Background Patients with segmental vitiligo (SV), unlike those with nonsegmental vitiligo (NSV), have a more predictable course and are more responsive to surgery. Objective To report 10 patients with SV treated with the melanocyte-keratinocyte transplantation procedure (MKTP), who responded with unusual responses not previously reported in the literature. Methods This is a retrospective, observational study that reports 10 patients with SV who underwent the MKTP between May 2003 and May 2012. Results Two patients had successful repigmentation after split-thickness skin grafting after failure of the MKTP. Two patients developed a hypopigmented ring at a margin of the MKTP-treated area. One patient had complete repigmentation after a second MKTP. Two patients developed koebnerization of the recipient site. Three patients developed new vitiligo patches in previously unaffected areas after the MKTP. Conclusions Uncommon and even suboptimal responses can occur following the MKTP in SV patients. There is a need for studies to provide better understanding and outcomes for SV patients undergoing the MKTP.

<p class="MDPI17abstract"><strong><span lang="EN-US">Tujuan:</span></strong><span lang="EN-US"> Penelitian ini bertujuan untuk menguji pakan penggemukan Domba Ekor Gemuk (DEG) ditinjau dari aspek konsumsi, kecernaan dan efisiensi penggunaan nutrien.</span></p><p class="MDPI17abstract"><strong><span lang="EN-US">Metode:</span></strong><span lang="EN-US"> Pakan penelitian menggunakan konsentrat mengandung menir kedelai tanpa proteksi dan terproteksi menggunakan formaldehid kadar 37%. Domba penelitian sebanyak 15 ekor dibagi dalam tiga macam perlakuan masing-masing perlakuan diulang satu kali menggunakan satu ekor DEG setiap ulangan. Perlakuan terdiri dari P1 = 30% rumput gajah (RG) + 55% konsentrat basal (KB) + 10% menir kedelai tanpa terproteksi (MKTP) + 5% menir kedelai proteksi (MKP), P2 = 30% rumput gajah (RG) + 55% konsentrat basal (KB) + 7.5% menir kedelai tanpa terproteksi (MKTP) + 7.5% menir kedelai proteksi (MKP), dan P3 = 30% rumput gajah (KB) + 55% konsentrat basal (KB) + 5% menir kedelai tanpa terproteksi (MKTP) + 10% menir kedelai proteksi (MKP). Parameter meliputi konsumsi, kecernaan dan efisiensi penggunaan nutrien bahan kering, bahan organik dan protein kasar.</span></p><p class="MDPI17abstract"><strong><span lang="EN-US">Hasil: </span></strong><span lang="EN-US">Hasil penelitian menunjukan bahwa perbedaan perlakuan berpengaruh nyata terhadap konsumi, kecernaan dan protein kasar dapat dicerna (P>0,05) dan tidak berpengaruh terhadap konsumsi dan kecernaan bahan kering dan bahan organik serta bahan kering dan bahan organik tercerna (P<0,05). Kecernaan protein kasar P1, P2 dan P3 berturut-turut 72,33%, 75,17%, dan 77,13%.</span></p><strong><span lang="EN-US">Kesimpulan: </span></strong><span lang="EN-US">Pakan P3 untuk penggemukan DEG dihasilkan konsumsi dan kecernaan protein kasar yang lebih tinggi dan efisien dari pada P1.</span>

Background: Melanocyte-keratinocyte transplant procedure (MKTP) successfully repigments postburn leukodermic scars. Objective: To further investigate the efficacy and limitations of MKTP for treatment of mechanically and chemically induced leukodermic scars. Methods: Ten patients with mechanically or chemically induced, depigmented or hypopigmented scars were preoperatively evaluated with Wood’s light examination, treated with MKTP, and followed for at least 6 months, with monitoring of repigmentation and colour matching. Results: Nine patients attended at least 6 months of follow-up. Six patients showed no fluorescence of scars under Wood’s lamp. All patients whose lesions didn’t fluoresce displayed less than 50% repigmentation, with 5 of 6 attaining 0% to 24%. Of the 3 patients displaying bright or some fluorescence, more than 95% repigmentation was achieved in 2 patients (skin phototypes V and VI), while less than 24% was attained for the third (skin phototype II). Conclusions: In this small case series, lack of fluorescence in leukodermic scars may be a useful negative prognostic indicator for MKTP, but additional trials are needed to verify that this is not due to melanocompetency.

<p><strong>Abstract.</strong> Due to the multiplicity of tombs in the area and the work of early archaeologists in Deir el-Bahari, the necropolis resembles a Swiss cheese. In addition, most of these monuments and their remains (coffin fragments, human remains, subsidiary structures) were left unpublished. A century later, the major purpose of the Middle Kingdom Theban Project of University of Alcala (MKTP-UAH) –led by the moudir (i.e. “director” in Arabic) Antonio Morales– is to document, understand, and publish all these monuments and findings left behind by previous expeditions. Such publications will shed light not only on the necropolis and owners of the monuments, but will also help to understand the beginning of the so-called Middle Kingdom, a golden age of the pharaonic period with a significant and impressive architecture. In fact, most of archaeologists take these tombs as patterns for this golden age, without taking into consideration that we do not know much about them.</p>

The US Pharmacopeia gives recommendations for the storage of pharmaceuticals in terms of mean kinetic temperature (MKT). This study's goal was to determine if color-changing time–temperature indicator (TTI) labels can reliably monitor the MKTs of medications used in ambulances. TTI labels and miniature electronic temperature recorders were placed into the drug storage boxes in five advanced life support units over 12 summer weeks in 1995 and in three advanced life support units over 4 weeks the following winter. The electronic temperature recorders measured temperatures several times per hour, and the color changes of the TTI labels were analyzed periodically. The MKTs calculated from the TTI labels differed from those calculated from the electronic temperature recorders in all cases by less than 1° C. The authors concluded that TTIs can accurately measure the MKTs of medications used in ambulances. A TTI label on each medication container may help insure that drugs are not damaged by temperture conditions.

Zusammenfassung. In den letzten zehn Jahren sind eine Reihe von psychotherapeutischen Interventionen entstanden, die sich als „metakognitiv“ bezeichnen. Dazu zählen unter anderem die Metakognitive Therapie nach Wells, die Metacognitive Reflection and Insight Therapy und das Metakognitive Training. Begriffliche Unschärfe und Überschneidungen führten zunehmend zu Unsicherheiten, wie diese neuen Konzepte einzuordnen und von bereits bestehenden abzugrenzen sind. Der vorliegende Artikel hat das Ziel zunächst eine Übersicht über den Begriff „Metakognition“ zu geben, metakognitive Ansätze in Bezug zur klassischen kognitiven Verhaltenstherapie einzuordnen und schließlich die oben genannten metakognitiven Interventionen vorzustellen. Abschließend wird das Metakognitive Training für Depression (D-MKT) als Beispiel für ein metakognitives Training näher vorgestellt und über aktuelle Entwicklungen des D-MKTs berichtet (Zusatzmodule zu Suizidalität, spezielle Versionen für ältere Menschen und für die Zielgruppe von Flüchtlingen).

Control of gene expression in kinetoplastids such as trypanosomes depends heavily on RNA-binding proteins that influence mRNA decay and translation. We previously showed that the trypanosome protein MKT1 forms a multicomponent protein complex: MKT1 interacts with PBP1, which in turn recruits LSM12 and poly(A)-binding protein. MKT1 is recruited to mRNAs by sequence-specific RNA-binding proteins, resulting in stabilization of the bound mRNA. We here show that PBP1, LSM12, and a 117-residue protein, XAC1 (Tb927.7.2780), are present in complexes that contain either MKT1 or an MKT1-like protein, MKT1L (Tb927.10.1490). All five proteins are present predominantly in the complexes, and we found evidence for a minor subset of complexes containing both MKT1 and MKT1L. XAC1-containing complexes reproducibly contained RNA-binding proteins that were previously found associated with MKT1. Moreover, XAC1- or MKT1-containing complexes specifically recruited one of the two poly(A)-binding proteins, PABP2, and one of the six cap-binding translation initiation complexes, EIF4E6–EIF4G5. Yeast two-hybrid assay results indicated that MKT1 directly interacts with EIF4G5. MKT1–PBP1 complexes can therefore interact with mRNAs via their poly(A) tails and caps, as well as through sequence-specific RNA-binding proteins. Correspondingly, MKT1 is associated with many mRNAs, although not with those encoding ribosomal proteins. Meanwhile, MKT1L resembles MKT1 at the C terminus but additionally features an N-terminal extension with low-complexity regions. Although MKT1L depletion inhibited cell proliferation, we found no evidence that it specifically interacts with RNA-binding proteins or mRNA. We speculate that MKT1L may compete with MKT1 for PBP1 binding and thereby modulate the function of MKT1-containing complexes.

