Abstract:
ABSTRACT An Epstein-Barr virus (EBV) recombinant (MS231) that expresses the first 231 amino acids (aa) of LMP1 and is truncated 155 aa before the carboxyl terminus transformed resting B lymphocytes into lymphoblastoid cell lines (LCLs) only when the infected cells were grown on fibroblast feeder cells (K. M. Kaye et al., J. Virol. 69:675–683, 1995). Higher-titer MS231 virus has now been compared to wild-type (WT) EBV recombinants for the ability to cause resting primary B-lymphocyte transformation. Unexpectedly, MS231 is as potent as WT EBV recombinants in causing infected B lymphocytes to proliferate in culture for up to 5 weeks. When more than one transforming event is initiated in a microwell, the MS231 recombinant supports efficient long-term LCL outgrowth and fibroblast feeder cells are not required. However, with limited virus input, MS231-infected cells differed in their growth from WT virus-infected cells as early as 6 weeks after infection. In contrast to WT virus-infected cells, most MS231-infected cells could not be grown into long-term LCLs. Thus, the LMP1 amino-terminal 231 aa are sufficient for initial growth transformation but the carboxyl-terminal 155 aa are necessary for efficient long-term outgrowth. Despite the absence of the carboxyl-terminal 155 aa, MS231- and WT-transformed LCLs are similar in latent EBV gene expression, in ICAM-1 and CD23 expression, and in NF-κB and c-jun N-terminal kinase activation. MS231 recombinant-infected LCLs, however, require 16- to 64-fold higher cell density than WT-infected LCLs for regrowth after limiting dilution. These data indicate that the LMP1 carboxyl-terminal 155 aa are important for growth at lower cell density and appear to reduce dependence on paracrine growth factors.

Abstract:
Diarylpentanoid (DAP), an analog that was structurally modified from a naturally occurring curcumin, has shown to enhance anticancer efficacy compared to its parent compound in various cancers. This study aims to determine the cytotoxicity, antiproliferative, and apoptotic activity of diarylpentanoid MS13 on two subtypes of non-small cell lung cancer (NSCLC) cells: squamous cell carcinoma (NCI-H520) and adenocarcinoma (NCI-H23). Gene expression analysis was performed using Nanostring PanCancer Pathways Panel to determine significant signaling pathways and targeted genes in these treated cells. Cytotoxicity screening revealed that MS13 exhibited greater inhibitory effect in NCI-H520 and NCI-H23 cells compared to curcumin. MS13 induced anti-proliferative activity in both cells in a dose- and time-dependent manner. Morphological analysis revealed that a significant number of MS13-treated cells exhibited apoptosis. A significant increase in caspase-3 activity and decrease in Bcl-2 protein concentration was noted in both MS13-treated cells in a time- and dose-dependent manner. A total of 77 and 47 differential expressed genes (DEGs) were regulated in MS13 treated-NCI-H520 and NCI-H23 cells, respectively. Among the DEGs, 22 were mutually expressed in both NCI-H520 and NCI-H23 cells in response to MS13 treatment. The top DEGs modulated by MS13 in NCI-H520—DUSP4, CDKN1A, GADD45G, NGFR, and EPHA2—and NCI-H23 cells—HGF, MET, COL5A2, MCM7, and GNG4—were highly associated with PI3K, cell cycle-apoptosis, and MAPK signaling pathways. In conclusion, MS13 may induce antiproliferation and apoptosis activity in squamous cell carcinoma and adenocarcinoma of NSCLC cells by modulating DEGs associated with PI3K-AKT, cell cycle-apoptosis, and MAPK pathways. Therefore, our present findings could provide an insight into the anticancer activity of MS13 and merits further investigation as a potential anticancer agent for NSCLC cancer therapy.

