Abstract:
The Drosophila POU gene miti-mere (previously known as pdm2) has a complex spatial and temporal pattern of expression during early development; initially it is expressed in gap-gene-like pattern, then in 14 stripes and finally in a subset of the cells in the developing CNS and PNS. To study the function of this gene during development, we generated a ‘synthetic anti-morphic mutation’ by expressing a truncated version of the miti protein from a constitutive hsp83 and an inducible hsp70 promoter. We show that these delta miti transgenes behave like classical antimorphic mutations. Using these dominant negative transgenes, together with deletions and a duplication for the gene, we show that miti is required during segmentation and neurogenesis. We have also used temperature-shift experiments with the hsp70 delta miti transgene to demonstrate that miti function in segmentation is distinct and separable from its function during neurogenesis. In segmentation, miti appears to be required in the specification of the segments A2 and A6. In the CNS, miti is required for the elaboration of the NB4-2-->GMC-1-->RP2/sib lineage. miti is initially required in this lineage to establish the identity of the parental ganglion mother cell, GMC-1. miti must then be down-regulated to allow the asymmetric division of GMC-1 into the RP2 and its sibling cell.

Besedilo utemeljuje možnost, da bi v sociologiji kulture uporabili Vernantovo koncepcijo mita v antični Grčiji za raziskovanje pojavov v sodobni popularni kulturi, ki jih lahko brez negativnih konotacij obravnavamo kot mite. Marxovo misel, da so miti še vedno gradivo umetnostne imaginacije, kar so bili že za Grke, so v kritičnem družboslovju različno predelovali. Praviloma je mit v sodobnosti pri tem reduciran na ideologijo - takšno je celo Barthesovo branje mitov v popularni kulturi v njegovih Mitologijah. Namesto tega se zavzemamo za branje, ki se navezuje na Vernanta in obravnava kot mite tiste polisemične zgodbe, katerih avtorstvo je nepomembno ali pozabljeno in ki jih je mogoče prenašati iz medija, jim odvzemati ali dodajati podrobnosti ter spreminjati smisel, ne da bi bila njihova vitalnost s tem kaj prizadeta.

Abstract:
An affinity purified trypsin inhibitor from the seed flour extracts ofMadhuca indica(MiTI) on denaturing polyacrylamide gel electrophoresis showed that MiTI consisted of a single polypeptide chain with molecular mass of ~19.8 kDa. MiTI inhibited the total proteolytic and trypsin-like activities of the midgut proteinases ofHelicoverpa armigeralarvae by 87.51% and 76.12%, respectively, at concentration of 5 µg/mL with an IC50of 1.75 µg/mL against trypsin like midgut proteinases. The enzyme kinetic studies demonstrated that MiTI is a competitive inhibitor with aKivalue of4.1×10−10 M forHelicoverpatrypsin like midgut proteinases.In vivoexperiments with different concentrations of MiTI in artificial diet (0.5, 1.0, and 1.5% w/w) showed an effective downfall in the larval body weight and an increase in larval mortality. The concentration of MiTI in the artificial diet to cause 50% mortality (LD50) of larvae was 1.5% w/w and that to cause reduction in mass of larvae by 50% (ED50) was 1.0% w/w. Nutritional indices observations suggest the toxic and adverse effects of MiTI on the growth and development ofH. armigeralarvae. The results suggest a strong bioinsecticidal potential of affinity purified MiTI which can be exploited in insect pest management of crop plants.

