperspektiv med den gröna marknadsföringen, det vill säga om de tydligt arbetar med miljöfrågorna både externt och internt.

Design/metodologi/tillvägagångssätt – Efter en kort historisk bakgrund redogör vi för den teoretiska bakgrunden till hur företag bör utforma den gröna marknadsföringen så att den effektivt når ut till kunder. Vi studerar sedan två företag, MTG Radio och TV4, för att se hur dessa arbetar med grön marknadsföring. Resultaten från studierna förs sedan samman med teorin till en analytisk diskussion.

Resultat – De studerade företagen har uppvisat delvis skilda tillvägagångssätt för hur de använder sig av grön marknadsföring. Största skillnaderna ligger i att MTG Radio arbetar internt i linje med vad dagens teoribildning menar att företag ska göra, och mycket av TV4:s externa arbete är utformat så att den enskilde individen kan se tydliga individuella fördelar med budskapen. Vi har också funnit en vilja hos företagen att påverka andra organisation-er.

Praktiska implikationer – Eftersom MTG Radio och TV4 arbetar med delvis olika områden inom den gröna marknadsföringen så skulle företagen kunna dra nytta av att studera varandras arbete.

Originalitet/värde – Vad vi erfar har inga studier gjorts utifrån hur svenska radio- och televisionsföretag använder sig av grön marknadsföring. Båda företagens önskan att påverka andra organisationer är något som inte heller finns diskuterat i dagens forskning om grön marknadsföring.

Abstract:
Un des problèmes majeurs en enzymologie est la fonctionnalisation de liaisons C-H peu réactives. Ces réactions nécessitent des cofacteurs spécialisés tels que l'hème, des Fer-oxo ou encore la vitamine B12. En 2001, il a été montré que des centres Fe-S particuliers liant la S-Adénosyle méthionine (SAM) pouvaient activer des liaisons C-H non réactives. Les enzymes utilisant ce cofacteur constituent une super-famille appelée " Radical SAM ". Les thiométhyltransférases (MTTases) sont des enzymes " Radical SAM " qui catalysent l'insertion d'un groupe thiométhyle (-SCH3) dans des liaisons C-H non réactives. Par des expériences in vivo et in vitro, nous avons montré qu'on pouvait les regrouper en trois classes. La première classe (RimO) catalyse la formation du β-thiométhylaspartate 89 sur la protéine ribosomale S12 (β-ms-D89-S12) alors que les deux dernières (MiaB et MtaB/eMtaB) catalysent respectivement la thiométhylation des nucléosides 2-methylthio-N6-isopentenyladenosine 37 (ms2i6A-37) et 2-methylthio-N6-threoninecarbamoyl adenosine 37 (ms2t6A-37) de certains ARNts. L'étude in vitro du mécanisme de ces enzymes a permis de démontrer que les MTTases catalysent l'insertion d'un groupement -SCH3 de façon catalytique invalidant l'hypothèse généralement retenue dans la littérature que le soufre inséré dérive de la destruction d'un centre Fe-S.

Abstract:
Mycobacterium tuberculosis (MTB) is a very successful pathogen, causing the deaths of approximately two million people a year world-wide. Survival of the pathogen in vivo is dependant on its ability to respond and adapt to changes within its environment. One method of adaptation is through two-component signal transduction systems, the phosphotransfer pathways that couple stimuli to responses. Generic two-component systems involve two conserved elements, a membrane bound histidine kinase, which is the sensory protein, and a response-regulator protein that controls the response usually by altering the expression of genes required for adaptive responses. The aim of this thesis is to investigate the structure and function of the sensory protein DosS (DevS). DosS is induced by exposure to hypoxia, NO and ethanol and is the only one of the 11 paired MTB two component systems for which inducers have been identified but the precise chemical nature of the signal is unknown. The N-terminal input region of the DosS sensor contains two putative GAF domains. Various fragments of the N-terminal region were cloned, expressed and purified to homogeneity. Ultraviolet-Visible spectral analysis reveals that full length DosS binds a classical high spin b haem cofactor. Mutagenesis identified histidine 149 of DosS, which is within the N-terminal GAF domain, as critical to haem-binding. This is the first known GAF domain to bind haem and the presence of a haem co-factor is consistent with the postulated involvement of DosS in oxygen and redox sensing. Based on this data a model for histidine kinase activation is suggested. The second GAF domain of DosS was analysed using NMR spectroscopy. Triple resonance NMR experiments enabled the identification and sequential assignment for 99 out of the 141 backbone amide proton and nitrogen resonances. In addition, both of the GAF domains were also subjected to protein crystallisation while the full length DosS was investigated using electron microscopy.

