Abstract:
Bacterial MinD and MinE form a standing oscillatory wave which positions the cell division inhibitor MinC, that binds MinD, everywhere on the membrane except at the midpoint of the cell, ensuring midcell positioning of the cytokinetic septum. During this process MinE undergoes fold switching as it interacts with different partners. We explore the exchange dynamics between major and excited states of the MinE dimer in 3 forms using15N relaxation dispersion NMR: the full-length protein (6-stranded β-sheet sandwiched between 4 helices) representing the resting state; a 10-residue N-terminal deletion (Δ10) mimicking the membrane-binding competent state where the N-terminal helix is detached to interact with membrane; and N-terminal deletions of either 30 (Δ30) or 10 residues with an I24N mutation (Δ10/I24N), in which the β1-strands at the dimer interface are extruded and available to bind MinD, leaving behind a 4-stranded β-sheet. Full-length MinE samples 2 “excited” states: The first is similar to a full-length/Δ10 heterodimer; the second, also sampled by Δ10, is either similar to or well along the pathway toward the 4-stranded β-sheet form. Both Δ30 and Δ10/I24N sample 2 excited species: The first may involve destabilization of the β3- and β3′-strands at the dimer interface; changes in the second are more extensive, involving further disruption of secondary structure, possibly representing an ensemble of states on the pathway toward restoration of the resting state. The quantitative information on MinE conformational dynamics involving these excited states is crucial for understanding the oscillation pattern self-organization by MinD–MinE interaction dynamics on the membrane.

Abstract:

Abstract:
Traditional cell culture conditions entail the use of gas atmosphere consisting of 5–6% CO2 in air. These same conditions have also been used universally for the culture of human embryonic stem cells (hES), despite the natural milieu of the embryos, from which these cells are derived, being slightly hypoxic. The aim of this work was to examine if human embryonic stem cells would benefit from the reduced oxygen culture environment, as used for human pre-implantation embryos. In Experiment 1, the relative growth in high and low oxygen atmosphere was compared by cutting undifferentiared hES-colonies into equal-size pieces and transferring them to two dishes, one into the conventional incubator in 6% CO2 in air (HIGH), the other into K-MINC-1000 (Eight Mile Plains, Queensland) mini-incubator in 5% O2, 6% CO2 and 89% N2 (LOW). After 8 days the colony sizes and differentiation status were measured. In Experiment 2, the absolute growth in high and low oxygen atmosphere was compared by cutting exactly same size fragments from undifferentiated hES-colonies and distributing them to two treatments as described. All colonies were measured and passaged at Day 7 and final measurements taken at Day 14. In Experiment 1, improved growth was observed in reduced oxygen, mean percentage of undifferentiated growth from original colony (assigned as 100%) being 217% for HIGH and 482% for LOW over three replicates. In Experiment 2, the total of 16.9 mm2 of undifferentiated colonies in both treatment groups (24 colonies per group over 3 replicates) had by Day 7 grown to total of 51.1 and 79.5 mm2 (P < 0.001), and by Day 14 to 216.8 and 373.3 mm2 (P < 0.0001) in HIGH and LOW, respectively (see Table 1). In neither experiment were there differences in the differentiation status of the colonies between the treatments (mean 6% and 5% at Day 7, and 9% and 9% at Day 14 in HIGH and LOW, respectively). We conclude that culture in reduced oxygen improves growth of human embryonic stem cells. Table 1. Abstract:
e13056 Background: Invasive lobular carcinoma (ILC) accounts for only 10-15% of all invasive breast cancers but has distinct clinicopathologic characteristics and genomic profiles. In particular, metastatic lobular cancers (mILC) have unique sites of metastasis and it is unclear if the response to initial endocrine therapy differs from metastatic ductal cancers (mIDC). Therefore we have undertaken a single-institution, retrospective analysis to compare overall outcomes and response to initial endocrine therapy (ET) in patients (pts) with metastatic ILC and IDC. Methods: An IRB approved retrospective review of medical records was conducted evaluating pts treated for metastatic IDC and ILC at The Ohio State University Comprehensive Cancer Center from January 1, 2004 to December 31, 2014. Pts diagnosed with mIDC were matched on age, year of diagnosis, estrogen receptor/progesterone receptor and HER2 status and site of metastasis 2:1 to patients diagnosed with mILC. Overall survival (OS) was defined as the time from metastasis to death or last known follow-up. Progression-free survival (PFS) was defined as time from metastasis to progression on first-line ET. Time to chemotherapy (TTC) was defined as time from starting ET for metastasis to initiation of chemotherapy. Kaplan Meier (KM) methods were used to calculate median OS, PFS and TTC. Results: A total of one hundred sixty one pts with metastatic breast cancer were included in this analysis. The demographic features between the two groups were well balanced and included in the table below. The median OS was 2.6 yrs (95% CI: 1.55, 3.22) for mILC and 2.2 yrs (95% CI: 1.95, 2.62) for mIDC. A subset of 111 patients who started on endocrine therapy were used in the PFS and TTC analyses. The median PFS following first-line ET was 2.2 yrs (95% CI: 0.1.0, 2.7) for mILC and 1.4 yrs (95% CI: 0.91, 1.90) for mIDC. Median TTC was 2.1 yrs (95% CI: 1.71, 4.92) for mILC and 2.4 yrs (95% CI: 1.90, 4.77) for mIDC. There was no statistically significant difference in outcomes between the two groups. Conclusions: Outcomes in patients with ILC and IDC have been varied, with several studies reporting that patients with ILC have worse outcomes and response to chemotherapy. Our retrospective study examining outcomes in mILC in comparison with mIDC showed no difference in OS. Given the concern of resistance to conventional therapies in patients with lobular cancers, it is reassuring to see that the median PFS on first line ET and TTC was similar to metastatic ductal cancers.[Table: see text]

