Bacterial MinD and MinE form a standing oscillatory wave which positions the cell division inhibitor MinC, that binds MinD, everywhere on the membrane except at the midpoint of the cell, ensuring midcell positioning of the cytokinetic septum. During this process MinE undergoes fold switching as it interacts with different partners. We explore the exchange dynamics between major and excited states of the MinE dimer in 3 forms using15N relaxation dispersion NMR: the full-length protein (6-stranded β-sheet sandwiched between 4 helices) representing the resting state; a 10-residue N-terminal deletion (Δ10) mimicking the membrane-binding competent state where the N-terminal helix is detached to interact with membrane; and N-terminal deletions of either 30 (Δ30) or 10 residues with an I24N mutation (Δ10/I24N), in which the β1-strands at the dimer interface are extruded and available to bind MinD, leaving behind a 4-stranded β-sheet. Full-length MinE samples 2 “excited” states: The first is similar to a full-length/Δ10 heterodimer; the second, also sampled by Δ10, is either similar to or well along the pathway toward the 4-stranded β-sheet form. Both Δ30 and Δ10/I24N sample 2 excited species: The first may involve destabilization of the β3- and β3′-strands at the dimer interface; changes in the second are more extensive, involving further disruption of secondary structure, possibly representing an ensemble of states on the pathway toward restoration of the resting state. The quantitative information on MinE conformational dynamics involving these excited states is crucial for understanding the oscillation pattern self-organization by MinD–MinE interaction dynamics on the membrane.

A Rede Saúde e Cultura publicou no ano de 2015 o segundo volume do Retratos da Interface Cultura e Saúde no Brasil, Experiências Premiadas e Selecionadas pelos editais Prêmio Cultura e Saúde 2008 e 2010, através da Fundação Oswaldo Cruz ‑ Fiocruz com o apoio do Ministério da Cultura – MINC e do Ministério da Saúde – MS. Com o objetivo de construir conhecimento sobre a apropriação social dos Prêmios Cultura e Saúde 2008 e 2010, a publicação analisou 149 destas experiências, identificando o perfil dos atores, a natureza das práticas que materializam na ponta a articulação saúde e cultura, os públicos para quem estas se voltam, as linguagens utilizadas e os parceiros no território.A obra começa apresentando o percurso da articulação interministerial a partir das iniciativas que antecederam o acordo de cooperação em 2010, entre a Fiocruz/MS e o MINC, que implementou o projeto “Rede Saúde e Cultura: promovendo inclusão e qualidade de vida”. Contextualizando conceitualmente a intrínseca relação entre os setores da saúde e da cultura relação através dos marcos conceituais da Reforma Sanitária Brasileira e da Convenção Internacional sobre Diversidade Cultural.A partir dos objetivos e resultados obtidos no primeiro volume, publicado em 2014, cujo foco da análise foi o perfil dos proponentes dos projetos selecionados e premiados, a distribuição regional da premiação, evidenciando os aspectos das formas de acesso a estes editais, o estudo realizado no segundo volume teve como foco a natureza das experiências selecionadas. Foi utilizado como fonte documental o conjunto de formulários preenchidos pelos proponentes selecionados ou premiados em 2008 e 2010. O objeto da análise foram as respostas do proponente às questões abertas do formulário que tratavam da descrição da atividade, do público a ser atendido, dos aspectos inovadores e do impacto da iniciativa na condição de saúde da comunidade.A publicação traz de forma detalhada, com tabelas explicativas, a metodologia utilizada para classificar e categorizar as finalidades, os temas, os grupos populacionais, as fases da vida, as práticas culturais, as parcerias desenvolvidas, dentre outras categorias criadas a partir do vasto material que as respostas do formulário trouxeram.Os resultados da análise dos dois editais são apresentados de forma concomitante, organizados a partir de perguntas geradoras como: Onde estão localizados os proponentes? O que foi proposto? Há variação do foco da ação segundo a região do país? Qual a natureza do foco das ações segundo as instituições proponentes? Para quem foram direcionadas as propostas? Que parcerias foram estabelecidas? No projeto foram realizados relatos fotográficos e reflexões sobre as memórias coletivas e individuais de ex-lavadeiras, a partir de duas oficinas de fotografias que [re] estabeleceram as relações de afeto e vínculo com trajetos, histórias e vivências diante das mudanças e permanências do Rio Sabará. As oficinas produzidas urdiram narrativas por meio do registro fotográfico de locais de memória criando mapas afetivos para a mediação de uma educação das sensibilidades e de patrimônios.