Cara yang ditempuh yaitu dengan pemakaian kontrasepsi.baik MKJPmaupunnon MKJP. Tujuan penelitian ini adalah untuk mengetahui ada atau tidak perbedaan pemilihan kontrasepsi MKJP dan non MKJP berdasarkan efek samping pada dua kelompok usia reproduksi. Penelitin ini menggunakan desain cross sectional, pengambilan data dengan kuesioner. Sampel dalam penelitian ini adalah akseptor KB baik MKJP maupun non MKJP pada bulan april sampai juni sebanyak 200 responden, dimana tekhnik pengambilan datanya dengan random sampling dan kuota sampling. Hasil penelitian kemudian diuji dengan mann-whitney test.Hasil penelitian dengan uji mann whitney test diperoleh p = 0.662 dengan kata lain p > α (0.05) yang berarti tidak ada perbedaan pemilihan MKJP dan non MKJP berdasarkan efek samping di Wilayah Kabupaten Semarang.
ABSTRACT
Reproductive-age category can be divided into three groups which are the group of delayed interval pregnancy (less than 20 years old), the group of intervalcontrol pregnancy (20 to 30 years old), and the group of high risk pregnancy (more than 30 years old). An alternative to avoid high risk pregnancy is by using contraception tool namely long-term contraception (MKJP) and non long-term contraception (non MKJP).The purpose of this research is to analysedwhether there are differences in choosing MKJP and non –MKJP based on side effects in the two reproductive-age groups.This research was an explanatory research with cross-sectional design. The population were all women of contraception acceptors in Semarang Regency.The samples were 200 respondents, used simple random sampling and quota sampling. This research used quisionaire instrument and analyze used mann whitney test (α=0,05). Theresult showed thatP = 0,662 meaning P > α = 0.05 which means there is no difference in choosing MKJP and non-MKJP based on side effects in the two reproduction-age groups in Semarang regency.

Pertimbangan dalam pemilihan MKJP diidentifikasi berhubungan dengan beberapa faktor diantaranya adalah dukungan suami. Namun, Dukungan suami dalam kepesertaan KeluargaBerencana khususnya kontrasepsi jangka panjang belum pernah dieksplorasi sebelumnya. Penelitian ini bertujuan untuk memahami secara mendalam dukungan suami terhadap pemilihan kontrasepsi jangka panjang.Metode: Penelitian deskriptif kualitatif dengan metode wawancara mendalam dan observasi langsung pada 16 partisipan pria pasangan usia subur. Partisipan dalam penelitian ini diperoleh dari teknik purposive sampling dengan kriteria, merupakan suami dari istri pengguna MKJP yang berusia 30-49 tahun dan bisa baca tulis. Penelitian dilaksanakan di desa Tlogopojok dengan tenik wawancara terbuka dan analisis ColaiziHasil: Terdapat 5 tema yaitu MKJP lebih mudah dan praktis, Pemilihan jenis MKJP menjadi keputusan bersama, MKJP memberikan pengalaman yang positif, MKJP pilihan yang aman dan menenangkan, dan dukungan suami dalam penggunaan MKJP.Kesimpulan: Penggunaan MKJP dipengaruhi oleh peran suami dalam pengambilan keputusan dan dukungan suami. Penelitian ini menemukan dukungan yang diberikan suami yaitu dalam bentuk transportasi, informasi dan diskusi bersama. Selain itu MKJP juga dinilai ungguluntuk menunda atau mencegah kehamilanyang tidak diinginkan dan aman untuk kesehatan.

Alat kontrasepsi merupakan produk yang digunakan oleh akseptor KB untuk memenuhi kebutuhan fertilitasnya. Dengan pilihan alat kontrasepsi yang beragam, akseptor KB memiliki peluang memilih jenis alat kontrasepsi yang digunakan. Berdasarkan tingkat efektifitas alat kontrasepsi untuk mencegah kehamilan, jenis kontrasepsi yang paling efektif adalah Metode Kontrasepsi Jangka Panjang (MKJP). Persepsi akseptor terhadap alat kontraepsi berpengaruh terhadap metode kontrasepsi yang dipilih. Persepsi akseptor yang tidak benar mengenai MKJP menyebabkan ketidaksesuain akseptor memilih alat kontrasepsi. Penelitian ini menggunakan data sekunder ICMM 2016. Penilitian ini menggunakan distribusi frekuensi dan mean untuk mengetahui gambaran persepsi terhadap ragam alat kontrasepsi MKJP dan Non MKJP. Sampel dalam penelitian ini adalah wanita usia subur (WUS) usia 15 – 49 Tahun yang menggunakan alat kontrasepsi di Provinsi Nusa Tenggara Barat (NTB) yang berjumlah 9100 responden. Penelitian ini menunjukan gambaran persepsi preferensi terhadap alat kontrasepsi, persepsi perbandingan kesamaan alat kontrasepsi MKJP dan Non MKJP, persepsi karakteristik alat kontrasepsi yang diinginkan responden terkait kemudahan dan efektivitas, dan persepsi perbandingan kesamaan karakteristik antara alat kontrasepsi MKJP dan Non MKJP. Dalam upaya peningkatan cakupan penggunaan MKJP diperlukkan strategi komunikasi dalam pembuatan program promosi dengan melihat gambaran persepsi akseptor terhadap ragam alat kontrasepsi yang akan mereka pilih atau mereka gunakan. Pendahuluan: MKJP adalah alat kontrasepsi untuk menunda, menjarangkan kehamilan serta menghentikan kesuburan yang digunakan dalam jangka panjang. Selain itu, MKJP lebih rasional dan mempunyai efek samping sedikit. Data Profil Kesehatan Nasional Tahun 2019 didapatkan hasil pencapaian peserta KB aktif per alat kontrasepsi sebagai berikut ; IUD 7,40%, MOW 2,70%, MOP 0,50%, implan 7,40%, kondom 1,20%, suntik 63,70%, dan pil 17,00%. Data tersebut menunjukkan bahwa kontrasepsi jangka pendek menjadi pilihan utama masyarakat (Kementerian Kesehatan RI, 2019). Metode: Literatur mancanegara ditelusuri melalui sarana media elektronik dengan penuntun kata kunci. Artikel terseleksi sejumlah 8, yang masing- masing mewakili faktor-faktor Yang Berhubungan Dengan Rendahnya Minat Ibu Terhadap Penggunaan Metode Kontrasepsi Jangka Panjang (MKJP) yang bervariatif. Hasil: Adanya perbedaan motivasi ibu dalam memilih alat kontrasepsi seperti, ibu, pekerjaan, paparan sumber informasi MKJP dan jenis kontrasepsi Non-MKJP. Pembahasan: Salah satu faktor yang paling mempengaruhi pemilihan alat kontrasepsi jangka Panjang (MKJP) adalah tingkat pengetahuan ibu hamil khususnya bagi kalangan menengah ke bawah. ABSTRACT Cells of budding yeast give rise to mother and daughter cells, which differ in that only mother cells express the HO endonuclease gene and are thereby able to switch mating types. In this study, we identified the MKT1 gene as a positive regulator of HO expression. The MKT1 gene encodes a protein with two domains, XPG-N and XPG-I, which are conserved among a family of nucleases, including human XPG endonuclease. Loss of MKT1 had little effect on HO mRNA levels but resulted in decreased protein levels. This decrease was dependent on the 3′ untranslated region of the HO transcript. We screened for proteins that associate with Mkt1 and isolated Pbp1, a protein that is known to associate with Pab1, a poly(A)-binding protein. Loss of PBP1 resembles an mkt1Δ deletion, causing decreased expression of HO at the posttranscriptional level. Mkt1 and Pbp1 cosedimented with polysomes in sucrose gradients, with Mkt1 distribution in the polysomes dependent on Pbp1, but not vice versa. These observations suggest that a complex of Mkt1 and Pbp1 regulates the translation of HO mRNA.

Penelitian yang digunakan adalah penelitianDeskriptif Kualitatif, data yang ditemukan dari hasil wawancara dan pengamatantersebut akan dianalisis secara Kualitatif dibantu dengan penggunaan Microsoft Excell2010. Hasil penelitian ini menjelaskan bahwa bahwa masyarakat kurang teredukasiperihal manfaat dan tata cara penggunaanya, disisi lain faktor budaya juga turutmendukung respon negatif masyarakat tersebut. Sedangkan faktor-faktor yang secaralangsung memengaruhi penggunaan MKJP di Kecamatan Batauga diataranya adalahkurangnya pengetahuan tentang MKJP, Kurangnya Komunikasi-Informasi danEdukasi terkait MKJP kepada masyarakat, mahalnya biaya penggunaan MKJPditengah ekonomi masyarakat kecamatan batauga yang tergolong rendah, dankurangnya dukungan pria/ suami dalam penggunaan MKJP.