Abstract:
The study presents a review of in-service models and a search for an effective in-service model for mathematics education in the primary school. The study considers the need to improve the teaching of mathematics in the primary school; the major reports that have made recommendations for the improvement of primary mathematics teaching and the opinions and findings of mathematics educators. The study examines the state of the art of in-service models and examines those models previously used. The study notes the failings of the in-service models past and present and examines elements within these models which have proven to be beneficial albeit in part. Throughout the study particular attention is given to recent studies concerned with implementing and sustaining change. The model designed for the in-service of the primary mathematics teacher, MINC (Mathematics In-service Network Course) is based on an examination of models past and present. Components and provisions that have met with success in these models were included in the design. The in-service model developed and described in this study includes four major components input from course leaders and teachers, reflection, feedback and the opportunity for teachers to work with others from the same school and neighbouring schools (networking). A major section of the study examines the responses of participants to an evaluation questionnaire given at the completion of the first in-service course, MINC 1 and the second, MINC 2. Data collected from the responses of participants to the questionnaire were analysed to see if there were differences in responses from teachers based on such factors as sex, the MINC network, the level to which participants had studied mathematics, the year level they taught and expressed confidence in teaching mathematics. In conclusion, the study reflects on the evaluation of the in-service course and addresses considerations for future in-service courses concerned with mathematics education for primary teachers.

Abstract:
A cut is a partition of a graph's nodes into two sets, and we say that an edge crosses the cut if it connects two nodes belonging to different sets. A maximum cut is a cut that maximises the number of crossing edges. We show that for any sufficiently small ε > 0 it is NP-hard to distinguish between graphs for which at least a fraction 1 - ε of all edges crosses the maximum cut and graphs for which at most a fraction 1 - 1.4568 ε of all edges crosses the maximum cut. The previous state of the art had a constant smaller than 1.375 in place of 1.4568.

Abstract:
I report 1684 new proper motion systems in the southern sky (declinations -90 degrees to -47 degrees) with 0.50 arcsec/yr > mu >= 0.18 arcsec/yr. This effort is a continuation of the SuperCOSMOS-RECONS (SCR) proper motion search to lower proper motions than reported in Hambly et al. (2004); Henry et al. (2004); Subasavage et al. (2005a,b). Distance estimates are presented for the new systems, assuming that all stars are on the main sequence. I find that 34 systems are within 25 pc, including three systems --- SCR 0838-5855, SCR 1826-6542, and SCR 0630-7643AB --- anticipated to be within 10 pc. These mini-motion (MINIMO) discoveries constitute a more than ten-fold increase in new systems found in the same region of sky searched for systems with mu >= 0.50 arcsec/yr, suggesting a happy hunting ground for new nearby slower proper motion systems in the region just north (declinations -47 degrees to 0 degrees), much of which has not been rigorously searched during previous efforts.