Abstract:
Traditional cell culture conditions entail the use of gas atmosphere consisting of 5–6% CO2 in air. These same conditions have also been used universally for the culture of human embryonic stem cells (hES), despite the natural milieu of the embryos, from which these cells are derived, being slightly hypoxic. The aim of this work was to examine if human embryonic stem cells would benefit from the reduced oxygen culture environment, as used for human pre-implantation embryos. In Experiment 1, the relative growth in high and low oxygen atmosphere was compared by cutting undifferentiared hES-colonies into equal-size pieces and transferring them to two dishes, one into the conventional incubator in 6% CO2 in air (HIGH), the other into K-MINC-1000 (Eight Mile Plains, Queensland) mini-incubator in 5% O2, 6% CO2 and 89% N2 (LOW). After 8 days the colony sizes and differentiation status were measured. In Experiment 2, the absolute growth in high and low oxygen atmosphere was compared by cutting exactly same size fragments from undifferentiated hES-colonies and distributing them to two treatments as described. All colonies were measured and passaged at Day 7 and final measurements taken at Day 14. In Experiment 1, improved growth was observed in reduced oxygen, mean percentage of undifferentiated growth from original colony (assigned as 100%) being 217% for HIGH and 482% for LOW over three replicates. In Experiment 2, the total of 16.9 mm2 of undifferentiated colonies in both treatment groups (24 colonies per group over 3 replicates) had by Day 7 grown to total of 51.1 and 79.5 mm2 (P < 0.001), and by Day 14 to 216.8 and 373.3 mm2 (P < 0.0001) in HIGH and LOW, respectively (see Table 1). In neither experiment were there differences in the differentiation status of the colonies between the treatments (mean 6% and 5% at Day 7, and 9% and 9% at Day 14 in HIGH and LOW, respectively). We conclude that culture in reduced oxygen improves growth of human embryonic stem cells. Table 1. e13056 Background: Invasive lobular carcinoma (ILC) accounts for only 10-15% of all invasive breast cancers but has distinct clinicopathologic characteristics and genomic profiles. In particular, metastatic lobular cancers (mILC) have unique sites of metastasis and it is unclear if the response to initial endocrine therapy differs from metastatic ductal cancers (mIDC). Therefore we have undertaken a single-institution, retrospective analysis to compare overall outcomes and response to initial endocrine therapy (ET) in patients (pts) with metastatic ILC and IDC. Methods: An IRB approved retrospective review of medical records was conducted evaluating pts treated for metastatic IDC and ILC at The Ohio State University Comprehensive Cancer Center from January 1, 2004 to December 31, 2014. Pts diagnosed with mIDC were matched on age, year of diagnosis, estrogen receptor/progesterone receptor and HER2 status and site of metastasis 2:1 to patients diagnosed with mILC. Overall survival (OS) was defined as the time from metastasis to death or last known follow-up. Progression-free survival (PFS) was defined as time from metastasis to progression on first-line ET. Time to chemotherapy (TTC) was defined as time from starting ET for metastasis to initiation of chemotherapy. Kaplan Meier (KM) methods were used to calculate median OS, PFS and TTC. Results: A total of one hundred sixty one pts with metastatic breast cancer were included in this analysis. The demographic features between the two groups were well balanced and included in the table below. The median OS was 2.6 yrs (95% CI: 1.55, 3.22) for mILC and 2.2 yrs (95% CI: 1.95, 2.62) for mIDC. A subset of 111 patients who started on endocrine therapy were used in the PFS and TTC analyses. The median PFS following first-line ET was 2.2 yrs (95% CI: 0.1.0, 2.7) for mILC and 1.4 yrs (95% CI: 0.91, 1.90) for mIDC. Median TTC was 2.1 yrs (95% CI: 1.71, 4.92) for mILC and 2.4 yrs (95% CI: 1.90, 4.77) for mIDC. There was no statistically significant difference in outcomes between the two groups. Conclusions: Outcomes in patients with ILC and IDC have been varied, with several studies reporting that patients with ILC have worse outcomes and response to chemotherapy. Our retrospective study examining outcomes in mILC in comparison with mIDC showed no difference in OS. Given the concern of resistance to conventional therapies in patients with lobular cancers, it is reassuring to see that the median PFS on first line ET and TTC was similar to metastatic ductal cancers.[Table: see text]

