Full textQuantification of specific biomarkers is an important diagnostic tool. Standard immunoassays such as ELISA require extensive washing steps and signal amplification, in particular when the biomarker of interest is only present at very low concentrations. On the other hand, non-radiative Förster resonance energy transfer (FRET) has been used to design one-step homogenous bioassays which do not require any washing steps, where the biomarker enables the formation of a sandwich complex involving donor-labeled and acceptor-labeled antibodies. FRET from the donor to the acceptor then provides an optical signature of the complex formation, hence of the biomarker of interest. However, FRET which is highly sensitive to the donor-acceptor distance, only occurs in a significant rate when the distance between the donor and acceptor is less than 10 nanometers; thus the large size of many biological complexes limits the efficiency of energy transfer, preventing sensitive detection. Here I propose a novel energy transfer modality that uses solution-phase optical microcavities to enhance energy transfer. Following that, I describe a bio-sensing scheme designed to detect a cancer biomarker DNA in solution.To this aim, I have designed microcavity structures in which fluorescent colloidal quantum dots are located inside dielectric polymer microspheres to enable strong coupling of their fluorescence emission with the cavity resonance modes or whispering gallery modes (WGMs) of the microspheres. A detailed study was carried out to comprehend the structural and optical properties of these optical microcavities. I also characterized the energy transfer between these modes and acceptor dye-loaded nanoparticles present in the evanescent field, within a few tens of nanometers above the microsphere surface. An analytical model was constructed to provide insights into the WGM mediated energy transfer (WGET) mechanisms. Moreover, a comparison between WGET and FRET revealed the superiority of WGET in the context of building sensors with improved sensitivity and longer range of detection. In the last part of the thesis, a strategy is discussed in detail to provide biological functionalities to these optical microcavities which would enable them to interact with target analytes such as DNA, RNA, and proteins with high specificity, and moreover to reduce non-specific interactions. This strategy then was adapted to attach DNA capture probes onto the WGM enabled microcavities. Using the DNA attached microspheres as optical donor in combination with probe-DNA functionalized dye nanoparticles as optical acceptors, a biosensing assay has been successfully demonstrated to detect a cancer biomarker DNA called survivin in the solution phase. This assay did not only show good sensitivity towards the target, but also it has proven to be highly specific. The detection scheme has been demonstrated in a sophisticated confocal microscope at the single microsphere level, then successfully translated to a much simpler spectrofluorometer that measures fluorescence from the whole sample solution; the signature of the sandwich complex formation was also effectively detected.In conclusion, I demonstrated that microcavity-assisted energy transfer has several advantages over regular FRET assays. A real bio-sensing assay based on the WGET principle has also been successfully designed to detect cancer biomarkers with high sensitivity and specificity. This study thus opens up many possibilities to design high-performing and more accurate assays to detect varieties of biological entities

Full textThe most stable oscillators use a sapphire resonator placed at low temperature. This resonator is used in a passive configuration and the oscillation is maintained by an amplifier at room temperature. To reach ultimate stabilities, several controls must be implemented in order to mitigate the phase and amplitude drifts of the signal in the oscillation loop. Such an oscillator is thus cumbersome and difficult to implement in ground applications. The main objective of this thesis is to validate another principle of the use of sapphire resonator: the Whispering Gallery modes MASER Oscillator (Whigmo). The idea is to use paramagnetic impurities that are present within the sapphire crystal to obtain a self-sustained MASER effect. The Whigmo principle has been demonstrated by using HEMEX sapphire resonators, containing paramagnetic ions (Fe3+ ions). Its principle is based on the three energy levels MASER at zero dc magnetic field. By pumping the ions thanks to a whispering gallery (WG) mode at 31 GHz, an oscillation is obtained via another WG mode at 12. 04 GHz. The maximum power delivred is of about 6. 10. 9 W (ten thousand times higher than an H-Maser) for ionic concentration as Iow as 10 ppb. A model combining the WG modes and the behaviour of the ions within the sapphire matrix, allows to describe the Maser power evolution according to various parameters, such as temperature. . Couplings of the WG modes (at 12 GHz and 31 GHz) and the ionic concentration. Measurements on the MASER instability for a power of about 10-12 W, showed that this oscillator is limited only at short term the thermal noise. An instability lower than 10. 14 at short term has been shown through another experiment, by comparing the Whigmo to the state of the art Cryogenic Sapphire Oscillator (CSO). This work suggests that the MASER working at power of 6. 10. 9 W could deliver short-term instability of about 1. 10. 1611/2. The Whigmo is then an oscillator that combine the performances of the traditional CSO and an extreme simplicity. Ln the near future it will elsewhere become the successor of the traditional CSO