Long-term contraception method (MKJP) is a type of contraception that is very effective for avoiding births, regulating birth intervals and not affecting sexual relations that can last for 3 years to a lifetime such as IUD, Implants, MOW and MOP. Long-term contraceptive methods (MKJP) have proven to be the most effective way to reduce pregnancy rates, but until now MKJP has not yet become the choice of the majority of couples of reproductive age in Indonesia. Kabupaten Banjar, Martapura are still included in the regions where the coverage of MKJP KB is low, it can be seen in the achievement figures which show a significant decrease in 2015 reached 6.6%, in year 2016 reached 7.0% and in year 2017, only 2.3%. This research is a quantitative study with a cross sectional approach. The population and sample are mothers with criteria for women of childbearing age (WUS) as family planning acceptors who visit and seek treatment at Puskesmas Paramasan Kabupaten Banjar. A large sample of 60respondents. Data was collected by interview using a structured questionnaire, the results of data collection were analyzed using univariate and bivariate statistics with a chi square test using a computer program with a significance value (α)≤0.05. The results of the univariate analysis showed that the majority of respondents did not use MKJP KB, most of the respondents had sufficient knowledge and most respondents' attitudes were sufficient. Bivariate analysis showed that there was a correlation between knowledge of the use of MKJP KB (p-value =0.001) and there was a relationship between attitudes towards the use of MKJP KB (p-value =0.000).It is recommended for agencies to be concerned about the need for cross-sector cooperation in an effort to increase respondents' knowledge and attitudes about the advantages and disadvantages of using MKJP KB.It is expected that with the increased knowledge held by respondents about MKJP KB, then the attitude of respondents in contraceptive use for KB MKJP is expected to be better.And will affect the increase in the number of MKJP KB use.

Pertumbuhan penduduk di Indonesia berkisar antara 2,15% hingga 2,49% per tahun. Strategi dalam pelayanan kontrasepsi yang dikembangkan saat ini adalah menggalakkan pemakaian Metode Kontrasepsi Jangka Panjang (MKJP). Menurut Data Puskesmas Tahtul Yaman Kota Jambi jumlah akseptor KB IUD sebanyak 0 akseptor, Suntik 23 akseptor, Pil 384 akseptor, Kondom 293 akseptor, Implant 54 akseptor, MOP 0 akseptor dan MOW 0 akseptor. Cara kontrasepsi yang termasuk MKJP adalah IUD, Implant, MOW, dan MOP. Berdasarkan 10 akseptor yang menggunakan kontrasepsi pil dan suntik ditanyakan pada saat pengamilan data mengatakan tidak mau menggunakan IUD karena merasa takut, disamping itu belum mendapat izin suami. Dampak pemakaian pil atau pun suntikan yang dipakai selama lebih 2 tahun akan menimbulkan tekanan darah dan kenaikan berat badan (Sulistyawati, 2013 : 56).

Penelitian dilakukan pada bulan Maret sampai Desember 2017 dan pengambilan sampel dilakukan pada tanggal 21 sampai 28 Agustus 2017 dengan menggunakan lembar pertanyaan (kuesioner), selanjutnya data dianalisis menggunakan analisis univariat dan bivariat.
Berdasarkan hasil penelitian diketahui sebagian responden (35,56%) menggunakan MKJP, sebagian (64,44%) responden menggunakan Non MKJP, sebagian (51,1%) responden memiliki persepsi kurang baik tentang pemilihan MKJP, sebagian (48,9%) responden memiliki persepsi baik tentang pemilihan MKJP dan hasil uji statistik diketahui bahwa p-value = 0,000 < 0,05, berarti ada hubungan yang bermakna antara persepsi dengan pemilihan MKJP.
Berdasarkan hasil penelitian masih ada ibu yang belum memakai KB MKJP. Disarankan kepada Kelurahan Tahtul Yaman untuk meminta staf puskesmas memberi penyuluhan lebih jelas lagi tentang pemakaian kontrasepsi khususnya IUD, Implan, dan sterilisasi laki-laki maupun perempuan dalam rangka meningkatkan pengetahuan dan persepsi Akseptor KB dalam pemilihan MKJP.

Pemakaian Metode Kontrasepsi JangkaPanjang (MKJP) memiliki banyak keuntungan, baik dilihat dari segi program, maupun dari sisi klien (pemakai). Di samping mempercepat penurunan Total Fertility Rate (TFR), penggunaan kontrasepsi MKJP juga lebih efisien karena dapat dipakai dalam waktu yang lama serta lebih aman dan efektif. Penelitian ini dilakukan untuk melihat faktor-faktor penyebab rendahnya penggunaan metode kontrasepsi jangka panjang (MKJP) di Desa Teupin Raya Peusangan Siblah Krueng Kabupaten Bireuen tahun 2018. Desain penelitian yang digunakan adalah analitik crosssectiona, yang dilakukan pada tanggal 10 Juli 2018. Populasi dalam penelitian ini yaitu seluruh akseptor KB yang ada di Desa Teupin Raya Kecamatan Peusangan Siblah Krueng Kabupaten Bireuen. Tehnik pengambilan sampel yaitu dengan cara total populasi sebanyak 41 orang. Hasil penelitian dilihat dari pengetahuan nilai p (0,124) > p value (0,05) berarti Ha ditolak dan Ho diterima, dengan demikian pengetahuan tidak menyebabkan pemakaian MKJP di Desa Teupin Raya Kecamatan Peusangan Siblah Krueng Kabupaten Bireuen. Dilihat dari sosial budaya, menunjukkan nilai p (0,001) < p value (0,05) berarti Ha diterima dan Ho ditolak, dengan demikian sosial budaya menyebabkan rendahnya pemakaian MKJP di Desa Teupin Raya Kecamatan Peusangan Siblah Krueng Kabupaten Bireuen. Dilihat dari peran serta tenaga kesehatan, hasil perhitungan menunjukkan nilai p (1,000) > p value (0,05) berarti Ha ditolak dan Ho diterima, dengan demikian peran serta tenaga kesehatan tidak menyebabkan rendahnya pemakaian MKJP di Desa Teupin Raya Kecamatan Peusangan Siblah Krueng Kabupaten Bireuen. Diharapkan kepada responden untuk lebih memilih MKJP khususnya bagi akseptor yang tidak mengalami penyakit dalam kontraindikasi MKJP, karena MKJP memiliki efek samping yang rendah dibandingkan kontrasepsi lainnya.

Muscle cells are frequently subjected to severe conditions caused by heat, oxidative, and mechanical stresses. The small heat shock proteins (sHSPs) such as αB-crystallin and HSP27, which are highly expressed in muscle cells, have been suggested to play roles in maintaining myofibrillar integrity against such stresses. Here, we identified a novel member of the sHSP family that associates specifically with myotonic dystrophy protein kinase (DMPK). This DMPK-binding protein, MKBP, shows a unique nature compared with other known sHSPs: (a) In muscle cytosol, MKBP exists as an oligomeric complex separate from the complex formed by αB-crystallin and HSP27. (b) The expression of MKBP is not induced by heat shock, although it shows the characteristic early response of redistribution to the insoluble fraction like other sHSPs. Immunohistochemical analysis of skeletal muscle cells shows that MKBP localizes to the cross sections of individual myofibrils at the Z-membrane as well as the neuromuscular junction, where DMPK has been suggested to be concentrated. In vitro, MKBP enhances the kinase activity of DMPK and protects it from heat-induced inactivation. These results suggest that MKBP constitutes a novel stress-responsive system independent of other known sHSPs in muscle cells and that DMPK may be involved in this system by being activated by MKBP. Importantly, since the amount of MKBP protein, but not that of other sHSP family member proteins, is selectively upregulated in skeletal muscle from DM patients, an interaction between DMPK and MKBP may be involved in the pathogenesis of DM.