Abstract Edward Halper’s “The Metaphysics of the Syllogism” argues that the ontological ground of valid inference is found in the necessity of the predications that constitute the premises of the sort of syllogism central to Aristotle’s theory: demonstration. I further support his conclusion on the basis of a consideration of the title and structure of Aristotle’s Analytics, as well as some recent analysis of Aristotle’s modal logic. Halper however suggests that the logical form of inference is a result of how the mind sorts out the elements involved in a complex unity. I suggest that it is not primarily the mind that does this work, but language. What the mind does is primarily to be understood as a reflection of what language does, not vice versa.

Rapid technology advancements, especially in the past decade, have allowed off-the-shelf unmanned aerial vehicles (UAVs) that weigh less than 250 g to become available for recreational use by the general population. Many well-known manufacturers (e.g., DJI) are now focusing on this segment of UAVs, and the new DJI Mini 2 drone is one of many that falls under this category, which enables easy access to be purchased and used without any Part 107 certification and Remote ID registration. The versatility of drones and drone models is appealing for customers, but they pose many challenges to forensic tools and digital forensics investigators due to numerous hardware and software variations. In addition, different devices can be associated and used for controlling these drones (e.g., Android and iOS smartphones). Moreover, according to the Federal Aviation Administration (FAA), the adoption of Remote ID is not going to be required for people without the 107 certifications for this segment at least until 2023, which creates finding personally identifiable information a necessity in these types of investigations. In this research, we conducted a comprehensive investigation of DJI Mini 2 and its data stored across multiple devices (e.g., SD cards and mobile devices) that are associated with the drone. The aim of this paper is to (1) create several criminal-like scenarios, (2) acquire and analyze the created scenarios using leading forensics software (e.g., Cellebrite and Magnet Axiom) that are commonly used by law enforcement agencies, (3) and present findings associated with potential criminal activities.

Abstract:
Penelitian ini bertujuan untuk mengetahui penerapan metode pembelajaran mind mapping di sekolah, untuk mengetahui kesulitan penerapan metode pembelajaran mind mapping di sekolah, untuk mengetahui kelebihan penerapan metode pembelajaran mind mapping di sekolah, untuk mengetahui kekurangan penerapan metode pembelajaran mind mapping disekolah. Jenis penelitian ini adalah penelitian kualitatif. Subyek dalam penelitian ini adalah guru kelas tinggi diSDN 2 Wakul dan SDN Gerintuk. Metode pengumpulan data yaitu wawancara dan dokumentasi. Wawancara digunakan untuk memperoleh data yang berkaitan dengan penerapan metode pembelajaran mind mapping, Sedangkan dokumentasi digunakan untuk menunjangbukti dari penelitian. Data yang diperoleh kemudian di analisis bagaimana pelaksanaan penerapan metode pembelajaran mind mapping, kesulitan penerapan pelaksanaan metode pembelajaran mind mapping, kelebihan penerapan metode pembelajaran mind mapping, kekurangan penerapan metode pembelajaran mind mapping.Maka dari data yang di dapatkan, dapat disimpulkan bahwa metodepembelajaran mind mapping baik digunakan di sekolah.