Abstract:
Background In 2016, a short message service text messaging intervention to titrate insulin in patients with uncontrolled type 2 diabetes was implemented at two health care facilities in New York City. Objective This study aimed to conduct a qualitative evaluation assessing barriers to and the facilitators of the implementation of the Mobile Insulin Titration Intervention (MITI) program into usual care. Methods We conducted in-depth interviews with 36 patients enrolled in the MITI program and the staff involved in MITI (n=19) in the two health care systems. Interviews were transcribed and iteratively coded by two study investigators, both inductively and deductively using a codebook guided by the Consolidated Framework for Implementation Research. Results Multiple facilitator themes emerged: (1) MITI had strong relative advantages to in-person titration, including its convenience and time-saving design, (2) the free cost of MITI was important to the patients, (3) MITI was easy to use and the patients were confident in their ability to use it, (4) MITI was compatible with the patients’ home routines and clinic workflow, (5) the patients and staff perceived MITI to have value beyond insulin titration by reminding and motivating the patients to engage in healthy behaviors and providing a source of patient support, and (6) implementation in clinics was made easy by having a strong implementation climate, communication networks to spread information about MITI, and a strong program champion. The barriers identified included the following: (1) language limitations, (2) initial nurse concerns about the scope of practice changes required to deliver MITI, (3) initial provider knowledge gaps about the program, and (4) provider perceptions that MITI might not be appropriate for some patients (eg, older or not tech-savvy). There was also a theme that emerged during the patient and staff interviews of an unmet need for long-term additional diabetes management support among this population, specifically diet, nutrition, and exercise support. Conclusions The patients and staff were overwhelmingly supportive of MITI and believed that it had many benefits and that it was compatible with the clinic workflow and patients’ lives. Initial implementation efforts should address staff training and nurse concerns. Future research should explore options for integrating additional diabetes support for patients.

O učenju tujih jezikov obstajajo določeni miti, nepreverjene »resnice«, ki so se ponekod »fosilizirali« in morda sploh več ne vzbujajo nobenih dvomov med običajnimi uporabniki jezikov. V članku osvetlimo izbor štirih mitov o učenju drugega/tujega jezika. V prvem mitu se sprašujemo, kakšno vlogo ima starost pri učenju drugega/tujega jezika. V drugem mitu se sprašujemo, če večina napak v drugem/tujem jeziku res nastane zaradi prenosa (ne)znanja iz prvega jezika v drugi/ tuji jezik oziroma transferja. S tretjim mitom preverjamo, ali naj res učitelj vsako napako popravi takoj z namenom, da si je učenec ne bo zapomnil. V četrtem mitu pa nas zanima, ali ima branje res največji vpliv na širjenje besedišča tako v prvem kot drugem/tujem jeziku. Miti o učenju tujega jezika so pogosto kompleksni, kar velja tudi za mite, predstavljene v tem članku. Glede njihove resničnosti ugotavljamo, da najverjetneje najmanj drži mit o nujnosti takojšnjega popravljanja napak, precej bolj pa prvi, drugi in četrti mit.

Seguendo l'ipotesi che la psicoanalisi sia una potente e preziosa mitologia contemporanea l'A. ripercorre le lotte fra generazioni dell'antica cosmogonia greca, che possono illuminare aspetti sottesi della vicenda edipica. I miti hanno una molteplicità di versioni, che sono tutte vere e tutte false, e la loro polisemia fa sì che, come nei sogni e nell'inconscio, allestiscano uno spazio nel quale può tran-sitare la follia. Nell'infanzia dell'A. il padre era sempre pronto ad ascoltarla e aveva una risposta per tutte le sue domande, tranne che sulla malattia del nonno, che periodicamente doveva essere ricoverato per crisi di psicosi maniaco depressiva. La psicoanalisi come il mito non esclude la follia, e riconosce per prima l'umanità profonda e perturbante di sofferenze che sono state sistematicamente rimosse dal senso comune e dalle istituzioni psichiatriche. Se mantiene come Freud lo sguardo sui sintomi nevrotici e sui deliri psicotici la psicoanalisi non può diventare un sistema compiuto e univoco, ma questo che sembra uno svan-taggio è invece la potenza della psicoanalisi, che è e resta una scienza delle do-mande, non delle risposte: sono le domande che aprono lo spazio metamorfico. Le analogie con la geometria non euclidea e la fisica quantistica sono importanti, ma è solo con le risorse originali della sua storia, della sua teoria e della sua clinica che la psicoanalisi potrà riconoscersi ed essere riconosciuta come scienza.