Devotion is done through counseling and examination of maternal health conditions that include examination of body weight, blood pressure, cholesterol. This devotion is done to 26 women of childbearing age in the village area candirejo kab. Semarang and implemented on 13 October and 5 November 2017.Evaluation conducted by giving questions on the pre and post counseling, while the examination data is written on the recap examination.The result of this devotion is from 11 IUD acceptors there are 17% have mild anemia while from 15 Acceptor Implant as much as 13% have anemia, equal to 36% IUD acceptor and 27% acceptor Implant has increased blood sugar level when, the increase of cholesterol level on Iud acceptor equal to 27% and 33% on the Implant acceptor. Melalui keluarga berencana sebuah keluarga merencanakan jumlah anak dan jarak kelahiran anak pertama dengan anak berikutnya. Salah satu yang berkaitan dengan kesadaran keluarga berencana pada masyarakat adalah pendidikan. Rendahnya fenomena pemakaian Metode Kontrasepsijangka Panjang (MKJP) pada akseptor KB aktif dan akseptor KB baru dikhawatirkan angka putus pakai yang tinggi sehingga lebih banyak terjadi drop out dan berkontribusi pada tingginya Laju Pertumbuhan Penduduk (LPP), angka Total Fertility Rate (TFR) dan Angka Kematian Ibu. Tujuan Penelitian ini adalah mengetahui Pengaruh Tingkat Pendidikan dan Usia Terhadap Penggunaan MKJP.
Desain penelitian ini Analitik korelasional dengan pendekatan Crosssectional. Populasi Penelitian adalah Seluruh Akseptor KB di RT/ RW 01/01 Desa Jatirejo Kecamatan Banyakan Kabupaten Kediri. Dengan Teknik Sampling Purposive Sampling didapatkan sampel 37 Responden. Instrumen yang digunakan berupa Kuesioner. Analisa Multivariat dengan Regresi Logistik Ganda.
Hasil Penelitian dari 37 Responden dengan tingkat pendidikan dasar sebanyak 22 responden (59,45%) . Mayoritas Berusia 36-45 tahun sejumlah 20 orang (54,05%). Pengguna Non Metode Kontrasepsi Jangka Panjang (MKJP) sebanyak 27 responden (72,97%) dan pengguna MKJP sebanyak 10 responden (27,02%). Berdasarkan Hasil Uji Statistik Regresi Logistik Ganda didapatkan Taraf Sig. 0,006 < 0,05 sehingga Tingkat Pendidikan amempengaruhi Penggunaan MKJP, Taraf Sig 0,823 > 0,05 sehingga Usia tidak mempengaruhi penggunaan MKJP. Penggunaan MKJP dipengaruhi Tingkat Pendidikan dan Usia sebesar 35,10 %.
Pendidikan tidak hanya didapatkan secara Formal,sedangkan Usia tidak mempengaruhi kematangan Akseptor dalam mengambil pilihan kontrasepsi. Sehingga hendaknya Tenaga Kesehatan dan Kader KB lebih secara mendalam memberikan motivasi Akseptor untuk memilih MKJP melalui Metode Pendekatan Keluarga.

Metode Kontrasepsi Jangka Panjang (MKJP) adalah alat kontrasepsi yang digunakan untuk menunda kehamilan. Puskesmas Pancur Batu Kabupaten Deli Serdang memiliki jumlah penduduk terbanyak yaitu 1.895 jiwa, dengan jumlah PUS 322 orang, dan peserta KB aktif 244 orang, jumlah pengguna MKJP 56 orang dan non MKJP 188 orang. Tujuan penelitian adalah mengetahui faktor-faktor yang mempengaruhi pemakaian alat MKJP pada isteri PUS di Puskesmas Pancur Batu Kabupaten Deli Serdang Tahun 2017. Populasi adalah ibu PUS yang menggunakan alat kontrasepsi sebanyak 244 orang. Teknik pengambilan sampel menggunakan metode systematic random sampling yaitu 71 orang. Hasil penelitian bahwa dari 71 responden, jumlah mayoritas umur>36 tahun 49 orang (69,0%), pendidikan menengah 40 orang (56,3%), paritas>2 orang adalah 50 orang (70,4%), pengetahuan kurang 46 orang (64,8%), sikap negatif 46 orang (64,8%), persepsi nilai anak baik 36 orang (50,7%), KIE (Komunikasi Informasi dan Edukasi) kategori baik 50 orang (70,4%), suami tidak mendukung 41 orang (57,7%) dan tidak menggunakan MKJP 40 orang (56,3%). Kesimpulan adanya pengaruh faktor predisposisi (umur, pendidikan, paritas, pengetahuan, sikap, persepsi nilai anak), pemungkin (KIE) dan pendorong (dukungan suami) terhadap pemakaian alat MKJP pada istri PUS. Disarankan bagi PUS lebih mencari informasi tentang kontrasepsi jangka panjang dan konsultasi kepada tenaga kesehatan terdekat, karena MKJP merupakan metode yang sangat efektif bagi PUS yang memiliki anak lebih dari 2 anak.

The use of MKJP in Indonesia has decreased every year from 13.7% in 1991 to 10.6% in 2012 (Ministry of Health, 2013). In 2013, based on a mini survey of the Population and Family Planning Agency (BKKBN), the coverage of MKJP in Indonesia was only 12.4% with a percentage based on the type of method, namely KB implants (5.2%), IUD (Contraceptive Devices in the Womb) / Intrauterine Device (IUD) (4.7%), and Women's Operations Method (MOW) (2.2%), whereas the expected MKJP target is 26.7%. This research is descriptive analytic, conducted to determine factors related to the interest in using MKJP (IUD Implants) in Perigi Mekar Village in 2016. The number of samples in this study were 104 people. Data were collected using a questionnaire. Of the 9 variables studied, 2 variables showed a significant relationship with the interest in using the MKJP is income with a p value of 0.018, and discussions with a partner with a p value of 0.024. From the above statement it can be concluded that the amount of income and the frequency of discussions with partners influences the interest in using the MKJP. It is expected that community leaders can work together with local health workers so that they can provide counseling to the community regarding long-term contraceptive methods. Penelitian ini bertujuan untuk mempelajari hubungan tingkat pendidikan dan pengetahuan dengan keikutsertaan penggunaan kontrasepsi jangka panjang (MKJP) di wilayah kerja Puskesmas Jembatan Kecil. Jenis penelitian ini adalah Survey Analitik dengan desain Cross Sectional. Populasi yang diteliti dalam penelitian ini adalah seluruh akseptor KB aktif di wilayah kerja Puskesmas Jembatan Kecil pada bulan Juli-Agustus 2018. Pengambilan sampel dalam penelitian ini menggunakan teknik Accidental Sampling sebanyak 39 akseptor. Penelitian ini menggunakan data primer data sekunder. Analisis data dilakukan secara univariat dan bivariat dengan Uji Chi-Square (c2) dan Contingency Coefficient (C). Hasil penelitian didapatkan dari 39 akseptor KB, terdapat 27 akseptor (69,2%) tidak menggunakan alat kontrasepsi MKJP, 16 akseptor (41,0%) berpendidikan dasar, 18 akseptor (46,2%) berpengetahuan cukup. Ada hubungan yang signifikan antara tingkat pendidikan dengan keikutsertaan penggunaan kontrasepsi MKJP di wilayah kerja Puskesmas Jembatan Kecil dengan kategori hubungan sedang dan ada hubungan yang signifikan antara pengetahuan dengan keikutsertaan penggunaan MKJP di wilayah kerja Puskesmas Jembatan Kecil dengan kategori hubungan sedang. Diharapkan tenaga kesehatan dapat memberikan pendidikan kesehatan bagi akseptor KB tentang alat kontrasepsi yang sesuai dan efektif dalam menjarangkan kehamilan dengan melakukan penyuluhan, membagikan leaflet, poster, dan buku. Kata Kunci: MKJP, pendidikan, pengetahuan ABSTRACT Long-term contraception (MKJP) is a contraception recommended in government family planning programs because it is proven to be more efficient and effective with lower costs, failure rates, side effects and complications. This study aims to study the relationship between education level and knowledge with the participation of MKJP usage in working area of Jembatan Kecil Public Health Center. The type of this research was Analytical Survey and the design was Cross Sectional. The population of this study was all active family planning acceptors in working area of Jembatan Kecil Public Health Center in July-August 2018. The Sampling technique in this study was Accidental Sampling. The sample total was 39 acceptors. This study used primary and secondary data. Data analysis was done by univariate and bivariate analyses with Chi-Square Test (c2) and Contingency Coefficient (C). The results were obtained : of 39 family planning acceptors, 27 acceptors (69.2%) did not use MKJP contraception, 16 acceptors (41.0%) had basic education, and 18 acceptors (46.2%) had sufficient knowledge. Efforts to reduce population growth are carried out by controlling births through family planning. The contraceptive method recommended by the government is the Long Term Contraception Method (MKJP) because it is more efficient, safe, and effective. Data at the state, provincial, district / city level, as well as data from the Community Health Center, the use of non-MKJP (73.4%) is more desirable than the Long-Term Contraception Method (MKJP) (26.6%). Many factors cause someone to choose to use MKJP based on Green's theory. The purpose of this study was to analyze the factors that influence EFA in the selection of the Long-Term Contraception Method (MKJP). This type of research is an analytic survey with a cross sectional design. The research was conducted in the working area of the Medan Teladan Community Health Center. The population of PUS mothers was 2,897 people, and the sample was obtained 110 people. Data analysis was univariate, bivariate using chi-square and multivariate using multiple logistic regression at the 95% confidence level ( = 0.05). The results showed that the variables that had an effect on the selection of MKJP in the Medan Teladan Community Health Center were knowledge (p = 0.002), family / husband support (p = 0.038), and the role of cadres (p = 0.001). The variables that had no effect were attitude (p = 0.153), accessibility (p = 0.061), and peer support (p = 0.098). The most dominant factor was the role of the cadre variable with a value of Exp (B) / OR = 8.819, which means that respondents who stated that the role of cadres was supportive had the opportunity to choose MKJP by 8.8 times higher than stating that the role of cadres was not supportive. The conclusion of this study is that the selection of MKJP by Fertile Age Couples (PUS) is influenced by knowledge, family / husband support, and the role of cadres. It is recommended that KB cadres increase counseling to husbands and wives about MKJP by offline (face to face, leaflets, brochures, etc.) and online (internet) during the Covid-19 period.