Introdução: O teste Xpert MTB / RIF está sendo cada vez mais utilizado em muitos países como diagnóstico inicial para a tuberculose (TB). Poucos estudos avaliaram o impacto do Xpert no diagnóstico em rotinas de programas de controle de TB no Brasil. O objetivo do presente estudo foi avaliar o impacto da introdução do Xpert MTB / RIF no diagnóstico de TB em uma cidade com alta incidência de TB no Brasil. Métodos: Incluímos pacientes avaliados com testes diagnósticos convencionais durante um ano antes da introdução do Xpert (grupo pré-Xpert) e pacientes avaliados usando Xpert durante um ano após a introdução do teste (grupo pós-Xpert). Resultados: 620 pacientes preencheram os critérios de inclusão (208 no grupo pré-Xpert e 412 no grupo pós-Xpert) e foram incluídos na análise. O tempo até o diagnóstico de TB foi menor no grupo pós-Xpert (0,7 dias, IQR: 0,5-1,0 dias) do que no grupo pré-Xpert (2,0 dias, IQR: 2,0-2,0 dias) (p <0,0001). Características atípicas da doença, como menor perda de peso, febre, dispneia, sudorese noturna e hemoptise; baciloscopia de escarro negativa; cultura negativa e radiografia de tórax atípica de TB foram mais comuns no grupo pós-Xpert do que no grupo pré-Xpert (p <0,0001 para todos). Conclusões: Observamos que a implementação do ensaio Xpert MTB / RIF, em rotinas de programas de controle de TB, melhora e facilita o diagnóstico de tuberculose, especialmente nos casos com manifestações da doença atípica. Esses resultados podem provavelmente ser generalizados para locais com incidência de TB similar.
Introduction: The receptor for advanced glycation end products (RAGE) is expressed in normal lungs and is upregulated during inflammation and infection. The interaction between AGEs and RAGE on the plasma membrane causes oxidative stress and apoptosis in lung cells. The objective of this study is to evaluate plasma levels of AGEs and its soluble receptor (sRAGE) in patients with active TB and healthy controls, and to investigate their relationship with food intake and nutritional status. Methods: Case-control study. AGE (carboxymethil lysine, CML) and RAGE were measured by Elisa. Nutritional assessment was performed by body mass index, triceps skin-fold thickness, mid-arm circumference, mid-arm muscle circumference, bioelectrical impedance analysis, and food frequency questionnaire. Results: 35 TB patients and 35 controls were included in the study. The mean S-RAGE levels were higher in TB patients than in controls (68.5 ± 28.1 vs 57.5 ± 24.0, p=0.046). Among cases that were current smokers, lower S-RAGE levels were associated with mortality (S-RAGE levels= 58.0 ± 36.5 [non-survivors] vs 71.3 ± 25.6 [survivors], p=0.006), and with weight loss (S-RAGE levels= 65.6 ± 27.4 [weight loss] vs 98.6 ± 16.7 [no weight loss], p=0.034). There was no statistically significant difference in CML levels and diet CML content between cases and controls. Malnutrition was more frequent in cases than in controls, but there was no correlation between nutritional parameters and CML or S-RAGE levels. Conclusions: TB patients had higher S-RAGE levels than controls. S-RAGE may play a role in disease manifestations and outcomes, being associated with weight loss and mortality.

Abstract:
This thesis is a study of how the Swedish media company MTG Radio has developed new strategies and production practices in relation to technological change, new competition and media convergence during the first decade of the 2000s. During this period the media landscape in general has been marked by digitization, the rise of new media platforms and competition from new media companies. The study engages in an ethnographical perspective on media production, but also takes its starting point in political-economic theories on media work (Banks 2007, Hesmondhalgh & Baker 2011, Ryan 1992) in order to raise questions about the relation between technological and organizational changes and relations of power in production. Empirically, the thesis builds on interviews with production staff as well as an analysis of production documents and content produced by MTG Radio. The analysis shows that digital production technologies contribute to anincreased automation and centralization of control over editorial decisions, and hence to “de-skilling” (Braverman 1974/1999, Örnebring 2010). On the other hand, strategies of multiplatform production and the organizational changes taking place contribute to an “upskilling” (Edgell 2012) and give DJs and presenters more autonomy and control within production. This strengthened autonomy involves their possibilities for reflexivity and critical self-evaluation, as well as their control over content and production. Finally, the thesis connects these results to the more overarching question of alienation, arguing that upskilling and increased autonomy do not automatically create better jobs within the media house, or necessarily represent emancipatory possibilities within media work, as has been argued in previous research and theory.