Abstract:
We have investigated (i) the role of pdm1, a Drosophila POU gene, during the elaboration of the GMC-1-->RP2/sib lineage and (ii) the functional relationship between pdm1 and the closely linked second POU gene, miti-mere, in this lineage. We show that deletion of pdm1 causes a partially penetrant GMC-1 defect, while deletion of both miti and pdm1 results in a fully penetrant defect. This GMC-1 defect in miti- and pdm1- embryos can be rescued by the pdm1 or miti transgene. Rescue is observed only when these genes are expressed at the time of GMC-1 formation. Overexpression of pdm1 or miti well after GMC-1 is formed results in the duplication of RP2 and/or sib cells. Our results indicate that both genes are required for the normal development of this lineage and that the two collaborate during the specification of GMC-1 identity.

Abstract:
This article describes the coding portion of a study to test the effectiveness of a motivational interviewing (MI) training program for probation officers. We describe some of the challenges with using the Motivational Interview Treatment Integrity (MITI) instrument to code interactions between probation officers and clients. Our team of raters was able to obtain adequate inter-rater reliability on most MITI scales, though reliability ratings on some of the specific behavior counts such as Giving Information, MI Adherent, and MI Non-adherent fell considerably lower than the original MITI norming study. Our results suggest that the MITI is a mostly reliable instrument for coding criminal justice interactions, though there were exceptions to this rule. Based on our experiences, we discuss some of the ways that probation interactions might be different from traditional counseling interactions, and identify some rules of thumb that helped us to code interactions. We end with suggestions for how MITI feedback can be used effectively in training and supervision in criminal justice and other non-traditional settings.

AbstractEfficient exogenous DNA integration can be mediated by Cas9 through the non-homology end-joining pathway. However, such integrations are often imprecise and contain a variety of mutations at the junctions between the external DNA and the genomic loci. Here we describe a microhomology-dependent targeted integration method, designated MITI, for precise site-specific gene insertions. We found that the MITI strategy yielded higher knock-in accuracy than Cas9 HITI for the insertion of external DNA and tagging endogenous genes. Furthermore, in combination with negative selection and four different CrRNAs targeting donor vectors and genome-targeted sites with a CrRNA array, MITI facilitated precise ligation at all junctions. Therefore, our Cas12a-based MITI method increases the repertoire of precision genome engineering approaches and provides a useful tool for various gene editing applications.

Abstract:
L'autore parte dalla constatazione che le ricerche sul ruolo dell'industria bellica italiana durante la Repubblica sociale italiana hanno a lungo riproposto i miti nati negli anni di guerra piuttosto che offrire dati sulla produzione sui quali riflettere. Molte affermazioni hanno ricalcato le memorie difensive degli industriali, interessati dopo il 1945 a svalutare il proprio contributo allo sforzo bellico del Terzo Reich e favoriti nella loro ricostruzione dall'indisponibilitŕ di fonti specifiche. Il tema viene cosě affrontato sulla base di una vasta ed eterogenea documentazione, utilizzata per disegnare il complesso mosaico del contributo che piccole, medie e grandi aziende italiane diedero alle forze armate tedesche, il cui obiettivo fu anzitutto rendere il Gruppo di armate B il piů possibile autonomo rispetto all'industria tedesca. L'autore ricostruisce i complessi rapporti tra gli occupanti, la Rsi e le industrie italiane, mettendo in rilievo l'importanza della mobilitazione dell'industria nazionale che, accanto a produzioni obsolete e volte a impiegare le maestranze per frenarne il malcontento, sviluppň, in sedi decentrate e coperte dal segreto, la produzione di componenti per i mezzi bellici piů avanzati di cui furono dotate le forze armate tedesche.