A tanulmány célja a Magyar Kétfarkú Kutya Párt (MKKP) 2015-ös ellen-plakátkampányának bemutatása és elemzése. A kutatás a relevancia-elmélet és a humor inkongruencia-feloldás elmélet segítségével próbálja feltárni azokat a magüzeneteket, amik a MKKP plakátjain megjelentek. A relevancia-elmélet keretében a vizsgálódás azokat az implikált premisszákat és implikált konklúziókat kívánja megtalálni, amik a humoros szövegekben rejlenek. A tanulmány két fő részből tevődik össze. Az első rész az elméleti háttérrel foglalkozik: a plakátok alkalmazását politológiai szempontok szerint vizsgálja. Említésre kerülnek a humoros plakátok fajtái Eduardo José Marcos Camilo írásai alapján, amik lehetnek fatikusak, kommerciálisak, érzelmiek és intertextuálisak. Az MKKP plakátjait az intertextuális kategóriába sorolhatjuk. Az elemzéshez nélkülözhetetlen relevancia-elmélet is megjelenik, ami Sperber és Wilson szándékközpontú, rámutató-következtetéses kommunikációs teóriája. A dolgozat második felében az MKKP plakát kampánya kerül elemzésre. Ezt három fő kategóriára oszthatjuk témájuk szerint, amelyek a korrupció, a szegénység és a szolidaritás. Ezek a témakörök ki is jelölik azokat a magüzeneteket, amikkel az MKKP alternatívát kínál a kormányzat, illetve a kormánypárt üzeneteire.

Banyak faktor yang mempengaruhi Penggunaan metode kontrasepsi jangka panjang antara lain pengetahuan, umur dan partisipasi suami. Berdasarkan data Dinas Kesehatan Kota Jambi didapatkan bahwa Puskesmas Paal V dengan jumlah PUS 7.220 orang, dengan KB aktif berjumlah 5.802 orang. Metode kontrasepsi jangka Panjang (MKJP) yang digunakan adalah Implant 3,01%, AKDR 9,5%, MOW 3,1%, dan MOP 0,3%. Sedangkang Non MKJP adalah suntik 60,1%, Pil 21,6% dan kondom 2,46%. Penelitian ini merupakan penelitian analitik dengan menggunakan rancangan cross sectional. Populasi adalah seluruh wanita usia subur di Puskesmas Paal V Kota Jambi Tahun 2018 yang berjumlah 5.802 orang. Penelitian ini menggunakan teknik Accidental sampling. Pengumpulan data primer dengan kuesioner. Penelitian dilakukan dari bulan Mei s/d Agustus 2018 dengan analisis Univariat dan Bivariat dengan uji chi squre.Hasil penelitian menunjukkan bahwa dari 95 responden yang diteliti tentang penggunaan metode kontrasepsi jangka panjang responden yang menggunakan MKJP sebanyak 52 responden. Hasil penelitian ini dapat dilihat bahwa responden yang memiliki pengetahuan kurang baik sebanyak 71 responden. Mayoritas umur sebanyak 50 responden yakni umur 25-35 tahun . mayoritas partisipasi suami yang mendukung sebanyak 52 responden. Ada hubungan pengetahuan dengan penggunaan MKJP dengan p-value (0,000) 0,05. Ada hubungan umur dengan penggunaan MKJP dengan p-value (0,000) 0,05. Ada hubungan partisipasi suami dengan penggunaan MKJP dengan p-value (0,000) 0,05.

Rendahnya angka pengguna MKJP ini dinilai menimbulkan beberapa permasalahan seperti tingginya angka kegagalan penundaan atau penjarangan memiliki anak.Pemilihan metode kontrasepsi ini tentunya dapat dipengaruhi oleh beberapa faktor.Tujuan penelitian ini adalah menentukan determinan yang berhubungan dengan pemilihan alat kontrasepsi pada akseptor KB di PMB Tuti Gambut Kabupaten Banjar Kalimantan Selatan. Penelitian ini menggunakan desain cross sectional dengan desain penelitian analitik.Populasi dalam penelitian ini adalah seluruh akseptor KB di PMB Tuti Gambut. Penelitian ini menggunakan tehnik simple random sampling dan didapatkan sampel yang diambil adalah sejumlah 75 orang.Tehnik pengambilan data menggunakan data sekunder, analisis data dilakukan dengan menggunakan uji chi square.Hasil penelitian ini yaitu dari segi usia akseptor KB yang ada di PMB Tuti terbanyak adalah pada usia tidak berisiko (usia reproduktif) yaitu usia 20 sampai 35 tahun. Akseptor dengan paritas yang tidak berisiko lebih banyak dari pada akseptor yang memiliki paritas yang berisiko (paritas lebih dari 3), sedangkan akseptor yang berpendidikan tinggi lebih sedikit dari pada akseptor yang memiliki pendidikan rendah.Akseptor yang menggunakan alat kontrasepsi jangka panjang lebih sedikit dibandingkan dengan akseptor yang menggunakan kontrasepsi non MKJP. Faktor determinan yang berhubungan dalam pemilihan kontrasepsi MKJP adalah usia, sedangkan faktor paritas dan pendidikan tidak berhubungan dengan pemilihan kontrasepsi MKJP.

Keluarga Berencana (KB) menurut World Health Organisation (WHO) adalah tindakan yang membantu individu atau pasangan suami istri untuk menghindari kelahiran yang tidak diinginkan, mendapatkan kelahiran yang diinginkan, mengatur interval kelahiran, mengontrol waktu saat kelahiran dalam hubungan dengan umur suami dan istri, menentukan jumlah anak dalam keluarga. Salah satu faktor yang memengaruhi keikutsertaan sebagai akseptor metode kontrasepsi jangka panjang adalah pengetahuan suami istri mengenai kontrasepsi tersebut. Tujuan penelitian ini adalah untuk mengetahui hubungan tingkat pengetahuan akseptor KB tentang metode kontrasepsi jangka panjang (MKJP) dengan keikutsertaan MKJP di banjar Air Kuning Kecamatan Jembrana. Penelitian ini menggunakan rancangan non eksperimen dengan pendekatan kuantitatif. Desain penelitian yang digunakan adalah penelitian analitik dengan pendekatan cross sectional yaitu melakukan pengumpulan data dalam satu waktu dan tiap subyek penelitian diobservasi sekali saja. Hasil uji statistik chi square (X2) didapatkan hasil hitung untuk tingkat pengetahuan akseptor KB tentang metode kontrasepsi jangka panjang (MKJP) sebesar 61,267 dengan tingkat kemaknaan 95% dengan nilai p<0.05 yang artinya terdapat hubungan yang signifikan antara tingkat pengetahuan akseptor KB tentang metode kontrasepsi jangka panjang (MKJP) dengan keikutsertaan terhadap MKJP. Whether the mitochondrial ATP-dependent potassium (mKATP) channel is the trigger or the mediator of cardioprotection is controversial. We investigated the critical time sequences of mKATP channel opening for cardioprotection in isolated rabbit hearts. Pretreatment with diazoxide (100 μM), a selective mKATP channel opener, for 5 min followed by 10 min washout before the 30-min ischemia and 2-h reperfusion significantly reduced infarct size (9 ± 3 vs. 35 ± 3% in control), indicating a role of mKATP channels as a trigger of protection. The protection was blocked by coadministration of the L-type Ca2+ channel blockers nifedipine (100 nM) or 5-hydroxydecanoic acid (5-HD; 50 μM) or by the protein kinase C (PKC) inhibitor chelerythrine (5 μM). The protection of diazoxide was not blocked by 50 μM 5-HD but was blocked by 200 μM 5-HD or 10 μM glybenclamide administrated 5 min before and throughout the 30 min of ischemia, indicating a role of mKATP opening as a mediator of protection. Giving diazoxide throughout the 30 min of ischemia also protected the heart, and the protection was not blocked by chelerythrine. Nifedipine did not affect the ability of diazoxide to open mKATP channels assessed by mitochondrial redox state. In electrically stimulated rabbit ventricular myocytes, diazoxide significantly increased Ca2+ transient but had no effect on L-type Ca2+ currents. Our results suggest that opening of mKATP channels can trigger cardioprotection. The trigger phase may be induced by elevation of intracellular Ca2+ and activation of PKC. During the lethal ischemia, mKATPchannel opening mediates the protection, independent of PKC, by yet unknown mechanisms.