Full textSemiconductor whispering-gallery-mode (WGM) microresonators have already gained a prominent place in today's photonics, as an ideal environment for highly enhanced light-matter interactions like nonlinear wave mixing and optomechanics. These optical cavities can store optical energy on a sub-pm3 volume, by trapping light via total internai reflection. The interest drawn by WGM microcavities originates from their high quality factors and promise of applications for fundamental science and engineering purposes. Thanks to their versatility and high performances, they can be integrated on a photonic chip and involved in a wide set of applications, such as quantum-dot lasers, optomechanics, nanosensing, or quantum optics. This doctoral work deals with the study of three wave mixing processes in two different frameworks. First, I show the development made to fabricate some original micro-resonators that have enabled us to observe second harmonic generation around 775 nm, with a pump wavelength around 1550 nm. The obtained nonlinear efficiency is comparable to the state-of-the-art devices in the III-V platform. The second part of the experiments finds its place within the European project TREASURE. The main goal is the demonstration of an electrically pumped, CW THz emitter, based on difference frequency generation at room temperature. I present the measurements in the near-IR and THz regions performed to compare our results and simulations

Full textThis work consisted in developing a fabrication process of silica microtoroids on a silicon chip (steps of lithography and etching in clean room for the realization of microdisks, set up of an optical bench to form a microtoroid with a reflow treatment of a silica microdisk by a CO2-laser), setting up an optical bench to measure the linewidth of their optical resonances at 1.55 µm and finally, exploring light emitters integration in these cavities such as silicon and germanium. Very high quality-factors (Q) close to 10^8 at 1.55µm have been measured on microtoroids. These realizations are very close to the State of the art and validate both the fabrication of these cavities and the optical bench to measure the linewidth of their Whispering Gallery Modes (WGM). With a precise control of the fabrication steps, new resonators have also been fabricated, silica microspheres on a chip with small radii (5 < r < 14µm). An in-depth study of these last ones is presented. Q-factors close to 10^8 have also been measured on microspheres. WGM cavities with a SiOx: Er layer (silicon nanoclusters in silica with erbium ions) are studied by photoluminescence. Coupling of these light emitters to WGM is observed in visible and near infrared at room temperature. A work of coupling of germanium to WGM began and seems promising

Full textLa première partie de l'étude concerne le développement de matériau émetteur utilisant l'erbium. Différentes matrices sont utilisées afin d'aboutir à une émission efficace à 1,54 µm à température ambiante : silice, silicium sur isolant (SOI) et silice non-stoechiométrique (SRO : nanoclusters de silicium en matrice de silice). Les émetteurs retenus pour l'étude, le silicium sur isolant et l'erbium associé au SRO, sont ensuite intégrés à des cavités de type microdisque par des procédés de lithographie et gravure dérivés de ceux utilisés en microélectronique. Dans le cas des disques à base de silice, un recuit au laser CO2 permet de s'affranchir des rugosités résiduelles par fusion des bords du disque conduisant à la formation de tores.
Les structures obtenues sont ensuite étudiées au moyen d'un banc de photoluminescence spécifique développé au laboratoire tenant compte des caractéristiques de l'émission dans les modes de galerie et permettant leur évaluation fine. Le couplage de l'émission du silicium et du SRO dopé erbium à des modes de galerie possédant des facteurs de qualité de plusieurs milliers (jusqu'à Q ≈ 90000) est ainsi observé à température ambiante. Ces résultats, associés à la possibilité de coupler les cavités à des guides démontrée pour les disques SOI, ouvrent des perspectives particulièrement intéressantes du point de vue de la recherche appliquée (sources intégrées) comme de la recherche fondamentale (effets CQED sur les terres rares).

Full textThis research work was done in order to use high-Q dielectric resonators for the characterization of semi-insulating materials and to build an ultra-low phase noise oscillator cooled at 77 Kelvin. The first chapter introduces the whispering gallery modes which are the heart of the work presented in this thesis. The second chapter presents a new method for the characterization of semi-insulating materials by using the whispering gallery modes. Measurements were made on gallium phosphide, gallium arsenide and silicon carbide 4H. Under light and at cryogenic temperatures, microwave properties of the samples change because of photoelectric effect. In the last and third chapter, we describe an ultra-low phase noise oscillator at 11GHz and cooled at 77 Kelvin. The ultralow phase noise oscillator is based on a double sapphire resonator. The whispering gallery modes are in the key element of this project. Phase noise measurement gives - 115dBc/Hz at 1 kHz of the carrier

