Abstract:
Laterální nehomogenity v zemském nitru významně ovlivňují časy příchodů seismických vln odečtené na stanicích při seismickém monitorování. Pozorované časy odečtů se liší od předpokládaných časů odvozených na základě globálních rychlostních modelů, přičemž zjištěné rozdíly jsou nazývány časovými rezidui (Tr). V článku je demonstrována problematika vztahu nehomogenit a časů detekce seismických vln na příkladu signálu severokorejské jaderné exploze ze září 2017. Šlo o silnou explozi (magnitudo mb počítané z objemových vln přesáhlo hodnotu 6), jejíž signál byl vzhledem k ostrému začátku a výrazné amplitudě první výchylky podélných vln výjimečně vhodný pro studium časových reziduí. Spolu s dobrou znalostí polohy hypocentra exploze dává zmíněný seismický jev mimořádnou příležitost k pozorování časových reziduí, které jsou zatíženy jen malou chybou a které tak jsou spolehlivějším odrazem vlivu litosférických nehomogenit, než jaký poskytují data jiných seismických jevů. Pro studium časových reziduí v prostoru střední Evropy byla použita data několika stovek seismických stanic. Vyšší kladné hodnoty časových reziduí Tr (tedy zpoždění času detekce oproti modelovému předpokladu), jejichž příčiny jsou stručně diskutovány v tomto článku, byly zjištěny v z. a s. části Západních Karpat a ve v. části Panonské pánve. Naopak výrazně záporné hodnoty časových reziduí byly pozorovány v širokém prostoru Jižních vápencových Alp a v. části Pádské nížiny. Pro regionální a globální seismické monitorování stanicemi situovanými ve střední Evropě (a především na území České republiky) je významným fenoménem především významné (až více než sekundové) zpoždění odečtů, oproti teoretickým odečtům plynoucím z modelu IASPEI, v západní části Západních Karpat. Toto zpoždění je způsobeno souběžně nejen mocnými polohami flyšových sedimentů, ale další příčiny lze hledat ve spodní kůře a nejsvrchnějších partiích pláště.

Abstract:
The project is focused on face detection in video. Firstly, it contains a summary of basic color models. Secondly, you can find the description and comparison of the basic methods for detection of human skin with a practical example of implementation of parametric detector. Thirdly, a theoretical basis for face detection and face tracking in a video containing a list of basic concepts and methods of this issue follows. Greater emphasis is placed on the description of machine learning algorithm AdaBoost and description of the possible application of the Kalman filter for the purpose of face tracking. Design, implementation and testing of library accomplished within the master thesis are listed in the final part of this thesis.

The main goal of this project was to familiarize readers with the techniques used in real-time animation of 3D characters. This work is focused on two types of animation: keyframe animation and skeletal animation. There are described algorithms for software and hardware accelerated model deformations, keyframe interpolations, animation blending, inverse kinematics and ragdoll. The result of this project is a framework, which consists of an animation library, examples demonstrating library functions and tools for export animations from 3D Studio Max and MilkShape 3D.