Family planning aims at reducing maternal mortality rates and reducing population growth rate with the main target of couples of childbearing age. One of the family planning options offered is long-term reversible contraceptive method (MKJP). The purpose of this study was to determine the factors associated with interest in using MKJP in the work area of Camplong health center, Fatuleu sub-district, Kupang regency in 2018. The research was quantitive study with cross-sectional approach. The sample of 77 people was selected using simple random sampling. The instrument uses was a questionnaire. Data analysis techniques consisted of descriptive and bivariate analysis (chi-square test) with α= 0,05. The study indicated that knowledge (p-value= 0,09) and the role of health workers (p-value =0,001) were associated with the use of the MKJP while attitude (p-value = 0,765) and husband support (p-value = 0,131) had no relationship with MKJP. Health center needs to regularly provide information to the community about the advantages of MKJP especially for couples of childbearing age who have already had more than three children.

Program KB (Keluarga Berencana) merupakan suatu bentuk upaya yang dilakukan oleh pemerintah untuk mengendalikan banyaknya kuota penduduk. Program KB selalu dikaitkan dengan alat kontrasepsi sebagai kendaraan untuk menyukseskan program tersebut. Metode kontrasepsi dibagi menjadi dua yaitu jangka panjang (MKJP) yang meliputi Intra Uterine Device, Implan, Metode Operasi Wanita dan Metode Operasi Pria dan jangka pendek (Non MKJP) yang meliputi Suntik, Kondom, dan Pil. Penelitian ini bertujuan untuk memetakan Kabupaten/Kota di Jawa Tengah berdasarkan metode kontrasepsi yang digunakan oleh peserta KB dalam dua dimensi. Metode pemetaan yang digunakan adalah Multidimensional Scaling Metrik dengan membangun suatu program berbasis Graphical User Interface (GUI) Python. Hasil penelitian ini memvisualisasikan karakteristik dari peserta KB pada Kabupaten/Kota berdasarkan jenis dan metode kontrasepsi yang digunakan. Pada kuadran I memiliki karakteristik penggunaan Non MKJP. Kuadran II memiliki karakteristik penggunaan PIL. Kuadran III merupakan kelompok dengan tingkat penggunaan kontrasepsi yang rendah baik MKJP maupun Non MKJP. Kuadran IV memiliki karakteristik penggunaan IUD. Dengan kriteria perceptual map yang dihasilkan sempurna, ditunjukan oleh nilai stress sebesar 0.4%.

Kgiatan Pengabdian masyarakat dilakukan di wilayah Kabupaten Sleman, dilaksanakan pada Bulan Agustus sampai dengan September 2020. Hasil kegiatan pengabdian kepada masyarakat ini di ikuti oleh 88 akseptor yang terdiri dari 60 akseptor IUD dan 28 akseptor implant. Kegiatan Pengabdian dapat memberikan informasi kepada WUS serta memberikan kontribusi yang besar terhadap peningkatan jumlah peserta KB MKJP ( IUD dan Implant) Installation of IUDs and Implants as a Baby Boom Prevention During the Covid-19 Pandemic The purpose of this community service is to increase the number of MKJP family planning participants by installing implants and IUDs. This service method uses field studies, direct examination with activity steps; Counseling, Health Education about KB MKJP IUD, Implants, and the installation of Implants and IUDs. Community service activities are carried out in the Sleman Regency area, carried out from August to September 2020. The results of this community service activity were participated in by 88 acceptors consisting of 60 IUD acceptors and 28 implant acceptors. Service activities can provide information to WUS and make a major contribution to increasing the number of MKJP family planning participants (IUD and Implant).

ABSTRACT Mitogen-activated protein (MAP) kinase phosphatases (MKPs) are dual-specificity phosphatases that dephosphorylate phosphothreonine and phosphotyrosine residues within MAP kinases. Here, we describe a novel posttranslational mechanism for regulating MKP-3/Pyst1/DUSP6, a member of the MKP family that is highly specific for extracellular signal-regulated kinase 1 and 2 (ERK1/2) inactivation. Using a fibroblast model in which the expression of either MKP-3 or a more stable MKP-3-green fluorescent protein (GFP) chimera was induced by tetracycline, we found that serum induces the phosphorylation of MKP-3 and its subsequent degradation by the proteasome in a MEK1 and MEK2 (MEK1/2)-ERK1/2-dependent manner. In vitro phosphorylation assays using glutathione S-transferase (GST)-MKP-3 fusion proteins indicated that ERK2 could phosphorylate MKP-3 on serines 159 and 197. Tetracycline-inducible cell clones expressing either single or double serine mutants of MKP-3 or MKP-3-GFP confirmed that these two sites are targeted by the MEK1/2-ERK1/2 module in vivo. Double serine mutants of MKP-3 or MKP-3-GFP were more efficiently protected from degradation than single mutants or wild-type MKP-3, indicating that phosphorylation of either serine by ERK1/2 enhances proteasomal degradation of MKP-3. Hence, double mutation caused a threefold increase in the half-life of MKP-3. Finally, we show that the phosphorylation of MKP-3 has no effect on its catalytic activity. Thus, ERK1/2 exert a positive feedback loop on their own activity by promoting the degradation of MKP-3, one of their major inactivators in the cytosol, a situation opposite to that described for the nuclear phosphatase MKP-1.

We have previously reported that MAPK phosphatase-1 (MKP-1/CL100) is a thrombin-responsive gene in endothelial cells (ECs). We now show that VEGF is another efficacious activator of MKP-1 expression in human umbilical vein ECs. VEGF-A and VEGF-E maximally induced MKP-1 expression in ECs; however, the other VEGF subtypes had no effect. Using specific neutralizing antibodies, we determined that VEGF induced MKP-1 specifically through VEGF receptor 2 (VEGFR-2), leading to the downstream activation of JNK. The VEGF-A165 isoform stimulated MKP-1 expression, whereas the VEGF-A162 isoform induced the gene to a lesser extent, and the VEGF-A121 isoform had no effect. Furthermore, specific blocking antibodies against neuropilins, VEGFR-2 coreceptors, blocked MKP-1 induction. A Src kinase inhibitor (PP1) completely blocked both VEGF- and thrombin-induced MKP-1 expression. A dominant negative approach revealed that Src kinase was required for VEGF-induced MKP-1 expression, whereas Fyn kinase was critical for thrombin-induced MKP-1 expression. Moreover, VEGF-induced MKP-1 expression required JNK, whereas ERK was critical for thrombin-induced MKP-1 expression. In ECs treated with short interfering (si)RNA targeting MKP-1, JNK, ERK, and p38 phosphorylation were prolonged following VEGF stimulation. An ex vivo aortic angiogenesis assay revealed a reduction in VEGF- and thrombin-induced sprout outgrowth in segments from MKP-1-null mice versus wild-type controls. MKP-1 siRNA also significantly reduced VEGF-induced EC migration using a transwell assay system. Overall, these results demonstrate distinct MAPK signaling pathways for thrombin versus VEGF induction of MKP-1 in ECs and point to the importance of MKP-1 induction in VEGF-stimulated EC migration.