Abstract:
Цель исследования – оценка комбинационной способности линий редьки черной по основным хозяйственно ценным признакам. Исследования проводили в 2019–2020 годах на базе ООО «Селекционная станция имени Н.Н. Тимофеева», подразделении ФГБОУ ВО РГАУ – МСХА имени К.А. Тимирязева, которая располагается на территории Москвы. Почвы опытного поля – среднесуглинистые дерново-подзолистые на покровном суглинке. Материалом служили линии редьки черной из коллекции ООО «Селекционная станция имени Н.Н. Тимофеева»: стерильные линии: МS 26, MS151 и фертильные линии: Рч65, Рч62, Рч64, Рч61, РчРын. Стандартом выбран популярный сорт редьки Зимняя круглая черная. Оценку комбинационной способности проводили методом скрещивания двух групп генотипов. Расчет комбинационной способности производили по первой математической модели, предложенной В.К. Савченко. Агроклиматические показатели 2019 и 2020 годов в целом несущественно отличались от среднемноголетних значений и были оптимальны для выращивания редьки. Опыты закладывали рендомизированным методом. Схема посева 20×50, однострочная, по 30 растений в варианте, в двукратной повторности. Уход за растениями – общепринятый. Оценку гибридных комбинаций проводили по следующим параметрам: масса корнеплода, масса листьев, диаметр и длина корнеплода, урожайность, индекс формы корнеплода. 5 октября 2020 года шесть гибридных комбинаций были заложены на хранение до 10 апреля 2021 года. Гибридную комбинацию MS26 × Рч65, обладающую комплексом ценных хозяйственных признаков (высокая урожайность, средняя масса листьев, округлая форма корнеплода), можно рекомендовать для станционного сортоиспытания. Наилучшая сохранность отмечена у гибридных комбинаций MS151 × Рч65 и MS151 × Рын (87,38 и 85,80% соответственно). Анализ эффектов ОКС выявил селекционную ценность линий MS151 и Рын (большая масса корнеплода), Рч61 (большая масса листьев), MS26, MS151 и Рч62 (округлая форма корнеплода). Гибридные комбинации MS26 × Рч65, MS26 × Рч62, MS151 × Рч62 обладают стабильностью в проявлении хозяйственных признаков при испытании в разные годы. The purpose of the study is to evaluate the combinational ability of black radish lines according to the main economically valuable characteristics. The research was carried out in 2019–2020 on the basis of the N.N. Timofeev Breeding Station LLC, a subdivision of the Russian State Agrarian University – MAA named after K.A. Timiryazev, which is located on the territory of Moscow. The soils of the experimental field are medium loamy, sod-podzolic on the cover loam. The material was the black radish lines from the collection of the N.N. Timofeev Breeding Station LLC: sterile lines: MS 26, MS151 and fertile lines: Rch65, Rch62, Rch64, Rch61, RchRyn. The standard is a popular variety of radish Zimnyaya kruglaya chernaya. The evaluation of the combinational ability was carried out by crossing two groups of genotypes. The calculation of the combinational ability was carried out according to the first mathematical model proposed by V.K. Savchenko. The agro-climatic indicators of 2019 and 2020 as a whole did not differ significantly from the long-term average values and are optimal for growing radishes. Bookmark the experience by a randomized method. The sowing scheme is 20 × 50, single-line, 30 plants in a variant, in double repetition. Plant care is generally accepted. Hybrid combinations were evaluated according to the following parameters: root crop mass, leaf mass, root crop diameter and length, yield, root crop shape index. On October 5, 2020, six hybrid combinations were put into storage until April 10, 2021. The hybrid combination MS26 × Rch65, which has a complex of valuable economic characteristics (high yield, average leaf weight, rounded shape of the root crop) can be recommended for station variety testing. The best preservation was noted in hybrid combinations MS151 × Rch65 and MS151 × Ryn (87.38 and 85.80%, respectively). The analysis of the effects of ACS revealed the breeding value of the lines MS151 and Ryn (large mass of root crop), Rch61 (large mass of leaves), MS26, MS151 and Rch62 (rounded shape of the root crop). Hybrid combinations MS26 × Rch65, MS26 × Rch62, MS151 × Rch62 have stability in the manifestation of economic signs when tested in different years.