This Master's thesis is focused on technical preparation of the MTB front wheel hub and application of SinuTrain 4.7 computer software. The thesis is divided into four thematic parts. The first part introduces the SinuTrain software and briefly describes the MTB wheel hub. The next section contains the hub design and selection of the suitable material for this component. The third part describes production technologies and selection of the turning machine with suitable tools. The last chapter deals with SinuTrain 4.7 software usage and also presents the results of the production simulations.

L’obiettivo di questo elaborato consiste nell’applicazione di alcune possibili tecniche di gestione ottimale del parco ricambi nel caso aziendale dell’affiliata bolognese di Philip Morris International. Nel corso del primo capitolo, verranno approfondite le nozioni teoriche provenienti dalla letteratura scientifica riguardo la gestione delle parti di ricambio. In particolare, saranno definite le caratteristiche distintive di questa particolare tipologia di codici, per poi descrivere in modo dettagliato i principali metodi statistici impiegabili per la previsione del consumo. Quindi, verranno esposte alcune delle metodologie utilizzate nel contesto aziendale odierno per la gestione ottimale del magazzino ricambi, focalizzandosi particolarmente sul criterio del costo globale minimo. Nel secondo capitolo, si descriverà brevemente l’azienda Philip Morris International, concentrandosi sulla sede bolognese chiamata PM MTB (Philip Morris Manufacturing & Technology Bologna). Dopo aver riportato qualche informazione circa il mercato di riferimento e la mission aziendale, verranno presentati gli obiettivi delle affiliate italiane del gruppo, i nuovi prodotti e, infine, il ciclo produttivo. Nel terzo capitolo, verrà innanzitutto analizzato lo stato attuale del magazzino ricambi dello stabilimento di Philip Morris a Valsamoggia, così da poter selezionare un set di ricambi definiti più critici su cui svolgere l’intero processo di ottimizzazione. Successivamente, verranno applicati al campione selezionato alcuni modelli di previsione del consumo, al fine di confrontarli per verificarne le performance. Nel quarto e ultimo capitolo verrà utilizzato il criterio del costo globale minimo per esaminare il problema di definizione del numero ottimale di ricambi da mantenere a scorta da un punto di vista prettamente economico. Saranno quindi confrontati i risultati ottenuti definendo l’impatto economico della soluzione proposta.

Lymphoblastic leukemia 1 (Lyl1) is a well-studied transcription factor known to exhibit oncogenic potential during various forms of leukemia. Since its discovery in 1989, many reports have been published describing its relationship with cancer as well as demonstrating its function during hematopoiesis. Lyl1 has been shown to serve a significant role during thymopoiesis by contributing to T-cell development. However, it has been recently reported that irrespective of its significance during T-cell development, mature comparable single positive T-cells are observed in mouse models. The use of murine models has been crucial in identifying potential targets for host-directed therapies (HDT) which has been shown to provide great potential in treating tuberculosis (TB). It is evident that Mycobacterium tuberculosis (Mtb), the causative agent for TB, is capable of developing resistance to various treatments that target the bacterium itself. Therefore, by designing therapies that directly target host factors could assist in circumventing Mtb resistance. By analyzing Mtb-infected bone marrow-derived macrophages (BMDM) that have been subjected to genome-wide transcriptional deep sequencing of total RNA using a single molecule sequencer in conjunction with the cap analysis gene expression (CAGE) technique, various differentially expressed genes were identified, including the oncogenic transcription factor, Lyl1. With the use of murine models, we investigated whether Lyl1 is important for various immunological responses at steady state, the regulation of Lyl1 in response to various immune stimulants including LPS and whether this transcription factor is relevant in bacterial infections including Listeria monocytogenes (Lm) and Mtb. The data in this thesis demonstrate comparable immunological responses, including cellular recruitment by means of flow cytometry and cytokine responses by means of ELISA, between naïve littermate control and Lyl1-deficient mice. Further evaluation of Lyl1 regulation revealed the influence of MAPk and NFκB signaling on Lyl1 expression upon LPS stimulation by significantly downregulating this transcription factor in immune stimulated macrophages. A role for Lyl1 during bacterial infections was observed in Lm-infected mice whereby Lyl1-/- mice succumbed earlier to listeriosis compared to the littermate controls. We further established a functional role for this transcription factor during Mtb infection in vitro and in vivo. The early surrender of Lyl1-deficient mice to Mtb HN878 infection, accompanied by increased bacterial burden during chronic Mtb infection, demonstrated enhanced susceptibility in the absence of Lyl1. We show that Lyl1-deficient host susceptibility is a consequence of enhanced inflammatory responses and increased bacterial growth. This is demonstrated by increased neutrophilic inflammation, pro-inflammatory cytokine and chemokine secretion along with a reduction in anti-inflammatory cytokine release during chronic Mtb infection. Here, we demonstrate the first non-leukemia role for Lyl1 by suggesting a role and requirement for this transcription factor during bacterial infections. Given the significant role during Mtb infection, our studies suggest the use of Lyl1 associated pathways as a potential HDT target for TB.