Full textGlass-based whispering gallery mode (WGM) microresonators are easy to produce by melting techniques. However, they suffer from surface contamination or residual absorption which limits their long term quality factor to only about 108. We show that an optical gain provided by erbium ions can compensate for residual losses. Moreover it is possible to control the coupling regime of an ultrahigh Q-factor microresonator from undercoupling to spectral selective amplification by changing the pumping rate. We demonstrate a critically coupled fluoride glass WGM microresonator with a diameter of 220 μm and a loaded Q-factor of 5,3×109. We also show that by introducing slow-light effects in a monolithic WGM microresonator it is possible to enhance the photon lifetime by several orders of magnitude and circumvent fabrication limitations. We experimentally demonstrate Erbium-doped fluoride glass microresonators with a photon lifetime up to 2,5 ms at room temperature, corresponding to a Q-factor of 3×1012 at 1530 nm, by combining WGM resonance effect and population oscillations

Full textPour réaliser une interaction atome-champ contrôlée, il est nécessaire de rendre la microsphère accordable. Par l'application d'une contrainte mécanique sur la sphère, nous avons pu accorder un mode de galerie sur 400 GHz. De plus, en miniaturisant le
dispositif de couplage, nous pouvons exciter un mode de la sphère dans une chambre ultra-vide, où les atomes froids sont manipulés.
D'autre part, une puce multi-couches nous permet de réaliser un convoyeur magnétique, transportant les atomes entre une zone de chargement du piège et la cavité, soit sur une distance de 6 cm à une vitesse de 10 cm/s. En effectuant 2 allers-retours,
les atomes ont parcouru une distance totale de 24 cm. Ce transport d'atomes piégés nous permet de résoudre l'un des principaux problèmes du couplage d'atomes froids
avec les modes d'une microsphère.

Full textWhispering galery mode resonators are dielectric cavities that support modes with ultra-high quality factor and small volume that remain confined in their inner periphery for time duratioons that can be as long as few microseconds. The strong confinement of these modes for such long durations strongly enhances nonlinear effect suchs as Kerr effect or Brillouin effect. These resonators can therefore be used for several applications such as spectroscopy, telecommunications or microwave generation. The objective of this thesis is to use Kerr and Brillouin nonlinearities in these resonators at the laser wavelength of 1550 nm, in order to generate high spectral purity microwave signals with frequencies rangong fros 5 to 30 GHz. The first chapter oh the thesis intriduces the theory, fabrication, coupling and characterisation of whispering gallery mode resonators. The second chapter is about the generation of Kerr optical frequency combs in these resonators and their application to the generation of microwave signals. We present our experimental resuktsdn the numerical modelling of Kerr combs, the analysis of oscillatory instabilities (due to thermal effect) observed during our experiments, and conclue. The third chapter concerns photon-phonon interactions via stimulated Brillouin scattering in these resonators and their application to the generation of microwave signals. We present our experimental results and the temporal model that we developed to track the dynamics of the forward and backscattered fields. The last chapter conclude the thesis. Full textThis thesis presents the studies of whispering gallery mode laser dynamics in order to realize a micro-wave source using simultaneous oscillations in a unique whispering gallery mode micro-laser. We show experimental results on the relative intensity noise (RIN) of a Whispering Gallery Mode Laser in ZBLALiP glass doped with Er3+ ions. Besides the pure laser specifications, the RIN spectrum gives informations about the dynamics inside the cavity, such as photon lifetime, effective pumping rate and noise sources. Moreover, we have shown that a single-mode emission comes with the presence of multiple harmonics of the relaxation frequency. The theoretical model taking account the non-linear coupling between photons and atoms allows us to determine the mode volume of the whispering gallery mode in laser regime, which is quite difficult to evaluate in this regime. On the other hand, we have studied the laser dynamics in an industrial IOG-1 glass codoped with Yb3+/Er3+ ions where the signature of a modal coupling, induced by Rayleigh scattering, lies in the RIN spectrum. In this particular case, the two coupled modes operate above threshold. The observed behavior is compared with thoses of a solid state gyro-laser

Full textThis study deals with the control of spontaneous emission, InAs quantum dot emitters, confined via original microstructures: GaAs photonic wires and micropillars with GaAs/AlAs Bragg mirrors. We present a higly efficient single-photon source based on a photonic wire. Correlation mesurements performed on a single quantum dot inserted in a photonic wire led to a pure and high broadband single-photon generation. The optimization of this geometry including an original mirror at the wire bottom and a taper at its top demonstrated a record efficiency of 70%. In addition, the systematic lifetime study of single quantum dots underlined the ability to observe a high inhibition of spontaneous emission in the leaky modes with this geometry. In the last part of this PhD Thesis, we show the laser effect in whispering gallery mode micropillars. Those modes have an almost stable behaviour in terms of wavelengh and a spectral narrowing maintained up to power corresponding to 7 times the laser threshold. This result confirms a better thermal stability of micropillars compared to microdisks, a geometry usually used to observe the whispering gallery modes

Nous avons réalisé un dispositif d'imagerie de corrélation afin de suivre, avec une résolution temporelle, la propagation d'un paquet d'onde laser. Des simulations FDTD ont permis d'interpréter les observations et de mettre en évidence un processus original de couplage de sortie, impliquant la réflexion de l'onde évanescente sur la discontinuité de la spirale.