Jarmila Zbytovská Title of thesis: In vitro models of skin barrier The aim of this work is to summarize information about various types of skin models which are used for testing of permeability, toxicity, irritability and other aspects of drugs, through professional, verified and reviewed literature. These characteristics are necessary to know in order to grant optimal safety, effectiveness nad quality of transdermally administered drugs. Transdermal administration of drugs has got lots of benefits in contrast with classic peroral administration. An administration of drugs through this way is quite simple and it can be interrupted quite easily. We can also easily change a place of administration in which a drug is released and the risk of overdosing is very low. Furthermore, transdermal administration makes possible to maintain constant plasmatic concentration of drug in a blood stream and also to prolong the duration of effect of drugs with small halftime thanks to constant releasing of drug. Experimental models of a transfer of stem cells for therapeutic purposes Abstract Stem cell therapy currently represents a standard procedure of treating a wide variety of hereditary diseases and serious injuries. Development of the most suitable way of transfer of stem cells into the patient body remains very important question concerning this type of therapy. In our experiments we used nanofiber scaffolds for stem cell cultivation and their subsequent transfer. These nanofibers were prepared by the original needleless electrospun NanospiderTM technology. Allogeneic cornea or skin graft were transplanted from B6 mice to BALB/c mice. The grafts were covered by a nanofibrous scaffold with cultivated stem cells. Stem cells were stained by an imunofluorescent dye to enable us to monitore their migration from nanofibers into tissues and consequent distribution in the body and characterize changes of this distribution in the time. The methods of ELISA and PCR were used to confirm that mesenchymal stem cells support the production of antiinflammatory cytokines IL-4 and IL-10 and contribute to inhibition of production of proinflammatory cytokines IL-1, IFNγ and inducible nitric oxide synthase. The skin barrier, which provides protection from water loss and harmful environmental influences is located in the stratum corneum. The dominant group of lipids within the stratum corneum are ceramides (Cer), which also have the most important role in ensuring the barrier properties of the skin. The enzymes sphingomyelin deacylase and glucosylceramide deacylase hydrolyze the amide bond of Cer precursors, which leads to highly polar metabolites, called lysolipids. The increased activity of these enzymes is considered to be one of the major factors leading to the development of a number of skin diseases characterized by a skin barrier disorder (for example atopic dermatitis). In this study we prepared model membranes mimicking a healthy skin barrier as an equimolar mixtures of human Cer, cholesterol, free fatty acids with the addition of 5 % cholesterol sulfate. Kallikrein-related peptidases (KLKs) constitute a highly conserved serine protease family. Based on in vitro experiments, KLKs are predicted to play an important role in a number of physiolog- ical and pathophysiological processes. However, their role in vivo remains not fully understood, partially due to a lack of suitable animal models. In this work, we aim to prepare a KLK5 and KLK14 double-deficient mouse model. Both KLK5 and KLK14 were proposed to be involved in epidermal proteolytic networks critical for maintaining skin homeostasis. However, both KLK5 and KLK14 single-deficient mouse models show minimal or no phenotype, likely due to similar substrate specificity resulting in functional compensation. Double-deficient mice cannot be easily obtained by crossing due to localization of the Klk5 and Klk14 genes within the same locus on chromosome 7. We report that KLK5 and KLK14 double-deficient mice were success- fully generated, mediated by transcription activator-like effector nucleases (TALENs) targeting Klk14 by microinjection of TALEN mRNA into KLK5-deficient zygotes. Furthermore, we show that KLK5 and KLK14 double-deficient mice are viable and fertile. We believe that these novel mouse models may serve as a useful experimental tool to study KLK5 and KLK14 in vivo.

Abstract:
The master's thesis consists of two parts. The first part contains the azimuth -- slowness analysis for period 3s<=T<=6s, which was conducted by the FK analysis for 10 stations of the WEBNET network. In the second part there is the surface wave group velocity measurement for every station pair of 10 WEBNET stations. The interstation group velocity was obtained by the cross--correlation of microseismic noise records. Local group velocities are connected with the instantaneous period data. The Love wave group velocities are visualised by the 2D tomography calculation. The thesis also contains the additional Love waves dispersion analysis for just one station pair. Epidermal homeostasis, including proper turnover of keratinocytes, plays important role in the barrier function and serine proteases and their inhibitors are the key players. Activated proteases cleave desmosomes in uppermost layer and thus shed the cells from the epidermal surface. Therefore the serine protease inhibitors are secreted in lower epidermal layers to prevent premature activation of proteases and consequent disruption of epidermal barrier. The most studied inhibitors in epidermis belong to Serine proteases inhibitors Kazal-type family (SPINK). This diploma thesis is aimed to investigate function of murine SPINK6 in epidermal compartment in vivo. To achieve this, the transgenic mice overexpressing mSPINK6 under modified human involucrin promoter was generated. Two of five transgenic lines exhibited higher expression of mSPINK6 at mRNA and protein levels. The mSPINK6 transgenic mice are viable with no apparent phenotype. The small but in most cases not significant differences were observed on microscopic level among mSPINK6 transgenic and wild type animals In conclusion, this work showed that mSPINK6 does not play major role in skin homeostasis but gains significant importance under specific challenges of epidermal barrier.