The aim of this thesis was to investigate the diagnostic accuracy of Xpert for musculoskeletal TB and for rifampicin resistance against a gold standard of culture or histology. Site of disease, HIV status, and age of patients, and accuracy in spinal compared to extraspinal TB were investigated as secondary objectives. The overarching hypothesis was that Xpert is more accurate and would provide results faster than the gold standard for musculoskeletal TB, and that it would have a higher yield in HIV infected patients, adult patients, and patients with spinal disease.

Abstract:
Background Xpert MTB/RIF (Xpert) is a rapid, automated, onsite nucleic acid amplification test for tuberculosis (TB). It is effective for the diagnosis of pulmonary TB but there is limited evidence for its usefulness in extrapulmonary TB, particularly musculoskeletal TB. Aims and hypothesis The aim of this thesis was to investigate the diagnostic accuracy of Xpert for musculoskeletal TB and for rifampicin resistance against a gold standard of culture or histology. Site of disease, HIV status, and age of patients, and accuracy in spinal compared to extraspinal TB were investigated as secondary objectives. The overarching hypothesis was that Xpert is more accurate and would provide results faster than the gold standard for musculoskeletal TB, and that it would have a higher yield in HIV infected patients, adult patients, and patients with spinal disease. Methods Prospective studies of patients with suspected musculoskeletal TB, at the tertiary care hospitals Groote Schuur and Red Cross Children’s Hospital in Cape Town, South Africa, were undertaken from June 2013 to March 2015. The diagnostic accuracy of Xpert was compared to culture or histopathology. Findings 206 biopsies of 201 patients older than 13 years of age (23% HIV positive) were analysed. The sensitivity and specificity of Xpert was 92.3% and 99.1% respectively. Xpert detected 8 cases more than culture (p = 0.069) and positive results were available 17 days earlier (<0.001). The sensitivity of Xpert in HIV positive patients was 96.9% (31/32) versus 89.6% (43/48) in HIV negative patients (p=0.225). The sensitivity of Xpert for spinal biopsies was 93.8% (95% CI 86.0-97.9) with specificity of 97.6% (95% CI 87.4 – 99.9), compared to extraspinal biopsies with a sensitivity of 81.8% (95% CI 48.2 – 99.7, p=0.164) and specificity of 100% (95% CI 95.1 – 100%, p=0.186). 109 osteoarticular samples of children 12 years of age or younger, with a median age of 5.6 years (IQR 2.2 – 8.7) were analysed. Xpert provided a sensitivity of 73.9% (95% CI 51.6-89.8) with a specificity of 100% (95% CI 95.7 - 100) and was available at a mean of 0.8 days (0.46- 1.4) compared to 21 days (19 – 30) for culture (p< 0.001). All rifampicin resistant cases were correctly diagnosed. A trend towards higher sensitivity in spinal tissue as well as HIV infected patients was observed. This study also provides evidence that Xpert has a lower sensitivity in children than in adults, yet, still detects more cases of paediatric musculoskeletal TB and is faster than culture. Histology was a useful test for the diagnosis of musculoskeletal TB, especially in children, and should be used alongside Xpert to provide the highest yield possible to detect TB. Conclusion: These first large studies on the accuracy of Xpert for musculoskeletal TB provide evidence for the usefulness of Xpert in the diagnosis of spinal TB, extraspinal TB, in HIV positive patients, and in childhood musculoskeletal TB. Based on these results, Xpert should be recommended as the initial test for diagnosis as it is more sensitive and faster than the gold standard of liquid culture.