Full textIn this work, feasibility of gas sensors based on Titanium Dioxide (TiO2) dielectric resonator, operating with whispering-gallery modes, is presented. The gas or humidity adsorption modifies the TiO2 dielectric permittivity and such modification induces variation in the resonant frequencies of high-Q whispering-gallery modes in the millimeter-wave frequency range. Full-wave electromagnetic simulation demonstrates that the measurement of this variation allows the derivation of few parts-per-million (ppm) fluctuations in gas or humidity concentration. For validation purposes very first prototypes of resonator operating with whispering-gallery modes in the millimeter-wave frequency range are micro-machined. The measured performances confirm that such high-Q resonant modes are very sensitive to small variations in dielectric resonator permittivity. Moreover we validate experimentally that these small variations can be remotely detected from the RADAR interrogation of an antenna loaded by the whispering-gallery modes resonator

Full textInvestigations towards a resonator featuring a high quality factor in a reduced volume are a very important and interesting problem in the microwave field. The microwave resonators used for high spectral purity microwave sources are reaching their limits when the goal is to get simultaneously high performance in terms of phase noise, small size and high frequency operation. Therefore, an original approach is to transpose the microwave wavelength in the optical field. This is the case, as an example, of microwave oscillators using as a frequency reference element an optical delay line in place of the conventional microwave resonator. However, these oscillators require a large volume, and are difficult to stabilize versus the temperature fluctuations. The aim of this study is thus to replace the optical fiber by an optical resonator, and to realize an high spectral purity microwave source referenced on this resonator. A characterization bench has been set up with nanometre scale precision 3D displacement stages to hold the coupling of the resonator (SiO2 mini-sphere, or monocrystalline disk). An optical Q factor of 4. 109 has been demonstrated on a quartz minidisk. However, it is difficult to get an efficient and stable resonator coupling, because of the mode shift with temperature when the light is induced in the resonator. Two techniques have been studied to solve these problems and are presented in this manuscript. .

Full textDans une première partie nous présentons un modèle matriciel pour décrire la propagation de la lumière dans des structures à anneaux couplés. L'utilisation de ce modèle se résume pour des structures dont on connaît les coefficients de couplage entre les guides.
Dans le cas de résonateurs constitués d'un milieu qui ne présente pas d'effets non-linéaires nous avons montré que les structures à anneaux couples permettent de réaliser la synthèse de filtres optiques.
Si les résonateurs sont le siège d'une non linéarité dispersive d'ordre 3 nous avons deux cas possibles en fonction du type d'excitation. Hors résonance nous avons montré que nous pouvons obtenir potentiellement des dispositifs /N+1/ stables en utilisant un système formé de /N/ anneaux couplés, ainsi que la tristabilité dans un seul anneau en utilisant un signal de commande. A résonance nous avons montré que l'on peut obtenir la fonction de remise en forme du signal pour des débits de 40 Gb/s.
La deuxième partie porte sur les travaux réalisés sur l'oscillateur optoélectronique. Dans le schéma classique d'un tel oscillateur la fonction de retard optique est obtenue en utilisant une ligne à fibre optique de longueur proportionnelle au retard nécessaire. Nous voulons remplacer cette ligne avec une ligne à retard à base de microrésonateurs. Nous présentons des procédés de fabrication de résonateurs sphériques et diverses méthodes de mesure de leurs facteurs de qualité, proportionnel au retard induit.

Full textThis thesis is dedicated to the study of the Kerr optical frequency combs in whispering gallery moderesonators, where the light can be excited by the extern pump. Due to the Kerr effect existing in theseresonators, the quasi-equidistant lines in the spectral domain are generated around the excited mode,that is the frequency comb. This thesis is devided in three chapters. The first one is dedicated to theintroduction of the Kerr comb generation and their applications.The second one presents the analysisof the Lugiato-Lefever equation used for the analytical study of the system, leading to the constructionof two bifurcation diagrams for the normal and anomalous dispersions. They are plotted for twoparameters, which can be controlled during experiments once the resonator has been fabricated,which are the pump power of the laser and its frequency detuning. These diagrams show the areas ofthe parameters for which one, two, or three solutions exist and their stability. The additional numericalsimulations show the exact type of the solution in each area (such as the bright and dark solitons,the breathers, the primary and secondary Kerr combs and chaotical regimes), finally these diagramsshow the parameters of the laser needed to be choosen for the generation of the desired solution.The third chapter is dedicated to the secondary Kerr combs, which are the additional lines generatedbetween the lines of the primary comb. They appear in the anomalous dispersion regime, when thequantity of the pump photons crosses the second-order threshold, which has been found numerically