Abstract Despite high dose chemotherapy and autologous stem cell transplant, multiple myeloma (MM) remains an incurable malignancy, with a median 5 year survival of less than 20%. With the exception of idiotype, few antigen targets have been identified that would facilitate specific immunotherapy of MM. We have previously described a murine monoclonal antibody that recognizes a conformation-dependent epitope on free human kappa light chains and a cell surface antigen, KMA, expressed on kappa MM plasma (MMκ) cells (Raison, RL and Boux, HA Mol Immunol 1985 22:1393). Here we show that the murine antibody, designated mKap, bound specifically to a range of MMκ cell lines and inhibited the in vitro growth of these cells. Flow cytometric analysis (Annexin-V and PI staining) of MMκcell lines incubated with mKap demonstrated a dose dependent induction of apoptosis. Furthermore, the presence of activated caspases in mKap treated cells was detected using the CaspACE™ FITC-VAD-FMK reagent. The induction, by mKap, of apoptosis in MMκ cells occured in the absence of cross-linking second antibody or effector cells. In vivo, anti-tumor activity by mKap was demonstrated in a SCID mouse tumor xenograft model. Tumor growth was measured by quantitation of secreted myeloma Ig over a period of 6 weeks. From week 4 onwards, significantly lower serum concentrations of myeloma Ig were detected in animal groups receiving 3.0, 1.5, 0.3 and 0.15 mg total mKap compared to the untreated control (P<0.005 at week 4; P<0.0001 at week 5). The tumor-restricted specificity of mKap, coupled with its ability to inhibit MMκ cell growth in vitro and in vivo, suggests the potential use of either a chimeric or humanised version of this antibody, alone or in combination with chemotherapy, for the treatment of kappa MM.

Indonesia is the nation with the third largest population in the world. One way to control the population is with the family planning program. The strategy of the implementation of KB program is increase the use of long-term contraceptive methods (MKJP) sterilization (MOW and MOP), IUD and implant. The research used descriptive research; it aimed to determine perception of acceptor KB about use of long-term contraceptive methods at the public health center Koni Jambi city year 2017. Populations were 1.686 people. It used accidental sampling technique i.e. acceptor KB who visit the public health center. Sample for this research was 42 womens. Analyzed univariate was used for statistical analysis. The collecting data used by questionnaire. The result showed 9 acceptors (21%) used MKJP and 33 acceptors (79%) choose to use contraceptive non MKJP. 29 acceptors (69%) have positive perception and 13 acceptors (31%) have negative perception about use of long-term contraceptive methods. It concluded that there are many acceptor more choose and use contraception non MKJP, it suggested to increase promotion in conducting counseling about contraception to increase awareness of acceptor for themselves especially MKJP contraceptives.

Program keluarga berencana merupakan satu strategi efektif dalam menurunkan angka kematian ibu. Hal ini akan tercapai dengan meningkatnya angka keberlangsungan kesertaan ber-KB. Metode kontrasepsi jangka panjang (MKJP) merupakan metode KB yang paling efektif dalam meningkatkan keberlangsungan kesertaan ber-KB. Provinsi Sumatera Barat merupakan satu provinsi dengan capaian peserta kb aktif terendah yaitu 63,73%. Begitu juga dengan kota Padang yaitu 15,50% ditahun 2016 dan 19,00% ditahun 2017. Penggunaan metode MKJP dipengaruhi oleh banyak faktor diantaranya kualitas dan akses pelayanan KB, keterbatasan sarana, kompetensi provider, pengetahuan, sosial-budaya, dan dukungan psikososial. Penelitian ini bertujuan menganalisis faktor yang berhubungan dengan keputusan memilih metode MKJP di Kampung KB. Penelitian ini menggunakan desain kualitatif dengan pendekatan fenomenologi terhadap 19 informan yang ditentukan dengan teknik snowball sampling. Pengumpulan data diperoleh dari wawancara mendalam, observasi dan studi dokumentasi. Analisis data dilakukan dengan constant comparative method. Hasil penelitian menunjukkan bahwa faktor yang berhubungan dengan keputusan memilih KB MKJP di wilayah Kampung KB Padang adalah dukungan psikososial dari pasangan dan orang tua, kompetensi provider, prosedur pencatatan dan pelaporan, kualitas pelayanan KB, budaya masyarakat, persepsi negative terhadap KB MKJP, dan tidak adanya indikator sasaran untuk metode kontrasepsi.

Jumlah penduduk di Indonesia dari tahun ke tahun semakin meningkat jumlahnya sehingga menempatkannya di urutan keempat sebagai negara dengan jumlah penduduk terbesar didunia. Menurut data Survei Demografi dan Kesehatan Indonesia (SDKI) 2017 menunjukkan bahwa pemakaian kontrasepsi di antara wanita berstatus kawin di Indonesia meningkat dari 60% pada tahun 2002-2003 menjadi 63,3 %. Peningkatan terbanyak terjadi pada pemakaian metode kontrasepsi suntik. Pemakaian IUD menurun selama kurun waktu 10 tahun terakhir. Dalam RPJMN 2015 – 2019 tertulis bahwa program pemerintah untuk lima tahun kedepan salah satunya adalah Peningkatan pelayanan KB dengan menggunakan MKJP. Dalam Notoatmodjo (2005) rendahnya jumlah Pengguna Metode Kontrasepsi Jangka Panjang (MKJP), dipengaruhi oleh beberapa faktor diantaranya adalah faktor predisposing yaitu faktor pengetahuan, nilai atau kepercayaan dan sikap, serta faktor pemungkin yaitu fasilitas, biaya, jarak dan ketersediaan trasnfortasi sedangkan faktor penguat yaitu dari dukungan suami dan keluargaTujuan penelitian ini adalah untuk mengetahui faktor-faktor yang berhubungan dengan partisipasi akseptor KB aktif terhadap penggunaan kontrasepsi jangka panjang di Desa Cigalontang Wilayah Kerja PKM Cigalontang tahun 2018. Jenis penelitian yang digunakan dalam penelitian ini adalah penelitian kuantitatif analitik dengan desain cross sectional. Analisa yang digunakan adalah analisa univariat dan bivariat. Populasi dalam penelitian ini adalah seluruh akseptor KB aktif dengan tekhnik pengambilan sampel menggunakan total sampling sebanyak 448 akseptor KB aktif.Berdasarkan hasil penelitian didapatkan bahwa 85% responden menggunakan metode kontrasepsi jangka panjang (MKJP) dan 15% responden tidak menggunakan MKJP. Hasil tabulasi silang didapatkan faktor yang berhubungan dengan partisipasi akseptor KB aktif terhadap penggunaan MKJP adalah paritas (p value 0,000), pekerjaan (p value 0,000), status sosio ekonomi (p value 0,000), dan dukungan suami (p value 0,000) dengan < alpha (0,05), sedangkan variabel pengetahuan (p value 0,164), dan sikap (p value 0,071) tidak berhubungan dengan penggunaan kontrasepsi jangka panjang dengan > alpha (0,05).Simpulan dalam penelitian ini adalah terdapat hubungan antara paritas, pekerjaan, status sosio ekonomi, dukungan suami dengan penggunaan kontrasepsi MKJP dan tidak ada hubungan antara pengetahuan dan sikap dengan penggunaan kontrasepsi MKJP. Saran meningkatkan konseling KB pada calon akseptor KB oleh tenaga kesehatan sehingga dapat meningkatkan pemahaman tentang metode MKJP dengan menggunakan media yang efektif serta meningkatkan keterlibatan suami dalam ber KB.

The mitogen-activated protein kinase (MAPK) pathway plays a critical role in Toll-like receptor (TLR) signaling. MAPK phosphatase-1 (MKP-1) inhibits the MAPK pathway and decreases TLR signaling, but the regulation of MKP-1 is not completely understood. We now show that MKP-1 is acetylated, and that acetylation regulates its ability to interact with its substrates and deactivate inflammatory signaling. We found that LPS activates acetylation of MKP-1. MKP-1 is acetylated by p300 on lysine residue K57 within its substrate-binding domain. Acetylation of MKP-1 enhances its interaction with p38, thereby increasing its phosphatase activity and interrupting MAPK signaling. Inhibition of deacetylases increases MKP-1 acetylation and blocks MAPK signaling in wild-type (WT) cells; however, deacetylase inhibitors have no effect in cells lacking MKP-1. Furthermore, histone deacetylase inhibitors reduce inflammation and mortality in WT mice treated with LPS, but fail to protect MKP-1 knockout mice. Our data suggest that acetylation of MKP-1 inhibits innate immune signaling. This pathway may be an important therapeutic target in the treatment of inflammatory diseases.