Full text
Abstract:
The Fully Encapsulated Resin Bolt (FERB) is widely utilised for strata control and ground support in civil and mining applications worldwide, with approximately 6 million installed per annum by the Australian coal mining industry. Independent studies have concluded that 30-35% of these rockbolts, which represents an annual expenditure of $40 million, are ineffective. The anchorage and failure mechanisms of FERB are yet to be quantified, and support systems are designed primarily from empirical rather than scientific methods. There are no standardised methods of assessing FERB components, installation techniques and support behaviour. The majority of research into FERB support systems remains commercial intellectual property, with little information released into the public domain. This thesis investigated several variables of FERB support systems, and also examined differences between field and laboratory pull-out test load distributions. This research was conducted in two phases, with Phase 1 seeking standardised methodology and repeatability in results, while Phase 2 further refined Phase 1 methods and extended the range of tests. The results in both phases were encouraging, with reasonable repeatability attained in all testing series. The findings included: ??? Annulus Thickness: There was little change in load capacity with small annulus thickness, however the maximum peak load (MPL) significantly reduced once annulus thickness exceeded 4mm ??? Resin Installation Spin Time: Underspinning of cartridge resin was found to have an insignificant effect on rockbolt load/deformation characteristics. Overspinning, however, led to a dramatic reduction in anchorage performance with a lessening in both MPL and stiffness ??? Rockbolt Load Transfer: The magnitude of an applied load reduced to zero along the length of the rockbolt, being greatest nearest the rock free surface (the point of load application). An exponential reduction was found when tested in the manner of laboratory tests, with the loading jack reacting against the free surface. This reduction was linear when the load was applied as in the field, with no load placed on the free surface This basic investigation into FERB support systems has validated many empirical understandings of rockbolts, while highlighting the need for further testing into several key areas.

Abstract:

Abstract:

Beck, Aaron. RiverOceanPlastic: Land-ocean transfer of plastic debris in the North Atlantic, Cruise No. AL534/2, 05 March – 26 March 2020, Malaga (Spain) – Kiel (Germany). GEOMAR Helmholtz Centre for Ocean Research Kiel, 2020. http://dx.doi.org/10.3289/cr_al534-2.

Full text
Abstract:
Cruise AL534/2 is part of a multi-disciplinary research initiative as part of the JPI Oceans project HOTMIC and sought to investigate the origin, transport and fate of plastic debris from estuaries to the oceanic garbage patches. The main focus of the cruise was on the horizontal transfer of plastic debris from major European rivers into shelf regions and on the processes that mediate this transport. Stations were originally chosen to target the outflows of major European rivers along the western Europe coast between Malaga (Spain) and Kiel (Germany), although some modifications were made in response to inclement weather. In total, 16 stations were sampled along the cruise track. The sampling scheme was similar for most stations, and included: 1) a CTD cast to collect water column salinity and temperature profiles, and discrete samples between surface and seafloor, 2) sediment sampling with Van Veen grab and mini-multi corer (mini-MUC), 3) suspended particle and plankton sampling using a towed Bongo net and vertical WP3 net, and 4) surface neusten sampling using a catamaran trawl. At a subset of stations with deep water, suspended particles were collected using in situ pumps deployed on a cable. During transit between stations, surface water samples were collected from the ship’s underway seawater supply, and during calm weather, floating litter was counted by visual survey teams. The samples and data collected on cruise AL534/2 will be used to determine the: (1) abundance of plastic debris in surface waters, as well as the composition of polymer types, originating in major European estuaries and transported through coastal waters, (2) abundance and composition of microplastics (MP) in the water column at different depths from the sea surface to the seafloor including the sediment, (3) abundance and composition of plastic debris in pelagic and benthic organisms (invertebrates), (4) abundance and identity of biofoulers (bacteria, protozoans and metazoans) on the surface of plastic debris from different water depths, (5) identification of chemical compounds (“additives”) in the plastic debris and in water samples.