ABSTRACT The role of the RecBCD recombination pathway in PilE antigenic variation in Neisseria gonorrhoeae is contentious and appears to be strain dependent. In this study, N. gonorrhoeae strain MS11 recB mutants were assessed for recombination/repair. MS11 recB mutants were found to be highly susceptible to DNA treatments that caused double-chain breaks and were severely impaired for growth; recB growth suppressor mutants arose at high frequencies. When the recombination/repair capacity of strain MS11 was compared to that of strains FA1090 and P9, innate differences were observed between the strains, with FA1090 and P9 rec + bacteria presenting pronounced recombination/repair defects. Consequently, MS11 recB mutants present a more robust phenotype than the other strains that were tested. In addition, MS11 recB mutants are also shown to be defective for pilE/pilS recombination. Moreover, pilE/pilS recombination is shown to proceed with gonococci that carry inverted pilE loci. Consequently, a novel RecBCD-mediated double-chain-break repair model for PilE antigenic variation is proposed.

Abstract:
The cytotoxic and apoptotic effects of turmeric (Curcuma longa) on colon cancer have been well documented but specific structural modifications of curcumin have been shown to possess greater growth-suppressive potential on colon cancer than curcumin. Therefore, the aim of this study is to identify the anti-cancer properties of curcumin analogue-MS13, a diarylpentanoid on the cytotoxicity, anti-proliferative and apoptotic activity of primary (SW480) and metastatic (SW620) human colon cancer cells. A cell viability assay showed that MS13 has greater cytotoxicity effect on SW480 (EC50: 7.5 ± 2.8 µM) and SW620 (EC50: 5.7 ± 2.4 µM) compared to curcumin (SW480, EC50: 30.6 ± 1.4 µM) and SW620, EC50: 26.8 ± 2.1 µM). Treatment with MS13 at two different doses 1X EC50 and 2X EC50 suppressed the colon cancer cells growth with lower cytotoxicity against normal cells. A greater anti-proliferative effect was also observed in MS13 treated colon cancer cells compared to curcumin at 48 and 72 h. Subsequent analysis on the induction of apoptosis showed that MS13 treated cells exhibited morphological features associated with apoptosis. The findings are also consistent with cellular apoptotic activities shown by increased caspase-3 activity and decreased Bcl-2 protein level in both colon cancer cell lines. In conclusion, MS13 able to suppress colon cancer cell growth by inhibiting cell proliferation and induce apoptosis in primary and metastatic human colon cancer cells.

ABSTRACT Lipooligosaccharide (LOS) heptose (Hep) glycan substitutions influence gonococcal serum resistance. Several gonococcal strains bind the classical complement pathway inhibitor, C4b-binding protein (C4BP), via their porin (Por) molecule to escape complement-dependent killing by normal human serum (NHS). We show that the proximal glucose (Glc) on HepI is required for C4BP binding to Por1B-bearing gonococcal strains MS11 and 1291 but not to FA19 (Por1A). The presence of only the proximal Glc on HepI (lgtE mutant) permitted maximal C4BP binding to MS11 but not to 1291. Replacing 1291 lgtE Por with MS11 Por increased C4BP binding to levels that paralleled MS11 lgtE, suggesting that replacement of the Por1B molecule dictated the effects of HepI glycans on C4BP binding. The remainder of the strain background did not affect C4BP binding; replacing the Por of strain F62 with MS11 Por (F62 PorMS11) and truncating HepI mirrored the findings in the MS11 background. C4BP binding correlated with resistance to killing by NHS in most instances. F62 PorMS11 and its lgtE mutant were sensitive to NHS despite binding C4BP, secondary to kinetically overwhelming classical pathway activation and possibly increased alternative pathway activation (measured by factor Bb binding) by the F62 background. FA19 lgtF (HepI unsubstituted) resisted killing by only 10% NHS, not 50% NHS, despite binding levels of C4BP similar to those of FA19 and FA19 lgtE (both resistant to 50% serum), suggesting a role for the proximal Glc in serum resistance independently of C4BP binding. This study provides mechanistic insights into how HepI LOS substitutions affect the serum resistance of N. gonorrhoeae.