Mitogen-activated protein (MAP) kinase phosphatase-1 (MKP-1) is a nuclear, dual-specificity phosphatase that has been shown to dephosphorylate MAP kinases. We used a “substrate-trap” technique involving a mutation in MKP-1 of the catalytically critical cysteine to a serine residue (“CS” mutant) to capture novel MKP-1 substrates. We transfected the MKP-1 (CS) mutant and control (wild-type, WT) constructs into phorbol 12-myristate 13-acetate (PMA)-activated COS-1 cells. MKP-1-substrate complexes were immunoprecipitated, which yielded four bands of 17, 15, 14, and 10 kDa with the CS MKP-1 mutant but not the WT MKP-1. The bands were identified by mass spectrometry as histones H3, H2B, H2A, and H4, respectively. Histone H3 was phosphorylated, and purified MKP-1 dephosphorylated histone H3 (phospho-Ser-10) in vitro; whereas, histone H3 (phospho-Thr-3) was unaffected. We have previously shown that thrombin and vascular endothelial growth factor (VEGF) upregulated MKP-1 in human endothelial cells (EC). We now show that both thrombin and VEGF caused dephosphorylation of histone H3 (phospho-Ser-10) and histone H3 (phospho-Thr-3) in EC with kinetics consistent with MKP-1 induction. Furthermore, MKP-1-specific small interfering RNA (siRNA) prevented VEGF- and thrombin-induced H3 (phospho-Ser-10) dephosphorylation but had no effect on H3 (phospho-Thr-3 or Thr-11) dephosphorylation. In summary, histone H3 is a novel substrate of MKP-1, and VEGF- and thrombin-induced H3 (phospho-Ser-10) dephosphorylation requires MKP-1. We propose that MKP-1-mediated H3 (phospho-Ser-10) dephosphorylation is a key regulatory step in EC activation by VEGF and thrombin.

Mitogen-activated protein kinase phosphatase (Mkp)-1 exerts its anti-inflammatory activities during Gram-negative sepsis by deactivating p38 and c-Jun N-terminal kinase (JNK). We have previously shown that Mkp-1+/+ mice, but not Mkp-1−/− mice, exhibit hypertriglyceridemia during severe sepsis. However, the regulation of hepatic lipid stores and the underlying mechanism of lipid dysregulation during sepsis remains an enigma. To understand the molecular mechanism underlying the sepsis-associated metabolic changes and the role of Mkp-1 in the process, we infected Mkp-1+/+ and Mkp-1−/− mice with Escherichia coli i.v., and assessed the effects of Mkp-1 deficiency on tissue lipid contents. We also examined the global gene expression profile in the livers via RNA-seq. We found that in the absence of E. coli infection, Mkp-1 deficiency decreased liver triglyceride levels. Upon E. coli infection, Mkp-1+/+ mice, but not Mkp-1−/− mice, developed hepatocyte ballooning and increased lipid deposition in the livers. E. coli infection caused profound changes in the gene expression profile of a large number of proteins that regulate lipid metabolism in wildtype mice, while these changes were substantially disrupted in Mkp-1−/− mice. Interestingly, in Mkp-1+/+ mice E. coli infection resulted in downregulation of genes that facilitate fatty acid synthesis but upregulation of Cd36 and Dgat2, whose protein products mediate fatty acid uptake and triglyceride synthesis, respectively. Taken together, our studies indicate that sepsis leads to a substantial change in triglyceride metabolic gene expression programs and Mkp-1 plays an important role in this process.

ATP-sensitive K+ (KATP) channels that contain K+ inward rectifier subunits of the 6.2 isotype (Kir6.2) are important regulators of the cardiac response to ischemia-reperfusion (I/R) injury. Opening of these channels is implicated in the cardioprotective mechanism of ischemic preconditioning (IPC), but debate surrounds the contribution of surface KATP (sKATP) versus mitochondrial KATP (mKATP) channels. While responses to I/R injury and IPC have been examined in Kir6.2−/− mice before, breeding methods and other technical obstacles may have confounded interpretations. The aim of this study was to elucidate the role of Kir6.2 in cardioprotection and mKATP activity, using conventionally bred Kir6.2−/− mice with wild-type littermates as controls. We found that perfused hearts from Kir6.2−/− mice exhibited a normal baseline response to I/R injury, were not protected by IPC, and showed a blunted response to the IPC mimetic drug diazoxide. These data suggest that the loss of IPC in Kir6.2−/− hearts is not due to an underlying difference in I/R sensitivity. Furthermore, mKATP channel activity was identical in cardiac mitochondria isolated from wild-type versus Kir6.2−/− mice, suggesting no role for Kir6.2 in the mKATP. Collectively, these data indicate that Kir6.2 is required for the full response to IPC or diazoxide but is not involved in mKATP formation.

Tingginya angka fertilitas dan laju pertumbuhan penduduk yang tidak seiring dengan peningkatan kualitas penduduk akan berimbas pada beratnya beban pemerintah dalam mensejahterakan rakyatnya. Berbagai cara telah diupayakan pemerintah guna menekan angka fertilitas, salah satunya melalui program Keluarga Berencana (KB) dengan penggunaan alat/cara kontrasepsi. Dari berbagai jenis metode, Badan Kependudukan dan Keluarga Berencana Nasional (BKKBN) mengemukakan bahwa Metode Kontrasepsi Jangka Panjang (MKJP) merupakan metode kontrasepsi paling efektif dengan tingkat keberhasilan melebihi 95 persen. Namun, hasil Survei Demografi dan Kesehatan Indonesia (SDKI) tahun 2017 menyatakan hanya 13,2 persen wanita usia subur (WUS) berstatus kawin yang menggunakan MKJP, padahal target paruh waktu Rencana Pembangunan Jangka Menengah Nasional (RPJMN) pada 2016 harus mencapai 21,1 persen. Melihat rendahnya capaian tersebut, penelitian ini bertujuan untuk menganalisis determinan status penggunaan MKJP di Indonesia tahun 2017 menggunakan regresi logistik biner yang mengakomodir penimbang survei. Data sekunder diperoleh dari raw data hasil SDKI 2017. Hasil analisis deskriptif menunjukkan bahwa secara umum hanya sebesar 21,05 persen pengguna MKJP di Indonesia tahun 2017. Selanjutnya, berdasarkan hasil analisis inferensia, diperoleh variabel umur WUS, tingkat pendidikan WUS, umur kawin pertama WUS, Anak Lahir Hidup, akses informasi KB, pengambil keputusan ber-KB, umur suami, dan tingkat pendidikan suami signifikan memengaruhi status penggunaan MKJP pada penelitian ini.

Geopolymer binders are a promising alternative to ordinary Portland cement (OPC) because they can significantly reduce CO2 emissions. However, to apply geopolymer in concrete, it is critical to understand the compatibility between the coarse aggregate and the geopolymer binder. Experimental studies were conducted to explore the effect of the size of the coarse aggregate on the mechanical properties and microstructure of a metakaolin-based geopolymer (MKGP) concrete and ordinary concrete. Three coarse aggregate size grades (5–10 mm, 10–16 mm, and 16–20 mm) were adopted to prepare the specimens. The microstructure of the concretes was investigated with scanning electron microscopy/energy-dispersive X-ray spectroscopy (SEM/EDS) and mercury intrusion porosimetry (MIP). Results showed an opposite coarse aggregate size effect between OPC and MKGP specimens in terms of compressive strength. SEM/EDS analysis indicated that the MKGP concrete has a weaker microstructure compared to OPC concrete induced by a higher porosity. The differences in mechanical properties and pore structure between the MKGP and OPC concrete are attributed to the greatly differing shrinkages triggered by the large surface area and penny-shaped particles of metakaolin. The findings in this work help tailor the mechanical properties and microstructure of MKGP concrete for future engineering applications.

ABSTRACT The mitogen-activated protein kinase (MAPK) phosphatase 1 (MKP-1) is an immediate-early gene comprised of a dual-specificity phosphatase domain and a noncatalytic NH2 terminus. Here, we show that the NH2 terminus of MKP-1, containing the cdc25 homology domains A (CH2A) and B (CH2B), mediates MKP-1 nuclear targeting and modulates MAPK-mediated gene expression. An LXXLL motif which is known to mediate protein-protein interactions with nuclear-targeted hormone receptors was identified proximal to the CH2A domain of MKP-1. The NH2 terminus alone of MKP-1 containing this LXXLL motif was sufficient to direct nuclear targeting, and mutating this motif to LXXAA resulted in the exclusion of MKP-1 from the nucleus. We found that the LXXLL motif proximal to the CH2A domain was present in other nuclear-localized MKPs but was absent in MKPs that localized to the cytoplasm. These data suggest that this LXXLL motif confers nuclear targeting properties to the MKPs. The NH2 terminus of MKP-1 was also found to inhibit the activation of the serum response element (SRE) by preventing MAPK-mediated phosphorylation of the regulatory serine 383 residue on Elk-1. Moreover, we show that MKP-1 plays a major role in the attenuation of serum-induced SRE activity, since MKP-1 null fibroblasts exhibited enhanced SRE activity in response to serum compared with wild-type fibroblasts. The NH2 terminus of MKP-1, when reconstituted into MKP-1 null fibroblasts to levels similar to endogenous MKP-1 following serum stimulation, reduced serum-mediated SRE activity. Collectively, these data reveal novel roles for the NH2 terminus of MKP-1 in nuclear targeting and transcriptional regulation.
